Self-patterning processes originated by physical stimuli have been extensively documented in thin films, whereas spontaneous wrinkling phenomena due to chemical transformation processes are, to the best of our knowledge, unprecedented. Herein we report a case of spontaneous polymerization-driven surface nano-patterning (B500 nm) that develops in smooth thin solid films of 5,6-dihydroxyindole (DHI), a major precursor of eumelanin polymers, over a time scale of 30 to 60 days in air at room temperature. The phenomenon can be observed only above a critical film thickness of B250 nm and it is affected by exposure to ammonia vapors causing acceleration of the oxidation process. The thickness-dependent onset of wrinkling can be attributed to non-homogeneous rates of oxidation through the film causing slow swelling/expansion of the inner layers followed by fast stiffening and cross-linking in the outer layer exposed to higher oxygen levels. Chiral porphyrin hetero-aggregates, produced from meso-tetrakis(4-N-methylpyridyl) porphyrin H2T4 and copper(II) meso-tetrakis(4-sulfonatophenyl)porphyrin CuTPPS by an imprinting effect in the presence of L-3,4-dihydroxyphenylalanine (L-DOPA), are shown herein to serve as templates for the generation of chiral structures during the oxidative conversion of the amino acid to melanin. This remarkable phenomenon is suggested to involve the initial role of L-DOPA and related chiral intermediates like dopachrome as templates for the production of chiral porphyrin aggregates. When the entire chiral pool from DOPA is lost, chiral porphyrin hetero-aggregate would elicit axially chiral oligomer formation from 5,6-dihydroxyindole intermediates in the later stages of melanin synthesis. These results, if corroborated by further studies, may open unprecedented perspectives for efficient strategies of asymmetric melanin synthesis with potential biological and technological applications.

Within the framework of neurodegenerative disorder therapies, the fabrication of 3D eumelanin architectures represents a novel strategy to realize tissue-engineering scaffolds for neuronal cell growth and control by providing both mechanical support and biological signals. Here, an appropriate procedure combining electrospinning, spin coating and solid-state polymerization process is established to realize the scaffolds. For biological analysis, a human derived cell line SH-SY5Y from neuroblastoma is used. Cell maturation on eumelanin microfibers, random and aligned, is evaluated by using confocal analysis and specific markers of differentiating neurons (beta III tubulin and GAP-43 expression). Cell morphology is tested by SEM analysis and immunofluorescence techniques. As results, eumelanin coated microfibers prove capable to support biological response in terms of cell survival, adhesion and spreading and to promote cell differentiation toward a more mature neuronal phenotype as confirmed by GAP-43 expression over the culture.

The recent development of eumelanin pigment-based blends integrating “classical” organic conducting materials is expanding the scope of eumelanin in bioelectronics. Beyond the achievement of high conductivity level, another major goal lays in the knowledge and feasible control of structure/properties relationship. We systematically investigated different hybrid materials prepared by in situ polymerization of the eumelanin precursor 5,6-dihydroxyindole (DHI) in presence of various amounts of graphene-like layers. Spectroscopic studies performed by solid state nuclear magnetic resonance (ss-NMR), x-ray photoemission, and absorption spectroscopies gave a strong indication of the direct impact that the integration of graphene-like layers into the nascent polymerized DHI-based eumelanin has on the structural organization of the pigment itself, while infrared, and photoemission spectroscopies indicated the occurrence of negligible changes as concerns the chemical units. A tighter packing of the constituent units could represent a strong factor responsible for the observed improved electrical conductivity of the hybrid materials, and could be possible exploited as a tool for electrical conductivity tuning.

In the human body, the black-brown biopigment eumelanin blocks the harmful ultraviolet (UV) radiation. In the plastics industry, additives are often added to polymers to increase their UV-absorption properties. We herein report an assessment of the biopigment eumelanin as a nature-inspired additive for plastics to enhance their UV-absorption. Since eumelanin is produced by natural sources and is non-toxic, it is an interesting candidate in the field of sustainable plastic additives. In this work, eumelanin-including films of commercial ethylene-vinyl acetate copolymer (EVA), a plastic used for packaging applications, were obtained by melt-compounding and compression molding. The biopigment dispersion in the films was improved by means of the Melanin Free Acid treatment (MFA). It was observed that eumelanin amounts as low as 0.8% wt. cause an increase of the UV absorption, up to one order of magnitude in the UVA range. We also evaluated the effect of eumelanin on the thermal and photostability of the films: the biopigment proved to be double-edged, working both as UV-absorption enhancer and photo-prooxidant, as thermogravimetric analysis (TGA) and infrared (IR) spectroscopy revealed.

The antioxidant activity of eumelanin, a ubiquitous pigment in flora and fauna, constitutes one of its most fascinating physicochemical properties. To shed light on free radical scavenging vs redox facets of such activity, we applied hydrogen atom transfer- and electron transfer-based assays to pristine Sepia ink, eumelanin from Sepia ink, chemically controlled eumelanins and their precursor building blocks. Our work contributes to the rational use of the antioxidant properties of eumelanin for health, cosmetics and environmental applications.

Melanin denotes a variety of mammalian pigments, including the dark electrically conductive eumelanin and the reddish, sulfur-containing, pheomelanin. Organic (bio)electronics is showing increasing interests in eumelanin exploitation, e.g., for bio-interfaces, but the low conductivity of the material is limiting the development of eumelanin-based devices. Here, for the first time, we report an abrupt increase of the eumelanin electrical conductivity, revealing the highest value presented to date of 318 S/cm. This result, obtained via simple thermal annealing in vacuum of the material, designed on the base of the knowledge of the eumelanin chemical properties, also discloses the actual electronic nature of this material's conduction.

Developing safe and high efficiency contrast tools is an urgent need to allow in vivo applications of photoacoustics (PA), an emerging biomolecular imaging methodology, with poor invasiveness, deep penetration, high spatial resolution and excellent endogenous contrast. Eumelanins hold huge promise as biocompatible, endogenous photoacoustic contrast agents. However, their huge potential is still unexplored due to the difficulty to achieve at the same time poor aggregation in physiologic environment and high PA contrast. This study addresses both issues through the design of a biocompatible photoacoustic nanoprobe, named MelaSil_Ag-NPs, relying on silica-templated eumelanin formation as well as eumelanins redox and metal chelating properties to reduce Ag + ions and control the growth of generated metal nanoparticles. This strategy allowed self-structuring of the system into a core-shell architecture, where the Ag core was found to boost PA signal, despite the poor eumelanin content. Obtained hybrid nanoplatforms, showed stable photoacoustic properties even under long irradiation. Furthermore, conjugation with rhodamine isothiocyanate allowed particles detection through fluorescent imaging proving their multifunctional potentialities. In addition, they were stable towards aggregation and efficiently endocytosed by human pancreatic cancer cells (BxPC3 and Panc-1)displaying no significant cytotoxicity. Such numerous features prove huge potential of those nanoparticles as a multifunctional platform for biomedical applications.

Melanins are a group of dark insoluble pigments found widespread in nature. In mammals, the brown-black eumelanins and the reddish-yellow pheomelanins are the main determinants of skin, hair, and eye pigmentation and play a significant role in photoprotection as well as in many biological functions ensuring homeostasis. Due to their broad-spectrum light absorption, radical scavenging, electric conductivity, and paramagnetic behavior, eumelanins are widely studied in the biomedical field. The continuing advancements in the development of biomimetic design strategies offer novel opportunities toward specifically engineered multifunctional biomaterials for regenerative medicine. Melanin and melanin-like coatings have been shown to increase cell attachment and proliferation on different substrates and to promote and ameliorate skin, bone, and nerve defect healing in several in vivo models. Herein, the state of the art and future perspectives of melanins as promising bioinspired platforms for natural regeneration processes are highlighted and discussed.

The origin of eumelanin optical properties remains a formidable conundrum preventing a detailed understanding of the complex photo-protective role of these widespread natural pigments and the rational design of innovative bioinspired materials for optoelectronic applications. Here we report the unusual kinetic and thickness-dependent evolution of the optical properties of black eumelanin polymers generated by spontaneous aerial polymerization of 5,6-dihydroxyindole (DHI) thin films (0.1–1 μm), consistent with peculiar solid state reorganization mechanisms governing broadband absorption. The complete reversal of eumelanin UV-visible transmittance spectrum curvature on passing from 0.2 to 0.5 μm thick films, the marked increase in visible extinction coefficients with increasing film thickness and the higher UV extinction coefficients in slowly vs. rapidly generated polymers concur to support distinct dynamic regimes of solid-state molecular reorganization at the nanoscale level and to do affect the development of broadband visible absorption. Solid state control of molecular reorganization disclosed herein may delineate new rational strategies for tuning optical properties in eumelanin thin films for optoelectronic applications.

We disclose herein the first example of stable monodispersed hybrid nanoparticles (termed MelaSil−NPs) made up of eumelanin biopolymer intimately integrated into a silica nanoscaffold matrix and endowed with high antioxidant and cytoprotective effects associated with a specific subcellular localization. MelaSil−NPs have been fabricated by an optimized sol−gel methodology involving ammonia-induced oxidative polymerization of a covalent conjugate of the eumelanin building block 5,6-dihydroxyindole-2-carboxylic acid (DHICA) with 3-aminopropyltriethoxysilanes (APTS). They displayed a round-shaped (ca. 50−80 nm) morphology, exhibited the typical electron paramagnetic resonance signal of eumelanin biopolymers, and proved effective in promoting decomposition of hydrogen peroxide under physiologically relevant conditions. When administered to human ovarian cancer cells (A2780) or cervical cancer cells (HeLa), MelaSil−NPs were rapidly internalized and colocalized with lysosomes and exerted efficient protecting effects against hydrogen peroxide-induced oxidative stress and cytotoxicity. A recently developed protocol allows to prepare high quality thin films integrating eumelanin pigment with standard commercial poly(3,4-ethylenedioxythiophene):poly(styrenesulfonate) (PEDOT:PSS). The new blend combines noteworthy water stability and substrate adhesion with valuable electro-optical properties, so it can be used as anodic material in indium tin oxide-free organic light emitting diodes. Starting from these results, the present work systematically addresses the parameters affecting the blend conductivity. In particular, X-ray scattering analysis discloses that low quantities of eumelanin modifies molecular packing of the ternary blend, mainly in intermolecular distances and possibly in the degree of crystalline order of PEDOT, having anyway very little effect on the decay of the blend conductivity. Because of the synergetic effects observed here between the eumelanin and the PEDOT:PSS, this study provides a promising ground to access multifunctional materials for applications in advanced electronic devices and bioelectronics.

Eumelanins, the black insoluble pigments of human skin, eyes and substantia nigra (neuromelanin), stand today as a unique source of inspiration for the design and implementation of soft biocompatible multifunctional materials for bio-optoelectronic devices. Interest in eumelanins stems from bioavailability, biocompatibility and a peculiar set of physicochemical properties, i.e. broadband absorption in the UV-visible range, intrinsic free radical character, water-dependent hybrid ionic-electronic conductor behaviour, supporting optimistic feelings about a possible rise of eumelanin-mimics as innovative bioinspired solutions for organic bioelectronics. However, a number of conceptual and technological gaps still hinder a rapid progress of melanin-based organic electronics and bioelectronics, including in particular the limited contribution of electronic conductivity and current decay with time under biasing. Herein, we provide a concise overview of the structural and optoelectronic properties of melanins with a view to bringing to focus main issues and challenges en route to bioelectronic applications. Eumelanin integration in silica aerogel (SA) was achieved via supercritical adsorption of 5,6-dyhydroxyindole (DHI) from CO2. Notably, after the supercritical treatment, DHI evolved towards spontaneous polymerization, which resulted in uniform pigment development over the SA. The new material was characterized for its morphological and physicochemical properties, disclosing the formation of a eumelanin-like coating, as confirmed by UV&ndash, vis and electron paramagnetic resonance (EPR) spectroscopy.

Relying on the water-dependent hybrid conductor properties of eumelanin biopolymers, a structurally controlled melanin thin film that can be reversibly switched-on and off by hydration-dehydration cycles and used as a biocompatible platform for stem cell growth and differentiation up to 11 days is reported. A key feature of the new eumelanin-based bioelectronic interface is its remarkable structural regularity and integrity, as the result of an ad hoc fabrication protocol involving ammonia-induced solid state polymerization (AISSP) technology of a 5,6-dihydroxyindole thin film. The AISSP technology is operationally simple and versatile, enabling the preparation of device-quality thin films (AFM and MALDI-MS evidence) on various substrates with efficient chemical control over molecular complexity. Overall the results of this study pave the way for the implementation of tailored eumelanin thin films for bioelectronic devices, e.g., in organic electrochemical transistors (OECTs).

Organic (bio)electronics appears to be the first target for competitive exploitation in the materials science of eumelanins, black insoluble photoprotective human biopolymers. Nonetheless, the low conductivity of these pigments is limiting the implementation of eumelanin-based devices. Here we present a novel organic/organic hybrid material (EUGL) by integration of conductive graphene-like (GL) layers within the EUmelanin pigment (EU). GL layers were obtained by a two-step oxidation/reduction of carbon black. The stability of GL layers over a wide pH range and the self-assembling tendency place this material in a leading position for the fabrication of hybrid materials in aqueous media. EUGL was obtained by inducing the polymerization of eumelanin precursors (5,6-dihydroxyindole, DHI and 5,6-dihydroxyindole-2 carboxylic acid, DHICA) in aqueous media containing GL layers. The new material featured promising biocompatibility and an increased conductivity with respect to eumelanin by four orders of magnitude.

The growing interest toward biocompatible and bioinspired materials is boosting the investigation and the engineering of natural products as active components in electronic devices. The human pigment melanin, and particularly its subgroup composed by the eumelanins, the black-brown pigments derived from the oxidative polymerization of L-3,4-dihydroxyiphenylalanine (l-DOPA) via 5,6-dihydroxy-indole intermediates are increasingly emerging as valuable candidates for organic (bio)electronics applications. Capitalizing on a recently developed protocol to prepare high quality eumelanin coatings, this paper reports herein the design and the integration of standard commercial poly(3,4-ethylenedioxythiophene) with the poly(styrenesulfonate) (PEDOT:PSS) with eumelanin pigment. The new blend has made it possible to obtain a water stable quite transparent thin film, able to operate as an electrode for organic devices, complementing the PEDOT:PSS conductivity with the peculiar eumelanin properties, including adhesion, water stability, and ionic–electronic conductivity. As proof of concept, an unprecedented indium tin oxide-free organic light emitting diode implementing an eumelanin–PEDOT layer as the anode is fabricated and characterized. © 2017 WILEY-VCH Verlag GmbH & Co. A bulk heterojunction of porous silicon and eumelanin, where the columnar pores of porous silicon are filled with eumelanin, is proposed as a new organic-inorganic hybrid material for photovoltaic applications. The addition of eumelanin, whose absorption in the near infrared region is significantly higher than porous silicon, should greatly enhance the light absorption capabilities of the empty porous silicon matrix, which are very low in the low energy side of the visible spectral range (from about 600 nm downwards). The experimental results show that indeed the photocarrier collection efficiency at longer wavelengths in eumelanin-impregnated samples is clearly higher with respect to empty porous silicon matrices.

Establishing structure-property relationships in the black insoluble eumelanins, the key determinants of human pigmentation and skin photoprotective system, is a considerable conceptual and experimental challenge in the current drive for elucidation of the biological roles of these biopolymers and their application as advanced materials for organoelectronics. Herein, we report a new breakthrough toward this goal by the first detailed investigation on the nanoscale level of the oxidative polymerization of 5,6-dihydroxyindole (DHI), a model process of eumelanin synthesis. On the basis of a combined use of spectrophotometry, dynamic light scattering (DLS), and small-angle neutron scattering (SANS) investigations, it was possible to unveil the dynamics of the aggregation process before precipitation, the key relationships with visible light absorption and the shape of fundamental aggregates. The results indicated a polymerization mechanism of the type: Polymer(n) + DHI(x) = Polymer(n+x), where DHI(x) indicates monomer, dimer, or low oligomers (x ≤ 5). During polymerization, visible absorption increases rapidly, reaching a plateau. Particle growth proceeds slowly, with formation of 2-D structures ~55 nm thick, until precipitation occurs, that is, when large aggregates with a maximum hydrodynamic radius (R(h)) of ~1200 nm are formed. Notably, markedly smaller R(h) values, up to ~110 nm, were determined in the presence of poly(vinyl alcohol) (PVA) that was shown to be an efficient aggregation-preventing agent for polymerizing DHI ensuring water solubilization. Finally, it is shown that DHI monomer can be efficiently and partially irreversibly depleted from aqueous solutions by the addition of eumelanin suspensions. This behavior is suggested to reflect oxidant-independent competing pathways of polymer synthesis and buildup via monomer conversion on the active aggregate surface contributing to particle growth. Besides filling crucial gaps in DHI polymerization, these results support the attractive hypothesis that eumelanins may behave as a peculiar example of living biopolymers. The potential of PVA as a powerful tool for solution chemistry-based investigations of eumelanin supramolecular organization and for technological manipulation purposes is underscored.

Eumelanin is an important pigment almost ubiquitous in animals and plants exhibiting interesting charge transport capabilities. Its poor solubility in common solvents represents a severe limitation for preparing thin films. It was recently demonstrated that eumelanin films can be successfully deposited with the MAPLE (Matrix Assisted Pulsed Laser Evaporation) technique starting from a frozen water suspension, using infrared laser radiation. The low laser absorption of ice together with the high absorption of eumelanin suggests that the target ablation is due to laser energy absorbed by the eumelanin molecules, followed by thermal energy transfer, and ejection of ice/water/vapor containing undamaged eumelanin molecules and supramolecular structures. Here, we report on the deposition of eumelanin thin films on substrates at different temperatures eventually followed by in-situ annealing. Structural characterization (UV-VIS, FTIR, AP-MALDI) confirms that the deposited films maintain the characteristics of the eumelanin biopolymer. Morphological characterization (AFM) shows that surface roughness increases with increasing substrate temperature during MAPLE deposition, but is not influenced by annealing. Preliminary electrical characterization shows that eumelanin films seem to obey Ohm's law without evidence that charge injection from gold electrodes is affected by the presence of significant energy barriers. Moreover, charge transport is drastically reduced in vacuum, even if the phenomenon is at least partially reversible.

Melanins constitute a very important class of organic pigments, recently emerging as a potential material for a new generation of bioinspired biocompatible electrically active devices. In this paper, we report about the deposition of synthetic melanin films starting from aqueous suspensions by matrix assisted pulsed laser evaporation (MAPLE). In particular, we demonstrate that it is possible to deposit melanin films by MAPLE even if melanin (a) is not soluble in water and (b) absorbs light from UV to IR. AFM images reveal that the film surface features are highly depending on the deposition parameters. UV-VIS and FTIR spectra show both the optical properties and the molecular structure typical of melanins are preserved.
