Photophysical properties of some azinium iodides in aqueous solution of nanostructured systems as DNA and nonionic micelles were investigated using steady-state and ultrafast time-resolved spectroscopy. Spectrophotometric and fluorimetric titrations of the investigated compounds with salmon testes DNA supplied evidence of a good interaction between the salts and DNA with binding constants of 10(4)-10(6) M(-1), making them interesting for pharmaceutical applications. The interaction with DNA also changes the photobehavior of the compounds, increasing the radiative deactivation pathway to the detriment of internal conversion and slowing down the excited state dynamics. The interaction of the azinium salts with the nonionic surfactant Triton X-100 from premicellar to postmicellar concentration was studied by spectrophotometric and fluorimetric titrations evidencing the ability of the micelles to associate the studied salts in their hydrophobic portion and to release them in the presence of DNA, acting as promising drug carriers. Also transient absorption spectroscopy with femtosecond resolution demonstrated the insertion of the investigated compounds into micellar aggregates. Preliminary measurements by confocal fluorescence microscopy on MCF-7 cells in the presence of the studied azinium salts showed that they are able to cross the cellular membrane and that their cytotoxicity can be expressed through interaction with DNA (RNA). In fact, they showed a significant fluorescence signal in all cell compartments, particularly (for 2 and 3) into punctuate structures within the nuclei compatible with a localization into the nucleoli.

The interaction of cationic amphiphiles with calf thymus DNA has been investigated by physicochemical techniques (surface tension, conductometry, UV spectroscopy, thermal denaturation) and morphological microscopies (AFM and TEM). The cationic molecules were the amphiphiles cetyltrimethylammonium and cetyltributylammonium bromides (CTAB and CTBAB, respectively), compared to the nonamphiphilic tetramethyl- and tetrabutylammonium bromides (TMAB and TBAB, respectively) and, as a transfection-efficient comparison, a commercial poliethyleneimine (PEI). As a result, well below their critical micelle concentrations (cmc), CTAB and CTBAB showed a peculiar, nonlinear adsorption profile with the nucleic acid, which showed a correlation with the melting temperatures and morphological changes observed with AFM and TEM microscopies. On the other hand, TMAB and TBAB interact much less with the DNA duplexes and do not induce any modifications of the structures. The same behavior was observed with PEI; however, CTAB and CTBAB proved much less effective in condensing the nucleic acid.

Here we describe uridine functionalization in the 5' position, which provides new classes of cationic and nonionic amphiphiles specifically designed as DNA transfection agents. The synthetic procedures developed to obtain the cationic uridine-head surfactants prevented intramolecular cyclization that occurs when uridine is functionalized in this position without using protecting groups in the uracil.

The use of Hoechst 33258 (HO) as a fluorescent probe to characterize the interactions between DNA and pH-sensitive amphiphiles is discussed. In the case of amine oxide amphiphiles dodecyldimethylamine oxide (DDAO) and p-dodecyloxybenzyldimethylamine oxide (pDoAO), the decrease in fluorescence emission, which signals DNA-amphiphile association, is accompanied by a large hypsochromic shift in the emission maximum of the bound probe; this eventually reaches a value characteristic of HO in a neutral or slightly basic environment. These findings are compared to results obtained by using the more common ethidium bromide (EB) probe, which shows no such shift. Circular dichroism and fluorescence depolarization experiments indicate that fluorescence emission only occurs from the DNA-bound probe, and the observed shift in emission maximum when using HO as a fluorescent probe is due to a variation in the local pH in the vicinity of the probe.
