Conselho Nacional de Desenvolvimento Científico e Tecnológico Os inibidores de protease (IP) produzidos pelos vegetais têm um amplo espectro de proteases- alvo. No entanto, as pragas agrícolas encontraram formas de debelar os efeitos negativos dos IP de suas plantas hospedeiras. Para compreender os mecanismos moleculares e fisiológicos envolvidos na interação inseto-planta, a lagarta da soja (Anticarsia gemmatalis) e a soja (Glycine max) foram utilizadas como animal e planta modelo. Por meio de diferentes abordagens bioquímicas e biológicas, este trabalho apresentou as hidrolases intestinais da A. gemmatalis com elevada cobertura mediante à metodologia de 1DE-LC/MS proposta. Essa estratégia, apresentada de forma inédita, possibilitou a identificação de 98 hidrolases não redundantes, compreendendo isoformas importantes no mecanismo adaptativo do inseto a IP de plantas. As variações ocorridas nesse proteoma foram avaliadas expondo a lagarta a IP. Os resultados mostraram modulações fisiológicas que foram capazes de driblar os efeitos negativos dos inibidores. Houve aumento de isoformas responsáveis por atividades proteolíticas bem como aumento de proteínas relacionadas a processos de estresse. A presença de IP na dieta causou respostas nas atividades enzimáticas que revelaram uma melhor adaptação da lagarta ao IP natural da soja quando comparado a inibidores sintéticos. As análises de docking molecular mostraram a termodinâmica e os sítios de ligação envolvidos no complexo enzima-inibidor (EI), revelando características importantes para um IP eficiente. Usando esse conhecimento como base, um tripeptídeo (GORE2) foi racionalmente desenhado para atuar como inibidor de tripsinas-like de A. gemmatalis. O transcriptoma do intestino da lagarta mostrou que a presença do GORE2 e do SKTI, inibidor natural da soja, foram responsáveis por 1474 genes diferencialmente expressos, revelando importantes enzimas envolvidas na resposta do inseto. O GORE2 proposto mostrou-se mais eficiente que o SKTI ao provocar danos celulares no epitélio intestinal. Os danos mais intensos e extensos provocados pelo inibidor peptídico foram acompanhados por estresse oxidativo e pela baixa eficiência na resposta do inseto para se desintoxicar do estresse oxidativo por meio de enzimas antioxidantes. Esses resultados permitiram um melhor entendimento sobre o arsenal bioquímico e molecular do herbívoro para propor um IP potente, abrindo caminho para novas abordagens no desenvolvimento de defensivos agrícolas ambientalmente corretos e eficientes no manejo de pragas. However, agricultural pests have overcome the negative effects of PI on their host plants. To understand the molecular and physiological mechanisms involved in the insect-plant interaction, the soybean caterpillar (Anticarsia gemmatalis) and soybean (Glycine max) were used as an animal and plant model. Through different biochemical and biological approaches, this work presented the intestinal hydrolases of A. gemmatalis with high coverage through the proposed 1DE-LC/MS method. This strategy, presented unprecedentedly, allowed the identification of 98 non-redundant hydrolases, comprising important isoforms in the adaptive mechanism of the insect to plant PI. The variations that occurred in this proteome were evaluated by challenging the caterpillar with PI. The results showed physiological modulations that aimed to circumvent the negative effects of inhibitors. There was an increase in isoforms responsible for proteolytic activities, as well as an increase in proteins related to stress processes. The presence of PI in the diet caused responses in the enzymatic activities that revealed a better adaptation of the caterpillar to the natural PI of soybean when compared to synthetic inhibitors. Molecular docking analyzes showed the thermodynamics and binding sites involved in the enzyme-inhibitor (EI) complex, shedding light on important features of an efficient IP. Using this knowledge as a basis, a tripeptide (GORE2) was rationally designed to act as a trypsin-like inhibitor of A. gemmatalis. The transcriptome of the caterpillar intestine showed that the GORE2 and SKTI, a natural soybean inhibitor, handled 1474 differentially expressed genes, revealing important enzymes involved in the insect’s response. The proposed GORE2 proved to be more efficient than SKTI, in causing cellular damage to the intestinal epithelium. The most intense and extensive damage caused by the peptide inhibitor was accompanied by oxidative stress and the low efficiency in the insect’s response to detoxify itself from oxidative stress through antioxidant enzymes. These results allowed a better understanding of the herbivore’s biochemical and molecular arsenal to propose a potent IP, opening the way for new approaches in the development of environmentally friendly and efficient pesticides in pest management. Coordenação de Aperfeiçoamento de Pessoal de Nível Superior Atualmente é consensual que as mudanças climáticas estão promovendo aumentos nas temperaturas globais, variabilidade na precipitação e surtos de pragas de insetos mais frequentes. Em adição, existem indicativos que a susceptibilidade das plantas ao ataque de insetos pragas pode ser aumentada em condições de estresses abióticos, tais como seca, e que estas respostas são variadas em função dos genótipos. Assim, a compreensão dos mecanismos moleculares de tolerância aos estresses bióticos e abióticos é crítica para o melhoramento genético das plantas cultivadas e sustentabilidade da produtividade. Estudos tem indicado que algumas respostas fisiológicas e cascatas regulatórias são compartilhadas quando desencadeadas por diferentes sinais de estresse. Desta forma, examinamos as cascatas regulatórias e as vias metabólicas de genótipos de soja tolerantes à seca (EMBRAPA 48), resistentes a infestação de A. gemmatalis (IAC17), comparando com um genótipo sensível à seca/ataque de insetos (BR16), e todos foram submetidos a ambos os sinais de estresse (abióticos e bióticos). Plantas em condições de seca foram menos suscetíveis ao ataque de insetos, promovendo menor sobrevivência da lagarta. As reduções de sobrevivência não foram dependentes dos fenótipos de tolerância à seca ou resistência a insetos, embora mais pronunciadas para IAC17. Além disso, perfis de metabólitos, expressão gênica e ensaios enzimáticos nos levam a concluir que apenas o sinal de seca não foi suficiente para explicar totalmente as reduções de sobrevivência. As atividades de inibição da protease e a expressão gênica se correlacionaram com os níveis de ABA, indicando que a sinalização de JA foi potencializada pelo ABA para aumentar a produção dos metabólitos dissuasores. Portanto, o aumento dos níveis de ABA durante o tratamento da seca pode estar agindo sinergicamente para aumentar a resposta de JA, não afetando os primeiros estágios das vias LOX e JA. Assim, “hub (s)” molecular (is) regulatório (s) integrando com múltiplos sinais podem ser alvos para engenharia genética de plantas com múltiplas tolerâncias a estresses ambientais. Currently is predicted that climate changes are promoting increases in global temperatures, variability in precipitation and more frequent insect pest outbreaks. Thus, understanding the molecular mechanisms for tolerance of both biotic and abiotic stresses in critical for development of improved genotypes and sustainability of the productivity. As, some physiological responses and regulatory cascades have been shared when triggered by different stress signals, we examine the regulatory cascades and metabolic pathways of drought-tolerant (EMBRAPA48), resistant to A. gemmatalis infestation (IAC17) genotypes, comparing with one sensitive to drought/insect attack (BR16) under both stress signals. Plants under drought were less susceptive to insect attack promoting lower caterpillar survival. Survival reductions were not dependent of the drought-tolerance or insect-resistance phenotypes, despite more pronounced for IAC17. Furthermore, metabolites profiles, gene expression and enzymatic assays lead us to concluded that only the drought signal not was enough to explain the survival reductions. Protease inhibition activities and gene expression correlated with ABA levels, indicating that JA-signaling were potentialized by ABA for enhance the production of the deterrent metabolites. Increased of ABA levels during drought treatment may be acting synergistically to increase JA response, did not affecting the first stages of LOX and JA pathways. Thus, regulatory molecular hub(s) integrating signals may be targets to genetic engineering of plants with multiple tolerances to environmental stresses. Therefore, in current studies, we examined whether signs of drought could affect caterpillar survival for other genotypes characterized as drought tolerant and resistant to insect attack. they can assist in the development of better management strategies and new varieties. O Brasil possui o maior rebanho comercial do mundo, e para garantir baixos custos na produção, usa as pastagens como a principal fonte de alimentação do rebanho. Em decorrência de ataques de insetos- praga, o nível de produtividade das pastagens pode sofrer declínio tanto na quantidade quanto na qualidade de produção. A seleção de cultivares de forrageiras resistentes, é uma alternativa de controle promissora, uma vez que o controle químico das cigarrinhas é economicamente inviável. São escassos os trabalhos que relatam a bioquímica da interação entre M. spectabilis com diferentes forrageiras. O objetivo, na primeira parte do trabalho, foi identificar as proteínas, metabólitos e hormônios presentes nas glândulas salivares quando a M. spectabilis se alimenta de plantas de capim- elefante (Cenchrus purpureus), cv. Roxo Botucatu e cv. Pioneiro, Brachiaria brizantha cv. Marandú e Brachiaria decumbens cv. Basilisk. Na segunda parte, objetivamos elucidar as possíveis respostas metabólicas nas forrageiras Brachiaria brizantha, Brachiaria decumbens, capim-elefante (Cenchrus purpureaus) cv. Roxo-de-Botucatu e cv. Pioneiro quando infestadas por M. spectabilis. Através das análises de proteômica foi possível identificar proteínas envolvidas em processos como controle da musculatura, síntese e metabolismo protéico, proteção contra estresse biótico/abiótico e termoregulação. As análises fitohormonais das glândulas salivares indicam uma variação nos níveis hormonais em função do tipo de forrageira usada na alimentação. Foram detectados nos componentes das glândulas salivares a presença de efetores, tais como ácidos graxos de cadeia longa que podem induzir ou suprimir as respostas de defesa das plantas. A presença dos efetores afetam as respostas de defesa da planta para permitir o melhor desempenho do inseto na infestação das plantas. De frente a esses resultados, nos é permitido propor que alguns efetores presentes nas glândulas salivares de cigarrinha das pastagens possam ser responsáveis por suprimir a resposta de defesa da planta, tornando-se uma estratégia bem sucedida do inseto na colonização do hospedeiro. The spittlebugs, Mahanarva spectabilis (Distant, 1909) is the main forage pest in Tropical America, causing severe damage to pastures and compromising the production of animal products. Brazil has the largest commercial herd in the world, and to ensure low production costs, it uses pasture as its primary feed source. As a result of insect pest attacks, pasture productivity may decline in both quantity and quality of production. Selection of resistant forage cultivars is a promising control alternative, since chemical control of leafhoppers is economically unfeasible. There are few studies reporting the biochemistry of the interaction between M. spectabilis with different forages. The objective, in the first part of the work, was to identify the proteins, metabolites and hormones present in the salivary glands when M. spectabilis feeds on elephant grass (Cenchrus purpureus), cv. Purple Botucatu and cv. Pioneer, Brachiaria brizantha cv. Marandú and Brachiaria decumbens cv. Basilisk In the second part, we aim to elucidate the possible metabolic responses in the forages Brachiaria brizantha, Brachiaria decumbens, elephant grass (Cenchrus purpureaus) cv. Botucatu purple and cv. Pioneer when infested by M. spectabilis. Through proteomics analyzes it was possible to identify proteins involved in processes such as muscle control, protein synthesis and metabolism, protection against biotic / abiotic stress and thermoregulation. Phytohormonal analyzes of salivary glands indicate a variation in hormone levels depending on the type of forage used in the diet. The presence of effectors such as long chain fatty acids that can induce or suppress plant defense responses has been detected in salivary gland components. The presence of effectors affects plant defense responses to allow the best insect performance in plant infestation. Given these results, we are allowed to propose that some effectors present in the leafhopper salivary glands of the pastures may be responsible for suppressing the defense response of the plant, becoming a successful insect strategy in host colonization. O controle-químico de cigarrinha-das-pastagens é economicamente inviável. Sendo assim, a seleção de cultivares forrageiras gramíneas resistentes é uma alternativa de controle muito importante. Estudos relatando a bioquímica da interação entre M. spectabilis e diferentes forrageiras gramíneas são raros. O objetivo foi avaliar o impacto do ataque de M. spectabilis na resposta defensiva de plantas de capim-elefante (Pennisetum purpureum), cv. Roxo Botucatu (RX) e cv. Pioneiro (PI), Brachiaria brizantha cv. Marandú (MA) e Brachiaria decumbens cv. Basilisk (DE). O impacto da alimentação em diferentes cultivares de forrageiras nas proteases digestivas do inseto também foi avaliado. O experimento foi realizado com os quatro genótipos distintos, infestados ou não por M. spectabilis. A atividade de lipoxigenases (LOX), inibidores de proteases e as concentrações de fitohormônios foram avaliadas nas folhas das forrageiras. A atividade de proteases totais e tripsina-like foram avaliadas na região do intestino de M. spectabilis. Os genótipos RX, MA e DE tiveram aumento na atividade de LOX após injúrias por M. spectabilis. A ativação da via da LOX está relacionada com o aumento nos níveis do hormônio jasmonato. O aumento na concentração de ácido abscisico aconteceu nos quatro cultivares. O ácido abscisico pode atuar sinergicamente com o jasmonato na transcrição de genes de defesa. Os cultivares MA e DE tiveram maiores níveis basais de ácido salicílico, mas apenas o cultivares DE foi capaz de aumentar os níveis do hormônio em resposta à herbivoria. A atividade de proteases totais foi maior em M. spectabilis que se alimentaram de DE, indicando secreção de enzimas. Há varialibidade na indução de LOX dentro da espécie Pennisetum purpureum. Todos os cultivares analisados apresentaram aumento nas concentrações de ABA e JA, quando atacados por M. spetabilis, indicando ações de defesa em comum. A indução nas concentrações de SA apenas em DE, pode indicar aumento nos níveis de radicais livres, que causam danos aos herbívoros. O aumento na atividade de proteases totais de M. spectabilis alimentadas com DE indica mecanismos de secreção de proteases para compensar àquelas inibidas, que não estão relacionado com enzimas tripsina-like. Mahanarva spectabilis are spittlebugs that cause several damages in tropical pastures, compromising cattle production. The chemical control of spittlebugs is economically unfeasible. Therefore, the selection of resistant forage grasses cultivars is an important control alternative. Studies reporting the biochemistry of the interaction between M. spectabilis and different forage grasses are rare. Thus, the goal was to assess the impact of M. spectabilis injury on the defensive response of elephant grass (Pennisetum purpureum), cv. Roxo Botucatu (RX) and cv. Pioneiro (PI), Brachiaria brizantha cv. Marandú (MA) and Brachiaria decumbens (DE). The impact of feeding on different cultivars of forages in the digestive proteases of the insect was also assessed. The experiment was carried out with four distinct cultivars, infested or not by M. spectabilis. The lipoxygenases activity, protease inhibitors and concentrations of phytohormones were evaluated in leaves of forage grasses. Total proteases and trypsin-like activities were evaluated in the gut region of M. spectabilis. RX, MA and DE genotypes showed increased LOX activity after injury by M. spectabilis. The LOX pathway activation is related to the increase in levels of the hormone jasmonate. The increase in abscisic acid concentration after injury occurred in the four genotypes. Abscisic acid acts synergistically with jasmonate in the transcription of defense genes. MA and DE cultivars had higher basal salicylic acid levels, but only DE was able to increase concentration in response to herbivory. There is variability in the induction of LOX within the species Pennisetum purpureum. All cultivares showed increased concentrations of ABA and JA when attacked by M. spectabilis, indicating common defense actions. Total proteases activity was higher in M. spectabilis fed with DE. The induction of SA concentration only in DE, may indicate an increase in free radical levels, which cause damage to herbivores. The increased activity of total proteases of M. spectabilis fed with DE indicates protease secretion mechanisms to compensate for those inhibited, which are not related to trypsin- like proteases.

Conselho Nacional de Desenvolvimento Científico e Tecnológico A água media as interações planta-praga afetando a produtividade e o sucesso reprodutivo dos organismos. As interações entre insetos e suas plantas hospedeiras são afetadas pela especialização na alimentação das espécies herbívoras, geralistas ou especialistas, assim como a disponibilidade de água. O objetivo deste trabalho foi comparar as respostas defensivas do tomateiro pela sinalização dos hormônios vegetais e compostos de defesa pela via das lipoxigenases, quando presentes estresses tanto biótico como a herbivoria por ácaros e insetos, assim como abiótico de água, especificamente o deficit hídrico, de forma simultânea, no campo aberto. Propõe-se responder: Como esses fatores podem afetar a produtividade do tomateiro e a susceptibilidade a insetos que broqueiam os frutos?; O deficit hídrico afeta as defesas induzidas no tomateiro, a expressão de hormônios vegetais e os compostos de defesa pela via das lipoxigenases e polifenol oxidases?; e se em combinação com a herbivoria as expressões de hormônios vegetais e compostos de defesa, podem aumentar ou diminuir?; também se o deficit hídrico afeta o desenvolvimento e sucesso reprodutivo de insetos pragas como a traça do tomateiro, Tuta absoluta (Lepidoptera: Gelechiidae). Para isto foi desenhada uma parcela experimental de tomate, onde um setor da parcela foi aplicado uma irrigação idónea, sendo do 100 % de demanda de água da cultura. No outro setor da parcela foi aplicado uma irrigação do 50% de demanda da cultura. Um desenho experimental de três fatores foi aplicado para os analises estatísticos. Cada setor da parcela composto de cinco linhas (blocos) com plantas nas que foram aplicados 10 tratamentos ao acaso, sendo cada planta uma unidade amostral. Os tratamentos consistiram de plantas sem infestar e infestadas com ácaros herbívoros especialistas, Tetranychus evansi e geralistas T. urticae (Acari: Tetranychidae) além de lagartas especialistas, T. absoluta, de forma isolada e em combinações das três espécies. Também, outras seis plantas de cada setor foram selecionadas para determinar taxas de oviposição e mortalidade de T. absoluta. Uma folha de cada planta foi coberta com uma sacola de organza e as mariposas liberadas dentro. Das folhas do tomateiro foram determinadas, as concentrações de hormônios vegetais e aminoácidos, também a atividade de lipoxigenases, polifenol oxidases e a concentração de inibidores de proteinase. Os frutos maduros foram classificados quanto ao tamanho, peso e danos pela broca-pequena do tomateiro Neoleucinodes elegantalis (Lepidoptera: Crambidae). Também, foram avaliadas as taxas de oviposição, e mortalidade da traça do tomateiro T. absoluta em plantas que tiveram irrigação idônea e plantas em deficit hídrico. Os perfis dos compostos bioquímicos mostram correlações positivas e negativas, indicando várias vias de sinalização e respostas defensivas mediadas pelos hormônios. O rendimento do tomateiro foi afetado pelo deficit hídrico, mas não pela herbivoria, podendo tornar as culturas mais suscetíveis a N. elegantalis. Interações entre as cascatas hormonais foram mediadas pelo deficit hídrico e a herbivoria, induzindo diferentes expressões dos compostos de defensa. O ácaro rajado T. urticae teve uma influência importante na ativação das lipoxigenases, polifenol oxidases e indução de inibidores de proteases do tomateiro, de forma tanto negativa como positiva. O ácaro vermelho T. evansi e a traça do tomateiro T. absoluta, marcaram principalmente, respostas cruzadas dos hormônios quando compartilharam plantas hospedeiras com outros herbívoros. O tomateiro sob deficit hídrico foi menos adequado para o desempenho de T. absoluta, devido as defesas mediadas pelo ácido jasmônico e compostos induzidos por herbivoria com alta produção de inibidores de proteases, coincidindo com a “hipótese da planta vigorosa”. As sinalizações cruzadas dos hormônios vegetais no tomateiro, mostraram grandes diferenças entre as condições hídricas e a herbivoria por T. absoluta. Conclui-se que, o tipo de mecanismo de alimentação e especialização do herbívoro foi um fator importante na variação destas interações. Assim como a quantidade de espécies que podem-se alimentar numa planta simultaneamente. A vias das lipoxigenases foi ativada com a produção de compostos de defesa, e o deficit hídrico influi na direção da atividade das lipoxigenases e polifenol oxidases. Water mediates plant-pest interactions affecting the productivity and reproductive success of organisms. Interactions between insects and their host plants are affected by the feeding mode of herbivorous species, generalists or specialists, as well as the availability of water. This work aimed to compare the defensive tomato responses by the phytohormones signalling and defense compounds of the lipoxygenases pathway, under biotic stress as herbivory by mites and insects, as well as water abiotic stress, specifically water deficit, simultaneously in the open field. It was proposed to answer: How can these factors affect tomato productivity and susceptibility to fruit-borer insects?; The water deficit affects the tomato induced defenses, as the phytohormones expression and the defense compounds through the lipoxygenases and polyphenol oxidases pathways?; and if in combination with herbivory can the expressions of plant hormones and defense compounds increase or decrease?; also if the water deficit affects the development and reproductive success of insect pests such as the tomato moth, Tuta absoluta (Lepidoptera: Gelechiidae). For this, a tomato experimental plot was designed, where a sector of the plot was applied a suitable irrigation, of 100% the water demand of the culture. In the other sector of the plot, an irrigation of 50% of crop demand was applied. An experimental design of three factors was applied for the statistical analyses. Each plot sector consisted of five lines (blocks) with plants on which 10 treatments were applied randomly, each plant being a sample unit. The treatments consisted of uninfested plants and infested with herbivorous specialist mites, Tetranychus evansi and generalists T. urticae (Acari: Tetranychidae) in addition to specialist caterpillars, T. absoluta, isolated and in combinations of the three species. In addition, six other plants from each sector were selected to determine T. absoluta oviposition rates and mortality. One leaf of each plant was covered with an organza bag and the moths released inside. From tomato leaves, the phytohormones, amino acids proteinase inhibitors concentrations, the lipoxygenases and polyphenoloxidases activities were determined. Mature fruits were classified according to size, weight and damage by tomato borer Neoleucinodes elegantalis (Lepidoptera: Crambidae). Also, the oviposition rates and mortality of the tomato leaf-miner Tuta absoluta (Lepidoptera: Gelechiidae) were evaluated, in plants that had adequate irrigation and plants in water deficit. Profile patterns of biochemical compounds show several positive and negative correlations, indicating several hormone crosstalk-signalling pathways mediating defensive responses. Tomato yield were affected by water deficit, but not by herbivory, and could turn crops more susceptible to N. elegantalis. The interactions of the hormonal cascades were mediated by water deficit and herbivory, resulted in different expressions of defense compounds. The two-spotted spider mite T. urticae had an important influence on the activation of lipoxygenases and polyphenol oxidases and proteinase inhibitors induction, both negative and positive. The red spider mite T. evansi (Acari: Tetranychidae) and tomato leafminer T. absoluta, mediated phytohormones cross- responses, mainly, when sharing host plants with other herbivores. The tomato under water deficit was less suitable for T. absoluta performance, due to the defenses mediated by the jasmonic acid and herbivory induced compounds with high production of protease inhibitors, coinciding with the "plant vigour hypothesis". Cross-talk phytohormones pathways in tomato plants, showed large differences between water conditions and herbivory by T. absoluta. It was concluded, that the type of feeding mechanism and specialization of the herbivore was an important factor in the variation of these interactions. As well as the quantity of species that can feed on a plant simultaneously. A lipoxygenases pathway was activated with the production of defense compounds, and the water deficit influences the lipoxygenase and polyphenol oxidases activity.

Coordenação de Aperfeiçoamento de Pessoal de Nível Superior A grande extensão de área com cultura de soja intensificoua ocorrência de diversas pragas, dentre elas, o percevejo-marrom, Euschistus heros (Fabricius, 1798) (Pentatomidae). Os danos desta praga vêm se tornando cada vez mais expressivos, devido ao seu aumento populacional, manejo inadequado e resistência a inseticidas, o que resulta em perda de produtividade. As plantas apresentam mecanismos de defesa contra os insetos e, por sua vez, estes tentam burlar estas defesas. Estudos sobre a interação inseto/planta têm sido realizados analisando a via de defesa da planta e/ou enzimas digestivas das pragas. Entretanto, não há relatos sobre enzimas digestivas presentes no intestino e nas glândulas salivares do percevejo- marrom após interação com a soja, ao longo do tempo. Assim, este estudo teve como objetivo avaliar o perfil das enzimas presentes no intestino médio e nas glândulas salivares após serem alimentados com as cultivares IAC 24 (resistente) e IAC Foscarin 31 (susceptível). Sendo este o início dos estudos em relação ao controle desta praga com base em aspectos bioquímicos. Plantas, no estágio R5, foram infestadas com machos do percevejo. As avaliações foram realizadas em diferentes tempos de alimentação (0, 24, 48, 72 e 96 horas). Após cada tempo, retirou-se os intestinos e as glândulas salivares. As atividades das enzimas proteolíticas, amilase e lipase, foram realizadas de acordo com protocolos específicos. Tanto no intestino como nas glândulas salivares não foi detectada atividade de cisteíno proteases. No intestino médio, a atividade de proteases totais apresentou diferença para tempo, cultivar e interação cultivar x tempo. A amilase foi diferente somente para o tempo e lipase diferiu no tempo e na interação. Nas glândulas salivares, a atividade de proteases totais apenas diferiu em relação às cultivares analisadas. A amilase mostrou significância tanto para o tempo quanto para a interação e lipase foi significativa para as três fontes de variações avaliadas. Assim, observou- se o comportamento das enzimas responsivo a alimentação com os distintos cultivares. Estudos mais aprofundados são necessários para entender o porquê das oscilações registradas nas condições analisadas. Great extension of soybean crop intensified the occurrence of several pests, including the brown stink bug Euschistus heros, which is considered one of the main soybean pests. Pest damage has become increasingly significant due to its population increase, inadequate management and resistance to insecticides, resulting in soybean productivity losses. Plants have defense mechanisms against insects, and these latter in turn try to cheat these defenses. Plant-insect interaction studies have been performed by analyzing plants defense-signaling pathways and/or pest’s digestive enzymes. However, digestive enzymes present in the gut and salivary glands of the stink bugafter his interaction with soybeans over the time has not been reported. Thus, the aim of this study was to assess the enzymatic profile present in the midgut and salivary glands of the stink bug after feeding with IAC 24 (resistant) and IAC Foscarin 31 (susceptible) soybean crop. This being the beginning of the studies in relation to the control of this pest based on biochemical aspects. Plants at the R5 stage were infected with male stink bug. Assessments were performed at different feeding times (0, 24, 48, 72 and 96 hours). After each time, midgut and salivary glands were removed. The proteolytic enzyme assays, amylase and lipase were performed according to specific protocols. Cysteine proteases activities were not detected in the midgut and salivary glands. In the midgut, total proteases activity presented difference for time, cultivar and cultivar x time interaction. Amylase activity was different only in time, meanwhile lipase differed in time and interaction. In the salivary glands, total proteases activity differed only in relation to the cultivars analyzed. Amylase activity showed significance for time and in interaction, while lipase was significant for the three sources of variations assessed. Thus, the behavior of the enzymes responsive to feeding with the different cultivars was observed. Further studies are needed to understand why the oscillations recorded under the conditions analyzed.

CAPES - Coordenação de Aperfeiçoamento de Pessoal de Nível Superior A lagarta-da-soja, Anticarsia gemmatalis, é considerada uma das principais pragas da cultura da soja. Os danos causados pelo ataque deste inseto associado à relevância econômica do cultivo da soja para o Brasil e para o mundo fomentam a busca por alternativas no controle deste inseto. Estratégias de controle de insetos-pragas baseadas no uso de inibidores de proteases têm sido estudadas, o conhecimento das enzimas digestivas e metabolismo do inseto tem se mostrado fundamental. Neste contexto, este trabalho teve como objetivo identificar proteínas com abundância diferencial no intestino de A. gemmatalis em resposta a inibidor de protease, visando obter dados que poderão contribuir para esclarecer o mecanismo adaptativo de A. gemmatalis em resposta a inibidores de proteases na interação planta-inseto. Foram dissecados intestinos de lagartas alimentadas com os inibidores de proteases berenil e o Kunitz, nas horas 6 h, 12 h, 24 h e 48 h e as proteínas extraídas por maceração. Os spots com abundância diferencial foram digeridos e os peptídeos analisados no espectrômetro de massa MALDI/TOF-TOF. Os espectros (MS/MS) foram pesquisados em bancos de dados usando a ferramenta MASCOT e as proteínas identificadas foram validadas usando o software Scaffold. Foram 44 spots de proteínas identificados, nos tratamentos com os inibidores berenil e Kunitz, nas diferentes horas, que correspondeu a 33 proteínas distintas. As proteínas identificadas pertenciam a diferentes categorias funcionais envolvidas no metabolismo energético, do carbono, de aminoácido, de carboidratos, de lipídeos, no estresse oxidativo e no turnover proteico e chaperonas moleculares. Algumas isoformas da proteína ATPase apresentaram menor abundância em relação ao controle, em ambos tratamentos. Enquanto a maioria das proteínas de choque térmico 70 apresentaram maior abundância que o controle. As proteínas piruvato desidrogenase, fosfoglicerato mutase e a ATP sintase aumentaram abundância e as proteínas aldeído desidrogenase e carbonil redutase diminuíram abundância no tratamento com berenil. No tratamento com Kunitz as proteínas arginina quinase e a choque térmico Hsp 90 aumentaram a abundância enquanto as proteínas enoil-CoA hidratase, ATP sintase e a aspartato aminotransferase diminuíram a abundância. A presença dos inibidores berenil e Kunitz na lagarta A. gemmatalis alterou o perfil de proteínas no intestino em ambos tratamentos. The soybean caterpillar, Anticarsia gemmatalis, is considered one of the main pests of soybean crop. The damage caused by the attack of this insect associated to the economic relevance of soybean cultivation to Brazil and to the world foments the search for alternatives in the control of this insect. Strategies for control of insect pests based on the use of protease inhibitors have been studied, knowledge of digestive enzymes and insect metabolism has been shown to be fundamental. In this context, this work aimed to identify proteins with differential abundance in the midgut of A. gemmatalis in response to protease inhibitor, aiming to obtain data that could contribute to scientifically elucidate the adaptive mechanism of A. gemmatalis in response to protease inhibitors in the interaction plant and insect. Midgut of caterpillars fed with a berenyl protease inhibitor and Kunitz type soybean inhibitor were dissected at 6h, 12 h, 24 h and 48 h and the proteins extracted. Protein extracts were submitted to two-dimensional gel electrophoresis. The differential abundance spots were digested and the peptides analyzed in the MALDI / TOF-TOF mass spectrometer. Fragmentation spectra (MS / MS) were searched in databases using the MASCOT tool, and the identified proteins were validated using the Scaffold software. A total of 44 protein spots were identified in the berenyl and Kunitz treatments at different times, which corresponded to 33 distinct proteins. The proteins identified belong to different functional categories involved in the energy, amino acids, carbohydrates, lipids metabolism, oxidative stress, protein turnover and chaperones. Some ATPase protein isoforms presented lower abundance than control in both treatments. While most heat shock 70 proteins were more abundant than control. Pyruvate dehydrogenase, phosphoglycerate mutase and ATP synthase proteins increased abundance, and aldehyde dehydrogenase and carbonyl reductase proteins decreased abundance in berenyl treatment. In Kunitz treatment arginine kinase and heat shock Hsp 90 proteins increased abundance, while enoyl-CoA hydratase, ATP synthase and aspartate aminotransferase proteins decreased abundance. Berenyl and Kunitz inhibitors in caterpillar A. gemmatalis altered the protein profile in the midgut in both treatments.
