The saphenous vein is the most commonly used bypass graft in patients with coronary artery disease. During routine coronary artery bypass, grafting the vascular damage inflicted on the vein is likely to stimulate the release of endothelin-1, a potent endothelium-derived vasoconstrictor that also possesses cell proliferation and inflammatory properties, conditions associated with vein graft failure. In both in vitro and in vivo studies, endothelin receptor antagonists reduce neointimal thickening. The mechanisms underlying these observations are multifactorial and include an effect on cell proliferation and cell/tissue damage. Much of the data supporting the beneficial action of endothelin-1 receptor antagonism at reducing intimal thickening and occlusion in experimental vein grafts were published over 20 years ago. The theme of the recent ET-16 conference in Kobe was "Visiting Old and Learning New". This short review article provides an overview of studies showing the potential of endothelin receptor antagonists to offer an adjuvant therapeutic approach for reducing saphenous vein graft failure and poses the question why this important area of research has not been translated from bench to bedside given the potential benefit for coronary artery bypass patients.

The purpose of this qualitative morphological study was the immunocytochemical and ultrastructural comparison of perivascular nerves of the basilar artery (BA) of young (6-month-old) and adult (12-month-old) capybaras - adult capybaras showed regression of the internal carotid artery (ICA). The study focused on immunolabeling for the vasoactive peptide endothelin-1 (ET-1) and endothelin A receptor (ETA) as well as for the synapse marker synaptophysin (SYP). In the BA of young capybaras, immunoreactivity for ET-1, ETA receptor and SYP was detected in perivascular nerve varicosities and/or intervaricosities. Immunoreactivity for ET-1 and ETA receptor was also displayed by some Schwann cells, which accompanied perivascular nerves. In addition to the presence of the above-described perivascular nerve characteristics, the BA of adult animals also revealed structurally altered perivascular nerves, where axon profiles were irregular in shape with dense axoplasm, while the cytoplasm of Schwann cells was vacuolated and contained myelin-like figures. These structurally altered perivascular nerves displayed immunoreactivity for ET-1, ETA receptor and SYP. These results show that the ET-1 system is present in some of the BA perivascular nerves and it is likely that this system is affected during animal maturation when ICA regression takes place. The role of ET-1 in cerebrovascular nerves is still unclear but its involvement in neural (sensory) control of cerebral blood flow and nerve function is possible., (Copyright © 2013 S. Since the discovery of endothelin in vascular endothelial cells and its pivotal role in vascular physiology (Yanagisawa and colleagues), a number of studies have focused on the localisation of this vasoconstrictor peptide in human and animal vascular tissue, largely in endothelial cells. Various vascular beds have been the subject of research in normal and pathophysiological conditions, for example in neonates, during ageing, pregnancy, hypertension, diabetes, heart failure, experimental metastases and neurological disorders. These studies have revealed the presence of endothelin in the blood vessel wall, suggesting the involvement of this peptide in vascular physiology in health and disease. This chapter reviews studies on the distribution of endothelin-1 (ET-1) and its receptors (ET(A) and ET(B)) in vascular tissue with emphasis on their ultrastructural localisation.

Little is known about cerebral vasculature of capybara, which seems may serve as a natural model of studying changes in cerebral circulation due to internal carotid artery atrophy at animal sexual maturation. This is the first study of the light- and electron-immunocytochemical localisation of endothelin-1 (ET-1) and ETA and ETB endothelin receptors in the basilar artery of capybaras (6 to 12-month-old females and males) using an ExtrAvidin detection method. All animals examined showed similar patterns of immunoreactivity. Immunoreactivity for ET-1 was detected in the endothelium and adventitial fibroblasts, whilst immunoreactivity for ETA and ETB receptors was present in the endothelium, vascular smooth muscle, perivascular nerves and fibroblasts. In endothelial cells immunoreactivity to ET-1 was pronounced in the cytoplasm or on the granular endoplasmic reticulum. Similar patterns of immunolabelling were observed for ETA and ETB receptors, though cytoplasmic location of clusters of immunoprecipitate seems dominant. These results suggest that the endothelin system is present throughout the wall of the basilar artery of capybara.

The saphenous vein (SV) is the most commonly used conduit for coronary artery bypass surgery. However, using traditional techniques, the occlusion rate for the SV is high, with over 50% of grafts failing within 10 years. In conventional coronary artery bypass surgery the SV is exposed to considerable damage during preparation for grafting. Recently, an increased graft patency has been described using a 'no-touch' technique, whereby the vein is prepared with minimal vascular trauma. There is evidence that the success of this form of coronary artery bypass surgery is a result, at least in part, of the retention of tissue-derived nitric oxide. We have examined the effects of conventional SV harvesting on vessel morphology, cell proliferation, endothelin-1 and its receptors. Considerable damage was observed in veins prepared using conventional surgery compared to 'no-touch' veins. The vessel wall exhibited evidence of surgical trauma, with regions of denudation of the luminal endothelium caused by distension. Endothelin-1 and endothelin-A receptors were present at subintimal regions of conventional SV segments where proliferating cells were identified. Endothelial endothelin-B receptors were also revealed that were absent at areas of distension-induced damage to the endothelium. These results suggest that endothelin-1 plays a role in vein graft failure, predominantly via the endothelin-A receptor.

In this study, we show the changes in the wall of the middle cerebral artery of a subject who suffered multiple system atrophy with autonomic failure. An electron-immunocytochemical approach was employed to reveal the presence of endothelin-1. Our results demonstrate the presence of immunoreactive endothelin-1 in the endothelial cells of the intima, vascular smooth muscle cells and macrophages of the media and neointima, and perivascular nerves/axons varicosities at the adventitial-medial border of the artery. It is concluded that endothelin-1 may, therefore, play a number of roles within diseased cerebral artery. The finding of endothelin-1-positive varicosities of autonomic innervation to this artery suggests an influence of neural endothelin on vascular smooth muscle in multiple system atrophy with autonomic failure. However, the presence of features such as neointima formation, wall irregularities and foam cells suggest the coexistence of atherosclerosis.

Novel data is presented from this electron-immunocytochemical study that demonstrates endothelin-containing cerebrovascular nerves in the human middle cerebral artery.

The co-localisation of neuronal nitric oxide synthase and endothelin-1 was studied in the rat supraoptic nucleus at the electron microscopy level. Double pre-embedding immunocytochemistry was performed using ExtrAvidin-horseradish peroxidase and immunogold-silver techniques. Immunoreactivities to neuronal nitric oxide synthase and endothelin-1 were co-localised in sub-populations of endocrine neurones (cell bodies) and dendrites. Double-labelled axon terminals making asymmetrical synapses on unlabelled dendrites were also observed. The findings are discussed in terms of the possible role and significance of nitric oxide and endothlin-1 in the hypothalamo-neurohypophysial system.
