Thermoanaerobacterium aotearoense SCUT27 is a prominent producer of biofuels from lignocellulosic materials. To provide sufficient NAD(P)H for ethanol production, redox-related genes, including lactate dehydrogenase (ldh), redox-sensing transcriptional repressor (rex), and hydrogenase (hfsB), were knocked out. However, the growth of strain PRH (Δldh/Δrex/ΔhfsB) was suppressed due to the intracellular redox state imbalance with the increased NADH concentration. Coincidentally, when the Bcd-EtfAB (BCD) complex was overexpressed, the resulting strain PRH-B3 (Δldh/Δrex/ΔhfsB::BCD) grew rapidly and produced ethanol with a high yield. With lignocellulosic hydrolysates, PRH-BA (Δldh/Δrex/ΔhfsB::BCD::adhE) demonstrated high ethanol productivity and yield, reaching levels of 0.45-0.51 g/L/h and 0.46-0.53 g/g sugars, respectively. The study results shed light on the cofactor balance for cell stability and the high ferredoxin-NAD + reductase activity of the BCD complex under an intracellular low redox state. The traditional strategy for isolating cellulose nanomaterials requires various chemicals and a high energy input, while achieving a low product yield owing to lignin removal during pulping and bleaching. Here, we propose a green ethanol pretreatment process with a synergistic effect using ultrasound and FeCl 3 to produce lignocellulose nanofibrils (LCNF) with a high yield (over 67 %) from thermomechanical pulp. Notably, a high-aspect-ratio LCNF with uniformly distributed lignin nanoparticles, a high lignin content, and excellent thermostability (T max  > 330 °C) with a typical cellulose I crystalline structure were successfully obtained. In addition, reduced distillation can easily retrieve the FeCl 3 solution and ethanol, reducing reagent waste. In general, FeCl 3 -catalyzed ethanol pretreatment can serve as a sustainable and environmentally friendly approach for converting sustainable lignocelluloses into high value-added nanomaterials. As the most abundant aromatic biopolymer, lignin presents great potential to produce valuable materials and chemicals. However, its large-scale value-added application is still facing many practical challenges and one of them is the unstable properties caused by lignin heterogeneity. Herein, we developed a novel "one-step-three-fractions" fractionation strategy to reduce lignin heterogeneity using aqueous two-phase system (ATPS) composed of (NH 4 ) 2 SO 4 and ethanol. In contrast to conventional step-wise fractionation processes, the proposed process subdivided heterogeneous lignin into three homogeneous fractions in only one step: the first fraction (F1) dissolved in the ethanol-rich top layer; the second fraction (F2) dissolved in the salt-rich bottom layer and the last fraction (F3) insoluble in both two layers. F2 presented the lowest molecular weight followed by F1 while F3 showed the highest molecular weight. With the increase of molecular weight, the contents of guaiacyl unit and β-O-4 linkage increased while the content of hydrophilic groups (carboxyl and aromatic hydroxyl) decreased significantly. Moreover, the ATPS exhibited satisfactory recyclability and the fractionation approach could be applied to different types/sources of lignin. In this study, two counterexamples of lignin preoxidation-hydroconversion were reported. First, two lignin feedstocks were preoxidized with 2,3-dichloro-5,6-dicyano-1,4-benzoquinone (DDQ) in acetonitrile with various dosages (15%, 30%, and 60%). Then, these preoxidized lignins (HELOs and MWLOs) were hydroconverted in supercritical ethanol catalyzed by Cu/CuMgAlO x . Total yields from HELOs were all higher than those from HEL, indicating the good promotion of DDQ preoxidation on the subsequent hydroconversion of HELOs, especially with the DDQ dosage of 15%. Differently, the promotion effect of DDQ preoxidation on the hydroconversion of MWLOs depended on the DDQ dosage as well as the reaction time. Through the comparison of two counterexamples, this work bursted the myth that preoxidation can always promote the subsequent hydroconversion of lignin, revealed the influence of lignin property, preoxidation degree, and reaction conditions on the subsequent hydroconversion of preoxidized lignin, and presented the new insight into the preoxidation-hydroconversion strategy for lignin., (Copyright © 2021 Elsevier Ltd. Lignocellulosic biomass fractionaion into its three major components is critically important for efficient feedstock utilization. The hydrothermal-ethanol method has broad application as its first step, hydrothermal treatment, provides high hemicellulose separation efficiency. However, it severely inhibits the delignification on the subsequent ethanol extraction. In this study, the second step, ethanol extraction, was facilitated by the addition of 3% NaOH and 3% H 2 O 2 , resulting in a significant improvement of lignin separation (by 48.2%). SEM, AFM, XPS, and XRD were used to characterize the surface composition of the remaining solids (crude cellulose) while the structure of isolated lignin was characterized by FT-IR, CP/MAS 13C NMR, GPC and TGA. The lignin samples isolated with both facilitated and non-facilitated ethanol extraction were compared to elucidate the lignin removal mechanism. In this work, lignin fractionation is proposed as an effective approach to reduce the heterogeneity of lignin and improve the adsorption and recycle performances of lignin as a cationic dye adsorbent. By stepwise dissolution of enzymatic hydrolysis lignin in 95% and 80% ethanol solutions, three lignin subdivisions (95% ethanol-soluble subdivision, 80% ethanol-soluble subdivision, and 80% ethanol-insoluble subdivision) were obtained. The three lignin subdivisions were characterized by gel permeation chromatography (GPC), FTIR, 2D-NMR and scanning electron microscopy (SEM), and their adsorption capacities for methylene blue were compared. The results showed that the 80% ethanol-insoluble subdivision exhibited the highest adsorption capacity and its value (396.85 mg/g) was over 0.4 times higher than that of the unfractionated lignin (281.54 mg/g). The increased adsorption capacity was caused by the enhancement of both specific surface area and negative Zeta potential. The maximum monolayer adsorption capacity of 80% ethanol-insoluble subdivision by adsorption kinetics and isotherm studies was found to be 431.1 mg/g, which was much higher than most of reported lignin-based adsorbents. Moreover, the 80% ethanol-insoluble subdivision had much higher regeneration yield (over 90% after 5 recycles) compared with the other two subdivisions. Consequently, the proposed fractionation method is proved to be a novel and efficient non-chemical modification approach that significantly improves adsorption capacity and recyclability of lignin.

The redox-sensing transcriptional repressor Rex (Rex) displayed diverse functions in different microbial species. Nowadays, only part function of rex has been verified in vitro and alcohol dehydrogenase gene (adhE) as the target of Rex has been widely reported. In this study, rex was knocked out in Thermoanaerobacterium aotearoense SCUT27 (GDMCC 60765) and the carbon metabolic distribution analysis was performed. Results showed that the ethanol yield (mol product/mol carbon) of SCUT27(Δrex) had increased by 75.00-90.91%, cell growth improved by 27.27-36.36%, and acetic acid and lactic acid decreased by 58.33-61.54% accompanied with the yield of hydrogen decreased by 46.15-58.35% within different carbon sources. The ability of sugar consumption of SCUT27(Δrex) had improved about 74.19-130.55% with the improvement of total ATP concentration and the cofactors NADH and NAD + concentrations. In addition, the specific activities of alcohol dehydrogenase of SCUT27(Δrex) with NADH and NADPH as cofactors were improved by 119.26-140.28% and 35.66-47.69%, respectively. After ldh was further knocked out in SCUT27(Δrex), SCUT27(ΔldhΔrex) showed higher ethanol production and yield when various carbon resources were used as substrates (including glucose, xylose, glucose/xylose mixture and three kinds of lignocellulosic hydrolysates). This study confirms that Rex is an important regulator for determining products distribution in SCUT27 and deletion of rex and ldh is a promising strategy for enhanced ethanol production.

Over the recent years, the exploitation of antibacterial performance of lignin and its subsequent usage as bacteriostatic additives has gained increasing interests. However, due to the restriction from structural heterogeneity, the antibacterial activity of lignin is always modest and unstable (especially against Gram-negative bacteria). In this regard, we proposed a facile one-step ethanol fractionation process to decrease the heterogeneity of lignin and, therefore, improve its antibacterial activity. Two fractions (Fs and Fi, 95% ethanol soluble and insoluble fraction, respectively) were obtained from bamboo kraft lignin (BKL) and their antibacterial effectiveness was compared. The results showed that the antibacterial activity of Fs increased significantly compared to that of BKL. On the contrary, Fi barely exhibited inhibitory effect on the growth of two Gram-positive bacteria and even promoted the growth of two Gram-negative bacteria. The much lower phenolic-OH content in Fi was an important reason for the absence of antibacterial activity. Besides, the growth promotion of Gram-negative bacteria by Fi was possibly caused by the formation of insoluble carriers for bacteria growth due to the poor water-solubility of Fi. This study was done with the aim of lignosulfonate and ethanol production from different spent liquors of bagasse pulping process. For this purpose, alkali lignin from bagasse alkali liquor was separated and was sulfomethylated to produce soda lignosulfonate (SLig). Furthermore, raw bagasse was directly treated with sodium sulfite in acidic and neutral conditions to produce BLig and NLig bagasse lignosulfonate, respectively. In addition, the pentoses and hexoses impurities in lignosulfonates were fermented to ethanol using Candida guilliermondii. Results showed that the molecular weight of NLig lignosulfonate was considerably high comparing to the SLig and BLig lignosulfonates. A high level of thermal resistance was found in case of SLig regarding to the other samples at 500 °C. Fermentation of the spent liquors with Candida guilliermondii led to a maximum ethanol yield of 7.0, 1.0 and 5.1 g L -1 in NLig, SLig and BLig, respectively., (Copyright © 2018 Elsevier Ltd. Consolidated bioprocessing (CBP) by micro-organisms is desired for efficient conversion of lignocellulosic biomass to bioethanol fuels. Potential candidates have been discovered, including cellulolytic bacteria and filamentous fungi. Genetic and metabolic manipulation of these organisms further promotes their fermentation capacities and the ethanol tolerance. In addition, Saccharomyces cerevisiae and several other yeasts were genetically modified to express recombinant cellulases in media or display them on the cell surface for CBP of cellulose. To compensate the insufficient capacity of a single strain, various microbial consortia have also been developed. In this article, we reviewed the recent advances in CBP microbes and focused on the efforts in strain improvement employing genetic engineering.

The fractionation of wheat straw by ethanol-water (EW) pre-treatment was studied regarding its main operating conditions: time, temperature, L/S ratio and ethanol percentage were optimized by using an orthogonal experimental design (Taguchi). Afterwards, diluted sulfuric acid (DSA) hydrolysis and EW treatments have been compared in terms of energy consumption and yield of a cellulosic solid residue able to be enzymatically hydrolyzed to glucose. Experimental results show that temperature is the only variable of EW with a significant effect on the quality of the pretreated solids. EW pre-treatment of wheat straw is more effective than DSA hydrolysis due to its higher capacity of delignification. Moreover, a high glucose yield (80%) can be obtained by enzymatic hydrolysis of a solid pretreated with a moderate energy input EW (160 °C, 45 min) while wheat straw needs of a higher energy input during DSA to produce a similar yield of glucose after saccharification., (Copyright © 2018 Elsevier Ltd. The alcoholic depolymerization and Mannich reaction were conducted to improve the chemical activity of biorefinery technical lignins and introduce amino groups into lignins, respectively. To understand the chemical structural transformations and examine the reaction mechanism, GPC and solution-state NMR techniques were performed. Element analysis was also used to quantify the amount of amine groups. The NMR characterization the depolymerized lignins indicated of the depolymerization, demethoxylation, and bond cleavage of linkages occurred during the depolymerization process. Results showed that the depolymerization temperature instead of the addition of capping reagents was the main factor for improving the reactivity of lignin under the given conditions. The Mannich reaction was very selective, primarily occurred at H 3,5 and G 5 positions, and the H units present a higher chemical reactivity. It is believed that the understanding of the fundamental chemistry of lignin during depolymerization and Mannich reaction process will contribute to the extension of high value-added applications of biorefinery lignin., (Copyright © 2017 Elsevier B.V. A sequential two-stage pretreatment process comprising alkaline pre-extraction and alkaline hydrogen peroxide pretreatment (AHP) was investigated to convert bamboo carbohydrates into bioethanol. The results showed that mild alkaline pre-extraction using 8% (w/w) sodium hydroxide (NaOH) at 100°C for 180min followed by AHP pretreatment with 4% (w/w) hydrogen peroxide (H 2 O 2 ) was sufficient to generate a substrate that could be efficiently digested with low enzyme loadings. Moreover, alkali pre-extraction enabled the use of lower H 2 O 2 charges in AHP treatment. Two-stage pretreatment followed by enzymatic hydrolysis with only 9FPU/g cellulose led to the recovery of 87% of the original sugars in the raw feedstock. The use of the pentose-hexose fermenting Saccharomyces cerevisiae SR8u strain enabled the utilization of 95.7% sugars in the hydrolysate to reach 4.6%w/v ethanol titer. A stepwise ethanol-precipitation (SEP) procedure was developed for the purification of sugar beet pectins (SBP) from a pectin-containing aqueous extract. Five fractions of different chemical and molecular characteristics were produced by stepwise elevating the alcohol concentration of the precipitation medium from 50% to 80% v/v. Comparison of chemical and macromolecular features between the obtained fractions indirectly suggested that the ability of pectin to solubilize in the ethanol-water binary mixture depended greatly on the polymer structure. Fractions rich in neutral sugars were precipitated at relatively high ethanol concentrations, probably due to the enhanced interactions generated between pectin side chains and solvent molecules. Furthermore, the obtained fractions displayed different surface activities. In the present study, a graded ethanol precipitation technique was employed to obtain hemicelluloses from the alkali-extracted corncob liquid. The relationship between the structural characteristics of alkali-soluble corncob hemicelluloses and the production of furfural was investigated by a heterogeneous process in a biphasic system. Results showed that alkali-soluble corncob hemicelluloses mainly consisted of glucuronoarabinoxylans and L-arabino-(4-O-methylglucurono)-D-xylans, and the drying way had less influence on the sugar composition, molecular weights and the functional groups of hemicelluloses obtained by the different ethanol concentration precipitation except for the thermal property, the amorphous structure and the ability for the furfural production. Furthermore, alkali-soluble corncob hemicelluloses with higher xylose content, lower branch degree, higher polydispersity and crystallinity contributed to the furfural production. A highest furfural yield of 45.41% with the xylose conversion efficiency of 99.06% and the furfural selectivity of 45.84% was obtained from the oven-dried hemicelluloses precipitated at the 30% (v/v) ethanol concentration., (Copyright © 2015 Elsevier Ltd. Background: Enantiopure (S)-1-(4-methoxyphenyl) ethanol {(S)-MOPE} can be employed as an important synthon for the synthesis of cycloalkyl [b] indoles with the treatment function for general allergic response. To date, the biocatalytic resolution of racemic MOPE through asymmetric oxidation in the biphasic system has remained largely unexplored. Additionally, deep eutectic solvents (DESs), as a new class of promising green solvents, have recently gained increasing attention in biocatalysis for their excellent properties and many successful examples in biocatalytic processes. In this study, the biocatalytic asymmetric oxidation of MOPE to get (S)-MOPE using Acetobacter sp. CCTCC M209061 cells was investigated in different two-phase systems, and adding DES in a biphasic system was also explored to further improve the reaction efficiency of the biocatalytic oxidation., Results: Of all the examined water-immiscible organic solvents and ionic liquids (ILs), 1-butyl-3-methylimidazolium hexafluorophoshpate ([C4MIM][PF6]) afforded the best results, and consequently was selected as the second phase of a two-phase system for the asymmetric oxidation of MOPE with immobilized Acetobacter sp. CCTCC M209061 cells. For the reaction performed in the [C4MIM][PF6]/buffer biphasic system, under the optimized conditions, the initial reaction rate, the maximum conversion and the residual substrate e.e. recorded 97.8 μmol/min, 50.5 and >99.9 % after 10 h reaction. Furthermore, adding the DES [ChCl][Gly] (10 %, v/v) to the aqueous phase, the efficiency of the biocatalytic oxidation was rose markedly. The optimal substrate concentration and the initial reaction rate were significantly increased to 80 mmol/L and 124.0 μmol/min, respectively, and the reaction time was shortened to 7 h with 51.3 % conversion. The immobilized cell still retained over 72 % of its initial activity after 9 batches of successive reuse in the [C4MIM][PF6]/[ChCl][Gly]-containing buffer system. Additionally, the efficient biocatalytic process was feasible up to a 500-mL preparative scale., Conclusion: The biocatalytic asymmetric oxidation of MOPE with Acetobacter sp. CCTCC M209061 cells was successfully conducted in the [C4MIM][PF6]-containing biphasic system with high conversion and enantioselectivity, and the reaction efficiency was further enhanced by adding [ChCl][Gly] to the reaction system. The efficient biocatalytic process was promising for the preparation of enantiopure (S)-MOPE.

This work reports on a full evaporation headspace gas chromatographic (FE HS-GC) method for simultaneously determining the ethanol (EtOH) and methanol (MeOH) content in wines. A small sample (10μL) was placed in a headspace sample vial, and a near-complete mass transfer of ethanol and methanol from the liquid sample to the vapor phase was obtained within three minutes at a temperature of 105°C, which allowed the measurement of the EtOH and MeOH content in the sample by GC. The results showed excellent precision and accuracy, as shown by the reproducibilities of 1.02% and 2.11% for EtOH and MeOH, respectively, and recoveries that ranged from 96.1% to 104% for both alcohols. In this study we investigated the technical feasibility of convert paper mill sludge into fuel ethanol. This involved the removal of mineral fillers by using either chemical pretreatment or mechanical fractionation to determine their effects on cellulose hydrolysis and fermentation to ethanol. In addition, we studied the effect of cationic polyelectrolyte (as accelerant) addition and hydrogen peroxide pretreatment on enzymatic hydrolysis and fermentation. We present results showing that removing the fillers content (ash and calcium carbonate) from the paper mill sludge increases the enzymatic hydrolysis performance dramatically with higher cellulose conversion at faster rates. The addition of accelerant and hydrogen peroxide pretreatment further improved the hydrolysis yields by 16% and 25% (g glucose / g cellulose), respectively with the de-ashed sludge. Sulfite pretreatment to overcome the recalcitrance of lignocelluloses (SPORL) was applied to poplar NE222 chips in a range of chemical loadings, temperatures, and times. The combined hydrolysis factor (CHF) as a pretreatment severity accurately predicted xylan dissolution by SPORL. Good correlations between CHF and pretreated solids enzymatic digestibility, sugar yield, and the formations of furfural and acetic acid were obtained. Therefore, CHF was used to balance sugar yield with the formation of fermentation inhibitors for high titer ethanol production without detoxification. The results indicated that optimal sugar yield can be achieved at CHF=3.1, however, fermentation using un-detoxified whole slurries of NE222 pretreated at different severities by SPORL indicated CHF≈2 produced best results. An ethanol titer of 41 g/L was achieved at total solids of approximately 20 wt% without detoxification with a low cellulase loading of 15 FPU/g glucan (27 mL/kg untreated wood)., (Published by Elsevier Ltd.)

This study demonstrated at the pilot-scale (50 kg) use of Douglas-fir forest harvest residue, an underutilized forest biomass, for the production of high titer and high yield bioethanol using sulfite chemistry without solid-liquor separation and detoxification. Sulfite Pretreatment to Overcome the Recalcitrance of Lignocelluloses (SPORL) was directly applied to the ground forest harvest residue with no further mechanical size reduction, at a low temperature of 145°C and calcium bisulfite or total SO2 loadings of only 6.5 or 6.6 wt% on oven dry forest residue, respectively. The low temperature pretreatment facilitated high solids fermentation of the un-detoxified pretreated whole slurry. An ethanol yield of 282 L/tonne, equivalent to 70% theoretical, with a titer of 42 g/L was achieved. SPORL solubilized approximately 45% of the wood lignin as directly marketable lignosulfonate with properties equivalent to or better than a commercial lignosulfonate, important to improve the economics of biofuel production., (Published by Elsevier Ltd.)

We present a general framework to predict the excess solubility of small molecular solids (such as pharmaceutical solids) in binary solvents via molecular simulation free energy calculations at infinite dilution with conventional molecular models. The present study used molecular dynamics with the General AMBER Force Field to predict the excess solubility of acetanilide, acetaminophen, phenacetin, benzocaine, and caffeine in binary water/ethanol solvents. The simulations are able to predict the existence of solubility enhancement and the results are in good agreement with available experimental data. The accuracy of the predictions in addition to the generality of the method suggests that molecular simulations may be a valuable design tool for solvent selection in drug development processes.

Sulfite pretreatment to overcome the recalcitrance of lignocelluloses (SPORL) was applied to an empty fruit bunches (EFB) for ethanol production. SPORL facilitated delignification through lignin sulfonation and dissolution of xylan to result in a highly digestible substrate. The pretreated whole slurry was enzymatically saccharified at a solids loading of 18% using a relatively low cellulase loading of 15 FPU/g glucan and simultaneously fermented without detoxification using Saccharomyces cerevisiae of YRH400. An ethanol yield of 217 L/tonne EFB was achieved at titer of 32 g/L. Furfural residues (FR), the abundant lignocellulosic residues from commercial furfural production, were delignified with alkaline peroxide process and then taken as substrates for ethanol production by simultaneous saccharification and fermentation (SSF). It was apparent that the delignification efficiency was increased with higher chemical addition and temperature, reaching the maximum removal (73.5%) of lignin. The widespread accessible-cellulose in FR favored the enzymatic hydrolysis and achieved the considerable bioconversion (75.7% with 5 FPU+10 IU/g substrate). The delignification process increased the relative glucose content and then the bioconversion efficiency, closely relating to the increased specific surface area. As the cellulose contents were higher than 60%, the final conversions conversely fell to around 75%, probably due to the insufficient utilization of all active cellulose with low enzyme cocktails addition. Although the SSF bioconversion slightly decreased as the elevated amount of fermentable cellulose, the maximum of ethanol concentration (16.9 g/L) was expectedly obtained., (Crown Copyright © 2013. Rejects from sulfite pulp mill that otherwise would be disposed of by incineration were converted to ethanol by a combined physical-biological process that was comprised of physical refining and simultaneous saccharification and fermentation (SSF). The energy efficiency was evaluated with comparison to thermochemically pretreated biomass, such as those pretreated by dilute acid (DA) and sulfite pretreatment to overcome recalcitrance of lignocelluloses (SPORL). It was observed that the structure deconstruction of rejects by physical refining was indispensable to effective bioconversion but more energy intensive than that of thermochemically pretreated biomass. Fortunately, the energy consumption was compensated by the reduced enzyme dosage and the elevated ethanol yield. Furthermore, adjustment of disk-plates gap led to reduction in energy consumption with negligible influence on ethanol yield. Concentrated dilute acid hydrolysate was obtained from hot water extracts of Acer saccharum (sugar maple) and was fermented to ethanol by Pichia stipitis in a 1.3-L-benchtop bioreactor. The conditions under which the highest ethanol yield was achieved were when the air flow rate was set to 100 cm(3) and the agitation rate was set to 150 rpm resulting in an overall mass transfer coefficient (K(L)a) of 0.108 min(-1). A maximum ethanol concentration of 29.7 g/L was achieved after 120 h of fermentation; however, after 90 h of fermentation, the ethanol concentration was only slightly lower at 29.1 g/L with a yield of 0.39 g ethanol per gram of sugar consumed. Using the same air flow rate and adjusting the agitation rate resulted in lower ethanol yields of 0.25 g/g at 50 rpm and 0.30 g/g at 300 rpm. The time it takes to reach the maximum ethanol concentration was also affected by the agitation rate. The ethanol concentration continued to increase even after 130 h of fermentation when the agitation rate was set at 50 rpm, whereas the maximum ethanol concentration was reached after only 68.5 h at 300 rpm.

Woody biomass is renewable only if sustainable production is imposed. An optimum and sustainable biomass stand production rate is found to be one with the incremental growth rate at harvest equal to the average overall growth rate. Utilization of woody biomass leads to a sustainable economy. Woody biomass is comprised of at least four components: extractives, hemicellulose, lignin and cellulose. While extractives and hemicellulose are least resistant to chemical and thermal degradation, cellulose is most resistant to chemical, thermal, and biological attack. The difference or heterogeneity in reactivity leads to the recalcitrance of woody biomass at conversion. A selection of processes is presented together as a biorefinery based on incremental sequential deconstruction, fractionation/conversion of woody biomass to achieve efficient separation of major components. A preference is given to a biorefinery absent of pretreatment and detoxification process that produce waste byproducts. While numerous biorefinery approaches are known, a focused review on the integrated studies of water-based biorefinery processes is presented. Hot-water extraction is the first process step to extract value from woody biomass while improving the quality of the remaining solid material. This first step removes extractives and hemicellulose fractions from woody biomass. While extractives and hemicellulose are largely removed in the extraction liquor, cellulose and lignin largely remain in the residual woody structure. Xylo-oligomers, aromatics and acetic acid in the hardwood extract are the major components having the greatest potential value for development. Higher temperature and longer residence time lead to higher mass removal. While high temperature (>200°C) can lead to nearly total dissolution, the amount of sugars present in the extraction liquor decreases rapidly with temperature. Dilute acid hydrolysis of concentrated wood extracts renders the wood extract with monomeric sugars. At higher acid concentration and higher temperature the hydrolysis produced more xylose monomers in a comparatively shorter period of reaction time. Xylose is the most abundant monomeric sugar in the hydrolysate. The other comparatively small amounts of monomeric sugars include arabinose, glucose, rhamnose, mannose and galactose. Acetic acid, formic acid, furfural, HMF and other byproducts are inevitably generated during the acid hydrolysis process. Short reaction time is preferred for the hydrolysis of hot-water wood extracts. Acid hydrolysis presents a perfect opportunity for the removal or separation of aromatic materials from the wood extract/hydrolysate. The hot-water wood extract hydrolysate, after solid-removal, can be purified by Nano-membrane filtration to yield a fermentable sugar stream. Hemicelluloses in industrially produced pre-hydrolysis liquor (PHL) were precipitated with ethanol. These PHL-derived hemicelluloses (PHL-EH) and a commercial, pure birch wood xylan sample (powder form) (BWX) were bleached using chlorine dioxide (D(0) and D(1)) and hydrogen peroxide (Ep) in the D(0)EpD(1) sequence, and the chemical compositions, molecular weights and charge densities of the treated samples were assessed. When applied to high-yield pulp (HYP) at 50 mg/g, 26 and 20 mg/g of the bleached PHL-EH and BWX, respectively, were adsorbed without significantly affecting paper properties. These results suggest that semi-bleached hemicelluloses could be used to increase the basis weight of paper products. Furthermore, an integrated process was proposed that converts the kraft-based dissolving pulp production process into a biorefinery unit with dissolving pulp, bleached hemicelluloses and lignin as main products., (Copyright © 2011 Elsevier Ltd. In this work, the potential of Acacia dealbata as raw material for ethanol production was evaluated, as well as its composition with regard to cellulose, hemicelluloses, lignin, extractives and ash. The tree samples were subjected to several dilute acid pretreatments using a combined severity parameter ranging from 0.7 to 3.7. The highest ethanol concentration obtained was 10.31 g ethanol/L within 24 h by using a separate hydrolysis and fermentation of the water insoluble fraction after pretreatment at 180 °C with 0.8% of sulfuric acid for 15 min. With simultaneous saccharification and fermentation, results obtained for the washed solids of water insoluble fraction were better than those obtained with the whole slurry., (Copyright © 2011 Elsevier Ltd. In this paper, we proposed a new modification for an ethanol-based pulping process, which would consist of the pre-hydrolysis (pre-extraction) of wood chips for removing hemicelluloses; the ethanol extraction of pre-hydrolyzed wood chips for removing lignin; and the post purification of cellulose, leading to the production of pure cellulose. We also experimentally evaluated the separation of hemicelluloses from the pre-hydrolysis liquor (PHL) obtained from a pulp mill. To remove lignin from the PHL, it was acidified to a pH of 2, which resulted in 47% lignin precipitation. The lignin separation from the acidified PHL was further improved via adding polyethylene oxide and poly aluminum chloride or adding ethyl acetate. To recover the hemicelluloses from the acidified PHL, ethanol was added to the acidified PHL with a volumetric ratio of 4 to 1. The isolated lignin and hemicelluloses were characterized by a Fourier transform infrared spectroscopy (FTIR) and a gas permeation chromatography (GPC)., (Copyright © 2010 Elsevier Ltd. Under the current energy scenario, the development of alternatives to fossil fuels, like bioethanol from lignocellulosic materials, is highly relevant. Therefore it is important to search and study new raw materials and to optimize the different steps that lead to bioethanol production. In this work, acid diluted pretreatment was optimized considering the release of sugars. Under the optimal conditions, the reducing sugars yield was of 293.4mg/g of dry biomass in liquid fraction. The tested pretreated samples of Pterospartum tridentatum that presented a higher glucose yield in enzymatic saccharification where those that were subject to a pretreatment at 180°C for 75min with 2.75% (w/w) of sulfuric acid when using a biomass/liquid ratio of 2.25g/10mL leading to a maximum yield of glucose that was 92% of the theoretical maximum. Hemicellulose is a potential by-product currently under-utilized in the papermaking industry. It is a hetero-carbohydrate polymer. For hardwood hemicelluloses, D-xylose is the major component upon depolymerization. At SUNY-ESF, wood extracts were obtained by extracting sugar maple wood chips with hot water at an elevated temperature. The wood extracts were then concentrated and acid hydrolyzed. Ethanologenic bacteria, E. coli FBR5, had a good performance in pure xylose medium for ethanol production. However, FBR5 was strongly inhibited in dilute sulfuric acid hydrolyzate of hot-water wood extract. FBR5 was challenged by hot-water wood extract hydrolyzate in this study. After repeated strain adaptation, an improved strain: E. coli FBHW was obtained. Fermentation experiments indicated that FBHW was resistant to the toxicity of hydrolyzate in the fermentation media of concentrated hydrolyzate, and xylose was completely utilized by the strain to produce ethanol. FBHW was grown in the concentrated hydrolyzate without any detoxification treatment and has yielded 36.8g/L ethanol., (Copyright 2010 Elsevier Inc. All rights reserved.)

A series of Ni-based catalysts were prepared using hydrogen reduction of Ni/Al hydrotalcite-like compounds (Ni/Al HTlcs) synthesized by coprecipitation. The physico-chemical properties of Ni/Al hydrotalcite-like compounds and the corresponding Ni-based catalysts were characterized using inductively coupled plasma (ICP), BET surface areas, X-ray diffraction (XRD), Fourier transform infrared (FTIR) spectroscopy and scanning electron microscopy (SEM) techniques. The results indicated that Ni/Al HTlcs with layered structures could be successfully prepared by the coprecipitation method, and the characteristic HTlcs reflections were also observed in the XRD analysis. The NiO and Ni0 phases were identified in all Ni-based catalysts, which displayed randomly interconnected pores and no layer structures. In addition, the studies also found the Ni/Al HTlcs and Ni-based catalysts had high specific surface areas, low pore volumes and low pore diameters. The catalytic hydrogenation of ethyl acetate to ethanol with Ni-based catalysts was also investigated. Among the studied catalysts, RE1NASH-110-3 showed the highest selectivity and yield of ethyl acetate to ethanol, which were 68.2% and 61.7%, respectively. At the same time, a major by-product, butyl acetate, was formed due to an ester-exchange reaction. A proposed hydrogenation pathway for ethyl acetate over Ni-based catalysts was suggested.

The two hemicellulosic fractions were subsequentially extracted with 5% and 8% NaOH aqueous solution at a solid to liquid ratio of 1:25 (g mL(-1)) at 50 degrees C for 3 h from the water, 1 and 3% NaOH-treated sugar cane bagasse, and subfractionated into six preparations by a graded ethanol precipitation method at concentrations of 15%, 30% and 60% (v/v). Sugar composition and molecular weight analysis showed that, with an increasing concentration of ethanol, hemicellulosic subfractions with both higher Ara/Xyl ratios and higher molecular weights were obtained. In other words, with an increasing ethanol concentration from 15% to 60%, the Ara/Xyl ratios increased from 0.043 in H(1) to 0.088 in H(3) and from 0.040 in H(4) to 0.088 in H(6), and the weight-average molecular weights of hemicellulosic subfractions increased from 42 430 (H(1)) to 85 510 (H(3)) g mol(-1) and from 46 130 (H(4)) to 64 070 (H(6)) g mol(-1), respectively. The results obtained by the analysis of Fourier transform infrared, sugar composition, and (1)H and (13)C nuclear magnetic spectroscopy showed that the alkali-soluble hemicelluloses had a backbone of xylose residues with a beta-(1-->4)-linkage and were branched mainly through arabinofuranosyl units at C-2 and/or C-3 of the main chain, whereas the differences may occur in the distribution of branches along the xylan backbone.

The sequential treatment of dewaxed sugarcane bagasse with H(2)O and 1 and 3% NaOH at a solid to liquid ratio of 1:25 (g mL(-1)) at 50 degrees C for 3 h yielded 74.9% of the original hemicelluloses. Each of the hemicellulosic fractions was successively subfractionated by graded precipitation at ethanol concentrations of 15, 30, and 60% (v/v). Chemical composition, physicochemical properties, and structures of eight precipitated hemicellulosic fractions were elucidated by a combination of sugar analysis, nitrobenzene oxidation of bound lignin, molecular determination, Fourier transform infrared (FT-IR), (1)H and (13)C nuclear magnetic spectroscopies, and thermal analysis. The results showed that the sequential treatments and graded precipitations were very effective on the fractionation of hemicelluloses from bagasse. Comparison of these hemicelluloses indicated that the smaller sized and more branched hemicelluloses were extracted by the hot water treatment; they are rich in glucose, probably originating from alpha-glucan and pectic polysaccharides. The larger molecular size and more linear hemicelluloses were dissolved by the alkali treatment; they are rich in xylose, principally resulting from l-arabino-(4-O-methylglucurono)-d-xylans. In addition, noticeable differences in the chemical composition and molecular weights were observed among the graded hemicellulosic subfractions from the water-soluble and alkali-soluble hemicelluloses. The Ara/Xyl ratio increased with the increment of ethanol concentration from 15 to 60%, and the arabinoxylans with higher Ara/Xyl ratios had higher molecular weights. There were no significant differences in the structural features of the precipitated hemicellulosic subfractions, which are mainly constituted of l-arabino-(4-O-methyl-d-glucurono)xylan, whereas the difference may occur in the distribution of branches along the xylan backbone.

This paper reports a full evaporation (FE) headspace gas chromatographic (GC) method for rapid determination of ethanol in fermentation liquor. The data show that ethanol in the fermentation liquor was transferred to the vapor phase (headspace) almost completely within 3 min at a temperature of 105 degrees C when a very small volume (< 50 microL) of sample was directly added to a sealed headspace sample vial (20 mL). The ethanol in the vapor phase was then measured by headspace GC using a flame ionization detector. The results show that the present method has an excellent measurement precision (RSD=1.62%) and accuracy (recovery=98.1 (+/-1.76%)) for the ethanol quantification in fermentation liquors. The method requires no sample pretreatment and is very simple and rapid.

Lignocellulose hydrolysates contain fermentation inhibitors causing decreased ethanol production. The inhibitors include phenolic compounds, furan aldehydes, and aliphatic acids. One of the most efficient methods for removing inhibiting compounds prior to fermentation is treatment of the hydrolysate with ion-exchange resins. The performance and detoxification mechanism of three different resins were examined: an anion exchanger, a cation exchanger, and a resin without charged groups (XAD-8). A dilute acid hydrolysate of spruce was treated with the resins at pH 5.5 and 10.0 prior to ethanolic fermentation with Saccharomyces cerevisiae. In addition to the experiments with hydrolysate, the effect of the resins on selected model compounds, three phenolics (vanillin, guaiacol, and coniferyl aldehyde) and two furan aldehydes (furfural and hydroxymethyl furfural), was determined. The cation exchanger increased ethanol production, but to a lesser extent than XAD-8, which in turn was less effective than the anion exchanger. Treatment at pH 10.0 was more effective than at pH 5.5. At pH 10.0, the anion exchanger efficiently removed both anionic and uncharged inhibitors, the latter by hydrophobic interactions. The importance of hydrophobic interactions was further indicated by a substantial decrease in the concentration of model compounds, such as guaiacol and furfural, after treatment with XAD-8.

Ethanol production from spent sulphite pulping liquor (SSL) was compared for four different yeasts. A second strain of S. cerevisiae as well as a 2-deoxyglucose-resistant strain formed through protoplast fusions between S. uvarum and S. diastaticus produced up to 27% more ethanol from SSL fortified with hydrolysis sugars than was produced by S. cerevisiae. The incremental improvement in ethanol yield appeared to vary with the degree of fortification, ranging from 5.8% for unfortified SSL, to 27% for the highest level of fortification tested. Decreasing fermentation rates were observed for SSL fortified with glucose, mannose and galactose, respectively. Sugar uptake rates in SSL fortified with glucose, galactose and mannose were 6.8, 2.8 and 2.0 g L-1 h-1, respectively. However, when these sugars were fermented along with a glucose cosubstrate, the rate at which the combined glucose/mannose medium was fermented was nearly identical to that of the glucose control.
