Introduction: Decreased physical activity is one of the main causes of the obesity epidemic. In the process of obesity, the secretion of some hepato-adipokines increases and causes metabolic disorders. Exercise plays an important role in improving the complications of obesity by regulating the levels of selected hepato-adipokine. The purpose of this study was to examine the effects of three types of resistance training methods on selected hepato-adipokine and lipid profiles in men with obesity. Materials and Methods: The sample of the study included 44 sedentary men with obesity divided into 4 groups of traditional resistance training (TRT), circuit resistance training (CRT), and interval resistance training (IRT) as well as one control group. Fetuin-A, Fetuin-B, high-density lipoproteins (HDL), low-density lipoproteins (LDL), total cholesterol (TC), and triglyceride (TG) were measured using the ELISA method. Results: The results of the mixed model ANOVA analysis showed a significant interaction between the type of training used and time at the levels of Fetuin-A (F (1, 40) = 94273.16, P = 0.001, ES = 1.00) and Fetuin-B (F (1, 40) = 49697.67, P = 0.001, ES = 0.99. In addition, within-group comparisons showed that lipid profile improved in TRT and CRT groups compared to the pretest (P = 0.001), while in the IRT group this improvement was not significant (P > 0.05). Conclusion: The result of the present study shows that three models of resistance training reduced the select hepato-adipokines level in comparison to the control group. However, IRT and CRT had the greatest effect on reducing Fetuin-A and Fetuin-B, respectively. [ABSTRACT FROM AUTHOR]

Context: Gait biomechanics are linked to biochemical changes that contribute to the development of posttraumatic knee osteoarthritis in individuals with anterior cruciate ligament reconstruction (ACLR). It remains unknown if modifying peak loading during gait using real-time biofeedback will result in acute biochemical changes related to cartilage metabolism. Objective: To determine if acutely manipulating peak vertical ground reaction force (vGRF) during gait influences acute changes in serum cartilage oligomeric matrix protein concentration (sCOMP) among individuals with ACLR. Design: Crossover study. Patients or Other Participants: Thirty individuals with unilateral ACLR participated (70% female, age = 20.43 ± 2.91 years old, body mass index = 24.42 ± 4.25, months post-ACLR = 47.83 ± 26.97). Additionally, we identified a subgroup of participants who demonstrated an increase in sCOMP after the control or natural loading condition (sCOMPCHANGE > 0 ng/mL, n = 22, 70% female, age = 20.32 ± 3.00 years old, body mass index = 24.73 ± 4.33, months post-ACLR = 47.27 ± 29.32). Main Outcome Measure(s): Serum was collected both prior to and immediately after each condition to determine SCOMPCHANGE. Intervention(s): All participants attended 4 sessions that involved 20 minutes of walking on a force-measuring treadmill consisting of a control condition (natural loading) followed by random ordering of 3 loading conditions with real-time biofeedback: (1) symmetric vGRF between limbs, (2) a 5% increase in vGRF (high loading) and (3) a 5% decrease in vGRF (low loading). A general linear mixed model was used to determine differences in sCOMPchange between altered loading conditions and the control group in the entire cohort and the subgroup. Results: The sCOMPCHANGE was not different across loading conditions for the entire cohort (F3.29 = 1.34, P = .282). Within the subgroup, sCOMPCHANGE was less during high loading (1.95 ± 24.22 ng/mL, t21 = -3.53, P = .005) and symmetric loading (9.93 ± 21.45 ng/mL, t21 =-2.86, P= .025) compared with the control condition (25.79 ± 21.40 ng/mL). Conclusions: Increasing peak vGRF during gait decreased sCOMP in individuals with ACLR who naturally demonstrated an increase in sCOMP after 20 minutes of walking. [ABSTRACT FROM AUTHOR]

Context: Military service members commonly sustain lower extremity stress fractures (SFx). How SFx risk factors influence bone metabolism is unknown. Understanding how SFx risk factors influence bone metabolism may help to optimize risk-mitigation strategies. Objective: To determine how SFx risk factors influence bone metabolism. Design: Cross-sectional study. Setting: Military service academy. Patients or other participants: Forty-five men (agepre = 18.56 ± 1.39 years, heightpre = 176.95 ± 7.29 cm, masspre = 77.20 ± 9.40 kg; body mass indexpre = 24.68 ± 2.87) who completed Cadet Basic Training (CBT). Individuals with neurologic or metabolic disorders were excluded. Intervention(s): We assessed SFx risk factors (independent variables) with (1) the Landing Error Scoring System (LESS), (2) self-reported injury and physical activity questionnaires, and (3) physical fitness tests. We assessed bone biomarkers (dependent variables; procollagen type I amino-terminal propeptide [PINP] and cross-linked collagen telopeptide [CTx-1]) via serum. Main outcome measure(s): A markerless motion-capture system was used to analyze trunk and lower extremity biomechanics via the LESS. Serum samples were collected post-CBT; enzyme-linked immunosorbent assays determined PINP and CTx-1 concentrations, and PINP : CTx-1 ratios were calculated. Linear regression models demonstrated associations between SFx risk factors and PINP and CTx-1 concentrations and PINP : CTx-1 ratio. Biomarker concentration mean differences with 95% confidence intervals were calculated. Significance was set a priori using α ≤ .10 for simple and α ≤ .05 for multiple regression analyses. Results: The multiple regression models incorporating LESS and SFx risk factor data predicted the PINP concentration (R2 = 0.47, P = .02) and PINP : CTx-1 ratio (R2 = 0.66, P = .01). The PINP concentration was increased by foot internal rotation, trunk flexion, CBT injury, sit-up score, and pre- to post-CBT mass changes. The CTx-1 concentration was increased by heel-to-toe landing and post-CBT mass. The PINP : CTx-1 ratio was increased by foot internal rotation, lower extremity sagittal-plane displacement (inversely), CBT injury, sit-up score, and pre- to post-CBT mass changes. Conclusions: Stress fracture risk factors accounted for 66% of the PINP : CTx-1 ratio variability, a potential surrogate for bone health. Objective: To determine whether or not self-selected walking speed associates with serum biomarkers of cartilage (collagen and proteoglycan) breakdown in anterior cruciate ligament reconstructed (ACLR) individuals.Methods: Twenty individuals with a history of a primary unilateral ACLR participated in this cross-sectional study. Resting blood was collected from each participant prior to completing 5 walking gait trials at a self-selected comfortable speed. Walking speed was evaluated in a 3-dimensional motion capture laboratory and determined from the velocity of the pelvic center of mass. Sera were assessed for collagen type II cleavage product (C2C) and proteoglycan (aggrecan) concentrations using commercially available specific enzyme-linked immunosorbent assays. Pearson's product-moment (r) and Spearman's (ρ) correlations were used to evaluate associations between walking speed and biomarkers of cartilage breakdown metabolism. Partial correlations were used to determine whether covariates influenced associations between walking speed and biomarkers of cartilage breakdown.Results: ACLR individuals with a slower walking speed demonstrated higher concentrations of serum C2C (r = -0.52, P = 0.02), while there was no significant association between walking speed and aggrecan concentrations (ρ = -0.29, P = 0.31). After accounting for the variance associated with stance phase duration, ACLR individuals with a slower walking speed still demonstrated greater serum C2C concentrations (partial r = -0.53, P = 0.02).Conclusion: ACLR individuals who habitually walk slower may experience a greater degree of collagen breakdown, suggesting that walking speed may be a future useful clinical indicator for identifying individuals with higher levels of cartilage breakdown and preradiographic osteoarthritic joint changes. Introduction: An in-depth understanding of the healthy cartilage response to activities of daily living is needed to better understand the complex relationship between cartilage health and loading. The purpose was to assess the role of loading on the acute serum cartilage oligomeric matrix protein (COMP) response in recreationally active individuals. Methods: Forty individuals without previous lower extremity injury participated in this repeated-measures study in which each participant completed all conditions during independent data collection sessions separated by at least 1 wk. An antecubital blood draw was performed before and after walking, drop-landing, and control (i.e., sitting) conditions. Commercially available enzyme-linked immunosorbent assays measured COMP concentration. The acute COMP response was quantified as the percent change of COMP concentration from before to after each condition. A one-way, repeated-measures ANOVA compared the acute COMP response between conditions. Post hoc Pearson product–moment correlation and chi-square analysis determined the relationship between the walking and drop-landing acute COMP response within individuals. Results: Acute COMP response was greater after walking (+4.2, P = 0.008) and drop landing (+4.6%, P = 0.002) compared with control (−2.3%), but did not differ between the walking and drop-landing conditions (P = 0.596). The magnitudes of the acute COMP response during walking and drop landing were correlated (r = 0.56, P < 0.001). However, the direction (i.e., either increase or decrease) of COMP was not the same after the walking and drop-landing conditions (χ1 = 0.870, P = 0.351). Conclusions: Walking and drop landing produced a greater acute COMP response when compared with a control condition in healthy individuals, but the acute COMP response was similar between the two physical activity conditions, although the conditions differed in magnitude and frequency of loading.
