Echinocandin B (ECB) is a key precursor of antifungal agent Anidulafungin, which has demonstrated clinical efficacy in patients with invasive candidiasis. In this study, the effects of microparticle-enhanced cultivation and methyl oleate on echinocandin B fermentation titer were investigated. The results showed that the titer was significantly influenced by the morphological type of mycelium, and mycelium pellet was beneficial to improve the titer of this secondary metabolism. First, different carbon sources were chosen for the fermentation, and methyl oleate achieved the highest echinocandin B titer of 2133 ± 50 mg/L, which was two times higher than that of the mannitol. The study further investigated the metabolic process of the fermentation, and the results showed that L-threonine concentration inside the cell could reach 275 mg/L at 168 h with methyl oleate, about 2.5 times higher than that of the mannitol. Therefore, L-threonine may be a key precursor of echinocandin B. In the end, a new method of adding microparticles for improving the mycelial morphology was used, and the addition of talcum powder (20 g/L, diameter of 45 µm) could make the maximum titer of echinocandin B reach 3148 ± 100 mg/L.

Background: Amphotericin B (AmB) is widely used against fungal infection and produced mainly by Streptomyces nodosus. Various intracellular metabolites of S. nodosus were identified during AmB fermentation, and the key compounds that related to the cell growth and biosynthesis of AmB were analyzed by principal component analysis (PCA) and partial least squares (PLS)., Results: Rational design that based on the results of metabolomics was employed to improve the AmB productivity of Streptomyces nodosus, including the overexpression of genes involved in oxygen-taking, precursor-acquiring and product-exporting. The AmB yield of modified strain S. nodosus VMR4A was 6.58 g/L, which was increased significantly in comparison with that of strain S. nodosus ZJB2016050 (5.16 g/L). This was the highest yield of AmB reported so far, and meanwhile, the amount of by-product amphotericin A (AmA) was decreased by 45%. Moreover, the fermentation time of strain S. nodosus VMR4A was shortened by 24 h compared with that of strain. The results indicated that strain S. nodosus VMR4A was an excellent candidate for the industrial production of AmB because of its high production yield, low by-product content and the fast cell growth., Conclusions: This study would lay the foundation for improving the AmB productivity through metabolomics analysis and overexpression of key enzymes.

Background: L-2-aminobutyric acid (L-ABA) is an unnatural amino acid that is a key intermediate for the synthesis of several important pharmaceuticals. To make the biosynthesis of L-ABA environmental friendly and more suitable for the industrial-scale production. We expand the nature metabolic network of Escherichia coli using metabolic engineering approach for the production of L-ABA., Results: In this study, Escherichia coli THR strain with a modified pathway for threonine-hyperproduction was engineered via deletion of the rhtA gene from the chromosome. To redirect carbon flux from 2-ketobutyrate (2-KB) to L-ABA, the ilvIH gene was deleted to block the L-isoleucine pathway. Furthermore, the ilvA gene from Escherichia coli W3110 and the leuDH gene from Thermoactinomyces intermedius were amplified and co-overexpressed. The promoter was altered to regulate the expression strength of ilvA* and leuDH. The final engineered strain E. coli THR ΔrhtAΔilvIH/Gap-ilvA*-Pbs-leuDH was able to produce 9.33 g/L of L-ABA with a yield of 0.19 g/L/h by fed-batch fermentation in a 5 L bioreactor., Conclusions: This novel metabolically tailored strain offers a promising approach to fulfill industrial requirements for production of L-ABA.

3-Hydroxypropionic acid (3-HP) is an important platform synthesis block for sets of chemicals, but the relatively low production of 3-HP from biological sources presented major barriers for its industrial applications. In this study, a dual-substrate fermentative strategy by glycerol and glucose was proposed, and the aim was to evaluate the effect of different substrate addition strategies on the fermentation process. The results indicated that the optimal cosubstrate was glucose (20 g/L), and the enzymatic activity of aldehyde dehydrogenase (AldH) could be improved 3.5-fold as compared with no glucose addition. Continuous fed-batch fermentation at a constant speed displayed better 3-HP production of 17.20 g/L and highest specific 3-HP productivity of 1.79 mmol/(g cell·H) than the other fed-batch mode. The addition of glucose could greatly reduce the imbalance of the activity between glycerol dehydratase and AldH and provide a feasible method for improving 3-HP production. Acarbose, a potent α-glucosidase inhibitor, is as an oral anti-diabetic drug for treatment of the type two, noninsulin-dependent diabetes. Actinoplanes utahensis ZJB-08196, an osmosis-resistant actinomycete, had a broad osmolality optimum between 309 mOsm kg(-1) and 719 mOsm kg(-1). Utilizing this unique feature, an fed-batch culture process under preferential osmolality was constructed through intermittently feeding broths with feed medium consisting of 14.0 g l(-1) maltose, 6.0 g l(-1) glucose and 9.0 g l(-1) soybean meal, at 48 h, 72 h, 96 h and 120 h. Improvement of dihydroxyacetone (DHA) production by mutagenesis of ion beam implantation and medium optimization using response-surface methodology (RSM) were investigated in this work. More than 1000 mutant strains were selected through a mutagenesis method using N(+) ions implantation with a dose of 60 × (2.6 × 10(13)) ions/cm(2) and energy of 10 keV. Several high-yield mutant strains were showed the potent application for DHA production and the genetically stable mutant strain G. oxydans ZJB09113 was selected for optimization of cultivation condition by RSM. The optimal medium for DHA fermentation is composed (in g/L) of yeast extract 4.88, CaCO(3) 2.00, and glycerol 52.86 mL/L (initial pH 4.89). The maximal DHA concentration of 40.0 g/L was achieved after 24 hr of shaken flask fermentation at 30°C with 150 rpm, and 196.3% increase in DHA production in comparison with unoptimized conditions.

Microbial fermentation for enzyme production and then whole-cell catalysis for l-2-aminobutyric acid (l-ABA) production have huge potential for industrial application, but the catalytic capacities of cells are directly related to the fermentation process. Using a 50 L fermenter, the effects of initial glycerol concentration in the medium and rotating speed on cell catalytic capacity were investigated. Fermentation cells showed the best catalytic activity when the initial glycerol concentration was 12 g/L and the rotating speed was 250 rpm. Furthermore, we studied the difference between glycerol and glycerol mixtures as fed-batch media in pH–stat fed-batch fermentation. Results showed that glycerol had better catalytic activity than the glycerol mixture, and the effect of fed-batch fermentation was better than batch fermentation. Meanwhile, the enzyme activities of leucine dehydrogenase and formate dehydrogenase reached 129.87 U/g DCW and 437.02 U/g DCW, respectively, and the intracellular NAD(H) concentration reached 14.94 μmol/g DCW. Using the optimized fermentation parameters, amplified fermentation was then carried out in a 5000 L fermenter to demonstrate the industrial production of l-ABA by Escherichia coli BL21. Objective: To solve the bottleneck of plasmid instability during microbial fermentation of l-DOPA with recombinant Escherichia coli expressing heterologous tyrosine phenol lyase. Results: The tyrosine phenol lyase from Fusobacterium nucleatum was constitutively expressed in E. coli and a fed-batch fermentation process with temperature down-shift cultivation was performed. Efficient strategies including replacing the original ampicillin resistance gene, as well as inserting cer site that is active for resolving plasmid multimers were applied. As a result, the plasmid stability was increased. The co-use of cer site on plasmid and kanamycin in culture medium resulted in proportion of plasmid containing cells maintained at 100% after fermentation for 35 h. The specific activity of tyrosine phenol lyase reached 1493 U/g dcw, while the volumetric activity increased from 2943 to 14,408 U/L for l-DOPA biosynthesis. Conclusions: The established strategies for plasmid stability is not only promoted the applicability of the recombinant cells for l-DOPA production, but also provides important guidance for industrial fermentation with improved microbial productivity. D-pantothenic acid (D-PA), as a crucial vitamin, is widely used in food, animal feed, cosmetics, and pharmaceutical industries. In our previous work, recombinant Escherichia coli W3110 for production of D-PA was constructed through metabolic pathway modification. In this study, to enhance D-PA production, statistical optimization techniques including Plackett–Burman (PB) design and Box-Behnken design (BBD) first were adopted to optimize the culture condition. The results showed that the glucose, β-alanine and (NH4)2SO4 have the most significant effects on D-PA biosynthesis. The response surface model based on BBD predicted that the optimal concentration is glucose 56.0 g/L, β-alanine 2.25 g/L and (NH4)2SO4 11.8 g/L, the D-PA titer increases from 3.2 g/L to 6.73 g/L shake flask fermentation. For the fed-batch fermentation in 5 L fermenter, the isoleucine feeding strategy greatly increased the titer and productivity of D-PA. As a result, titer (31.6 g/L) and productivity (13.2 g/L·d) of D-PA were achieved, they increased by 4.66 times and 2.65 times, respectively, compared with batch culture. At the same time, the accumulation of acetate reduced from 29.79 g/L to 8.55 g/L in the fed-batch fermentation. These results demonstrated that the optimization of medium composition and the cell growth rate are important to increase the concentration of D-PA for microbial fermentation. Objective: O-acetylhomoserine (OAH) is an important platform chemical to produce high-valuable chemicals. To improve the production of O-acetylhomoserine from glycerol, the glycerol-oxidative pathway was investigated and the optimization of fermentation with crude glycerol was carried out. Results: The glycerol-uptake system and glycerol-oxidative pathway were modified and O-acetyltransferase from Corynebacterium glutamicum was introduced into the engineered strain to produce O-acetylhomoserine. It was found that overexpression of glycerol 3-phosphate dehydrogenase improved the OAH production to 6.79 and 4.21 g/L from pure and crude glycerol, respectively. And the higher OAH production depending on higher level of transcription of glpD. Two-step statistical approach was employed to optimize the fermentation conditions. The significant effects of glycerol, ammonium chloride and yeast extract were screened applying Plackett–Burman design and were optimized further by employing the Response Surface Methodology. Under optimized conditions, the OAH production was up to 9.42 and 7.01 g/L when pure and crude glycerol were used in shake flask cultivations, respectively. Conclusions: The enzymatic step catalyzing the oxidation of glycerol through GlpD was the key step for OAH production, which served the foundation for realization of a consistent OAH production from crude glycerol in the future. R-2-(4-hydroxyphenoxy)propionic acid (R-HPPA) is a key chiral intermediate for phenoxypropionic acid herbicide synthesis. In this study, to improve the production of R-HPPA with B. bassiana ZJB16007, the cultivation conditions in solid-state fermentation (SSF) were investigated. The effects of various substrates on R-HPPA production were evaluated and the process parameters were also optimized. The results showed that rice bran was the optimal substrate for R-HPPA production. The optimal medium components and cultivation conditions were: rice bran: silkworm chrysalis powder = 5.25: 2.25 (g: g), nutrient salts solution 12 mL which contained 50 g/L R-PPA, pH 5.0, and cultivated at 28 °C for 11 days. Under the optimized conditions, the transformation of R-HPPA was significantly improved and the yield of R-HPPA reached 77.78%, which was 15.14% higher than that of the control (67.55%). Tyrosine phenol lyase (TPL) is a robust biocatalyst for the production of L-dihydroxyphenylalanine (L-DOPA). The improvement of TPL production is conducive to the industrial potential. In this study, the optimization of culture medium of recombinant Escherichia coli harboring TPL from Fusobacterium nucleatum (Fn-TPL) was carried out. Sucrose and combination of yeast extract and peptone were selected as carbon and nitrogen source, respectively. Their optimal concentrations were determined by Box-Behnken design and the synergistic effect between yeast extract and peptone was found to be significant, with p-value < 0.05. The DO-STAT fed-batch fermentation under optimized culture condition was established and the oxygen level was fixed at 20%. Both the biomass and Fn-TPL activity were significantly increased, which were 35.6 g dcw/L and 12292 U/L, respectively. To biosynthesize the (3R,5S)-CDHH in an industrial scale, a newly synthesized stereoselective short chain carbonyl reductase (SCR) was successfully cloned and expressed in Escherichia coli. The fermentation of recombinant E. coli harboring SCR was carried out in 500 L and 5000 L fermenters, with biomass and specific activity of 9.7 g DCW/L, 15749.95 U/g DCW, and 10.97 g DCW/L, 19210.12 U/g DCW, respectively. The recombinant SCR was successfully applied for efficient production of (3R,5S)-CDHH. The scale-up synthesis of (3R,5S)-CDHH was performed in 5000 L bioreactor with 400 g/L of (S)-CHOH at 30°C, resulting in a space-time yield of 13.7 mM/h/g DCW, which was the highest ever reported. Abstract: Valienamine is an important medicinal intermediate with broad use in the synthesis of some stronger α-glucosidase inhibitors. In order to improve valienamine concentration in the fermentation broth and make the downstream treatment easy, a fed-batch process for the enhanced production of valienamine by Stenotrophomonas maltrophilia in a stirred tank bioreactor was developed. Results showed that supplementation of validamycin A in the process of cultivation could increase the valienamine concentration. One-pulse feeding was observed to be the best strategy. The maximum valienamine concentration of 2.35gL−1 was obtained at 156h when 86.4g of validamycin A was added to a 15-L bioreactor containing 8L fermentation medium with one-pulse feeding. The maximum valienamine concentration had a great improvement and was increased above 100% compared to batch fermentation in the stirred tank bioreactor. The pH-controlled experiments showed that controlling the pH in the process of one-pulse feeding fermentation had not obvious effect on the production of valienamine. [Copyright &y& Elsevier]

Abstract: The effects of initial culture pH ranging from 3.5 to 6.5 on biomass content and astaxanthin concentration by Xanthophyllomyces dendrorhous were investigated in shake flask fermentation. An optimal initial pH of 6.0 for cell growth was obtained with cell density at 17.21g/l, while an optimum pH of 5.0 for astaxanthin accumulation was obtained with volumetric concentration of astaxanthin at 20.48mg/l. Batch bioprocess of astaxanthin fermentation by X. dendrorhous at various pH values ranging from 4.0 to 6.0 was examined. In batch fermentation under a controlled pH, the optimum pH for cell growth was 6.0 with specific cell growth 0.049h−1, and the optimum pH for astaxanthin formation in cell was 4.0 with specific product yield (Y p/x) at 1.52mg/g. A pH-shift strategy was developed to gain higher volumetric concentration of astaxanthin in batch fermentation. The maximum volumetric concentration of astaxanthin reached 27.05mg/l, increased by 24.1% as comparison with that of constant pH fermentation. [Copyright &y& Elsevier]

Microbial fermentation for L-methionine (L-Met) production based on natural renewable resources is attractive and challenging. In this work, the effects of medium composition and fermentation conditions were investigated to improve L-Met production by genetically engineered Escherichia coli MET-3. Statistical optimization techniques including Plackett-Burman (PB) design and Box-Behnken design (BBD) were adopted first to optimize the culture medium. Results of PB-designed experiments indicated that the culture medium components including glucose, yeast extract, KH2PO4, and MgSO4.7H2O had significant effects on L-Met biosynthesis. With their best-predicted concentration established by BBD (glucose 37.43 g/L, yeast extract 0.95 g/L, KH2PO4 1.82 g/L, and MgSO4.7H2O 4.51 g/L), L-Met titer was increased to 3.04 g/L from less than 2.0 g/L. For further enhancement of L-Met biosynthesis, the fermentation conditions of batch cultivation carried out in a 5-L fermentor were optimized, and the optimum results were obtained at an agitation rate of 300 rpm, medium pH of 7.0, and induction temperature of 28 °C. Based on the optimization parameters, fed-batch fermentation with the modified medium was conducted. As a result, great improvement of L-Met titer (12.80 g/L) and yield (0.13 mol/mol) were achieved, with an increase of 38.53% and 30.0% compared with those of the basal medium, respectively. Furthermore, higher L-Met productivity of 0.261 g/L/h was obtained, representing 2.13-fold higher in comparison to the original medium. Microbial production of D-pantothenic acid (D-PA) has become an important research area, as D-PA is an essential vitamin widely applied in food, feed, chemical and pharmaceutical industries. Here both dynamic regulations of redox metabolism and fermentation strategies were applied to enhancing D-PA biosynthesis in Escherichia coli W3110. Firstly, a quorum-sensing (QS) system for redox regulation developed, in which zwf and nadK overexpression were shown to be effective for enhancing D-PA production. Based on this, a strain DPA11/p LBCWK was constructed with D-PA titer at 4.19 g/L in flask. Moreover, the fermentation strategies including pH-feedback feeding and gradient β-alanine feeding were introduced, under optimal conditions, the strain DPA11/p LBCWK could produce 48.21 g/L D-PA in 5 L fermenter, with the specific productivity of 0.72 g/L·h and the yield of 0.33 g/g glucose. These modification strategies were proved effective and would provide valuable reference for biosynthesis of other chemicals. The biosynthetic pathway of l -methionine in microorganisms was complex and regulated at multiple levels. In this study, a two-step method for l -methionine production combined fermentation and biocatalysis was realized in one pot. The O -succinyl- l -homoserine (OSH) producing strain Escherichia coli W3110(DE3) ΔIJB∗Trc metL /pTrc- metA fbr-Trc- thrA fbr- yjeH (ΔIJB) was constructed initially. OSH in the fermentation supernatant was then converted to l -methionine in the presence of O -succinyl- l -homoserine sulfhydrylase (OSHS) and sodium methanethiol. The titer of l -methionine could reach 21.1 g/L after 88 h (84 h fermentation and 4 h catalysis) in a two-step method (process 1). In a one-pot two-strain system (process 2), two strains ΔIJB and E. coli BL21(DE3)/pET28b–OSHS–cutinase were co-cultured, and 8.24 g/L l -methionine was obtained. In another one-pot one-strain system (process 3), strain E. coli ΔIJB/pET28b–OSHS–cutinase could co-express OSHS and cutinase during ΔIJB fermentation at the same time, obtaining 13.6 g/L l -methionine in a 5 L fermentor after 84 h. By comparing the three processes for l -methionine production based on the process 1, the simplified process in process 3 provided in this study showed potent in the large-scale production of l -methionine with convenient handling and production efficiency, but further works still need to be carried out to improve the l -methionine production. • High-yield O-succinyl- l -homoserine (OSH) producing strain was constructed in Escherichia coli W3110(DE3) (ΔIJB). • Enzyme O-succinyl- l -homoserine sulfhydrylase and cutinase were co-expressed in E. coli BL21(DE3) (OSHS–Cut). • A one-pot two-strain system was established for l -methionine production with co-cultivation of ΔIJB and OSHS–Cut. • A system combining of OSH production and enzymatic catalysis in one pot was established. • Amino acids involved in the biosynthesis of PQQ framework were disproportionately utilized in yeast extract. • Glutamate 0.1% addition significantly improved mSLDH activity in a PQQ-dependent manner within 20 g/L yeast extract. • A low-cost nitrogen source was developed by partial substitution of yeast extract (15 g/L) with glutamate (2 g/L). • Cells obtained from low-cost nitrogen source exhibit an equally high mSLDH activity towards miglitol precursor 6NSL. Existing industrial fermentations for Gluconobacter oxydans require a rich nitrogen source, which results in a relatively high production cost. In present study, amino acids utilization from yeast extract (YE) (20 g·L−1) was investigated, revealing that glutamate is disproportionately utilized, and consequently becomes a rate-limiting factor for the activity of membrane-bound sorbitol dehydrogenase (mSLDH). Additional supplementation of glutamate (0.1%) improved the abundance of coenzyme pyrroloquinoline quinone (PQQ) by 1.51-fold. Meanwhile, enzyme activity of mSLDH was improved by 1.21-fold, which was mainly dependent on the induction of PQQ. Partial substitution of YE (15 g·L−1) with glutamate (2 g·L−1) as nitrogen source was tested in G. oxydans fermentations and the thus-obtained resting-cells exhibited an high mSLDH activity towards the miglitol precursor N-2-hydroxyethyl-glucamine with an increased productivity (59.2 vs. 42.1 mg· (g DCW)-1 h-1) and an equally high yield (81.56% vs. 80.35%) compared with cells fermented with 25 g·L−1 YE. Taken together, the results provide a feasible strategy for economical fermentation of G. oxydans in achieving high mSLDH activity for biotransformation applications in large-scale. Abstract Methionine is an essential sulfur-containing amino acid for organisms. The separation and purification are important for the production of l -methionine from fermentation broth. In this work, the adsorption properties for l -methionine separation of ten macroporous resins were firstly evaluated. Macroporous cation resin D72 showed the best adsorption capacity (52.37 mg/g) and desorption rate (99.12%). The adsorption kinetics of l -methionine on D72 resin followed the pseudo second-order model and adsorption isotherm fitted well to the Sips model, the adsorption process mainly followed a physisorption mechanism. D72 resin packed columns were then used in the batch separation of fermentation broth, the operation parameters were optimized during the dynamic adsorption and desorption experiments. Under the optimized conditions: fermentation broth adjusted to pH = 2, loading flow rate at 2 BV/h, loading volume 55 mL, 1 mol/L NH 3 ·H 2 O as eluent, elution flow rate at 2 BV/h, column height-diameter ratio at 14:1, excellent recovery and purity of l -methionine (82.37% and 85.69%, respectively) could be achieved. Abstract A colorimetric assay has been developed for quantitative analysis of active biomass of Fusarium fujikuroi , based on the reduction of the tetrazolium salt 2,3-Bis-(2-methoxy-4-nitro-5-sulfophenyl)-2H-tetrazolium-5-carboxanilide (XTT) when menadione was present as an electron coupling agent. The optimum assay-conditions were set as 200 μg/ml XTT, 5 μM menadione and one-hour reaction time. Under these settings, the produced formazan displayed a linear relationship with F. fujikuroi biomass. This method was subsequently applied to evaluate the cell growth behavior, which showed a positive correlation with the carbon source consumption and gibberellin biosynthesis under the industrial fermentation conditions. Our results showed that the XTT-menadione assay is a valuable tool in analyzing the industrial fermentation process of F. fujikuroi , especially when the medium contains insoluble and complex components. In this work, a robust brefeldin A-synthesizing fungus, Eupenicillium brefeldianum ZJB082702, was bred from a Murraya paniculata endophytic fungus E. brefeldianum A1163. Using one-factor-at-a-time experimental design, optimization of media composition for E. brefeldianum ZJB082702 fermenting brefeldin A was conducted. Outcomes indicated that mixed carbon source and mixed nitrogen source were of c ritical importance to brefeldin A fermentation. After 6d culture in the optimized fermentation media, composed of (gl−1) 13.33 starch, 26.67 glucose, 1.0 yeast extract powder, 1.0 corn steep liquor, 0.5 soybean meal, 0.75 NaNO3, 2.5 malt extract, 6.0 CaCO3, 3.0 MgSO4, 4.0 KH2PO4, 1.0 × 10−2 CuSO4, brefeldin A yield peaked at 1304.7mgl−1, 648.2mgl−1 in 500ml baffled flask and 15l stirred fermentor respectively, formed as a growth associated type of secondary metabolite based on fermentation profile analysis.
