The poultry red mite, Dermanyssus gallinae, is one of the most economically deleterious ectoparasites affecting egg-laying hens in many parts of the world. New approaches to control D. gallinae often require the maintenance of colonies of D. gallinae under laboratory conditions. In the present study, we present an efficient rearing system for D. gallinae , consisting of a metal cage, a plastic storage box and a tray filled with water. Chicks were raised in the cage as host animals. A novel trap was developed to monitor the dynamic changes of mite populations, made with a plastic centrifuge tube and a disposable breathing mask with folds. Mite parameters were analyzed, including number of mites and eggs, survival and feeding rates, oviposition, hatchability and the proportion of D. gallinae at different life stages. The results show that the rearing system had a 53.5-fold increase in the number of mites over a period of six weeks after the introduction of mites. The survival rates of mites were above 94%, and the mean feeding rates ranged from 22.57% to 37.30%. The mean number of eggs per female ranged from 3.42 to 3.50, with the hatchability of eggs above 97%. Nymphs made up most of the population, ranging from 71.46% to 81.37%, while the population of larvae was minor and ranging from 7.54% to 13.04%. The mask trap used in this study was an effective and convenient device to shelter D. gallinae and monitor the dynamic changes of the mite population. The rearing system proved very effective in maintaining and reproducing colonies of D. gallinae , with great potential for the evaluation of the efficacy of vaccines or compounds against D. gallinae under laboratory conditions. Background: The soil-transmitted nematodes (STNs) or helminths (hookworms, whipworms, large roundworms) infect the intestines of ~1.5 billion of the poorest peoples and are leading causes of morbidity worldwide. Only one class of anthelmintic or anti-nematode drugs, the benzimidazoles, is currently used in mass drug administrations, which is a dangerous situation. New anti-nematode drugs are urgently needed. Bacillus thuringiensis crystal protein Cry5B is a powerful, promising new candidate. Drug combinations, when properly made, are ideal for treating infectious diseases. Although there are some clinical trials using drug combinations against STNs, little quantitative and systemic work has been performed to define the characteristics of these combinations in vivo. Methodology/Principal findings: Working with the hookworm Ancylostoma ceylanicum-hamster infection system, we establish a laboratory paradigm for studying anti-nematode combinations in vivo using Cry5B and the nicotinic acetylcholine receptor (nAChR) agonists tribendimidine and pyrantel pamoate. We demonstrate that Cry5B strongly synergizes in vivo with both tribendimidine and pyrantel at specific dose ratios against hookworm infections. For example, whereas 1 mg/kg Cry5B and 1 mg/kg tribendimidine individually resulted in only a 0%-6% reduction in hookworm burdens, the combination of the two resulted in a 41% reduction (P = 0.020). Furthermore, when mixed at synergistic ratios, these combinations eradicate hookworm infections at doses where the individual doses do not. Using cyathostomin nematode parasites of horses, we find based on inhibitory concentration 50% values that a strongylid parasite population doubly resistant to nAChR agonists and benzimidazoles is more susceptible or “hypersusceptible” to Cry5B than a cyathostomin population not resistant to nAChR agonists, consistent with previous Caenhorhabditis elegans results. Conclusions/Significance: Our study provides a powerful means by which anthelmintic combination therapies can be examined in vivo in the laboratory. In addition, we demonstrate that Cry5B and nAChR agonists have excellent combinatorial properties—Cry5B combined with nAChR agonists gives rise to potent cures that are predicted to be recalcitrant to the development of parasite resistance. The raccoon ( Procyon lotor ) is a North American carnivore introduced to Europe in the 20th Century. Raccoons are believed to be the potential hosts of many parasites, or to be involved in their transmission to other animals. Nematodes of the genus Trichinella can infect many carnivorous and omnivorous animals worldwide. The aim of the present study was to determine the occurrence of Trichinella spp. infection in raccoons in Central Europe. Muscle samples were collected from various regions of Poland, the Czech Republic and Germany during the years 2012–2016. The larvae of Trichinella spp. were detected in 11 raccoons, and these were identified as T. spiralis and T. pseudospiralis by multiplex PCR (89.9% and 9.1%, respectively). No mixed infection was observed. This is the first report describing the occurrence of T. spiralis and T. pseudospiralis in P. lotor in Central Europe. All Theileria parasites have definitive natural hosts that act as carriers. Incidental infections of uncommon hosts do occur raising questions regarding host specificity and its drivers. Reported hosts for Theileria taurotragi include bushbuck, cattle and eland. More recently T. taurotragi was detected in African buffalo, which may have implications for accurate diagnostics of T. parva . The current study therefore investigated the host specificity of T. taurotragi by developing a specific and sensitive real-time Taqman PCR assay. Animals were screened from areas where Rhipicephalus appendiculatus , the tick vector for both T. parva and T. taurotragi was present. While T. taurotragi was detected in cattle, eland, kudu and nyala, African buffalo (n = 352) was negative. Conversely, these same buffalo showed a prevalence of 72–100% for T. parva . While transmission of T. taurotragi to cattle was successful using the same infected tick batch, transmission to African buffalo did not occur. The results suggest that African buffalo is not a natural host of T. taurotragi and would probably not harbor anti-schizont antibodies against T. taurotragi . This would preclude T. taurotragi as possible source of cross-reactivity in the T. parva immunofluorescent antibody test. Host specificity of T. taurotragi for members of the Tragelaphini, but not buffalo also suggests that host specificity may have been an important driver in the speciation of the T. taurotragi clade. Zoonotic visceral leishmaniasis (ZVL) is a parasitic disease affecting dogs and humans, which is transmitted by female sandflies. Over the last decade, disease prevalence has increased fivefold in parts of southern Europe, where an estimated 2.5 million dogs are infected. This increase is mainly due to an expansion in sandfly distribution due to climate change and to the greater numbers of dogs travelling among European countries. To combat the spread of ZVL in Europe, international guidelines have been drawn up that describe strategies to prevent, control and monitor the disease. To investigate whether these strategies are being implemented in the field, we conducted an online survey among veterinarians in Spain (endemic for ZVL) and France (south: emerging; north: non-endemic). Of the 889 respondents, 459 veterinarians completed all questions. Although 60% of all veterinarians were aware of the current ZVL increase in Europe, 70% were not familiar with any guidelines for controlling the disease. Most of their preventive and treatment actions were, however, in line with intervention strategies recommended by the guidelines. From the veterinarians in this survey, 76% had received no reports regarding confirmed cases of canine leishmaniosis (CanL) or human visceral leishmaniasis in their region or country. The fact that 88% of confirmed cases of clinical CanL were not reported suggests inadequate disease monitoring and evaluation. We therefore recommend that an easy-to-use and accessible international online network be developed, where both veterinarians and physicians can report confirmed cases of leishmaniosis in dogs and humans. Filarial nematodes of the Eulimdana genus inhabit subcutaneous tissue of various avian species, mostly Charadriiforme birds. In domestic pigeons, E. clava is the only species recorded in the subcutaneous tissue in a number of isolated cases. In the present study, we discuss the morphology and histopathology of filarial nematodes recovered from subcutaneous tissue of domestic pigeons in Bosnia and Herzegovina. In total 110 pigeons were submitted to necropsy at the Department of Pathology of the Faculty of Veterinary Medicine in Sarajevo. At necropsy, in four pigeons (3.6%) numerous thread-like 0.9–2.1 cm long nematode parasites were observed in the subcutaneous tissue, peritracheal and periesophageal connective tissue. In one pigeon, the parasites were also found free in the body cavity around the heart and lungs. In addition, several 80–90 μm long microfilariae were noted in the tissue cross-sections. No significant lesions were observed associated with adult parasites or microfilariae. Based on morphology, host species and localization detected parasites were identified as E. clava . Molecular analyses of the cox 1 and 12S rRNA nucleotide sequences herein generated revealed the close genetic relationship to other filarioid nematodes. The importance of the nematodes in pigeons and the lack of sequences in genetic databases for comparison of avian filarial parasites are emphasized. [ABSTRACT FROM AUTHOR]

Aim: Parasitic infections, especially of the zoonotic-parasitic type, are the most important health, economic, and social problems in developing countries, including Iran. The aim of this study was to review systematically the available data on gastrointestinal parasites of carnivores in Iran and their ability to infect humans. Materials and Methods: Studies reporting intestinal parasites of carnivores were systematically collected from nine electronic English and Persian databases and Proceedings of Iranian parasitology and veterinary congresses published between 1997 and 2015. A total of 26 studies issued from 1997 to 2015 met the eligibility criteria. Results: The pooled proportion of intestinal parasites of carnivores was estimated as 80.4% (95% confidence interval=70.2-88.8%). The overall prevalence of gastrointestinal parasites in dogs, cats, foxes, and jackals were 57.89%, 90.62%, 89.17%, and 97.32%, respectively. Dipylidium caninum (20.45%), Toxocara spp. (18.81%), Taenia hydatigena (15.28%), Mesocestoides lineatus (11.83%), Echinococcus granulosus (10%), and Toxascaris leonina (8.69%) were the most frequently observed parasites. Conclusion: High prevalence rates of zoonotic intestinal parasites of carnivores particularly Echinococcus spp. and Toxocara spp. increase the risk of acquiring zoonotic infections such as cystic hydatid, alveolar cysts, and visceral or ocular larva migrants in Iranian people. In the period from January 2004 to December 2015, 56 dogs were diagnosed with rangeliosis in the Setor de Patologia Veterinária at Universidade Federal do Rio Grande do Sul (SPV-UFRGS). The main hematological abnormalities were thrombocytopenia and anemia. The affected dogs showed signs of apathy, anorexia, fetid and bloody diarrhea, vomiting, and dehydration. At necropsy, the main changes were jaundice, splenomegaly, hepatomegaly, and lymphadenomegaly. Histological analyses revealed parasitophorous vacuoles of Rangelia vitalii in cytoplasmic endothelial cells, mainly in the heart, kidneys, lymph nodes, intestines, and pancreas. Inflammation characterized by mononuclear cells was predominant in the analysis, and most was due to the presence of plasma cells. Other lesion types observed were lymphoid hyperplasia, extramedullary hematopoiesis, erythrophagocytosis, and erythroid lineage hyperplasia in bone marrow. Of the total number of animals, 49 were diagnosed using necropsy and histological analysis, and seven were diagnosed using a molecular analysis (i.e., PCR and genetic sequencing of blood samples). This paper presented a different method of diagnosing rangeliosis in canines. Infection patterns of the invasive Anguillicola crassus nematode were investigated in a population of the European eel Anguilla anguilla where parasite invasion is very recent , Loch Lomond, Scotland. Intensity levels of the parasite were associated with differences in fish ontogeny and trophic ecology. Although eels foraged on both fish and invertebrates, individuals which were smaller and fed on invertebrates (>70% contribution to diet) were found to contain a greater number of swim bladder parasites compared to larger eel with a predominance of fish (>60% contribution) in their diet. Within affected fish, a significant negative relationship was found between fish length and parasite intensity, with smaller individuals having higher parasite intensity than larger individuals. This study indicates that food intake and infection risk are linked in this recently infected host-parasite system. From a management perspective increasing our understanding of how infection intensity and repeated exposure is linked to resource use in an ecosystem is important for the future management of this endangered species in Europe. Sarcoptic mange is a highly contagious zoonotic skin disease that can have severe effect on population dynamics of many wild mammals. However, very little is known about its effect on the activity and space use of infected animals. In this study we equipped two raccoon dogs ( Nyctereutes procyonoides ) in a mange outbreak area with radio-collars and observed the spatial and temporal effects of sarcoptic mange to these individuals. Initially, the raccoon dogs had no external symptoms of mange infection, but developed these during the study period. One of the raccoon dogs died just 32 and the other 52 days after collaring. During a relatively short period before their death, there was an abrupt and drastic decline in their home range size. For one of the animals it started about 1 month before its death and the home range size reduced >1000×, whereas for the other raccoon dog it took place within the last week, decreasing >200×. The daily covered distances also declined considerably. These results indicate that at the later stage sarcoptic mange affected the physiological state of the diseased animals so forcefully that made them almost immobile. Our results show for the first time how rapid and severe could the impact of sarcoptic mange be for one of its most important reservoir and vector species. [ABSTRACT FROM AUTHOR]

Background: Diseases associated to external parasitosis are scarcely reported in sea turtles. During the last decades several organism have been documented as a part of normal epibiont community connected to sea turtles. The copepod Balaenophilus manatorum has been cited as a part of epibiont fauna with some concern about its parasitic capacity. This study serves three purposes, i.e. (i) it sheds light on the type of life style that B. manatorum has developed with its hosts, particularly turtles; (ii) it makes a cautionary note of the potential health risks associated with B. manatorum in sea turtles under captivity conditions and in the wild, and (iii) it provides data on effective treatments against B. manatorum. Results: We report for the first time a massive infestation of the copepod B. manatorum and subsequent acute mortality in a group of loggerhead sea turtle hatchlings. Four-month-old turtles from a head-starting program started exhibiting excitatory and fin rubbing behavior preceding an acute onset of lethargy, skin ulceration and death in some animals. All the individuals (n = 57) were affected by severe copepod load and presented different degrees of external macroscopic skin lesions. The ventral area of front flippers, axillar and pericloacal skin were mostly affected, and were the main parasite distribution regions. Copepods were also detected on plastron and carapace sutures. The gut contents of B. manatorum reacted positively for cytokeratin, indicating consumption of turtle skin. Severe ulcerative necrotic dermatitis and large amount of bacteria presence were the major histopathological findings. Conclusions: Individual fresh water immersion for 10 min and lufenuron administration (0.1 ppm) to the water system every 2 weeks proved effective for removing turtle parasites and to control re-infestation, respectively. The results from our study clearly indicated that B. manatorum individuals consume turtle skin. Background: Anecdotal reports support the position that the adulticidal heartworm treatment utilizing doxycycline and Advantage Multi®/Advocate® for Dogs (10% imidacloprid + 2.5% moxidectin) has successfully converted antigen-positive dogs to antigen-negative. To date, no controlled experimental studies have demonstrated the adulticidal efficacy of this treatment regimen. The aim of this study was to evaluate the parasitological and clinical efficacy of Advantage Multi® for Dogs (IMD + MOX) and doxycycline in heartworm-infected beagles. Methods: This study utilized 16 dogs, 8 dogs in each of non-treated control and treated groups. A total of 16 adult Dirofilaria immitis (Missouri strain) were surgically transplanted into the jugular vein of each study dog. The treatment regimen of monthly IMD + MOX topically (per labeled dosage and administration) for 10 months and 10 mg/kg doxycycline BID orally for 30 days was initiated 30 days post-surgical transplant. Echocardiograms, radiographs, complete blood counts, clinical chemistry profiles, heartworm antigenemia and microfilaremia were evaluated every 4 weeks. Serum samples were assayed for heartworm antigen using the DiroCHEK® heartworm antigen test. The DiroCHEK® was performed according to the manufacturer's recommendations and read using a spectrophotometer at 490 nm. Results: All dogs tested positive for the presence of heartworm antigen post-surgical transplant and prior to treatment. Heartworm antigen levels began declining in treated dogs 3 months post-treatment. Non-treated control dogs remained antigen-positive. No microfilariae were detected in treated dogs after 21 days post-treatment. At necropsy, adult heartworms were recovered from all non-treated control dogs with a range of 10-12 adult worms/dog for an average recovery of 10.6 adult heartworms/dog. In the IMD + MOX- and doxycycline-treated dogs, the range of adult heartworms recovered was 0-2 adult worms/dog, with five dogs having no adult heartworms present. The average adult heartworm recovery was 0.6/dog in the treated group. This treatment regimen demonstrated a 95.9% efficacy in eliminating adult heartworms (P < 0.0001). Conclusions: This study demonstrated that this treatment regimen successfully eliminated D. immitis microfilariae by 21 days post-treatment, reduced heartworm antigen concentration over time, and had a 95.9% efficacy in the elimination of mature adult heartworms. Based on this study, we conclude that this treatment regimen is a relatively quick, reliable and safe option to treat canine heartworm infection as compared to other treatment regimens involving macrocyclic lactones, when the approved drug melarsomine dihydrochloride is unavailable, contraindicated or declined by an owner unable to afford the more costly treatment or concerned about the potential side effects. Proliferative kidney disease (PKD) is an endoparasitic disease of salmonid fish caused by Tetracapsuloides bryosalmonae (Myxozoa: Malacosporea). This study presents a comprehensive view on PKD development in rainbow trout ( Oncorhynchus mykiss ) reared at an intensive fish breeding facility, with focus on mortality, pathology/histopathology, haematological findings and immune functions. Diseased and reference fish were sampled monthly and time course of natural infection was followed up from the onset of clinical signs (September 2014) to full recovery (January 2015). PKD- associated cumulative mortality was 30% with a peak value in October, while immunohistochemical testing indicated a continuous significant decrease in T. bryosalmonae numbers from September to December; with no parasites detected in January. During peak clinical infection, a significant decrease in red blood cell counts, haematocrit values, haemoglobin concentration, along with a reduction in lymphocytes and a significant phagocyte elevation corresponding with an increase in phagocyte oxidative burst were measured in comparison to control animals. Complement activity and total immunoglobulin plasma concentrations were also elevated, though only during the initial monitoring period (September). Individuals surviving PKD, recovered and were able to fully regenerate both renal structure and haematopoietic parameters to normal levels. Changes in the red blood cell parameters indicate anaemia and a decreased oxygen transportation capacity during the clinical disease phase. Together with an increased oxygen demand at higher temperatures and decreased oxygen solubility this could lead to decompensation and elevated mortality. The stimulation of immune parameters, and especially oxidative phagocytic burst, is likely to have had a strong effect on both, regeneration and elimination of the pathogenic agent. Fasciola hepatica infestation is reported as a cause of severe eosinophilic cholangiohepatitis in a 28-year-old Warmblood cross pony gelding. The gelding presented initially for investigation of acute laminitis found to be secondary to pars pituitary intermedia dysfunction ( PPID). Six weeks of treatment including administration of pergolide achieved good control of the laminitis but there was a marked general clinical deterioration over this period with notable weight loss, tachycardia, anorexia and ventral oedema. Hepatobiliary enzymes were increased significantly and there was mild elevation of plasma bile acid concentration. An eosinophilia in both blood and peritoneal fluid specimens and histopathological confirmation of an eosinophilic cholangiohepatitis on examination of liver biopsies supported a primary parasitic aetiology. The pony was treated for Fasciola hepatica with triclabendazole 12 mg/kg bwt but clinical signs continued to progress. Due to poor clinical response the pony was subjected to euthanasia and post mortem examination revealed an overwhelming infestation of the biliary tracts with Fasciola hepatica and a secondary chronic and severe cholangiohepatitis with marked fibrosis. To the authors' knowledge this is the first equine report demonstrating the relationship between the clinical disease syndrome, eosinophilic cholangiohepatitis and hepatic fluke infestation. Concurrent infection of Leptospira interrogans, Babesia gibsoni, and Dirofilaria repens in a properly vaccinated and dewormed Labrador retriever dog and its successful clinical management are described. The dog was presented with a history of anorexia, vomiting, lethargy, brownish yellow urine, frequent shivering, and weakness. Clinical and laboratory investigations revealed pyrexia, spleenomegaly, icterus, lymphadenopathy, dehydration, hemoglobinuria, anemia, neutrophilia, thrombocytopenia, and hyperbilirubinemia. Concurrent infection of Leptospira interrogans var pyrogenes and autumnalis, Babesia gibsoni and Dirofilaria repens, was confirmed by dark field microscopy of urine sample and blood smear examination, respectively. The animal was stabilized and undergone a combination therapy using Metronidazole (20 mg/kg bid i/v) and Clindamycin (12 mg/kg bid i/v) for 7 days, Doxycycline (5 mg/kg bid po) for 21 days, single dose of Ivermectin was given at 100 μg/kg orally on day 7. Supportive therapy using plasma volume expander, polyionic electrolytes, antiemetics, hepatotonics, vitamin supplements, and hematinics was provided during the course of treatment. The animal made a dramatic recovery by 3 months of care. The case point towards the need of selection of appropriate clinical materials for differential diagnosis, the control of mosquito borne diseases based on ecological characteristics of vector, and the need of incorporating emerging virulent serovars of leptospira in vaccines. Toxoplasma gondii , an obligate intracellular protozoan parasite, is responsible for one of the most common zoonotic parasitic diseases in almost all warm-blooded vertebrates worldwide, and it is estimated that about one-third of the world human population is chronically infected with this parasite. Little is known about the circulation of T. gondii in snakes and this study for the first time aimed to evaluate the infection rates of snakes by this parasite by PCR methods. The brain of 68 Snakes, that were collected between May 2012 and September 2015 and died after the hold in captivity, under which they were kept for taking poisons, were examined for the presence of this parasite. DNA was extracted and Nested-PCR method was carried out with two of pairs of primers to detect the 344 bp fragment of T. gondii GRA6 gene. Five positive nested-PCR products were directly sequenced in the forward and reverse directions by Sequetech Company (Mountain View, CA). T. gondii GRA6 gene were detected from 55 (80.88%) of 68 snakes brains. Sequencing of the GRA6 gene revealed 98–100% of similarity with T. gondii sequences deposited in GenBank. To our knowledge, this is the first study of molecular detection of T. gondii in snakes and our findings show a higher frequency of this organism among them. [ABSTRACT FROM AUTHOR]

Background: Infection of dogs with the cardiopulmonary nematode Angiostrongylus vasorum may result in severe clinical disease therefore adequate prevention is necessary. A randomized, negative control, blinded study was conducted to evaluate the efficacy in the prevention of canine A. vasorum infection after monthly administrations of NexGard Spectra®, a novel chewable tablet formulation combining the insecticide and acaricide afoxolaner and the anthelmintic milbemycin oxime, in a multiple challenge (trickle infection) model. Methods: Twenty beagle dogs were challenged orally with doses of approximately 32–43 third-stage larvae of A. vasorum once every other week on seven occasions (Study Days -7, 7, 21, 35, 49, 63 and 77). Ten dogs were administered NexGard Spectra® as close as possible to the minimum recommended dose of afoxolaner and milbemycin oxime, i.e. 2.5 mg/kg body weight and 0.5 mg/kg body weight, respectively, four times at monthly intervals (Study Days 0, 28, 56 and 84) while the remaining ten dogs served as untreated controls. For parasite recovery and count, dogs were euthanized humanely and necropsied six to eight days following the last treatment (Study Days 90–92). Beginning six weeks after first inoculation, faeces were collected on a bi-weekly basis and examined for first-stage larvae of A. vasorum. Results: Untreated dogs harboured 39–95 adult A. vasorum (geometric mean, 66.4), while zero to 24 adult A. vasorum were recovered from the treated dogs (geometric mean, 3.4; P < 0.0001). Thus, efficacy of NexGard Spectra® administered at monthly intervals against incoming A. vasorum was 94.9 %. Compared to the untreated controls, larval excretion of the treated dogs was reduced by 99.9 % (P < 0.0001). Cryptosporidium is a protozoan parasite that infects a wide range of hosts, yet relatively little is known about the epidemiology of cryptosporidiosis in fish. Here we report a disseminated Cryptosporidium infection in a male Koi carp ( Cyprinus carpio ), with parasite stages identified deep within the epithelium of the intestine, kidneys, spleen, liver and gills causing severe granulomatous inflammatory lesions. Molecular characterization at two loci; 18S ribosomal RNA (rRNA) and actin, revealed this to be a novel Cryptosporidium genotype, most closely related to Cryptosporidium molnari . Cryptocaryoniasis is a severe parasitic disease caused by ciliate Cryptocaryon irritans in farmed marine fish. To control this disease, we established a method to interrupt the life cycle of C. irritans via the removal of its tomonts. Marine fish Plectorhinchus cinctus were divided randomly into four groups, including placemat removal group (Group I), rotational culturing group (Group II), infection control group (IF group) and blank control group (Control); the former three groups were infected with C. irritans at low dose. A daily food consumption (DFC), relative infection intensity (RII) and survival rate of each group were observed. At 4-week post infection, some fish were challenged with C. irritans theronts at a lethal dose. Results showed that the fish' DFC in IF Group, Group I and II decreased significantly on 2nd day post infection, and gradually restored food intake when the trophonts exited the fish on the 5th day. However, their DFC in IF Group decreased significantly on 7th day, and eventually stopped feeding. Trophonts on fish' body of the three groups began to reduce from the 4th day and those of Group I and II almost disappeared on the 7th day, conversely the trophonts of IF group increased ten times. At 2-week post infection, the survival rates of Group I, II, IF and Control were 97%, 98%, 0%, and 100%, respectively. The survival rates of Group I, II and Control were 88.3%, 93.3%, 66.7%, respectively after challenge. Results demonstrate that placemat and rotational culturing method to remove tomonts can effectively control C. irritans infection. Aim: The study was conducted to know the prevalence of gastrointestinal parasites of captive wild animals at Bir Moti Bagh Mini Zoo (Deer Park), Patiala, Punjab. Materials and Methods: A total of 31 fecal samples from eight species of captive animals including Civet cat (Viverra zibetha), Porcupine (Hystrix indica), Nilgai (Boselaphus tragocamelus), Spotted deer (Axis axis), Black buck (Antelope cervicapra), Sambar deer (Cervus unicolor), Hog deer (Axis porcinus), and Barking deer (Muntiacus muntjak) were screened using classical parasitological techniques including sedimentation and floatation technique. Results: Out of 31 fecal samples examined, 20 were positive for parasitic ova/oocysts of different species indicating an overall prevalence of 68.0%. The six different types of parasites observed in the study included strongyle (67%), Strongyloides spp. (14%), coccidia (38%), Trichuris spp. (19%), ascarid (10%), and Capillaria spp. (10%). Strongyles were the most common parasites observed (67%) followed by coccidia (38%). Mixed helminth and protozoan infection were observed in 48% of animals. No cestode or trematodes were detected during the study. Conclusion: The high prevalence of gastrointestinal parasites without overt clinical signs of disease or mortality as observed in this study is suggestive of subclinical infection. Out of 92 donkeys examined for gastrointestinal parasites, 90 animals were found infected by one or more gastrointestinal parasites with an overall prevalence rate of 97.78%. The distributions of the recovered parasites in the different parts of the body were as follows: stomach, 92.4%, small intestine, 19.6%, caecum, 88%, colon, 80.4%, rectum, 73.9%, and cranial mesenteric artery, 64.1%. A significant difference was found between mean parasite counts and seasons. Hot wet season had higher mean parasites count (5411.5±1694.4) in comparison with hot dry (1795.9±399.6) and cool dry (1719.9±522.4) seasons. Although there was no significant difference between age and mean parasite count, animals more than four years old had high mean count (3361.3±921.8) in comparison with 2330±744.3 and 2030.2±873.1 for young and adults animals, respectively. No significant positive or negative correlation was found between total parasite counts of infected animals and any of the climatic factors. The parasites identified were Habronema spp. (40.2%), Trichostrongylus axei (30.4%), Parascaris equorum (18.5%), Anoplocephala perfoliata (4.35%), Gastrodiscus aegyptiacus (8.7%), large strongyles (84%), small strongyles (72%), and Oxyuris equi (1.1%). Neosporosis is a parasitic disease caused by the protozoan Neospora caninum which results in major economic losses for cattle breeding due to abortion and other reproductive disorders. Gerbils (Meriones unguiculatus) are commonly used as experimental models for neosporosis due to their high susceptibility to N. caninum infection, both by oocysts ingestion as by tachyzoites/bradyzoites parenteral inoculation. However, the risk of transmission by tachyzoites ingestion is not fully elucidated. In this study, infection of neonate gerbils by N. caninum (NC-1 strain) tachyzoites inoculated by the oral route and the parasite distribution in gerbils' tissues were evaluated by protozoan DNA detection. Seventeen neonate gerbils, aged 4-5 days, were inoculated with 4x1 05 tachyzoites by the oral route and one gerbil was kept as uninfected control. N. caninum DNA was detected in 100% of the inoculated gerbils, showing that the oral route is effective as a potential route of infection of neonates by N. caninum tachyzoites. N. caninum DNA was reported in all organs evaluated (heart, lungs, kidneys, liver, spleen and brain), with different frequencies. Wildcats are endangered felid species living in Europe, Asia, and Africa. Regrettably, scientific information on parasites of wildcats is particularly meager and they often rely on data gained by necropsies of a small number of animals. In the present study, scat collection was used to assess the parasite spectrum of European wildcats living in the Etna Park (Sicily, Italy). Scat collection was performed from May to September 2010 by weekly walking four transects for a total of 391 km. Samples were then analyzed by flotation and sedimentation techniques to investigate wildcat parasitic fauna. A total of 121 scats of wildcats were collected, and parasitic forms (i.e., oocysts, eggs, and larvae) were retrieved in 110 (90.9 %) of the samples. Parasites found were Physaloptera sp. (52.1 %), tapeworms (45.5 %), Toxocara cati (43.8 %), Eucoleus aerophilus (27.3 %), Ancylostoma sp. (22.3 %), Troglostrongylus brevior (15.7 %), trematodes (9.9 %), Isospora felis (4.1 %), Cylicospirura sp. (1.7 %), and Acanthocephala (0.8 %). The prevalence of endoparasitic infections herein recorded is similar to that described in other studies conducted using necropsy technique. The species richness of parasites found in the present survey, with a total of nine helminths and one protozoon, is the highest ever reported for wildcat in Europe. Scat collection and examination are reliable and rapid non-invasive tools which can be used in a systematic survey design to study the parasite spectrum of wildcat as well as that of other endangered wild species. Angiostrongylus vasorum is a parasitic nematode that infects the heart and pulmonary artery and its branches of domestic and wild canids. The parasite can use several species of terrestrial and aquatic molluscs as intermediate hosts, although susceptibility varies. Pomacea canaliculata is a mollusc found in lakes, swamps and rivers in South America. In this study, we evaluated the susceptibility, parasite growth, oviposition and larval development of 282 P. canaliculata infected with 500 A. vasorum first-instar larvae (L1). From day 5 post-infection (pi) to day 30 pi, seven specimens per day were sacrificed to recover the larval instars. We compared 50 egg masses from infected and uninfected molluscs to determine the number of eggs per clutch, the hatching rate and the growth of the molluscs. The percentage of recovered larvae ranged from 39.17% to 67.5%. First-stage larvae (L1) were found until day 19 pi, second-stage larvae (L2) were found from days 11 to 25 pi, and third-stage larvae (L3) were recovered only after day 19 pi. Infected snails exhibited the most eggs during spawning, although the rate of hatching and shell size were lower in the infected snails compared with controls. This is the first report of an experimental infection of P. canaliculata with A. vasorum, and the results confirm the non-specificity of the nematode in relation to the intermediate host and indicate the importance of epidemiological surveys of this parasite and mollusc. The work describes a case of Sarcocystis gigantea infection in a 3-years-old Corriedale ewe from Buenos Aires Province, Argentina. The ewe was found dead with a poor body condition. Pathological and molecular studies were carried out in order to try and confirm the causative agent of the infection. At necropsy, approx. 100 whitish elliptic (3–5 mm to 5–8 mm) macrocysts with a hard consistency were observed along the esophageal and pharyngeal muscular layers. Microscopically, the macrocysts consisted of an eosinophilic wall, internal septa originated from the eosinophilic wall and basophilic parasitic cells were located among the septa. The sarcocysts were identified molecularly through PCR amplification and sequencing of a short segment of the 18S rRNA gene. Sequence analysis of the amplified DNA demonstrated 100% identity to S. gigantea sequences previously published. Current knowledge on bovine besnoitiosis, caused by the emerging apicomplexan pathogen Besnoitia besnoiti, is still fragmentary. So far, studies dealing with ultrastructural pathology focused mainly on the easily accessible chronic stage, whereas ultrastructural investigations of tachyzoites were confined to in vitro studies. In the study presented here, the ultrastructural pathology of naturally B. besnoiti-infected cattle in the acute and chronic disease stages and experimentally B. besnoiti-infected mice was monitored. Further, the ultrastructure of tachyzoites and bradyzoites was investigated. Skin samples of two adult Limousin cows and one adult Limousin bull naturally infected with B. besnoiti and liver and skin samples of gamma-interferon knockout mice infected with B. besnoiti were examined in semithin sections stained with toluidine blue and safranin and in ultrathin sections contrasted with uranyl acetate and lead citrate. Samples of vessel walls of the bull and nasal mucosa of one cow were examined by scanning electron microscopy. Few tachyzoites-like endozoites were detected for the first time in bovine skin, and large numbers of tachyzoites were detected in murine skin and liver. Within tissue cysts in bovine skin, numerous bradyzoites were observed displaying signs of degeneration. Tachyzoites had apicomplexan endozoite ultrastructure. B. besnoiti tachyzoites and bradyzoites differed in shape and the number of amylopectin granules. Transmission and scanning electron microscopy confirmed the presence of two different cyst wall layers, and the present results on cyst wall ultrastructure were in accordance with those previously obtained by histological sections. Little information is available on the occurrence of endoparasites in pet guinea pigs ( Cavia porcellus). The purpose of this study was to evaluate the prevalence of intestinal parasites in cavies kept as pets in southern Italy. Fresh fecal samples were randomly collected from 60 guinea pigs housed in pet shops or privately owned. All fecal samples were processed using the FLOTAC pellet technique to identify and count helminthic eggs/larvae and protozoan cysts/oocysts. In addition, the specimens were analyzed also by the Remel Xpect® Giardia/Cryptosporidium immunoassay. Intestinal parasites were detected in 19 out of 60 guinea pigs (31.7 %). Paraspidodera uncinata eggs were found in 13.3 % (8/60) of the rodents examined, Nippostrongylus-like eggs in 10 % (6/60), and finally Eimeria caviae oocysts were found in 10 % (6/60) of the animals. In one case, both E. caviae oocysts and P. uncinata eggs were found. None of the samples was positive for Cryptosporidium or Giardia. The purpose of the present study was to determine the prevalence of tick infestation in buffalo and goats of Punjab Province (District Sargodha), Pakistan. There were a total of twelve hundred buffalo and goats examined from October 2012 to September 2013 to determine the prevalence of ticks infestation. The results showed that the prevalence of tick infestation in buffalo was 84.33% (1012/1200). The ticks were collected and identified. In buffalo, the higher prevalence rate was Hyalomma aegyptium (37.91 %) followed by Rhipicephalus (Boophilus) microplus (22.59%), Rhipicephalus (Boophilus) annulatus (17.15%), Hyalomma marginatum (9.45%), Hyalomma anatolicum (9.03%) and Rhipicephalus sanquineus (3.84%) respectively. The breed wise prevalence was in Chenab breed (84.59%), Ravi breed (7910%) and Hybrid breed (91.66%) respectively. The sex wise prevalence was in male (87.71%) and in female (83.53%). The prevalence based on age showed that the rate of infestation in buffalo having age group (1-4 year) was 85.48%, (4-8 year) was 86.02% and (>8 year) was 66.66%, respectively. The prevalence based on grazing pattern showed that rate of infestation in intensive grazing 85.28%, extensive grazing 86.41% and both (intensive and extensive) was 81.94%.While in case of goats the tick infestation was 86.50% (1038/1200). In goats, the higher prevalence was Hyalomma anatolicum (31.56%), Rhipicephalus spp. (25.95%), Haemophysalis spp. (21.07%), Ixodes spp (1546%) and Ambylomma spp. (5.93%) respectively. The breed wise prevalence was in Desi breed (91.12%), Taidi breed (90.97%), Lail Poori breed (91.78%) and Hybrid breed (66.66%) respectively. The sex wise prevalence was in male (79.16%) and in female (88.81%). The prevalence based on age showed that the rate of infestation in goats having age group (1-4 year) was 86.17%, (4-8 year) was 91.66%, respectively. The prevalence based on grazing pattern showed that rate of infestation in intensive grazing 86.71%, extensive grazing 85.97% and both (intensive grazing and extensive grazing) was 87.12%.The present study showed that these epidemiological factors have a significant effect on the prevalence of tick infestation in buffalo and goats of Sargodha district, Pakistan. The results of this survey showed that the ticks are active throughout the year except January but highly active from June to August. This study was carried out to assess the levels of parasitic infection in domestic ducks ( Anas platyrhynchas) in Gilan Province, Northern Iran. During the summer of 2015, samples were collected randomly from local markets at different villages in Gilan Province. Blood samples were collected in EDTA-treated tubes, blood smear slides created, stained with giemsa and examined for the presence of blood protozoa. Ectoparasites samples were collected and identified according to Wall and Shearer (2001). To assess endoparasitic infection, the ducks' gastrointestinal tracts were removed and the various parts identified and opened. Parasites were collected, fixed in methanol, cleared with lactophenol and stained with borax carmine. Collected parasites were identified according to the descriptions given by Soulsby (1982) and Yamaguti (1958a, b). 56.6 % of ducks were infected with protozoan parasites, with the most prevalent of which being Tricomonas gallinae (prevalence 23.3 %, intensity 18 ± 2.5) followed by Cryptosporidium bailey (prevalence 16.6 %, intensity 15.44 ± 16.5) and Eimeria oocyst (prevalence 16.6 %, intensity 17.56 ± 12.5). Also found on the feathers and body surface were two species of lice ( Menocanthus stranineus and Liperus squalidus), one species of tick ( Argas persicus) and one of flea ( Ctenocephalides canis). Prevalence and intensity of the parasite infestation varied from species to species: M . stranineus (prevalence 100 %, intensity 43.33 ± 17.5), L. squalidus (prevalence 100 %, intensity 39.20 ± 12.42), A. persicus (prevalence 75 %, intensity 35.24 ± 12.7) and C. canis (prevalence 60 %, intensity 30.16 ± 15.5). Helminthes parasite infection rate was 50 % in collected samples which consisted of Railletina tetragona (prevalence 26.6 %, intensity 19.34 ± 16.5), Heterakis gallinarum (prevalence 13.33 %, intensity 14.65 ± 12.5) and Capillaria (prevalence 10 %, intensity 12.56 ± 16.5). This study shows the prevalence of parasitic infection in domesticated ducks in Gilan Province, Iran. Findings indicate that applying protocols of management and applying preventative methods and treatment of infected cases are necessary to restrict parasitic infections in domestic ducks and other birds in this region. Pitted darkling beetles ( Adesmia cancellata) were infected with nematode eggs found in the alimentary tract of a gyrfalcon ( Falco rusticolus) naturally infected with Paraspiralatus sakeri. Third-stage larvae in numbers between 1 and 84 were removed from the beetles 5 weeks postinfection and were used for morphological studies as well as to infect domestic chicken, yellow-bellied geckos ( Hemidactylus flaviviridis) and fringe-toed lizards ( Acanthodactylus schmidti). All experimental animals, necropsied 4-38 weeks later, were positive for spirally coiled nematode larvae located under the skin and in the interstitium of skeletal muscles. Despite similarities in general morphology, larvae from beetles and reptiles and chicken differed strikingly in the total body length and body width. Differences in length of the muscular oesophagus and distances of cervical papillae, nerve ring and excretory pore from the anterior end were less distinct. Morphology of these larvae matched with larvae found in subcutaneous cysts in naturally infected houbara bustards ( Chlamydotis macqueeni) from Pakistan and UAE as well as with those detected in the muscles of an ocellated skink ( Chalcides ocellatus). Pulmonary protostrongyliasis of hare is a parasitic disease caused by nematodes belonging to the genus Protostrongylus (Nematoda, Protostrongylidae). During survey of wildlife disease in the South-East of France, pathologic examination of lungs from European hares found dead or hunter-killed between 2009 and 2012 was performed. Adult male worms were morphologically characterized and the identification confirmed by molecular biology (D2 domain of the 28S and ITS2 of rDNA). Two different species were identified: the first one, Protostrongylus pulmonalis, is identical with the haplotype previously deposited in GenBank. Based on morphological criteria of copulatory bursa of adult male worms (especially length of spicules and gubernaculum structure), we identified a second species found in France as Protostrongylus oryctolagi. This is the first report of P. oryctolagi in France from European hare and rabbit. P. oryctolagi was isolated from 248 hares and 3 rabbits in the South of France. P. pulmonalis was isolated from four hares found dead in the Northern France and from one hare in the South, which was co-parasitized by P. oryctolagi and P. pulmonalis. The European eel (Anguilla anguilla) is a critically endangered species. Red anus syndrome (RAS) is known to be associated with parasitic infections of the eel, particularly with Anguillicola crassus, but the full range of causative pathogenic organisms has not been systematically investigated. Here we examined the infective organisms and histopathological and ultrastructural features of seventy eels with RAS. In total, nine different pathogens were detected in association with RAS: Pseudomonas aeruginosa were present in twelve specimens (17%), the metacercaria of Euclinostromum heterostomum in three cases (4%), Gastrostome (Bucephalidae family) in seven cases (10%), A. crassus in forty-five cases (64%), Bothriocephalus in seventeen cases (24%), and Proteocephalus in twenty-three cases (32%). Yeast, amoeba, and myxobolus-like pathogens were seen in the anal skin in all cases when examined in combination with electron microscopy. Histopathologically, the lesions appeared as anoproctitis of varying severity from mild anusitis to severe haemorrhagic anoproctitis, with severe perianal oedema, haemorrhage, and proctoptosis. Gut inflammation ranged from mild catarrhal enteritis to severe haemorrhagic enteritis with mucosal sloughing. RAS is associated with a range of parasitic infections, not only A. crassus, some of which we describe here for the first time. Since RAS is not associated with direct invasion by parasites, it is likely that RAS is a secondary phenomenon caused by superadded infection on a background of generalised immunosuppression, or indirect local toxic effects. RAS may be used as a non-invasive indicator of underlying parasitic infection, but further investigations are required to establish the causative organisms for effective fishery management. Abstract: The establishment of Taenia solium adult parasite in the human intestine causes taeniosis. Importantly, the immunological mechanisms occurring at the interface between the parasite and its host are not fully known. The development of experimental animal models has facilitated the understanding of the host–parasite relationship. In this study we standardized a quantitative RT-PCR method for analyzing hamster messenger RNA for interferon-gamma (IFN-γ) and interleukins (IL): IL-4 IL-10, IL-12 and IL-13. This method was then used to evaluate the local cytokine response elicited against the adult parasite at the attachment site in the intestine of infected hamsters. The results showed an intense IFN-γ response, as well as an up-regulation of IL-4 as early as three days post-infection, permanence of IL-10 until the end of the experiment and down regulation of IL-12. Red squirrels (Sciurus vulgaris) declined in Great Britain and Ireland during the last century, due to habitat loss and the introduction of grey squirrels (Sciurus carolinensis), which competitively exclude the red squirrel and act as a reservoir for squirrelpox virus (SQPV). The disease is generally fatal to red squirrels and their ecological replacement by grey squirrels is up to 25 times faster where the virus is present. We aimed to determine: (1) the seropositivity and prevalence of SQPV DNA in the invasive and native species at a regional scale; (2) possible SQPV transmission routes; and, (3) virus degradation rates under differing environmental conditions. Grey (n = 208) and red (n = 40) squirrel blood and tissues were sampled. Enzyme-linked immunosorbent assay (ELISA) and quantitative real-time polymerase chain reaction (qPCR) techniques established seropositivity and viral DNA presence, respectively. Overall 8% of squirrels sampled (both species combined) had evidence of SQPV DNA in their tissues and 22% were in possession of antibodies. SQPV prevalence in sampled red squirrels was 2.5%. Viral loads were typically low in grey squirrels by comparison to red squirrels. There was a trend for a greater number of positive samples in spring and summer than in winter. Possible transmission routes were identified through the presence of viral DNA in faeces (red squirrels only), urine and ectoparasites (both species). Virus degradation analyses suggested that, after 30 days of exposure to six combinations of environments, there were more intact virus particles in scabs kept in warm (25°C) and dry conditions than in cooler (5 and 15°C) or wet conditions. We conclude that SQPV is present at low prevalence in invasive grey squirrel populations with a lower prevalence in native red squirrels. Abstract: The domestic South American camelids (SACs), llama (Lama glama) and alpaca (Lama paco), are frequently found to be infected with Sarcocystis parasites. Infections give rise in skeletal muscle to macroscopic cysts (1–5mm long) that resemble rice seeds, each containing several million living bradyzoites. The finding of cysts prevents commercialization of SAC meat, an important source of income for rural families in the Andean flatlands. Thus, development of diagnostic methods to facilitate the control of these infections is highly desirable, and the first step to this end is the unequivocal species identification of the causative agent. Based on the cyst form and size, the infecting parasite has been described as Sarcocystis aucheniae; however, this traditional approach is not reliable as similar cysts may contain different species. To date, molecular identification has been done for a single isolate of S. aucheniae from an alpaca in Australia. In order to verify the identity of the species present in SACs of South America, the complete 18S rRNA gene was PCR-amplified and sequenced from macrocyst DNA obtained from three llamas of the Andean flatlands. A phylogenetic Bayesian analysis was carried out using the analyzed and available 18S rRNA sequences of Sarcocystis spp. In the constructed tree, all of the new 18S rRNA gene sequences segregated in a single clade together with the 18S rRNA gene sequence reported from an alpaca in Australia, demonstrating that the isolated parasite is S. aucheniae, and that this parasite indiscriminately infects both domestic SACs. This work represents the first molecular identification of the causative agent of SAC sarcocystiosis in South America, and can contribute to the development of control methods for this neglected parasitosis. Abstract: Fasciolosis is an emerging or reemerging human and animal disease in numerous parts of the world. In Galicia (NW, Spain), the wild boar (Sus scrofa) is the main wild ungulate in terms of abundance and distribution. Its population has continuously increased over the past decades and this population growth has been accompanied by a reduction of habitats, so that the wild boar populations encroach more and more frequently onto agricultural lands. The increase of the interface area between livestock and the wild boars frequently involves the sharing of pastures and water sources, so that the circulation of common pathogens is propitiated. This is the first report concerning the importance of the wild boar as a possible reservoir of Fasciola hepatica infection in Spain. Livers from 358 hunted wild boars were analyzed showing that 11.2% were parasitized by F. hepatica, with burdens ranging from 1 to 14 flukes (mean=2.3). Fecal analysis demonstrated that 40.0% of parasitized animals shed F. hepatica eggs with a mean excretion of 6.1 eggs per gram of feces (epg). The presence of coproantigens analyzed by MM3-COPRO ELISA was positive in 62.9% of infected wild boars. After incubation, the percentage of hatched eggs ranged between 41.0% and 90.0% suggesting that the wild boar is very likely to contribute to the environmental contamination with viable parasite eggs. Comparative morphometric data were obtained using a computer image analysis system (CIAS) on the basis of standardized measurements. F. hepatica from cattle, sheep and wild boars from the same geographical area presents a similar body development and gravidity. Our study shows for the first time that the F. hepatica uterus from the wild boar presents an intermediate size between that found in primary reservoir hosts such as cattle and sheep, i.e., the individual potential egg output capacity of the wild boar does not greatly differ from that detected in Galician livestock. These results show that F. hepatica in Galicia has a normal development in wild boars, presenting its own characteristics in shape and size in comparison with other host species. The high prevalence of infection detected in the wild boar, the normal fluke development in the liver, and the possibility of shedding F. hepatica eggs capable of embryonating and giving rise to viable miracidia with the potential to infect intermediate hosts suggest a possible role of this species as a secondary reservoir in this Spanish region. The aim of this work is to know the prevalence of Fasciola hepatica in 301 roe deer and in 676 beef cattle kept in an endemic area. Detection of antibodies was determined in roe deer using a homemade ELISA with excretory/secretory antigens (FhES) and a recombinant protein (FhrAPS). None of the deer passed eggs by faeces and none flukes in their livers were found. The seroprevalence of F. hepatica was 29% using FhES, with significantly higher values in the oldest ones (36%). Twenty-eight percent of the samples were positive to FhrAPS. Twenty-three percent of the cows eliminated eggs of F. hepatica and the seroprevalence was 67% using FhrAPS. No relationship between the seropositivity values of deer and cattle was demonstrated. The role of wild ruminants as reservoirs of F. hepatica is discussed. Abstract: The present report describes the reappearance of Taenia ovis krabbei in a roe deer from Denmark after more than 60 years. The cysticerci were isolated from the thigh muscle of the deer, and the diagnosis was based on histostological analysis, morphology of the rostellar-hooks as well as molecular typing of the mitochondrial cytochrome c oxidase I (cox1) gene. The exact definitive host was not revealed in this report, but domestic dogs may play a role of the definitive host in the area. This finding is of concern to hunters and deer meat producers, since the infected meat is usually condemned due to esthetic reasons. [Copyright &y& Elsevier]

Abstract: Intestinal contents of 218 roe deer hunted in the northwest (NW) of the Iberian Peninsula during the 2008–2009 hunting seasons were examined in order to provide information on the gastrointestinal (GI) nematode prevalence and intensity of infection and the possible influence of some environmental and intrinsic factors such as climatic conditions, age and sex. All the animals studied harboured GI nematodes, and a total of 20 different species belonging to ten genera were identified. Spiculopteragia spiculoptera/Spiculopteragia mathevossiani, Ostertagia leptospicularis/Ostertagia kolchida and Nematodirus filicollis were the most common. This is the first citation for Chabertia ovina, Cooperia pectinata, Cooperia punctata, Cooperia oncophora, Haemonchus contortus, Nematodirus spathiger, Oesophagostomum venulosum, Teladorsagia trifurcata, Trichostrongylus capricola, Trichostrongylus colubriformis, Trichostrongylus vitrinus and Trichuris capreoli in roe deer from the Iberian Peninsula. Prevalence and intensity were significantly higher in the abomasum, where infections with more than one GI nematode species were the most common; in the other intestinal segments infections with only one GI nematode species were the most prevalent. When considering the influence of the different risk factors on the prevalence of GI nematodes, the highest prevalence for most of the genera were observed in roe deer from coastal areas, where climatic conditions are more favourable for the development and survival of third stage larvae in the environment. Regarding the sex of the animals, the prevalence was, in general, higher in males than in females, probably due to behavioural and physiological sex-related differences. On the contrary, no differences were found in relation to the age of the animals. This study reveals that roe deer from the NW of the Iberian Peninsula are widely and intensely infected with gastrointestinal nematodes, which probably affect the health status of these ungulates. Abstract: Hares are important game species in Europe and their meat is consumed by humans. Toxoplasmosis is an important cause of mortality in hares in Nordic countries but little is known of Toxoplasma gondii infection in the Iberian hare (Lepus granatensis). Sera from 298 Iberian hares from Navarra (North Spain) were assayed for antibodies against T. gondii by the modified agglutination test. The samples were collected at necropsy during three consecutive years (2009–2011). Titers of 1:25 and higher were found in 34 hares (11.4%; CI95%: 7.8–14.9). Significant differences in seroprevalence were observed among geographical areas, years of sampling and age groups. The highest seroprevalence and the highest titers were observed in 2009, indicating fluctuating rates of transmission. Significantly higher seroprevalence was observed in the juvenile age group compared to the adult hare group. More than 40% of juvenile hares were seropositive the first year of study, seroprevalence drastically decreased in the juvenile animals sampled in the second year and was negligible in those sampled in the third year in this group. No clinical cases were detected in the study area and T. gondii was not detected by PCR in the liver of the 34 seropositive hares tested. These data may indicate that most juvenile seropositive hares did not survive as adults in the consecutive year or that there is a short-term humoral immune response against T. gondii in Iberian hares. Abstract: Blood samples and ticks were collected from 100 shepherd dogs, 12 hunting dogs and 14 stray dogs in southern Hungary, in order to screen them for the presence of Hepatozoon spp. by PCR. Out of 126 blood samples, 33 were positive (26%). Significantly more shepherd dogs (31%) were infected, than hunting (8%) and stray dogs (7%). Three genotypes of Hepatozoon canis were identified by sequencing, differing from each other in up to six nucleotides in the amplified portion of their 18S rRNA gene. In Dermacentor marginatus larvae/nymphs and Dermacentor reticulatus nymphs, H. canis was present only if they had been collected from PCR-positive dogs, and the genotypes were identical in the ticks and their hosts. However, two Haemaphysalis concinna nymphs removed from a PCR-negative dog were found positive for H. canis, and the genotype detected in specimens of this tick species differed from that in the blood of their respective hosts. These results indicate that canine hepatozoonosis may be highly prevalent in regions where Rhipicephalus sanguineus is considered to be non-endemic. In addition, H. canis was identified for the first time in Hungary, as well as in D. marginatus, D. reticulatus and Ha. concinna ticks. Canine hepatozoonosis was significantly more prevalent west of the Danube river (where higher densities of red fox and golden jackal populations occur), suggesting a role of wild carnivores in its epidemiology. Abstract: The prevalence of piroplasm (order Piroplasmida) infection was assessed in blood and bone marrow samples from 91 red foxes (Vulpes vulpes) from northern, central and southern Portugal by means of molecular methods. PCR for the 18S rRNA gene of Babesia spp. followed by sequencing revealed 63 foxes positive for the Babesia microti-like piroplasm (syn. Theileria annae) (69.2%; 95% confidence interval [CI]: 58.7–78.5%) and one fox positive for Babesia canis (1.1%; 95% CI: 0.0–6.0%). Positivity to the B. microti-like piroplasm or B. canis in 43 blood samples (83.7%) was significantly higher (p <0.001) than in 43 paired bone marrow samples (20.9%). There were no statistically significant differences in the prevalence of infection between genders (p =0.219) or age groups (<2 years vs. ≥2 years) (p =1.0). This is the first report of the B. microti-like piroplasm in foxes from Portugal as well as the first report on detection by PCR and genotyping of B. canis in a red fox worldwide. A natural cycle of the B. microti-like piroplasm is suggested in red fox populations based on the high prevalence of the protozoan. Background: Infectious diseases pose one of the greatest threats to endangered species, and a risk of gastrointestinal parasite transmission from humans to wildlife has always been considered as a major concern of tourism. Increased anthropogenic impact on primate populations may result in general changes in communities of their parasites, and also in a direct exchange of parasites between humans and primates. Aims: To evaluate the impact of close contact with humans on the occurrence of potentially zoonotic protists in great apes, we conducted a long-term monitoring of microsporidia, Cryptosporidium and Giardia infections in western lowland gorillas at different stages of the habituation process, humans, and other wildlife in Dzanga-Sangha Protected Areas in the Central African Republic. Results: We detected Encephalitozoon cuniculi genotypes I and II (7.5%), Enterocytozoon bieneusi genotype D and three novel genotypes (gorilla 1–3) (4.0%), Giardia intestinalis subgroup A II (2.0%) and Cryptosporidium bovis (0.5%) in gorillas, whereas in humans we found only G. intestinalis subgroup A II (2.1%). In other wild and domestic animals we recorded E. cuniculi genotypes I and II (2.1%), G. intestinalis assemblage E (0.5%) and C. muris TS03 (0.5%). Conclusion: Due to the non-specificity of E. cuniculi genotypes we conclude that detection of the exact source of E. cuniculi infection is problematic. As Giardia intestinalis was recorded primarily in gorilla groups with closer human contact, we suggest that human-gorilla transmission has occurred. We call attention to a potentially negative impact of habituation on selected pathogens which might occur as a result of the more frequent presence of humans in the vicinity of both gorillas under habituation and habituated gorillas, rather than as a consequence of the close contact with humans, which might be a more traditional assumption. The sucking louse Haematopinus tuberculatus (Burmeister 1839) is an ectoparasite of buffaloes, cattle, camels, and American bison. Alphacypermethrin (ACYP) is a pyrethroid insecticide commonly used to control arthropods of veterinary and public health interest. Therapeutics, such as antiparasitic compounds, is often administered to buffaloes based on dosage and intervals recommended for cattle because very few drugs have buffalo-specific label indications. A trial was conducted on 20 louse-infested buffaloes at a farm to assess the efficacy and safety of ACYP pour-on, at the manufacturer's recommended dose for cattle, on buffaloes naturally infested by H. tuberculatus. Ten animals were assigned to ACYP-treated group (ACYP-group) and ten to untreated control group (C-group). On day 0, all ACYP-group buffaloes received alphacypermethrin pour-on. Louse counts were performed on days −1, 7, 14, 21, 28, 35, 42, 49, and 56 at eight predilection sites on the skin of each buffalo. ACYP was completely effective (100 %) at day 7, highly effective (99.8 %) at day 14, and completely effective (100 %) from day 21 until the end of the study (day 56 post-treatment). During the trial, ACYP was well tolerated by all animals as there were no observed clinically adverse reactions. The results of this trial suggest that ACYP is an effective, safe, and user-friendly compound suitable for treatment of buffaloes with natural louse infestations. [ABSTRACT FROM AUTHOR]

Aphanomyces astaci, the crayfish plague pathogen, first appeared in Europe in the mid-19th century and is still responsible for mass mortalities of native European crayfish. The spread of this parasite across the continent is especially facilitated by invasive North American crayfish species that serve as its reservoir. In France, multiple cases of native crayfish mortalities have been suggested to be connected with the presence of the signal crayfish Pacifastacus leniusculus, which is highly abundant in the country. It shares similar habitats as the native white-clawed crayfish Austropotamobius pallipes and, when infected, the signal crayfish might therefore easily transmit the pathogen to the native species. We investigated the prevalence of A. astaci in French signal crayfish populations to evaluate the danger they represent to local populations of native crayfish. Over 500 individuals of Pacifastacus leniusculus from 45 French populations were analysed, plus several additional individuals of other non-indigenous crayfish species Orconectes limosus, O. immunis and Procambarus clarkii. Altogether, 20% of analysed signal crayfish tested positive for Aphanomyces astaci, and the pathogen was detected in more than half of the studied populations. Local prevalence varied significantly, ranging from 0% up to 80%, but wide confidence intervals suggest that the number of populations infected by A. astaci may be even higher than our results show. Analysis of several individuals of other introduced species revealed infections among two of these, O. immunis and P. clarkii. Our results confirm that the widespread signal crayfish serves as a key reservoir of Aphanomyces astaci in France and therefore represents a serious danger to native crayfish species, especially the white-clawed crayfish. This paper describes a cross-sectional study in buffaloes of district Toba Tek Singh from April, 2009 to March, 2010 which aimed to identify prevalent species of Eimeria (E) and risk factors associated with subclinical coccidiosis. Overall prevalence of Eimeria in buffaloes was 49.6%. Six species of Eimeria were identified in total infected buffaloes. E. bovis was the commonest one among the species identified during survey followed in order by E. zuernii, E. canadensis, E. ellipsoidalis, E. alabamensis and E. cylindrica. Peak prevalence was observed in August. Wet season was found favorable for Eimeria. Odds of finding oocysts were higher in young stock and females (P<0.05) as compared to those of adults and males respectively. Among management and husbandry practices, five variables were found significantly associated with status. These included housing system, feeding system, watering system, floor type and herd size. Open housing system, trough feeding, tap watering, partially cemented floor type and smaller herds showed protective effect against coccidiosis with Odds of detection being higher in closed housing system, ground feeding, pond watering, non-cemented floor type and larger herd size, respectively. Body condition and breed of buffaloes were not found associated with prevalence of Eimeria. Value of feeding in troughs, provision of clean tap water, cementing floor of farms in reducing the extent of infection should be communicated to farmers. A study was undertaken on clinico-biochemical status in healthy and naturally infected buffaloes with fasciolosis and its therapeutic management through different anthelmintic combination. TSP, albumin, globulin and A/G ratio were found to be reduced as compared to healthy, whereas SGOT and SGPT were found to be significantly higher in infected buffaloes. Recovery was observed in different clinico-biochemical parameters on treatment with combination of oxyclozanide and albendazole @ 15 mg/kg.b.wt., followed by oxyclozanide + tetramisole and oxyclozanide alone. It is also revealed that apart from chemotherapy, supportive therapy helped in speedy recovery of different biochemical parameters and in gaining normal health after elimination of parasite. Hypodermosis is an endemic disease in semi-hilly and mountainous areas of Pakistan. Keeping in view the importance of buffaloes an epidemiological survey was conducted to determine the prevalence of hypodermosis in district Jhelum Punjab, Pakistan, during the year 2010-2011. Out of 1000 buffaloes examined clinically in the study area, 32 (3.2%) found to be positive for the warble fly infestation. The number of nodules in the infested animals ranged from 1-5 (2.7±1.1). There were significant differences (P<0.05) in the prevalence of Hypoderma spp. when the sex, age and different geographic areas were considered. The Prevalence was higher in males and young animals and also in those animals grazing in hilly and semi-hilly areas. The climatic conditions (temperature, humidity, sunshine and wind speed) favoured the warble fly activity and contributed in the onset of disease. [ABSTRACT FROM AUTHOR]

Abstract: Bovine besnoitiosis, caused by Besnoitia besnoiti, is considered to be emergent in Europe and responsible for severe economic losses due to the chronic and debilitating course of the disease but has not been reported in North America. Besnoitia tarandi is a related species and it has been reported in reindeer and caribou from different locations of the Arctic Pole, including North America. Diagnosis of clinical besnoitiosis is largely based on the recognition of dermal grossly visible tissue cysts of Besnoitia. Nothing is known of cross reactivity between B. besnoiti and B. tarandi species. Here, we evaluated the use of serological tests employed in the diagnosis of bovine besnoitiosis for the detection of Besnoitia spp. infections in different wild ruminant species (caribou, elk, mule-deer, white-tailed deer, moose, muskox and bison) from Canada and investigated cross-reactivity between B. besnoiti and B. tarandi species by indirect immunofluorescence antibody test and Western blot. For this, species-specific antibodies were obtained in rabbits experimentally infected with B. besnoiti and B. tarandi. Marked cross reactivity was found between B. besnoiti and B. tarandi. For the first time, antibodies to Besnoitia spp. infection were found in 16 of 20 caribou (Ranginfer tarandus), seven of 18 muskox (Ovibos moschatus), one of three bison (Bison bison), but not in 20 elk (Cervus canadensis), 20 white tailed deer (Odocoileus virginianus), and 20 moose (Alces alces) in Canada; results were similar using B. besnoiti and B. tarandi as antigen. There was no cross reactivity between the two Besnoitia species, Neospora caninum and Toxoplasma gondii with the cut-offs applied that prevented to observe it. The present study provides evidence that the serological assays can be useful to accomplish large scale prevalence studies in caribou and other wildlife species.
