Unlabelled: Occasional transmission of highly pathogenic avian H5N1 influenza viruses to humans causes severe pneumonia with high mortality. To better understand the mechanisms via which H5N1 viruses induce severe disease in humans, we infected cynomolgus macaques with six different H5N1 strains isolated from human patients and compared their pathogenicity and the global host responses to the virus infection. Although all H5N1 viruses replicated in the respiratory tract, there was substantial heterogeneity in their replicative ability and in the disease severity induced, which ranged from asymptomatic to fatal. A comparison of global gene expression between severe and mild disease cases indicated that interferon-induced upregulation of genes related to innate immunity, apoptosis, and antigen processing/presentation in the early phase of infection was limited in severe disease cases, although interferon expression was upregulated in both severe and mild cases. Furthermore, coexpression analysis of microarray data, which reveals the dynamics of host responses during the infection, demonstrated that the limited expression of these genes early in infection led to a failure to suppress virus replication and to the hyperinduction of genes related to immunity, inflammation, coagulation, and homeostasis in the late phase of infection, resulting in a more severe disease. Our data suggest that the attenuated interferon-induced activation of innate immunity, apoptosis, and antigen presentation in the early phase of H5N1 virus infection leads to subsequent severe disease outcome., Importance: Highly pathogenic avian H5N1 influenza viruses sometimes transmit to humans and cause severe pneumonia with ca. 60% lethality. The continued circulation of these viruses poses a pandemic threat; however, their pathogenesis in mammals is not fully understood. We, therefore, investigated the pathogenicity of six H5N1 viruses and compared the host responses of cynomolgus macaques to the virus infection. We identified differences in the viral replicative ability of and in disease severity caused by these H5N1 viruses. A comparison of global host responses between severe and mild disease cases identified the limited upregulation of interferon-stimulated genes early in infection in severe cases. The dynamics of the host responses indicated that the limited response early in infection failed to suppress virus replication and led to hyperinduction of pathological condition-related genes late in infection. d‐Aspartate oxidase (DDO) is a degradative enzyme that acts stereospecifically on free acidic D‐amino acids such as d‐aspartate and d‐glutamate. d‐Aspartate plays an important role in regulating neurotransmission, developmental processes, hormone secretion, and reproductive functions in mammals. In contrast, the physiological role of d‐glutamate in mammals remains unclear. In Caenorhabditis elegans, the enzyme responsible for in vivo metabolism of d‐glutamate is DDO‐3, one of the three DDO isoforms, which is also required for normal self‐fertility, hatching, and lifespan. In general, eukaryotic DDOs localize to subcellular peroxisomes in a peroxisomal targeting signal type 1 (PTS1)‐dependent manner. However, DDO‐3 does not contain a PTS1, but instead has a putative N‐terminal signal peptide (SP). In this study, we found that DDO‐3 is a secreted DDO, the first such enzyme to be described in eukaryotes. In hermaphrodites, DDO‐3 was secreted from the proximal gonadal sheath cells in a SP‐dependent manner and transferred to the oocyte surface. In males, DDO‐3 was secreted from the seminal vesicle into the seminal fluid in a SP‐dependent manner during mating with hermaphrodites. In both sexes, DDO‐3 was secreted from the cells where it was produced into the body fluid and taken up by scavenger coelomocytes. Full‐length DDO‐3 transgene rescued all phenotypes elicited by the deletion of ddo‐3, whereas a DDO‐3 transgene lacking the putative SP did not. Together, these results indicate that secretion of DDO‐3 is essential for its physiological functions. Acidic sphingomyelinase (ASMase) catalyses the generation of ceramide from sphingomyelin. Ceramide is a lipid mediator and is implicated in mediating and regulating various cellular processes including cell proliferation, differentiation, stress response and inflammation. We have previously reported that electrophiles including diethyl maleate (DEM), heavy metals and cigarette smoke extracts induced ASMase expression in human bladder carcinoma ECV-304 cells, but the mechanism of ASMase mRNA induction by electrophiles remains unknown. In this study, we clarified the involvement of NF-E2-related factor 2 (Nrf2) in the induction of ASMase mRNA by DEM. Promoter analysis using a series of deletion mutants of the human ASMase gene showed that ARE-like element1 located in a region between -200 and -160 bp upstream of the transcription start point is mainly a DEM-responsive element. Moreover, an electrophoretic mobility shift assay using ARE-like element1 revealed that Nrf2 is a candidate transcription factor that binds to ARElike element1 in response to DEM. Finally, alteration of Nrf2 expression by overexpression and knockdown could regulate the induction of ASMase mRNA by DEM. This is the first evidence that supports the possibility that sphingolipid metabolism is affected via the induction of ASMase by the Nrf2 pathway. [ABSTRACT FROM AUTHOR]

Purpose: To evaluate the influence of glutathione peroxidase 4 (GPx4) expression in retinal pigment epithelium (RPE)/choroid tissue using a mouse model of laser-induced choroidal neovascularization (CNV). Methods: In this study, GPx4+/−, GPx4+/+, and GPx4-overexpressing transgenic mice were created for comparison. The mRNA and protein expression of vascular endothelial growth factor (VEGF)-A in RPE/choroid tissue were evaluated before and after CNV induction by laser. Moreover, we investigated the changes in the VEGF-A mRNA level in RPE/choroid tissue in the CNV model that have not been clearly shown previously. Lipid peroxidation in RPE/choroid tissue was evaluated by immunohistochemistry using antibody against 4-hydroxy-2-nonenal. To investigate the protective role of GPx4, the size of laser-induced CNV was compared on day 7 among the mice expressing different levels of GPx4. Results: In the laser-induced CNV mouse model, laser treatment reduced the VEGF-A mRNA level in RPE/choroid tissue, while it increased the VEGF-A protein level. Evaluation of VEGF-A expression in RPE/choroid tissue of the GPx4+/−, GPx4+/+, and GPx4 transgenic mice revealed that GPx4 increased the VEGF-A protein level under physiological conditions (i.e., without laser treatment), while GPx4 suppressed the increase in the VEGF-A protein level under pathological conditions (i.e., after CNV induction by laser). In addition, GPx4 reduced the CNV size in a dose-dependent manner in vivo. Conclusions: GPx4 suppresses the increase in the VEGF-A protein level, which occurs during the development of pathological CNV, thus partly explaining the protective effect of GPx4 against CNV. Abstract: Mitochondria are major compartments in cells responsible for generating reactive oxygen species, which can cause the development of diabetes, Parkinson''s disease and premature aging. Antioxidant systems in mitochondria are important for the prevention of diseases and reduction in the speed of aging. We investigated whether the reactive oxygen species generated in mitochondria induced the expression of metallothionein as an antioxidant. We compared the expression level of metallothionein mRNA in mitochondrial phospholipid hydroperoxide glutathione peroxidase (PHGPx)-overexpressed (PHGPx-ov) cells with that in control cells. These cells were treated with respiratory inhibitors, including rotenone and 2, 4-dinitrophenol; under these conditions, the PHGPx-ov cells were more resistant to cell death than the control cells. In addition, the intracellular reactive oxygen species level that was induced by these inhibitors was lower in PHGPx-ov cells than in control cells. This indicates that PHGPx degrades the membrane phospholipid hydroperoxide that is formed via the reactive oxygen species generated in mitochondria. The enhanced expression of metallothionein-I and metallothionein-II mRNA in rotenone-treated control cells was significantly decreased in rotenone-treated PHGPx-ov cells, suggesting that the hydrogen peroxide that is formed by superoxide anions generated in mitochondria diffuse into the cytosol and induce metallothionein mRNA expression. Conversely, the expression of manganese-superoxide dismutase (Mn-SOD) mRNA, which is localized in mitochondria, was not correlated with the intracellular reactive oxygen species level that was induced by rotenone treatment. These results suggest that metallothionein expression is sensitively and strictly regulated by the oxidative state that is induced by mitochondrial respiration. [Copyright &y& Elsevier]
