In-home pet food testing has the benefit of yielding data which is directly applicable to the pet population. Validated and standardised in-home test protocols need to be available, and here we investigated key protocol requirements for an in-home canine food digestibility protocol. Participants were recruited via an online survey. After meeting specific inclusion criteria, sixty dogs of various breeds and ages received, during 14 consecutive days, a relatively low and high digestible complete dry extruded food containing titanium (Ti) dioxide. Both foods were given for 7 d in a cross-over design. Owners collected faeces daily allowing daily faecal Ti concentrations and digestibility of nitrogen (N), dry matter (DM), crude ash, organic matter (OM), crude fat (Cfat), starch and gross energy (GE) to be determined. Faecal Ti and digestibility values for all nutrients were not different ( P > 0·05) from the second day onwards after first consumption for both foods. One day of faecal collection yielded reliable digestibility values with additional collection days not reducing the confidence interval around the mean. Depending on the accepted margin of error, the food and the nutrient of interest, the minimal required sample size was between 9 and 43 dogs. Variation in digestibility values could in part be explained by a dog's neuter status (N, crude ash) and age (crude ash, Cfat) but not sex and body size. Future studies should focus on further identifying and controlling sources of variation to improve the in-home digestibility protocol and reduce the number of dogs required.

In current feed evaluation systems, the nutritional value of protein sources in diets for pigs is based on the ileal digestibility of protein and amino acids, which does not account for the kinetics of protein digestion along the gastrointestinal tract. The objective of the present study was to determine the in vitro protein digestion kinetics of different protein sources (soya bean meal (SBM), wheat gluten (WG), rapeseed meal (RSM), whey powder (WP), dried porcine plasma protein, yellow meal worm larvae and black soldier fly larvae (BSF)). Protein sources were incubated with pepsin at pH 3.5 for 0 to 90 min and subsequently with pancreatin at pH 6.8 for 0 to 210 min at 39°C. The in vitro protein digestion kinetics were described as the kinetics of nitrogen (N) solubilisation and the release of low molecular weight peptides (LMW) (<500 Da). The N solubilisation rate ranged from 0.025 min-1 for BSF to 0.685 min-1 for WP during the incubation with pepsin, and from 0.027 min-1 for RSM to 0.343 min-1 for WP during the incubation with pancreatin. The release rate of LMW peptides ranged from 0.027 min-1 for WG to 0.093 min-1 for WP during the incubation with pepsin, and from 0.029 min-1 for SBM to 0.385 min-1 for WP. Black soldier fly larvae showed a similar release rate of LMW peptides as WP during the incubation with pancreatin. At the end of the sequential incubation with pepsin (90 min) and pancreatin (210 min), WG and WP showed the highest percentage of N present in LMW peptides relative to total N (78% and 79%, respectively), whereas SBM showed the lowest (35%). In conclusion, protein sources for pig diets show substantial differences in in vitro protein digestion kinetics as measured by the kinetics of N solubilisation and the release of LMW peptides. The rate of release of LMW peptides was not correlated to the rate of N solubilisation for each of the protein sources evaluated.

In equines, Cr2O3 is widely accepted as an indigestible marker, but there are health concerns regarding the carcinogenic properties of Cr2O3. Recently, TiO2 has been suggested to be an alternative digestibility marker in equines. However, a comparison between Cr2O3 and TiO2 has not been made in equines. Six Welsh pony geldings (initial BW: 254±3 kg; 7 years of age) fed chopped alfalfa hay were used to evaluate the use of TiO2 (Ti) and Cr2O3 (Cr) as markers for calculating apparent digestibility and to investigate the effect of frequency of marker administration on the measurement of digestibility values. Diets contained 4.65 kg dry matter (DM) chopped alfalfa hay supplemented with minerals, vitamins, TiO2 (3.3 g Ti/day) and Cr2O3 (3.2 g Cr/day). Ponies were dosed with either 3.3 g Ti and 3.2 g Cr once daily (DF1) or with 1.65 g Ti and 1.60 g Cr twice daily (DF2). After adaptation to the diets and procedures for 14 days, voluntary voided faeces were collected quantitatively over 7 days and analysed for moisture, ash, Ti and Cr. Apparent total tract DM digestibility (DMD) and organic matter digestibility (OMD) were calculated using the total faecal collection (TFC) and marker method (Ti and Cr). The overall mean cumulative faecal recovery of Cr and Ti (as % of intake) were 102.0% and 96.6%, respectively. Mean daily faecal recoveries of Cr as well as of Ti were not different (P=0.323; P=0.808, respectively) between treatments. Overall daily faecal recovery of Cr differed (P=0.019) from 100% when the marker was dosed once daily, whereas overall daily faecal recovery was similar to 100% for both administration frequencies when Ti was used as a marker. For both markers, the coefficient of variation of the mean faecal marker recovery between horses was lower when the markers were administrated twice per day. Across treatments, cumulative DMD and OMD estimated with Ti were similar (P=0.345; P=0.418, respectively) compared with those values determined by TFC method. When Cr was used, the calculated cumulative DMD tended (P=0.097) to be greater compared with those estimated with TFC, and cumulative OMD values were overestimated (P=0.013). Orally supplemented Ti recovery in the faeces of ponies fed chopped alfalfa hay with Ti administered once or twice daily was close to 100%, making it the preferred marker for digestibility trials in equines.

The hypothesis was tested that butyrate presence in the digesta of distinct gastrointestinal tract (GIT) segments of broilers leads to differential effects on digesta retention time, gut morphology, and proteolytic enzymatic activities, ultimately resulting in differences in protein digestibility. A total of 320 male day-old Ross 308 broilers were randomly assigned to 5 dietary treatments: 1) control (no butyrate), 2) unprotected butyrate (main activity in the crop and gastric regions), 3) tributyrin (main activity in the small intestine), 4) fat-coated butyrate (activity in the whole GIT) and 5) unprotected butyrate combined with tributyrin, each replicated 8 times. Rapeseed meal was used in combination with a fine dietary particle size in order to challenge the digestive capacity of young broilers. Birds were dissected at 22, 23, and 24 d of age and samples of digesta at various GIT locations as well as tissues were collected. Butyrate concentration varied significantly across GIT segments depending on treatment, indicating that the dietary contrasts were successful. The apparent ileal digestibility of methionine tended to increase when butyrate and/or propionate was present in colonic and cecal contents, possibly due to modifications of GIT development and digesta transit time. Butyrate presence in the digesta of the crop, proventriculus and gizzard, on the contrary, decreased the apparent ileal digestibility of several amino acids (AA). In addition, butyrate presence beyond the gizzard elicited anorexic effect that might be attributable to changes in intestinal enteroendocrine L-cells secretory activities. Methodological aspects of digestibility measurements were studied in four Welsh pony geldings consuming haylage-based diets with increasing proportions of a pelleted concentrate according to a 4×4 Latin square design experiment. Ponies were fed four experimental, iso-energetic (net energy (NE) basis) diets (i.e. 22 MJ NE/day) with increasing proportions of a pelleted concentrate (C) in relation to haylage (H). The absolute amounts of diet dry matter fed per day were 4.48 kg of H (100H), 3.36 and 0.73 kg of H and C (75H25C), 2.24 and 1.45 kg of H and C (50H50C) and 1.12 and 2.17 kg of H and C (25H75C). Diets were supplemented with minerals, vitamins and TiO2 (3.7 g Ti/day). Voluntary voided faeces were quantitatively collected daily during 10 consecutive days and analysed for moisture, ash, ADL, acid-insoluble ash (AIA) and Ti. A minimum faeces collection period of 6 consecutive days, along with a 14-day period to adapt the animals to the diets and become accustomed to the collection procedure, is recommended to obtain accurate estimations on dry matter digestibility and organic matter digestibility (OMD) in equids fed haylage-based diets supplemented with concentrate. In addition, the recovery of AIA, ADL and Ti was determined and evaluated. Mean faecal recovery over 10 consecutive days across diets for AIA, ADL and Ti was 124.9% (SEM 2.9), 108.7% (SEM 2.0) and 97.5% (SEM 0.9), respectively. Cumulative faecal recovery of AIA significantly differed between treatments, indicating that AIA is inadequate to estimate the OMD in equines. In addition, evaluation of the CV of mean cumulative faecal recoveries obtained by AIA, ADL and Ti showed greater variations in faecal excretion of AIA (9.1) and ADL (7.4) than Ti (3.7). The accuracy of prediction of OMD was higher with the use of Ti than ADL. The use of Ti is preferred as a marker in digestibility trials in equines fed haylage-based diets supplemented with increasing amounts of pelleted concentrate.

Methodological aspects of digestibility measurements of feedstuffs for equines were studied in four Welsh pony geldings consuming four grass-hay diets in a 4 × 4 Latin square design. Diets contained either a low (L), medium (M), high (H), or very high (VH) ADF content (264, 314, 375, or 396 g·kg DM, respectively). Diets were supplemented with minerals, vitamins, and TiO (3.9 g Ti·d). Daily feces excreted were collected quantitatively over 10 consecutive days and analyzed for moisture, ash, ADL, AIA, and titanium (Ti). Minimum duration of total fecal collection (TFC) required for an accurate estimation of apparent organic matter digestibility (OMD) of grass hay was assessed. Based on literature and the calculated cumulative OMD assessed over 10 consecutive days of TFC, a minimum duration of at least 5 consecutive days of fecal collection is recommended for accurate estimation of dry matter digestibility (DMD) and OMD in ponies. The 5-d collection should be preceded by a 14-d adaptation period to allow the animals to adapt to the diets and become accustomed to the collection procedures. Mean fecal recovery over 10 d across diets for ADL, AIA, and Ti was 93.1% (SE 1.9), 98.9% (SE 5.5), and 97.1% (SE 1.8), respectively. Evaluation of CV of mean fecal recoveries obtained by ADL, AIA, and Ti showed that variation in fecal Ti (6.8) and ADL excretion (7.0) was relatively low compared to AIA (12.3). In conclusion, the use of internal ADL and externally supplemented Ti are preferred as markers to be used in digestibility trials in equine fed grass-hay diets.

In vitro gas production studies are routinely used to assess the metabolic capacity of intestinal microbiota to ferment dietary fibre sources. The faecal inocula used during the in vitro gas production procedure are most often obtained from animals adapted to a certain diet. The present study was designed to assess whether 19 days of adaptation to a diet are sufficient for faecal inocula of pigs to reach a stable microbial composition and activity as determined by in vitro gas production. Eighteen multiparous sows were allotted to one of two treatments for three weeks: a diet high in fibre (H) or a diet low in fibre (L). After this 3-week period, the H group was transferred to the low fibre diet (HL-treatment) while the L group was transferred to the diet high in fibre (LH-treatment). Faecal samples were collected from each sow at 1, 4, 7, 10, 13, 16 and 19 days after the diet change and prepared as inoculum used for incubation with three contrasting fermentable substrates: oligofructose, soya pectin and cellulose. In addition, inocula were characterised using a phylogenetic microarray targeting the pig gastrointestinal tract microbiota. Time after diet change had an effect (P<0.05) on total gas production for the medium-fast fermentable substrates; soya pectin and oligofructose. For the more slowly fermentable cellulose, all measured fermentation parameters were consistently higher (P<0.05) for animals in the HL-treatment. Diet changes led to significant changes in relative abundance of specific bacteria, especially for members of the Bacteroidetes and Bacilli, which, respectively, increased or decreased for the LH-treatment, while changes were opposite for the HL-treatment. Changing the diet of sows led to changes in fermentation activity of the faecal microbiota and in composition of the microbiota over time. Adaptation of the microbiota as assessed by gas production occurred faster for LH-animals for fast fermentable substrates compared with HL-animals. Overall, adaptation of the large intestinal microbiota of sows as a result of ingestion of low and high fibre diets seems to take longer than 19 days, especially for the ability to ferment slowly fermentable substrates.

Several in situ studies have been conducted on maize silages to determine the effect of individual factors such as maturity stage, chop length and ensiling of maize crop on the rumen degradation but the information on the relationship between chemical composition and in situ rumen degradation characteristics remains scarce. The objectives of this study were to determine and describe relationships between the chemical composition and the rumen degradation characteristics of dry matter (DM), organic matter (OM), CP, starch and aNDFom (NDF assayed with a heat stable amylase and expressed exclusive of residual ash) of maize silages. In all, 75 maize silage samples were selected, with a broad range in chemical composition and quality parameters. The samples were incubated in the rumen for 2, 4, 8, 16, 32, 72 and 336 h, using the nylon bag technique. Large range was found in the rumen degradable fractions of DM, OM, CP, starch and aNDFom because of the broad range in chemical composition and quality parameters. The new database with in situ rumen degradation characteristics of DM, OM, CP, starch and aNDFom of the maize silages was obtained under uniform experimental conditions; same cows, same incubation protocol and same chemical analysis procedures. Regression equations were developed with significant predictors (P<0.05) describing moderate and weak relationships between the chemical composition and the washout fraction, rumen undegradable fraction, potentially rumen degradable fraction, fractional degradation rate and effective rumen degradable fraction of DM, OM, CP, starch and aNDFom.

Marker methodologies to measure ileal and total tract digestibilities of diets varying in content and degradability of dietary fiber in broiler chickens were evaluated. Chromium sesquioxide (Cr2O3) and cobalt-ethylenediamine tetraacetic acid (Co-EDTA) were used as markers of solid and soluble fractions, respectively, and compared with digestibility values obtained with the total collection method. Groups of broilers (n = 17, 11 broilers/group) were assigned to a low-fiber diet or 1 of 2 high-fiber diets, the latter 2 containing 35% rapeseed meal (RSM). Pectolytic enzymes were added to one RSM diet to improve degradability of the fiber fraction. Excreta were quantitatively collected for 96 h, and contents from ileum and ceca were collected at slaughter at 29, 30, or 31 d of age. Chromium recovery in excreta ranged between 86 and 95%, whereas cobalt recovery was considerably lower (66 to 70%). Chromium:cobalt ratio was higher in ileal digesta than in feed. Hardly any chromium was found in the ceca, indicating that separation of the marker and specific digesta fractions occurs. Estimates of apparent total tract digestibility (ATTD) were lower when calculated using the marker method compared with the total collection method, particularly in high-fiber diets. Using Cr2O3 as a marker, differences were relatively small and effects due to enzyme addition were generally similar. Using Co-EDTA as a marker, ATTD of all components were lower compared with values obtained using the collection method (3 to 45 percentage units, P < 0.001), likely related to the low Co recovery. When estimating apparent ileal digestibility (AID), separation of marker and digesta resulted in unrealistically high estimates for the digestibility of nonglucose polysaccharides (54 to 66%), exceeding ATTD values by 16 to 42 percentage units. Moreover, the effect of pectolytic enzyme addition on the AID of nonglucose polysaccharides was in the opposite direction when compared with total collection. The data illustrate that fractionation of digesta, particularly in high-fiber diets, complicates accurate AID measurements in broilers, regardless of the choice of markers used., (© Poultry Science Association Inc.)

The apparent total tract and ileal digestibility assays to measure AA absorption in commercial canine diets were compared in the present study. Five ileal cannulated dogs were fed 5 commercial dry canine foods selected to contain 19 to 30% CP in a 5 × 5 Latin square design. Ileal and total tract digestibility (apparent and standardized) of DM, OM, CP, crude fat (CF), carbohydrate, and AA (including reactive Lys) were calculated using Cr2O3 as an indigestible marker. Greater apparent total tract digestibility values were found for DM, OM, and CP (P ≤ 0.034) compared with ileal digestibility values; however, CF (P = 0.058) had a greater ileal apparent digestibility. Apparent and standardized CP digestibility values were, respectively, 5.7 and 7.4 percentage units greater when measured over the total digestive tract compared with measurement at the ileum (P = 0.034 and 0.011, respectively). Ileal apparent digestibility for N of AA (P = 0.009) and most AA (P < 0.05), except for Met, Ile, Lys, Phe, and Ala, was decreased if measured at the ileum. However, correction for endogenous losses showed only Met digestibility did not differ between measurement sites. Differences between sites in excess of 15 percentage units were recorded for AA. Apparent and standardized ileal reactive Lys digestibility was 3.1 to 15.3 percentage units greater than corresponding total tract digestibility values. For several indispensable AA, the bioavailability estimates currently used by the 2006 NRC and the 2011 Association of American Feed Control Officials to derive allowance estimates for canine adult maintenance were greater than the digestibility values of these AA in the commercial dog foods evaluated. Although the canine large intestine is relatively short, the total tract digestibility assay in dogs can overestimate the digestibility of dietary AA and CP and may not be an accurate method for the measurement of absorption. In this study, bioavailability estimates of AA appeared to be less than those used to derive allowance estimates for commercial dog foods. Further work is required if current recommendations warrant adjustment.

Fractional passage rates are required to predict nutrient absorption in ruminants but data on nutrient-specific passage kinetics are largely lacking. With the use of the stable isotope ratio (δ) as an internal marker, we assessed passage kinetics of fiber and fiber-bound nitrogen (N) of intrinsically labeled grass silage from fecal and omasal excretion patterns of δ(13)C and δ(15)N. In a 6×6 Latin square, lactating dairy cows received grass silages [455 g/kg of total diet dry matter (DM) ] in a 2×3 factorial arrangement from ryegrass swards fertilized at low (45 kg of N/ha) or high (90 kg of N/ha) levels of N and harvested at 3 maturity stages. Feed intake (16.7±0.48 kg of DM/d; mean ± standard error of the mean) and milk yield (26.7±0.92 kg/d) increased at the high level of N fertilization and at decreasing maturity. Nutrient digestibility decreased with increasing plant maturity, particularly at the high level of N fertilization, essentially reflecting dietary treatment effects on the nutritional composition of the grass silage. Fractional rumen passage rates (K1) were highest and total mean retention time in the gastrointestinal tract (TMRT) was lowest when based on the external marker chromium mordanted fiber (Cr-NDF; 0.047/h and 38.0 h, respectively). Fecal δ(13)C in the acid detergent fiber fraction ((13)CADF) provided the lowest K1 (0.023/h) and the highest TMRT (61.1 h) and highest peak concentration time (PCT; 24.3h) among markers. In comparison, fecal fiber-bound N ((15)NADF) had a considerably higher K1 (0.032/h) and lower TMRT (46.4 h) than (13)CADF. Total N (measured with (15)NDM) had a comparable K1 (0.034/h) to that of (15)NADF but provided the highest fractional passage rates from the proximal colon-cecum (K2; 0.37/h) and lowest PCT (17.4 h) among markers. A literature review indicated unclear effects of grass silage maturity on K1 and unknown effects of N fertilization on K1. Our study indicated no effect of advancing maturity on fecal K1 and a trend for K1 to increase with the high level of N fertilization. Parameter K2 increased, whereas PCT and TMRT generally decreased with the high level of N fertilization. Omasal digesta sampling largely confirmed results based on fecal sampling. Goat milk is used as an alternative to cow milk for the production of infant formulas. However, little is known about the protein quality and, specifically, about the digestible AA pattern of goat milk formulas compared with their cow milk counterparts. In this study, the true ileal AA digestibility of a goat milk infant formula was compared with a premium cow milk infant formula. The 3-wk-old piglet was used as a model for the 3-mo-old infant. Both milk formulas were prepared as described by the manufacturer, with titanium dioxide added as an indigestible marker. The formulas were fed to the piglets over a 2-wk trial period. Digesta from the terminal ileum were collected post euthanasia and analyzed for AA content, along with samples of the formulas. True AA digestibility was determined after correcting for endogenous AA loss at the terminal ileum of pigs fed an enzyme-hydrolyzed casein-based diet, followed by ultrafiltration (5,000 Da) of the digesta. Total urine and feces collection was also undertaken to determine the nitrogen retention from the diets. The true ileal AA digestibility was similar between the goat and cow milk infant formulas for all AA except Gly and Trp. There was no significant difference in the nitrogen retention of piglets fed the two different formulas. The goat milk infant formula and the premium cow milk infant formula were similar in terms of protein quality.

The influence of sex of broilers and dietary phosphorus (P) level on energy utilization and apparent ileal digestibility of N, P and phytate-P were investigated. The AME(N) was determined using 3- and 6-week old broilers, while the apparent ileal digestibility were determined only with 6-week old birds. Sex of broilers had no effect on the AME(N) values determined during week 3. During week 6, the AME(N) values for male broilers were higher (P < 0.01) than those for the females. An interaction (P < 0.05) between sex and dietary P level was also observed. AME(N) values determined with male broilers were lower in the adequate-P diet compared to those in the low-P diet, whereas no effect of P level was observed in females. Female broilers tended (P < 0.10) to have a higher ileal N digestibility than the males, but a significant (P < 0.01) sex x P level interaction was observed. Males receiving the adequate-P diet had a lower N digestibility than those receiving the low-P diet, whereas the opposite was true in the females. Ileal phytate P degradation in males was higher than in females (0.282 vs. 0.234), but the differences were not significant. Increasing dietary P level increased ileal P digestibility in both the males and females, but the improvements were greater in the females, as indicated by a significant sex x P level interaction.

1. The influence of a microbial phytase on the performance, toe ash contents and nutrient utilisation of male broilers fed diets based on maize and wheat was investigated. The experiment was conducted as 2 x 2 x 2 factorial arrangement of treatments. Within the factorial, two diet types (maize-soy or wheat-soy) containing two levels of non-phytate phosphorus (3.0 or 4.5 g/kg) were evaluated and each level of non-phytate phosphorus was supplemented with 0 or 500 PU phytase/kg diet. Each of the 8 dietary treatments were fed to 6 pens of 8 birds from d 1 to 21 post-hatching. 2. Main effects of diet type and phytase were observed for all parameters. Main effect of non-phytate phosphorus was significant only for feed/gain and toe ash contents. Phytase addition improved weight gains irrespective of diet type or non-phytate phosphorus level, but the magnitude of improvement in the phosphorus-deficient wheat-soy diet was greater, resulting in a diet type x non-phytate phosphorus interaction. Responses in toe ash contents were noted only in phosphorus-deficient diets, as indicated by a non-phytate phosphorus x phytase interaction. 3. Phytase addition improved apparent metabolisable energy values of wheat-based diets, but had little effect on the apparent metabolisable energy of maize-based diets as shown by a diet type x phytase interaction. The apparent metabolisable energy was not influenced by dietary non-phytate P. 4. Phytase improved ileal nitrogen digestibility in both diet types, but the responses to added phytase tended to be higher in wheat-based diets, as shown by a diet type x phytase interaction. 5. Increasing the dietary non-phytate phosphorus level reduced phosphorus digestibility and increased excreta phosphorus content. Addition of phytase improved phosphorus digestibility, but the increments were higher in low phosphorus diets resulting in a non-phytate phosphorus x phytase interaction. Phytase addition tended to lower the excreta phosphorus content, but the effects were greater in birds fed low phosphorus diets, as shown by a non-phytate phosphorus x phytase interaction.

The article discusses the nutritional value of vegetable feed ingredients used in swine diets. It refers to protein structural changes during the process of vegetable feed ingredients and outlines its impact on protein digestibility. It examines the effect of shear on protein structure using extrusion and mentioned the nutritional consequences of the processing condition.

An experiment was conducted to determine protein quality in processed protein sources using the content of AA, o-methylisourea (OMIU)-reactive Lys, Maillard reaction products (MRP), and cross-link products; the standardized ileal digestibility (SID) of CP and AA; and growth performance in growing pigs as criteria. Differences in protein quality were created by secondary toasting (at 95°C for 30 min) of soybean meal (SBM) and rapeseed meal (RSM) in the presence of lignosulfonate resulting in processed SBM (pSBM) and processed RSM (pRSM). The processing treatment was used as a model for overprocessed protein sources. Ten growing pigs were each fed 1 of the 4 diets containing SBM, pSBM, RSM, or pRSM in each of 3 periods. Ileal chyme was collected at the end of each period and analyzed for CP, AA, and OMIU-reactive Lys. Diets were analyzed for furosine and carboxymethyllysine (CML) as an indicator for MRP and lysinoalanine (LAL), which is a cross-link product. The SBM and RSM diets contained furosine, CML, and LAL, indicating that the Maillard reaction and cross-linking had taken place in SBM and RSM, presumably during the oil extraction/desolventizing process. The amounts of furosine, CML, and LAL were elevated in pSBM and pRSM due to further processing. Processing resulted in a reduction in total and OMIU-reactive Lys contents and a decrease in G:F from 0.52 to 0.42 for SBM and 0.46 to 0.39 for RSM (P = 0.006), SID of CP from 83.9 to 71.6% for SBM and 74.9 to 64.6% for RSM (P < 0.001), and SID of AA (P < 0.001), with the largest effects for total and OMIU-reactive Lys. The effects of processing could be substantial and should be taken into account when using processed protein sources in diets for growing pigs. The natural diet of felids contains highly digestible animal tissues but also fractions resistant to small intestinal digestion, which enter the large intestine where they may be fermented by the resident microbial population. Little information exists on the microbial degradability of animal tissues in the large intestine of felids consuming a natural diet. This study aimed to rank animal substrates in their microbial degradability by means of an in vitro study using captive cheetahs fed a strict carnivorous diet as fecal donors. Fresh cheetah fecal samples were collected, pooled, and incubated with various raw animal substrates (chicken cartilage, collagen, glucosamine-chondroitin, glucosamine, rabbit bone, rabbit hair, and rabbit skin; 4 replicates per substrate) for cumulative gas production measurement in a batch culture technique. Negative (cellulose) and positive (casein and fructo-oligosaccharides; FOS) controls were incorporated in the study. Additionally, after 72 h of incubation, short-chain fatty acids (SCFA), including branched-chain fatty acids (BCFA), and ammonia concentrations were determined for each substrate. Glucosamine and glucosamine-chondroitin yielded the greatest organic matter cumulative gas volume (OMCV) among animal substrates (P < 0.05), whereas total SCFA production was greatest for collagen (P < 0.05). Collagen induced an acetate production comparable with FOS and a markedly high acetate-to-propionate ratio (8.41:1 ) compared with all other substrates ( 1.67:1 to 2.97:1 ). Chicken cartilage was rapidly fermentable, indicated by a greater maximal rate of gas production (Rmax) compared with all other substrates (P < 0.05). In general, animal substrates showed an earlier occurrence for maximal gas production rate compared with FOS. Rabbit hair, skin, and bone were poorly fermentable substrates, indicated by the least amount of OMCV and total SCFA among animal substrates (P < 0.05). The greatest amount of ammonia production among animal substrates was measured after incubation of collagen and rabbit bone (P < 0.05). This study provides the first insight into the potential of animal tissues to influence large intestinal fermentation in a strict carnivore, and indicates that animal tissues have potentially similar functions as soluble or insoluble plant fibers in vitro. Further research is warranted to assess the impact of fermentation of each type of animal tissue on gastro-intestinal function and health in the cheetah and other felid species. Availability of amino acids include digestibility, chemical integrity (i.e., absorbed in an utilizable form) and freedom from interference in metabolism (i.e., presence of specific dietary anti-nutritional factor limiting protein synthesis). The greatest source of variation in amino acid availability is usually digestibility. The most susceptible amino acid to heat damage is Lys which is involved via its ε-amino group in the Maillard reaction. Lysine can also react with other amino acid side chains such as the reaction with cysteine to form lysinoalanine. Analysis of reactive lysine allows accurate estimation of the available Lys content of heat-damaged ingredients although recent research has identified some inaccuracies in the reactive Lys assay. Met and Cys can be oxidised via their side chain sulphur atom upon heat treatments with some of the oxidised forms being poorly available. Heating of proteins can convert the L- to the D-enantiomer of an amino acid (racemization), the latter which is poorly available. Free asparagine, present in relatively large amounts in certain plant-based feedstuffs, can form acrylamide during heat processing. Before any feed processing occurs, various feed ingredients used in animal nutrition have already been processed to a smaller or larger extent which often affects the bioavailability of amino acids. Examples include soybean meal, DDGS, rapeseed meal but also commonly used grains (e.g. corn and wheat).
