DL-β-amino-butyric acid (BABA) has been found to protect potato plants against late blight caused by Phytophthora infestans. This paper compares gene expression profiles of a potato clone harboring horizontal resistance to P. infestans in response to BABA and the pathogen by using cDNA-amplified fragment length polymorphism (cDNA-AFLP), aiming at fudermental elucidation of molecular mechanisms of BABA induced resistance (BABA-IR). In total, 101 transcript derived fragments (TDFs) showed significant differential expression among the treated and their respective control samples. About half of differentially expressed fragments (49) were overlapped in both BABA and P. infestans induction systems, and homology analysis showed that these TDFs in common involved in signaling, cell wall strengthening and synthesis of antimicrobial compounds, reflecting both BABA-IR and the natural resistance shared similar defense mechanisms to a great extent. Analysis of the transcription profiles demonstrated that an early activation of plant basal defense system could be crucial for BABA-IR. Some differentially expressed TDFs homologous to genes encoding proteins related to jasmonic acid–and salicylic acid–dependent signaling pathways, were up-regulated by BABA. Similar genes associated with these pathways were also identified in the pathogen inoculated samples, reinforcing that the extensive cross-talk between BABA-IR and horizontal resistance may be important to coordinate the genes responsible for P. infestans infection in potato. Common scab, caused by Streptomyces scabies, is a disease that produces scab-like surface lesions on potato tubers. Testing for susceptibility/resistance of breeding lines at the Potato Research Centre of Agriculture & Agri-Food Canada (AFC) is carried out in a scab nursery maintained at AFC for the annual scab evaluation field trials. A replicated field trial routinely consists of breeding lines from previous testing season(s) plus newly selected lines. Data of scab scores generated from long-term experiments thus formulate an incomplete 2-way table over combinations of breeding lines (genotypes) and trials (years). This requires an advanced statistical method to estimate genetic parameters for evaluation purposes. A data base with 1,435 scab index scores from 344 breeding lines were extracted from 5 years (1995–1999) of field experiments in the scab nursery maintained at AFC. The statistical method Residual Maximum Likelihood (REML) was employed to estimate variance components of the breeding population and Best Linear Unbiased Predictor (BLUP) was used to predict genetic merit of breeding lines. High heritability was obtained from variance components estimated by REML. The BLUP scores of breeding lines provided reliable evaluation of their responses to common scab. Two data base sub-sets were separately formulated from the original data base for those parents and grandparents of the breeding lines having pedigree records available. They were again subjected to REML and BLUP analyses to compare the responses to common scab and identify sources of resistance at the parental and grandparental levels. Two random data sets with equal sized samples of breeding lines were also generated from the over-all data base. Calcium signalling has profound implications in the fungal infection of plants and animals, during which a series of physiological and morphological transitions are required. In this article, using a model fungal pathogen, Magnaporthe oryzae, we demonstrate that the regulation of the intracellular calcium concentration ([Ca2+]int) is essential for fungal development and pathogenesis. Imaging of [Ca2+]int showed that infection-specific morphogenesis is highly correlated with the spatiotemporal regulation of calcium flux. Deletion of the fungal phospholipase C gene ( M.  oryzae phospholipase C 1, MoPLC1) suppressed calcium flux, resulting in a fungus defective in developmental steps, including appressorium formation and pathogenicity. Surprisingly, the PLC-δ1 gene of mouse was able to functionally substitute for MoPLC1 by restoring the calcium flux, suggesting the evolutionary conservation of the phospholipase C-mediated regulation of calcium flux. Our results reveal that MoPLC1 is a conserved modulator of calcium flux that is essential for the regulation of key steps in fungal development and pathogenesis. [ABSTRACT FROM AUTHOR]

Tomato production in West Africa has been severely affected by begomovirus diseases, including yellow leaf curl and a severe symptom phenotype, characterized by extremely stunted and distorted growth and small deformed leaves. Here, a novel recombinant begomovirus from Mali, Tomato yellow leaf curl Mali virus (TYLCMLV), is described that, alone, causes tomato yellow leaf curl disease or, in combination with a betasatellite, causes the severe symptom phenotype. TYLCMLV is an Old World monopartite begomovirus with a hybrid genome composed of sequences from Tomato yellow leaf curl virus-Mild (TYLCV-Mld) and Hollyhock leaf crumple virus (HoLCrV). A TYLCMLV infectious clone induced leaf curl and yellowing in tomato, leaf curl, crumpling and yellowing in Nicotiana benthamiana and common bean, mild symptoms in N. glutinosa, and a symptomless infection in Datura stramonium. In a field-collected sample from a tomato plant showing the severe symptom phenotype in Mali, TYLCMLV was detected together with a betasatellite, identified as Cotton leaf curl Gezira betasatellite (CLCuGB). Tomato plants co-agroinoculated with TYLCMLV and CLCuGB developed severely stunted and distorted growth and small crumpled leaves. These symptoms were more severe than those induced by TYLCMLV alone, and were similar to the severe symptom phenotype observed in the field in Mali and in other West African countries. TYLCMLV and CLCuGB also induced more severe symptoms than TYLCMLV in the other solanaceous hosts, but not in common bean. The increased symptom severity was associated with hyperplasia of phloem-associated cells, but relatively little increase in TYLCMLV DNA levels. In surveys of tomato virus diseases in West Africa, TYLCMLV was commonly detected in plants with leaf curl and yellow leaf curl symptoms, whereas CLCuGB was infrequently detected and always in association with the severe symptom phenotype. Together, these results indicate that TYLCMLV causes tomato yellow leaf curl disease throughout West Africa, whereas TYLCMLV and CLCuGB represent a reassortant that causes the severe symptom phenotype in tomato. Pantoea ananatis causes disease symptoms in a wide range of economically important agricultural crops and forest tree species worldwide. It is regarded as an emerging pathogen based on the increasing number of reports of diseases occurring on previously unrecorded hosts in different parts of the world. Its unconventional nature lies in the fact that, unlike the majority of plant pathogenic microbes, P. ananatis is capable of infecting humans and occurs in diverse ecological niches, such as part of a bacterial community contaminating aviation jet fuel tanks and contributing to growth promotion in potato and pepper. Taxonomy: Bacteria; Gammaproteobacteria; family Enterobacteriaceae; genus Pantoea. Microbiological properties: Gram-negative; facultatively anaerobic; most strains are motile and produce a yellow pigment in culture; indole positive. Biology: Pantoea ananatis is a common epiphyte; it also occurs endophytically in hosts where it has been reported to cause disease symptoms and in hosts where no such symptoms have been described. Some strains are ice-nucleating, a feature which has been used as a biological control mechanism against some insect pests of agricultural crops and by the food industry. Disease symptoms: Pantoea ananatis infects both monocotyledonous and dicotyledonous plants. The symptoms are diverse depending on the host infected, and include leaf blotches and spots, die-back, and stalk, fruit and bulb rot. Biological control agent: Pantoea ananatis has both antifungal and antibacterial properties. These characteristics have the potential of being exploited by biological control specialists. A questionnaire was sent to all participating countries to gain some background information on rape growing and on pollen beetle ( Meligethes aeneus). Information was sought on the use of thresholds, the availability of active substances and on the year in which resistance, if present, had been observed. A summary of the main findings of the questionnaire are presented. In total 20 countries responded and resistance in pollen beetle was reported in 12 of those countries. The article focuses on the discovery of rhynchophorus ferrugineus (Olivier), a dangerous pest of palms. R. ferrugineus is composed of common named such as Asiatic palm weevil, coconut weevil, red palm weevil and red stripe weevil. This pest also attacks in Agave Americana and sugarcane. It geographically distributed in different countries including Cyprus, France and Israel. The Coconut Research Institute introduces Davis Red Weevil detector to detect R. ferrugineus larva attack in palms.

The article focuses on the discovery of hesperophanes campestris Faldermann commonly known as mulberry beetle, a pest larva in plants/trees. H. campestris usually approaches in fruit and deciduous trees. H. campestris geographically distributed in several countries including Eastern Siberia, Armenia and Japan. Common symptoms of H. campestris includes holes in trunks, peeling bark and leaves yellowing. To prevent spreading of H. campestris, plants for planting must come from a pest-free area.

Sugar beet root maggot (SBRM; Tetanops myopaeformis) is a major insect pest of sugar beet throughout much of North America. Two insecticides with the same mode of action are used almost exclusively for SBRM control. Alternative control strategies would be required if insecticide-resistant SBRM developed or the insecticides were no longer available due to regulatory actions. Germplasm lines with SBRM resistance are available, but information on the SBRM resistance of hybrid cultivars created by crossing these lines with a SBRM-susceptible cms parental line is lacking. This study compared the performance of four hybrids with SBRM-resistant pollinators to two susceptible hybrids, with and without insecticide, in four environments. In all environments, the hybrids with the SBRM-resistant pollinators and no insecticide had root yields equal to the SBRM-susceptible commercial hybrid with insecticide. In two environments, the root yields of the SBRM-susceptible hybrids were low and the difference between the root yields with and without insecticide was relatively small, indicating the insecticide was ineffective. In contrast, the root yields of the SBRM-resistant hybrids, with or without insecticide, in these trials were substantially higher than the susceptible hybrids and similar to corresponding root yields at the other sites. Based upon these trials, it appears that hybrids produced with the currently available resistance sources would have sufficient SBRM resistance to prevent catastrophic yield reductions due to SBRM feeding, could reduce uncertainty caused by fluctuations in insecticide effectiveness, would provide protection against the development of insecticide-resistant SBRM strains, and also provide a useful level of SBRM protection, if the current insecticides were no longer available. Aluminum being the third most abundant metal in the earth's crust poses a serious threat to crop productivity in acid soils, which comprise almost half of the arabIe land. This review travels across time and updates research done on aluminum stress in plants. In its phytotoxic forms, aluminum affects root growth by acting in the root apical zone, resulting in growth inhibition in a very short time at micromolar concentrations. The mechanisms of aluminum toxicity in plants may proceed by growth inhibition, callose accumulation, cytoskeletal distortion, disturbance of plasma membrane surface charge, and W-ATPase activity, lipid peroxidation of membranes, production of reactive oxygen species in cytosol and mitochondria, respiratory dysfunction, opening of mitochondrial permeability transition pores, collapsing of inner mitochondrial membrane potential, activation of mitochondrial protease, and induction of nuclear apoptosis, resulting ultimately in programmed cell death. In contrast, the mechanism of tolerance involves the exudation of organic acid anions, complexation of aluminum with organic acids, and subsequent detoxification. Many oxidative stress genes and other metabolically important genes have also been found to be induced under aluminum stress, and overexpression analyses have also shown some plants to develop some degree of tolerance. In the future, researchers in the area of aluminum research should investigate more basic mechanisms of aluminum toxicity and discover and study more aluminum-responsive genes that confer resistance against this toxic metal, to ensure food security for ever-increasing human populations in the future. To identify genes involved in trifluralin herbicide-induced resistance of melon to Fusarium oxysporum f. sp. melonis, suppression subtractive hybridization (SSH) and cDNA-amplified fragment-length polymorphism (cDNA-AFLP) were used. A total of 123 clones—60 of which have never been isolated from melon—were isolated, sequenced and annotated. A significant proportion (35%) of the total 123 clones exhibited similarity to genes that have been formerly described as stress- or defence-related. Thirty-two selected clones were subjected to a detailed expression analysis, one-third of which were found to be up-regulated in response to trifluralin treatment and/or fusarium inoculation. The putative roles of seven of these clones in stress are discussed. Furthermore, the expression of four stress-related and up-regulated genes was enhanced when the plants were subjected to salinity stress, suggesting that trifluralin induces a general stress response which protects the plant against fusarium wilt. Alternaria brassicicola is a necrotrophic pathogen causing black spot disease on virtually all cultivated Brassica crops worldwide. In many plant pathosystems fungal secondary metabolites derived from non-ribosomal peptide synthetases (NPSs) are phytotoxic virulence factors or are antibiotics thought to be important for niche competition with other micro-organisms. However, many of the functions of NPS genes and their products are largely unknown. In this study, we investigated the function of one of the A. brassicicola NPS genes, AbNPS2. The predicted amino acid sequence of AbNPS2 showed high sequence similarity with A. brassicae, AbrePsy1, Cochliobolus heterostrophus, NPS4 and a Stagonospora nodorum NPS. The AbNPS2 open reading frame was predicted to be 22 kb in length and encodes a large protein (7195 amino acids) showing typical NPS modular organization. Gene expression analysis of AbNPS2 in wild-type fungus indicated that it is expressed almost exclusively in conidia and conidiophores, broadly in the reproductive developmental phase. AbNPS2 gene disruption mutants showed abnormal spore cell wall morphology and a decreased hydrophobicity phenotype. Conidia of abnps2 mutants displayed an aberrantly inflated cell wall and an increase in lipid bodies compared with wild-type. Further phenotypic analyses of abnps2 mutants showed decreased spore germination rates both in vitro and in vivo, and a marked reduction in sporulation in vivo compared with wild-type fungus. Moreover, virulence tests on Brassicas with abnps2 mutants revealed a significant reduction in lesion size compared with wild-type but only when aged spores were used in experiments. Erwinia amylovora is a plant pathogenic enterobacterium that causes fire blight disease of apple, pear and other rosaceous plants. A type III (T3) secretion system, encoded by clustered, chromosomal hrp genes (hypersensitive response and pathogenicity), is essential for infection, but only a few proteins are known that are secreted through this pathway (the T3 ‘secretome’). We developed an efficient protocol for purification and concentration of extracellular proteins and used it to characterize the T3 secretome of E. amylovora Ea273 by comparing preparations from the wild-type strain with those from mutants defective in hrp secretion, regulation, or in genes encoding putative T3-secreted proteins. Proteins were resolved by gel electrophoresis and identified using mass spectrometry and a draft sequence of the E. amylovora genome. Twelve T3-secreted proteins were identified, including homologues of known effector and helper proteins, and HrpJ, a homologue of YopN of Yersinia pestis. Several previously uncharacterized T3-secreted proteins were designated as Eops for Erwinia outer proteins. Analysis of the secretome of a non-polar hrpJ mutant demonstrated that HrpJ is required for accumulation of wild-type levels of secreted harpins. The gene-for-gene interaction triggering resistance of wheat against first-instar Hessian fly larvae utilizes specialized defence response genes not previously identified in other interactions with pests or pathogens. We characterized the expression of Hfr-3, a novel gene encoding a lectin-like protein with 68–70% identity to the wheat germ agglutinins. Within each of the four predicted chitin-binding hevein domains, the HFR-3 translated protein sequence contained five conserved saccharide-binding amino acids. Quantification of Hfr-3 mRNA levels confirmed a rapid response and gradual increase, up to 3000-fold above the uninfested control in the incompatible interaction 3 days after egg hatch. Hfr-3 mRNA abundance was influenced by the number of larvae per plant, suggesting that resistance is localized rather than systemic. In addition, Hfr-3 was responsive to another sucking insect, the bird cherry-oat aphid, but not to fall armyworm attack, wounding or exogenous application of methyl jasmonate, salicylic acid or abscisic acid. Western blot analysis demonstrated that HFR-3 protein increased in parallel to mRNA levels in crown tissues during incompatible interactions. HFR-3 protein was detected in both virulent and avirulent larvae, indicating ingestion. Anti-nutritional proteins, such as lectins, may be responsible for the apparent starvation of avirulent first-instar Hessian fly larvae during the initial few days of incompatible interactions with resistant wheat plants. The flax rust resistance gene L, a nucleotide binding site, leucine-rich repeat (NBS-LRR) class of plant resistance gene, has 12 characterized alleles with different gene-for-gene resistance specificities. Here the specificities of presumptive L1, L5, L8 and L11 genomic clones are confirmed by transgenic expression. L6 and L11 differ by 33 amino acids, 32 in the LRR region and one in the C-terminal non-LRR region, and recognize unrelated avirulence proteins, AvrL567 and AvrL11, respectively. To analyse the specificity differences, 13 L6L11 recombinant genes were constructed in vitro and tested in transgenic flax for resistance to F2 progeny of rust strain CH5, in which the unlinked avirulence genes AvrL567 and AvrL11 segregate. The data show that the single C-terminal non-LRR region polymorphism is not involved in L6–L11 specificity differences, that polymorphisms necessary for specificity are spread throughout the LRR region and that some polymorphisms essential for L11 are not essential for L6. Seven ‘null’ recombinants expressed no resistance when tested with CH5-derived rusts. These were tested for new resistance specificities by inoculation with a strain of rust, Bs-1, which is distantly related to CH5 and which potentially carries a different range of avirulence specificities. The ‘null’ recombinant L6L11RV, which differs from L6 and L11 by its susceptibility to CH5, was resistant to strain Bs-1. The specificity difference is due to a reduction in the number of AvrL567 variants recognized by L6L11RV compared with L6 and not due to recognition of an unrelated Avr gene product in strain Bs-1.
