Soil-borne plant diseases cause major economic losses globally. This is partly because their epidemiology is difficult to predict in agricultural fields, where multiple environmental factors could determine disease outcomes. Here we used a combination of field sampling and direct experimentation to identify key abiotic and biotic soil properties that can predict the occurrence of bacterial wilt caused by pathogenic Ralstonia solanacearum. By analyzing 139 tomato rhizosphere soils samples isolated from six provinces in China, we first show a clear link between soil properties, pathogen density and plant health. Specifically, disease outcomes were positively associated with soil moisture, bacterial abundance and bacterial community composition. Based on soil properties alone, random forest machine learning algorithm could predict disease outcomes correctly in 75% of cases with soil moisture being the most significant predictor. The importance of soil moisture was validated causally in a controlled greenhouse experiment, where the highest disease incidence was observed at 60% of maximum water holding capacity. Together, our results show that local soil properties can predict disease occurrence across a wider agricultural landscape, and that management of soil moisture could potentially offer a straightforward method for reducing crop losses to R. solanacearum.

Recent studies have observed differing microbiomes between disease-suppressive and disease-conducive soils. However, it remains unclear whether the microbial keystone taxa in suppressive soil are critical for the suppression of diseases. Bacterial wilt is a common soil-borne disease caused by Ralstonia solanacearum that affects tobacco plants. In this study, two contrasting tobacco fields with bacterial wilt disease incidences of 0% (disease suppressive) and 100% (disease conducive) were observed. Through amplicon sequencing, as expected, a high abundance of Ralstonia was found in the disease-conducive soil, while large amounts of potential beneficial bacteria were found in the disease-suppressive soil. In the fungal community, an abundance of the Fusarium genus, which contains species that cause Fusarium wilt, showed a positive correlation (p < 0.001) with the abundance of Ralstonia. Network analysis revealed that the healthy plants had more complex bacterial networks than the diseased plants. A total of 9 and 13 bacterial keystone taxa were identified from the disease-suppressive soil and healthy root, respectively. Accumulated abundance of these bacterial keystones showed a negative correlation (p < 0.001) with the abundance of Ralstonia. To complement network analysis, culturable strains were isolated, and three species belonging to Pseudomonas showed high 16S rRNA gene similarity (98.4–100%) with keystone taxa. These strains displayed strong inhibition on pathogens and reduced the incidence of bacterial wilt disease in greenhouse condition. This study highlighted the importance of keystone species in the protection of crops against pathogen infection and proposed an approach to obtain beneficial bacteria through identifying keystone species, avoiding large-scale bacterial isolation and cultivation.

Bacterial traits for virulence of Ralstonia solanacearum causing lethal wilt in plants were extensively studied but are not yet fully understood. Other than the known virulence factors of Ralstonia solanacearum, this study aimed to identify the novel gene(s) contributing to bacterial virulence of R. solanacearum. Among the transposon-inserted mutants that were previously generated, we selected mutant SL341F12 strain produced exopolysaccharide equivalent to wild type strain but showed reduced virulence compared to wild type. In this mutant, a transposon was found to disrupt the murI gene encoding glutamate racemase which converts L-glutamate to D-glutamate. SL341F12 lost its motility, and its virulence in the tomato plant was markedly diminished compared to that of the wild type. The altered phenotypes of SL341F12 were restored by introducing a full-length murI gene. The expression of genes required for flagella assembly was significantly reduced in SL341F12 compared to that of the wild type or complemented strain, indicating that the loss of bacterial motility in the mutant was due to reduced flagella assembly. A dramatic reduction of the mutant population compared to its wild type was apparent in planta (i.e., root) than its wild type but not in soil and rhizosphere. This may contribute to the impaired virulence in the mutant strain. Accordingly, we concluded that murI in R. solanacearum may be involved in controlling flagella assembly and consequently, the mutation affects bacterial motility and virulence.

Enset (Ensete ventricosum (Welw.) Cheesman) is one of the Ethiopia’s indigenous sustainability crops supporting the livelihoods of about 20 million people, mainly in the densely populated South and Southwestern parts of the country. Enset serves as a food security crop for humans, animal feed, and source of fiber for the producers. The production of enset has been constrained by plant pests, diseases, and abiotic factors. Among these constraints, bacterial wilt disease has been the most important limiting factor for enset production since its outbreak five decades ago. There is no known bacterial wilt disease resistant genetic material in the enset genetic pool to transfer this trait to susceptible enset varieties through conventional breeding. Moreover, the absence of effective chemicals against the disease has left farmers without means to combat bacterial wilt for decades. Genetic engineering has been the alternative approach to develop disease resistant plant materials in other crops where traditional breeding tools are ineffective. This review discusses enset cultivation and recent developments addressing the control of bacterial wilt disease in enset and related crops like banana to help design effective strategies.

Inhibitory effect of pace extraction on growth of pathogen and development of bacterial wilt of banana disease. Bacterial wilt of banana (Ralstonia solanacearum) is one of banana diseases that potentially destructs banana plants in some areas. A study on the effect of extract of pace plant against bacterial wilt of banana were carried out in laboratory and experimental farm of Plant Protection Department, College of Agriculture, University of Lampung from May until October 2003. The study consisted of in vitro and intact plant experiments. The treatments for both in vitro and intact plants were (1) steril water (control), (2) extract of pace fruit, (3) extract of pace fruit leaves, (4) extract of pace roots and (5) bactericide (agrymycin sulfate). In vitro study was conducted by using completely randomized design, while intact plant study was conducted by using completely randomized block design. The results showed that extract of fruit, leaf and root of pace suppressed the growth of R. solanacearum in vitro. Fruit extract had the highest ability in suppressing the growth of bacterial wilt. However, the application of all kind of extiact did not suppress the incident of bacterial wilt of banana plant.

Bacterial wilt disease caused by Ralstonia solanacearum race 4 is a yield-limiting disease of ginger. A study to investigate the ability of indigenous Arbuscular Mycorrhizae Fungi (AMF) to reduce bacteria wilt disease has been done. This research was designed by using Randomized Block Design (RBD) at greenhouse experiments, using eight AMF isolates with six replication. As supporting data, population R. solanacearum race 4 after treatments was assesed. The results showed that aplication of indigenous AMF to ginger rhizome as seed treatment dicreased disease incidence, support ginger growth and production such as the seedling formation 150%, plant height 98.6%, the number of leaf 115,7%. As the result yield of ginger rhizome increased by 400 % and phyloplan 598,3%, respectively, compared to the control.

International audience; Bacterial wilt caused by Ralstonia solanacearum is one of the most destructive bacterial plant diseases. Although many molecular determinants involved in R. solanacearum adaptation to hosts and pathogenesis have been described, host components required for disease establishment remain poorly characterized. Phenotypical analysis of Arabidopsis mutants for leucine-rich repeat (LRR)-receptor-like proteins revealed that mutations in the CLAVATA1 (CLV1) and CLAVATA2 (CLV2) genes confer enhanced disease resistance to bacterial wilt. We further investigated the underlying mechanisms using genetic, transcriptomic and molecular approaches. The enhanced resistance of both clv1 and clv2 mutants to the bacteria did not require the well characterized CLV signalling modules involved in shoot meristem homeostasis, and was conditioned by neither salicylic acid nor ethylene defence-related hormones. Gene expression microarray analysis performed on clv1 and clv2 revealed deregulation of genes encoding nuclear transcription factor Y subunit alpha (NF-YA) transcription factors whose post-transcriptional regulation is known to involve microRNAs from the miR169 family. Both clv mutants showed a defect in miR169 accumulation. Conversely, overexpression of miR169 abrogated the resistance phenotype of clv mutants. We propose that CLV1 and CLV2, two receptors involved in CLV3 perception during plant development, contribute to bacterial wilt through a signalling pathway involving the miR169/NF-YA module.

Expression of several virulence factors in the plant pathogen bacterium Ralstonia solanacearum is controlled by a complex regulatory network, at the center of which is PhcA. We provide genetic evidence that PhcA also represses the expression of hrp genes that code for the Type III protein secretion system, a major pathogenicity determinant in this bacterium. The repression of hrp genes in complete medium is relieved in a phcA mutant and two distinct signals, a quorum-sensing signal and complex nitrogen sources, appear to trigger this PhcA-dependent repression. This control of hrp gene expression by PhcA is realized at the level of the HrpG regulatory protein.
