Introduction: Many countries use commercial foot-and-mouth disease (FMD) vaccines to prevent FMD pandemics, but these vaccines have disadvantages, such as repeated vaccinations due to the short persistence of antibody (Ab) titers and incomplete host defense despite high Ab titers. To address these shortcomings, we aimed to develop a novel FMD vaccine containing furfurman as an adjuvant., Method: To demonstrate the efficacy of the test vaccine, adaptive immunity was evaluated by measuring Ab and neutralizing Ab titers and host defense against viral infections in experimental and target animals. In addition, the expression levels of cytokines [interferon (IFN)α, IFNβ, IFNγ, interleukin (IL)-1β, IL-2, and IL-12p40] were evaluated at the early stages of vaccination to confirm the simultaneous induction of cellular and humoral immune responses induced by the test vaccine., Result: The groups that received vaccine containing furfurman showed a strong early, mid-term, and long-term immune response and host defense against viral infections compared to the control groups. The significant upregulation observed in cytokine levels in the furfurman group compared to those in the control groups strongly suggest that the test vaccine strengthens cellular immune response and effectively induces a humoral immune response., Conclusion: Our study demonstrated that furfurman, as an FMD vaccine adjuvant, achieves long-lasting immunity and host defense against viral infections by eliciting potent cellular and humoral immune responses. Objective: The aim: The aim of this study was to determine the content of interleukins (IL) 1β, 2, 4, and 10, as well as the generally accepted marker of inflammation - C-reactive protein (CRP) - in the peripheral blood on the first and tenth days of ischemic stroke (IS)., Patients and Methods: Materials and methods: The study involved 25 patients with IS (including 8 people with mild case of neurological disorders, 9 - moderate case and 8 - severe case) and 14 people of the control group. The levels of IL-1β, IL-2, IL-4 and IL-10 in the blood were determined by the immunoenzyme method., Results: Results: It was found that on the first day in patients with IS an increase in the concentration of CRP and all the studied cytokines, especially pro-inflammatory cytokines IL-1β and IL-2, is marked. On the tenth day, the content of pro-inflammatory cytokines and CRP significantly decreases compared to the first day, but remains higher than in the control, but the concentration of anti-inflammatory cytokines (IL-4 and IL-10) continues to increase., Conclusion: Conclusions: The results obtained on the first day of IS indicate the development of neuroinflammation. On the tenth day the severity of the inflammatory process is significantly reduced, but it still occurs. It was also shown that the outcome of IS depends on the concentration of cytokines in the blood: the higher the level of pro-inflammatory interleukins on the first day, the lower the content of anti-inflammatory interleukins and the higher the amount of pro-inflammatory interleukins on the tenth day, the more pronounced the neurological deficit.

Purpose: Particular interest is now given to the potential of dietary supplements as alternative non-pharmacological approaches in intestinal inflammation handling. In this aim, this study evaluates the efficiency of fish collagen peptides, Naticol®Gut, on colonic inflammation. Methods: Wild type and Mannose receptor-deficient in the myeloid lineage C57BL/6 mice were administered with Dextran Sodium Sulfate (DSS), Naticol®Gut, DSS, and Naticol®Gut or only water for 4 or 8 days. Inflammatory status was evaluated by establishing macroscopic and microscopic scores, by measuring cytokine and calprotectin production by ELISA and the myeloperoxidase activity by chemiluminescence. Colonic macrophages were phenotyped by measuring mRNA levels of specific markers of inflammation and oxidative status. Colonic immune populations and T-cell activation profiles were determined by flow cytometry. Mucosa-associated gut microbiota assessment was undertaken by qPCR. The phenotype of human blood monocytes from inflammatory bowel disease (IBD) subjects was characterized by RT-qPCR and flow cytometry and their oxidative activity by chemiluminescence. Results: Naticol®Gut-treated DSS mice showed attenuated colonic inflammation compared to mice that were only exposed to DSS. Naticol®Gut activity was displayed through its ability to orient the polarization of colonic macrophage towards an anti-inflammatory and anti-oxidant phenotype after its recognition by the mannose receptor. Subsequently, Naticol®Gut delivery modulated CD4 T cells in favor of a Th2 response and dampened CD8 T-cell activation. This immunomodulation resulted in an intestinal eubiosis. In human monocytes from IBD subjects, the treatment with Naticol®Gut also restored an anti-inflammatory and anti-oxidant phenotype. The work provides information on the results of the Joint Immunological Forum, which was held from June 24 to 29, 2019 in Novosibirsk. The modern directions of the development of fundamental and applied immunology are analyzed. Particular attention is paid to the discussion of the most issues identified in the section "Immunopathogenetic bases of tumor growth", which also presented the results of studies conducted at the National Medical Research Centre for Oncology of the Ministry of Health of Russia for the study of isolation and study of the biological properties of tumor stem cells. Noteworthy are the new advances in modern immunology, which clarify the hierarchical structure of lymphocyte populations, with the separation of various minor subpopulations based on the phenotypic, molecular genetic and functional properties of cells, whose role in ensuring the integrity of the body has not been fully studied. In addition to theoretical reports, during this Forum the results of using new methodological approaches to study the structural and functional organization of individual links of innate and adaptive immunity both under model conditions and during the development of various human diseases were presented, the most promising ways to improve analytical and technological platforms were identified. In the crayfish of the Forum, several advanced training programs were implemented for employees of various levels of practical health care and fundamental science.

Researchers employ various study-specific screening criteria to reduce potential variables associated with subsequent re-use of NHPs. However, criteria set for NHP re-assignments largely neglect the impact of previous exposures on overall biology. Since the immune system is a key determinant of overall biological outcome, an altered biological state could be predicted by monitoring global changes in the immune profile. We postulate that every different exposure or a condition can generate a unique global immune profile in NHPs. Methods Changes in the global immune profile were evaluated in three different groups of rhesus macaques previously enrolled in dengue or malaria vaccine studies over six months after their last exposure. Naïve animals served as the baseline. Fresh blood samples were stained with various immune cell surface markers and analyzed by multi-color flow-cytometry to study immune cell dynamics in the peripheral blood. Serum cytokine profile in the pre-exposed animals were analyzed by mesoscale assay using a customized U-PLEX NHP biomarker panel of 12 cytokines/chemokines. Results Pre-exposed macaques showed altered dynamics in circulating cytokines and certain innate and adaptive immune cell subsets such as monocytes, HLA-DR+NKT cells, B cells and T cells. Some of these changes were transient, while some lasted for more than six months. Each group seemed to develop a global immune profile unique to their particular exposure. Conclusion Our data strongly suggest that re-used NHPs should be evaluated for long-term, overall immunological changes and randomly assigned to new studies to avoid study bias.

Vaccinal prophylaxis is the most effective way to reduce incidence rates. However, the immune response developing after injection of most inactivated vaccines is insufficient when used without adjuvants. Special attention should be paid to the issue of immunoprophylaxis of high-risk patients with impaired immune status. Use of appropriate adjuvants during vaccination can optimize development of protective immunity in immunocompromised patients. Cytokines that regulate in vivo immune-response cascades to any antigenic stimulus and influence the development of an antigen-specific immune response can be considered as potential immunoadjuvants. The present review analyzes briefly studies of the immunoadjuvant properties of cytokines used to increase the effectiveness of regularly scheduled vaccines and those used for epidemic indications. The idea that bacterial toxins are not only killers but also execute more sophisticated roles during bacteria–host interactions by acting as negotiators has been highlighted in the past decades. Depending on the toxin, its cellular target and mode of action, the final regulatory outcome can be different. In this review, we have focused on two families of bacterial toxins: genotoxins and pore-forming toxins, which have different modes of action but share the ability to modulate the host’s immune responses, independently of their capacity to directly kill immune cells. We have addressed their immuno-suppressive effects with the perspective that these may help bacteria to avoid clearance by the host’s immune response and, concomitantly, limit detrimental immunopathology. These are optimal conditions for the establishment of a persistent infection, eventually promoting asymptomatic carriers. This immunomodulatory effect can be achieved with different strategies such as suppression of pro-inflammatory cytokines, re-polarization of the immune response from a pro-inflammatory to a tolerogenic state, and bacterial fitness modulation to favour tissue colonization while preventing bacteraemia. An imbalance in each of those effects can lead to disease due to either uncontrolled bacterial proliferation/invasion, immunopathology, or both.

Disorders of immune state in desynchronosis may be associated with reduced concentrations of melatonin in blood, thus being a prerequisite for pharmacological correction of appropriate homeostatic changes. The purpose of this work was to explore some mechanisms of exogenous melatonin actions upon parameters of innate and adaptive immunity in experimental model of desynchronosis under the conditions of LED illumination. The study was performed with 196 adult guinea pigs. Light desynchronosis was produced by day-and-night illumination of the animals having been continued for 30 days. Melatonin was administered applied per os daily at the total dose of 30 mg/kg. A solution of melatonin in isotonic NaCl solution was prepared from the Melaxen drug (INN: melatonin, “Unipharm Inc.,” USA) ex tempore. To study innate immunity of blood cells, we determined leukocyte numbers, WBC differential counts, and functional activity of phagocytes, as spontaneous and induced NBT test, as well as engulfment of polystyrene latex particles. Th1-specific immune response was studied according to degree of delayed type hypersensitivity reaction; Th2-dependent response was assessed as the numbers of antibody-forming cells in the spleen of the animals after immunization with allogeneic erythrocytes. Serum concentrations of interleukin 4 (IL-4), interferon-gamma (IFNγ), melatonin, and cortisol were measured by enzyme immunoassay, using the “Immulayt 2000” (USA) with guinea pigspecific test systems. It was found that experimental desynchronosis was associated with leukocytosis, lymphoand monocytopenia, activation of oxygen-dependent metabolism of blood phagocytes, suppression of Th1-and Th2-dependent immune response. Desynchronosis was also accompanied by decreased concentrations of serum melatonin, IFNγ and IL-4, along with increased cortisol concentrations. Reduced IFNγ and IL-4 amounts was associated with decreased melatonin concentrations. Suppression of Th1- and Th2-dependent immune response is found to develop in accordance with reduction in IFNγ and IL-4. Melatonin administration in desynchronosis (a total of 30 mg/kg) resulted in recovery of blood leukocyte counts, ROS generation by phagocytes, higher intensity of Th1- and Th2-dependent immune response, cortisol, IFNγ and IL-4 concentrations in blood serum, like as stimulation of blood phagocytic capacity. Restoration of Th1- and Th2-dependent immune response is associated with normalized concentrations of, respectively, IFNγ and IL-4 levels in blood serum.

Background and aim Lipoprotein-apheresis (LA) is a therapeutic approach used against severe forms of dyslipidemia in patients who are non-responders or intolerant to pharmacological treatments. However, little is known about the potential pleiotropic effects of LA, particularly regarding the immune system and its regulation. Thus, in an attempt to analyse the potential effects of dyslipidemia and LA on the regulation of CD4 + T cells activation and lineage differentiation, we compared the CD4 + T cells cytokines secretion profiles of dyslipidemic patients before and after LA with the profiles observed in healthy donors. Methods CD4 + T cells were isolated from 5 LA patients and 5 healthy donors and activated with anti-CD3 or anti-CD3 + anti-CD46 antibodies. The supernatants were collected after 36 h incubation and levels of secreted cytokines analysed by flow cytometry. Results Our results revealed a deep remodelling of CD4 + T cells cytokines secretion patterns in dyslipidemic patients compared to healthy donors, as reflected by a 15 times higher IFN-γ secretion rate after CD3 + CD46 co-activation in dyslipidemic patients after LA compared to healthy subjects and 8 times higher after CD3 activation alone ( p = 0.0187 and p = 0.0118 respectively). Moreover, we demonstrated that LA itself also modifies the phenotype and activation pattern of CD4 + T-cells in dyslipidemic patients. Conclusion These observations could be of fundamental importance in the improvement of LA columns/systems engineering and in developing new therapeutic approaches regarding dyslipidemia and associated pathologies such as atherosclerosis and type 2 diabetes. Elevated concentrations of Nt-pro-BNP were revealed in 11% of apparently healthy subjects aged 19-24 years and 22% of middle-aged (34-55 years) subjects. An increase in Nt-pro-BNP blood concentration was detected in 90% of patients with metabolic syndrome. Elevated levels of Nt-pro-BNP in peripheral blood occur with age. Higher concentrations of the peptide were revealed in women. Inhibition of lymphocytes proliferation and differentiation accompanied by increasing concentration of Nt-pro-BNP in blood is associated with a deficiency of the IL-2 level due to increased IL-10 concentration. Immunosuppressive effect of IL-10 declares itself at the level of innate immune reactions as a decrease in blood counts of naive CD45RA+ T lymphocytes, natural killer cells, T cells with adhesion receptor ( CD56+) and adhesion receptor ligand ( CD62L+). To prevent a loss of the intercellular pool, the effect of Nt-pro-BNP is activated. No statistically significant correlations between the increased Nt-pro-BNP concentration in blood and the content of serum IgM, IgG, IgA, IgE, IL-6, TNF-α, IFN-γ, CEC C1q, or С3d, as well as glucose, hemoglobin, transferrin, iron, or free fatty acids were revealed in apparently healthy subjects. Studies over the last two decades have revealed profound immunomodulatory aspects of vitamin D on various aspects of the immune system. This review will provide an overview of Vitamin D metabolism, a description of dendritic cell subsets, and highlight recent advances on the effects of vitamin D on dendritic cell function, maturation, cytokine production and antigen presentation. The active form of vitamin D, 1,25(OH)2D3, has important immunoregulatory and anti-inflammatory effects. Specifically, the 1,25(OH)2D3-Vitamin D3 complex can affect the maturation and migration of many dendritic cell subsets, conferring a special immunoregulatory role as well as tolerogenic properties affecting cytokine and chemokine production. Furthermore, there have been many recent studies demonstrating the effects of Vitamin D on allergic disease and autoimmunity. Since the discovery of interleukin-33 (IL-33) ligand for the ST2 receptor, scarce studies have implicated this cytokine in the pathogenesis of Behçet’s disease (BD). IL-33 is member of the IL-1 superfamily of cytokines that is expressed by epithelial cells, endothelial cells, inflammatory cells and central nervous tissue. Its expression is upregulated following pro-inflammatory situations making IL-33 essential to innate immunity. IL-33 can function both as a traditional cytokine and as a nuclear factor regulating gene transcription. It is thought to function as an “alarmin” which is released following cell necrosis to alert the immune system of tissue damage or stress. Over the last 20 years it has become recognized that low-grade inflammation plays a role in cardiovascular disease. More recently, participation of the innate and the adaptive immune response in mechanisms that contribute to inflammation in cardiovascular disease has been reported in atherosclerosis and hypertension. Different subsets of lymphocytes and their cytokines are involved in vascular remodelling in hypertension, chronic kidney disease and heart disease. Effector T-cells include Th1 (interferon-γ -producing) and Th2 (interleukin-4 producing) lymphocytes, as well as Th17 (which produce interleukin-17) and T-suppressor lymphocytes such as Treg-cells (regulatory T-cells), which express the transcription factor Foxp3 (forkhead box P3) and participate respectively as pro- and anti-inflammatory cells. Pro-inflammatory T-lymphocytes participate in mechanisms of cardiovascular disease in part by mediating the effects of angiotensin II and mineralocorticoids. Involvement of immune mechanisms in cardiac, vascular and renal changes in hypertension has been demonstrated in many experimental models, an example being the Dahl-salt sensitive rat and the spontaneously hypertensive rat. How activation of immunity is triggered remains unknown, but neo-antigens could be generated by elevated blood pressure through damage-associated molecular pattern receptors or other mechanisms. Once activated, Th1 cells may contribute to blood pressure elevation by affecting the kidney, vascular remodelling of blood vessels directly via the effects of the cytokines produced or through their effects on perivascular fat. Treg-cells protect from blood pressure elevation by acting upon similar targets. Recent data suggests that participation of these mechanisms that have been demonstrated already in murine models also occurs in humans. Abstract: The mechanisms of immune response following yellow fever (YF-17DD) vaccination are still poorly understood. In this study, we have performed a longitudinal investigation (days 0, 7, 15 and 30) to characterize the cytokine profile of innate and adaptive immunity following YF-17DD first-time vaccination. Data from non-stimulated cultures demonstrated a prominent participation of the innate immunity with increased frequency of TNF-α+ neutrophils and IFN-γ+ NK-cells at day 7 besides TNF-α+ monocytes at day 7, day 15 and day 30. Increased frequency of IL-10+ monocytes was observed at day 15 and day 30, and decreased percentage of IL-4+ NK-cells were detected at day 7, day 15 and day 30. Time-dependent and oscillating cytokine pattern was observed in CD4+ T-cells, with low percentage of IL-12+, IL-4+ and IL-10+ cells at day 7 and increased frequency of TNF-α+ cells at day 15 besides IFN-γ+ and IL-5+ cells at day 15 and day 30. Later changes with increased percentage of IL-12+ and IL-5+CD8+ T-cells were observed at day 30. Increased frequency of IL-10+ B-cells was observed at day 15, when seroconversion was detected in all vaccinees. The overall cytokine analysis of non-stimulated leukocytes showed a transient shift towards a pro-inflammatory profile at day 7, mainly due to changes in the innate immunity, which draws back toward a mixed/regulatory pattern at day 15 and day 30. The changes induced by the in vitro YF-17DD vaccine-stimulation were mainly observed at day 0 and day 7 (before seroconversion) with minor changes at day 15 and day 30 (after seroconversion). These data support the hypothesis that a complex network with mixed pro/anti-inflammatory cytokine profile is associated with the establishment of the protective immunity following YF-17DD primo-vaccination, free of adverse events. STAT3 has been known as a mediator for gene expression induced by many important cytokines. Recent studies have suggested that STAT3 has important functions in regulation of both innate and adaptive immunity. Loss of STAT3 in immune cells caused severe inflammation in response to pathogens. and The crosstalk between innate adaptive immunity is regulated by cytokines and complex interactions between cells of the immune system. A variety of endogenous agents are involved in the regulation of the cytokine network. Especially, eicosanoids and ROIs have a great impact on the regulation of cytokine production. Eicosanoids (prostanoids, leukotrienes and lipoxins) are produced mainly by inflammatory cells while their receptors are distributed on the cells of both arms of the immune system. Depending on the predominant prostanoid produced and the profile of prostanoid receptors expression on immune cells, eicosanoids can selectively regulate the production of Th1 and Th2 driven cytokines. Inflammatory cells (neutrophils, macrophages), are also a rich source of large amounts of ROIs. In this paper we have focused on the role of taurine chloramine (TauCl), the physiological product of neutrophil MPO-halide system, in the regulation of immune system. It is well documented that TauCl has pleiotropic effects on the inductive phase of the immune response. TauCl''s immunoregulatory properties result from its ability to modulate the production of cytokines and eicosanoids.
