Sheath rot disease seriously limits rice productivity in several countries. In this study, Fusarium spp. were proven to be major pathogens responsible for sheath rot disease in Indonesia. The disease was initially found in 13 provinces in Indonesia. A total of 287 fungal isolates was obtained from 900 sampled leaf sheaths, and all of them were shown to be pathogenic to rice plants by artificial inoculation. Molecular identification of sheath rot pathogens was performed using the translation elongation factor 1-alpha (TEF1-α) gene sequencing. Morphological characterization was conducted using synthetic low-nutrient agar (SNA) using filter paper. Species within the Fusarium incarnatum-equiseti species complex (FIESC) and the Fusarium fujikuroi species complex (FFSC) were the major taxa found to be associated with the disease. The FIESC included Fusarium bubalinum, F. hainanense, F. sulawesiense and F. tanahbumbuense. The FFSC included F. andiyazi, F. fujikuroi, F. proliferatum, F. pseudocircinatum, and F. sacchari. Fusarium grosmichelii, a member of the Fusarium oxysporum species complex (FOSC), and a species within the Fusarium solani species complex (FSSC) were also found to be associated with the disease. The species in the FIESC and the FFSC are morphologically difficult to distinguish; however, some FFSC species have some diagnostic traits. This is the first study to identify the Fusarium spp. causing sheath rot disease in Indonesia through field survey, fungal isolation, pathogenicity test, molecular identification, and morphological characterization. Two extracellular alkalistable 1,4-β-d-glucan-4-glucanohydrolase (EC 3.2.1.4) fractions, namely Endo A and Endo B were separated from the culture filtrate of an alkalotolerant Fusarium strain by gel chromatography on Bio Gel P-100 column. The viscometric characterization of enzyme indicated that the Endo B was 1.9 times more random in action as compared to Endo A. Both the endoglucanases showed comparable patterns of their ability to release short fibers in the reaction mixture when filter paper was used as the substrate. The average size of released short fibers was determined by scanning electron microscopy ranged between 20 and 100 μm. Adsorption characteristics of the endoglucanases indicated that the enzyme complex acts prominently at the surface of substrate rather than at their cracked edges. The X-ray diffractograms showed that the crystallinity of the substrates does not vary significantly in the initial stages of the enzymatic action. The extracellular enzyme preparation was found to be suitable for deinking of mixed office waste papers. The enzyme treatment resulted in the increase in brightness with the reduction in ink counts of the recycled paper. Pea (Pisum sativum) is an economically important legume crop, cultivated worldwide due to its high nutritional value and diverse culinary applications. However, like any agricultural crop, peas are vulnerable to various biotic and abiotic stresses, including fungal infection. The present paper deals with a study to identify the mycoflora associated with pea seed. The seed borne mycoflora of pea was examined by agar plate method, blotter method and Deep freezing blotter paper method. A total 17 species of fungi viz. Alternaria alternata, Aspergillus niger, Aspergillus flavus, Aspergillus fumigatus, Aspergillus candidus, Curvularia sp., Penicillium expansum, Mucor spp., Rhizopus nigricans, Rhizophus stolnifer, Fusarium monliforme, Fusarium oxysporum, Fusarium solani, Macrophomina phaseolina, Rhizoctonia solani, Stemphylium sp., Trichoderma harzianum and some unidentified species were reported. Among the method used for detection of seed borne fungi, the standard blotter paper method is more effective followed by agar plate method and deep freeze blotter paper method. This study was conducted to investigate the antagonistic potential of endophytic and rhizospheric bacterial isolates obtained from Citrullus colocynthis in suppressing Fusarium solani and Pythium aphanidermatum and promoting the growth of cucumber. Molecular identification of bacterial strains associated with C. colocynthis confirmed that these strains belong to the Achromobacter, Pantoea, Pseudomonas, Rhizobium, Sphingobacterium, Bacillus, Sinorhizobium, Staphylococcus, Cupriavidus, and Exiguobacterium genera. A dual culture assay showed that nine of the bacterial strains exhibited antifungal activity, four of which were effective against both pathogens. Strains B27 (Pantoea dispersa) and B28 (Exiguobacterium indicum) caused the highest percentage of inhibition towards F. solani (48.5% and 48.1%, respectively). P. aphanidermatum growth was impeded by the B21 (Bacillus cereus, 44.7%) and B28 (Exiguobacterium indicum, 51.1%) strains. Scanning electron microscopy showed that the strains caused abnormality in phytopathogens' mycelia. All of the selected bacterial strains showed good IAA production (>500 ppm). A paper towel experiment demonstrated that these strains improved the seed germination, root/shoot growth, and vigor index of cucumber seedlings. Our findings suggest that the bacterial strains from C. colocynthis are suppressive to F. solani and P. aphanidermatum and can promote cucumber growth. Background: Vermicomposting is the agricultural technique of conversion of organic wastes to a fertile product, which can result in better crop growth and production. However, even though earthworms are the main organisms participating in the process, the microbes associated with it also have an important role to play. These microbes degrade the waste products biochemically and are responsible of the conversion processes. Few studies are carried out on microbial diversity and related enzymes activities in the vermicompost prepared from different organic waste materials. Methods: In this paper, we isolated both bacteria and fungi from seven different types of vermicompost, using different selective media. We also studied the activity of hydrolytic enzymes that are associated with the isolated microbes. Result: It was observed that bacteria like Bacillus sp., Pseudomonas sp., Klebsiella sp., Staphylococcus aureus, Streptococcus, Micrococcus, Actinomycetes, Pigment producing Actinomycetes, Streptomyces, Azotobactor and fungi like Penicillium purpurogenum, Aspergillus sp., Alternaria alternata, Fusarium solani, Rhizopus sp., Mucor hiemalis, Myrothecium verrucaria etc. were present in our vermicompost preparations. The presence of nitrogen fixing bacteria, phosphate solubilizing microorganisms and PGPR indicated the good fertilizer value of the vermicompost samples. Background: Apple (Malus domestica Borkh.) is an important economic crop. The pathological effects of Fusarium solani, a species complex of soilborne pathogens, on the root systems of apple plants was unknown. It was unclear how mycorrhizal apple seedlings resist infection by F. solani. The transcriptional profiles of mycorrhizal and non-mycorrhizal plants infected by F. solani were compared using RNA-Seq. Results: Infection with F. solani significantly reduced the dry weight of apple roots, and the roots of mycorrhizal apple plants were less damaged when the plants were infected with F. solani. They also had enhanced activity of antioxidant enzymes and a reduction in the oxidation of membrane lipids. A total of 1839 differentially expressed genes (DEGs) were obtained after mycorrhizal and non-mycorrhizal apple plants were infected with F. solani. A gene ontogeny (GO) analysis showed that most of the DEGs were involved in the binding of ADP and calcium ions. In addition, based on a MapMan analysis, a large number of DEGs were found to be involved in the response of mycorrhizal plants to stress. Among them, the overexpressed transcription factor MdWRKY40 significantly improved the resistance of the apple 'Orin' callus to F. solani and the expression of the resistance gene MdGLU by binding the promoter of MdGLU. Conclusion: This paper outlines how the inoculation of apple seedlings roots by arbuscular mycorrhizal fungi responded to infection with F. solani at the transcriptional level. Cereal caryopses are rich in 5-n-alk(en)ylresorcinols, antimicrobial compounds. In this paper, the correlation between the presence of resorcinolic lipids on the surface of cereal grains and the susceptibility of their seedlings to infection by Rhizoctonia solani or Fusarium culmorum was evaluated. The declines in length of both the roots and coleoptiles were observed in barley seedlings of Scarlett and Rabel cultivars grown from the wax-impoverished seeds infected with F92 and F93 strains of Rhizoctonia solani, respectively. Regarding wheat, R. solani F93 significantly reduced only the coleoptile growth. Resorcinolic lipids, being the mixture of homologues with C17–C25 carbon chains, were the only compounds washed off wheat caryopses by chloroform. Moreover, the better anti-Rhizoctonia solani F93 activity of 5-n-alk(en)ylresorcinols of wheat grains than that of rye caryopsis lipids was proven by the poisoned medium technique. Two saturated homologues (C21:0 and C23:0) were the most effective inhibitors of the mycelial growth of this fungus. Thus, the susceptibilities of barley and wheat seedlings to some fungal pathogens have been found to be related to the content and composition of 5-n-alk(en)ylresorcinols in the waxy layer of cereal grains, confirming the protective role of these compounds, during the early stages of cereal development. The food and pharmaceutical industries are searching for natural colour alternatives as required by consumers. Over the last decades, fungi have emerged as producers of natural pigments. In this paper, five filamentous fungi; Penicillium multicolour, P. canescens, P. herquie, Talaromyces verruculosus and Fusarium solani isolated from soil and producing orange, green, yellow, red and brown pigments, respectively, when cultured on a mixture of green waste and whey were tested. The culture media with varying pH (4.0, 7.0 and 9.0) were incubated at 25 °C for 14 days under submerged and solid-state fermentation conditions. Optimal conditions for pigment production were recorded at pH 7.0 and 9.0 while lower biomass and pigment intensities were observed at pH 4.0. The mycelial biomass and pigment intensities were significantly higher for solid-state fermentation (0.06-2.50 g/L and 3.78-4.00 AU) compared to submerged fermentation (0.220-0.470 g/L and 0.295-3.466 AU). The pigment intensities were corroborated by lower L* values with increasing pH. The γmax values for the pigments were all in the UV region. Finally, this study demonstrated the feasibility of pigment production using green waste:whey cocktails (3:2). The potentials of Luffa aegyptiaca and its rhizospheric, non-mycorrhizal fungi in biodegrading and bio-remediating hydrocarbon contaminated soil were investigated in-vitro and in-situ. Biodegradation study was done in two stages: preliminary study using hydrocarbon treated filter paper and in-vitro with Mineral Salt Media (MSM) read on Spectrophotometer at two photo synthetically active wavelengths (530 nm and 620 nm) while rhizoremediation study was done in-situ in contaminated plot of land. Hydrocarbon utilization ability of the fungi and plant were confirmed using total petroleum hydrocarbon (TPH) analysis and gas chromatography mass spectroscopy (GC-MS). Results show differing rates of hydrocarbon utilization by isolated fungi. In-vitro biodegradation study showed that Aspergillus niger, Fusarium solani, Curvularia lunata and Trichoderma harzianum were best in degrading kerosene (78%), diesel (70%), spent engine oil (83%) and crude oil (77%) respectively. Rhizoremediation study using L. aegyptiaca and C. lunata show that remediation was enhanced to 72.15% as against 32.32% and 14% when only the plant or fungus is used respectively. Hydrocarbon accumulation by L. aegyptiaca also decreased in the presence of the fungus. Curvularia lunata is shown in this study to enhance the germination, survival, growth and bioremediation efficiency of L. aegyptiaca in polluted environment. Novelty statement The potentials of Curvularia lunata, a non-mycorrhizal fungi associated with L. aegyptiaca in survival, growth and phytoremediation of petroleum hydrocarbon polluted soil by L. aegyptiaca is highlighted in this study. The herbicide butachlor has been used in huge quantities worldwide, affecting various environmental systems. Butachlor residues have been detected in soil, water, and organisms, and have been shown to be toxic to these non-target organisms. This paper briefly summarizes the toxic effects of butachlor on aquatic and terrestrial animals, including humans, and proposes the necessity of its removal from the environment. Due to long-term exposure, some animals, plants, and microorganisms have developed resistance toward butachlor, indicating that the toxicity of this herbicide can be reduced. Furthermore, we can consider removing butachlor residues from the environment by using such butachlor-resistant organisms. In particular, microbial degradation methods have attracted much attention, with about 30 kinds of butachlor-degrading microorganisms have been found, such as Fusarium solani, Novosphingobium chloroacetimidivorans, Chaetomium globosum, Pseudomonas putida, Sphingomonas chloroacetimidivorans, and Rhodococcus sp. The metabolites and degradation pathways of butachlor have been investigated. In addition, enzymes associated with butachlor degradation have been identified, including CndC1 (ferredoxin), Red1 (reductase), FdX1 (ferredoxin), FdX2 (ferredoxin), Dbo (debutoxylase), and catechol 1,2 dioxygenase. However, few reviews have focused on the microbial degradation and molecular mechanisms of butachlor. This review explores the biochemical pathways and molecular mechanisms of butachlor biodegradation in depth in order to provide new ideas for repairing butachlor-contaminated environments. Cnidium officinale Makino, a perennial crop in the Umbeliperae family, is one of Korea's representative forest medicinal plants. However, the growing area of C. officinale has been reduced by plant disease and soil sickness caused by fusarium wilt. This study isolated rhizosphere bacteria from C. officinale, and their antagonistic activity was evaluated against Fusarium solani. Particularly, four isolated strains, namely, PT1, ST7, ST8, and SP4, showed a significant antagonistic activity against F. solani. An in planta test showed that the mortality rates of shoots were significantly low in the PT1-inoculated group. The fresh and dry weights of the inoculated plants were also higher than that of the other groups. The 16S rRNA gene sequencing identified the strain PT1 as Leclercia adecarboxylata, and downstream studies confirmed the production of antagonism-related enzymes such as siderophore and N-acetyl-β-glucosaminidase. The phosphorous solubilizing ability and secretion of related enzymes were also analyzed. l-Tyrosine is a versatile compound used in the fine chemical, pharmaceutical, and functional food industries. Here, we report a bi-enzymatic cascade involving alanine racemase (ALR) and d-amino acid oxidase (DAAO) to produce pyruvate, as co-substrate for l-tyrosine production, from the cheap substrate l-alanine. The BpALR (ALR from Bacillus pseudofirmus) was used as a whole-cell biocatalyst, converting l-alanine to d, l-alanine. The FsDAAO (DAAO from Fusarium solani) was immobilized to oxidize the d-alanine generated in the first step to pyruvate. Both systems were combined as a continuous-flow reactor for maximized l-alanine-to-pyruvate conversion rates. The optimal parameters and appropriate conditions for FsDAAO immobilization were investigated. The pyruvate concentration of 86.6 g/L was achieved within 17 h. Subsequently, a whole-cell biocatalyst system for l-tyrosine production, catalyzed by the tyrosine phenol-lyase (TPL) from Erwinia herbicola (EhTPL), was developed, and a fed-batch approach was applied with phenol and the pyruvate produced with the ALR/DAAO system mentioned above. The concentration of phenol and pyruvate in the reactor should not exceed 7.5 g/L and 10 g/L, respectively. Significantly, the l-tyrosine concentration of 152.5 g/L was achieved within 10 h, demonstrating the great potential for high-efficiency production of l-tyrosine through the approach we established in this paper. The fungal kingdom has been widely studied as a source of bioactive compounds of interest to the pharmaceutical and food industry. This paper studies the production of natural red pigments by Fusarium solani BRM054066 in the submerged fermentation system, using Doehlert experimental design to determine optimal cultivation conditions. The chemical composition of the red pigment was determined by Nuclear Magnetic Resonance spectroscopy (NMR) and Ultra-Performance Liquid Chromatography coupled to Mass Spectrometry (UPLC-MS). Antioxidant activity was assessed by the ability to sequester of free radical DPPH. In the analysis of anti-inflammatory activity, murine peritoneal macrophages activated by LPS were used, and the gene expression of TNF-α, IL-1β, IL-6, IL-10 and IL-17 was determined using qPCR. As a result, it was found that agitation at 200 rpm and glucose concentration ≥ 20 g/L promote the best results in the production of red pigment. The chemical compounds identified were two naphthoquinones, fusarubin and dihydrofusarubin, and an anthraquinone, a bostrycoidin, being fusarubin the majority compound. The red pigment showed antioxidant activity by scavenge 50% of the DPPH radical, in a concentration of 24 µg/mL. The pigment also showed an effective anti-inflammatory capacity by reducing the overexpression of the pro-inflammatory cytokines TNF-α, IL-1β and IL-6 and promoting the production of anti-inflammatory IL-10 and IL-17, in murine macrophages activated by LPS (p < 0.05). According to the results, the fungus F. solani BRM054066, under optimized conditions of cultivation, proved to be a promising source of biologically active natural pigments with wide industrial applicability. This paper documents six new saprobic marine fungi and one new genus based on morphology and multi-gene phylogenies. Three Dothideomycetes, and members of the Pleosporales, are introduced: Pseudomassariosphaeria triseptata sp. nov. was recognized as a mangrove species in Amniculicolaceae, and Salsuginea phoenicis sp. nov. was discovered as a second member of Salsugineaceae. A new genus Raghukumaria with Raghukumaria keshaphalae sp. nov., recovered from mangroves, is phylogenetically sister to Halomassarina and nests in the Trematosphaeriaceae. Three new species are referred to the Sordariomycetes: Coniochaeta marina (Coniochaetales, Coniochaetaceae) on driftwood; Fusicolla bharatavarshae (Hypocreales, Nectriaceae) is introduced with asexual and sexual morphs, on decayed mangrove wood of Avicennia marina; and Fusarium sedimenticola (Hypocreales, Nectriaceae) is new to the Fusarium solani species complex (FSSC) from deep-sea sediment. This study was conducted to evaluate the efficiency of the bioagent Trichoderma longibrachiatum in controlling cucumber root rot disease caused by Fusarium solani. The pathogenic fungus was isolated from the roots of cucumber plant infected with root rot disease. The pathogenicity of F. solani was tested in Petri dishes and it was found that it reduced the germination percentage to 50% compared to the control treatment, which was 90%. The results of dual culture showed that the bioagent T. longibrachiatum inhibited the growth of F. solani and the degree of antagonism reached 1 according to Bell scale. The results of the field experiment which conducted in the greenhouse showed that the bioagent T. longibrachiatum led to increase the germination percentage of cucumber plants, reaching 96.7%, which significantly differed from the pathogenic fungus treatment, which amounted to 63.3%. The results indicated that the bioagent T. longibrachiatum significantly reduced the percentage of disease severity to 20.33%, compared with the treatment of the pathogenic fungus F. solani alone, which reached 65.67%. bioagent T. longibrachiatum also increased the plant height significantly up to 177 cm, compared to control treatment and pathogenic fungus treatment, which were 142 and 116 cm, respectively. It was also found that bioagent T. longibrachiatum when it was interacted with the pathogenic fungus led to increase the plant height up to 167 cm, compared with the treatment of pathogenic fungi alone, which was 116 cm. Fresh and dry weight of shoot and root systems also increased significantly in the treatment of T. longibrachiatum compared to the treatment of pathogenic fungus, followed by the treatment of the interaction between T. longibrachiatum and the pathogenic fungus treatments. The results also revealed the efficiency of T. longibrachiatum in increasing plant productivity represented by the number of fruits, the average weight of the fruits and the fruit yield per plant, which were significantly differed from the pathogenic fungus treatment alone. Fritillaria taipaiensis P. Y. Li (F. taipaiensis) is a traditional Chinese herbal medicine that has been used for over two millennia to treat cough and expectoration. However, the increasing cultivation of F. taipaiensis has led to the spread of bulb rot diseases. In this study, pathogens were isolated from rotten F. taipaiensis bulbs. Through molecular identification, pathogenicity testing, morphological assessment, and microscopy, Fusarium solani was identified as the pathogen causing bulb rot in F. taipaiensis. The colonization of F. solani in the bulbs was investigated through microscopic observation. The rapid and accurate detection of this pathogen will contribute to better disease monitoring and control. Loop-mediated isothermal amplification (LAMP) and qPCR methods were established to quickly and specifically identify this pathogen. This study delves into the dynamic interaction between various fungal strains, substrates, and treatment durations to optimize the nutritional value of these by‐products. Six fungi, including Penicillium chrysogenum, Fusarium sp., Fusarium oxysporum, Fusarium solani, Penicillium crustosum, and Cosmospora viridescens, were evaluated across three substrates: wheat straw (WS), cedar sawdust (CW), and olive pomace (OP) over treatment periods of 4, 8, and 12 weeks. The study discerned profound impacts of these fungi across multiple parameters, including cellulose variation (C.var), lignin variation (L.var), and in vitro true digestibility variation (IVTD.var). Our results demonstrated that the various fungi had a significant effect on all parameters (p <.001). Noteworthy, F. oxysporum and F. solani emerged as exemplars, displaying notable lignin degradation, cellulose liberation, and IVTD enhancement. Importantly, P. crustosum demonstrated substantial cellulose degradation, exhibiting optimal efficacy in just 4 weeks for all substrates. Notably, F. sp. excelled, yielding favorable results when treating WS. P. chrysogenum achieved optimal outcomes with 8‐week treatment for WS. Both Fusarium sp. and P. chrysogenum exhibited slight cellulose release, with remarkable reduction of WS lignin compared to other substrates. Especially, WS and OP displayed superior digestibility enhancements relative to CW. It should be noted that the treatment duration further shaped these outcomes, as prolonged treatment (12 weeks) fostered greater benefits in lignin degradation and digestibility, albeit with concomitant cellulose degradation. These findings underscore the intricate balance between fungal strains, substrates, and treatment durations in optimizing the nutritional value of lignocellulosic agro‐industrial by‐products. Strawberry red core root rot disease affects the growth and yield of strawberry (Fragaria ananassa Duch), which leads to economic losses in China. The study employed a tissue separation method to isolate and identify the causal agent responsible for strawberry red core root rot. This was achieved by the observation of its morphological characteristics, sequencing analyses, and pathogenicity tests. The sensitivity of five chemical fungicides against the two species of Fusarium was determined using the mycelial growth rate method, and the biological characteristics of the two species were examined. The pathogens were identified as Fusarium solani and Fusarium oxysporum. The optimal conditions for the mycelial growth of F. solani and F. oxysporum were determined to be potato sucrose agar at 25 °C and pH 6, and potato dextrose agar at 30 °C and pH 8, respectively, with a 24:24 light cycle. The most suitable carbon and nitrogen sources for the mycelial growth of F. solani were sucrose and sodium nitrate (NaNO3), while for F. oxysporum, they were glucose and peptone. A fungicide sensitivity test indicated that Prochloraz had a good inhibitory effect on the growth of F. solani and F. oxysporum with EC50 values of 0.088 mg L−1 and 0.162 mg. The growth inhibition effect of Azoxystrobin to F. solani and Carbendazim to F. oxysporum was not obvious. Lebanon is a small Mediterranean country with different pedoclimatic conditions that allow the growth of both temperate and tropical plants. Currently, few studies are available on the occurrence and diversity of Fusarium species on Lebanese crops. A wide population of Fusarium strains was isolated from different symptomatic plants in the last 10 years. In the present investigation, a set of 134 representative strains were molecularly identified by sequencing the translation elongation factor, used in Fusarium as a barcoding gene. Great variability was observed, since the strains were grouped into nine different Fusarium Species Complexes (SCs). Fusarium oxysporum SC and Fusarium solani SC were the most frequent (53% and 24%, respectively). Members of important mycotoxigenic SCs were also detected: F. fujikuroi SC (7%), F. sambucinum SC (5%), F. incarnatum-equiseti SC (3%), and F. tricinctum SC (4%). Two strains belonging to F. lateritium SC, a single strain belonging to F. burgessii SC, and a single strain belonging to F. redolens SC were also detected. This paper reports, for the first time, the occurrence of several Fusarium species on Lebanese host plants. The clear picture of the Fusarium species distribution provided in this study can pose a basis for both a better understanding of the potential phytopathological and toxicological risks and planning future Fusarium management strategies in Lebanon. Metabolites exploration of the ethyl acetate extract of Fusarium solani culture broth that was isolated from Euphorbia tirucalli root afforded five compounds; 4-hydroxybenzaldehyde (1), 4-hydroxybenzoic acid (2), tyrosol (3), azelaic acid (4), malic acid (5), and fusaric acid (6). Fungal extract as well as its metabolites were evaluated for their anti-inflammatory and anti-hyperpigmentation potential via in vitro cyclooxygenases and tyrosinase inhibition assays, respectively. Azelaic acid (4) exhibited powerful and selective COX-2 inhibition followed by fusaric acid (6) with IC50 values (2.21 ± 0.06 and 4.81 ± 0.14 μM, respectively). As well, azelaic acid (4) had the most impressive tyrosinase inhibitory effect with IC50 value of 8.75 ± 0.18 μM compared to kojic acid (IC50 = 9.27 ± 0.19 μM). Exclusive computational studies of azelaic acid and fusaric acid with COX-2 were in good accord with the in vitro results. Interestingly, this is the first time to investigate and report the potential of compounds 3–6 to inhibit cyclooxygenase enzymes. Medicinal plants are hosts to an infinite number of microorganisms, commonly referred to as endophytes which are rich in bioactive metabolites yielding favorable biological activities. The endophytes are known to have a profound impact on their host plant by promoting the accumulation of secondary metabolites which are beneficial to humankind. In the present study, the fungal endophyte, Fusarium solani (ABR4) from the medicinal plant Tinospora cordifolia, was assessed for its bioactive secondary metabolites employing fermentation on a solid rice medium. The crude ABR4 fungal extract was sequentially purified using the solvent extraction method and characterized using different spectroscopic and analytical techniques namely TLC, UV spectroscopic analysis, HRESI-MS, FTIR, and GC-MS analysis. The GC-MS analysis revealed the presence of pyridine, benzoic acid, 4-[(trimethylsilyl)oxy]-trimethylsilyl ester, hexadecanoic acid trimethylsilyl ester, and oleic acid trimethylsilyl ester. The cytotoxic ability of ABR4 was evaluated by MTT assay against lung cancer (A549) and breast cancer (MCF-7) cell lines. The compounds did not exhibit significant cytotoxicity against the tested cell lines. The endophytic ABR4 extract was evaluated for its antimicrobial potential against human pathogens (S. aureus, B. cereus, E. coli, S. typhimurium, P. aeruginosa, and C. albicans) by recording 47 to 54% inhibition. Taken together, the endophytic fungal strain ABR4 demonstrated a remarkable antimicrobial activity against the tested pathogens. Furthermore, the functional metabolites isolated from the endophytic strain ABR4 reveal its broader usage as antimicrobial agents for newer drug development in the pharmaceutical industry. Shrimp aquaculture is one of the fastest-growing food production sectors in the world, due to its economic importance and ability to meet the protein needs of a growing population. However, bacterial and fungal infection has caused significant problems in the shrimp sector which instigated enormous economic loss for the main producer countries. Probiotic feed additives can enhance the gut health benefits and immune status of shrimp, thus boosting the growth, production efficacy, and disease resistance. Thereafter, the effects of different levels of dietary probiotic, Pediococcus acidilactici (PA), on growth performance, feed consumption, immunological response, antioxidant activities, and disease resistance in Litopenaeus vannamei juveniles against the pathogen, Fusarium solani, were investigated in this study. In 12 hapas (4 treatments × 3 replicates), 240 L. vannamei juveniles (4.0 ± 0.03 g) were randomly allocated into four treatments (T0 = 0 g PA/kg as control, T1 = 2.0 g PA/kg, T2 = 2.5 g PA/kg, and T3 = 3.0 g PA/kg of diet). The results provided that all PA-supplemented groups had considerably higher growth performance (final body weight, weight gains, and percentages), the final number, shrimp biomass (m3), survival rate, and feed intake (p < 0.05) than the control, with T3 being much higher. Regarding the body chemical composition, protein percentage was increased significantly in T3, while lipid was decreased significantly by dietary PA inclusion (p < 0.05). Feed conversion ratio (FCR, ↓) and nonspecific immune responses (↑), beneficial bacteria were improved significantly by dietary PA supplementation (p < 0.05). When challenged with the fungus F. solani, shrimp treated with dietary PA had significantly (p < 0.05) decreased fatality rates. Overall, the current investigation discovered that high doses of dietary PA increased growth status, feed utilization, gut microbiota, and health status of L. vannamei and disease resistance against F. solani. The thermophilic fungus Gymnascella thermotolerans GTE-21 produces a significant amount of both volatile and nonvolatile compounds with potent antifungal and anticancer properties. Gas chromatography-mass spectrometry (GC-MS) was utilized to identify 14 volatile organic compounds (VOCs) released by G. thermotolerans. The most predominant compound identified was diisooctyl phthalate, constituting 71.99% of the total VOCs, followed by phenol, 2-methoxy-4-(2-propenyl) at 7.12%. The GTE-21's VOCs demonstrated significant antifungal growth inhibition activity of 42.5% against Fusarium solani and 39.03% against Alternaria alternata. Additionally, GTE-21's crude extract exhibited substantial antibacterial effects, evidenced by inhibition zones of 1.5 cm against S. epidermidis, 1.4 cm against C. ciferrii, and 1.3 cm against S. aureus. The CHR2RClR2R:MeOH (1:1) fraction was more effective against human pathogens, producing the largest zones of inhibition of 1.1 and 1.03 cm against S. epidermidis and C. ciferrii, respectively. The evaluation of GTE-21's antifungal activity using the Evans blue assay showed that fungal spores treated with GTE-21's crude extract exhibited 96.2% inhibition at a concentration of 25 mg/ml. Similarly, the fractions CH2Cl2:MeOH (1:1) and CH2Cl2:MeOH (9:1) demonstrated spore inhibition rates of 96.3% and 94.3%, respectively. The EtOAc crude extract demonstrated pronounced cytotoxic effects against the SKOV-3 and MDA-MB-231 cancer cell lines. For SKOV-3, cell viability decreased to 3.40% and 0.47% at concentrations of 50 and 100 μL, respectively. Panax notoginseng (Burke) F. H. Chen, which is commonly referred as ''Sanqi'', is the most widely used Chinese medicinal herb. Root rot can have deleterious effects on the success of Sanqi cultivation, including reductions in yield and quality. Thaumatin-like proteins (TLPs) are large, complex proteins in the pathogenesis-related family 5 (PR-5) that play key roles in the responses of plants to environmental stress (e.g., pathogen infection) and other physiological processes. The aim of this study was to identify the TLP gene family members in the genome of Sanqi and characterize their responses to Fusarium solani infection. A total of 20 PnTLP genes were identified in the Sanqi genome, which were phylogenetically divided into 10 subfamilies (Group I-X). Subcellular localization analysis revealed that most of the PnTLP proteins were in the extracellular space, while a few located in chloroplasts. MEME Suite software was used to characterize the structure of PnTLP genes and identify the conserved domains of PnTLP proteins. Analysis of gene duplications revealed that segmental duplication has played a key role in driving the expansion of the PnTLP family. Analysis of collinear relationships revealed 36 homologous genes between Sanqi and Arabidopsis and nine homologous genes between Sanqi and rice. Prediction of the cis-acting elements of the promoters of PnTLP genes indicated that PnTLP genes are involved in biotic, abiotic stress, and hormone induction. Analysis of expression profiles revealed that TLP genes are involved in plant development and the response of plants to fungal infection. qRT-PCR analyses indicated that the expression of PnTLP genes was up-regulated or down-regulated in response to F. solani infection. The results of this study provide new insights on PnTLP genes and their potential roles in the responses of plants to pathogenic fungi.
