Forages cut in the afternoon (p.m.) generally yield a higher concentration of nonstructural carbohydrates (NSC) than those cut in the morning (a.m.). We aimed to compare the effects of p.m.-cut red clover baleage (p.m.-RC) versus a.m.-cut RC baleage (a.m.-RC) on milk yield, concentrations and yields of milk components, and apparent total-tract digestibility of nutrients in late-lactation Holstein cows. Twelve multiparous and 2 primiparous Holstein cows received a total mixed ration containing, on a dry matter (DM) basis, 65% p.m.-RC or a.m.-RC plus 35% concentrate in a crossover design with 14 d for diet adaptation and 7 d for data and sample collection. One RC field was split in 2 with the first half cut in the afternoon (1600 h) and the second half in the following morning (0600 h). The p.m.-RC and a.m.-RC contained (% of DM): 12.6 versus 9.43% NSC in samples collected before the beginning of the experiment and 7.49 versus 7.79% NSC in samples collected during the sampling periods (i.e., d 14 to 21). The total mixed rations averaged 18.2 and 17.5% NSC for the p.m.-RC and a.m.-RC, respectively. Feeding p.m.-RC or a.m.-RC did not improve feed intake or milk yield and composition in late-lactation dairy cows. However, milk urea N and plasma urea N were both lowest in cows offered p.m.-RC. With the exception of the apparent total-tract digestibility of DM, which was highest in cows fed p.m.-RC, no other changes in nutrient digestibility were observed. Similarly, no treatment effect was observed for the urinary excretion of N and purine derivatives. Maceration and evening-cutting are 2 forage management techniques that have independently improved forage quality and nutrient utilization in ruminants, but have not been evaluated in combination. Using a dual-flow continuous culture fermenter system, this preliminary study was designed to evaluate the individual and combined effects of time of cutting and maceration on in vitro ruminal digestion, nutrient flows, and microbial protein synthesis. Forages were harvested as hay from a timothy (Phleum pratense L.)-birdsfoot trefoil (Lotus corniculatus L.) stand in the morning (AM) or evening (PM). Half of each morning- and evening-cut treatment was macerated (AM-M, PM-M). The chemical composition (DM, OM, CP, NDF, ADF), including nonstructural carbohydrates (NSC) and water-soluble carbohydrates (WSC), was determined for each of the 4 treatments (AM, AM-M, PM, PM-M). Forages were ground to 2 mm and allocated to separate fermenters at 60 g of DM/d in a 4 × 4 Latin square design. Fermenters were operated over four 10-d periods with the first 7 d for adaptation followed by 3 d of sampling. Evening-cutting enhanced the apparent digestibility of NDF (P = 0.02) and ADF (P = 0.006), with a tendency (P < 0.10) for improved true DM digestibility and microbial protein synthesis. Molar proportions of individual VFA were not affected (P > 0.10) by time of cutting, though evening-cutting increased (P = 0.02) total concentration of VFA. Maceration had no effect (P > 0.10) on true nutrient digestibility or microbial protein synthesis. An interaction of time of cutting and maceration (P < 0.05) was observed whereby maceration decreased true DM and OM digestibilities in evening-cut treatments, but had no effect in morning-cut treatments. Similarly, maceration reduced total N supply (P < 0.001) and molar proportions of acetate (P = 0.04) and increased molar proportions of propionate (P = 0.01) in evening-cut treatments with no effect on morning-cut treatments. These results indicate that independent use of evening cutting increased fiber digestibility and total VFA concentration, and independent use of maceration shifted molar proportions of VFA toward glucogenic fermentation. The combined use of these management techniques afforded no improvement for in vitro digestibility or metabolism when applied to morning-cut hay, and decreased nutrient digestibility when applied to evening-cut hay. Due to inherent limitations of in vitro systems, the results of this study should be interpreted with caution. Further in vivo studies are needed to support our conclusions.

Alfalfa (Medicago sativa L.) cut at sundown has been shown to contain greater concentration of total nonstructural carbohydrates (TNC) than that cut at sunup. Fourteen multiparous (8 ruminally cannulated) and 2 primiparous lactating dairy cows were randomly assigned to 2 treatments in a crossover design (2 periods of 24 d) to investigate the effects of alfalfa daytime cutting management on ruminal metabolism, nutrient digestibility, N balance, and milk yield. Half of each alfalfa field (total of 3 fields) was cut at sundown (PM) after a sunny day, whereas the second half was cut at sunup (AM) on the following day. Both PM and AM cuts were field-wilted and harvested as baleage (531 +/- 15.0 g of dry matter/kg of fresh matter). Bales (PM and AM) were ranked according to their concentrations of TNC, paired, and each pair of PM and AM baleages was then assigned to each experimental day (total of 48 d). The difference in TNC concentration between PM and AM baleages fed during the 10 d of data and sample collection varied from -10 to 50 g/kg of dry matter. Each pair of baleage was fed ad libitum to cows once daily with no concentrate. Ruminal molar proportion of acetate and total volatile fatty acid concentration were greater in animals fed the AM baleage, whereas the proportion of valerate was greater with PM baleage; no other significant changes in ruminal molar proportions of volatile fatty acids were observed between forage treatments. Digestible organic matter intake, organic matter digestibility, and plasma Lys concentration were significantly greater in cows fed PM alfalfa, suggesting that more nutrients were available for milk synthesis. Significantly lower body weight gain and retained N as a proportion of N intake were observed in cows fed PM alfalfa, thus suggesting that nutrients were channeled to milk synthesis rather than to body reserves. Intake of dry matter (+1.0 kg/d), and yields of milk (+1.0 kg/d), milk fat (+70 g/d), and milk protein (+40 g/d) were significantly greater in cows fed PM vs. AM alfalfa. Concentration of milk urea N and excretion of urea N as a proportion of total urinary N were significantly reduced, and milk N efficiency was increased when feeding PM vs. AM alfalfa, indicating an improvement in N utilization. Increasing the TNC concentration of alfalfa by shifting forage cutting from sunup to sundown improved N utilization and milk production in late-lactation dairy cows.

Six ruminally fistulated midlactating multiparous Holstein cows were used in a double 3 x 3 Latin square design (35-d periods) to study the effects of lasalocid (LAS) and monensin (MON) supplemented at 24 mg/ kg of dry matter on digestion, ruminal fermentation, blood metabolites, and milk production. Cows were blocked according to milk production and fed a red clover silage-based total mixed ration (17.8% crude protein) without supplementation or supplemented with LAS or MON. Daily dry matter intake, milk production, and milk fat and protein concentrations were similar among treatments and averaged 23.5 kg, 36.6 kg, 3.36%, and 3.38%, respectively. Rumen lipogenic:glucogenic volatile fatty acids and NH(3)-N concentration were lower, and apparent digestibility of dry matter, organic matter, crude protein, and gross energy were higher with than without ionophore supplementation. Compared with LAS, MON increased concentrations of plasma urea-N and milk urea-N, and excretion of urinary urea-N and total N. Monensin also decreased N retention and tended to reduce plasma concentration of nonessential AA in comparison with LAS. Both ionophores reduced daily fecal excretion of N by 13 g compared with the control, but MON increased daily losses of urinary N by 36 g compared with LAS. Results from this study suggest that postabsorptive metabolism of N might be altered by the type of ionophore fed.

Four Holstein cows fitted with ruminal cannulas were used in a 4 x 4 Latin square design (28-d periods) with a 2 x 2 factorial arrangement of treatments to investigate the effects of addition of a specific mixture of essential oil compounds (MEO; 0 vs. 750 mg/d) and silage source [alfalfa silage (AS) vs. corn silage (CS)] on digestion, ruminal fermentation, rumen microbial populations, milk production, and milk composition. Total mixed rations containing either AS or CS as the sole forage source were balanced to be isocaloric and isonitrogenous. In general, no interactions between MEO addition and silage source were observed. Except for ruminal pH and milk lactose content, which were increased by MEO supplementation, no changes attributable to the administration of MEO were observed for feed intake, nutrient digestibility, end-products of ruminal fermentation, microbial counts, and milk performance. Dry matter intake and milk production were not affected by replacing AS with CS in the diet. However, cows fed CS-based diets produced milk with lower fat and higher protein and urea N concentrations than cows fed AS-based diets. Replacing AS with CS increased the concentration of NH(3)-N and reduced the acetate-to-propionate ratio in ruminal fluid. Total viable bacteria, cellulolytic bacteria, and protozoa were not influenced by MEO supplementation, but the total viable bacteria count was higher with CS- than with AS-based diets. The apparent digestibility of crude protein did not differ between the AS and CS treatments, but digestibilities of neutral detergent fiber and acid detergent fiber were lower when cows were fed CS-based diets than when they were fed AS-based diets. Duodenal bacterial N flow, estimated using urinary purine derivatives and the amount of N retained, increased in cows fed CS-based diets compared with those fed AS-based diets. Feeding cows AS increased the milk fat contents of cis-9, trans-11 18:2 (conjugated linoleic acid) and 18:3 (n-3 fatty acid) compared with feeding cows CS. Results from this study showed limited effects of MEO supplementation on nutrient utilization, ruminal fermentation, and milk performance when cows were fed diets containing either AS or CS as the sole forage source.

Four ruminally cannulated, lactating Holstein cows were used in a 4 x 4 Latin square design (28-d periods) with a 2 x 2 factorial arrangement of treatments to study the effects of dietary addition of essential oils (0 vs. 2 g/d; EO) and monensin (0 vs. 350 mg/d; MO) on digestion, ruminal fermentation characteristics, milk production, and milk composition. Intake of dry matter averaged 22.7 kg/d and was not significantly affected by dietary additives. Apparent digestibilities of dry matter, organic matter, neutral detergent fiber, and starch were similar among treatments. Apparent digestibility of acid detergent fiber was increased when diets were supplemented with EO (48.9 vs. 46.0%). Apparent digestibility of crude protein was higher for cows fed MO compared with those fed no MO (65.0 vs. 63.6%). Nitrogen retention was not changed by additive treatments and averaged 27.1 g/d across treatments. Ruminal pH was increased with the addition of EO (6.50 vs. 6.39). Ruminal ammonia nitrogen (NH3-N) concentration was lower with MO-supplemented diets compared with diets without MO (12.7 vs. 14.3 mg/100 mL). No effect of EO and MO was observed on total volatile fatty acid concentrations and molar proportions of individual volatile fatty acids. Protozoa counts were not affected by EO and MO addition. Production of milk and 4% fat-corrected milk was similar among treatments (33.6 and 33.4 kg/d, respectively). Milk fat content was lower for cows fed MO than for cows fed diets without MO (3.8 vs. 4.1%). The reduced milk fat concentration in cows fed MO was associated with a higher level of trans-10 18:1, a potent inhibitor of milk fat synthesis. Milk urea nitrogen concentration was increased by MO supplementation, but this effect was not apparent when MO was fed in combination with EO (interaction EO x MO). Results from this study suggest that feeding EO (2 g/d) and MO (350 mg/d) to lactating dairy cows had limited effects on digestion, ruminal fermentation characteristics, milk production, and milk composition.

Four lactating Holstein cows with ruminal and duo-denal cannulas were used in a 4 x 4 latin square design to determine the effects of feeding heat-treated flaxseed on ruminal fermentation and site and extent of nutrient utilization. Four diets were formulated: a control diet with no flaxseed, a raw flaxseed diet (RF), a micronized flaxseed diet (MF), and an extruded flaxseed diet (EF). Flaxseed diets contained 12.6% flaxseed (dry matter [DM] basis). Ruminal pH, NH3 N, and total concentration of volatile fatty acids were not affected by dietary treatments. However, feeding flaxseed decreased the molar proportion of acetate and increased that of propionate. Flaxseed supplementation had no effect on ruminal digestion of DM, organic matter (OM), neutral detergent fiber (NDF), crude protein (CP), fatty acids (FA), and gross energy. However, ruminal digestion of acid detergent fiber (ADF) was lower for cows fed the flaxseed diets than for cows fed the control diet. Feeding flaxseed tended to increase post-ruminal and total tract digestibilities of DM, OM, NDF, and gross energy. Feeding heat-treated flaxseed diets relative to RF had no effect on ruminal, post-ruminal, and total tract nutrient digestibilities. Cows fed EF had higher ruminal and lower post-ruminal digestibilities of DM, OM, ADF, CP, and FA than cows fed MF. However, total tract digestibilities were similar for the 2 heat treatments. It was concluded that flaxseed supplementation improved total tract nutrient utilization with no adverse effects on ruminal fermentation. Extrusion failed to protect flaxseed from ruminal digestion. However, micronization can be used to increase the ruminal undegraded protein value of flaxseed.

We designed a switchback expriment to compare the intestinal disappearance of ruminally protected Met determined in situ or in vivo over three consecutive periods. Four nonlactating Holstein heifers (477+/-36 kg) with cannulae in the rumen, duodenum, and ileum were fed a diet based on timothy silage to meet requirements for maintenance (dry matter intake = 8.72+/-0.15 kg). A total of 16 bags, containing 1.5 g of ruminally protected Met, were incubated in the rumen (4.5 h) of each cow and transferred to an acid-pepsin solution to simulate the abomasum (2.5 h). Following each incubation, bags were recovered and three bags were dried at 55 degrees C and analyzed for Met. Remaining bags were introduced directly into the duodenal or ileal cannula for the in situ method, while for the in vivo method, the content of remaining bags was transferred into gelatin capsules before their introduction in the duodenal or ileal cannula. Spot samples of digesta were collected during a 96-h period, with Co-EDTA and Cr-mordanted fiber used as indigestible markers to estimate in vivo digestibility. The disappearance of Met in the small intestine determined in vivo tended to be higher than in situ (74.45 vs. 43.65+/-1.79%). Our results indicate that when used to assess intestinal availability of ruminally protected Met, the mobile nylon bag technique can underestimate the true bioavailability of Met.

In an attempt to develop a non-invasive, accurate technique to determine the effect of treatment of soybean meal (SBM) on lysine (Lys) availability, the increment in plasma Lys irreversible loss rate (ILR) was compared with the increment in Lys intestinal flow and apparent digestibility. Four multiparous Holstein cows (173 DIM) with ruminal and duodenal cannulas were used in a 4 x 4 Latin square with 14-d periods. They were fed either solvent extracted SBM (SE), expeller SBM (EP) or lignosulfonate treated SBM (LS) as 23% of the diet. The fourth treatment (SE20) consisted in a 20 mmol/h omasal infusion of Lys to cows fed the SE diet. On the last day of each period, a pulse dose of [2-15 N] Lys was given in the jugular vein and jugular plasma samples were collected at 1, 2, 3, 4, 5, 6, 7, 8, 9, 10, 13, 16, 19, 25 and 31 min after the injection. Chromic oxide was included as an indigestible marker to asses DM flow and 8 separated digesta samples were collected on d 13 and 14 and pooled by period. The decay curve of 15 N Lys enrichment was explained by a two exponential equation. The plasma Lys ILR was not different (P>0.10) between the different SBM (84, 89 and 80 ± 3.9 mmol/h for SE, EP and LS, respectively). However, the SE20 treatment increased (P<0.04) Lys ILR to 104 mmol/h, i.e. exactly 20 mmol/h above the SE treatment. No differences were observed on duodenal Lys flow (213, 203 and 213± 11.9 g/d) or apparent intestinal digestion (79, 75 and 77 ± 1.8 %) between the SBM diets. Plasma Lys ILR was an accurate technique to estimate the increment of Lys availability, as it responded quantitatively to Lys infusion. Although the treatment of SBM should have increased Lys availability (NRC 2001), differences were too small or too variable to be detected. Further studies should be performed to test the sensitivity of this method, increasing the power of detection with larger amount of animals. [ABSTRACT FROM AUTHOR]
