The use of a new atmospheric-pressure chemical ionization source for gas chromatography (APGC) coupled with a tandem quadrupole mass spectrometry (MS/MS) system, as an alternative to high-resolution mass spectrometry (HRMS), for the determination of PCDDs/PCDFs is described. The potential of using atmospheric-pressure chemical ionization (APCI) coupled to a tandem quadrupole analyzer has been validated for the identification and quantification of dioxins and furans in different complex matrices. The main advantage of using the APCI source is the soft ionization at atmospheric pressure, which results in very limited fragmentation. APCI mass spectra are dominated by the molecular ion cluster, in contrast with the high energy ionization process under electron ionization (EI). The use of the molecular ion as the precursor ion in MS/MS enhances selectivity and, consequently, sensitivity by increasing the signal-to-noise ratios (S/N). For standard solutions of 2,3,7,8-TCDD, injections of 10 fg in the splitless mode on 30- or 60-m-length, 0.25 mm inner diameter (id), and 25 μm film thickness low-polarity capillary columns (DB5MS type), signal-to-noise (S/N) ratios of >10:1 were routinely obtained. Linearity was achieved in the region (correlation coefficient of r(2) > 0.998) for calibration curves ranging from 100 fg/μL to 1000 pg/μL. The results from a wide variety of complex samples, including certified and standard reference materials and samples from several QA/QC studies, which were previously analyzed by EI HRGC/HRMS, were compared with the results from the APGC/MS/MS system. Results between instruments showed good agreement both in individual congeners and toxic equivalence factors (TEQs). The data show that the use of APGC in combination with MS/MS for the analysis of dioxins has the same potential, in terms of sensitivity and selectivity, as the traditional HRMS instrumentation used for this analysis. However, the APCI/MS/MS system, as a benchtop system, is much easier to use.

Persistent organic pollutants (POPs) are a large group of chemicals widely used and produced in various industrial applications. Many cell culture/animal studies have shown that POPs can induce oxidative stress. Since such data is lacking in humans, we conducted a large population-based study to analyze associations between POPs and oxidative stress markers. We measured following POPs; 16 polychlorinated biphenyls (PCBs), 5 organochlorine (OC) pesticides, octachlorinated dibenzo-p-dioxin, and polybrominated diphenyl ether 47, and oxidative stress markers; homocysteine, reduced [GSH] and oxidized glutathione [GSSG], glutathione ratio [GSSG/GSH], total glutathione, oxidized low-density lipoprotein [ox-LDL], ox-LDL antibodies, conjugated dienes, baseline conjugated dienes of LDL, and total anti-oxidative capacity in plasma samples collected from 992 70-year old individuals (50% women) from the population-based Prospective Investigation of the Vasculature in Uppsala Seniors (PIVUS) cohort. Linear regression analyses were performed to study the associations between oxidative stress markers and summary measures of POPs including the total toxic equivalence (TEQ), sums of PCBs and OC pesticides (main exposures) while adjusting for potential confounders. In multivariable-adjusted analyses, sum of PCBs showed strong associations with ox-LDL (β=0.94; P=2.9*10(-6)). Further, sum of PCBs showed association with glutathione-related markers (GSSG: β=-0.01; P=6.0*10(-7); GSSG/GSH: β=-0.002; P=9.7*10(-10)), although in reverse direction. Other summary measures did not show any significant association with these markers. In our study of elderly individuals from the general population, we show that plasma levels of POPs are associated with markers of increased oxidative stress thereby suggesting that even low dose background exposure to POPs may be involved in oxidative stress., (Copyright © 2014 Elsevier Ltd. Use of in vitro assays as screening tool to characterize contamination of a variety of environmental matrices has become an increasingly popular and powerful toolbox in the field of environmental toxicology. While bioassays cannot entirely substitute analytical methods such as gas chromatography-mass spectrometry (GC-MS), the increasing improvement of cell lines and standardization of bioassay procedures enhance their utility as bioanalytical pre-screening tests prior to more targeted chemical analytical investigations. Dioxin-receptor-based assays provide a holistic characterization of exposure to dioxin-like compounds (DLCs) by integrating their overall toxic potential, including potentials of unknown DLCs not detectable via e.g. GC-MS. Hence, they provide important additional information with respect to environmental risk assessment of DLCs. This review summarizes different in vitro bioassay applications for detection of DLCs and considers the comparability of bioassay and chemical analytically derived toxicity equivalents (TEQs) of different approaches and various matrices. These range from complex samples such as sediments through single reference to compound mixtures. A summary of bioassay derived detection limits (LODs) showed a number of current bioassays to be equally sensitive as chemical methodologies, but moreover revealed that most of the bioanalytical studies conducted to date did not report their LODs, which represents a limitation with regard to low potency samples., (Copyright © 2014 Elsevier B.V. Dioxin exposure has experimentally been associated with changes in DNA methylation, an epigenetic change that is associated with disease. The present study aims to investigate if serum levels of dioxin and other persistent environmental pollutants are related to global DNA methylation in a human sample. In the population-based Prospective Investigation of the Vasculature in Uppsala Seniors (PIVUS) study (all aged 70), global DNA methylation was measured by the Luminometric Methylation Assay in 524 subjects. Twenty-three different POPs, including 16 PCBs, five pesticides, one dioxin (OCDD) and one brominated flame retardant (BDE47) were analysed by HRGC/HRMS. Ten single nucleotide polymorphisms (SNPs) in the Aryl hydrocarbon (Ah)-receptor were analysed by mini-sequencing. High levels of toxic equivalency (TEQ) for PCBs and dioxin were associated with DNA hypermethylation (p=0.030). This was mainly attributed to coplanar non-ortho PCBs. While no significant associations were found between DNA methylation and SNPs in the Ah-receptor, an interaction was found between the SNP rs2237297 and TEQ so that TEQ was associated with hypermethylation (p=0.009) only in subjects with one G-allele (n=103). Also high levels of the PCB126 congener, the OCDD, and the pesticide metabolite p,p'-DDE were related to DNA hypermethylation (p=0.01, 0.03 and 0.003, respectively). In conclusion, in a sample of elderly subjects, high TEQ including PCBs and the dioxin OCDD and high serum levels of PCB126, OCDD, and p,p'-DDE were related to global DNA hypermethylation in a cross-sectional analysis., (© 2013.)

Background, Aim, and Scope: As a consequence of flood events, runoff and remobilized sediments may cause an increase of ecotoxicologically relevant effects from contaminant reservoirs. Aquatic and terrestrial organisms as well as cattle and areas of settlement are exposed to dislocated contaminants during and after flood events. In this study, the impacts of two flood events triggered by intense rain at the rivers Neckar and Rhine (Southern Germany) were studied. Effects in correlation to flood flow were assessed at the river Neckar using samples collected at frequent intervals. River Rhine suspended particulate matter (SPM) was sampled over a longer period at normal flow and during a flood event. Three cell lines (H4L1.1c4, GPC.2D.Luc, RTL-W1) were used to compare Ah receptor agonist activity in different biotest systems. Multilayer fractionation was performed to identify causative compounds, focusing on persistent organic contaminants., Materials and Methods: Native water and SPM of flood events were collected at the river Neckar and at the monitoring station (Rheinguetestation, Worms, Germany) of the river Rhine. Water samples were XAD-extracted. SPM were freeze-dried and Soxhlet-extracted using acetone and finally dissolved in dimethyl sulfoxide. Resulting crude extracts were analyzed for cytotoxicity with the neutral red assay. Aryl hydrocarbon receptor (AhR) agonist activity was measured in a set of biological test systems (DR-CALUX, GPC.2D, and ethoxyresorufin-O-deethylase (EROD) assay) and different cell lines. In addition, crude extracts were fractionated using a combined method of multilayer (sequence of acidified silica layers) and carbon fractionation. Fractions from the multilayer fractionation contained persistent organic compounds (polychlorinated dibenzo-p-dioxins and dibenzofurans (PCDD/Fs), polychlorinated biphenyls (PCBs), and some polycyclic aromatic hydrocarbon (PAHs)); fractions from the carbon fractionation were separated into a PCDD/F and a PCB fraction. Dioxin-like activity of multilayer and carbon fractions was determined in the EROD assay and expressed as biological toxicity equivalency concentrations of 2,3,7,8-tetrachlorodibenzo-p-dioxin (bio-TEQs). The calculation of chemical equivalency concentrations (chem-TEQs) and comparison to bio-TEQ values allowed the determination of the contribution of the analyzed persistent compounds to the total biological effects measured., Results: Soluble compounds in native and extracted water samples resulted in no or minor activity in the toxicity tests, respectively. Filter residues of native water caused increased AhR-mediated activity at the peak of the flood. Activities of SPM of the river Neckar correlated well with the flow rate indicating a flood-dependent increase of toxicity culminating at the peak of flow. River Rhine SPM showed a decrease of activity regarding an SPM sample of the flood event compared to a long-term sample. Excellent correlations with AhR agonistic activity were determined for DR-CALUX and EROD assay, while the GPC.2D assay did not correlate with both other biotests. The activity of persistent dioxin-like acting compounds in multilayer and carbon fractionated PCDD/F and PCB fractions was low if compared to corresponding crude extracts. The congener pattern of PCDD/F revealed that the contaminations mainly originated from products and productions of the chlorine and organochlorine industries., Discussion: Native and extracted water samples could be shown to contain little or no cytotoxic or AhR agonistic compounds. In contrast, particle-bound compounds were shown to be the relevant effect-causing fraction, as indicated by the activities of filter residues of native water and SPM. Compounds other than fractionated persistent PCBs and PCDD/Fs were more relevant to explain AhR-mediated activities of crude flood SPM at both rivers assessed. Biologically detected activities could at least in part be traced back to chemically analyzed and quantified compounds., Conclusions: The calculation of the portion of persistent PCBs and PCDD/Fs in multilayer fractions causing the high inductions in the EROD assay in combination with chemical analysis provides a suitable tool to assess dioxin-like activity of persistent compounds in SPM sampled over the course of flood events. Depending on the catchment area and annual course of flood events, end points may either indicate an increase or a decrease of activity. In order to determine the ecological hazard potential of mobilized contaminants during flood events, the focus should be set on particle-bound pollutants. Furthermore, PCDD/Fs and PCBs, commonly expected to be the most relevant pollutants in river systems, could be shown to contribute only to a minor portion of the overall AhR-mediated activity. However, they might be most relevant for human exposure when considering persistence and bioaccumulation-biomagnification in the food chain., Recommendations and Perspectives: As a consequence of climate change, flood events will increase in frequency and intensity at least in some regions such as Central Europe. Thus, it is crucial to identify the potential hazard of (re-)mobilized contaminants from reservoirs dislocated via floods and threatening especially aquatic organisms and cattle grazing in flood plains. Since other less persistent compounds seem to be more relevant to explain AhR-mediated activities in flood SPM, nonconventional PAHs and more polar compounds also need to be considered for risk assessment. Effect-directed analysis using broad-range fractionation methods taking into account compounds from polar to nonpolar should be applied for identification of pollutants causing biological effects, thus integrating biological and chemical parameters.

Elevated concentrations of dioxins in ancient ball clay from the Mississippi Embayment suggest natural formation of dioxins in the environment. Evidence for such natural formation in ball clay derives from unique congener profiles in undisturbed ancient clay deposits and from the lack of other anthropogenic contaminants. Here we present novel evidence of natural formation of dioxins based on congener-specific carbon isotopic analysis of octachlorodibenzo-p-dioxin (OCDD) in ball clays from the USA and Japan. The analyses were performed using a combination of double-column high performance liquid chromatography clean-up and two-dimensional gas chromatography-isotope ratio mass spectrometry. Elevated concentrations of OCDD found in ball clays from the USA and Japan were isotopically distinguished from the anthropogenic source materials (fly ash and pentachlorophenol) and environmental samples (sediment and soil). The isotopic signatures and the occurrence of OCDD in ancient ball clays deposited in the Tertiary Era provide evidence for the in situ formation of dioxins.

Dioxin levels measured after wet scrubbing systems have been found to be higher than levels measured before the scrubber. It is believed that there is an adsorption of PCDD/Fs on plastic materials in the scrubber. The PCDD/F levels after a polishing wet scrubber were followed continuously for 18 months using long-time sampling equipment at a hazardous waste incineration facility in Sweden. Each sampling period lasted two weeks. It was found that the levels during and shortly after start-up periods were elevated. The decline was very slowly, which supports a memory effect in the scrubber. Further, a multivariate model showed that the relation between different homologues changed over time, which is in agreement with a desorption model, taking into account the vapour pressures for different congeners.

This study is a consequence of a distinct fish decline in the Danube river since the beginning of the 1990s. In contrast to the decline of fish population, former studies have repeatedly documented that the water quality along the Danube river is improving. However, the conclusion of a pilot study in 2002 was that a high hazard potential is associated with local sediments. The present study documents that sediment samples from the Danube river showed comparatively high aryl hydrocarbon receptor mediated activity in biotests, using the cell lines GPC.2D.Luc, H4IIE (DR-CALUX®) and RTL-W1. The combination of chemical analysis, fractionation techniques and different in vitro tests revealed that priority pollutants could not explain the main induction, even though the concentrations of priority polycyclic aromatic hydrocarbons (PAHs) were very high (maximum in the tributary Schwarzach, sum of 16 EPA PAHs 26 μg/g). In conclusion, this investigation shows that nonpriority pollutants mainly mediate the high induction rates. Nevertheless, owing to the effects of PAHs towards fish and the connection between dioxin-like activity and carcinogenicity, the link between contamination and the fish population decline cannot be ruled out. As a consequence of ubiquitous use of brominated organic chemicals, there is a concern for persistent or increasing environmental levels of polybrominated dibenzo-p-dioxins/furans (PBDD/Fs) and mixed polychlorinated and polybrominated dibenzo- p-dioxins/furans (PXDD/Fs). Hence, there is a need to broaden the toxicological and environmental knowledge about these compounds, as a basis for risk assessment. In the study presented here, the relative potencies (REPs) for 18 PBDD/F and PXDD/ F congeners were determined in four dioxin-specific bioassays from different species: dioxin receptor chemically activated luciferase expression assay (DR-CALUX, rat hepatoma cells), TV101L (human hepatoma cells), and GPC.2D (guinea pig adenoma cells), as well as ethoxyresorufin-O-deethylase induction in the fish cell line RTL-W1 (rainbow trout liver cells). The bioassay specific REP factors presented here enable the assessment of the contribution from PBDD/Fs and PXDD/Fs to total 2,3,7,8-tetrachlorodibenzop- dioxin (TCDD) equivalents (TEQs: toxic equivalents), using bioassay analysis. The PBDD/Fs were found to be equally potent as their chlorinated analogues in the three mammalian assays, whereas the PXDD/Fs showed relatively higher potencies. Of special concern were the 2,3,7,8-substituted penta- and tetrahalogenated congeners, for which mean REPs were ≥1. The 2-B-1,3,7,8-CDD (2-bromo-1,3,7,8-tetrachlorodibenzo-p-dioxin) was up to three times more potent than TCDD in individual experiments (on weight basis). The RTL-W1 was less sensitive to the tested compounds with overall 10-fold lower REPs than the mammalian cell lines. The article reports on Ah receptors agonists in UV-exposed toluene solutions of decabromodiphenyl ether and in soils contaminated with polybrominated diphenyl ethers. Mechanism-specific dioxin bioassays were used to study photolytic formation of Ah receptors agonists in toluene solutions of decaBDE. Soils were exposed to UV-light to study changes in Ah receptors agonist levels and laboratory experiments show that high levels of AhR agonists can be formed as photoproducts of decaBDE. Bioassays are important for studying environmental transformation processes where chemical analysis and toxicological evaluation requires available standard compounds and information on toxicological potency.

Several of the polychlorinated biphenyls (PCBs), i.e. the dioxin-like PCBs, are known to induce the P450 enzymes CYP1A1, CYP1A2 and CYP1B1 by activating the aryl hydrocarbon receptor (Ah)-receptor. We evaluated if circulating levels of PCBs in a population sample were related to genetic variation in the genes encoding these CYPs. In the population-based Prospective Investigation of the Vasculature in Uppsala Seniors (PIVUS) study (1016 subjects all aged 70), 21 SNPs in the CYP1A1, CYP1A2 and CYP1B1 genes were genotyped. Sixteen PCB congeners were analysed by high-resolution chromatography coupled to high-resolution mass spectrometry (HRGC/ HRMS). Of the investigated relationships between SNPs in the CYP1A1, CYP1A2 and CYP1B1 and six PCBs (congeners 118, 126, 156, 169, 170 and 206) that captures >80% of the variation of all PCBs measured, only the relationship between CYP1A1 rs2470893 was significantly related to PCB118 levels following strict adjustment for multiple testing ( p =0.00011). However, there were several additional SNPs in the CYP1A2 and CYP1B1 that showed nominally significant associations with PCB118 levels ( p -values in the 0.003–0.05 range). Further, several SNPs in the CYP1B1 gene were related to both PCB156 and PCB206 with p -values in the 0.005–0.05 range. Very few associations with p <0.05 were seen for PCB126, PCB169 or PCB170. Genetic variation in the CYP1A1 was related to circulating PCB118 levels in the general elderly population. Genetic variation in CYP1A2 and CYP1B1 might also be associated with other PCBs.
