Background Canine brucellosis , caused by the bacterium Brucella canis , is a zoonotic and largely reproductive disease of dogs. The disease is a recognized problem in canine breeding populations, and the risk to individuals assisting with birthing is well described. Prior to 2015, all cases of canine brucellosis reported to the Minnesota Board of Animal Health were in dogs used for breeding. In 2015, canine brucellosis was identified in eight Minnesota rescue dogs, all originating from specific geographic areas in South Dakota. Our objective was to measure the seroprevalence of B. canis in stray and previously owned dogs entering a large Minnesota animal rescue organization to determine if our observations represented a localized or generalized disease issue among rescue dogs. Methods A stratified random sample of stray and previously owned dogs entering the largest Minnesota animal rescue organization between November 1, 2016 and November 7, 2017, was tested for B. canis antibodies by the 2-Mercaptoethanol Rapid Slide Agglutination Test (2ME-RSAT) (Zoetis d -TEC® CB kit). Sample sizes for each strata were calculated using previously published seroprevalence estimates. Blood from selected dogs was collected, serum harvested, and transported to the Minnesota Veterinary Diagnostic Laboratory for testing. Positive samples in the 2ME-RSAT were shipped to Cornell University for confirmation by Agarose Gel Immunodiffusion (AGID) testing. Demographics, state and setting of origin, and health status were collected on study-dogs. Results Of the 10,654 dogs accepted by AHS during the study period, 943 (8.9%) were selected for testing. Most study dogs arrived from Oklahoma (28%), Alabama (18%), and Minnesota (12%). The median age of study dogs was 1.5 years; 303 (32%) were intact males and 294 (31%) were intact females. Most study dogs were strays (n = 716, 76%). Of the total, 22 (3.1%) stray and eight (3.5%) owner-surrendered dogs were presumptively positive by RSAT; one (0.11%) of the stray dogs was positive by 2ME-RSAT and confirmed by AGID. The positive dog was a healthy-appearing 1 year-old neutered male beagle from Texas. Conclusions The seroprevalence of canine brucellosis in dogs entering Minnesota for adoption from multiple states was low. Never-the-less, care must to be taken to consider all potential risks and outcomes of interstate and international dog trade, including the spread of infectious diseases such as canine brucellosis.

Optimal age for ovariohysterectomy or castration has not been defined in the scientific literature. Bitches and queens are significantly less likely to develop mammary neoplasia, which has a high incidence and potentially high morbidity and mortality, if spayed when young. Tom cats exhibit undesirable behaviors that preclude them being good pets and should be castrated young. There is no compelling reason to castrate male dogs when young unless it is needed to control reproductive behaviors or prevent indiscriminate breeding. Alternatives to surgical sterilization that may be available in the future include intratesticular injection and immunization against gonadotropin-releasing hormone.

A massive open online course (MOOC) in canine theriogenology was offered for dog owners and breeders and for veterinary professionals as a partnership between the University of Minnesota and Coursera. The six-week course was composed of short video lectures, multiple-choice quizzes with instant feedback to assess understanding, weekly case studies with peer evaluation to promote integration of course materials, and discussion forums to promote participant interaction. Peak enrollment was 8,796 students. The grading policy for completion was strict and was upheld; completion rate for all participants was 7.5%. About 12% of participants achieved a grade of over 90% in the course, with those who had any deficiency mostly missing one quiz or assignment. Ninety-nine individuals were enrolled in a for-cost, credentialed pathway, and 50% of those individuals completed all required course components. Pre- and postcourse surveys were used to demonstrate that learning objectives were met by the participants and to identify that lack of time to commit to study was the biggest impediment to completion. Positive aspects of the course were active engagement by participants from all over the world and the ability of this university and instructor to reach those learners. Negative aspects concerned technical support and negative feedback from some participants who were unable to meet course requirements for reasons beyond the control of the instructor.

Pediatric gonadectomy is most commonly performed by humane organizations as a means of population control. Benefits and detriments of gonadectomy are reviewed, with special attention to literature describing effects specific to age at gonadectomy. Techniques for pediatric anesthesia and surgery are reviewed.

Two castrated male dogs presented for evaluation of tenesmus. Presurgical evaluations included complete physical examinations, serum biochemistry, abdominal ultrasonography, and MRI (case 2 only). Paraprostatic cysts were diagnosed in both cases based on the results of abdominal ultrasonography, MRI, and histopathology of tissue samples obtained during exploratory laparotomy. To the authors' knowledge, the two cases presented herein are the first documented cases of paraprostatic cysts that developed after castration in male dogs. Paraprostatic cysts should be considered in the differential diagnoses for castrated male dogs with prostatic disease.

Semen was collected from eight dogs after SC administration of 0.1 mg/kg PGF2alpha or 0.6 mL 0.9% NaCl solution 15 min prior to collection in the presence or absence of an estrous teaser bitch (switchback design; all dogs given all four treatments in random sequence). There were more spermatozoa (P = 0.02) in ejaculates collected after administration of PGF2alpha in the presence of an estrous teaser bitch ((852 ± 736) × 106, mean ± S.D.) than in ejaculates collected in saline-treated dogs in the absence of a teaser bitch ((371 ± 620) × 106). However, the number of spermatozoa in the ejaculate of dogs given PGF2alpha in the absence of a teaser bitch and in dogs given saline in the presence of a teaser bitch ((556 ± 494 and 600 ± 622) × 106, respectively) were not significantly different from each other or from the other two groups. The percentage of morphologically normal spermatozoa did not vary by treatment (P = 0.51). In conclusion, treatment with PGF2alpha and presence of a teaser bitch had an additive effect on the number of spermatozoa. This, coupled with relatively minor side-effects, suggests this is a useful technique to increase number of spermatozoa in a single canine ejaculate.

Definitive diagnosis of reproductive tract infection or other disease often requires sampling of tissue, either for culture or histopathology. Indications, sample collection technique, possible side-effects and interpretation of results are reviewed. Pertinent facts include: (1) collection of uterine biopsy specimens via laparotomy was associated with higher yield of diagnostic samples and fewer side-effects than other less invasive techniques; (2) vaginal culture samples should be collected from the anterior vagina to minimize number of contaminants in the sample; (3) collection of culture samples from the anterior vagina during proestrus or estrus, in the presence of discharge originating in the uterus, was a non-invasive technique for assessment for uterine infection; (4) samples for bacterial culture from mucosal surfaces, including the vagina and penis, must be quantitated to allow interpretation, with moderate to heavy growth of any single aerobic bacterial organism relevant; (5) mycoplasma and ureaplasma are part of the normal flora of the genitourinary tract in dogs and bitches and, because most laboratories cannot give reliable quantitative results, interpretation of positive results often is difficult; (6) collection of prostatic tissue samples for cytology or culture was more likely to yield a correct diagnosis than submission of ejaculated prostatic fluid.

Seminal fluid was collected by manual ejaculation from 95 dogs. Quantitative aerobic bacterial, qualitative anaerobic bacterial and mycoplasma cultures were performed on the seminal fluid, and their association with presence of inflammatory cells present in the pellet formed after centrifugation of the fluid was investigated. There was a clinically meaningful aerobic bacterial growth in 28.4%, anaerobic bacterial growth in 13.7%, and mycoplasma growth in 57.9% of the seminal fluid samples. Presence of inflammatory cytology was statistically associated with clinically meaningful aerobic bacterial growth. However, of the 78 dogs (82.1%) with clinically meaningful growth of at least one aerobic, anaerobic or mycoplasma organism, 43 (55.1%) had non-inflammatory seminal fluid cytology.

Normal and abnormal reproductive behavior of dogs and cats, and known and proposed hormonal bases for these behaviors are addressed. Emerging information includes use of oxytocin to promote pair-bonding between dam and offspring and the possible effect of prolactin in inhibiting sexual behaviors.

New and traditional techniques for pregnancy diagnosis in the dog are reviewed. Imaging techniques yield the most information, with ultrasonography the preferred technique for assessment of fetal viability and radiography the preferred technique for assessment of litter size. Diabetes mellitus and pregnancy toxemia are uncommon complications of pregnancy. Pathogenesis, diagnosis and treatment of infectious and non-infectious causes of pregnancy loss are reviewed.

Early spay-neuter is ovariohysterectomy or castration of puppies or kittens 6 to 14 weeks of age. Pediatric animals may have an enhanced response to relatively low doses of anesthetic agents. Animals should be fasted no more than 3 to 4 hours before surgery to prevent hypoglycemia, and hypothermia should be avoided. Heart and respiratory rates must be monitored carefully throughout anesthesia. Pediatric gonadectomy surgeries are quick with minimal bleeding. Anesthetic recovery is rapid. No significant short-term or long-term effects have been reported. Prepuberal gonadectomy is most useful for humane organizations and conscientious breeders wishing to preclude reproduction of pet dogs and cats while placing animals at a young enough age to optimize socialization and training.

The canine estrous cycle is discussed with special emphasis on endocrinology. Breeding management and chemical and management strategies for estrus induction and suppression are described.

Elective gonadectomy of dogs and cats is one of the most common veterinary procedures in the United States. Increasingly, dog owners and members of the veterinary profession throughout the world have questioned the optimal age for performance of these procedures and whether these procedure should even be performed as elective surgeries.

To test the hypothesis that male dogs treated with smooth muscle contracting drugs have an increase in the total number of spermatozoa in the ejaculate but no change in all other ejaculate characteristics, such as progressive motility of spermatozoa or percentage morphologically normal spermatozoa, dogs were treated with oxytocin or prostaglandin F2alpha (PGF2alpha) and compared to saline treatments. Semen was collected from each of the 3 dogs once every 3 to 4 d for a total of 6 collections per dog. Ten minutes before each collection, 1 of 3 injections (oxytocin 10 IU [0.5 mL], IM; PGF2alpha 2.5 mg [0.5 mL], IM; or saline 0.5 mL, IM) was administered. Compared to the saline controls, neither treatment had any significant effect on any measured variable when collected in this manner with an estrus bitch present. Therefore, the use of these drugs does not appear to be a viable treatment to increase the number of spermatozoa.

Objective—To determine the effect of finasteride on programmed cell death (apoptosis) of prostatic cells during prostatic involution in dogs with benign prostatic hypertrophy (BPH). Animals—9 dogs with BPH. Procedure—Dogs were randomly assigned to treatment or control groups. Treatment dogs (n = 5) were administered finasteride (0.1 to 0.5 mg/kg, PO, q 24 h) for 16 weeks, whereas the 4 control dogs were administered an inert compound. Prostatic cells from the prostatic fluid portion of the ejaculate of treatment and control dogs were obtained before and 1, 2, 3, 4, 8, and 16 weeks after initiation of treatment. Cells were concentrated by use of centrifugation. Prostatic cells were examined for indications of apoptosis by use of a terminal deoxyribonucleotidyl transferase- mediated deoxyuracil triphosphate nick-end labeling technique. After receiving the inert compound for 16 weeks, the 4 control dogs were administered finasteride for 16 weeks, and evaluations were repeated. Results—Percentage of apoptotic prostatic cells in ejaculated prostatic fluid of treatment dogs increased significantly (from 9% before treatment to 33, 31, 26, and 27% after 1, 2, 3, and 8 weeks of treatment, respectively). There was no significant change in percentage of apoptotic prostatic cells in the ejaculated prostatic fluid of control dogs. Conclusions and Clinical Relevance—Finasterideinduced prostatic involution appears to be via apoptosis in dogs with BPH. Finasteride treatment of dogs with BPH causes prostatic involution by apoptosis rather than necrosis. (Am J Vet Res 2002;63:495–498)

Objective—To determine the effect of the 5α-reductase inhibitor finasteride on prostatic diameter and volume, semen quality, and serum dihydrotestosterone (DHT) and testosterone concentrations in dogs with spontaneous benign prostatic hypertrophy (BPH). Design—Double-blind placebo-controlled trial. Animals—9 dogs with BPH. Procedure—Five dogs were treated with finasteride for 16 weeks (0.1 to 0.5 mg/kg [0.05 to 0.23 mg/lb] of body weight, PO, q 24 h); the other 4 received a placebo. Prostatic diameter, measured radiographically, prostatic volume, measured ultrasonographically, semen quality, and serum DHT and testosterone concentrations were evaluated before and during treatment. After receiving the placebo for 16 weeks, the 4 control dogs were treated with finasteride for 16 weeks, and evaluations were repeated. Results—Finasteride significantly decreased prostatic diameter (mean percentage decrease, 20%), prostatic volume (mean percentage decrease, 43%), and serum DHT concentration (mean percentage decrease, 58%). Finasteride decreased semen volume but did not adversely effect semen quality or serum testosterone concentration. No adverse effects were reported by owners of dogs in the study. Conclusions and Clinical Relevance—Results suggest that finasteride can be used to reduce prostatic size in dogs with BPH without adversely affecting semen quality or serum testosterone concentration.
