Blood transcriptomics has emerged as a vital tool for tracking the immune system and supporting disease diagnosis, prognosis, treatment, and research. The present study was conducted to analyze the gene expression profile and potential biomarker candidates using the whole blood of mandarin fish (Siniperca chuatsi) infected with LPS or poly (I:C) at 0 h, 3 h, 6 h, and 12 h. Our data suggest that 310 shared differentially expressed genes (DEGs) were identified among each comparison group after LPS infection, and 137 shared DEGs were identified after poly (I:C) infection. A total of 62 shared DEGs were differentially expressed in all compared groups after LPS or poly (I:C) infection. Pathways analysis for DEGs in all different compared groups showed that cytokine-cytokine receptor interaction was the most enrichment pathway. The expression levels of genes C-X-C chemokine receptor type 2-like (cxcr2), chemokine (C-C motif) receptor 9a (ccr9a), chemokine (C-C motif) receptor 9b (ccr9b), chemokine (C-X-C motif) receptor 4b (cxcr4b), and interleukin 10 receptor alpha (il10ra) were significantly different in all compared groups and most enriched in cytokine-cytokine receptor interaction pathway. The protein-protein interactions analysis among all shared DEGs showed that cxcr4 was the hub gene with the highest degree. The waste recycling of lemon peel, as a functional feed additive in aquafeed was evaluated by estimating the effects of fermented lemon peel (FLP) supplementation in diet on growth performance, innate immune responses, and susceptibility to Photobacterium damselae of grouper, Epinephelus coioides. A basal diet was added FLP at 0%, 1%, 3%, and 5%. Four tested diets were each fed to juvenile grouper (initial weight: 15.89 ± 0.10 g, triplicate groups) in a recirculation rearing system for eight weeks. Fish fed diets with 0%-3% FLP exhibited higher (p < 0.05) final weight, weight gain, and feed efficiency than fish fed the 5% FLP-diet. After challenge test, fish fed the 3% FLP-diet appeared the lowest mortality, followed by fish fed the 1% FLP-diet, and lowest in fish fed 0% and 5% FLP-diets. Plasma lysozyme activities were higher in fish fed diets with FLP than in fish fed the FLP-free control diet before challenge test. After challenge, fish fed diets with 1% and 3% FLP showed highest lysozyme activities, followed by fish fed the diet with 5% FLP, and lowest in fish fed the control diet. Hepatic malondialdehyde content was higher in fish fed the control diet than in fish fed diets with 1%-3% FLP. Bacterial ghosts (BGs) can be generated by the controlled expression of the PhiX174 lysis gene E in gram-negative bacteria. They are intriguing vaccine candidates since ghosts retain functional antigenic cellular determinants often lost during traditional inactivation procedures. Here we prepared Edwardsiella tarda ghost (ETG) and tested different concentrations in vaccination trials. The results showed that serum IgM antibody titers were significantly higher in three different concentration immunization groups than control group (P < 0.05), However, there was no significant (P > 0.05) difference between the immunized groups. The phagocytic percentage (PP) was significantly higher (P < 0.05) in ETG immunized groups than in the control group from 3 days post-treatment. The PP continued to rise with time until day 21, when the values of three ETG immunized groups were 45.7%,51.2% and 50.7%, respectively. In addition, phagocytic index (PI) was significantly higher (P < 0.05) in ETG immunized groups than in the control group after 7 days post-treatment. However, there was no significant (P > 0.05) difference of PP or PI between immunized groups. In addition, non-specific immune immunity, such as acid phosphatase, alkaline phosphatase, superoxide dismutase and lysozyme activities displayed a similar pattern in all immunized groups, all immunized fish showed significantly higher activities than control group fish (P < 0.05). Most importantly three ETG immunized groups were all significantly more protected against the E. tarda challenge (19/25, 76% survival), (21/25, 84% survival) and (20/25, 80% survival) respectively, compared to (9/25, 36% survival) survival in the control group, but there was no significant (P > 0.05) difference of survival rate (SR) or relative percent survival (RPS) between immunized groups. All these results suggest that an ETG could stimulate cellular and humoral immunity, and could be used as a vaccine candidate in S.m. In summary, ETG can protect fish from Edwardsiellosis, and there is no significant difference in SR and RPS when three different concentrations of ETG are used, so it can easily be developed as a vaccine for mechanical and artificial operations., (Copyright © 2018 Elsevier Ltd. Epizootic Ulcerative Syndrome (EUS) caused by Aphanomyces invadans which is a primary fungal parasitic pathogen, inflicts serious economic loss in tropical freshwater fish including snakehead murrel, Channa striatus. In the present study with an aim to circumvent the adverse effects of the traditional measures in graded levels (2%, 4%, and 6%) of Zeolite enriched diet on growth performance, hematology, immunological response, and disease resistance in C. striatus against A. invadans is reported. The final weight (FW), specific growth rate (SGR), feed conversion ratio (FCR), protein efficiency ratio (PER), and average daily gain (ADG) were significantly high in infected fish fed with 4% or 6% Zeolite incorporated diets on 4th week. The maximum survival rates (SR) of 96% and 98% were observed when fed with 2% or 4% diets on 4th week. Similarly, the white blood cell (WBC), red blood cell (RBC), hematocrit (Hct), mean corpuscular volume (MCV), and mean corpuscular haemoglobin concentration (MCHC) were significantly high when fed with any Zeolite enriched diet. However, the haemoglobin (Hb) and mean corpuscular haemoglobin (MCH) were significantly high with 4% and 6% Zeolite diets. The total protein and globulin were significantly high with 4% and 6% diets; the albumin, glucose, cholesterol, and triglyceride were significantly elevated with any enriched diet. The 4% and 6% Zeolite diets significantly enhanced the phagocytic activity on 2nd week but the 2% diet could increase it on 4th week. The respiratory burst (RB) activity, complement activity, and lymphocyte proliferation level were significantly enhanced with 4% and 6% Zeolite diets on weeks 1 and 2 while with 2% diet on 4th week. All enriched diets significantly increased the lysozyme activity during the experimental period. Superoxide anion (SOA) production significantly enhanced with 6% diet on weeks 1 and 2 whereas with 2% diet on week 4. Lower cumulative mortality of 10% and 15% was found with 4% and 6% Zeolite diets whereas a higher mortality (20%) was observed with 2% diet. The present study suggests that dietary supplementation with 4% or 6% Zeolite significantly increase growth performance, hemato-biochemical changes, immunological response, and disease resistance in C. striatus against A. invadans., (Copyright © 2016 Elsevier Ltd. Objective: The aim of this study was to study the pathophysiological effects on Channa punctatus due to the nematode parasite Eustrongylides sp., Methods: A total of 250 fish were examined during the period January 2012-2014. Hematological, serum biochemical, histological, and scanning electron microscopic studies were performed on normal and infected hosts to study the effects caused by the nematode., Results: The mean values of red blood corpuscle [RBC] count, hematocrit, and hemoglobin were significantly higher (P<0.01) in noninfected fish, while the values of white blood corpuscle [WBC] count, mean corpuscular volume [MCV], and mean corpuscular hemoglobin [MCH] were significantly higher (P<0.01) in infected fish. In infected fish, the average values of aspartate amminotransferase [AST] (416 UL-1), alanine amminotransferase [ALT] (73.35 UL-1), alkaline phosphatase [ALP] (161.6 mg dl-1), and cholesterol (154.82 mg dl-1) were significantly higher (P<0.01) than those in noninfected fish. Significant differences were also observed in total protein and glucose levels between the infected and noninfected fish. Histological and scanning electron microscopic studies of the host tissues revealed a series of pathological changes and mechanical damage., Conclusion: It can be concluded that Eustrongylides sp. has a significant impact on its host and thus the parameters outlined in the present paper may be employed as tools in monitoring the health status of fish in culture practices.

The immunoglobulin (Ig) from the serum of Channa striata was isolated by gel electroelution and characterized further to understand its nature and subsequent applications in studying the immune response. The purity of the sample was confirmed with the presence of a single band on native gradient PAGE and the molecular weight of ∼897 kDa was determined from the gel. In SDS-PAGE, C. striata Ig was reduced to produce two bands corresponding to H (heavy) (∼72 kDa) and L (light) (∼27 kDa) chain subunits. Polyclonal antiserum against the purified Ig was raised in a rabbit and adsorbed with 10% liver tissue homogenate of C. striata to enhance its specificity. By an indirect ELISA standardized using the adsorbed rabbit antiserum, the normal serum Ig concentration in C. striata was estimated to be 3.48 mg/mL. Further, a kinetic study of specific immunoglobulin response to formalin-killed Aeromonas hydrophila antigen was undertaken using another indirect ELISA, which showed a significant increase in serum immunoglobulin titer from day 2 onwards and reached its peak at day 14. Subsequently, the Ig titer was dropped from day 21 onwards till the completion of the experiment at day 42, although it was at a significantly higher level than the control.

The serum of rabbitfish (Siganus oramin) has been confirmed previously to have killing effect to Cryptocaryon irritans, an important marine ciliate protozoan that causes a disease referred to as "marine white spot disease". Herein, we find the serum of the rabbitfish also shows antibacterial activity against both gram-positive and gram-negative bacteria and has killing effect on two other parasites: Trypanosoma brucei brucei, Ichthyophthirius multifiliis. Results of scanning electron microscopy indicated that after treating with rabbitfish serum, the surface of the Staphylococcus aureus was wrinkled and pores were formed on the surface of Escherichia coli. Serum of the rabbitfish possesses a strong killing effect to Ichthyophthirius multifiliis in vitro, causing a similar effect as to C. irritans. The serum of rabbitfish also showed strong killing effect to T. b. brucei in vitro, with the minimus trypanocidal titre (MTT) only to be 1.5% in 1 h. Results of laser confocal fluorescence microscopy indicated that rabbitfish serum could also induce cell rupture of T. b. brucei. A novel antimicrobial protein (SR-LAAO) was isolated from the serum of rabbitfish by using ultrafiltration, reversed phase high performance liquid chromatography (RP-HPLC) and Native polyacrylamide gel electrophoresis (Native-PAGE). Results of gel overlay assay showed that the protein could act alone to inhibit the growth of S. aureus and E. coli. Results of western blot and automated Edman degradation showed that it was the same as the antiparasitic protein (APP) reported before to have killing effect on C. irritans. Full length cDNA sequence of the SR-LAAO was cloned. BLAST research suggested that the cDNA of SR-LAAO has a close similarity with a number of L-amino acid oxidases (LAAOs) and possesses two conserved motifs that exist in LAAOs. Combined, these results demonstrate that this protein which has antimicrobial activity to some pathogenic organisms was a novel LAAO found in the serum of rabbitfish. Immunohistochemical analysis demonstrated tissue specific expression and localization of SR-LAAO in the spleen, kidney, gill and blood of the rabbitfish, but was not found in other tissues. These results suggest that this protein may contribute considerably to the host non-specific immune defense mechanism to combat microbes of the rabbitfish and has the potency for using in future drug development., (Crown Copyright © 2011. This study determines the effect of diet enriched with the herb Baical skullcap Scutellaria baicalensis, and/or probiotics Lactobacillus sakei BK19 in rock bream, Oplegnathus fasciatus (32 ± 3 g) against Edwardsiella tarda. The changes in haematological parameters, innate immune response, and disease resistance were investigated after 1, 3, and 6 weeks. The white blood cell count (WBC: 10(4) mm(-3)), red blood cell count (RBC: 10(6) mm(-3)), and haemoglobin (Hb: g dl(-1)) levels significant increased (P < 0.05) with mixed diet on 3rd and 6th week and probiotics enriched diet on 6th week. The haematocrit (Ht: %) level significantly increased (P < 0.05) when fed with mixed diet on weeks 1-6. Interestingly, in mixed diet group the lymphocytes (LYM), monocytes (MON), and neutrophils (NEU) significantly increased from week 1-6. The eosinophils (EOS) significantly increased in all the treated groups. In the probiotics or mixed diet groups the total protein (TP: g dl(-1)) increased significantly on weeks 3 and 6. The serum lysozyme activity significantly was enhanced in all the treated groups indicating an increase in the innate immunity level. Serum complement, antiprotease activities, reactive oxygen species (ROS) and reactive nitrogen species (RNS) production significantly increased from week 1-6 with mixed diet. The maximum protection against E. tarda was recorded in mixed diet group with a minimum cumulative mortality of 20% and a high relative percent survival (RPS) of 72.84. In the probiotics and herbal diet groups the cumulative mortality was 25% and 35% and RPS was 68.63 and 59.42, respectively. This study indicates that administration of probiotics or mixed diets can effectively minimize the mortality and restore the altered hematological parameters and enhancing the innate immunity in O. fasciatus against E. tarda., (Copyright © 2011 Elsevier Ltd. The greater amberjack, Seriola dumerilii (Risso 1810) is a semi-pelagic fish and a worldwide species; it is considered a promising candidate for the diversification of Mediterranean aquaculture. In this paper an experimental injection with Listonella (Vibrio) anguillarum was performed to study the immune response of S. dumerilii. Antibody titres to L. anguillarum O1 were determined with indirect-ELISA at different times over a period of 42 days. Results showed that the antibody levels against L. anguillarum were significantly higher in the challenged fish compared to the control. They started developing since the 5th day reaching the highest peak on day 20 after injection, indicating a fast response of the immune system. The observed antibody titre was very high versus L. anguillarum if compared to other fish species.

Gnathiid isopods are common ectoparasites of fish on the Great Barrier Reef, Australia. While screening for appropriate markers for phylogenetic studies of gnathiids, we found that primers for 12S and 16S rDNA preferentially amplified the host fish DNA instead of gnathiid DNA. This amplification occurred even when using gnathiids that were not engorged with host blood and adult gnathiids that do not feed on fish blood. This method could be used in host-parasite studies to identify hosts without having to sample parasites directly from the host (which can be costly and requires considerable skill in a marine environment). Target ribosomal DNA sequences can be amplified from total DNA extracted from parasites that are captured in funnel traps or plankton tows. Sequence data from these can be used to identify the hosts that gnathiids were feeding on before capture.

A Vibrio strain Ls001, originally isolated from a body surface lesion of a moribund black porgy (Acanthopagrus schlegeli) in 1994 in Taiwan, was identified as Vibrio vulnificus. The extracellular products (ECP) of the strain were lethal to the fish, and its effects on fish serum in vitro and in vivo are described in the present study. Nine major precipitation arcs were visualized in normal fish serum in a crossed immunoelectrophoresis (CIE) gel using rabbit antiserum to the fish normal serum and staining with Coomassie brilliant blue. Only four and six of the nine major arcs could be tentatively identified by CIE following in vivo and in vitro ECP treatment, respectively. The same two major arcs were both missing following either in vivo or in vitro treatment with ECP. These complex events may significantly contribute to the pathogenesis of V. vulnificus in A. schlegeli.
