Maize (Zea mays L.) is one of the most widely cultivated grain crops globally. In sub-Saharan Africa (SSA), it plays an important role in ensuring both food and income security for smallholder farmers. This study was conducted to (i) assess the performances of testcross hybrids constituted from maize lethal necrosis (MLN) tolerant doubled haploid (DH) lines under various management conditions; (ii) estimate the combining ability effects and determine the nature of gene action in the DH lines; and (iii) identify DH lines and testcross hybrids for resistance to MLN, high grain yield, and other important traits. Eleven DH lines were crossed with 11 single-cross testers using the line-by-tester mating design, and 115 successful testcross hybrids were generated. These hybrids, along with five commercial check hybrids, were evaluated across four optimum management conditions, two MLN artificial inoculations, and one managed drought environment in Kenya. Under each management condition, the effects of genotypes, environments, and genotype-by-environment interactions were significant for grain yield (GY) and most other traits. Hybrids T1/L3, T10/L3, and T11/L3 exhibited higher grain yields under at least two management conditions. A combining ability analysis revealed that additive gene effects were more important than non-additive effects for GY and most other traits, except for leaf senescence (SEN) and MLN disease severity score. DH line L3 exhibited a desirable general combining ability (GCA) effect for GY, while L5 was the best general combiner for anthesis date (AD) and plant height (PH) across all management conditions. DH lines L2, L6, and L7 showed negative GCA effects for MLN disease severity. Single-cross testers T11 and T10 were good general combiners for GY under all management conditions. Hybrids T2/L11, T9/L10, and T2/L10 demonstrated high specific combining ability (SCA) effects for GY under all conditions. This study identified DH lines and testers with favorable GCA effects for grain yield, MLN resistance, and other agronomic traits that can be used in breeding programs to develop high-yielding and MLN-resistant maize varieties. Better-performing testcross hybrids identified in the current study could be verified through on-farm testing and released for commercial production to replace MLN-susceptible, low-yield hybrids grown in the target ecologies. Summary: Incorporating the dominant male sterile gene, Ms44, in new maize varieties results in 50% non-pollen producing (FNP) varieties. This makes the varieties more nitrogen efficient and increases yield directly by an average of 200 kg ha−1 across yield levels. However, as half of the plants do not shed pollen, the presence of Ms44 in an FNP variety is clearly visible. This technology can improve food production and security in the African maize-based agri-food systems, but only if accepted by farmers. Farmers were therefore invited to 11 on-farm, researcher managed trial sites of FNP varieties in Kenya over 2 years. They were asked to identify the traits they find important in evaluating maize varieties and to score the FNP varieties, as well as their conventional counterparts, on these criteria (including yield, resistance to pests, and cob size) and overall, using a five-point hedonic scale. In total, 2,697 farmers participated, of which 62% were women. Farmers mentioned many traits they find important, especially yield and related traits, early maturity, and drought resistance, but also tassel and pollen formation. In 2017, mid-season, participants scored FNP varieties lower than conventional varieties on tassel and pollen formation, indicating that farmers could distinguish the trait. FNP varieties still received higher scores for yield and overall evaluation. In mid-season 2018, participants no longer scored FNP varieties lower for pollen formation as they now understood the technology. In both years, at the end-season evaluation, scores for tassel formation were not different, but participants scored FNP varieties higher for yield and overall. We conclude that farmers recognized the FNP trait but did not mind it as they clearly favored its yield advantage. The FNP technology, therefore, has high potential not only to increase maize yields, food production, and food security in the agricultural systems of Africa but also to increase varietal turnover and the adoption of new, high-yielding, climate-smart maize hybrids. Key message: Analysis of the genetic architecture of MCMV and MLN resistance in maize doubled-haploid populations revealed QTLs with major effects on chromosomes 3 and 6 that were consistent across genetic backgrounds and environments. Two major-effect QTLs, qMCMV3-108/qMLN3-108 and qMCMV6-17/qMLN6-17, were identified as conferring resistance to both MCMV and MLN. Maize lethal necrosis (MLN) is a serious threat to the food security of maize-growing smallholders in sub-Saharan Africa. The ability of the maize chlorotic mottle virus (MCMV) to interact with other members of the Potyviridae causes severe yield losses in the form of MLN. The objective of the present study was to gain insights and validate the genetic architecture of resistance to MCMV and MLN in maize. We applied linkage mapping to three doubled-haploid populations and a genome-wide association study (GWAS) on 380 diverse maize lines. For all the populations, phenotypic variation for MCMV and MLN was significant, and heritability was moderate to high. Linkage mapping revealed 13 quantitative trait loci (QTLs) for MCMV resistance and 12 QTLs conferring MLN resistance. One major-effect QTL, qMCMV3-108/qMLN3-108, was consistent across populations for both MCMV and MLN resistance. Joint linkage association mapping (JLAM) revealed 18 and 21 main-effect QTLs for MCMV and MLN resistance, respectively. Another major-effect QTL, qMCMV6-17/qMLN6-17, was detected for both MCMV and MLN resistance. The GWAS revealed a total of 54 SNPs (MCMV-13 and MLN-41) significantly associated (P ≤ 5.60 × 10−05) with MCMV and MLN resistance. Most of the GWAS-identified SNPs were within or adjacent to the QTLs detected through linkage mapping. The prediction accuracy for within populations as well as the combined populations is promising; however, the accuracy was low across populations. Overall, MCMV resistance is controlled by a few major and many minor-effect loci and seems more complex than the genetic architecture for MLN resistance. [ABSTRACT FROM AUTHOR]

Breeding for nitrogen use efficiency (NUE) is important to deal with food insecurity and its effect on grain quality, particularly protein. A total of 1679 hybrids were evaluated in 16 different trials for grain yield (GY), grain quality traits (protein, starch and oil content) and kernel weight (KW) under optimum and managed low soil nitrogen fields in Kiboko, Kenya, from 2011 to 2014. The objectives of our study were to understand (i) the effect of low soil N stress on GY and quality traits, (ii) the relationship between GY and quality traits under each soil management condition and (iii) the relationship of traits with low-N versus optimum conditions. From the results, we observed the negative effects of low N on GY, KW and the percentage of protein content, and a positive effect on the percentage of starch content. The correlation between GY and all quality traits was very weak under both soil N conditions. GY had a strong relationship with KW under both optimum and low soil N conditions. Protein and starch content was significantly negative under both optimum and low-N conditions. There was no clear relationship among quality traits under optimum and low N, except for oil content. Therefore, it seems feasible to simultaneously improve GY along with quality traits under both optimum and low-N conditions, except for oil content. However, the negative trend observed between GY (starch) and protein content suggests the need for the regular monitoring of protein and starch content to identify varieties that combine both high GY and acceptable quality. Finally, we recommend further research with a few tropical maize genotypes contrasting for NUE to understand the relationship between the change in grain quality and NUE under low-N conditions. In sub-Saharan Africa, maize is the key determinant of food security for smallholder farmers. The sudden outbreak of maize lethal necrosis (MLN) disease is seriously threatening the maize production in the region. Understanding the genetic basis of MLN resistance is crucial. In this study, we used four biparental populations applied linkage mapping and joint linkage mapping approaches to identify and validate the MLN resistance-associated genomic regions. All populations were genotyped with low to high density markers and phenotyped in multiple environments against MLN under artificial inoculation. Phenotypic variation for MLN resistance was significant and heritability was moderate to high in all four populations for both early and late stages of disease infection. Linkage mapping revealed three major quantitative trait loci (QTL) on chromosomes 3, 6, and 9 that were consistently detected in at least two of the four populations. Phenotypic variance explained by a single QTL in each population ranged from 3.9% in population 1 to 43.8% in population 2. Joint linkage association mapping across three populations with three biometric models together revealed 16 and 10 main effect QTL for MLN-early and MLN-late, respectively. The QTL identified on chromosomes 3, 5, 6, and 9 were consistent with the QTL identified by linkage mapping. Ridge regression best linear unbiased prediction with five-fold cross-validation revealed high accuracy for prediction across populations for both MLN-early and MLN-late. Overall, the study discovered and validated the presence of major effect QTL on chromosomes 3, 6, and 9 which can be potential candidates for marker-assisted breeding to improve the MLN resistance.
