Diatoms are one of the dominant groups in phytoplankton communities of the western Antarctic Peninsula (WAP). Although generally well-studied, little is known about size dependent photophysiological responses in diatom bloom formation and succession. To increase this understanding, four Antarctic diatom species covering two orders of magnitude in cell size were isolated in northern Marguerite Bay (WAP). Fragilariopsis sp., Pseudo-nitzschia cf. subcurvata, Thalassiosira cf. antarctica, and Proboscia cf. alata were acclimated to three different irradiances after which photophysiology, electron transport, carbon fixation, and growth were assessed. The small species Fragilariopsis sp., Pseudo-nitzschia cf. subcurvata, and large species Proboscia cf. alata showed similar photoacclimation to higher irradiances with a decrease in cellular chlorophyll a and an increase in chlorophyll a specific absorption and xanthophyll cycle pigments and activity. In contrast, pigment concentrations and absorption remained unaffected by higher irradiances in the large species Thalassiosira cf. antarctica. Overall, the small species showed significantly higher growth rates compared to the large species, which was related to relatively high light harvesting capacity and electron transport rates in the smaller species. However, photophysiological responses related to photoinhibition and photoprotection and carbon fixation showed no relationship with cell size. This study supports the dominance of small diatoms at low irradiances during winter and early spring, but does not provide photophysiological evidence for the dominance of large diatoms during the phytoplankton bloom in the WAP. Photoacclimation properties were investigated in two marine microalgae exposed to four ambient irradiance conditions: static photosynthetically active radiation (PAR: 400-700 nm), static PAR + UVR (280-700 nm), dynamic PAR and dynamic PAR + UVR. High light acclimated cultures of Thalassiosira weissflogii and Dunaliella tertiolecta were exposed outdoors for a maximum of 7 days. Dynamic irradiance was established by computer controlled vertical movement of 2 L bottles in a water filled basin. Immediate (<24 h), short-term (1-3 days) and long-term (4-7 days) photoacclimation was followed for antioxidants (superoxide dismutase, ascorbate peroxidase and glutathione cycling), growth and pigment pools. Changes in UVR sensitivity during photoacclimation were monitored by measuring UVR-induced inhibition of carbon assimilation under standardized UV conditions using an indoor solar simulator. Both species showed immediate antioxidant responses due to their transfer to the outdoor conditions. Furthermore, upon outdoor exposure, carbon assimilation and growth rates were reduced in both species compared with initial conditions; however, these effects were most pronounced in D. tertiolecta. Outdoor UV exposure did not alter antioxidant levels when compared with PAR-only controls in both species. In contrast, growth was significantly affected in the static UVR cultures, concurrent with significantly enhanced UVR resistance. We conclude that antioxidants play a minor role in the reinforcement of natural UVR resistance in T. weissflogii and D. tertiolecta.

The synergistic effects of iron limitation and irradiance dynamics on growth, photosynthesis, antioxidant activity and excessive PAR (400-700 nm) and UV (280-400 nm) sensitivity were investigated for the Antarctic marine diatom Chaetoceros brevis. Iron-limited and iron-replete cultures were exposed to identical daily irradiance levels, supplied as dynamic (20-1350 micromol m(-2) s(-1)) and constant (260 micromol m(-2) s(-1)) irradiance. After acclimation, growth, maximal quantum yield of PSII (F(v)/F(m)), pigment composition, and the activities of the antioxidant enzymes superoxide dismutase (SOD), ascorbate peroxidase (APX) and glutathione reductase (GR) were determined. Then, excessive irradiance sensitivity was assessed by monitoring pigment composition, F(v)/F(m) and viability loss during a single excessive PAR and UV treatment. Iron limitation reduced growth rates, F(v)/F(m) dynamics, and cellular pigments pools. Cellular pigment concentrations were higher under dynamic irradiance than under constant irradiance but this difference was less pronounced under iron limitation compared to iron-replete conditions. SOD and APX activities increased during dynamic irradiance under iron limitation, suggesting increased radical formation around PSII. Despite these physiological differences, no effects on growth were observed between constant and dynamic irradiance cultivation in iron-limited and iron-replete cells. The applied culturing conditions did not affect glutathione reductase activity in C. brevis. F(v)/F(m) and xanthophyll de-epoxidation dynamics during excessive irradiance were not different for iron-limited and replete cells and viability loss was not found during excessive irradiance. This study revealed photoacclimation differences between iron-limited and iron-replete C. brevis cultures that did not affect growth rates and excessive irradiance sensitivity after acclimation to constant and dynamic irradiance.

Iron (Fe) is a key limiting nutrient for Southern Ocean phytoplankton. Input of Fe into the Southern Ocean is projected to change due to global warming, yet the combined effects of a concurrent increase in temperature with dissolved Fe (dFe) addition on phytoplankton growth and community composition have not been extensively studied. To improve our understanding of how Antarctic phytoplankton communities respond to Fe and enhanced temperature, we performed four full factorial onboard bioassays under trace-metal-clean conditions with phytoplankton communities from different regions of the Weddell Sea and the Amundsen Sea in the Southern Ocean. Treatments consisted of 2 nM Fe addition with 2 °C warming (TF), Fe addition at in situ temperature (F) + 2 °C warming with no Fe addition (T) and a control at in situ temperature with no Fe addition (control, C). Temperature had a limited effect by itself but boosted the positive response of the phytoplankton to Fe addition. Photosynthetic efficiency, phytoplankton abundances and chlorophyll a concentrations typically increased (significantly) with Fe addition (F and/or TF treatment), and the phytoplankton community generally shifted from haptophytes to diatoms upon Fe addition. The < 20 µ m phytoplankton fraction displayed population-specific growth responses, resulting in a pronounced shift in community composition and size distribution (mainly towards larger-sized phytoplankton) for the F and TF treatments. Such a distinct enhanced impact of dFe supply with warming on Antarctic phytoplankton size, growth and composition will likely affect trophic transfer efficiency and ecosystem structure, with potential significance for the biological carbon pump. Effects of prolonged darkness on Arctic phytoplankton composition were investigated with lab experiments and a pigment time series in Kongsfjorden, Spitsbergen (78°55′N). Chlorophyll a (Chl a), pigment composition, particulate organic carbon, cell numbers, and photosynthetic characteristics were studied in Arctic diatoms (Thalassiosira antarctica, Thalassiosira nordenskioeldii) and flagellates (Rhodomonas sp., Micromonas sp.) during 8 weeks of darkness and subsequent recovery in irradiance. Loss of photosynthetic functionality after 2 weeks of darkness was reversible in all species when returned to irradiance. Diatoms were more resistant to prolonged darkness (> 2 weeks) compared to the flagellates, with lower decline rates of Chl a and maximum quantum yield of PSII. T. nordenskioeldii showed rapid growth during recovery throughout 8 weeks of dark incubation, whereas recovery of flagellates diminished within 4 weeks. Ratios of taxonomic marker pigments relative to Chl a of all species showed limited variation during 8 weeks of dark incubation. The experimentally observed enhanced dark survival of diatoms was in agreement with pigment observations during four polar nights (2013–2017) in Kongsfjorden, which showed increased relative diatom abundance during declining biomass (down to 0.02 mg Chl a m−3). Therefore, a period of prolonged darkness gives Arctic diatoms a head start during the early stages of the spring bloom. The taxon‐specific survival traits can influence the geographical distribution of diatoms and flagellates within the polar oceans and their phenology. Abstract: Enhanced or reduced pCO2 (partial pressure of CO2) may affect the photosynthetic performance of marine microalgae since changes in pCO2 can influence the activity of carbon concentrating mechanisms, modulate cellular RuBisCO levels or alter carbon uptake efficiency. In the present study we compared the photophysiology of the Antarctic diatom Chaetoceros brevis at two pCO2 extremes: 750ppmv (2× ambient) and 190ppmv (0.5× ambient) CO2. Cultures were acclimated to four irradiance regimes: two regimes simulating deep or shallow vertical mixing, and two regimes mimicking limiting and saturating stable water column conditions. Then, growth rate, pigmentation, RuBisCO large subunit expression, RuBisCO activity, photosynthesis vs irradiance curves, effective quantum yield of PSII (Fv/Fm), and POC were measured. The four irradiance regimes induced a suite of photophysiological responses, ranging from low light acclimation to efficient photoprotection. Growth was reduced under the low constant and the deep mixing regime, compared to the shallow mixing and the stable saturating regime. Low stable irradiance resulted in higher light harvesting pigment concentrations, lower RuBisCO activity and a lower light saturation point (Ek) compared to the other irradiance regimes. Highest RuBisCO activity as well as P max levels was measured in the shallow mixing regime, which received the highest total daily light dose. Photoprotection by xanthophyll cycling was observed under all irradiance regimes except the low stable irradiance regime, and xanthophyll cycle pool sizes were higher under the dynamic irradiance regimes. For the fluctuating irradiance regimes, Fv/Fm was hardly affected by previous excess irradiance exposure, suggesting minimal PSII damage. No significant differences between the two pCO2 levels were found, with respect to growth, pigment content and composition, photosynthesis, photoprotection and RuBisCO activity, for all four irradiance regimes. Short-term photosensitivity and oxidative stress responses were compared for three groups of marine microalgae: Antarctic microalgae, temperate diatoms and temperate flagellates. In total, 15 low-light-acclimated species were exposed to simulated surface irradiance including ultraviolet radiation (SSI). Photosensitivity was assessed as the rate of recovery of Fv/Fm in the hours following SSI treatment. Before, during and after the SSI treatment, oxidative stress responses were assessed by following xanthophyll content and cycling, and activities of superoxide dismutase, ascorbate peroxidase and glutathione reductase, and glutathione redox status. When acclimated to low irradiance, antioxidant levels were not group specific. Superoxide dismutase activity was positively correlated with cell size, whereas in general, ascorbate peroxidase activity appeared to be lower and glutathione redox status appeared to be higher in the Antarctic than in the temperate species. After SSI exposure, the strong inhibition of PSII was followed by variable rates of recovery, although four species remained photosynthetically inactive. SSI tolerance appeared unrelated to geographic or taxonomic background, or to cell size. PSII recovery was enhanced in species with decreasing superoxide dismutase activity, glutathione redox status and increased xanthophyll cycle activity. We conclude that antioxidant responses are highly species specific and not related to the geographic or taxonomic background. Furthermore, xanthophyll cycling seems more important than antioxidants. The enzyme superoxide dismutase (SOD) holds a key position in the microalgal antioxidant network. The present research focused on oxidative stress responses in the Antarctic diatom Chaetoceros brevis F. Schütt during transition to excess (including ultraviolet radiation [UVR]) and limiting irradiance conditions. Over a 4 d period, cellular responses of thiobarbituric acid reactive substances (TBARS, a general oxidative stress indicator), SOD activity, photosynthetic and xanthophyll cycle pigments, PSII efficiency, and growth were determined. In addition, oxidative responses were measured during a daily cycle. Changing irradiance conditions significantly affected growth rates of C. brevis. PSII efficiency decreased significantly during periodic excess irradiance and increased under low irradiance conditions. Transition to excess irradiance increased the ratio of xanthophyll to light-harvesting pigments, whereas the opposite was observed for cultures transferred to low irradiance. This acclimation process was completed after 2 d in the new irradiance environment. SOD activity increased significantly after the first day regardless of the new irradiance environment but returned to preexposure values on the fourth day. We hypothesize that SOD activity may be temporarily elevated in C. brevis after irradiance shifts, thereby reducing oxidative stress when photoacclimation is in progress. [ABSTRACT FROM AUTHOR]

The effects of high PAR (400–700 nm), UVA (315–400 nm), and UVB (280–315 nm) radiation on viability and photosynthesis were investigated for Chaetoceros brevis Schütt. This Antarctic marine diatom was cultivated under low, medium, and high irradiance and nitrate, phosphate, silicate, and iron limitation before exposure to a simulated surface irradiance (SSI) treatment, with and without UVB radiation. Light-harvesting and protective pigment composition and PSII parameters were determined before SSI exposure, whereas viability was measured by flow cytometry in combination with a viability stain after the treatment. Recovery of PSII efficiency was measured after 20 h in dim light in a separate experiment. In addition, low and high irradiance acclimated cells were exposed outdoors for 4 h to assess the effects of natural PAR, UVA, and UVB on viability. Low irradiance acclimated cells were particularly sensitive to photo induced viability loss, whereas no viability loss was found after acclimation to high irradiance. Furthermore, nutrient limitation reduced sensitivity to photo induced viability loss, relative to nutrient replete conditions. No additional viability loss was found after UVB exposure. Sunlight exposed cells showed no additional UVB effect on viability, whereas UVA and PAR significantly reduced the viability of low irradiance acclimated cells. Recovery of PSII function was nearly complete in cultures that survived the light treatments. Increased resistance to high irradiance coincided with an increased ratio between protective- and light-harvesting pigments before the SSI treatment, demonstrating the importance of nonphotochemical quenching by diatoxanthin for survival of near-surface irradiance.
