Fascioliasis is a zoonotic disease caused by Fasciola hepatica or F. gigantica. There are two phases of Fasciola infection. In the acute phase, the patient has a wide spectrum of clinical manifestations while the later phase of infection is usually related to inflammatory changes in the bile duct and the mechanical effects of the worm, which can lead to biliary obstruction. The presence of Fasciola inside the biliary system has rarely been reported in the literature. In this report, we presented a case of bile duct obstruction in a 36-year-old male patient undergoing cholecystectomy for gallstones and with a history of worsening jaundice for one month. Extensive laboratory testing failed to clarify the cause of jaundice. Three living liver flukes were subsequently retrieved from the right hepatic duct and later confirmed as F. gigantica by morphology and internal transcribed spacer (ITS2) sequence analysis.

Fasciolosis is one of the most important causes of reduction in milk production, growth and fertility in cattle and causes considerable economic losses generated by liver seizures in slaughterhouses. The objective of this study was to determine the occurrence of livers seized byFasciola hepaticain the Lorenzo Gonzales bovine slaughterhouse in Sagua la Grande municipality in Villa Clara province, Cuba. A retrospective observational analysis of the seizures of livers by F. hepatica in the period 2016-2020 was carried out. The prevalence, seasonality, trend and enzootic channels of the occurrence of livers seizures due to fasciolosis in slaughtered cattle were determined. In addition, the annual economic losses as a consequence of fasciolosis seizures were estimated. The sanitary impact of seizures due to fasciolosis showed a prevalence of 18.93%, which is a reflection of the epizootic situation of the units of origin. Losses due to liver seizures as a consequence of F. hepatica infestation amounted to 1 088 532 cup in the five-year period evaluated. The trend in the incidence of F. hepatica seizures is decreasing and shows a marked seasonal pattern in the months of March, April, May, June, October, November and December. The usual expected behavior of F. hepatica seizures of livers in the Sagua bovine slaughterhouse for a period of five years shows a wide range of monthly occurrences.

Protein phosphorylation plays key roles in a variety of essential cellular processes. Fasciola gigantica is a tropical liver fluke causing hepatobiliary disease fascioliasis, leading to human health threats and heavy economic losses. Although the genome and protein kinases of F. gigantica provided new insights to understand the molecular biology and etiology of this parasite, there is scant knowledge of protein phosphorylation events in F. gigantica. In this study, we characterized the global phosphoproteomics of adult F. gigantica by phosphopeptide enrichment-based LC–MS/MS, a high-throughput analysis to maximize the detection of a large repertoire of phosphoproteins and phosphosites. A total of 1030 phosphopeptides with 1244 phosphosites representing 635 F. gigantica phosphoproteins were identified. The phosphoproteins were involved in a wide variety of biological processes including cellular, metabolic, and single-organism processes. Meanwhile, these proteins were found predominantly in cellular components like membranes and organelles with molecular functions of binding (51.3%) and catalytic activity (40.6%). The KEGG annotation inferred that the most enriched pathways of the phosphoproteins included tight junction, spliceosome, and RNA transport (each one contains 15 identified proteins). Combining the reports in other protozoa and helminths, the phosphoproteins identified in this work play roles in metabolic regulation and signal transduction. To our knowledge, this work performed the first global phosphoproteomics analysis of adult F. gigantica, which provides valuable information for development of intervention strategies for fascioliasis. Significant disease burden is caused by infections with human liver flukes, including Clonorchis sinensis, Opisthorchis viverrini and O. felineus. Epidemiology is characterized by high burden in the male than the female, and in the elder than the younger. Chemotherapy is the mainstream for morbidity control, but it could not prevent re-infection. Meanwhile, behavioral change on the dietary habit of ingesting raw freshwater fish is challenging. In this opinion, we argue why it is important to educate children for sustainable control of liver fluke infections. Then, the design, development and key messages of an educational cartoon for preventing liver fluke infections is introduced. Technical focuses are emphasized based on a pilot using the education cartoon for preventing clonorchiasis in China. Finally, how to further develop and verify this education strategy is discussed. The aim of the present study was to determine the effective dose of fosfatriclaben through a field study in sheep naturally infected with F. hepatica. Thirty crossbred sheep positive for fluke eggs were selected for inclusion in the trial. On day 0, 5 groups of 6 animals each were formed for treatments: group 1 (G1), G2, and G3 received fosfatriclaben at 4, 6, and 8 mg/kg/IM, respectively. G4 received triclabendazole at 10 mg/kg/PO, and G5 was the untreated control group. Fecal samples of the sheep were analyzed to count the number of fluke eggs to evaluate the percentage of egg reduction. Twenty-one days after treatment, all sheep were humanely euthanized to extract the flukes from the bile ducts. They were counted to assess the percentage of fluke reduction. Fosfatriclaben reduced fluke eggs by 99.6, 99.6, and 100% and flukes by 94.3, 100, and 100%, respectively. Triclabendazole reduced fluke eggs by 95.2% and flukes by 100%. The fosfatriclaben injectable prodrug showed a high fasciolicidal efficacy similar to triclabendazole, with advantages over its predecessor, since only half the dose as compared to triclabendazole was required to remove eggs and flukes in the sheep that were studied.

A retrospective study was carried out from registers of the slaughterhouses of Constantine, Algeria, on 145,919 cattle and 345,282 sheep apparently healthy, slaughtered between 2009 and 2018. The objective was to estimate the prevalence of the two dominant parasitic infections which cause gross lesions in liver and lungs, i.e. cystic echinococcosis (CE) and Fasciola hepatica infection. The postmortem examination consisted in a visual inspection of the organs, palpation of lungs and livers, and systematic incision of livers, performed according to FAO procedures. The prevalence of F. hepatica infection was significantly higher in cattle (2.7%) than in sheep (0.2%). There was a non-significant decrease from 4.5 to 2.0% (p > 0.05) in the prevalence of F. hepatica in cattle during the 10-year period. The prevalence of CE in the livers and lungs was significantly higher in cattle (3.3% and 9.7%, respectively) than in sheep (0.6% and 1.1%, respectively). The prevalence of CE was significantly higher in lungs than in livers. These results showed the importance of these parasitic infections in the province of Constantine and its surroundings, which requires the implementation of control programs against these infections in both sheep and cattle.

Clonorchis sinensis, a liver fluke parasite, infects humans through ingestion of raw or undercooked fish, crabs, or crayfish in endemic areas where the parasite is found. Clonorchis sinensis infects the liver, gallbladder, and bile duct in humans, causing Clonorichiasis. Although the majority of patients are asymptomatic, long-lasting infections may cause severe disease. Without treatment, human infection may persist for the parasite's lifespan (25-30 years). Pathologic diagnosis can be challenging as sampling may demonstrate limited cellularity with minuscule eggs that may be overlooked. Here, we report a rare case of liver fluke eggs diagnosed in bile duct brush cytology.

Calicophoron daubneyi (rumen fluke) is an emerging parasitic infection of livestock across Europe. Despite increasing in prevalence, little is known about the level of awareness of rumen fluke or current control practices used by UK farmers. Fasciola hepatica (liver fluke) is a common parasitic infection of cattle and sheep in the UK. Co-infections with these parasites can present in sheep and cattle, but the only drug with reported efficacy against rumen fluke is oxyclozanide. Between December 2019 and March 2020, 451 sheep and/or cattle farmers completed an online questionnaire, capturing their awareness and current means of control for liver fluke and rumen fluke. Most respondents (70%) were aware of rumen fluke, with 14% recording its presence on their farms and 18% having previously treated for rumen fluke. Almost all respondents (99%) were aware of liver fluke and higher numbers of respondents reported its presence on farm (67%) with 88% having previously treated for liver fluke. Respondents who were aware of rumen fluke said they were concerned about the parasite (81%), although rumen fluke was less of a concern than liver fluke (p 0.05). Of respondents who reported rumen fluke presence on their farm, 42% cited incorrect diagnostic methods, including those traditionally used to detect liver fluke. Respondents were more likely to treat annually for liver fluke, as opposed to rumen fluke (p 0.05). The most frequently used drug for the treatment of liver fluke infection was triclabendazole (53% sheep treatments, 34% cattle treatments) and only a minority of farmers treated with a product effective against rumen fluke (oxyclozanide; 42% cattle treatments, 35% sheep treatments). A small proportion of farmers stated that they used a non-flukicide drug to treat sheep for liver fluke infection (1.6% sheep treatments). These results demonstrate a broad awareness of liver and rumen fluke in sheep and cattle, but reveal confusion amongst farmers about their diagnosis and treatment, highlighting the need to provide best practice advice to the livestock industry for the control of both parasites.

La fasciolosis es una zoonosis provocada por el parásito Fasciola hepatica Linnaeus, 1758, éste para poder desarrollar su ciclo biológico, necesita de un hospedero intermediario (molusco gasterópodo, Lymnaeidae), y un hospedero definitivo (mamíferos, incluyendo al hombre). Esta enfermedad, ha causado pérdidas económicas anuales de $ 200 M en el mundo. El objetivo de la presente investigación fue identificar la prevalencia de F. hepatica en los centros de rastro bovino, determinar las pérdidas económicas, e identificar las características agroecológica en donde se desarrollan los hospederos intermediarios, en la provincia de Imbabura, Ecuador. Se identificó una prevalencia de F. hepatica, de 10,9%, donde el Cantón Otavalo presentó 190 muestras positivas (19,7%), considerándose el punto con mayor prevalencia dentro del estudio. Los bovinos positivos fueron, 102 machos (26,9%) y 277 hembras (73,1%), los cuales pertenecen al biotipo Mestizo (53,5%), Holstein (42%) y Normando (4,5 %), siendo el 78,6% mayores a dos años y 21,4% menores a dos años. Se presentó una pérdida económica anual de $ 69.547,5 dólares americanos. Los 20 biotopos muestreados, se mantuvieron dentro de los valores promedio; distancia del centro poblado 0,7 km, área de 42,8 m2; 31 moluscos por biotopo, pH del suelo 7,1; profundidad del agua 2,3 cm; altitud de 2.019 a 2.772 msnm; temperatura de 20 °C y humedad relativa de 74%. Los moluscos se ubicaron en acequias (55%), charcos (25%), pantanos (10%), riachuelos y el suelo (5%), con presencia de kikuyo (Pennisetum clandestinum Hochst. Ex Chiov, 1903) y totora (Schoenoplectus californicus C. A. Mey., 1850) como especies dominantes. La provincia de Imbabura posee una prevalencia moderada en cuanto a epidemiología por parte de fasciolosis, se confirma que estas zonas presentan un ecosistema apto para el desarrollo del ciclo biológico de la fasciolosis.

Fascioliasis is a zoonotic disease caused by Fasciola hepatica that infects mainly cattle, sheep, and goats. Humans can be infected by water or aquatic plants contaminated with metacercariae. The authors encountered two cases of F. hepatica infection. One patient reported abdominal discomfort with marked eosinophilia. The other patient had chest discomfort with marked eosinophilia. The abdominal CT images revealed hypodense lesions in the liver. The ultrasonography-guided liver biopsy findings in both patients were indicative of parasitic infections. Serological tests confirmed the definite diagnoses. Both patients were treated with a single dose of triclabendazole, which is the treatment of choice for fascioliasis. These findings suggest that a diagnosis of fascioliasis, particularly in the acute phase, should be considered in patients with abdominal pain, marked eosinophilia, and hypodense hepatic lesions on CT.

The purpose of the research: is to conduct a comparative assessment of the infection of cattle with helminths and the structure of helminthic complexes in various natural provinces of Gorny Altai.Materials and methods. Coproovolarvoscopic examinations of about 2.4 thousand of cattle from various farms of the Altai Republic were carried out. Based on their results, the animal’s infection (the prevalence of infection, PI, %) with helminths in various natural provinces was characterized. The structural indices of various taxa of helminths (SI) have been calculated, which reflect their weight in the structure of the parasite complex.Results and discussion. Cattle in the Altai Republic are mostly infected with nematodes (51.4%). The indicators of the animal infection with trematodes and cestodes were significantly lower and amounted to 17.4 and 5.2% respectively. Nematodes dominate in the structure of animal helminthocenosis in various provinces. Their total SI varies from 66.8 to 76.8. The classes of trematodes and cestodes account for 8.8–26.3 and 4.5–14.4 respectively. The maximum SI was registered in Strongylata of the digestive tract (45.2), the subdominant position is occupied by trematodes: Dicrocelium sp. (SI – 13.2) and Fasciola hepatica (SI – 11.5). Moniezia sp. SI was quite low and amounted to 4.7.

The liver fluke, Fasciola hepatica, is an obligate blood-feeder, and the gastrodermal cells of the parasite form the interface with the host’s blood. Despite their importance in the host–parasite interaction, in-depth proteomic analysis of the gastrodermal cells is lacking. Here, we used laser microdissection of F. hepatica tissue sections to generate unique and biologically exclusive tissue fractions of the gastrodermal cells and tegument for analysis by mass spectrometry. A total of 226 gastrodermal cell proteins were identified, with proteases that degrade haemoglobin being the most abundant. Other detected proteins included those such as proton pumps and anticoagulants which maintain a microenvironment that facilitates digestion. By comparing the gastrodermal cell proteome and the 102 proteins identified in the laser microdissected tegument with previously published tegument proteomic datasets, we showed that one-quarter of proteins (removed by freeze–thaw extraction) or one-third of proteins (removed by detergent extraction) previously identified as tegumental were instead derived from the gastrodermal cells. Comparative analysis of the laser microdissected gastrodermal cells, tegument, and F. hepatica secretome revealed that the gastrodermal cells are the principal source of secreted proteins, as well as showed that both the gastrodermal cells and the tegument are likely to release subpopulations of extracellular vesicles (EVs). Microscopical examination of the gut caeca from flukes fixed immediately after their removal from the host bile ducts showed that selected gastrodermal cells underwent a progressive thinning of the apical plasma membrane which ruptured to release secretory vesicles en masse into the gut lumen. Our findings suggest that gut-derived EVs are released via a novel atypical secretory route and highlight the importance of the gastrodermal cells in nutrient acquisition and possible immunomodulation by the parasite.

Liver flukes, Fasciola spp., are veterinary and medically important parasites infecting numerous species of economically important animals in addition to humans on a global scale. The components of transforming growth factor beta (TGF-β) signalling are widely distributed throughout the animal kingdom and are considerably conserved. Through shared common signal transduction mechanisms, crosstalk of TGF-β signalling between a host and the parasite during infection is possible. Herein, we have identified and undertaken the molecular characterisation of a putative TGF-β homologue from the tropical liver fluke F. gigantica (FgTLM). A FgTLM cDNA was 3557 bp in length, it encoded for 620 amino acid polypeptide which consisted of 494 amino acids of prodomain and 126 amino acids comprising the mature protein. FgTLM displayed characteristic structures of mammalian TGF-β ligands that were unique to the inhibin-β chain, monomer of activin. A phylogenetic analysis revealed the high degree of conservation with TGF-β molecules from trematode species. Interestingly, the sequence of amino acid in the active domain of FgTLM was completely identical to FhTLM from F. hepatica. FgTLM expressed throughout the lifecycle of F. gigantica but was highly expressed in developmental active stages. The dynamics of expression of FgTLM during the developmental stages of F. gigantica was comparable to the pattern of TGF-β expression in F. hepatica. Our findings demonstrated that FgTLM exhibits a high level of similarity to FhTLM in the context of both amino acid sequence and the life stage expression patterns. These similarities underline the possibility that the FgTLM molecule might have the same properties and functions as FhTLM in biological processes of the immature parasites and host immune evasion. Consequently, the specific biological functions of FgTLM on either parasite or relevant hosts need to be defined experimentally.

RESUMEN Objetivo : Determinar el rendimiento diagnóstico de la técnica de inmnunocromatografía o flujo lateral para la detección de anticuerpos en pacientes con Fasciolosis humana. Materiales y métodos : Estudio observacional, prospectivo y de corte transversal. Hemos desarrollado una prueba de flujo lateral (Fasciorap) para el diagnóstico serológico de las Fasciolosis humana por Fasciola hepatica, compuesta por antígenos de excreción-secreción de formas adultas conjugadas con orocoloidal de 40 nm y una proteína A e IgG de conejo anti Fasciola hepatica como reactivos detectores en la línea de prueba y control, flanqueados por almohadillas en un cassette. Se evaluaron 240 sueros, 120 positivos, 50 sueros de pacientes con otras parasitosis, 20 de pacientes con enfermedades infecciosas y 50 sueros de personas no parasitadas, la interpretación de resultados se realizó por inspección visual a los 15 minutos de aplicada las muestras. Resultados : La prueba detectó la presencia de anticuerpos en el suero de pacientes con fasciolosis, alcanzando una sensibilidad de 92,5%, una especificidad de 94,17%, un valor predictivo positivo de 94,07% y negativo de 92,62%; con 100% de concordancia en la repetibilidad y reproducibilidad. Conclusiones : Fasciorap detecta casos de fasciolosis, por lo tanto, es una potencial prueba diagnóstica en zonas endémicas donde se requiere pruebas de punto de atención ABSTRACT Objective : To determine the diagnostic performance of the immunochromatography technique or lateral flow for the detection of antibodies in patients with human fasciolosis. Materials and methods : Observational, prospective and cross-sectional study. We have developed a lateral flow test (Fasciorap) for the serological diagnosis of fasciolosis due to F. hepatica, composed of excretion-secretion antigens of adult forms conjugated with orocolloid of 40 nm and a protein A and IgG of rabbit anti F. hepatica as detector reagents in the test and control line, flanked by pads in a cassette. 240 sera were evaluated, 120 positive, 50 sera from patients with other parasites, 20 from patients with infectious diseases and 50 sera from non-parasitized people, the interpretation of results was performed by visual inspection 15 minutes after applying the samples. Results : The test detected the presence of antibodies in the serum of patients with fasciolosis, reaching a sensitivity of 92.5%, a specificity of 94.17%, a positive predictive value of 94.07% and a negative predictive value of 92.62%; with 100% agreement on repeatability and reproducibility. Conclusions : Fasciorap detects cases of fascioliasis, therefore, it is a potential diagnostic test in endemic areas where point-of-care testing is required

Fasciola hepatica is a liver parasite of ruminants whose distribution is determined by its intermediate host, the freshwater snail Galba truncatula. In Europe, F. hepatica is mostly associated with lowlands. Infection from sympatric domestic reservoirs is rarely reported in wild mountain ungulates. This study explores F. hepatica in a multi-host system in a European alpine area. Serum samples (n = 1,209) from Pyrenean chamois (Rupicapra p. pyrenaica), European mouflon (Ovis aries musimon), domestic sheep (Ovis aries) and domestic cattle (Bos taurus) were collected in the National Game Reserve of Freser-Setcases (NGRFS) in Catalonia, Northeastern Spain, from 2008 to 2019, and tested for antibodies against F. hepatica. During the same period, the livers of 214 chamois hunted in the NGRFS were inspected for F. hepatica and associated pathological changes. Finally, 907 freshwater snails were collected in summer 2016 between 1559 and 2,224 metres above sea level (asl) in the NGRFS, and F. hepatica DNA sought by PCR. Antibodies against F. hepatica were detected in all four species, with a higher prevalence in cattle and sheep than in chamois. Fasciola hepatica and hepatic lesions were concurrently observed in 13/214 of the chamois livers inspected (6.1%, CI95 2.9%-9.3%). Fasciola hepatica DNA was detected in one out of the 907 snails (0.1%, Cl95 0.1% - 0.3%; Ct value 33.3) and collected at 2054 m asl. Fasciola hepatica was consistently detected in a high mountain multi-host system, suggesting that its life cycle is completed and that it occurs endemically at the highest elevation reported in Europe. Transhumant livestock are the likely source in this alpine ecosystem, which according to rare occurrence of F. hepatica DNA in G. truncatula is still a suboptimal habitat for F. hepatica life cycle. Studying parasites at their highest distribution range can be useful to monitor climate change in seasonal mountain environments.

The objective of the present study was to determine the seroprevalence of Fasciola hepatica infection in horses, donkeys and mules from different climate regions in two states of Mexico. A total of 594 serum samples were analysed for immunoglobulin G (IgG) antibodies using enzyme-linked immunosorbent assay (ELISA), with excretory-secretory (E/S) products as the antigen. The diagnostic sensitivity and specificity of serum IgG ELISA were 100% and 97.2%, respectively. We collected data using a questionnaire. The overall prevalence of the parasite in equids between May 2018 and April 2019 was 13.1% (78 out of 594). The highest prevalence was found in mules (17.9%), followed by that in donkeys (13.9%) and horses (10.4%). In addition, the highest percentage of positive equines was detected in samples from the tropical climate (17.5%). The identification of risk factors was assessed by bivariate logistic regression analysis. Mules had 8.608 times higher risk for F. hepatica infections as compared with horses (odds ratio (OR) 8.608; 95% confidence interval (CI): 2.809–26.377), while the OR for 6–10-year-old equines was 93.375 compared with that of young equines (≤ 5 years old) (OR 93.375; 95% CI: 11.075–787.239). Likewise, tropical climate equines had 83.7% increased odds for fasciolosis (OR 1.83).

Objective This study was conducted to determine the prevalence of distomatosis in cattle in the Agri province in Turkey. Methods The livers and bile ducts of 200 slaughtered cattle were examined macroscopically, and the gall bladders were examined by sedimentation. The presence of Fasciola hepatica eggs and antigens in 188 cattle were investigated using sedimentation and coproantigen ELISA, respectively. Egg counts per gram of faeces (EPG) was determined using the modified McMaster sedimentation method. Results Adult Fasciola hepatica and Dicrocoelium dendriticum were found in the bile ducts of 47 and 25 cattle, respectively. Examination of the gall bladders of cattle revealed the presence of eggs of Fasciola hepatica and D. dendriticum in 63 and 48 cattle, respectively. The coproantigens of Fasciola hepatica were found in 148 (78.7%) cattle using ELISA. Additionally, the eggs of F. hepatica were found in 63 (33.5%) cattle by sedimentation, with the value of EPG between 17 and 83. The prevalence of D. dendriticum was determined as 25.5%, with the value of EPG between 17 and 67. Conclusion In this study, distomatosis was detected in cattle in Agri province using coproantigen ELISA and sedimentation methods along with macroscopic examination. The findings necessitate an urgent implementation of an effective prevention and control program, which can increase the awareness of cattle breeders regarding these diseases. Furthermore, there is also a need for more detailed studies on these diseases.

Fasciola hepatica (F. hepatica) is a well-known zoonotic parasite that is crucial for economic and public health worldwide. Quantitative proteomics studies have been performed on proteins expressed by F. hepatica to investigate the differential expression of proteomes in different growth phases. And the screening of several marker proteins for use as early diagnostic antigens is essential. In this study, high-performance liquid chromatography-tandem mass spectrometry (LC-MS/MS) was conducted to analyze the differences in the expression of F. hepatica somatic proteins in different growth phases. Furthermore, gene ontology (GO) functional annotation, KEGG metabolic pathway, and clustering analyses were also performed. LC-MS/MS identified 629, 2286, 2254, and 2192 proteins in metacercariae, juvenile flukes 28dpi, immature flukes 59dpi, and adult phases, respectively. GO analysis revealed that differentially expressed proteins (DEPs) were mainly involved in transport, localization, metabolism, enzyme regulation, protein folding and binding, and nucleoside and nucleotide binding. The DEPs were enriched in cells, intracellular components, organelles, cytoplasm, vesicles, and membranes. KEGG pathway annotation results showed that the DEPs were involved in metabolism, genetic information processing, environmental information processing, cellular processes, organismal systems, and other processes. These findings provide a theoretical basis for vaccine development and establishing early diagnostic methods in the future. The purpose of the research is evaluating histological and histochemical research methods as information criteria in identifying the effect of chemicals on trematodes. Materials and methods. The study material was trematodes Fasciola hepatica, F. gigantica, Paramphistomum cervi and Dicrocoelium lanceatum collected during dissecting spontaneously infected animals after they had been treated with anthelmintics (Triclabendazole, Dimezole, Polytrem, Antitrem, Tetraxichol, Tegalid and Fascocid) using various therapeutic doses with appropriate regimens. Trematodes from untreated animals were used as control. The detected material was processed under the generally accepted histological procedure and embedded in paraffin. Microslides, namely, slices of 5–7 µm thick, were stained using histological and histochemical methods and studied under a light microscope. Results and discussion. Morphologically, when the microstructure of organs and tissues of trematodes F. hepatica, F. gigantica, P. cervi and D. lanceatum treated with anthelmintics (Triclabendazole, Dimezole, Polytrem, Antitrem, Tetraxichol, Tegalid and Fascocid) was studied, edema, vacuoles, lysis of tissue and cell structures, and severe basophilia were observed in their body. Protein metabolism imbalance and distorted protein synthesis were recorded histochemically. As an outcome of pathophysiological processes after anthelmintic treatment, necrobiosis detected in trematodes and the developing severe edema lead to colliquative necrosis caused by irreversible dystrophic processes occurred at the level of water and protein metabolism imbalance. Misdirected histochemical reaction of trematodes’ tissues and organs when stained for proteins marks nuclear degeneration, termination of protein molecule synthesis, and protein metabolism failure.

In this work, we compare and evaluate the efficiency of fosfatriclaben with three commercial fasciolicides in experimentally infected sheep. Fosfatriclaben is a novel prodrug derived from triclabendazole; it is highly water-soluble with excellent aqueous stability at pH 7, properties that make it ideal for developing intramuscular pharmaceutical compositions in the form of solutions. In order to compare, 30 mixed breed sheep, previously diagnosed negative to fluke eggs, were infected with 200 metacercariae of Fasciola hepatica, twice. Five groups of six animals/each were formed for treatments. Group 1 (G1) was treated with closantel 5% injectable at 5 mg/kg subcutaneously, G2 with clorsulon at 2 mg/kg subcutaneously, G3 with triclabendazole at 12 mg/kg per os, G4 with fosfatriclaben at 6 mg/kg intramuscularly (dose equivalent to triclabendazole content), and G5 remained as the non-treated control. On day 110, fecal samples were examined to determine the percentage of egg reduction after treatment, and sheep were humanely euthanized. The livers were collected, the flukes were extracted, measured, and counted. Efficiency in egg reduction was of 86.8, 90.5, 98.4, and 97.3% for closantel, clorsulon, triclabendazole, and fosfatriclaben, respectively, and efficiency against flukes was of 96.2, 91.9, 99.4, and 95.7%, respectively. No statistical differences were found between treatments. It is concluded that fosfatriclaben at 6 mg/kg intramuscularly presented a high fasciolicide efficiency, similar to the best commercial fasciolicides, having advantage over its predecessor since it uses half of the dose required by triclabendazole to remove flukes in sheep under study.

The purpose of the research is to study the seasonal and age dynamics of infection of cattle and sheep with Fasciola hepatica L., 1758 in the Central economic region of Russia in the Bryansk Region.Materials and methods. The study of the seasonal dynamics of infection of cattle and small ruminants with F. hepatica was carried out by studying the feces of animals by means of coprological methods. The age dynamics of infection of cattle and sheep with F. hepatica was studied in the farms of the Bryansk region based on the results of veterinary-sanitary inspection.Results and discussion. When studying the effect of the season of the year on infection of cattle and sheep with F. hepatica, it was established that the adult stock was infected with F. hepatica in all seasons of the year, ranging from 9.0 % in June to 16.6 % in February of cattle and from 8.5% in August to 17.6% in January of sheep. The highest infection rate of the animals was noted in winter, which, in our opinion, is due to the fasciola reaching a new generation of maturity. With the age of the animals, their F. hepatica infection rate increases due to the accumulation of infection and the realization of the possibility of reinfection in subsequent years. The maximum F. hepatica infection rate of cattle with was observed in the animals over the age of 8 years, and sheep – of 5 years and older. They, in turn, play a major role in the circulation of fasciolosis infection.

Human fascioliasis is a rare parasitic disease outside of countries in which it is endemic. The diagnosis of hepatic-phase fascioliasis by diagnostic imaging alone is challenging. A 69-year-old female was referred to our hospital for the treatment of a solitary solid cystic mass lesion, 6 cm in diameter, accompanied with mild symptoms and minimal changes in laboratory parameters. Intrahepatic cholangiocarcinoma was suspected, and she underwent extended posterior sectionectomy. Four months later, she was re-admitted because of fatigue, high fever, and epigastric pain. Her eosinophil fraction and immunoglobulin E levels were extremely elevated (49.1% and 6833 IU/ml, respectively). She was found to have two new reticular cystic hepatic tumors. Serum dot enzyme-linked immunosorbent assay for parasites revealed strong positivity for Fasciola hepatica. Praziquantel was ineffective, and multi-cystic tumors rapidly developed in the left lateral section, requiring emergency left lateral sectionectomy. An F. hepatica helminth, approximately 3 cm in size, was observed on the cut liver surface during hepatic resection. Prophylactic triclabendazole (1,000 mg/day) was administered twice. She has been well for over 10 years without relapse of fascioliasis. In patients with hepatic tumors accompanied by inflammatory changes and eosinophilia, detailed medical history and serological testing by dot enzyme-linked immunosorbent assay for parasites are strongly recommended.

The foodborne trematodiases refer to a cluster of zoonotic neglected tropical diseases caused by trematodes, with transmission involving ingestion of contaminated plants, fishes, and crustaceans. Over 40 million people are infected with foodborne trematodes and 750 million are at risk of infection. From a public health point of view, important species include Clonorchis sinensis, Opisthorchis viverrini, Opisthorchis felineus, Fasciola hepatica, and Fasciola gigantica. Infection with C. sinensis and O. viverrini is classified as a group 1 biological carcinogen and a major risk factor for cholangiocarcinoma. The carcinogenic potential of the infection with O. felineus is less clear but recent biochemical and histopathological findings revealed that opisthorchiasis felinea also fits this pattern. By contrast, evidence of carcinogenic potential of infection with F. hepatica or F. gigantica, close phylogenetics relatives of Opisthorchis, is less certain. Oxysterols have been essentially described in animal model of opisthorchiasis and associated cholangiocarcinoma. Several oxysterol-like metabolites have been detected not only on developmental stages of O. viverrini and O. felineus but also on biofluids from experimentally infected hamsters as products of the activities of the liver flukes. These sterol derivatives are metabolized to active quinones that can modify host DNA. We have postulated that helminth parasite-associated sterols might induce tumor-like phenotypes in biliary epithelia, the cells of origin of liver fluke infection-associated cholangiocarcinoma, through the formation of DNA adducts, dysregulation of apoptosis, and other homeostatic pathways. Here we review, interpret, and discuss findings of oxysterol-like metabolites detected in liver flukes and their role in carcinogenesis, aiming to enhance understanding the pathogenesis of foodborne trematodiasis caused by Opisthorchis and Fasciola species. In future, further investigations will be necessary in order to comprehend relationship between liver flukes' oxysterols and their role in infection-associated diseases in humans.

The worldwide occurring common liver fluke Fasciola hepatica can infect humans and animals and leads to considerable illness and economic loss annually. The aim of this study was to determine the genetic diversity of F. hepatica in Austria. In total, 31 adult flukes isolated from cattle from various regions in Austria were investigated for their cytochrome oxidase subunit 1 (cox1) and nicotinamide dehydrogenase subunit 1 (nad1) gene sequences. It was shown that Austrian isolates of F. hepatica reveal extensive genetic diversity. To the best of our knowledge, these are the first data on the diversity of F. hepatica in Austria.

Fasciola gigantica faces a series of threats from various free radicals produced by the host immune system during its invasion through the abdominal cavity and establishment in the bile duct of ruminants, limiting the fluke viability. The role of the superoxide radical produced by Muzaffarnagari sheep immune effector cells against F. gigantica newly excysted juveniles (NEJs) is highlighted in this study, as is the critical role of superoxide dismutase enzyme (SOD) in dismutation of superoxide radicals derived from host immune effector cells in vitro. Fasciolosis is a neglected zoonotic parasitic disease caused by liver flukes, Fasciola hepatica. F. hepatica is harmful to livestock and human health. However, changes in host metabolism caused by F. hepatica infection are unclear. An artificial sheep model was established as follows. The sheep in the infection group were fed with 220 metacercariae obtained by incubating F. hepatica miracidia with the intermediate host snail (Galba pervia). Thereafter, serum and blood were collected from these sheep periodically. Changes in 31 biochemical parameters were systematically tested over different periods of infection. Metabolomic analysis was performed based on liquid chromatography/mass spectrometry (LC-MS) technology using a UHPLC system. Differentially expressed metabolites were analyzed for biomarkers, and changes in the metabolic pathways of the host were evaluated. Ten biochemical parameters (TP, ALB, GLB, DBIL, IBIL, GGT, LDH, CHOL, HDL-C, and BUN) showed significant dynamic changes during the study period. For metabolomic analysis: 13, 27, and 82 differential metabolites (ESI+ mode) and 0, 37, and 83 differential metabolites (ESI- mode) were found on 7, 56, and 98 dpi, respectively. The number of different metabolic pathways increased with disease development. Five metabolites had the highest area under the curve (AUC) value as joint diagnostic factors, indicating their potential use as biomarkers for diagnosing F. hepatica infection. This study establishes the F. hepatica life cycle in an artificial model of sheep infected with F. hepatica to identify changes in metabolic pathways in the host due to infection. Biochemical parameters and metabolomic analysis revealed that not only the biomarkers screened by differentially expressed metabolites have the potential to diagnose F. hepatica infection in sheep, but the differential pathways and biochemical parameters also explain the metabolic pathway changes in the sheep infected with F. hepatica. F. hepatica absorbs the nutrients of the host and destroys the essential metabolic pathways of the host. This result suggests that animal metabolism can be altered in the host as a response to parasitic infections such as F. hepatica. In addition, this finding will provide the basis for studying the pathogenic mechanisms and biomarkers for F. hepatica infection.

The clinical manifestations of fascioliasis hepatica in humans are unspecific. Traditional diagnosis relies on evidence of live parasites or eggs in the bile or feces. However, due to similar imaging manifestations, they are often misdiagnosed as malignant tumors. Here, we report a case of a 43-year-old woman with fever and space-occupying liver disease. Liver biopsy, parasite-specific antibody screening, and stool testing did not find any pathogens. Therefore, metagenomic next-generation sequencing (mNGS) and routine microbiological examinations were performed. Finally, Fasciola hepatica was only identified by mNGS. The body temperature of the patient and the eosinophil count remained normal, and the space-occupying liver lesions were significantly absorbed after more than 7 months of treatment with albendazole. The details of this case highlight the timely use of mNGS to identify parasites and judge therapeutic effects after treatment, providing important help for clinical decision-making.

Fasciola hepatica, a worldwide-distributed liver fluke, is one of the causative agents of fasciolosis, a zoonotic disease that affects livestock and humans. In livestock, fasciolosis causes huge economic losses worldwide, reducing animal fertility, milk production, weight gain and condemnation of livers. In spite of the availability of drugs, such as triclabendazole (TCZ), for the treatment of fasciolosis, they do not necessarily prevent liver damage or parasite reinfection and can eventually increase parasite resistance. The aim of this research was to relate the hepatic function, haematological parameters, leukocyte counts in circulation and parasite egg shedding during F. hepatica acute and chronic phases of infection in cattle as well as to determine how these parameters change with TCZ-treatment of chronically infected cattle. Our results show that increased levels of serum aspartate aminotransferase (AST) and gamma glutamyltransferase (GGT) were detected in early stages of the experimental infection. Moreover, high circulating eosinophil count and plateletcrit levels were correlated with fluke number in livers from infected cattle. On the other hand, although TCZ-treatment in the chronic phase of infection reduced parasite burden and damage in the liver, it was not able to completely avoid them. In conclusion, our work sheds light into the physiopathological mechanisms induced during fluke infection in cattle, revealing the complexity of the host response to the infection, together with the effects of TCZ-treatment in chronically infected animals.

Understanding drug penetration, distribution, and metabolization is fundamental for understanding drug efficacy. This also accounts for parasites during antiparasitic treatment. Recently, we established matrix-assisted laser desorption/ionization (MALDI) mass spectrometry imaging (MSI) in blood flukes and liver flukes. This label-free technique is capable of visualizing the molecular distribution of endogenous and exogenous molecules, such as drug compounds. Here, we conducted atmospheric-pressure scanning microprobe MALDI MSI (AP-SMALDI MSI) of tissue sections of adult Fasciola hepatica that have been treated in vitro with 100 µM of triclabendazole (TCBZ), the drug of choice for treatment of fasciolosis, and its main metabolite triclabendazole sulfoxide (TCBZ-SO). Measurements covered an m/z mass range of 250–1,000 and provided a high spatial resolution using a pixel size of 10 µm. To support the interpretation of drug distribution, we first identified endogenous lipids that mark characteristic tissues such as the gastrodermis, the tegument, and the parenchyma. The obtained results suggested an early tegumental route of TCBZ uptake within 20 min, followed by spreading throughout the parasite after 4 h, and an even distribution in most tissues after 12 h. This coincided with a strong reduction of parasite vitality. TCBZ-SO treatment demonstrated the accumulation of this metabolite in the same tissues as the parent drug compound. These data demonstrate the auspicious potential of MALDI MSI to visualize uptake and distribution patterns of drugs or drug-candidate compounds in parasites, which might contribute to preclinical drug discovery in liver fluke research and beyond.

Control of liver fluke infections remains a significant challenge in the livestock sector due to widespread distribution of drug resistant parasite populations. In particular, increasing prevalence and economic losses due to infection with Fasciola hepatica is a direct result of drug resistance to the gold standard flukicide, triclabendazole. Sustainable control of this significant zoonotic pathogen, therefore, urgently requires the identification of new anthelmintics. Plants represent a source of molecules with potential flukicidal effects and, amongst their secondary metabolites, the diterpenoid abietic acids can be isolated in large quantities. In this study, nineteen (19) chemically modified abietic acid analogues (MC_X) were first evaluated for their anthelmintic activities against F. hepatica newly excysted juveniles (NEJs, from the laboratory-derived Italian strain); from this, 6 analogues were secondly evaluated for their anthelmintic activities against adult wild strain flukes. Australian livestock are challenged by liver fluke (Fasciola hepatica) in grazing regions endemic to the intermediate snail host. Liver fluke infests a wide range of herbivores including free-roaming wildlife such as kangaroos (Macropods). The role played by Macropods in cross-species transmission and as vectors for anthelmintic resistance is largely unknown. In Phase 1 of this study, liver fluke of Eastern grey kangaroo (Macropus giganteus Shaw, 1790) origin (Kangaroo isolate) were artificially infected in sheep to confirm establishment and cross-species transmission. In Phase 2, the efficacy of triclabendazole (TCBZ) was assessed in vivo against the Kangaroo isolate to identify any drug resistance. Forty (40) merino sheep were housed in pens and allocated to one of 4 groups (Groups 1–4). Groups 1 and 2 were artificially infected with a TCBZ resistant liver fluke isolate (Oberon) originating from sheep whilst Groups 3 and 4 were infected with the Kangaroo isolate (Phase 1). At 9 weeks post infection (wpi), sheep in Groups 2 and 4 were treated with 10 mg/kg TCBZ (Phase 2). Sheep were subsequently euthanased at 11 wpi to conduct total fluke counts (TFC) in the liver. Faecal samples were collected fortnightly to measure fluke egg counts and coproantigens. Individual blood samples were collected, concurrently with faecal sampling, to monitor haematocrit and plasma proteins levels. Liver fluke of kangaroo origin established to patent infections in sheep with similar establishment and pathogenicity to the Oberon isolate. TCBZ achieved an 86 % reduction in TFC (99.8 % - adult fluke, 0 % - immature fluke) in sheep with the Kangaroo isolate and a 28 % reduction in the Oberon isolate (37 % - adult, 0 % - immature fluke). An 89 % reduction in faecal coproantigens was observed in sheep with the Kangaroo isolate and no reduction in sheep with Oberon. This study confirmed cross-species transmission of liver fluke from a kangaroo to sheep. When cohabiting the livestock grazing environment, kangaroos may act as reservoirs for liver fluke and vectors for drug resistance within liver fluke endemic areas.

La infección humana por Fasciola hepatica en el Perú es una enfermedad infecciosa emergente. En estarevisión describimos la situación de la fasciolosis en el Perú, en base a los estudios mas recientes sobreepidemiología, métodos de diagnóstico y tratamiento. Proponemos una nueva clasificación clínica en baseal estado de la enfermedad. Estudios recientes han destacado la variabilidad clínica de esta infección quepuede comprender desde una infección indolente a una severa que puede comprometer la vida del paciente. Un incremento en el número de casos humanos ha sido reportado en todo el mundo, y en especial en la Región Andina de Latinoamérica. Las manifestaciones clínicas más comunes en la fase aguda son: hepatomegalia, fiebre, pérdida de peso y eosinofilia. En la fase crónica, esta puede ser leve e inespecífica, pero también severa como obstrucción biliar, colangitis, quistes calcificados hepáticos, cálculos vesiculares y fibrosis hepática. La Técnica de Sedimentación Rápida descrita por Lumbreras debe ser aplicada para el diagnóstico de la fase crónica y para estudios epidemiológicos en zonas endémicas. El examen directo solo detecta 2 % de los casos. El nuevo examen diagnostico ELISA Fas2 (catepsina L1) tiene una sensibilidad del 92,4 % y especificidad del 83,6 %, y esto fue observado en 634 sujetos peruanos en áreas endémicas y es útil tanto para la fase aguda como la crónica. Las anormalidades radiológicas más comunes en la fase aguda son: lesiones hipodensas hepáticas en forma de “camino”, abscesos hepáticos y hematoma subcapsular. La ecografía abdominal tiene una baja sensibilidad en casos crónicos y no es recomendada para tamizaje. El triclabendazol es el tratamiento de elección con una dosis única (tasa de curación ≥ 90 %) para ambas fases, pero casos de resistencia a la droga es ahora una preocupación aunque solo en animales. El nuevo arsenal de información disponible puede ser aplicado a los programas de prevención y control en el Perú.

La utilización de extractos vegetales representa una alternativa para el manejo integrado de parásitos enSalud Pública, debido a su bajo costo y al menor impacto sobre el ambiente al conservar la biodiversidad. El presente trabajo tuvo como objetivo determinar el efecto de los extractos de la planta amazónica Paullinia clavigera Simpson “Sacha yoco” (Sapindaceae) sobre la eclosión de huevos de Fasciola hepatica L. (Trematoda: Fasciolidae). Los huevos fueron obtenidos de bovinos procedentes del camal de Yerbateros, Lima, Perú. Se realizó el tamizaje fitoquímico y se prepararon los extractos hexánico, clorofórmico e hidroalcohólico a partir de la corteza y hoja de P. clavigera. Se realizaron bioensayos para determinar el efecto de Sacha yoco sobre la eclosión de los huevos, los cuales fueron incubados en los extractos a 22° ± 2º C durante 21 días y finalmente expuestos a la luz por 12 h. No existieron diferencias significativas entre el número de huevos de F. hepatica expuestos en cada una de las concentraciones en los bioensayos. Solo existió efecto en la eclosión de los huevos de F. hepatica por el extracto hexánico de corteza de P. clavigera a las dos concentraciones más altas. La toxicidad observada en el extracto hexánico de corteza de P. clavigera pudiera deberse a la presencia de triterpenos, flavonoides y quinonas.

Background The popularity of new world camelids, particularly alpacas, is growing rapidly in Ireland, presenting a clinical challenge to veterinary practitioners who may not have worked with these species previously. To the authors’ knowledge, the clinical course of a case of acute fasciolosis in an alpaca has not previously been reported, and fasciolosis has not been reported at all in alpacas in Ireland, making this case report a valuable addition to the current literature. Case presentation A three-year-old male castrated huacaya alpaca was admitted to UCD Veterinary Hospital with a two-day history of colic and tenesmus. He had been treated with albendazole, dexamethasone and potentiated amoxycillin by the referring veterinary practitioner with no response. On initial clinical exam, sensitivity to abdominal palpation was the only abnormality. However, the alpaca proceeded to show abnormal lying positions, tenesmus and reduced faecal output over the next 24 h. A general blood panel demonstrated moderate anaemia, marked hyperglobulinaemia and moderately increased hepatocellular and hepatobiliary enzyme activity. Abdominal radiography revealed enlargement of the first forestomach compartment without evidence of gastrointestinal obstruction or peritonitis. An abdominal ultrasound exam revealed an elongated, heterogenous mass in the caudoventral abdomen that appeared to be contiguous with the liver. FNA of this mass revealed that it was in fact a liver lobe with biliary stasis and inflammation. Faecal sedimentation demonstrated Fasciola hepatica eggs. In spite of treatment with triclabendazole and supportive treatment including blood transfusion, the alpaca’s condition continued to deteriorate and he was euthanised. On post-mortem exam, acute fasciolosis was diagnosed. Conclusions The clinical presentation and course of a case of acute fasciolosis in an individual alpaca is described, including the results of a range of diagnostic tests that were carried out. The final diagnosis is supported by a description of post-mortem findings. This information will serve as a resource for veterinary practitioners involved in the diagnosis and treatment of similar cases.

Information is provided on the causative agents of fasciolosis and the developmental biology of Fasciola hepatica. Fasciolosis of cattle is widespread in the conditions of the Kaluga Region and the Central Federal District on farms of different forms of ownership. The greatest number of F. hepatica eggs in 1 g of feces in cattle is established in JSC "Iskra" - 175,4 specimens, the lowest - in the samples from cows from private farms - 87,6 sp. At the opening slaughter of cattle at meat processing plants region and slaughterhouses invasion of their fasciolae was 34.8%. The average number fascial one infected animal was 32.1 specimens, including adult - and larval specimens of 25.2 and 6.9 copies At veterinary-sanitary expertise and helminthological autopsy of the liver, gallbladder and bile ducts installed year-round invasion fasciolae mainly in the adult stage. Young animals of the current year are infected with fasciola in the pasture massly in August-September, and the extent of invasion in November was 4.3%, for winter-stabling months an average of 12.6%, in March - 17.9% and in April after de-worming - 5.9%.The data obtained in the study of the distribution of the small pond has shown that in the Kaluga region these mollusks are widespread. In this case, the invasion of the small pond snail by larval forms of fasciolae in different years has its own peculiarities, which are influenced by climatic factors.The different dosage forms of clozantel - Rolenol and Santel and Genamectin with mixed fascioliasis and hypodermatosis of bovine in the recommended dose against subcutaneous gadfly larvae - provided 100% efficacy. The tested dosage forms of clozantel are similar in effectiveness against subcutaneous gadfly larvae (EE-100%) and fasciol (EE-95 and 90%, IE-97.4 and 96.3%).The therapeutic and economic effectiveness of complex domestic preparations of Helmicide and Fascocida in spontaneous fascioliasis of cattle is studied.

Fasciolosis is a re-emerging disease of livestock and rarely human, being endemic in Iran. Herein, we aimed to quantitatively assess the human seropositivity and prevalence of animal fasciolosis in our country. English and Persian databases were searched for online literature. In total, 10 human seroprevalence studies and 49 animal investigations were obtained from January 1999 to March 2019. Only animal studies were included in meta-analysis. The highest human seroprevalence was detected in Guilan province (326/452; 50%). The pooled prevalence of the animal infections was calculated as 6.2% (95% CI = 5.8%–6.5%). In detail, the prevalence ranges in three hosts included as 4.2% (95% CI = 3.8%–4.5%) in sheep, 9% (95% CI = 8.0%–9.9%) in cattle and 3.1% (95% CI = 2.4%–3.7%) in goat. The highest prevalence rate was observed in the north, 11.8% (95% CI = 8.4%–15.1%), while the lowest prevalence was detected in the central Iran with 1.8% (95% CI = 1.3%–2.3%). Egger’s regression test revealed no significant publication bias (P = 0.307). Also, there was no remarkable correlation between weighted prevalence and sample size (P = 0.249) as well as year of study (P = 0.172). These findings would be necessary for better preventive strategies in case of human and animal Fasciola infections as well as snail intermediate hosts.

Background Fasciola hepatica (liver fluke) affects grazing animals including horses but the extent to which it affects UK horses is unknown. Objectives To define how liver fluke affects the UK horse population. Study design Descriptive, cross-sectional, observational study. Methods An F. hepatica excretory-secretory antibody detection ELISA with a diagnostic sensitivity of 71% and specificity of 97% was validated and used to analyse serum samples. An abattoir study was performed to determine prevalence. A case-control study of 269 horses compared fluke exposure between horses with liver disease and controls. Data on clinical signs and blood test results were collected for sero-positive horses. Genotyping of adult fluke was used to produce a multilocus genotype for each parasite. Results Four (2.2%) of 183 horses registered in the UK, sampled in the abattoir, had adult flukes in the liver, and the sero-prevalence of F. hepatica was estimated as 8.7%. In the case-control study, horses showing signs consistent with liver disease had significantly higher odds of testing positive for F. hepatica on ELISA than control horses. In 23 sero-positive horses, a range of non-specific clinical signs and blood test abnormalities was reported, with a third of the horses showing no signs. Genotypic analysis of liver flukes from horses provided evidence that these came from the same population as flukes from sheep and cattle. Main limitations Bias could have arisen in the prevalence and case-control studies due to convenience sampling methods, in particular the geographic origin of the horses. Only a small number of horses tested positive so the data on clinical signs are limited. Conclusions Exposure to liver fluke occurs frequently in horses and may be an under-recognised cause of liver disease. Flukes isolated from horses are from the same population as those found in ruminants. When designing and implementing parasite control plans, fluke should be considered, and horses should be tested if appropriate.

The purpose of the research is to study morphofunctional changes in tegument, intestines and parenchyma of Fasciola hepatica after triclabendazole influence.Materials and methods. The objects of the research were F. hepatica, taken from the liver of spontaneously infected sheep in 24 hours after triclabendazole influence at a dose of 10 mg/kg. Trematodes were fixed in methyl alcohol and glacial acetic acid solution at the ratio 3 : 1, and then in 70% alcohol. The material was treated by the generally accepted histologic procedure and submerged into paraffin. Sections with thickness 5–7 mkm were dyed with histological and histochemical methods and were studied in a light microscope.Results and discussion. Morphofunctional research of tegument and intestines of F. hepatica – organs which perceived anthelminthic first, showed its deep structural changes after triclabendazole influence. Parenchyma – a tissue of internal environment, which is a system regulating parasite’s organism metabolic processes and homeostasis is also exposed to anthelminthic influence. Parenchyma’s decay products increase toxic properties of anthelminthic and his metabolites, and in common this greatly influences on reproductive organs, including a female with eggs.

peer-reviewed Background Quantitative genetic studies suggest the existence of variation at the genome level that affects the ability of cattle to resist to parasitic diseases. The objective of the current study was to identify regions of the bovine genome that are associated with resistance to endo-parasites. Methods Individual cattle records were available for Fasciola hepatica-damaged liver from 18 abattoirs. Deregressed estimated breeding values (EBV) for F. hepatica-damaged liver were generated for genotyped animals with a record for F. hepatica-damaged liver and for genotyped sires with a least one progeny record for F. hepatica-damaged liver; 3702 animals were available. In addition, individual cow records for antibody response to F. hepatica on 6388 genotyped dairy cows, antibody response to Ostertagia ostertagi on 8334 genotyped dairy cows and antibody response to Neospora caninum on 4597 genotyped dairy cows were adjusted for non-genetic effects. Genotypes were imputed to whole-sequence; after edits, 14,190,141 single nucleotide polymorphisms (SNPs) and 16,603,644 SNPs were available for cattle with deregressed EBV for F. hepatica-damaged liver and cows with an antibody response to a parasitic disease, respectively. Association analyses were undertaken using linear regression on one SNP at a time, in which a genomic relationship matrix accounted for the relationships between animals. Despite the presence of some information about the study of fascioliasis in theAbsheron region (Absheron Peninsula and the Khizi district), where private farms are widely developed, until recent years this problem has remained virtually unexplored. And the studies conducted 40–50 years ago are historically outdated and do not provide a complete picture of thecurrent economic system. Taking all this into account, by applying appropriate methods, theextensiveness and intensity of the spread of fascioliasis (Fasciola hepatica L.) in different villages and settlements of the Absheron region, as well as along various altitude belts, were determined. As a result of the research it was found that the greatest prevalence of the extensiveness of this parasite in various villages and towns fell on the following territories: Gyzyldere village — 33.9%, Altagach village — 30.2%, Tudar village — 29.2%. Also, as a result of the research, it was found that infection is highest in the lowland zone (31.4%). And this is closely related to theenvironmental impact of biotic and abiotic factors in that zone. The results of the study are reflected in the tables and charts.

The liver fluke Fasciola hepatica is the main cause of fasciolosis in North Africa leading to significant economic losses and public health problems. In this study, the ribosomal internal transcribed spacer (ITS), cytochrome c oxidase I (COI), the mitochondrial region spanning the COI-trnT-rrnL, and the NADH dehydrogenase subunit I (NADI) markers were used to characterize Fasciola flukes from Algeria. Fasciola appeared widespread from the east to the west of Algeria. Among 1701 sampled cattle from 8 Algerian provinces, 5% were infected. Using morphological and morphometric analysis, one morphotype of Fasciola was observed. Nuclear ITS marker indicated that all collected flukes belong to F. hepatica. Multiple alignments of ITS dataset revealed two haplotypes, one described here for the first time. Analysis of molecular variance (AMOVA) of mitochondrial markers revealed weak population structure in Algeria. Mismatch distributions, neutrality tests, and median-joining network analysis all were compatible with a recent expansion of Algerian F. hepatica population. Fasciolosis appeared common in Algerian cattle, it seems that the absence of control strategy coupled to the favorable Mediterranean climate may lead to a reconstruction and dispersion of its populations. This study provides important results concerning the genetic characterization and variability of F. hepatica in Algeria as well as the significant role of cattle importation in shaping its dispersal route worldwide.

The immunogenicity and efficacy of Fasciola DNA vaccines have not yet been comprehensively summarised in the form of a systematic review and meta-analysis. Though multiple vaccine studies with respect to Fasciola vaccines exist, the variance in the experimental parameters has made comparison difficult. We conducted a bibliographic database search in Scopus, PubMed, Science Direct, Cochrane Library, EMBASE and Web of Science databases, limited to publications from 1998 to 2017. The key words: Liver fluke, Fasciola hepatica, Fasciola gigantica, DNA vaccination, and immunogenicity were used in combination to form search strings. A total of 4760 studies were identified after initial screening, of which 14 qualified for systematic review and 7 for meta-analysis. The mean Odds Ratio (OR) for all studies was 0.565 (95% confidence interval (CI) of 0.293 to 1.087), which means the percentage of protection in terms of decreased fluke burden in animals vaccinated with DNA vaccines was 43.5%. A moderate protective efficacy was observed for cysteine protease and phosphoglycerate kinase vaccine antigen candidates (pooled OR and 95% CI, [0.542; 0.179-1.721] and [0.616; 0.219-1.735], respectively). Vaccine effectiveness was observed in individual studies and cohorts; however, the overall pooled efficacy for all vaccine candidates was found to be non-significant. Despite multiple individual studies showing promising results for various DNA vaccine candidates against fascioliasis, the pooled studies showed the non-significant effect of the vaccine formulations against fluke burden, and displayed minimal protective efficacy against Fasciola infection. Though promising results are observed in isolated studies, further animal trials with standardised experimental parameters are required to develop new vaccine candidates effective against Fasciola.

Infections with the liver fluke Fasciola hepatica may result in considerable economic losses in the dairy livestock industry worldwide. Infections have been associated with detrimental impacts on milk production and milk quality as well as reduced fertility. However, most related data rely on examinations on herd level and the rather few studies on individual cow level are based solely on antibodies as measure for F. hepatica infections. This entails the risk of including false-positives as anti-F. hepatica antibodies persist for months even if the infection is cleared. Therefore, the presented study aimed to overcome this issue by assessing the association between F. hepatica infections measured via faecal egg counts (FEC) and milk production as well as fertility parameters in individual dairy cows. In total, 2006 faecal samples from 1166 Black and White dairy cows from 17 small and medium-sized German grassland farms were examined in July and September 2015. The relationship between patent F. hepatica infections and the milk production parameters milk yield, milk protein content, milk fat content and somatic cell score (SCS) was assessed in a linear mixed model using test-day records of individual cows. Patent F. hepatica infections were found on 35.3% (7/17) of farms with an individual cow prevalence of 10.1% (97/963) in July and 9.1% (95/1036) in September. Patent rumen fluke infections were detected on 17.6% (3/17) farms with an individual cow prevalence of 0.4% (4/963) in July and 0.7% (9/1036) in September. No significant association was found between F. hepatica infection status and either SCS as an indicator of udder health or milk production parameters, despite 0.06 and 0.10% lower values for milk protein and fat content in patently infected cows. Linear mixed models and generalized linear mixed models were established to estimate the impact of fasciolosis on the fertility parameters calving to first service (CTFS), calving interval (CI), success in first insemination (SFI) and 56-day nonreturn rate (NRR56). A significantly higher average CTFS of 4.69 days was detected in F. hepatica infected cows (P = 0.025), but no significant relationship was found for the other fertility parameters.

Objective: The aim of this study was to investigate the prevalence of fascioliasis in humans, cattle, sheep, and goats, done by a systematic review in Iran. Methods: Fifty articles were extracted including Iranian papers such as Google scholar, Magiran, Iran Medex, SID, and Pubmed. Out of these, 21 articles were selected for meta-analysis. Essential information for meta-analysis was extracted from papers and archived in Excel software for calculating by statistical analysis. The variance of each study was obtained using the binomial distribution. Heterogeneity of the studies was surveyed using I2 index. Data were analyzed using a random effect model. Results: Of 21 collected papers, 1,275,506 samples from cow (507,152), sheep (454,882), goat (207,925), and human (105,547) had been surveyed in Iran. Eight studies were conducted on humans and 13 on animals. The prevalence rate obtained in humans was 3% with a confidence interval (CI) of 95% (1%–7%). Prevalence rate obtained in cows, sheep, and goats was 13% with CI of 95% (10%–16%). The highest level of prevalence was reported from cities in the North in animals with a prevalence of 14%. The highest level of prevalence was reported from Gilan in humans with a prevalence of 0.1%. Conclusion: The prevalence of the Fascioliasis in Iran has reduced in recent years, But the importance of the disease has not reduced and there is a possibility of an epidemic. Furthermore, in many cities of Iran, there is no study on the disease.

El analisis e interpretacion de los resultados demostro que 146 muestras fueran positivas a F. hepatica (32,09 %). Todas las vaquerias presentaron animales infectados con rangos de prevalencia entre las vaquerias que oscilaron de 9,5 % al 84 %. Tambien fue posible encontrar las dos especies de hospederos intermediarios de este parasito en Cuba, Galba cubensis y Pseudosuccinea columella. Ten dairy farms in western Cuba were studied to assess the prevalence in both intermediate and definitive hosts. A single stool sample from 455 dairy cows was taken and studied. The animals were randomly selected and the samples were kept at -20oC until infection was determined. Detection of the Fasciola hepatica infection was carried out using a non-commercial sandwich-ELISA called FasciDIG®. Results showed that 146 samples were positive for F. hepatica (32.09%) with every farm testing positive for F. hepatica infection, while prevalence ranged from 9.5% to 84% among farms. It was also possible to detect the presence of the two species of intermediate hosts of this trematode in Cuba, Galba cubensis and Pseudosuccinea columella. Bovine tuberculosis (bTB), caused by Mycobacterium bovis, remains a persistent problem for cattle industries in endemic countries. The frequency, quality, and performance of tests, and the presence of wildlife reservoirs, have been identified as impediments to eradication. Recently, exposure to helminth infection (Fasciola hepatica) has been associated negatively with the disclosure of bTB. Here, for the first time, we assess impact of concurrent infections of Fasciola hepatica and the disclosure of bTB at the animal-level using large surveillance datasets. We utilized a dataset of 138,566 animal records from an abattoir from Northern Ireland (2011-2013). The presence of F. hepatica infection was assessed from macroscopic tissue inspection at abattoir. Multivariable models were developed to assess co-infection associations with bTB status based on: Single Intradermal Comparative Tuberculin Test (SICTT), lesion, bacteriological confirmation, including either all animals, or only skin-test negative animals (lesions at routine slaughter; LRS; confirmed nonreactors at routine slaughter; cNRs) or positive (reactors) animals alone, respectively. The relationship between skin tuberculin reaction sizes and fluke status was also explored for a subset of animals with field recordings (n = 24,680). Controlling for known risk factors (e.g., climatic, herd, and individual level characteristics), we did not find significant associations between the SICTT (standard or severe interpretation), lesion, nor confirmation status of animals and their liver fluke status. The only exception was a negative association between liver fluke positivity, and LRS or cNRs, respectively; though effect-sizes were small (e.g., LRS Odds-Ratio: 0.87; 95% CI: 0.76-1.00). There was limited evidence of a relationship between tuberculin reaction sizes during SICTT testing and liver fluke infection status. These data do not support the contention that the detection of bTB using skin-tests or reactor postmortem follow-up may be compromised by co-infection at a population level, but the relationship with lesion formation (pathogenesis) may indicate an impact for postmortem surveillance.

Entre mayo y agosto del 2006, se realizó un estudio sobre prevalencia del parasitismo intestinal y factores epidemiológicos en 205 escolares del centro poblado de Pucchún del distrito Mariscal Cáceres, Provincia Camaná, Departamento de Arequipa, Perú. Se aplicó una encuesta epidemiológica y se recolectó una sola muestra de heces a cada escolar que fue procesada en el laboratorio mediante los métodos de Teleman modificado y de sedimentación rápida. Se encontró un 92,68% de prevalencia a uno o más parásitos intestinales, con el predominio de los protozoos frente a los helmintos: Entamoeba histolytica/E.dispar (33,17%), Giardia intestinalis (31,71%), Hymenolepis nana (47,17%), Diphyllobothrium pacificum (0,49%) y Fasciola hepatica (0,49%); siendo más frecuente el poliparasitismo con 42,63%. La prevalencia para los parásitos no patógenos fue: Entamoeba coli (72,68%), Blastocystis hominis (48,29%), Endolimax nana (35,61%), Iodamoeba butschlii (23,90%) y Chilomastix mesnilli (16,58%). Los principales factores epidemiológicos que influyen en la prevalencia del parasitismo intestinal en los escolares son: edad entre 8 a 10 años, eliminación de basuras en el peridomicilio y campo abierto, presencia de roedores y vectores, y consumo de verduras tanto crudas como cocidas.

Background Pasture management influences the prevalence and impact of the pasture parasites (PP) in cattle herds, which cause production-limiting disease worldwide. Evaluating farmer management strategies is vital when considering sustainable PP control practices. The aim of this questionnaire-based study was to describe the pasture management and control strategies regarding PP in Norwegian beef cattle (BC) and dairy cattle (DC) production systems with a focus on gastrointestinal nematodes (GIN) and Fasciola hepatica. Results A total of 745 responses from BC (return rate 20.5%) and 1347 responses from DC farmers (30.7%) were included. The mean total pasture time for DC was 4.2 months for first-season grazers and 4.3 months for second-season grazers and cows, while the corresponding finding in BC was 5.4 months. Home pasture was used for most of the pasture period, particularly for first-season grazer dairy heifers (81%), which were also commonly grazed on the same pasture every year (79%). For most farmers it was necessary for grazing areas to be used for cattle for more than one season (77% of BC farmers and 89% of DC farmers). However, changing the pasture during the season was common in both DC (67%) and BC (70%) herds. The majority of DC farmers (60%) stated that they did not consider that they had a problem with PP. Of the remaining 40%, few respondents could specify whether their herds had a problem due to infection by GIN (11%) or liver flukes (12%). Treatment for GIN was performed by 52% of DC and 34% of BC farmers. Diagnostic faecal samples were collected upon suspicion of parasitic disease by 5% of DC and 16% of BC farmers. Veterinarians were stated as a central source of information about parasite management and treatment. Conclusions Potential risks for exposure to PP were identified, such as use of the same pasture every year for first-season grazers and frequent use of home pasture. The perception of problems related to PP appeared low. Regular anthelmintic treatment without concurrent use of diagnostic faecal samples seems to be common practice.

Fasciolosis, a global parasitic disease of agricultural livestock, is caused by the liver fluke Fasciola hepatica. Management and strategic control of fasciolosis on farms depends on early assessment of the extent of disease so that control measures can be implemented quickly. Traditionally, this has relied on the detection of eggs in the faeces of animals, a laborious method that lacks sensitivity, especially for sub-clinical infections, and identifies chronic infections only. Enzyme linked immunosorbent assays (ELISA) offer a quicker and more sensitive serological means of diagnosis that could detect early acute infection before significant liver damage occurs. The performance of three functionally-active recombinant forms of the major F. hepatica secreted cathepsins L, rFhCL1, rFhCL2, rFhCL3, and a cathepsin B, rFhCB3, were evaluated as antigens in an indirect ELISA to serologically diagnose liver fluke infection in experimentally and naturally infected sheep. rFhCL1 and rFhCL3 were the most effective of the four antigens detecting fasciolosis in sheep as early as three weeks after experimental infection, at least five weeks earlier than both coproantigen and faecal egg tests. In addition, the rFhCL1 and rFhCL3 ELISAs had a very low detection limit for liver fluke in lambs exposed to natural infection on pastures and thus could play a major role in the surveillance of farms and a 'test and treat' approach to disease management. Finally, antibodies to all three cathepsin L proteases remain high throughout chronic infection but decline rapidly after drug treatment with the flukicide, triclabendazole, implying that the test may be adapted to trace the effectiveness of drug treatment.
