In the light of the fast growing several applications of selenium nanoparticles (SeNPs) in different industrial and agricultural sectors, this paper was conducted to explore the suitability of endophytic fungi as nano-factories for SeNPs. Thus, 75 fungal isolates were recovered from plant tissues and tested for their efficacy to biosynthesize SeNPs. Four promising strains were found able to synthesis SeNPs with different characteristics and identified. These strains were Aspergillus quadrilineatus isolated from the twigs of Ricinus communis, Aspergillus ochraceus isolated from the leaves of Ricinus communis, Aspergillus terreus isolated from the twigs of Azadirachta indica, and Fusarium equiseti isolated from the twigs of Hibiscus rose-sinensis. The synthesized SeNPs were characterized by several techniques viz., UV–Vis, X-ray diffraction, Dynamic light scattering analyses, High resolution transmission electron microscopy, and Fourier transform infrared spectroscopy, to study their crystalline structure, particle sized distribution, and morphology. Furthermore, the in vitro antimicrobial and antioxidant activities were evaluated. SeNPs synthesized by the four strains showed potent antifungal and antibacterial potentials against different human and phyto- pathogens. Moreover, SeNPs synthesized by the respective strains showed promising antioxidant power with IC50 values of 198.32, 151.23, 100.31, and 91.52 µg mL− 1. To the best of our knowledge, this is the first study on the use of endophytic fungi for SeNPs' biosynthesis. The presented research recommends the use of endophytic fungi as facile one-pot production bio-factories of SeNPs with promising characteristics. In order to explore the antibiotics development potential of the endophytic fungus Apiospora malaysiana from Thamnolia subuliformis, the antimicrobial spectrum of Apiospora malaysiana against common pathogen was determined after comparing the effects of six commonly used methods for determination of in vitro antimicrobial activity. The effects of storage time, temperature, ultraviolet irradiation time, and pH on the stability of antimicrobial activities of Apiospora malaysiana in vitro were further investigated by using the most sensitive Staphylococcus aureus as the indicator bacterium. The results showed that there was no significant difference in the antimicrobial activity of Apiospora malaysiana in vitro by three methods (plate punching method, filter paper method, and Oxford cup method) (P>0.05). However, the results of the three fungal inhabition methods showed that the mycelial growth rate method was optimal (inhibition rate of 100%), the effect of the plate confrontation method was much lower than the former (inhibition rate of only 24.0%), and the filter paper diffusion method had no inhibiting effect. The results of in vitro antimicrobial test showed that Apiospora malaysiana had strong inhibiting effects on a variety of Gram-positive bacteria and pathogenic fungi, among which Staphylococcus aureus was the most sensitive (the diameter of the inhibition zone was 18.3 mm, highly sensitive). In addition, Apiospora malaysiana also had strong antimicrobial activity against pathogenic fungi such as drug-resistant Candida albicans (highly sensitive), Botrytis cinerea, and Rhizoctonia solani (inhibition rate of 100%). The search for new bioactive compounds with innovative modes of action and chemistry are desperately needed to tackle the increased emergence of drug-resistant microbes. With this view, this paper was conducted for the isolation, identification, and biological evaluation of fungal endophytes of eleven different plant species. A total of 69 endophytic strains were isolated and tested for the presence of bioactive metabolites with antifungal, antibacterial, anticancer, and antioxidant properties in their extracts. Upon screening, two promising strains were found to have all the before-mentioned activities. These strains were Aspergillus sydowii isolated from the bark of Ricinus communis and Aspergillus flavus isolated from the twigs of Psidium guajava. Major compounds present in extracts of the two strains were identified by GC-Mass analyses. Several well-known bioactive compounds as well as unreported ones were identified in the fungal extracts of the two strains. Furthermore, gamma irradiation (at 1000 Gy) of the fungal cultures resulted in improved bioactivities of extracts from the two strains. These findings recommend the two fungal strains as sources of antimicrobial, anticancer, and antioxidant compounds which may aid in the development of novel drugs. The presented research also explains the high-value of fungal endophytes as untapped sources of bioactive metabolites. Coffee is grown in more than 80 countries as a cash crop and consumed worldwide as a beverage and food additive. It is susceptible to fungal infection during growth, processing and storage. Fungal infections, in particular, can seriously affect the quality of coffee and threaten human health. The data for this comprehensive review were collected from the United States Department of Agriculture, Agricultural Research Service (USDA ARS) website and published papers. This review lists the fungal species reported on coffee based on taxonomy, life mode, host, affected plant part and region. Five major fungal diseases and mycotoxin-producing species (post-harvest diseases of coffee) are also discussed. Furthermore, we address why coffee yield and quality are affected by fungi and propose methods to control fungal infections to increase coffee yield and improve quality. The present paper deals with the endophytic fungi of Litsea glutinosa, a medicinal plant highly valued in Ayurvedic system. A total of 21 fungal endophytes were extracted from different parts of the plant. The following eight species viz, Aspergillus oryzae, A. terreus, Curvularia sp, Fusarium oxysporum, Gliocladium solani, Penicillium citrinum, Trichoderma viride and Verticillium dahliae were selected for testing the production of secondary metabolites including hydrolytic enzymes. All the eight endophytic fungal species produced amylase, cellulase, protease and lipase. Except for Curvularia species, others did not elaborate laccase. All the test fungi produced antibacterial and antifungal metabolites; however, the crude extracts exhibited poor antimicrobial activity. The present endophytic fungi also produced IAA, ammonia and phosphate solubilizing substances. They were also capable to produce IAA with or without adding the tryptophan. An affect was also made to assess the influence of crude extracts on the root growth of maize seedlings. These studies revealed the enhancement effect of extracts on root growth. [Objective] The present paper aimed to screen, isolate and identify the high-yield lignocellulose-degrading endophytic fungi from Taxillus chinensis (DC.) seeds, and provide technical support for the acquisition of lignocellulose-degrading enzymes with strong capacity and T. chinensis artificial cultivation. [Method] The endophytic fungi of T. chinensis seeds were isolated by conventional tissue separation method. Wheat bran was used as substrate for lignin biomass. The activities of laccase, lignin peroxidase, manganese peroxidase and cellulose degrading enzyme were determined by ABTS method, veratrol method, phenol red method and cellulase (CL) activity detection kit. The strains with strong lignocellulosic degradation ability were screened and their classification status was determined according to morphological characteristics and phylogenetic tree analysis. (Result) Four high-yielding lignocellulosic degrading strains (15H3, 2013, 106 and 10y11) were selected from the 57 strains of endophytic fungi from T. chinensis seeds. Among them, the lignin peroxidase activity of strain 15H3 and the manganese peroxidase activity of strain 2013 reached the peak when cultured in the crude enzyme solution of endophytic fungi from T. chinensis seeds for 11 days, they were 7.14 and 11.69 U/mL, respectively. At 9 days, the laccase activity of strain 106, cellulase activity of strain 2013, the manganese peroxidase activity of strain 10y11 were the highest, which were 57.98,0.32 and 8.56 U/mL, respectively. By morphological observation and molecular biology identification, strain 15H3 and 2013 could be identified as Pestalotiopsis arceuthobii and Coniella pseudogranati, respectively. Strains 106 and 10y11 were identified as Fusarium incarnatum. [Conclusion] The endophytic fungi P. arceuthobii, C. pseudogranati and F. incarnatum with high production of lignocellulosic degrading enzymes were screened and identified from T. chinensis seeds, which could provide theoretical basis for the study of the degradation mechanism of T. chinensis invasion into host cell wall and provide data support for the breeding of T. chinensis. Rosa roxburghii Tratt. is widely applied in food, cosmetics, and traditional medicine, and has been demonstrated to possess diverse bioactivities. Plant endophytic fungi are important microbial resources with great potential for application in many fields. They not only establish mutualistic symbiosis with host plants but also produce a variety of bioactive compounds. Therefore, in the present study, endophytic fungi were isolated from R. roxburghii , the diversity and antimicrobial activities were evaluated. As a result, 242 strains of endophytic Sordariomycetes were successfully isolated. Multigene phylogenetic analyses showed that these isolates included eight orders, 19 families, 33 genera. The dominant genera were Diaporthe (31.4%), Fusarium (14.4%), Chaetomium (7.9%), Dactylonectria (7.0%), Graphium (4.5%), Colletotrichum (4.1%), and Clonostachys (4.1%). For different tissues of R. roxburghii , alpha diversity analysis revealed that the diversity of fungal communities decreased in the order of root, fruit, stem, flower, leaf, and seed, and Clonostachys and Dactylonectria exhibited obvious tissue specificity. Meanwhile, functional annotation of 33 genera indicated that some fungi have multitrophic lifestyles combining endophytic, pathogenic, and saprophytic behavior. Additionally, antimicrobial activities of endophytic Sordariomycetes against Lasiodiplodia theobromae , Botryosphaeria dothidea , Colletotrichum capsici , Pyricularia oryzae , Rhizoctonia solani , Fusarium oxysporum , Pseudomonas syringae , Pantoea agglomerans , Staphylococcus aureus , Bacillus subtilis , Escherichia coli , and Pseudomonas aeruginosa were screened. Dual culture test assays showed that there were 40 different endophytic species with strong inhibition of at least one or moderate inhibition of two or more against the 12 tested strains. The results from the filter paper diffusion method suggested that extracellular metabolites may be more advantageous than intracellular metabolites in the development of antimicrobial agents. Eleven isolates with good activities were screened. In particular, Hypomontagnella monticulosa HGUP194009 and Nigrospora sphaerica HGUP191020 have shown promise in both broad-spectrum and intensity. Finally, some fungi that commonly cause disease have been observed to have beneficial biological activities as endophytic fungi. In conclusion, this study showed the species composition, alpha diversity, and lifestyle diversity of endophytic Sordariomycetes from R. roxburghii and demonstrated these isolates are potential sources for exploring antimicrobial agents. In the past few decades, the control of pests and diseases of cultivated plants using natural and biological measures has drawn increasing attention in the quest to reduce the level of dependence on chemical products for agricultural production. The use of living organisms, predators, parasitoids, and microorganisms, such as viruses, bacteria, and fungi, has proven to be a viable and sustainable pest management technique. Among the aforementioned, fungi, most importantly the insect-pathogenic species, have been in use for more than 150years. These include the most popular strains belonging to the genera Beauveria , Metarhizium , Isaria , Hirsutella , and Lecanicillium. Their application is usually through an inundative approach, which inherently involves exposure of the fungal spores to unfavorable humidity, temperature, and solar radiation conditions. These abiotic factors reduce the persistence and efficacy of these insect-pathogenic fungi. Despite these limitations, over 170 strains have been formulated as mycopesticides and are available for commercial use. In the last few decades, numerous studies have suggested that these species of entomopathogenic fungi (EPF) offer far more benefits and have broader ecological functions than hitherto presumed. For instance, aside from their roles as insect killers, it has been well established that they also colonize various host plants and, hence, provide other benefits including plant pathogen antagonism and plant growth promotion and serve as sources of novel bioactive compounds and secondary metabolites, etc. In this light, the potential of EPF as alternatives or perhaps as supplements to chemical pesticides in plant protection is discussed in this review. The paper highlights the numerous benefits associated with endophytic fungal entomopathogen and host plant associations, the mechanisms involved in mediating plant defense against pests and pathogens, and the general limitations to the use of EPF in plant protection. A deeper understanding of these plant host-fungus-insect relationships could help unveil the hidden potentials of fungal endophytes, which would consequently increase the level of acceptance and adoption by users as an integral part of pest management programs and as a suitable alternative to chemical inputs toward sustainable crop production. Ageratina adenophora is an invasive plant of global importance and has a broad distribution in the Western Himalayas. Endophtytic fungus Coniochaeta sp.F-8 was found in Ageratina adenophora. In this paper, we aim to investigate the antioxidative activity and cytotoxicity of the metabolites from Coniochaeta sp.F-8. Consequently, two compounds Phomoxanthone A and Penialidin A were isolated from the endophytic fungus of Ageratina adenophora for the first time. The structures of compounds were identified by IR, NMR and ESI-MS methods. Moreover, both of those compounds showed considerable antioxidative activity in vitro and resulted cytotoxicity in mouse embryo fibroblasts cell line Balb/c3T3. Endophytes are barely untapped as vital sources in the medicine. They are microorganisms which mostly exist in plants. As they are exploited, it is accepted that endophytes can produce active metabolites that possess same function as their hosts such as taxol, podophyllotoxin, hypericin, and azadirachtin. These metabolites have been promising potential usefulness in safety and human health concerns. We are supposed to adopt measures to raise production for the low yield of metabolites. This paper summarizes the latest advances in various bioprocess optimization strategies. These techniques can overcome the limitations associated with rare pharmaceutical metabolite-producing endophytic fungi. These strategies include strain improvement, genome shuffling, medium optimization, fermentation conditions optimization, addition of specific factor, addition of solid sorbent, and co-culturing. Beauveria bassiana is an entomopathogenic fungus widely used as a biopesticide for insect control; it has also been shown to exist as an endophyte, promoting plant growth in many instances. This study highlights an alternative potential of the fungus; in the production of an industrially important biocatalyst, xylanase. In this regard, Beauveria bassiana SAN01 xylanase was purified to homogeneity and subsequently characterized. The purified xylanase was found to have a specific activity of 324.2 U.mg-1 and an estimated molecular mass of ~37 kDa. In addition, it demonstrated optimal activity at pH 6.0 and 45 °C while obeying Michaelis-Menton kinetics towards beechwood xylan with apparent Km, Vmax and kcat of 1.98 mg.mL-1, 6.65 μM.min-1 and 0.62 s-1 respectively. The enzyme activity was strongly inhibited by Ag2+ and Fe3+ while it was significantly enhanced by Co2+ and Mg2+. Furthermore, the xylanase was shown to effectively deink wastepaper at an optimal rate of 106.72% through its enzymatic disassociation of the fiber-ink bonds as demonstrated by scanning electron microscopy and infrared spectroscopy. Objective: The present investigation primarily focusses on enhancement of resveratrol production by endophytic production from the endophytic fungus, Arcopilus aureus via one variable at a time approach (OVAT) followed by statistical approach using response surface methodology (RSM). The paper also highlights the characterization of fungal resveratrol using spectroscopic techniques. Further the tyrosinase inhibitory property was also explored in this communication for its possible use as a cosmeceutical ingredient. Results: Optimization of physicochemical and nutritional parameters using OVAT approach exhibited 1.23-fold enhancement in production of resveratrol when compared to initial yield, 89.1 ± 0.08 µg/mL. Further RSM resulted in 1.49-fold enhancement in resveratrol production i.e. 133.53 µg/ml. Further, 25 mg of fungal resveratrol in pure form was obtained from the spent broth of Arcopilus aureus by column chromatography using a mobile phase comprising of MeOH: DCM in a ratio of 1.75:98.25. Further its purity on TLC was checked using 5% MeOH: DCM as mobile phase. Symmetrical peak with Rt of 3.36 min using a C18 reverse phase column confirmed the homogeneity of the purified fungal resveratrol with standard resveratrol and further corroborated by 1H-NMR, 13C-NMR and HR-MS analysis. Fungal resveratrol exhibited a good tyrosinase inhibition with an IC50 of 2.654 ± 0.432 µg/mL as compared to Kojic acid (1.329 ± 0.333). Conclusions: The present study has provided sufficient lead that process optimization techniques can complement each other for optimized production of bioactive compounds by microorganisms apart from strain improvement techniques which are generally adopted in the industry. The enhancement of resveratrol production by Arcopilus aureus by process optimization further opens up avenues for strain improvement for commercial resveratrol production through fermentation for nutraceutical and cosmeceutical applications. Model of 2′-deoxyguanosine involvement in plant immune response. Endophytic fungus-derived 2-dG directly activates ET signaling, EIT and PTI. Moreover, pathogen invasion induces the expression of the plant 2-dG biosynthesis gene VEN4 and promotes 2-dG accumulation in plant leaves. However, 2-dG-mediated plant resistance against Pst DC3000 is conferred in an ET-, NPR1 -, PRR/coreceptor- and P2K1 -dependent manner. [Display omitted] • We identified for the first time that nucleoside 2′-deoxyguanosine from endophytic fungal metabolites can effectively stimulate classical immune responses in plants. • We demonstrated that 2-dG-induced plant resistance depends on salicylic acid (SA) receptor NPR1 (but not SA), ethylene (ET) signaling, pattern-recognition receptors/coreceptors (PRRs/coreceptors) and the ATP receptor P2K1. • We provide the first demonstration that VEN4 is 2-dG biosynthesis gene and involved in pathogen-induced 2-dG accumulation, VEN4 also plays an essential role for plant immunity. Beneficial microorganisms play essential roles in plant growth and induced systemic resistance (ISR) by releasing signaling molecules. Our previous study obtained the crude extract from beneficial endophyte Paecilomyces variotii , termed ZNC (ZhiNengCong), which significantly enhanced plant resistance to pathogen even at 100 ng/ml. However, the immunoreactive components of ZNC remain unclear. Here, we further identified one of the immunoreactive components of ZNC is a nucleoside 2′-deoxyguanosine (2-dG). This paper intends to reveal the molecular mechanism of microbial-derived 2′-deoxyguanosine (2-dG) in activating plant immunity, and the role of plant-derived 2-dG in plant immunity. The components of ZNC were separated using a high-performance liquid chromatography (HPLC), and 2-dG is identified using a HPLC–mass spectrometry system (LC-MS). Transcriptome analysis and genetic experiments were used to reveal the immune signaling pathway dependent on 2-dG activation of plant immunity. This study identified 2′-deoxyguanosine (2-dG) as one of the immunoreactive components from ZNC. And 2-dG significantly enhanced plant pathogen resistance even at 10 ng/ml (37.42 nM). Furthermore, 2-dG-induced resistance depends on NPR1, pattern-recognition receptors/coreceptors, ATP receptor P2K1 (DORN1), ethylene signaling but not salicylic acid accumulation. In addition, we identified Arabidopsis VENOSA4 (VEN4) was involved in 2-dG biosynthesis and could convert dGTP to 2-dG, and vne4 mutant plants were more susceptible to pathogens. In summary, microbial-derived 2-dG may act as a novel immune signaling molecule involved in plant-microorganism interactions, and VEN4 is 2-dG biosynthesis gene and plays a key role in plant immunity. Endophytes provide a plethora of potential substances that are used in modern medicine. There is an emergence of interest to discover potential compounds from endophytes due to their abundant availability from renewable sources, and low cost relative to chemically synthesized compounds. Studies have shown that Morus alba plant has potential antioxidant and antibacterial activities. Hence, in the present study we deals with the bioprospecting of endophytes of Morus alba. Stem bark of M. alba was collected from different regions of Gujarat. Total 13 bacteria and 14 fungal endophytes were isolated from the collected samples. Among 27 endophytic isolates, the extract of four isolates showed significant antibacterial activity with a diameter zone ranged from 8.0±0.0 to 22.33±1.262 mm with considerable minimum inhibitory concentration (MIC) values. These four extracts also showed significant inhibition of reactive oxygen species in 1,1-diphenyl-2-picrylhydrazyl (DPPH) assay. Triterpenoids, flavonoids, and phenols were present in these extracts which might be responsible for these bioactivities. Results of this study concluded that these endophytes isolated from the stem bark of Morus alba L. have an important bioactivities in vitro. Many problems caused by antibiotic resistance encourage the exploration of new antibiotic compounds from natural materials, especially from endophytic fungal cultures. This study aims to evaluate different culture media that can support endophytic fungi to produce bioactive compounds that have antibacterial and antioxidant activity. The method used in this study was to ferment endophytic fungi from gall rust sengon in different media, namely PDB, PDB plus sengon wood powder, and PDB plus yeast, for 9 days, followed by extraction using ethyl acetate solvent and testing of antioxidant and antibacterial activity. The DPPH radical was used to measure antioxidant activity, and Bacillus subtillis was used as a representative for Gram-positive and Gram-negative For testing of antimicrobials, Escherichia coli and Pseudomonas aeruginosa were used. The experiment showed that the addition of yeast and sengon wood to PDB medium increased the growth and production of bioactive compounds extracted from fungal cultures. A fungal endophyte extract in ethyl acetate from all media used produced antioxidant compounds with high activity. Antibacterial activity was obtained from the extract of fungal cultures grown on PDB plus Sengon wood floor on day 6 for P. aeruginosa and E. coli, and day 3 for B. subtillis. 【Objective】The present paper aimed to analyze the effects of endophytic fungus Y118 on the growth promotion and quality of its host, Zanthoxylum nitidum（Rox.） DC,so as to provide a reference for improving comprehensive benefits of Z.nitidum.【Method】Z.nitidum plants were cultured with endophytic fungus Y118 solid fermentation medium.The growth indexes（fresh weight, plant height, stem base width, stem middle width and fresh and dry weight of roots, stems and leaves） of Z.nitidum plants were measured at different growth stages.The contents of nitrine chloride and chelidonine in roots of Z.nitidum were determined by chromatographic method.【Result】After 3 months of culture, the fresh weight, plant height, stem base width and stem middle width of the fungus Y118 treatment group were 34.76 g, 34.47 cm, 6.54 mm and 4.25 mm, respectively, which were extremely significantly higher than those of the control group（P＜0.01,the same as below）.The fresh weight and dry weight of roots, stems and leaves were significantly increased（P＜0.05,the same as below） or extremely significantly increased.After 6 months of culture, the fresh weight and plant height of Z.nitidum were 68.33 g and 47.85 cm, respectively, which were extremely significantly higher than control group. The fresh weight and dry weight of roots, stems and leaves were extremely significantly increased. After 9 months of culture, the fresh weight, plant height and stem base width of Z.nitidum treated with fungus Y118 were 73.80 g, 49.22 cm and 9.79 mm, respectively, which were extremely significantly higher than those of the control group. The fresh weight and dry weight of roots, stems and leaves were extremely significantly increased. After 12 months of culture, the fresh weight of Z.nitidum in Y118 treatment group was 78.71 g, which was extremely significantly higher than control group, and the fresh and dry weight of root, stem and leaf were significantly increased or extremely significant. After 9 and 12 months of co-culture, Z.nitidum alkali content in fungus Y118 treatment group was 0.783 and 0.904 mg/g, respectively, which increased by 42.11% and 44.18% compared with the control group. The content of chelidonine at 9 and 12 months of co-culture were 3.704 and 4.201 mg/g, respectively, which increased by 40.73% and 26.57% compared with the control.【Conclusion】Inoculation of endophytic fungus Y118 could significantly or extremely significantly increase the fresh weight, plant height, stem base width and stem middle width of Z.nitidum seedlings, which was beneficial to promote the growth and development of Z.nitidum seedlings, and could improve the fresh and dry weight and quality of roots, stems and leaves. Endophytic fungi isolated from medicinal plants have high diversity and the secondary metabolites produced have biological activity. Endophytic fungi isolated from medicinal plants have been the subject of many studies on their diversity and biological activity, one of which is jambu mawar (Syzygium jambos (L.) Alston). S. jambos has long been used as a traditional medicine to treat infections caused by pathogenic bacteria in many parts of the world, including South Sumatra. This study purposed to explore the diversity of endophytic fungi isolated from the branches of S. jambos, the antibacterial activity of endophytic fungi extracts, the determination of the structure of the compounds, and the activity of the active compounds of the selected endophytic fungi. Observations of the morphological characteristics of endophytic fungi were macroscopically and microscopically. The endophytic fungal extracts were then tested for antimicrobial activity against test bacteria Staphylococcus aureus, Bacillus subtilis, Salmonella typhi, and Escherichia coli using the Kirby-Bauer paper disk diffusion method. Isolation of pure compounds using a gravity chromatography column, the determination of the structure of the compound is determined based on 1H-NMR, 13C-NMR, HMQC, and HMBC spectroscopy. Four endophytic fungi, code SJC1-4, were isolated from a branch of S. jambos. The results of macroscopic and microscopic morphological characterization showed three genera of Botryosphaeria, Trichothecium, and Aspergillus. The endophytic fungal isolate SJC1 exhibited strong activity against Gram-positive bacteria and moderate activity against Gram-negative bacteria, while SJC2-4 showed moderate activity against the four bacteria. Molecular identification of SJC1 revealed that it was Botryosphaeria mamane. Isolation of SJC1's pure compound yielded compound 1, which was identified as 5-acetyl-6-hydroxy-3-methyl-2H-pyran-2-one. Plant seeds are not merely reproductive organs, they are also carriers of microorganism, particularly, inherent and non-invasive characteristic endophytes in host plant. Therefore, in this study, the endophytic diversity of Angelica seeds was studied and compared with endophytes isolated from healthy leaves, stems, roots, and seeds of A. sinensis using 20 different media. The metabolites of endophytic strains were evaluated with six different methods for their antioxidant activity and the paper disc diffusion method for antimicrobial activities. As a result, 226 endophytes were isolated. Compared with the biodiversity and abundance of uncultured fungi from Angelica seed, the result showed that the most frequent endophytic fungi were Alternaria sp. as seen in artificial media; moreover, compared with artificial media, the pathogenic fungi, including Fusarium sp. and Pseudallescheria sp., were not found from the Angelica seed, the results suggested it may not be inherent endophytes in plants. In addition, bacteria from seven phyla were identified by high-throughput sequencing, while five phyla of endophytic bacteria were not isolated on artificial media including Proteobacteria , Actinobacteria , Bacteroidetes , Microgenomates , and Saccharibacteria. Furthermore, the sample JH-4 mycelium displayed the best antioxidant activity, and the active constituent may be a flavonoid as determined by total phenol and flavonoid content. Moreover, YH-12-1 mycelium had strong inhibitory activity against the five tested strains and the minimum inhibitory concentration (MIC) against Pseudomonas aeruginosa and Streptococcus pneumoniae was found to be 25 μg/mL. How community-level specialization differs among groups of organisms, and changes along environmental gradients, is fundamental to understanding the mechanisms influencing ecological communities. In this paper, we investigate the specialization of root-associated fungi for plant species, asking whether the level of specialization varies with elevation. For this, we applied DNA barcoding based on the ITS region to root samples of five plant species equivalently sampled along an elevational gradient at a high arctic site. To assess whether the level of specialization changed with elevation and whether the observed patterns varied between mycorrhizal and endophytic fungi, we applied a joint species distribution modeling approach. Our results show that host plant specialization is not environmentally constrained in arctic rootassociated fungal communities, since there was no evidence for changing specialization with elevation, even if the composition of root-associated fungal communities changed substantially. However, the level of specialization for particular plant species differed among fungal groups, root-associated endophytic fungal communities being highly specialized on particular host species, and mycorrhizal fungi showing almost no signs of specialization. Our results suggest that plant identity affects associated mycorrhizal and endophytic fungi differently, highlighting the need of considering both endophytic and mycorrhizal fungi when studying specialization in root-associated fungal communities. Taxol, a phyto-extracted diterpenoid, is the most commercially needed drug in cancer chemotherapy. In spite of the microbial production of taxol being successful and prospective, the reported yields are still not sufficient for large-scale production. Thus, the discovery of new taxol-producing microbial strains and production enhancement methodologies such as process optimization, strain improvement, and immobilization technique are the main objectives. In this paper, a taxol-producing start strain Epicoccum nigrum TXB502 (initial yield 61.35 μg L−1) was isolated from Taxus baccata and identified by morphological and molecular tools. The optimum cultivation and nutritional conditions were assessed by testing one parameter at a time approach that resulted in 88.59% significant production increase. In addition, a stable mutant with improved productivity (40.07% yield increase in comparison with the parent strain) was successfully developed after gamma irradiation mutagenesis of the start strain. The taxol titer was further improved via testing different immobilization carriers for both spores and mycelia of this mutant. Over taxol production was achieved using alginate-immobilized mycelia with the feasibility of conducting six successive production cycles in a semi-continuous form. The final total concentration reached 8187.77 μg taxol 6 L−1 which represents approximately 22-fold increase, as compared to the initial titer of the start strain. These findings can pave the way for the prospective industrial manufacturing of taxol, as the achieved taxol production in this study is the highest reported by academic laboratories for microbial cultures. This study was conducted to evaluate the insecticidal activity of the endophytic fungus Isaria fumosorosea Wize isolated from Ricinus communis L. against Locusta migratoria L. and Acanthoscelides obtectus Say. To evaluate the insecticidal activity, a mixture of fungal extract and spore suspensions (1x107, 1x106 and 1x105 spores/ml of dissolved extract) were applied against the insects. It was found that I. fumosorosea had a good insecticidal activity, positively proportional to concentration and time after exposure (24, 48, 72 hours). A maximum mortality rate of 80% for A. obtectus and 70% for L. migratoria at 1x107 spores/ml of dissolved extract was recorded. Preliminary phytochemical analyses of the fungal extract revealed the presence of alkaloids, flavonoids and phenols. Among the non-chemical insect control methods, biological control is one of the most effective human and environmentally friendly alternatives. One of the main biological control methods is the application of entomopathogenic fungi (EPF). Today, biological crop protection with EPF plays a key role in projects for the sustainable management of insect pests. EPF have several advantages over conventional insecticides, including cost-effectiveness, high yield, absence of harmful side-effects for beneficial organisms, fewer chemical residues in the environment and increased biodiversity in ecosystems. Apart from direct application as contact bioinsecticides, EPF are able to colonize plants as endophytes acting not only as pest and disease control agents but also as plant growth promoters. The present paper presents an outline of the biocontrol potential of several EPF, which could be harnessed for the development of new integrated pest Management (IPM) strategies. Emphasis is given on benefits of endophytic EPF, on issues for practical application and in fields in need of further research. Our findings are discussed in the context of highlighting the value of entomopathogenic fungal endophytes as an integral part of pest management programs for the optimization of crop production. Endophytes are one of the symbiotic associations which can be successfully utilized to mitigate negative effects of abiotic stress in agricultural crops. Habitat adapted symbiotic fungal endophytes are known to impart drought tolerance in host plants. This study aimed at evaluating fungal endophytes of drought adapted plants against drought stress in maize. Fungal endophytes were isolated from arid and semi-arid regions of Karnataka and screened for their ability to grow in matric modified media using Polyethylene glycol (PEG 8000). Out of 65 isolates, six isolates (P1R1, P4L5, P6R3, P6R4, P10R1 and P12R2) showed increased mycelial growth up to -10.31 MPa (30 % PEG). The six fungal endophytes were evaluated for their ability to alleviate drought stress and improve growth of maize seedlings at PEG induced drought stress under in vitro condition. Maize seedlings inoculated with fungal endophytes showed significant increase in shoot length under drought stress after 10 days compared to control. Among six endophytes, the isolate P10R1 significantly enhanced shoot and root length of maize. The fungus was identified as Alternaria burnsi by using ITS region sequences. This study revealed that the fungal endophyte, Alternaria burnsi isolated from drought adapted plant has ability to mitigate drought stress and improve the maize growth under drought condition. White root disease is a significant disease of cashew caused by Rigidoporus sp. Five endophytic fungal isolates, namely AR31D (Fusarium proliferatum), AR42D (Penicillium citrinum), BR32C (Trichoderma asperellum), VNTB1 (Chaetomium sp.), and EAGS14 (Curvularia lunata), were assessed as the biocontrol agents against Rigidoporus sp. in vitro and in planta. The research objective was to obtain endophytic fungi that effectively control Rigidoporus sp. and their mechanisms. The in vitro test results showed that all isolates could inhibit Rigidoporus sp. and promote plant growth by producing volatile organic compounds, chitinase enzymes, and indole acetic acid. Meanwhile, only four isolates could solubilize phosphate with low-medium solubilization efficiency. The isolates successfully colonized the root of cashew saplings in 10-65%. The effectiveness of endophytic fungal isolates in controlling white root disease was determined by the mechanisms involved, such as resistance induction (increased activity of defense enzymes like polyphenol oxidase), chitinase enzyme production, indole acetic acid production, phosphate solubilization, and suppression of plant stress which observed from decreased malondialdehyde concentrations in saplings' roots. Laccase belongs to a small group of enzymes called the blue multicopper oxidases, having the potential ability of oxidation. It belongs to enzymes, which have innate properties of reactive radical production, but its utilization in many fields has been ignored because of its unavailability in the commercial field. There are diverse sources of laccase producing organisms like bacteria, fungi and plants. In fungi, laccase is present in Ascomycetes, Deuteromycetes, Basidiomycetes and is particularly abundant in many white-rot fungi that degrade lignin. Laccases can degrade both phenolic and non-phenolic compounds. They also have the ability to detoxify a range of environmental pollutants. Due to their property to detoxify a range of pollutants, they have been used for several purposes in many industries including paper, pulp, textile and petrochemical industries. Some other application of laccase includes in food processing industry, medical and health care. Recently, laccase has found applications in other fields such as in the design of biosensors and nanotechnology. Endophytic fungi are microorganisms that, exhibiting within the plant tissues without causing any apparent harm to the host, establish a symbiotic relationship with plants. Host plants provide endophytic fungi with essential nutrients and a protected environment. In exchange, the fungi can enhance the plant's ability to acquire nutrients. They can also play a crucial role in increasing the host plant's tolerance to various abiotic and biotic stresses. Endophytic fungi can produce a wide range of bioactive compounds, some similar to those found in the host plant. In Iraq's Duhok province of the Kurdistan region, the plant species Vitis vinifera has been explored as a habitat for diverse endophytic microorganisms across various ecological environments. During the period from 2021 to 2022, a total of 600 samples were collected from four distinct locations: Bagera, Besfke, Barebhar, and Atrush. From these samples, twelve endophytic fungal species were isolated, including Aspergillus flavipes, Botryosphaeria dothidea, Fusarium oxysporum, Fusarium ruscicol, Fusarium venenatum, Chaetomium globosum, Clonostachys rosea, Mucor racemosus, Penicillium glabrum, Aspergillus terreus, Aspergillus nidulans, and Aspergillus niger, Alternaria alternata, Paecilomyces maximus, Curvularia buchloes. These fungi were introduced for their potential as biocontrol agents against grapevine trunk diseases and grape rotting fungi, which pose significant risks to grapevine health and productivity. Penicilium radiatolobatum, Botrysphaeria dothidea, Fusarium ruscicola, Fusarium venenatum, and Paecilomyces maximus represented the first record as endophytes on grapevine in Iraq. Based on ITS and SSU sequencing, molecular identification confirmed these fungi's presence with sequence identities ranging from 99% to 100%. Phylogenetic analysis revealed that these endophytes could be categorized into five main clusters (A, B, C, D, and E), showing high intra-group similarity. Utilizing the Dual Culture method, the endophyte Paecilomyces maximus demonstrated a 70.83% inhibition rate against Ilyonectria destructans. In the Food Poisoning method, A. flavipes and P. maximus emerged as the most effective inhibitors of Ilyonectria destructans, whereas A. terreus, M. racemosus, and P. maximus achieved complete inhibition (100%) of Botrytis cinerea. Additionally, M. racemosus was identified as the most effective biocontrol agent against Neoscytalidium dimidiatum. In conclusion, the study emphasizes the potential of endophytic fungi from Vitis vinifera as effective biocontrol agents against grapevine diseases, highlighting their role in sustainable vineyard management. Three sulfur-containing alkaloids aplospojaveedins A-C (1-3) with a hitherto undescribed carbon skeleton comprising octahy-dronaphthalene, a, ß-unsaturated lactam and glycine-cysteine moieties were isolated from Aplosporella javeedii. Their structures were elucidated by 1D and 2D NMR spectroscopy, HR-MS, X-ray diffraction analysis, DFT-NMR and TDDFT-ECD calculations. A plausible biosynthetic pathway and putative targets are described. Introduction: Understanding the microbial diversity and potential functional dynamics within the rhizocompartments of Dendrobium huoshanense is crucial for unraveling the plant--microbe interactions that influence its medicinal properties. Methods: This study is the first to characterize the microbiome associated with the rhizocompartments of D. huoshanense, including its cultivation medium, rhizosphere, rhizoplane, and root endosphere, using high-throughput sequencing and subsequent bioinformatic analysis. Results: Bacterial phylogenetic diversity was significantly higher in the endosphere than in the rhizosphere, while fungal α-diversity significantly decreased from the cultivation medium to the endosphere. Both bacterial and fungal niche widths decreased from the cultivation medium to the endosphere. β-Diversity analysis revealed distinct spatial patterns in both bacterial and fungal communities across the rhizocompartments, with the most pronounced differences between the cultivation medium and the endosphere. Taxonomically, Proteobacteria and Ascomycota were predominant in the endosphere for bacterial and fungal communities, respectively. Functional predictions showed significant enrichment of pathways related to xenobiotics biodegradation, lipid metabolism, and nitrogen fixation in the endosphere, while functions associated with plant pathogens and saprotrophs were significantly reduced. Discussion: The results indicate a shift from generalist to specialist microbes from the cultivation medium to the endosphere, suggesting that D. huoshanense exerts strong selective pressure for endophytic fungi. Interestingly, a high proportion of fungi with unknown functions were found in the endosphere, highlighting an area for further research regarding the medicinal efficacy of D. huoshanense. Background: Hazardous synthetic dye wastes have become a growing threat to the environment and public health. Fungal enzymes are eco-friendly, compatible and cost-effective approach for diversity of applications. Therefore, this study aimed to screen, optimize fermentation conditions, and characterize laccase from fungal endophyte with elucidating its ability to decolorize several wastewater dyes. Results: A new fungal endophyte capable of laccase-producing was firstly isolated from cladodes of Opuntia ficus-indica and identified as T. harzianum AUMC14897 using ITS-rRNA sequencing analysis. Furthermore, the response surface methodology (RSM) was utilized to optimize several fermentation parameters that increase laccase production. The isolated laccase was purified to 13.79-fold. GFC, SDS-PAGE revealed laccase molecular weight at 72 kDa and zymogram analysis elucidated a single band without any isozymes. The peak activity of the pure laccase was detected at 50 °C, pH 4.5, with thermal stability up to 50 °C and half life span for 4 h even after 24 h retained 30% of its activity. The Km and Vmax values were 0.1 mM, 22.22 µmol/min and activation energy (Ea) equal to 5.71 kcal/mol. Furthermore, the purified laccase effectively decolorized various synthetic and real wastewater dyes. Conclusion: Subsequently, the new endophytic strain produces high laccase activity that possesses a unique characteristic, it could be an appealing candidate for both environmental and industrial applications. Endophytic fungi are promising producers of phenolic compounds; however, the process of extracting these molecules from the culture medium is a crucial step for obtaining these metabolites. In this context, the best conditions for extracting phenolic compounds produced by Cophinforma mamane CF2-13, isolated from an Amazonian plant, were evaluated. The fungus was cultivated in a liquid medium to obtain its metabolites. The effects of the solvent, time, and temperature were investigated, as well as the use of ultrasound. The total phenolics and the antioxidant activity were assessed to define the best extraction conditions. The optimized extract was chemically characterized and used to analyze its potential in protecting olive oil from lipid oxidation. The best results were found using ethyl acetate at 20 °C for 2 h. These conditions led to 61.61 mg GAEq/g of extract of total phenolics, and an antioxidant capacity of 0.47, 0.45, and 0.29 mmol TEq/g of extract using the DPPH, ABTS, and FRAP methods, respectively. In addition, the optimized extract presented caffeic acid and was able to delay the lipid oxidation of olive oil. The growing emergence of multi-drug resistant microbial strains has kept the scientific world searching for novel bioactive compounds with specific chemical characteristics. Accordingly, researchers have started exploring the understudied metabolites from endophytes as a new source of bioactive compounds. In this context, the current study was designed to evaluate the bioactive properties of endophytic fungi from the Mokrzański forest in Wrocław, Poland that have not yet been fully researched. Forty-three endophytic fungi were isolated from twelve distinct plants. Following their cultivation, fungal extracts were separately prepared from biomass and cell-free filtrates, and their antibacterial, antifungal (against human and plant pathogens), and antioxidant properties were examined. Five promising fungi after screening were identified to possess all of these activities. These strains and their respective plant hosts were Trichoderma harzianum BUK-T (Fagus sylvatica), Aspergillus ochraceus ROB-L1 (Robinia pseudoacacia), Chaetomium cochliodes KLON-L1, Fusarium tricinctum KLON-L2 (Acer platanoides), and Penicillium chrysogenum SOS-B2 (Pinus sylvestris). Moreover, gamma irradiation at several doses (Gy) was separately applied to the fungal cultures to study their effects on the recorded activities. Finally, compounds after preparative thin-layer chromatography fractionation of the five fungal strains were identified by GC-MS. These findings suggest that the isolated endophytic fungi could serve as novel sources of bioactive metabolites with antibacterial, antifungal, and antioxidant properties, potentially paving the way for future research and the development of new bioactive compounds. The search for natural product-based biopesticides from endophytic fungi is an effective tool to find new solutions. In this study, we studied a pre-selected fungal endophyte, isolate YCC4, from the paleoendemism Persea indica, along with compounds present in the extract and the identification of the insect antifeedant and nematicidal ones. The endophyte YCC4 was identified as Phyllosticta sp. by molecular analysis. The insect antifeedant activity was tested by choice bioassays against Spodoptera littoralis, Myzus persicae, and Rhopalosiphum padi, and the in vitro and in vivo mortality was tested against the root-knot nematode Meloidogyne javanica. Since the extract was an effective insect antifeedant, a strong nematicidal, and lacked phytotoxicity on tomato plants, a comprehensive chemical study was carried out. Two new metabolites, metguignardic acid (4) and (-)-epi-guignardone I (14), were identified along the known dioxolanones guignardic acid (1), ethyl guignardate (3), guignardianones A (5), C (2), D (7), and E (6), phenguignardic acid methyl ester (8), the meroterpenes guignardone A (9) and B (10), guignarenone B (11) and C (12), (-)-guignardone I (13), and phyllomeroterpenoid B (15). Among these compounds, 1 and 4 were effective antifeedants against S. littoralis and M. persicae, while 2 was only active on the aphid M. persicae. The nematicidal compounds were 4, 7, and 8. This is the first report on the insect antifeedant or nematicidal effects of these dioxolanone-type compounds. Since the insect antifeedant and nematicidal activity of the Phyllosticta sp. extract depend on the presence of dioxolanone components, future fermentation optimizations are needed to promote the biosynthesis of these compounds instead of meroterpenes. Exopolysaccharides (EPS) are bioactive metabolites with high molecular weight and are produced by several microorganisms such as endophytic fungi. Thus, the aim of the present study was designed to explore the biosynthesis and characteristics of EPS by the endophytic fungus Neopestalotiopsis clavispora AUMC15969 and then investigate some environmental and nutritional factors that affect their production. Moreover, we estimated an additional value, namely the production of biodiesel. Maximum production of EPS was 7.86 g/L when N. clavispora was grown on lactose as the sole carbon source and peptone as the nitrogen source, respectively, and pH 7 at 35 °Cfor 10 days. The produced EPS had a total sugar content of 0.93 g/g where protein content was 0.076 g/g. It revealed a strong antioxidant activity that improved with increasing sample concentration, with the optimum concentration of 10 mg/mL producing 83.1% DPPH radical scavenging activity with an IC50 equal to 3.89 mg/mL. The extracted lipid from the fungal mycelia at the end of the fermentation process was 31.76% w/w. The biodiesel produced from the transesterification of lipids was 87.4% total fatty acid methyl esters. The present study revealed the consequences of the interaction of a widely used bioinsecticide and endophyte Metarhizium anisopliae with the hazardous mycotoxin zearalenone (ZEN) as a pure substance and with ZEN as a native component of a crude Fusarium extract. In the environment, microorganisms encounter a mixture of metabolites secreted by other organisms living in the same area, not single substances. The obtained results suggest that M. anisopliae, exposed to a variety of active substances produced by Fusarium graminearum, is able to eliminate ZEN. Within 14 days, M. anisopliae biotransformed 90.8% and 85.8% of ZEN as a pure substance and ZEN as a native component of the F. graminearum extract from Rice Medium (E-Fg-RM), respectively, through reduction predominantly to α-epimers of zearalenols and zearalanols, considered more estrogenic than ZEN, which can raise concerns. Compared to pure ZEN, E-Fg-RM significantly affected the production of Metarhizium secondary metabolites by increasing the destruxins amount by approximately 20–25% and reducing the swainsonine content by 96.2%. All these findings provide a possible picture of the interaction of M. anisopliae with ZEN in the wild, mainly as a result of the use of crude extract from Fusarium, which contained a mixture of different metabolites. Aquilaria sinensis is a significant resin-producing plant worldwide that is crucial for agarwood production. Agarwood has different qualities depending on the method with which it is formed, and the microbial community structures that are present during these methods are also diverse. Furthermore, the microbial communities of plants play crucial roles in determining their health and productivity. While previous studies have investigated the impact of microorganisms on agarwood formation, they lack comprehensiveness, particularly regarding the properties of the microbial community throughout the entire process from seedling to adult to incense formation. We collected roots, stems, leaves, flowers, fruits and other tissues from seedlings, healthy plants and agarwood-producing plants to address this gap and assess the dominant bacterial species in the microbial community structures of A. sinensis at different growth stages and their impacts on growth and agarwood formation. The bacteria and fungi in these tissues were classified and counted from different perspectives. The samples were sequenced using the Illumina sequencing platform, and sequence analyses and species annotations were performed using a range of bioinformatics tools to assess the plant community compositions. An additional comparison of the samples was conducted using diversity analyses to assess their differences. This research revealed that Listeria, Kurtzmanomyces, Ascotaiwania, Acinetobacter, Sphingobium, Fonsecaea, Acrocalymma, Allorhizobium, Bacillus, Pseudomonas, Peethambara, and Debaryomyces are potentially associated with the formation of agarwood. Overall, the data provided in this article help us understand the important roles played by bacteria and fungi in the growth and agarwood formation process of A. sinensis, will support the theoretical basis for the large-scale cultivation of A. sinensis, and provide a basis for further research on microbial community applications in agarwood production and beyond. Root-associated fungal endophytes may facilitate nitrogen (N) absorption in plants, leading to benefits in photosynthesis and growth. Here, we investigated whether endophytic insect pathogenic fungi (EIPF) are capable of transferring soil N to the crop species Chenopodium quinoa. We evaluated nutrient uptake, carbon allocation, and morpho-physiological performance in C. quinoa in symbiosis with two different EIPF (Beauveria and Metarhizium) under contrasting soil N supply. A controlled experiment was conducted using two plant groups: (1) plants subjected to low N level (5 mM urea) and (2) plants subjected to high N level (15 mM urea). Plants from each group were then inoculated with different EIPF strains, either Beauveria (EIPF1+), Metarhizium (EIPF2+) or without fungus (EIPF-). Differences in N and C content, amino acids, proteins, soluble sugars, starch, glutamine synthetase, glutamate dehydrogenase, and physiological (photosynthesis, stomatal conductance, transpiration), and morphological performance between plant groups under each treatment were examined. We found that both Beauveria and Metarhizium translocated N from the soil to the roots of C. quinoa, with positive effects on photosynthesis and plant growth. These effects, however, were differentially affected by fungal strain as well as by N level. Additionally, an improvement in root C and sugar content was observed in presence of EIPF, suggesting translocation of carbohydrates from leaves to roots. Whereas both strains were equally effective in N transfer to roots, Beauveria seemed to exert less demand in C. quinoa for photosynthesis-derived carbohydrates compared to Metarhizium. Our study revealed positive effects of EIPF on N transfer and morpho-physiological performance in crops, highlighting the potential of these fungi as an alternative to chemical fertilizers in agriculture systems. Polygonum cuspidatum, alternatively known as Fallopia japonica or Reynoutria japonica, is a perennial herb belonging to the Polygonaceae family. Commonly called Japanese knotweed or Asian knotweed, this plant is native to East Asia, particularly in regions such as Korea, China, and Japan. It has successfully adapted to a wide range of habitats, resulting in it being listed as a pest and invasive species in several countries in North America and Europe. This study focuses on analysing the composition of the bacterial and fungal endophytic communities associated with Japanese knotweed growing in the Russian Far East, employing next-generation sequencing (NGS) and a cultivation-based method (microbiological sowing). The NGS analysis showed that the dominant classes of endophytic bacteria were Alphaproteobacteria (28%) and Gammaproteobacteria (28%), Actinobacteria (20%), Bacteroidia (15%), and Bacilli (4%), and fungal classes were Agaricomycetes (40%), Dothideomycetes (24%), Leotiomycetes (10%), Tremellomycetes (9%), Pezizomycetes (5%), Sordariomycetes (3%), and Exobasidiomycetes (3%). The most common genera of endophytic bacteria were Burkholderia-Caballeronia-Parabukholderia, Sphingomonas, Hydrotalea, Methylobacterium-Metylorubrum, Cutibacterium, and Comamonadaceae, and genera of fungal endophytes were Marasmius, Tuber, Microcyclosporella, Schizothyrium, Alternaria, Parastagonospora, Vishniacozyma, and Cladosporium. The present data showed that the roots, leaves, and stems of P. cuspidatum have a greater number and diversity of endophytic bacteria and fungi compared to the flowers and seeds. Plant tissues harbor abundant endophytes, which are crucial for plant growth. Endophytes present in Alsophila spinulosa, which is enriched with medicinal components, have not been isolated and characterized yet. Here we employed meta-amplicon sequencing to identify endophytic species and examined their diversity in the leaves, petioles, roots and stems of A. spinulosa. Our findings revealed 1,247 operational taxonomic units (OTUs) for endophytic bacteria across 210 species and 476 OTUs for endophytic fungi across 222 species. Alpha diversity analysis showed the highest endophytic bacterial diversity in A. spinulosa roots, whereas fungal diversity was similar across the leaf, petiole and root tissues. Fungal diversity in the leaves and petioles was markedly higher than that in the stems. Furthermore, beta diversity analysis revealed similarities in the endophytic bacterial and fungal compositions between the leaves and petioles, whereas the compositions in roots and stems considerably differed from those in the leaves and petioles. At the genus level, the predominant endophytic bacteria were Methylobacterium-Methylorubrum and Pseudomonas, whereas the predominant endophytic fungi were Cutaneotrichosporon and Pseudofabraea. Linear discriminant analysis effect size revealed characteristic endophytic bacterial genera specific to each tissue type and characteristic endophytic fungal genera specifically in the leaves, petioles and roots. The co-occurrence network analysis indicated that the complexity of endophyte networks was the highest in the leaves and the lowest in the stems of A. spinulosa. Overall, this study elucidates the distribution patterns of endophytes in A. spinulosa across various tissues, offering valuable microbial resources for the development of natural products for medicinal application. The increasing emergence and re‐emergence of resistant pathogenic microbes causes a health threat to the human population. Scientists have been striving to find novel bioactive compounds and drugs to overcome these obstacles. This study aimed to characterize mangrove endophytic fungi and evaluate their antibacterial activity. Heritiera littoralis, Rhizophora mucronata, Bruguiera gymnorrhiza, Avicennia marina, and Xylocarpus granatum species were collected from Tudor Creek, Mida Creek, and Gazi Bay. A total of 30 fungal isolates were subjected to molecular identification based on analysis of their ITS gene region. The isolates in the inferred phylogenetic trees were affiliated with the genus Aspergillus. Ethyl acetate and butanol crude extracts of 38.2% of the 76 isolated fungal endophytes and eight mycelia samples were screened for antibacterial activity against Staphylococcus aureus (ATCC 27853), Escherichia coli (ATCC 25922), and Pseudomonas aeruginosa (ATCC 25923) using the disc diffusion method. A. marina and R. mucronata harbored the most fungal endophytes that showed the highest antibacterial activity. Seven fungal broth extracts exhibited higher antibacterial activities against the tested microorganisms than the positive control. The minimum inhibitory concentration (MIC) activity for the isolates demonstrated that the ethyl acetate extract of a root endophytic fungal isolate (RC6) (3.31 ± 0.01) of A. marina is a strong inhibitor since it showed significantly lower MIC activity compared to the positive control (3.84 ± 0.00) against Pseudomonas aeruginosa (P < 0.05). Therefore, this study confirms that mangrove species harbor fungal isolates that have antibacterial activity and hence could serve as a novel source of antibiotics. It is recommended that the pure compounds from these extracts be isolated for further bioactivity tests and structural elucidation for consideration as lead molecules in drug discovery. Endophytic fungi are considered an eco-friendly and bio-safe alternative to increase agricultural productivity. The study objectives were to isolate and identify endophytic fungi from the roots of Rhizophora racemosa and to assess their effect on the growth of Cucurbita maxima plants. Molecular identification of the endophytic fungal isolates revealed five fungal species: Aspergillus aculeatus, Aspergillus fumigatus, Fusarium equiseti, Penicillium citrinum, and Talaromyces albobiverticillius. A. aculeatus induced the best improvement rate of plant development with an increase of shoot length (159 %), petiole length (171%), internode length (155%), leaf number (133%) and leaf area (149%) at 84 days after the fungal treatment. This research highlights the importance of A. aculeatus, which can be an eco-friendly bio-fertilizer that can enhance the production of Cucurbita maxima and improve the agricultural sector. Dichrocephala integrifolia is a wild medicinal plant utilised in traditional healing and Ayurveda to cope up with several health issues by various groups of people around the world. Medicinal plants are associated with diverse fungal endophytes with potential bioactive properties. In this investigation, 26 fungal endophytes were isolated from D. integrifolia and three sterile forms using the Petri plate culture method. The endophytic isolation rate was highest for inflorescence (35.97%) and lowest for stem (15.61%). The highest colonization frequency was shown by F. solani (8.37%) and the lowest by Sterile morphotype 3 (1.36%). Out of the four plant parts, inflorescence was found to be highly infected, displaying an infection rate of 93.43% and the least infection occurred in the stem with 45%. The maximum number of isolated endophytic fungi belongs to the class Sordariomycetes, with a relative occurrence (%) of 71.72%. The Simpson's diversity index reveals that the leaf endophytes were more diverse (0.94). Qualitative antifungal activity of the sporulating isolates against Curvularia lunata has shown that the maximum number of endophytes possessed Class 3 antagonism. Four isolates were selected based on screening of their antagonistic activity and their antifungal inhibition was calculated against nine fungal phytopathogens. Maximum inhibition (100%) was shown by Trichoderma sp. 2 (S2B2) against Alternaria alternata, A. brassicicola, Colletotrichum capsici, C. lunata, and Ustilaginoidea virens and least inhibition by Gliocladium sp. 1 (19.78%) against C. lunata. The four isolates were found to produce protease, lipase, amylase and cellulase enzymes. The isolates produced ammonia and hydrogen cyanide, but none of the isolates could solubilize phosphate. Corylus heterophylla, the majestic hazelnut tree, stands as one of the esteemed quartets of grand nuts reigning across the landscapes of temperate Asia, Europe, and North America. However, its relationship with endophytic fungi remains thinly explored. This study successfully identified a new species designated as Nigrospora coryli through conjoint analysis of morphology and systematics. Endophytic fungi isolated from Terminalia chebula exhibit potent antimicrobial activity, showcasing their potential to develop novel therapeutic agents. Additionally, these fungi produce diverse phytochemicals, which may contribute to their medicinal properties and offer new avenues for natural product research. This research study investigated the endophytic fungi isolated from T. chebula and assessed their antimicrobial and phytochemical potential. A total of 28 endophytic fungal species were isolated from leaf and bark tissues. Antimicrobial assays revealed significant inhibitory effects against pathogenic fungi Candida albicans - MTCC 183 & Saccharomyces cerevisiae - MTCC 174 and bacteria Escherichia coli - MTCC 2939, & Bacillus subtilis - MTCC 1305, with endophytic Alternaria sp. exhibiting maximum inhibition zone for antifungal activity and endophytic Aspergillus flavus demonstrating potent activity for their antibacterial analysis. A wide array of phytochemical compounds like alkaloids, flavonoids, phenols, steroids, terpenoids, tannin, saponin, etc, were observed whereas, phytochemical screening indicated the presence of alkaloids and flavonoids in all selected endophytes. Endophytic microorganisms are promising sources for new biocatalysts as they must deal with their host plants' chemicals by developing adaptative strategies, such as enzymatic pathways. As part of our efforts in selecting endophytic strains as biocatalysts, this study describes the screening of endophytic fungi isolated from Handroanthus impetiginosus leaves for selective bioreduction of Acetophenone. The bioreductions were monitored by chiral gas chromatography and conducted to the selection of the endophyte Talaromyces sp. H4 as capable of reducing acetophenone to (S)-1-phenylethanol in excellent conversion and enantiomeric excess rates. The influence of seven parameters on the stereoselective bioreduction of acetophenone by Talaromyces sp. H4 was studied: reaction time, inoculum charge, shaking, pH, temperature, substrate concentration, and co-solvent. The optimal conditions were then used to reduce substituted acetophenones and Acetophenone scale-up, which furnished (S)-1-Phenylethanol in 73% yield and 96% ee. The results highlight the endophytic fungus Talaromyces sp. H4 as an excellent biocatalyst for stereoselective reduction of prochiral carbonyls. [ABSTRACT FROM AUTHOR]

Our research group isolated an endophyte yeast from the endemic plant Echinopsis chiloensis. This yeast, identified as Naganishia sp, produces volatile organic compounds (VOC) with antifungal activity against Botrytis cinerea. Due to the need for alternative control methods for this pathogen, the effect of VOC on B. cinerea was analyzed. On the fourth day of cultivation, in the presence of VOCs, the mycelial growth of B. cinerea stopped. VOCs inhibited 32.8% of the conidia germination and reduced sporulation by 70.6%. These compounds promoted the formation of infection cushions. VOCs caused damage to the cell wall and plasma membrane in B. cinerea. On the other hand, the volatile compounds induced oxidative stress, and led to membrane lipid peroxidation after 16 and 24 h of incubation in the presence of VOCs, because an increased level of malondialdehyde content was observed. These compounds exerted a fungistatic effect on B. cinerea. The volatile compounds emitted by Naganishia sp were identified by gas chromatography coupled with mass spectrometry as 3-methylbutyl acetate, ethylbenzene, 1-ethyl-3-methylbenzene, 2-phenylethanol, 2-heptanone and (5E)-6,10-dimethylundeca-5,9-dien-2-one. The most abundant volatile compound, 2-phenylethanol, inhibited the mycelial growth of B. cinerea with an IC50 value of 0.21 µL mL−1. The effect of this pure compound was also fungistatic. Finally, it was shown that volatile compounds decreased the severity of B. cinerea infection in tomatoes and grapes by 64.7 and 43,1%, respectively.
