Ochratoxin A (OTA) is a mycotoxin produced by two main types of fungi, Aspergillus and Penicillium species. OTA is a natural contaminant found in a large number of different matrices and is considered as a possible carcinogen for humans. Hence, low maximum permitted levels in foods have been established by competent authorities around the world, making essential the use of very sensitive analytical methods for OTA detection. Sample treatment is a crucial step of analytical methodology to get clean and concentrated extracts, and therefore low limits of quantification. Solid phase extraction (SPE) is a useful technique for rapid and selective sample preparation. This sample treatment enables the concentration and purification of analytes from the sample solution or extract by sorption on a solid sorbent. This review is focused on sample treatment procedures based on SPE prior to the determination of OTA in food matrices, published from 2010.

An analytical method for the determination of eleven aminoglycosides in different types of milk and milk-based functional products has been optimized and validated. A hydrophilic interaction chromatography (HILIC) column was proposed for the separation of analytes by ultra-high performance liquid chromatography-tandem mass spectrometry (UHPLC-MS/MS). A commercially molecularly imprinted polymer has been used for the solid phase extraction of the analytes, in order to achieve high selectivity in the sample treatment. The proposed method was characterized for different types of milk (whole cow milk, skimmed cow milk, whole goat milk) and functional dairy products, such as follow-on milk, Omega 3-enriched milk and isoflavones-enriched milk. Matrix effect was studied in the different matrices, being lower than │15│% in all cases, showing that the proposed procedure provided very clean extracts. Limits of quantification in the range 4.2-49µgkg -1 were estimated, well below the maximum residue limits established by the European Union. Recoveries ranged from 70% to 106% with RSD lower than 13%, in compliance with the current legislation. The combination of HILIC to solve the difficulties of the separation of these very polar compounds in reverse phase with the use of MISPE for sample treatment and MS/MS detection provided a very sensitive, highly selective, robust and useful method for identification and quantification of aminoglycoside residues in different types of milk and milk-based products., (Copyright © 2017 Elsevier B.V. A selective and rapid method has been developed to determine 15 antibiotic residues (eight tetracyclines and seven quinolones) in milk samples by capillary zone electrophoresis coupled with quadrupole time-of-flight mass spectrometry (CZE-Q-TOF-MS). The use of this hybrid mass spectrometer allowed obtaining full scan and full MS/MS spectra for quantification/confirmation purposes in a single run. In addition, solid phase extraction (SPE) using the new Oasis PRiME HLB cartridge was proposed for the extraction, achieving excellent results in terms of sample throughput. The proposed method was validated using whole cow milk as representative matrix. Good linearity was obtained (R 2 >0.99) for all the studied compounds. The precision, expressed as relative standard deviation (%, RSD), at two concentration levels (50 and 100μgkg -1 ) was below 13%. Recoveries obtained from goat milk, whole cow milk and semi-skimmed cow milk, at two concentration levels, ranged from 76 to 106%, while limits of quantification ranged from 1.5 to 9.6μgkg -1 , being lower than the established maximum residue limits in the European legislation. Matrix effect was negligible in all cases, showing that with this new SPE sorbent cleanest extracts were obtained with a minimum number of steps in the sample treatment. A simple and efficient method for the determination of 28 carbamates in high-fat cheeses is proposed. The methodology is based on a modified quick, easy, cheap, effective, rugged, and safe procedure as sample treatment using a new sorbent (Z-Sep + ) followed by ultra-high performance liquid chromatography with tandem mass spectrometry determination. The method has been validated in different kinds of cheese (Gorgonzola, Roquefort, and Camembert), achieving recoveries of 70-115%, relative standard deviations lower than 13% and limits of quantification lower than 5.4 μg/kg, below the maximum residue levels tolerated for these compounds by the European legislation. The matrix effect was lower than ±30% for all the studied pesticides. The combination of ultra-high performance liquid chromatography and tandem mass spectrometry with this modified quick, easy, cheap, effective, rugged, and safe procedure using Z-Sep + allowed a high sample throughput and an efficient cleaning of extracts for the control of these residues in cheeses with a high fat content., (© 2016 WILEY-VCH Verlag GmbH & Co. A reliable and rapid ultra high performance liquid chromatography tandem mass spectrometry (UHPLC-MS/MS) method has been developed for the determination of the eight quinolones of veterinary use regulated by European Union (marbofloxacin, ciprofloxacin, danofloxacin, enrofloxacin, sarafloxacin, difloxacin, flumequine and oxolinic acid). Chromatographic conditions were optimized in order to increase sample throughput and sensitivity. The antibiotics were detected by electrospray ionization in positive ion mode with multiple reaction monitoring (MRM) and MS/MS conditions were optimized in order to increase selectivity, selecting the corresponding product ions for quantification and identification. The separation was achieved in 3 min, using a Zorbax Eclipse Plus C18 column (50 mm × 2.1mm, 1.8 μm), with a mobile phase of 0.02% aqueous formic acid solution and acetonitrile. A dispersive solid phase extraction methodology, often referred to as the "QuEChERS" (quick, easy, cheap, effective, rugged, and safe) method, was optimized for extraction of the quinolones from honey and also it was evaluated for other bee products such as royal jelly and propolis. The method was validated for each matrix in terms of linearity, trueness, precision, limits of detection (LODs) and quantification (LOQ). LODs ranged between 0.2 and 4.1 μg kg(-1) with precision lower than 12% and satisfactory recoveries in most cases. MEKC coupling with LIF detection has been used for the determination of four aflatoxins (B(1), B(2), G(1) and G(2)). Separations were performed in an uncoated fused-silica capillary (70 cm x 75 microm id, 55 cm effective length), using 20 mM borate buffer with 30 mM SDS (pH 8.5) and 7% ACN. In order to increase sensitivity, an on-line preconcentration procedure was applied, based on sweeping, using the same separation buffer without SDS as solvent of the sample. The precision of the method was evaluated in terms of repeatability and intermediate precision and the results were acceptable in all cases (RSD<12%). With the on-line preconcentration LODs (obtained as 3 x S/N) were as low as 0.11, 0.52, 0.04 and 0.10 microg/L for G(2), G(1), B(2) and B(1), respectively. Recovery studies were developed with extracts of rice samples spiked with aflatoxins, being in the range between 93.0 and 105.4%. The method has also been applied to the determination of aflatoxins in rice samples, and the results compared with those obtained by a standard method, being in good agreement.

A method for the simultaneous determination of four fluoroquinolones of veterinary use (ciprofloxacin, danofloxacin, enrofloxacin and sarafloxacin) in two complex matrixes, such as bovine raw milk and pig kidney, has been established and validated. The method is based on the use of capillary electrophoresis (CE) coupled with a very sensitive detection mode, such as laser induced fluorescence (LIF) detection, due to the fact the all the compounds selected show native fluorescence. In order to achieve high selectivity in the sample treatment procedure, a commercially available molecularly imprinted polymer has been used for the solid phase extraction of the analytes. Once the retention and elution processes were optimized, the final extract was analyzed by CE-LIF using a 325 nm He-Cd laser. Optimum separation was obtained in a 70 cm x75 microm capillary using a 125 mM phosphoric acid solution at pH 2.8 with 36% methanol as background electrolyte. The method provided very low detection limits, ranging from 0.17 to 0.98 microg/kg for milk and 1.10 to 10.5 microg/kg for kidney, with acceptable precision and satisfactory recoveries., (Copyright 2010 Elsevier B.V. A sensitive and reliable method using capillary HPLC with UV-diode array detection (DAD) has been developed and validated for the trace determination of residues of 10 beta-lactam antibiotics of human and veterinary use, in milk, chicken meat and environmental water samples. The analytes included ampicillin, amoxicillin, penicillin V, penicillin G, cloxacillin, oxacillin, dicloxacillin, nafcillin, piperacillin and clavulanic acid. Legal levels are regulated by the EU Council regulation 2377/90 in animal edible tissues for these compounds. For food analysis, a solid-phase extraction (SPE) procedure consisting in a tandem of Oasis HLB and Alumina N cartridges was applied for off-line preconcentration and cleanup. For water analysis, the first step was only necessary. The limits of detection for the studied compounds were between 0.04-0.06 microg l(-1) for water samples and 0.80-1.40 microg l(-1) (or microg kg(-1)) in the case of foods derived from animals. Average recoveries for fortified samples at different concentration levels ranged between 82.9% and 98.2%, with relative standard deviations (RSDs) lower than 9%. The method showed the advantages of capillary HPLC for the detection of these widely applied antibiotics in different samples at very low concentration levels.

In this paper we review applications of capillary electrophoresis (CE) to the determination of antibiotic residues in food derived from animals and in environmental samples. Although many CE methods have been used to determine antibiotics in the pharmaceutical field (drug quality control or therapeutic monitoring in biological samples), food and environmental applications have been increasing in recent years. Due to the maximum residue limits established by the EU, in Directive 2377/90/EEC, for foodstuffs of animal origin and considering the low levels that can be found in environmental or waste waters or soils, different strategies to increase sensitivity have been developed, including off-line preconcentration, on-line stacking modes to use higher sample volumes, or in-line solid-phase extraction. Also, several detection techniques, such as fluorescence, laser-induced fluorescence, electrochemical detection, or mass spectrometry have been used; the last of these also enables unequivocal identification of the residues, required by Commission Decision 2002/657/EC. All these aspects will be discussed in this paper, in relation to the main groups of antibiotics used in veterinary and human medicine, for which applications in food and environmental samples have been developed by using CE as an efficient alternative to liquid chromatography.

Chemiluminescence (CL)-based detection has become in the last years quite a useful detecting tool in liquid chromatography (HPLC) due to its simplicity, low cost and high sensitivity and selectivity, and the development in instrumentation. Minimal instrumentation is required and no external light source is needed; thus, the optical system is quite simple. As a consequence, a wide variety of analytical methods have been developed in clinical, pharmaceutical, environmental and food analysis. In this review, applications of the HPLC-CL coupling in those different fields have been included and classified in relation to the different CL systems employed (namely peroxyoxalate reaction, tris(2,2'-bipyridine) ruthenium (II) reaction, luminol system and direct oxidations) and also sub-classified according to the group of analyte. The review covers the literature from 2000 until the end of 2008.

This paper presents two methods based on CZE-MS detection and CZE-MS/MS detection developed for the multiresidue determination of ten sulfonamides (sulfapyridine, sulfadoxin, sulfamethazine, sulfadimethoxine, sulfameter, sulfamerazine, sulfachlorpyridazine, sulfadiazine, sulfamethoxazole, and sulfamethizole) and a potentiator, trimethoprim (TMP), whose contents are regulated by the EU Council Regulation no. 2377/90 in animal edible tissues. Experimental designs were employed to optimize the electrospray conditions. MS/MS experiments using an IT as analyzer operating in multiple reaction monitoring (MRM) mode were carried out to achieve the minimum number of points according to the 2002/657/EC European Decision for unambiguous identification. The proposed procedures have been compared in terms of the performance characteristics and trueness. The limits of detection and quantification were in all cases lower than the maximum residue limits legislated for these compounds and the recoveries were satisfactory, being possible the application for their monitoring in foodstuff of animal origin and in environmental samples, allowing the determination of sulfonamides and TMP residues in meat and in superficial water in the low microg/L range.
