Fine particulate matter, a major air pollutant across the world, causes a series of pulmonary diseases. Vitamin D is a typical vitamin with emerging roles in inflammation and fibrosis. Different situations and diseases need different doses and modes of vitamin D administration, which challenges the existing vitamin D supplementary rules. Thus, studies of vitamin D applications and their mechanisms in various diseases are important for its future therapeutic applications. In this study, the therapeutic application of vitamin D3 in chronic particle-exposure-associated lung fibrosis and tissue remodeling was investigated. In vivo studies showed that vitamin D3 significantly attenuated fibrosis effects by decreasing α-smooth muscle actin-regulated extracellular matrix deposition and restoring expressions of E-cadherin and N-cadherin. With the importance of activated macrophage in the regulation of local epithelium and fibroblast in the process of tissue fibrosis, two separate in vitro systems of co-culture of macrophages with lung epithelium or fibroblast were built. The results confirmed that vitamin D3 promoted the proliferation of lung epithelium and depressed the fibrosis effects of fibroblasts as well. In addition, our results indicated that the therapeutic effects of vitamin D3 were through Nrf2 signals. Our work provides convincing experimental evidence for vitamin D therapeutic application to promote tissue repair and improve particle-associated lung fibrosis.

Lead exposure is known to affect the pituitary-thyroid axis. Likewise, ascorbic acid (AA) has a protective action against lead poisoning. We examine the protective role of AA in lead-induced damage to the thyroid gland. The Wistar rats were divided into three groups: control that received 0.2% AA in drinking water throughout the experiment (15 days), intoxicated with lead acetate (20 mg/kg) intraperitoneally every 48 h for 15 days, and the experimental group treated with lead acetate and 0.2% AA in drinking water throughout the experiment. Plasma thyroid-stimulating hormone, triiodothyronine, thyroxine, and lead were determined. The thyroid gland was weighed, then epithelial cell height and nuclear volume were measured on histological slides. The results show that AA reduced the thyroid atrophy caused by lead acetate, as well as the loss of weight of the gland. In addition, it prevented the decrease of the hormone triiodothyronine, although the thyroxine hormone remained lower than the control values ​​and the thyroid-stimulating hormone remains high. Our results indicated that AA could play a protective role in lead poisoning in the thyroid gland.

Purpose: Prostate cancer primarily arises from the glandular epithelium. Histomophometric techniques have been used to assess the glandular epithelium in automated detection and classification pipelines; however, they are often rigid in their implementation, and their performance suffers on large datasets where variation in staining, imaging, and preparation is difficult to control. The purpose of this study is to quantify performance of a pixelwise segmentation algorithm that was trained using different combinations of weak and strong stroma, epithelium, and lumen labels in a prostate histology dataset. Approach: We have combined weakly labeled datasets generated using simple morphometric techniques and high-quality labeled datasets from human observers in prostate biopsy cores to train a convolutional neural network for use in whole mount prostate labeling pipelines. With trained networks, we characterize pixelwise segmentation of stromal, epithelium, and lumen (SEL) regions on both biopsy core and whole-mount H&E-stained tissue. Results: We provide evidence that by simply training a deep learning algorithm on weakly labeled data generated from rigid morphometric methods, we can improve the robustness of classification over the morphometric methods used to train the classifier. Conclusions: We show that not only does our approach of combining weak and strong labels for training the CNN improve qualitative SEL labeling within tissue but also the deep learning generated labels are superior for cancer classification in a higher-order algorithm over the morphometrically derived labels it was trained on.

This study aims to determine if light scatter parameters measured with spatial frequency domain imaging (SFDI) can accurately predict stromal, epithelial, and adipose fractions in freshly resected, unstained human breast specimens. An explicit model was developed to predict stromal, epithelial, and adipose fractions as a function of light scattering parameters, which was validated against a quantitative analysis of digitized histology slides for N = 31 specimens using leave-one-out cross-fold validation. Specimen mean stromal, epithelial, and adipose volume fractions predicted from light scattering parameters strongly correlated with those calculated from digitized histology slides (r = 0.90, 0.77, and 0.91, respectively, p-value

The presence of unidentified estrogens and androgens in effluents from pulp and paper mills is well documented. However, their role in effluent effects on fish reproduction remains unclear. The objective of this study was to investigate the hypothesis that reproductive impacts of a modern pulp mill effluent are mediated by androgens and/or estrogens in the effluent. Male and female threespine stickleback were exposed to biologically treated Canadian bleached kraft mill effluent under flow-through conditions in the laboratory at 0, 1, 10 and 100% (v/v) dilutions. After 7 and 21 d of exposure, steroidogenesis was assessed using in vitro incubations of gonadal tissue in both males and females. mRNA expression of the estrogen-regulated gene vitellogenin, and the androgen-responsive gene spiggin were assessed using quantitative RT-PCR in the livers of male and posterior kidneys of female stickleback, respectively. Hepatic 7-ethoxyresorufin-O-deethylase (EROD) activity was assessed in both sexes. Effluent extracts were examined for estrogenic and androgenic bioactivity using receptor binding bioassays, and were screened for pulp and paper related extractives and steroidal androgens using GC-MS. This effluent up-regulated spiggin mRNA in the kidney of female stickleback at 10% and 100% (v/v) effluent at 21 d, but not at 7 d of exposure. This change at the mRNA expression of the gene was associated with an increase in cell height in kidney proximal tubule epithelial cells at 100% effluent after both 7 and 21 d. Liver vitellogenin mRNA in male stickleback was not induced at either 7 or 21 d. EROD was induced at 10 and 100% after 21 d of exposure in both sexes, but not after 7 d of exposure. Despite evidence of exposure to androgens, there was no reduction in steroidogenic capacity at any effluent dilution. Effluent extracts were capable of eliciting the displacement of androgens and estrogens from receptors, but androgenic potency was 4-fold greater. A screen of more than 30 androgenic androstane steroids showed no detections. Hence, the androgenic constituents in this particular effluent remain unknown.

Bacterial pathogens have evolved two major strategies to colonise the intestinal epithelium. Adherent microorganisms bind to the apical pole of the intestinal epithelium, whereas invasive microorganisms disrupt and invade the epithelium. Recognition of the genetic bases of bacterial pathogenicity and analysis of the molecular cross talks established between pathogens and their mammalian target cells have illuminated this diversity of interactions. We have compared the strategies of enteroinvasive pathogens, with emphasis on bacterial species such as Shigella, Yersinia, and Salmonella, that represent paradigms of interaction. Cross talks leading to alteration of the epithelial cell actin cytoskeleton appear as a recurrent theme during entry and dissemination into epithelial cells. Other cross talks alter the trafficking of cellular vesicles and induce changes in the intracellular compartment in which they reside, thus creating niches favourable to bacterial survival and growth. Finally, a variety of strategies also exist to deal with other components of the epithelial barrier, such as macrophages. Pro-phagocytic, anti-phagocytic, and pro-apoptotic processes appear to be of particular importance.

A number of phenanthrolines and benzoquinolines were examined for their ability to activate epithelial chloride secretion by measuring short circuit current (SCC) using the mouse colon epithelium. 1,10 phenanthroline stimulated electrogenic chloride secretion with an EC50 of 612±10 μ and a Hill slope of 4.9±0.3. A similar pharmacology was demonstrated by both 1,7 and 4,7 phenanthrolines, 7,8 benzoquinoline and phenanthridine. Evidence that the increase in SCC caused by 1,10 phenanthroline was due to chloride secretion is based upon (a) inhibition of the current by furosemide, (b) failure of cystic fibrosis (CF) colons to respond and (c) an associated net flux of 36Cl−. 1,10 Phenanthroline affected neither the generation of cyclic AMP or the concentration of intracellular Ca2+ in colonic epithelial cells. 1,10 phenanthroline affected the chloride conductance of the apical membrane, as shown by an increase in chloride current in ‘apical membrane only' preparations in the presence of an apical to basolateral chloride gradient. The increase in chloride current was inhibited by 5-nitro-2-(3-phenylpropylamino) benzoic acid (NPPB) and was not present in CF colons. Additionally, 1,10 phenanthroline activated basolateral K+ channels, both Ca2+- and cyclic AMP-sensitive channels, as shown by inhibitor studies with charybdotoxin (ChTX) and XE991, and after the apical membrane was permeabilized with nystatin. The phenanthrolines and benzoquinolines described here, with dual actions affecting CFTR and basolateral K+ channels, may constitute useful lead compounds for adjunct therapy in CF. Keywords: CFTR, cystic fibrosis, chloride channel openers, phenanthrolines, 7,8 benzoquinoline, phenanthridine, potassium channels, cyclic AMP, intracellular Ca2+ Introduction Increased interest in agents affecting epithelial chloride channels arose after it was shown that the cystic fibrosis transmembrane conductance regulator (CFTR) is also a chloride channel in many epithelial membranes (Anderson et al., 1991). Not only have blockers of CFTR been described but considerable interest has also focused on CFTR chloride channel openers (Schultz et al., 1999). The most common mutation in cystic fibrosis (CF) is ΔF508, in which phenylalanine is absent from position 508. This mutant protein (ΔF508CFTR) can function as a chloride channel (Drumm et al., 1991), however only a minute fraction of this protein is transported to the apical domain of epithelial cells (Ward & Kopito, 1994). Instead the quality control mechanisms of the cell cause the mutant protein to be degraded in the proteasome. Therefore attempts are being made to increase the delivery of ΔF508 CFTR to the membrane (Brodsky, 2001; Fischer et al., 2001; Fuller & Cuthbert, 2000). Thus agents which open CFTR channels are of interest, particularly so if they also activate ΔF508 CFTR. The present classes of CFTR openers are as follows; phosphatase inhibitors (Becq et al., 1998), xanthines, particularly CPX (Guay-Broder et al., 1995), benzimidazolones such as EBIO (Devor et al., 1996; Cuthbert et al., 1999), flavones, for example genistein (Illek et al., 1995), benzoxazolones like chlorzoxazone (Singh et al., 2000) and finally the MPB compounds described by Becq et al. (1999). The purpose of this study was to further investigate the actions of 1,10 phenanthroline on chloride secretion in the mouse colon following the observations made by Duszyk et al. (1999) that 1,10 phenanthroline, a powerful inhibitor of matrix metalloproteases (MMPs) activated a chloride conductance, measured as whole cell current, in human airway epithelial cells, Calu-3. No effect was found in airway cells lacking CFTR, namely A549 cells. Furthermore, an antibody to MMP2 caused a similar effect in Calu-3 cells as did 1,10 phenanthroline, suggesting metalloproteinase inhibition was responsible. A further aim was to explore structure activity relations in the phenanthroline isomers and to ask if both heterocyclic nitrogens in the tricyclic aromatic molecules were necessary for the effect. From various experimental approaches we conclude that the phenanthrolines and related structures constitute a new class of agents that stimulate chloride secretion.

The purpose of this study is to investigate the recovery rate in paper-patch and fat-plug myringoplasty in rats.Small and large perforations were inflicted on the tympanic membranes of 60 rats. Fat-plug and paper-patch myringoplasties were performed to different groups and another group was left for spontaneous healing.We found that for small perforations, the recovery rate was 94.7% in fat-plug myringoplasty, 94.4% in paper-patch myringoplasty, and 66.6% in control group. The recovery rates in large perforations were 52.9%, 56.2%, and 26.6%, respectively. On the other hand, in larger perforations, paper-patch or fat-plug myringoplasty have not been found effective.We believe that due to significant operational advantages, fat-plug or paper-patch myringoplasty can be suggested for the reconstruction of small and dry perforations of the tympanic membrane.

Ocular signs and serum vitamin A concentrations are commonly used for the diagnosis ofvitamin A deficiency. Recently, impression cytology was suggested as a method for early detection of this condition. A clinical trial was conducted to compare the results of impression cytology with the other two indicators of vitamin A status. A total of 246 children aged 1-10 y were investigated. About 25% ofthe children with normal eyes showed abnormal cytology. Their mean vitamin A concentrations were significantly lower compared with those with normal cytology, which suggests that abnormal cytology reflects subclinical deficiency. After treatment with vitamin A, most of the children who had abnormal cytology at baseline examination showed improvement. These observations suggest that abnormal cytology is spe- cific to vitamin A deficiency. Impression cytology is a relatively simple technique and can be used for assessing the magnitude of the problem, especially in communities where the preva- lence ofclinical disease is low. Birch pollen and dog epithelium allergen extracts were studied by ultracentrifugation in linear sucrose gradients. The various sucrose-gradient fractions were then tested for allergenic activity by the paper disc radioallergosorbent test, using sera from allergen sensitive patients.

Central corneal alkali burn injuries were induced in rabbits by applying five NaOH concentrations on uniformly soaked 7 mm filter paper discs. Clinical parameters were evaluated daily by microscopic examination and photography, and corneal myeloperoxidase levels were measured periodically. Satisfactory alkali burn models of corneal inflammation, vascularization and ulceration were developed by manipulating the alkali concentration.
