Applications of proteomics tools revolutionized various biomedical disciplines such as genetics, molecular biology, medicine, and dentistry. The aim of this review is to highlight the major milestones in proteomics in dentistry during the last fifteen years. Human oral cavity contains hard and soft tissues and various biofluids including saliva and crevicular fluid. Proteomics has brought revolution in dentistry by helping in the early diagnosis of various diseases identified by the detection of numerous biomarkers present in the oral fluids. This paper covers the role of proteomics tools for the analysis of oral tissues. In addition, dental materials proteomics and their future directions are discussed.

The possibilities to increase adhesion of glass-ionomer cements to dentine with the help of conditioning of dentine surface by 20% orthophosphoric acid were analysed. Adhesion of glass-ionomer cements to dentine was determined by microtenzile strength method (mTSB). It was established that dentine etching for 15 sec led to improved adhesion; etching time increasing did not improve the adhesion.

Background: The purpose of this in vitro study was to compare and evaluate the stress distribution of maxillary first premolar residual crowns restored with post-core crowns, endocrowns and inlay crowns after deep margin elevation, to explore the fitting restoration for residual crowns using finite element analysis. Methods: A healthy complete right maxillary first premolar from a male adult was scanned by cone beam computed tomography (CBCT). The finite element model of the tooth was established by reverse engineering software such as Mimics, Geomagic and Hypermesh. On this basis, the residual crown model after deep margin elevation was made, and the experimental group models were divided into three groups, those restored with post core crowns, endocrowns and inlay crowns. Vertical and oblique static loads were applied to the experimental models to simulate the force on the tooth during mastication (the loading position was located in the central fossa of the occipital surface, and the load was 100 N) using Abaqus software. Results: The peak value and distribution of von Mises stress in each part of the experimental model were observed. After deep margin elevation, the peak dentin von Mises stresses were lower than the tensile strength of normal dentin in the post-core crown, endocrown, and inlay crown groups; the lowest stress results were found in the post-core crown group for the dentin, restoration, enamel, and deep margin elevation (DME) layers under vertical and oblique loading. In terms of stress distribution clouds, the peak stresses in the dentin tissue were located in the apical 1/3 of the root after postcore crown restorations for both loads, while stress concentrations were evident in the cervical and root areas after endocrown and inlay crown restorations; regardless of the load and restoration method, the corresponding stress concentration areas appeared at the junction of the DME and dentin tissue at the loading site of the restorations; Conclusions: Post-core crowns, endocrowns and inlay crowns can be used to restore residual crowns after deep margin elevation, and post-core crowns can better protect the residual tooth tissue. This study evaluated bond strength of glass fiber posts to root dentin using push‐out (PO) and diametral compression (DC), testing glycolic acid as a conditioner and varying dentin moisture. An additional aim was to test whether DC can be an alternative test to PO for bond strength assessment. Eighty bovine teeth were divided into eight groups (n = 10) defined by the use of either 37% glycolic acid or 37% phosphoric acid (PA) on moist or wet dentin before bonding with either Adapter SingleBond/RelyX ARC or One Step Plus/Duo‐Link Bisco. Each tooth provided discs with an internal diameter of 2 mm, external diameter of 5 mm, and height of 2 mm, which underwent PO and DC. Finite element analysis (FEA) was carried out on 3D models. When analyzing PO results through linear regression, the highest values of bond strength were observed using glycolic acid on wet dentin in the cervical and middle thirds of the teeth. Analyzing DC results, the only statistical influence on values was the dental thirds. The scatterplot of the DC results and the PO bond strength values indicated no relationship between the results of the two tests (r = 0.03; p = 0.64). Odontodysplasia is an uncommon dental developmental disorder associated with enamel, dentin, pulp abnormalities, and overall tooth morphology. The affected tooth is grossly abnormal in size and contour and is commonly associated with swelling of the affected area and failure of eruption. Histologically, the enamel and dentin are hypoplastic and hypomineralized. Odontodyplasia occurs most commonly in response to direct trauma to the developing tooth bud (enamel organ and dental follicle). Data on the prevalence and features of non-traumatic odontodysplasia are lacking. Medical records of dogs diagnosed with odontodysplasia were reviewed at the William R. Pritchard Veterinary Medical Teaching Hospital (VMTH), University of California, Davis, for 10 years (from 2013 to 2023). Dogs with a known history of facial trauma, persistent deciduous tooth or teeth over the region of odontodysplastic tooth or teeth, and endodontic disease of the persistent deciduous tooth or teeth were excluded from the study. Twenty-eight dogs were included in this retrospective study, representing an incidence of 1.4 per 100 dogs presenting over 10 years. Regional odontodysplasia (RO) was identified in twenty-two dogs, and generalized odontodysplasia (GO) was found in six dogs. Both comprehensive oral examination and diagnostic imaging were essential in diagnosing and assessing the presence of odontodysplasia. Awake oral examination failed to identify odontodysplasia in almost 70% of the RO cases. Secondary diseases or lesions in odontodysplastic teeth, such as periodontal disease, endodontal disease, and perio-endo lesions, were commonly seen and were particularly more frequently identified in strategic teeth (canine and carnassial teeth) than non-strategic ones. Similarities, such as female predilection, maxilla more commonly affected, and clinical signs, were observed between RO in dogs and those reported in people. Objectives: The aim of this study was to assess the lightness difference perceptibility and acceptability thresholds, for a single maxillary central incisor, and to investigate possible differences in these thresholds regarding the direction (+ΔL* vs. −ΔL*), the observer group (dentists vs. laypersons), and the gender and age of the observers. Materials and Methods: A series of images with varying lightness (L*), were created by altering the right maxillary central incisor of a male Caucasian, on a frontal view full‐portrait image. Digital modification of one central incisor by 1 ΔL* unit resulted in 15 different images: one control, seven with increased and seven with decreased lightness. The images were presented in random order, on a digitally calibrated monitor, to 158 observers, grouped into 79 dentists, and 79 laypersons, who were asked to evaluate every image and answer whether they perceive a difference and if yes, whether they accept this difference. A multifactorial analysis of covariance was performed to analyze the perception and acceptance of any difference in the central incisors and to estimate the thresholds. The statistical significance level was α = 0.05. Results: Overall, the magnitude of ΔL*, direction of ΔL*, observer group and age were each significant (p < 0.001), as was gender (p = 0.03). The effect of the magnitude of ΔL* interacted with its direction and the observer group (p = 0.045), and the effect of age interacted with the direction of ΔL*, and the gender and the group of the observers. Estimated 50%:50% perceptibility thresholds ranged from 0.1 to 2.7, and 50%:50% acceptability thresholds ranged from 2.3 to 4.5. Overall, dentists perceived lightness differences at lower magnitudes than laypersons; difference in lightness was perceived and accepted at lower magnitudes when the tooth become lighter instead of darker; and female observers perceived and accepted lightness differences at lower magnitudes compared to males. As age increased overall, differences of lightness were perceived and accepted at higher magnitudes. Conclusions: Perceptibility and acceptability of lightness differences of a single maxillary central incisor are affected by the magnitude and the direction of lightness change, as well as the gender, the age, and the type of the observers. Clinical Significance: Even small changes in lightness are perceivable; however, thresholds of perception and acceptance are dependent on the magnitude and the direction of change and are also dependent on the observer, with women, dentists and younger people perceiving and accepting changes at lower magnitudes of ΔL*. Background: Antibiotic pastes used as intracanal medication in cases of revascularization therapy might cause negative effects on tooth properties, such as a reduction in dentin microhardness. This in vitro study investigated dentin microhardness in three different locations distancing from the canal lumen after 20 days of treatment with a tri-antibiotic paste (ciprofloxacin, metronidazole, and minocycline), and with a double-antibiotic paste (ciprofloxacin and metronidazole), with calcium hydroxide [Ca(OH)2] UltracalTM XS-treated dentin as comparison. Material and Methods: Human mandibular premolars (n = 48) had the root canals cleaned and shaped and were used to produce dentin slices. Dentin slices remained immersed in the medications for 20 days. The Knoop microhardness (KHN) test was performed before (baseline/Day-0) and after treatment (Day-20) with the medications. Indentations were made at 25 µm, 50 µm, and 100 µm distances from the root canal lumen. The KHN was compared intra-group using Wilcoxon's test. Independent groups were compared using Mann–Whitney's and Kruskal–Wallis' tests, at α = 5%. Results: The microhardness in all the tested groups was reduced at Day-20 in comparison with Day-0 (p < 0.001) (intra-group comparison/same distances). The Day-0 values were similar, and the Day-20 values were higher for the Ca(OH)2 group (p < 0.05) (comparison between groups/same distances). Conclusions: Calcium hydroxide for 20 days would be preferred rather than antibiotic pastes to minimize the expected reduction in dentin microhardness during regenerative procedures. Statement of the Problem: Immediate dentin sealing (IDS) was introduced to overcome the disadvantages of delayed dentin sealing like pollution of dentin tubules, microleakage, and bond strength destruction over time. The effect of IDS on the bond strength of indirect restorations is still debatable.Purpose: This study was conducted to determine the effect of IDS on the bond strength of ceramic restorations to dentin.Materials and Method: In this systematic review and meta-analysis, the study protocol was registered on the PROSPERO database under the registration number CRD420202014 27. MEDLINE (PubMed), Web of Science, Scopus, and ProQuest databases were searched until January 2021 and updated in January 2022. Worldcat.org and Opengrey.eu, ProQuest dissertation and thesis, and Google Scholar were searched to explore the grey literature. The in vitro studies evaluating the bond strength of ceramic restoration to dentin with and without IDS were included. Seven criteria were assessed to evaluate the risk of bias in the study. Statistical analyses were conducted using RevMan 5.3. The inverse variance method was used to determine the mean difference of micro-tensile bond strength (µTBS) and shear bond strength (SBS).Results: A total of 10 studies (20 datasets) were included in the meta-analysis. Regarding the µTBS analysis, IDS had a significantly higher bond strength than Delayed Dentin Sealing (DDS) (MD:1.16, 95%CI:0.28_2.03, I2=0%). However, no significant difference was found between them in the SBS analysis (MD:0.25, 95%CI: -0.56-1.06, I2=96%). All studies were categorized to have a moderate or high risk of bias.Conclusion: Most in vitro evidence showed favorable results for the effect of IDS on the bond strength and durability of indirect restorations. The adhesive system and the type of ceramic and its treatment before cementation are determining factors. Due to the heterogeneity of the outcomes and studies with a moderate/high risk of bias, the quality of the evidence was low.

Abstract Background The healing process after tooth removal involves bone remodelling which implies some loss of alveolar bone volume. Among materials proposed for minimising this remodelling and preserving the bone, autologous dental tissue is a promising option, but more data are needed. In this context, we evaluated size and density changes using cone beam computed tomography in autologous dental material (ADM)-preserved sockets compared to controls, and assessed biological responses by histological analysis. Methods A split-mouth study was conducted including 22 patients, who underwent removal of ≥ 2 single-rooted teeth with intact sockets, assigning one socket to the experimental group which received ADM for alveolar preservation and another to the control group, which only underwent blood clot stabilisation. Cone beam computed tomography was performed postoperatively (week 0) and at weeks 8 and 16 to assess socket size and bone density. Histological analysis was carried out on trephine biopsies taken (Ø4 × 4.5 mm) from the experimental group. Background/purpose: The dental adhesive market is constantly evolving to meet the demands of dentists and patients, but new products and upgrades should be rigorously evaluated before being used in clinical practice. This study investigated the physicomechanical properties and dentin bonding efficacy of a newly upgraded universal adhesive compared to its predecessor. Materials and methods: Twenty-four molars were divided into four groups (n = 6/group) based on adhesive (new vs. predecessor) and application mode [self-etch (SE) vs. etch-and-rinse (ER)] for evaluating their dentin microtensile bond strength (μTBS), failure pattern, and bonding interface. Additional thirty-six molars’ crowns were perpendicularly sectioned to obtain flat mid-coronal dentin discs. The opposing dentin surfaces of each disc received contrasting treatments (new/predecessor adhesive applied in SE/ER mode), resulting in six interventions. The bonded discs (n = 6/intervention) were used to assess the adhesives’ survival probability employing a double-sided μTBS test. The other physicomechanical properties examined were adhesives’ oxygen inhibition layer (OIL), viscosity, hardness, elastic modulus, degree of conversion (DC), and in-situ DC. Results: Both adhesive versions showed similar μTBS (P > 0.05), failure pattern (P > 0.05), and survival probability (P > 0.008). Background/purpose: 4-methacryloyloxyethyl trimellitate anhydride/methyl methacrylate-tri-n-butyl borane (4-META/MMA-TBB) resin is used for indirect restorations. We aimed to evaluate effects of immersion in 4-META/MMA-TBB-activated liquid on the bond strength of root canal dentin. Materials and methods: We used freshly extracted single-rooted human teeth. After decoronation, each root was vertically sectioned into halves; their dentin walls were polished and flattened. The control group underwent dentin treatment with Green Activator. The immersion group was treated with Green Activator and Teeth Primer and immersed in 4-META/MMA-TBB-activated liquid. After bonding the resin blocks with Super-Bond, microtensile bond strength (μTBS) tests were performed (n = 6), and fracture surfaces were analyzed. Before surface treatment, dentin was immersed in a sodium fluorescein solution for 3 h, and resin blocks were bonded with Super-Bond with rhodamine B as in the bond strength test. Background: The long-term success of adhesive restorations is contingent on minimizing microleakage, which can lead to postoperative sensitivity and secondary caries. Sodium hypochlorite (NaOCl) and ethylenediaminetetraacetic acid (EDTA) irrigation are commonly employed during adhesive restorative procedures, but their impact on microleakage remains a concern. Proanthocyanidin (PA), a naturally derived agent, has shown potential in enhancing bond strength. Aim: This study aimed to compare and evaluate the effect of treatment with or without 30% PA of the pulp chamber dentin after 1% NaOCl and 17% EDTA irrigation on the microleakage of adhesive restorations. Materials and Methods: Forty caries-free, freshly extracted permanent human molars were used. The pulp chamber roof was removed, and the roots were resected. Canal orifices were enlarged and sealed. Specimens were divided into four groups based on adhesives (OptiBond All-In-One and Adper Easy One) and irrigation regimens. After irrigation, pulp chambers were restored with Filtek Bulk-Fill composite resin. Microleakage was assessed using methylene blue dye penetration. Results: Microleakage was observed in all groups, irrespective of irrigation protocol and adhesives. PA application showed reduced dye penetration compared to without PA, although not statistically significant. Statistical analysis was performed using Kruskal–Wallis and Mann–Whitney tests at P < 0.05. Conclusion: Irrigation protocols and the use of PA had no significant effect on the microleakage of self-etch adhesives to pulp chamber dentin. A 1-min application of 30% PA solution after NaOCl and EDTA irrigation did not improve the bonding of self-etch adhesives to pulp chamber dentin.

Background: Nonsurgical periodontal therapy results in the formation of a smear layer which inhibits tissue regeneration. Root biomodification (RB) using various agents has been tried for the enhancement of new attachment formation. However, no substantial therapeutic advantages of currently available root conditioning agents have been reported emphasizing the need for additional biologically acceptable agents. Glycolic acid (GA) due to its antimicrobial nature and ability of initiation and proliferation of fibroblasts may potentially modify root surface enabling regeneration. Materials and Methods: Eighty specimens from 40 single-rooted teeth were treated with 17% ethylenediaminetetraacetic acid (EDTA) and 5% GA and scanning electron microscopy analysis was done. The micrographs were examined for the evaluation of smear layer removal, total number of dentinal tubules, total number of patent dentinal tubules, mean diameter and surface area of dentinal tubules, and dentin erosion. Statistical analysis was done using unpaired t-test for intergroup comparison. Results: The efficacy of smear layer removal (P = 0.01) and dentin erosion (P = 0.042) was significantly better in the GA group. Both the groups showed no difference in dentinal tubule-related parameters. Conclusion: GA showed improved RB with greater smear layer removal and lesser dentin erosion, indicating its use as a potent alternative to the conventional EDTA root conditioning.

Dental stains are pigmented deposits that can be intrinsic or extrinsic in origin and are seen on the surface of the teeth. Discoloration of teeth can be due to developmental defects, traumatic injuries, or the intake of several medications. Extrinsic stains create discoloration outside the tooth surface and lay on the surface or in the acquired pellicle, whereas stains within dentin or intrinsic discoloration sometimes come from pulpal or systemic origin. To plan a specific treatment approach and its associated result, it is important to identify the etiologic cause that is creating the stain, in an attempt to provide esthetic treatment for the same. This case report describes the successful management of intrinsic staining of anterior teeth in a 10-year-old girl. The treatment plan included esthetic rehabilitation using composite veneer. The fluoride-releasing composite used provided an acceptable color match and exceptional handling properties and was relatively easy to maintain with its antiplaque effect. This case also represents a unique histopathological finding from ground sections of a retained extracted deciduous tooth.

Statement of the Problem: Although various kinds of research have been conducted to compare the physical and chemical properties of dentin and enamel in animal and human samples, proving the ability of animal dentin material as a good substitute for human specimens is always a challenge for experimental studies.Purpose: The aim of the present study is to investigate whether the changes in the dentin microhardness of animal samples are similar to those of human samples or not.Materials and Method: In this in vitro study, sixty single-rooted human, bovine, and ovine teeth (n=20 in each group) were decoronated at CEJ. The remaining roots were embedded in acrylic resin and a cross-section cut was made in the middle of the samples in order to achieve dentin disks. All of the 120 samples were randomly assigned to three control (n=20 for each group) and three experimental groups (n=20 for each group). In the experimental groups, calcium hydroxide with a creamy consistency was prepared and the disks were embedded in dishes containing calcium hydroxide. Control groups were embedded in physiological saline. The samples were incubated for seven days at the 37oC and Vickers microhardness test was performed immediately. The average of three yielded values was considered as the final value of microhardness. Data were analyzed using two-way ANOVA, one-way ANOVA, and Tukey’s post hoc tests.Results: In the control group, the human samples showed the highest microhardness value, while the bovine teeth had the lowest microhardness value (p< 0.001). In the calcium hydroxide group, the human samples showed the highest microhardness value in comparison to bovine and ovine to teeth. However, no significant difference was observed between the bovine and ovine samples in microhardness value.Conclusion: Based on our research, substituting bovine and ovine samples with human samples in experimental studies is not recommended. Nevertheless, more studies are needed in this regard.

Background: The stability of resin–dentin interfaces is still highly questionable. The aim of this study was to evaluate the effect of Salvadora persica on resin–dentin bond durability. Materials and methods: Extracted human third molars were used to provide mid-coronal dentin, which was treated with 20% Salvadora persica extract for 1 min after acid-etching. Microtensile bond strength and interfacial nanoleakage were evaluated after 24 h and 6 months. A three-point flexure test was used to measure the stiffness of completely demineralized dentin sticks before and after treatment with Salvadora persica extract. The hydroxyproline release test was also used to measure collagen degradation by endogenous dentin proteases. Statistical analysis was performed using two-way ANOVA followed by post hoc Bonferroni test and unpaired t-test. P-values < 0.05 were considered statistically significant. Results: The use of Salvadora persica as an additional primer with etch-and-rinse adhesive did not affect the immediate bond strengths and nanoleakage (p > 0.05). After 6 months, the bond strength of the control group decreased (p = 0.007), and nanoleakage increased (p = 0.006), while Salvadora persica group showed no significant difference in bond strength and nanoleakage compared to their 24 h groups (p > 0.05). Salvadora persica increased dentin stiffness and decreased collagen degradation (p < 0.001) compared to their controls. Conclusion: Salvadora persica extract pretreatment of acid-etched dentin preserved resin–dentin bonded interface for 6 months. Clinical significance: Durability of resin-dentin bonded interfaces is still highly questionable. Endogenous dentinal matrix metalloproteinases play an important role in degradation of dentinal collagen within such interfaces. Salvadora persica may preserve resin-dentin interfaces for longer periods of time contributing to greater clinical success and longevity of resin composite restorations. [ABSTRACT FROM AUTHOR]

Abstract This study evaluated the effect of fluoride varnishes containing micrometric or nanosized sodium trimetaphosphate (TMP) on dentin erosive wear in vitro. Bovine root dentin blocks were selected by surface hardness and randomly divided into five experimental groups/varnishes (n = 20/group): placebo, 5% sodium fluoride (NaF); 5% NaF+5% micrometric TMP; 5% NaF+2.5% nanosized TMP; and 5% NaF+5% nanosized TMP. Half of the surface of all blocks received a single application of the assigned varnish, with subsequent immersion in artificial saliva for 6 h. Varnishes were then removed and the blocks were immersed in citric acid (90 s, 4×/day, 5 days). After each erosive cycle, ten blocks of each group were immersed in a placebo dentifrice for 15 s (ERO), while the other ten blocks were subjected to abrasion by brushing (ERO+ABR). Objective: Some endodontic procedures require using intracanal medicament between treatment sessions. The effectiveness of these agents on dentin microhardness is crucial for deciding whether to use them or not. This study aimed to compare the effectiveness of three intracanal medicaments including Triphala, calcium hydroxide (Ca(OH)2) paste, and chlorhexidine (CHX) gel on the root dentin microhardness.Methods: Forty-eight single canal mature permanent teeth were selected. Mechanical preparation was done using RaCe rotary files. The samples were randomly allocated to four equal groups (n=12), according to the applied intracanal medicament. Group 1 received no medicament, whereas the root canals in groups 2, 3, and 4 were filled with Triphala, Ca(OH)2, and CHX, respectively. Specimens were stored for one week. Then, the roots were sectioned and the Vickers microhardness value was recorded at 0.5 mm from the pulp–dentin interface. Data were analyzed by one-way ANOVA and Tukey test and a P-value < 0.05 was considered statistically significant.Results: The mean microhardness values in the Triphala and calcium hydroxide groups were comparable to each other (P>0.05) and significantly lower compared to the control and CHX groups (P0.05).Conclusions: Triphala and Ca(OH)2 had similar effects on root dentin microhardness. Given the favorable characteristics of Triphala medicament, it can be considered a suitable alternative to Ca(OH)2 for intracanal application.

Over the last few decades, several new materials and techniques have been developed for bone regeneration. Scaffolds based on demineralized dentin matrix (DDM) present an attractive option due to their availability and several animal and human studies have been conducted to ascertain their utility in regenerative dentistry. The aim of this review was to summarize the recent studies conducted on DDM and used for bone grafts. PubMed, Web of Science, and Scopus were used to search for studies published within the last 10 years. The keywords and terms used were: “demineralized dentine matrix”, “bone grafting”, “bone augmentation” and “guided tissue regeneration” in various combinations. Original studies (in vitro, animal and human) and systematic reviews were included in the literature search. The literature search initially identified 23 studies (16 animal studies and 7 clinical reports. Most studies included in this review indicate that DDM has demonstrated promising results in a variety of dental and regenerative medicine applications. Further studies are required to completely comprehend its characteristics and prospective applications. Abstract Background Sodium hypochlorite (NaOCl) is the most efficient root canal irrigant to date. The aim of this study was to compare the effect of NaOCl used at different temperatures and concentrations on the compressive strength of root dentin. Materials and methods Seventy-two extracted human single-canaled straight roots of comparable size and length were selected and randomly divided into six groups (n = 12): Group (A) served as a control with unprepared canals. The other groups were instrumented with rotary ProTaper Universal files up to size F3. Group (B) was irrigated with 1% NaOCl at room temperature, Group (C) with 1% NaOCl heated to 70 °C, Group (D) with 5.25% NaOCl at room temperature, and Group (E) with 5.25% NaOCl heated to 70 °C. Saline was used in Group (F). The roots were sectioned into 2-mm-thick disks that underwent compression testing using a universal testing machine. Data were analyzed using one-way ANOVA and post hoc Tukey tests. The significance level was set at p ≤ 0.05. Results A total of 255 disks were tested. The control group showed the highest compressive strength (p = 0.0112). However, this did not differ significantly from that of heated (p = 0.259) or unheated (p = 0.548) 1% NaOCl. There were no statistically significant differences between the groups of instrumented teeth. Conclusion Within the conditions of this study, irrigation with NaOCl at different concentrations and temperatures during root canal preparation did not affect the compressive strength of root dentin. Clinical Relevance: This study demonstrates that the use of NaOCl as a root canal irrigant is not associated with a clinically relevant decrease in root compressive strength, especially when compared to saline.

Background: The selection of post-core material holds significant importance in endodontically treated teeth, influencing stress distribution in the dental structure after restoration. The use of computer-aided design/computer-aided manufacturing (CAD/CAM) glass fiber post-core possesses a better adaptation for different root canal morphologies, but whether this results in a more favorable stress distribution has not been clearly established. Materials and methods: This study employed finite element analysis to establish three models of post-core crown restoration with normal, oversized, and dumbbell-shaped root canals. The three models were restored using three different materials: CAD/CAM glass fiber post-core (CGF), prefabricated glass fiber post and resin core (PGF), and cobalt-chromium integrated metal post-core (Co-Cr), followed by zirconia crown restoration. A static load was applied and the maximum equivalent von Mises stress, maximum principal stress, stress distribution plots, and the peak of maximum displacement were calculated for dentin, post-core, crown, and the cement acting as the interface between the post-core and the dentin. Results: In dentin of three different root canal morphology, it was observed that PGF exhibited the lowest von Mises stresses, while Co-Cr exhibited the highest ones under a static load. CGF showed similar stress distribution to that of Co-Cr, but the stresses were more homogeneous and concentrated apically. In oversized and dumbbell-shaped root canal remnants, the equivalent von Mises stress in the cement layer using CGF was significantly lower than that of PGF. Conclusions: In oversized root canals and dumbbell-shaped root canals, CGF has shown good performance for restoration of endodontically treated teeth. Clinical relevance: This study provides a theoretical basis for clinicians to select post-core materials for residual roots with different root canal morphologies and should help to reduce the occurrence of complications such as root fracture and post-core debonding. Objectives: Determine visual 50:50% color difference acceptability thresholds (AT) for regions of the dental color space with varying chromaticity. Methods: A 40‐observer panel belonging to two different groups (dentists and laypersons) evaluated 144 dental resin composites pairs (divided in three different sets of 48 pairs according to chroma value: Low Chroma (LC), Medium Chroma (MC) and High Chroma (HC) placed 40 cm away and inside of a viewing cabinet (D65 Standard light source; diffuse/0° geometry). A Takagi–Sugeno–Kang (TSK) fuzzy approximation was used for fitting the data points and calculate the 50:50% acceptability thresholds in CIEDE2000. A paired t‐test was used to evaluate the statistical significance between thresholds differences and Bonferroni correction was applied. Results: The CIEDE2000 50:50% AT were ∆E00 = 2.84, ∆E00 = 2.31 and ∆E00 = 1.80 for LC, MC and HC sets of sample pairs, respectively. The 50:50% AT values were statistically significant between the different sets of sample pairs, as well as the 50:50% AT values obtained for different observer groups. Conclusions: 50:50% CIEDE2000 acceptability thresholds for dentistry are significantly different depending on the chromaticity of the samples. Observers show higher acceptability for more achromatic samples (low chroma value) than for more chromatic samples. Clinical Significance: The difference in the AT for distinct regions of the dental color space can assist professionals as a quality control tool to assess clinical performance and interpret visual and instrumental findings in clinical dentistry, dental research, and subsequent standardization processes. Objective: To investigate the effect of ceramic thickness and dental substrate (enamel vs. dentin/enamel) on the survival rate and failure load of non‐retentive occlusal veneers. Materials and Methods: Human maxillary molars (n = 60) were divided into five test‐groups (n = 12). The groups (named DE‐1.5, DE‐1.0, DE‐0.5, E‐1.0, E‐0.5) differed in their dental substrate (E = enamel, DE = dentin/enamel) and restoration thickness (standard: 1.5 mm, thin: 1.0 mm, ultrathin: 0.5 mm). All teeth were prepared for non‐retentive monolithic lithium‐disilicate occlusal veneers (IPS e.max Press, Ivoclar). Restorations were adhesively cemented (Syntac Classic/Variolink II, Ivoclar) and exposed to thermomechanical fatigue (1.2 million cycles, 1.6 Hz, 49 N/ 5–55°C). Single load to failure was performed using a universal testing‐machine. A linear‐regression model was applied, pairwise comparisons used the Student–Newman–Keuls method (p < 0.05). Results: Three dentin‐based occlusal veneers (one DE‐1.0, two DE‐0.5) revealed cracks after fatigue exposure, which corresponds to an overall‐survival rate of 95%. Load to failure resulted in the following ranking: 2142 N(DE‐0.5) > 2105 N(E‐1.0) > 2075 N(E‐0.5) > 1440 N(DE‐1.5) > 1430 N(DE‐1.0). Thin (E‐1.0) and ultrathin enamel‐based occlusal veneers (E‐0.5) revealed high failure loads and surpassed the standard thickness dentin‐based veneers (DE‐1.5) significantly (p = 0.044, p = 0.022). Conclusion: All tested monolithic lithium disilicate occlusal veneers obtained failure loads above physiological chewing forces. Thin and ultrathin enamel‐based occlusal veneers outperformed the standard thick dentin‐based occlusal veneers. Aim: To incorporate different concentrations of zirconia/silver phosphate (ZrO2/Ag3PO4) nanoparticles (NPs) in self-etch (SE) adhesive. Surface characterization, elemental analysis, survival rate assessment of S. mutans, micro-tensile bond strength (μTBS), and the degree of conversion (DC) of composite bonded to caries-affected dentin (CAD) were determined. Material and Methods: This study employed a comprehensive methodological approach, incorporating a variety of analytical techniques, including scanning electron microscopy (SEM) combined with energy-dispersive X-ray spectroscopy (EDX), Fourier transform infrared (FTIR) spectroscopy, and μTBS testing. Eighty human third molars that had caries advancement up to the middle third of the dentin were included. Samples were distributed into four groups based on concentration of ZrO2/Ag3PO4 NPs in the primers of the two-step SE adhesive: Group 1 (Control): unmodified SE adhesive; Group 2: 0.15 wt% ZrO2/Ag3PO4 + SE adhesive; Group 3: 0.25 wt% ZrO2/Ag3PO4 + SE adhesive; Group 4: 0.5 wt% ZrO2/Ag3PO4 + SE adhesive. SEM was employed to investigate the morphological characteristics of ZrO2/Ag3PO4 NPs. For elemental distribution EDX spectroscopy and to assess the of cured and uncured adhesive with changed concentrations of NPs, FTIR spectroscopy were performed. Antibacterial efficacy was calculated in adhesives with different concentrations of ZrO2/Ag3PO4 using the pour plate method. For μTBS assessment, a compressive force was applied at the material–dentin interface at a crosshead speed of 0.5 mm/min. The debonding process of each specimen was measured in MegaPascals (MPa). One-way analysis of variance (ANOVA) and Tukey's post hoc test were used to compare the means and standard deviation (SD) between groups. Results: The samples from Group 4, which were applied with 0.5 wt% ZrO2/Ag3PO4 + SE, displayed the lowest survival rate (0.12 ± 0.01 CFU/mL) of S. mutans. The strongest bond of composite to the CAD surface was observed in Group 4 (0.5 wt% ZrO2/Ag3PO4 + SE) (20.12 ± 0.79 MPa). The highest DC was observed in the control group (unmodified SE (69.85 ± 8.37)). Conclusion: The self-etch adhesive modified with ZrO2/Ag3PO4 nanoparticles showed a favorable effect on micro-tensile bond strength (μTBS) and demonstrated enhanced antibacterial efficacy against S. mutans. Objectives: To evaluate minimally invasive restorations' capacity to mask discolored teeth and explore the impact of ceramic thickness, translucency, and cement color. Materials and Methods: Twenty‐four assessment pairs of naturally colored and discolored bovine dentin samples were formed, using lithium disilicate specimens in six different thicknesses (0.3–0.8 mm), two different translucencies (high, low), and two cements (transparent, tooth‐colored). Evaluators assessed the color differences in each assessment pair, and the threshold for detecting a color difference was determined using sequential testing and the Bonferroni‐Holm method. Results: A thickness of 0.6 mm effectively masked color differences using high translucent ceramic with transparent cement, detectable differences were still observed at 0.7/0.8 mm. A threshold thickness of 0.4 mm was seen using high translucent ceramic and tooth‐colored cement, with color differences still discernible at 0.5 and 0.8 mm. A threshold thickness of 0.4 mm was detected using low translucent ceramic and transparent cement, while detectable differences persisted at 0.5, 0.7, and 0.8 mm. A 0.5 mm threshold thickness was observed when using low translucent ceramic and tooth‐colored cement, and no detectable color differences were detected beyond this thickness. Conclusions: Masking can be achieved with a thickness of 0.4–0.5 mm using a low translucent material and tooth‐colored cement. Clinical Significance: Understanding the impact of ceramic thickness, translucency, and cement color can aid clinicians in making informed decisions for achieving the best esthetic outcomes while preserving tooth structure. Effective masking can be accomplished with ceramic thicknesses starting at 0.4 mm, especially when employing a low translucent material and tooth‐colored cement. Objectives: To evaluate the influence of the group and individual education of students on shade matching quality. Secondary study objective included a survey‐assessment of quality and difficulty level of color matching. Materials and Methods: Forty dental student participants in Integrated Academic Studies of Dentistry at the Faculty of Medicine, University of Niš (26 female, 14 male) 21–25 years old, signed consent to voluntarily participate in the research. The Ethics Committee of the Clinic of Dentistry, Faculty of Medicine, University of Niš, approved the study under number 14/11–2019‐2EO. They were randomly divided into two groups (n = 20), experimental (IE, individual education) and control (GE, group education). Each group was divided into four subgroups (n = 5). The study was performed in three phases. In Phase I (I) and III (III), students matched the shade of the maxillary central left incisor (#9) using three different shade guides. Each of the students served as a "patient" for shade matching, while the other members of the subgroup, individually (VMI) and as a team (VMT), visually matched the tooth shade. In Phase II (between phases I and III), group IE received standard group and individual education and training, while group GE had only group education. Shade matching results in before and after education were compared with results obtained by an intraoral spectrophotometer. CIELAB (∆E*), and CIDE2000 color differences (∆E00) were calculated. The color differences between VMI or VMT and ES readings were graded. In addition, the students rated the level of difficulty and quality of shade matching from 1 to 5, signifying the most to least difficult, and the worst to best match, respectively. Means and standard deviations were calculated. Differences were compared using the Student t‐test. Results: Means (s.d.) of CIEDE2000 color differences for VMI before and after IE were 4.5 (1.5) and 3.3 (0.9), respectively. Corresponding values for group GE were 4.4 (1.4) and 4.0 (1.0), respectively. As compared with GE, group IE achieved 15% better results for the best matches and 27.5% for one of the three best matches in by‐point comparison. Mean (s.d.) results for VMT in the experimental group before and after IE were 3.6 (2.4) and 2.8 (1.6), respectively. Corresponding results for the control group were 4.4 (2.1) and 3.5 (1.7), respectively. Group IE had statistically higher VMI and VMT scores than GE, while, for GE, only VMT scores were statistically higher. Group IE achieved better results in by‐point comparison. After education and training, the students rated the shade matching as more precise and less difficult. Results were significantly better for Quality (IE) (sig = 0.010) and Difficulty (GE) (sig = 0.021). Conclusions: Within the limitations of the study, the experimental group achieved significantly better results in individual color matching after education and training than the control group, while both groups made progress in team color matching. Both groups rated shade matching more accurate and easier after education. Regenerative dentistry (RD) is an innovative strategy for treating necrotic teeth and regenerating damaged dental tissue. Biocompatible materials are pivotal for the advancement of RD, and the rising interest in environmental sustainability drives exploration of sustainable materials for dentistry. Bacterial nanocellulose (BNC) has emerged as a promising eco-friendly option and this study aims to assess BNC's suitability as scaffolds for regenerative dentistry applications. Different in vitro methods have been utilized to characterize the properties of BNC scaffolds in regenerative dentistry, such as scanning electron microscopy (SEM) to analyse surface property and porosity, as well as examining their absorption behaviour using phosphate-buffered saline and bovine serum. Dental pulp stem cell (DPSCs) attachment, viability, and proliferation were evaluated using SEM, live and dead, and tetrazolium reduction assays. The odontogenic potential of the scaffold was evaluated using Alizarin Red staining and qPCR (14 and 21 days). Scanning electron microscopy (SEM) images and ethanol displacement method demonstrated the porous architecture of the BNC scaffold with an average porosity of 70.02 ± 4.74% and 50.26 ± 1.43% respectively. The scaffold absorbed 2846.54 ± 258.95 of BSA and 1648.63 ± 50.37% PBS after immersion in solution for 1 h, following pseudo first and second order kinetics. The biocompatibility assay indicated that cell density increased with time and that the scaffold was appropriate for cell adhesion and migration. Moreover, the BNC led to significantly higher mineralization and odontogenic expression compared to the control (BNC in conditioned media). BNC showed fast adsorption of bovine serum, allowed DPSC attachment, migration, and odontogenic differentiation. (1) Background: The objective of the current investigation was to determine how different dark chocolate concentrations impacted the enamel and dentine microhardness. (2) Methods: Twenty noncarious premolar teeth extracted for orthodontic reasons were used in this study. Each tooth was divided mesiodistally to obtain 40 specimens of enamel and dentine 4 × 4 mm. Initial and final assessments of the surface microhardness of the enamel and dentine were created using a Digital Micro Vickers Hardness Tester (Laryee, Beijing, China) under (0.24 N load for 15 s). Four random groups (n = 10) were created: G1 (control, immersed in artificial saliva), G2 (70%), G3 (85%), and G4 (100% dark chocolate). Each sample underwent four cycles of 60-s immersion in suggested concentrations, followed by 30 min in artificial saliva. The attained values underwent statistical analysis employing SPSS version 26, with the utilization of T-tests and ANOVA. (3) Results: The results revealed a significant, incremental increase in the average microhardness values for enamel, specifically 61.72 ± 22.729, 64.17 ± 23.397, and 109.15 ± 34.625, and for dentin from 57.91 ± 8.332 to 72.29 ± 2.752 and ultimately reaching 88.67 ± 8.602, corresponding to the escalating concentrations of dark chocolate (70%, 85%, and 100%) (p < 0.001). Aim: Long-term clinical success on indirect restorations is largely determined by bonding efficiency of the luting agent, with adhesion to dentin being the main challenge. Therefore, aim of this study was to assess the microtensile bond strength when using flowable resin composite, preheated resin composite and dual self-adhesive resin cement as dentin luting agents. Materials and Methods: Occlusal thirds of molar teeth were cut and randomly divided into 3 groups to be cemented: RelyX™U200, Filtek™ Z250 XT- preheated to 70° and Filtek Flow™ Z350XT. They were then thermocycled 5000 times between 5+/-2°C and 55+/-2°C. Subsequently, 10 microbars per group were prepared. The 30 samples were placed in saline solution for 24 hours at room temperature prior to microtensile test. This was performed with a digital universal testing machine at a crosshead speed of 0.5 mm/min. The bond strength values obtained were analyzed in Megapascals (MPa). Measures of central tendency such mean and measures of dispersion such standard deviation were used. In addition, the Kruskall Wallis non-parametric test with Bonferroni post hoc test was applied, considering a significance value of 5% (P < 0.05), with type I error. Results: The dentin microtensile bond strengths of preheated resin composite, flowable resin composite and dual self-adhesive cement were 6.08 ± 0.66 Mpa, 5.25 ± 2.60Mpa and 2.82 ± 1.26Mpa, respectively. In addition, the preheated resin composite exhibited significantly higher microtensile bond strength compared to the dual self-adhesive cement (P < 0.001). While the flowable resin composite showed no significant difference with the dual self-adhesive cement (P = 0.054) and the preheated resin composite (P = 0.329). Conclusion: The microtensile bond strength in dentin was significantly higher when using a preheated resin composite at 70°C as a luting agent compared to dual self-adhesive cement. Abstract This study aimed to evaluate the influence of the Er,Cr:YSGG irradiation and 980-nm diode lasers on the surface roughness (SR) and volume loss (VL) of dentin subjected to cariogenic challenge. Subsequently, 130 specimens of bovine dentin were divided into the following 13 groups: NT: no treatment; FG: fluoride gel; FV: fluoride varnish; Di: 980-nm diode; Di + FG; Di + FV; FG + D; FV + Di; Er: Er,Cr:YSGG; Er + FG; Er + FV; FG + Er and FV + Er. Er,Cr:YSGG laser parameters were as follows: 0.25 W; 5.0 Hz; 4.46 J/cm2 without water and 55% air. Furthermore, the 980-nm diode laser parameters were 2.0 W; 2.0 Hz; 21.41 J/cm2. The samples from each group were subjected to pH cycling. A confocal laser scanning microscope was used to evaluate SR and VL. Difference between the volume of the reference and treated areas + DES/RE was used to determine SR and VL. The mean values of the different groups were subjected to analysis of variance and Tukey’s post-hoc test. The VL values were analyzed using the Kruskal–Wallis and Dunn post-hoc test (p < 0.05). The SR of the reference area did not show a statistically significant 1807-3107-bor-38-e025treatment and cariogenic challenge (p > 0.05). Moreover, VL in the FV + Di and FV + Er groups showed a statistically significant difference compared with areas submitted to different types of treatment and cariogenic challenge (p > 0.05). Er,Cr:YSGG and 980-nm diode lasers associated with fluoride varnishes decreased dentin VL in bovine teeth submitted to cariogenic challenge.

Objective: To evaluate the influence of antimicrobial photodynamic therapy (aPDT) on the bond strength of adhesive restorative materials to healthy and caries-affected dentin. Material and Methods: 04 electronic databases and 02 additional searches were assessed. In vitro studies considering sound or caries-affected human or bovine dentin (population), use of aPDT before restorative treatment (intervention), use of other dentin disinfection techniques (comparison), and bond strength to dentin (outcome) were included. The risk of bias for in vitro studies was used as a method to check the evidence. The method used to present and synthesize results was data extraction. Results: The total number of included studies was 11, including 550 permanent teeth. Bond strength was evaluated mainly through the shear bond strength (SBS) test, with dentin affected by caries being the substrate most frequently used in studies. Six studies demonstrated that aPDT with methylene blue (MB) negatively affected the SBS values of resin-modified glass ionomer cement (RMGIC) to dentin. Regarding risk of bias assessment, all studies were at high risk of bias. Conclusion: The effects of aPDT on adhesion to dentin vary depending on the type of substrate and the photosensitizer used. Mainly, MB negatively affected the SBS values from RMGIC to dentin. The conclusion should be interpreted considering the high risk of bias in the studies.

Objectives: This study assessed the effect of chlorhexidine (CHX) and isopropyl alcohol (IA) on immediate and late pushout bond strength (PBS) of fiber posts to dentin. Materials and Methods: In this in vitro study, 54 single-canal premolars were endodontically treated, and randomly assigned to 3 groups (N=18) for root dentin conditioning with distilled water (control), 2% CHX, and 70% IA after post space preparation. Fiber posts were cemented with TheraCem self-adhesive cement, and each group was subdivided into two subgroups (N=9) for PBS measurement immediately after bonding, and after 5000 thermal cycles (5-55°C). The roots were then sectioned, and their PBS was measured. The mode of failure was evaluated under a stereomicroscope at ×40 magnification. Data were analyzed by repeated measures ANOVA and Tukey’s test (alpha=0.05). Results: The highest PBS was noted in the IA group (21.12 MPa) after 24 hours and the lowest PBS belonged to the control group after thermocycling (7.48 MPa). ObjectivesThe purpose of this study was to show that optical coherence tomography (OCT) and thermal imaging can be used to monitor changes in the structure and activity of caries lesions over time after treatment with silver diamine fluoride (SDF).MethodsArtificial caries lesions were formed on enamel and dentin bovine blocks. Each block was partitioned into five windows with the central three windows exposed to a demineralization solution to create lesions: one sound window served as a sound control (SC), one sound window was exposed to SDF to serve as a test control (SCT), one lesion window served as a lesion control (LC), one lesion window received one application of SDF (L1), while the other lesion window received two applications of SDF (L2). Each window was scanned using OCT before SDF application, and every week subsequently, for 12 weeks after initial SDF treatment. Changes in the mean intensity and the width of the peak of increased reflectivity due to the lesion and SDF along with the intensity at a depth of 180 µm from the surface representing optical penetration through the lesion were monitored. Changes in the heat lost, ΔQ (temperature integrated over time) of each window during drying with air were also monitored using a thermal imaging camera. Transverse microradiography (TMR), and high-resolution microscopy were also used for the analysis of selected samples.ResultsThe reflectivity and optical penetration of sound and lesion areas of enamel and dentin manifested significant changes in OCT images after SDF application. Thermal imaging showed significant differences in ΔQ indicative of permeability changes in the sound and lesion areas of enamel and dentin after SDF application.

Objective: The access cavity preparation technique might influence the fracture resistance of endodontically treated teeth. This study evaluated the impact of preserving the enamel-dentin bridge on fracture resistance of endodontically treated mandibular molars.Methods: A total of 42 mandibular molars were randomly divided into three groups according to the access cavity design: Traditional endodontic access cavity (TEC), truss endodontic access cavity (TREC), and control (CON) (n=14). The teeth in each group were divided into two equal subgroups (with and without thermocycling). The control group was stored in saline over the experiment, whereas class II mesio-occlusal access cavities were prepared in the two experimental groups. In the TEC design, a conventional access cavity was prepared. In the TREC design, the occlusal enamel and dentin between the mesial and distal root canal orifices were not removed. Endodontic treatment, and composite resin restoration were performed similarly in the experimental groups. Successful management of these lesions is often challenging, and given that dentin can be remineralized, adverse consequences due to progression of these lesions can be prevented or minimized as such.Purpose: This study aimed to assess the effect of casein phosphopeptide amorphous calcium phosphate (CPP-ACP) and Remin-Pro remineralizing agents on dentin microhardness of non-carious dentin lesions.Materials and Method: This in vitro, experimental study evaluated 36 extracted sound human premolars. The teeth were decoronated at the cementoenamel junction. Enamel was removed, and dentin was exposed at the cervical third of the buccal surface. The primary microhardness of dentin was then measured. The teeth, standardized in terms of dentin microhardness, then underwent demineralization by acid etching and were subjected to microhardness test again. They were then randomized into three groups for treatment with CPP-ACP, Remin-Pro, and artificial saliva (control), and dentin microhardness was measured for the third time after treatment. Data were analyzed using ANOVA.Results: Within group comparisons showed a significant difference in microhardness at the three time points in all three groups (p< 0.005). Between-group comparisons revealed that the microhardness of the three groups was not significantly different at baseline or after demineralization. However, the microhardness of the three groups was significantly different after the intervention (p= 0.000). Pairwise comparisons revealed significantly higher microhardness in the CPP-ACP group than the other two groups (p= 0.003). Remin-Pro and the control groups were not significantly different in this respect (p= 0.340).Conclusion: CPP-ACP can be used for remineralization of non-caries dentin lesions; however, Remin-Pro does not appear to be effective for this purpose.

Background/purpose: One of the challenges in adhesive ceramic restorations is deterioration of tooth-cement-ceramic interfaces. This study was to quantitatively investigate lipopolysaccharide (LPS) penetration through adhesive ceramic restorations. Materials and methods: Standardized holes were formed on ceramic (IPS Empress CAD, Ivoclar Vivadent) and human dentin discs. Prepared discs were randomly assigned to experimental groups (n = 6/group): SC, self-adhesive resin cement (SmartCem 2, Dentsply Sirona) bonded to ceramic discs; ST, self-adhesive resin cement bonded to dentin discs; VC, etch-and-rinse resin cement (Variolink II, Ivoclar Vivadent AG) bonded to ceramic discs; VT, etch-and-rinse resin cement bonded to dentin discs. The specimens underwent thermocycling (10,000 cycles; 5–55 °C) then LPS penetration test until 5-weeks. A mixed effect analysis using R statistical language was performed for data analysis. Results: Dentin and ceramic bonded with etch-and-rinse resin cements (groups VT and VC) showed significantly less penetration than those with self-adhesive resin cement (groups ST and SC) (P 0.05). Stem cells are present in most of the tissues in the craniofacial complex and play a major role in tissue homeostasis and repair. These cells are characterized by their capacity to differentiate into multiple cell types and to self-renew to maintain a stem cell pool throughout the life of the tissue. The authors discuss original data from experiments and comparative analyses and review articles describing the identification and characterization of stem cells of the oral cavity. Every oral tissue except enamel, dentin, and cementum contains stem cells for the entire life span. These stem cells self-renew to maintain a pool of cells that can be activated to replace terminally differentiated cells (for example, odontoblasts) or to enable wound healing (for example, dentin bridge in pulp exposures and healing of periodontal tissues after surgery). In addition, dental stem cells can differentiate into functional blood vessels and nerves. Initial clinical trials have shown that transplanting dental pulp stem cells into disinfected necrotic teeth has allowed for the recovery of tooth vitality and vertical and horizontal root growth in immature teeth with incomplete root formation. As a consequence of these groundbreaking discoveries, stem cell banks are now offering services for the cryopreservation of dental stem cells. The future use of stem cell–based therapies in the clinic will depend on the collaboration of clinicians and researchers in projects designed to understand whether these treatments are safe, efficacious, and clinically feasible. Background: Endodontic shaping causes stress and strain in the root canal dentin. Dentin microcracks have the potential to be later followed by root fractures occurring under the occlusal load. The aim of our research was to theoretically determine the values of such dentinal states of stress and strain during the endodontic shaping of curved root canals using finite element analysis (FEA). Methods: To highlight the stress concentrations in dentin, two geometric models were created considering the volume of the curved dental root and the contact between the endodontic file and the root canal walls. The application of forces with different values was simulated both on a uniform curved root canal and on a root canal with an apical third curvature of 25° as they would be applied during the preparation of a root canal. Results: In the case of the first model, which was acted upon with a force of 5 N, the deformations of the root canal appeared along the entire working length, reaching the highest values in the apical third of the root, although there were no geometric changes in the shape of the root canal. Regarding the second root model, with an apical third curvature of 25°, although the applied force was 2 N, the deformations were accompanied by geometric changes in the shape of the root, especially in the upper part of the apical third. At a higher force of 7 N exerted on the endodontic file, the geometric shape changed, and the deformation reached extreme critical values. The resulting tensile stresses appearing in the experimental structure varied similarly to the deformations. Conclusions: Significant stress and strain can develop, especially in the apical third of curved root canals during their shaping, and the risk of cracks is higher for endodontically treated teeth presenting severe curvatures in the apical third of the root. The aim is to quantitatively analyze the diameter and tubular density and semi-quantitatively analyze the elemental composition (Calcium and Phosphorus) in the dentin of primary teeth of children with Osteogenesis Imperfecta (OI) in comparison with a control group. Material and methods: A microstructural (in vitro) analysis of primary teeth of children with OI was performed with SEM and EDS. The variables measured were the tubule count (tubule/mm2) and diameter (µm) at 2000 times magnification at four points of the dentin of different depths. A semiquantitative analysis of the elemental composition of the dentin was performed with EDS of Calcium and Phosphorus (cps). Descriptive and inferential analysis (Fisher's exact test, the Mann–Whitney U test, the Kruskal–Wallis test, the Bonferroni post hoc test, the ANOVA test of repeated measures, a test of inter- and intra-subject effects, and the Geisser–Greenhouse test) were carried out. Data were analyzed with a 95% confidence level (p-value < 0.05). Results: A total of 25 deciduous teeth from 17 patients with OI and 30 teeth from healthy children were studied. There were differences in the count and tubular diameter for the control group with OI; in addition, the behavior curve changed when the systemic disease was severe. While there were no differences in the amount of dentinal Calcium, the OI tooth showed a significantly lower amount of Phosphorus (p < 0.05), except in the pulpal dentin (p > 0.05). Conclusion: The alterations of the dentinal tubules (density and diameter) were more pronounced in the most severe phenotypes of systemic disease. The amount of Phosphorus was decreased in the dentin of the primary teeth of children with OI compared to the control group. [ABSTRACT FROM AUTHOR]
