Narrow intraspecific variation for sugar content in pepino ( Solanum muricatum Aiton) hinders the development of new pepino cultivars with improved fruit quality. However, some wild related species have high soluble solids concentration (SSC) and titratable acidity (TA). Generation means analysis was used to estimate genetic parameters for SSC, TA and ascorbic acid concentration (AAC) in two families of interspecific crosses between a S. muricatum parent (Sm-32) and one accession of each of the wild species Solanum caripense (Sc-4) and Solanum tabanoense (St-1). In both families, the additive effect [d] was the only significant parameter for SSC, the alleles of wild species contributing positively to increasing SSC values. For TA, genetic effects associated with additivity [d] and dominance [h] were detected in both families. Additionally, in Sm-32 × Sc-4 nonallelic interactions associated with dominance ([j] and [l]) were significant. For this trait, alleles from the wild species contribute additively to a high value of the character, but are recessive to those of the cultivated S. muricatum. No genetic variation was found for AAC. Broad-sense heritabilities for SSC and TA were intermediate (0.40 to 0.50). Positive significant genetic correlations (around 0.68 in both families) were found between SSC and TA. The results obtained in backcrosses of interspecific hybrids to S. muricatum, together with the estimates of genetic advance after selection, indicate that recovery of individuals with higher SSC and TA and adequate SSC/TA ratio can be achieved in a few backcrosses. The use of hybrids as a new type of cape gooseberry (Physalis peruviana L.)cultivars could improve yield in this crop, but little or no information is available on hybrid performance. We studied several vegetative characters, yield, fruit weight and fruit shape, soluble solids content (SSC), tiltratable acidity (TA)and ascorbic acid content (AAC) in three hybrids of cape gooseberry and their parents grown outdoors and in a glasshouse. Interaction dominance X environment for yield was very important; a higher dominance effect was detected in the glasshouse, than that observed outdoors. Quality characters were highly affected by the environment and showed variable results for the different families. For fruit comparison traits, the additive and additive X environment interactions were most important. Broad-sense heritability for all characters was high to medium (0.48-0.91), indicating that a high response to selection would be expectecd. Eggplant (Solanum melongena) is amenable to grafting and this technique can be exploited to improve production of this vegetable crop. Here, we study the performance of the 'Crista! F l ' eggplant cultivar grafted onto a total of 1 7 rootstocks, including five families, each o f which is made by two parents and their hybrid, as well a commercial eggplant rootstock and a self-grafted control. Three families consist of eggplant rootstocks, including a S. melongena intraspecific family and two interspecific families of S. melongena with S. incanum and S. aethiopicum. Two other families are made of interspecific hybrids between tomato (S. lycopersicum) and S. habrochaites. Overall, eggplant rootstocks showed a good compatibility and graft success, while tomato rootstocks had a poor compatibil ity and only S. habrochaites rootstocks could be evaluated for yield and fruit production. Important differences were found for susceptibility to nematodes, with some S. melongena accessions showing the best results. Eggplant hybrids were intermediate to their parents in nematode resistance and tomato rootstocks proved to be very susceptible to nematodes. Yield, fruit number and earliness were higher in the most vigorous rootstocks, in particular in the S. melongena x S. aethiopicum hybrid. Good performance was also observed for some S. melongena accessions, but poor results were obtained when using S. incanum, S. aethiopicum and S. habrochaites rootstocks. Some differences were observed for fruit size and shape among the different rootstocks, but for the best rootstocks no differences of commercial relevance were apparent. The results suggest that S. melongena germplasm and both intraspecific and interspecific eggplant hybrids seem to be promising materials for developing new rootstocks for eggplant production. Greenhouse-grown tomatoes (Lycopersicon esculentum) often have poor internal organoleptic and nutritional qualities. "Heirloom" (traditional) varieties of tomato possess unique or superior fruit quality characteristics and may be useful for the development of new greenhouse tomato cultivars with improved internal quality. A collection of 55 North American "heirloom" varieties was evaluated for fruit colour, shape, weight, firmness (external quality traits) and total soluble solids (TSS) content, titratable acidity (TA), TSS:TA ratio, flavour intensity (FI; TSS×TA) and ascorbic acid (AA) concentration (internal quality traits). The F1 hybrid 'Daniela' and pure line 'NE-1' were used as controls. A wide diversity was found among "heirlooms" for all traits studied, and several sources of variation (e.g., varieties 'Moira', 'Paragon', 'White Queen', 'Black Prince' and 'Green Zebra') useful for the improvement of fruit quality were identified. Brightness (L*), hue angle and chroma parameters facilitated the classification of varieties into colour groups (white, yellow, orange, red or black) and the identification of the varieties with better colour characteristics within each colour group. Although all "heirloom" varieties had lower firmness than the delayed ripening cv. 'Daniela', some of them (e.g., 'Moira' and 'Paragon') were very firm. Values obtained for TSS, TA, FI and AA demonstrated that many "heirloom" varieties had better organoleptic and nutritional qualities than modern varieties. Ripening fruit of pepino (Solanum muricatum) are susceptible to sunscald. We studied the effects of the sun over 6 d in detached mature-green fruit of cultivars 'Puzol' (used in salads) and 'Sweet Long' (used in desserts) from a summer 2001 greenhouse crop in Valencia, Spain. In both cultivars, severe sunscald symptoms developed and the skin turned yellow and brown and papery thin, while the flesh softened and browned. Infrared thermography of fruits showed that 'Puzol' is hotter in the sun (44.5 °C) than 'Sweet Long' (44.1 °C) and that maximum surface temperature increased 1 K over the 6 d. The mesocarp in the exposed part of the fruit (M1) was 2.2 K wanner than in the shaded part (M2). Average electrolyte leakage (EL) increased from 70.3% to 76.2% with sun exposure, and was as a mean higher in 'Sweet Long' (76.4%) than in 'Puzol' (69.3%), and in M1 (73.9%) than in M2 (71.7%). Ascorbic acid content (AAC) decreased over the 6 d, especially in 'Sweet Long' (from 22.0 to 7.8 mg 100 g-1) and in M1 (from 17.8 to 3.6 mg 100 g-1). Higher EL and lower AAC after exposure in M1 (78.1% and 3.6 mg 100 g-1, respectively), compared with M2 (74.4 % and 10.6 mg 100 g-1), indicate that small differences in temperature with sun exposure affect the mesocarp. Changes in AAC reflect damage (r=-0.71) more than changes in EL (r=0.49). Milder symptoms of sunscald in M2 than in M1 suggest that temperatures above 40-42°C are necessary for inducing severe sunscald symptoms. [ABSTRACT FROM AUTHOR]

Participatory plant breeding (PPB) has the potential to be an alternative to a centralized breeding model for niche markets that are historically underserved. Our objectives were to evaluate the performance of two parental cultivars from a Canadian PPB program: a modern spring wheat (Triticum aestivum L.) cultivar (5602HR) and a landrace (Red Fife)—with the progeny of the cross when selected by two farmers located 1800 km apart (denoted Farm1 and Farm2) and evaluate progeny differences from each other under organic conditions. Red Fife was 18 cm taller, matured 5 days later, and had greater lodging susceptibility, greater seed mass, and 3.5% lower protein concentration than 5602HR. Farmer genotypes were similar to 5602HR in protein concentration and lodging severity, and similar to Red Fife in plant height (14 cm taller than 5602HR) and seed mass. Farmer genotypes did not yield differently from either parent. Farmer genotypes did not differ from each other in most parameters measured; however, Farm1 had greater lodging resistance under high fertility conditions than Farm2 despite similar plant height. This research provides a proof of concept for the role that farmer selectors can play in selecting for positive traits for organic production and provide insight into organic farmers' preferences. Tomato (Solanum lycopersicum L.) is one of the most important cultivated vegetables in the world. However, in some countries such as Mexico the lack of cultivars adapted to different environmental production conditions is a limitation. Moreover, recent studies have indicated that breeding aimed at increasing yield has led to a loss of genetic diversity. Therefore, it is necessary to explore and characterize new sources of germplasms. This study aimed to characterize new sources of germplasm and identify the most transcendental traits for distinguishing tomato types and lines that are useful for the genetic improvement of the species. Sixty characters were evaluated in 16 advanced lines of native tomatoes from Central and Southern Mexico during the fall–winter cycles 2023–2024 at the Bajío Experimental Station, Celaya, Guanajuato, Mexico, based on the guidelines of the International Union for the Protection of New Varieties of Plants (UPOV) and the International Plant Genetic Resources Institute (IPGRI). The data were analyzed using descriptive statistics, analysis of variance and post hoc tests, canonical discriminant analysis, and the Eigenanalysis selection index method (ESIM). Morphological variation showed that five qualitative traits were determinant factors in distinguishing tomato types and lines, whereas agronomic discriminant traits were the equatorial and polar diameters of the fruit and its ratio, number of locules, pedicel length, stem length, and internode distance. In addition, significant positive correlations were found between leaf length and width, equatorial diameter of the fruit, and polar diameter of the fruit. Lines JCM-17, JMC-10, and JCM-01 were the most selectable lines according to the ESIM values. The morphological variation found and the characteristics with higher selection values identified may be valuable for optimizing the tomato genetic improvement process in general. Objective: Cucumber mosaic virus (CMV) is a significant pathogen causing quality loss in spinach. Although host genetic resistance is the primary method of managing CMV infection in this crop, CMV resistance genes are not widely utilized in spinach breeding programs as the genetic and molecular mechanisms underlying resistance are not yet fully understood. CMV infections were therefore studied in different lines of spinach plants, and their progeny, to develop a model of the genetic basis of CMV resistance. Results: Visual observations and RT-PCR assays revealed that three monoecious lines (03-258, 03-263, and 03-336) were susceptible to CMV, while three traditional resistant cultivars and a near-isogenic line (NIL-M) exhibited resistance. A dioecious line (03–009) consisted of susceptible and resistant plants. Notably, resistant plants did not exhibit the lesions typical of the hypersensitive response. Genetic analysis of progeny from the cross NIL-M × 03-336 indicated that a single dominant allele (designated SRCm1, standing for Spinach Resistance to CMV 1) controlled CMV resistance; analysis of sib-cross progeny populations derived from line 03–009 supported this conclusion. Climate change significantly threatens crops, mainly through drought stress, affecting barley, which is essential for food and feed globally. Ten barley cultivars were evaluated under normal and drought stress conditions during the 2019/20 and 2020/21 seasons, focusing on traits such as days to heading and maturity, plant height, number of spikes m−2, spike length, 1000-kernel weight, and biological and grain yield. Drought stress significantly reduced most of these traits. The genotypes showed significant differences in their responses to irrigation treatments, with the interaction between seasons and cultivars also being significant for most traits. The grain yield and 1000-kernel weight were among the least affected traits under drought stress, respectively. Notably, Giza138 and Giza126 showed strong drought tolerance, suitable for drought-resilient breeding. In season one, Giza126, Giza134, and Giza138 yielded 13%, 9%, and 11%, respectively, while Giza135 and Giza129 showed higher reductions at 31% and 39%. In season two, Giza126, Giza134, and Giza138 had reductions of 14%, 10%, and 13%, respectively, while Giza135 and Giza129 again exhibited higher reductions at 31% and 38%. These cultivars also showed strong performance across various stress tolerance indices, including the MP, YSI, STI, GMP, and YI. Giza 134 demonstrated the lowest values for the SDI and TOL, indicating superior drought stress tolerance. On the other hand, Giza 129 and Giza 135 were the most sensitive to drought stress, experiencing significant reductions across critical traits, including 6.1% in days to heading, 18.37% in plant height, 28.21% in number of kernel spikes−1, 38.45% in grain yield, and 34.91% in biological yield. In contrast, Giza 138 and Giza 2000 showed better resilience, with lower reductions in the 1000-kernel weight (6.41%) and grain yield (10.61%), making them more suitable for drought-prone conditions. Giza 126 and Giza 132 also exhibited lower sensitivity, with minimal reductions in days to heading (2%) and maturity (2.4%), suggesting potential adaptability to water-limited environments. Giza 126 maintained the highest root lengths and had the highest stomatal conductance. Giza 138 consistently had the highest chlorophyll content, with SPAD values decreasing to 79% under drought. Despite leading in shoot length, Giza 135 decreased to 42.59% under drought stress. In conclusion, Giza 126 and Giza 138 showed adaptability to water-limited conditions with minimal impact on phenological traits. Giza 126 had the longest roots and highest stomatal conductance, while Giza 138 consistently maintained a high chlorophyll content. Producing uniform Cannabis sativa (Cannabis) for medicinal/recreational flower production through sexual propagation has been problematic, leading to dominance of clonal propagation from "mother plants" in the cannabinoid industry, which also faces significant limitations. Cannabis tissue culture (TC) methods have been developed to overcome these challenges, but the long-term health and maintenance of Cannabis explants in TC have been largely overlooked in previous studies. The current study focused on the development of an efficient and optimized micropropagation protocol covering the entire process, with a specific focus on the health and performance in the multiplication stage. Multiplication media were formulated hormone-free to avoid longer-term vitrification issues, resulting in single-main-shoot cultures rather than multiple-shoot cultures. This instigated the use of stage II explant types different from the standard shoot tips previously used for multiple shoot cultures. Multiplication media were further improved from the basal salt composition via nitrogen and calcium additives. The optimized protocol was used on eight diverse Cannabis cultivars to test its applicability across various genetic backgrounds. Results indicated that the protocol was effective for conservation purposes across all cultivars and achieved good long-term multiplication rates for some but not all. We conducted an analysis on the combined effects of two light conditions (L1: greenhouse natural lighting; L2: greenhouse natural lighting plus supplemental lighting (SL)) and three nutrient solution concentrations (EC, NS1: 3.2 dS/m; NS2: 3.7 dS/m; NS3: 4.2 dS/m) on the growth, fruit production, and quality of two cherry tomato cultivars with different fruit coloring ('Baiyu' and 'Qianxi'). The plants subjected to NS2 exhibited enhanced growth, photosynthetic parameters, and fruit production. The utilization of SL further enhanced stem diameter, leaf number, and single fruit weight, resulting in higher fruit weight per plant in 'Baiyu', which was not observed in 'Qianxi'. The growth, fruit size, and fruit weight of both cultivars cultivated under NS3 conditions were suppressed, while these fruits exhibited elevated levels of total soluble solids (TSS), soluble sugars, vitamin C, polyphenols, fructose, glucose, sucrose, citric acid, and carotenoids. These levels were further enhanced by SL treatment. The improvement of fruit quality through the application of SL was found to be cultivar and EC dependent. In 'Baiyu', SL at NS1 significantly enhanced the accumulation of fruit water, minerals (N, P, K, Ca, and Mg), TSS, vitamin C, fructose, sucrose, and carotenoids. However, this effect was not observed in 'Qianxi'. The combination of SL and EC 4.2 dS/m (NS3) generally contributes to the enhancement of fruit quality, while SL and EC 3.7 dS/m can ensure consistent fruit production. The yellowish-white fruit cultivar exhibited higher levels of soluble sugars, vitamin C, and polyphenols under L2NS3 conditions compared to the red fruit cultivar, whereas the carotenoid content showed an opposite trend. Myrteae is the most species-rich tribe in the Myrtaceae family, represented by a range of socioeconomically and ecologically significant species. Many of these species, including commercially relevant ones, have become increasingly threatened in the wild, and now require conservation actions. Tissue culture presents an appropriate in vitro tool to facilitate medium-term and long-term wild germplasm conservation, as well as for commercial propagation to maintain desirable traits of commercial cultivars. So far, tissue culture has not been extensively achieved for Myrteae. Here, tissue culture for Eugenia, one of the most species-rich genera in Myrteae, is reviewed, giving directions for other related Myrteae. This review also focuses on ex situ conservation of Australian Myrteae, including using seed banking and field banking. Despite some progress, challenges to conserve these species remain, mostly due to the increasing threats in the wild and limited research. Research into in vitro methods (tissue culture and cryopreservation) is paramount given that at least some of the species are 'non-orthodox'. The close interconnection of plants with rhizosphere- and root-associated microorganisms is well recognized, and high expectations are raised for considering their symbioses in the breeding of future crop varieties. However, it is unclear how consistently plant-mediated selection, a potential target in crop breeding, influences microbiome members compared to selection imposed by the agricultural environment. Landraces may have traits shaping their microbiome, which were lost during the breeding of modern varieties, but knowledge about this is scarce. We investigated prokaryotic community composition along the radial root axis of two European maize (Zea mays L.) landraces. A sampling gradient included bulk soil, a distal and proximal rhizosphere fraction, and the root compartment. Our study was replicated at two field locations with differing edaphic and climatic conditions. Further, we tested for differences between two plant developmental stages and two precipitation treatments. Community data were generated by metabarcoding of the V4 SSU rRNA region. While communities were generally distinct between field sites, the effects of landrace variety, developmental stage, and precipitation treatment were comparatively weak and not statistically significant. Under all conditions, patterns in community composition corresponded strongly to the distance to the root. Changes in a- and b-diversity, as well as abundance shifts of many taxa along this gradient, were similar for both landraces and field locations. Most affected taxa belonged to a core microbiome present in all investigated samples. Remarkably, we observed consistent enrichment of Actinobacteriota (particularly Streptomyces, Lechevalieria) and Pseudomonadota (particularly Sphingobium) toward the root. Further, we report a depletion of ammoniaoxidizers along this axis at both field sites. We identified clear enrichment and depletion patterns in microbiome composition along the radial root axis of Z. mays. Many of these were consistent across two distinct field locations, plant developmental stages, precipitation treatments, and for both landraces. This suggests a considerable influence of plant-mediated effects on the microbiome. We propose that the affected taxa have key roles in the rhizosphere and root microbiome of Z. mays. Hardy geranium is a popular ornamental plant with a high market demand. As a result, there is a strong need for continuous innovation to improve its ornamental qualities. Interspecific hybridization is a widely used technique for introducing desirable traits and creating new cultivars with added value. To explore the possibilities of creating novelty within the Geranium genus, we conducted 2438 interspecific crosses over three flowering seasons, resulting in the successful harvest of 445 seeds and the final production of 82 seedlings. Using embryo rescue increased the germination rate of a seedling from 0.17% to 1.01%. To validate the paternal genetic contribution in the new seedlings, we used amplified fragment length polymorphism (AFLP) to assess the proportion of uniquely inherited paternal markers in all seedlings. AFLP analysis confirmed that 54 of the 82 seedlings were indeed true hybrids (65.8% hybridization success rate). A morphological evaluation of the flowers and leaves, including flower diameter (mm), style length (mm), number of flowers, flowering period (weeks), and color of both flowers and leaves, provided further confirmation of the F1 hybrid status of the seedlings. The results of our interspecific breeding demonstrated the efficiency of interspecific hybridization in hardy geraniums and its potential to create distinctive and novel cultivars for the ornamental market. Tomatoes (Solanum lycopersicum L.) represent a valuable crop species cultivated on a global scale, with Thailand playing a notable role in Southeast Asia's agricultural landscape. However, a comprehensive understanding of the genetic underpinnings, population dynamics, and genetic correlations among tomato cultivars within Thailand remains relatively nascent. This study conducted an extensive analysis of 283 tomato accessions sourced from Thailand's largest germplasm repository, employing approximately 2.4 million single-nucleotide polymorphisms (SNPs) obtained through whole-genome sequencing (WGS). The results of the genetic diversity assessment demonstrate a consistent delineation of groups, as indicated by UPGMA clustering, revealing five distinct clusters, while population structure analysis unveils twelve subpopulations. These findings underscore the limited genetic reservoir within Thai tomato germplasm, providing valuable insights for germplasm management strategies and offering a promising avenue for the refinement of breeding programs aimed at enhancing tomato genetics. The main production area of Camellia vietnamensis (C. vietnamensis) is in the low mountain and hilly areas of southern China. The low survival rate of seedlings caused by drought is one of the main obstacles restricting the development of the C. vietnamensis industry. An exploration of the key adaptation mechanism of C. vietnamensis to drought stress is important in order to improve its drought resistance. We conducted a study on the morphological, physiological, biochemical, and drought resistance-related genes of five C. vietnamensis cultivars grown in Hainan province under varying degrees of drought stress. The results indicate that drought stress can lead to a decrease in the relative water content and photosynthetic capacity of C. vietnamensis leaves. Compared with the control, the drought damage index, malondialdehyde, relative electrical conductivity, soluble protein, soluble sugar and proline contents of the five C. vietnamensis cultivars increased with drought-stress duration and degree. With increasing drought-stress intensity, the activity of antioxidant enzymes and the content of related metabolites (total polyphenols, total flavonoids, tea saponins) gradually increased, and the expression levels of phenylpropanoid pathway-related genes (Cv4CL1, CvCAD1, CvCAD2, CvPOX1, CvPOX2, CvPOX3) were upregulated. Based on the results of the drought tolerance coefficients, principal component analysis, and hierarchical cluster analysis, we classified five C. vietnamensis cultivars into drought-tolerant cultivars ('Haida 1'); moderately drought-tolerant cultivars ('Haida 4' and 'Wanhai 4'); and drought-sensitive cultivars ('Wanhai 3' and 'Wanhai 1'). The present study aims to establish the application of fluorescence spectroscopy as a mobile field method in the determination of varietal differences in Spinach (Spinacia oleracea L.). The experimental studies were conducted on site at the farm where the spinach accessions were grown. Fluorescence analysis was conducted using a light source emitting at a wavelength of 285 nm and a mobile fiber-optic experimental setup developed by the authors. The subjects of this research are leaves from Rembord, Marishka, Marquisa, and Victoria. The correlation between the emission signals of the samples was established. This discovery enables the successful application of mobile fluorescence spectroscopy as a rapid tool for determining the origin of unknown leaves. The results from the experiment can be used to optimize the timing for analyzing varietal differences in spinach genotypes after harvest. Fluorescence spectroscopy in a fiber-optical configuration will support the process of determining the belonging of a specific variety to a given variety, even for samples of unknown origin, when it is necessary to qualify the result of accessions in a short time. The potato (Solanum tuberosum L.), an important field crop consumed extensively worldwide, is adversely affected by abiotic stress factors especially drought. Therefore, it is vital to understand the genetic mechanism under drought stress to decrease loose of yield and quality. This trial aimed to screen drought-responsive gene expressions of potato and determine the drought-tolerant potato cultivar. The trial pattern is a completely randomized block design (CRBD) with four replications under greenhouse conditions. Four cultivars (Brooke, Orwell, Vr808, Shc909) were irrigated with four different water regimes (control and three stress conditions), and the gene expression levels of 10 potato genes were investigated. The stress treatments as follows: Control = 100% field capacity; slight drought = 75% field capacity; moderate drought = 50% field capacity, and severe drought 25% field capacity. To understand the gene expression under drought stress in potato genotypes, RT-qPCR analysis was performed and results showed that the genes most associated with drought tolerance were the StRD22 gene, MYB domain transcription factor, StERD7, Sucrose Synthase (SuSy), ABC Transporter, and StDHN1. The StHSP100 gene had the lowest genetic expression in all cultivars. Among the cultivars, the Orwell exhibited the highest expression of the StRD22 gene under drought stress. Overall, the cultivar with the highest gene expression was the Vr808, closely followed by the Brooke cultivar. As a result, it was determined that potato cultivars Orwell, Vr808, and Brooke could be used as parents in breeding programs to develop drought tolerant potato cultivars. [ABSTRACT FROM AUTHOR]

Chilean peppers, cultivated from Capsicum pubescens, are globally renowned as popular vegetable and spice crops. C. pubescens belongs to the Capsicum L. (pepper) family and is one of the five pepper cultivars grown in China. In this study, we assembled and annotated the complete mt genome of C. pubescens. We investigated several aspects of its genome, including characteristics, codon usage, RNA editing sites, repeat sequences, selective pressure, gene clusters, and phylogenetic relationships. Furthermore, we compared it with other plant mt genomes. The data we obtained will provide valuable information for studying evolutionary processes in the Capsicum genus and will assist in the functional analysis of Capsicum mitogenomes. Background: Genomewide prediction estimates the genomic breeding values of selection candidates which can be utilized for population improvement and cultivar development. Ridge regression and deep learning-based selection models were implemented for yield and agronomic traits of 204 chile pepper genotypes evaluated in multi-environment trials in New Mexico, USA. Results: Accuracy of prediction differed across different models under ten-fold cross-validations, where high prediction accuracy was observed for highly heritable traits such as plant height and plant width. No model was superior across traits using 14,922 SNP markers for genomewide selection. Bayesian ridge regression had the highest average accuracy for first pod date (0.77) and total yield per plant (0.33). Multilayer perceptron (MLP) was the most superior for flowering time (0.76) and plant height (0.73), whereas the genomic BLUP model had the highest accuracy for plant width (0.62). Using a subset of 7,690 SNP loci resulting from grouping markers based on linkage disequilibrium coefficients resulted in improved accuracy for first pod date, ten pod weight, and total yield per plant, even under a relatively small training population size for MLP and random forest models. Genomic and ridge regression BLUP models were sufficient for optimal prediction accuracies for small training population size. Combining phenotypic selection and genomewide selection resulted in improved selection response for yield-related traits, indicating that integrated approaches can result in improved gains achieved through selection. Conclusions: Accuracy values for ridge regression and deep learning prediction models demonstrate the potential of implementing genomewide selection for genetic improvement in chile pepper breeding programs. Mulberry fruits are rich in bioactive components renowned for their antioxidant properties and potential health benefits. This study thoroughly investigated the impact of cultivation modes on the phenological stages, yield, bioactive components, and antioxidant activity of two mulberry cultivars, Yueshen Dashi (YS-DS) and Xinjiang Baisang (XJ-BS). Notably, greenhouse cultivation led to earlier phenology and shorter fruit development maturation durations compared to field cultivation. Despite a decrease in fruit production and firmness, the greenhouse-grown mulberries exhibited higher individual fruit fresh weight. The content of bioactive components, encompassing anthocyanins, polyphenols, flavonoids, and vitamin C, and of antioxidant activity (measured in the FRAP and DPPH radical scavenging assays) was found to be lower in the greenhouse-grown mulberries than in those cultivated in the field. The contents of total polyphenols and flavonoids showed robust positive correlations in the FRAP and DPPH radical scavenging assays, which suggests that the antioxidant activity of mulberry fruit might be primarily attributable to the bioactive components of total polyphenols and flavonoids. Interestingly, the sugar content and hydroxyl radical scavenging activity (HRSA) displayed an inverse relationship between the two cultivars in the greenhouse versus field conditions. The multivariate analysis highlighted distinct patterns for different cultivars under varying cultivation modes. This study underscores the potential to enhance bioactive components and antioxidant activity through effective manipulation of climate conditions, thereby unlocking the full nutritional potential of mulberry fruits on a large scale in greenhouse environments. Sorghum holds the potential for enhancing food security, yet the impact of the interplay of water stress and salinity on its growth and productivity remains unclear. To address this, we studied how drought and salinity affect physiological traits, water use, biomass, and yield in different tropical sorghum varieties, utilizing a functional phenotyping platform, Plantarray. Cultivars (Kuali, Numbu, Samurai2) were grown under moderate and high salinity, with drought exposure at booting stage. Results showed that Samurai2 had the most significant transpiration reduction under moderate and high salt (36% and 48%) versus Kuali (22% and 42%) and Numbu (19% and 16%). Numbu reduced canopy conductance (25% and 15%) the most compared to Samurai2 (22% and 33%) and Kuali (8% and 35%). In the drought*salinity treatment, transpiration reduction was substantial for Kuali (54% and 57%), Samurai2 (45% and 60%), and Numbu (29% and 26%). Kuali reduced canopy conductance (36% and 53%) more than Numbu (36% and 25%) and Samurai2 (33% and 49%). Biomass, grain yield, and a-100 grain weight declined in all cultivars under both salinity and drought*salinity, and Samurai2 was most significantly affected. WUEbiomass significantly increased under drought*salinity. 1. Introduction: Nowadays, the cultivation of vegetables in greenhouses using soilless beds has become a widely adopted practice for vegetable production worldwide. The inclusion of new greenhouse crops like melons offers the potential to supply consumers with fresh and high-quality produce. Greenhouse melon cultivation is a relatively new practice in Iran, and there is a scarcity of published information regarding its various aspects of production in a greenhouse setting. One crucial factor that significantly influences the quality and quantity of greenhouse crops, such as melons, when employing soilless cultivation techniques, is the selection of appropriate nutrient solutions. Therefore, the current study was conducted to investigate the influence of different nutrient solutions on vegetative growth, seed and fruit characteristics, and biochemical status of three melon cultivars (Ahlam, Polinica, Curie) under greenhouse condition. 2. Materials and Methods: The present study was conducted at the Horticultural Sciences Department of Shahid Chamran University of Ahvaz in southwestern Iran during the 2017-2018. A factorial experiment based on randomized complete blocks design with three replications, was carried out to assess the impact of three nutrient solution treatments (Pars, Shizuoka, Hachmath) and three different cultivars (Ahlam, Polinica, Curie) on key characteristics of greenhouse melon. The melon plants of the three cultivars were cultivated in pots containing 8 L of a growth medium consisting of a 4:1 ratio of cocopeat to perlite. At the end of experiment, four characteristic groups including vegetative growth parameters (dry weight of leaf, root, stem and plant and the ratio of leaf dry weight to root dry weight), seed characteristics (seed dry weight and hundred seeds weight), physical parameters of fruit (length, diameter, weight, wide of seed cavity and flesh thickness) and biochemical traits (ascorbic acid content, carotenoid content, taste index, total soluble solid content and titratable acidity) were measured. 3. Results and Discussion: The findings demonstrated that in the Hochmuth and Pars nutrient solutions, the Ahlam cultivar exhibited the highest total dry weight (63.66 g and 37.65 g, respectively). However, the Polinica cultivar showed the highest total dry weight in the Pars nutrient solution. The Hochmuth nutrient solution resulted in the highest leaf dry weight for the Ahlam cultivar (43.77 g). The Ahlam cultivar also displayed the highest fruit weight (1584 g and 1510 g, respectively), dry weight of hundred seeds (4.04 g and 3.56 g, respectively), seed dry weight per fruit (11.56 g and 10.75 g), weight of seed cavity contents (104.10 g and 114 g, respectively), fruit diameter (13.51 cm and 13.58 cm, respectively), and fruit lengths (17.58 cm and 18.28 cm, respectively). The weight of seed cavity content was influenced by both the cultivar and the nutrient solution. The Ahlam cultivar, when fed with the Pars nutrient solution, exhibited the highest total titratable acidity (0.07 %) and seed cavity thickness (6.38 cm). Regarding the Polinica cultivar, the Pars nutrient solution resulted in the highest total soluble solids (11.98 %B) and taste index (337.3), while the Hochmuth nutrient solution led to the highest ascorbic acid content (1.05 mg g-1 FW). The nutrient solution treatments did not have a significant effect on carotenoid content of the melon cultivars. Different amounts of elements in different nutrient solutions cause different growth behaviors of plants. For instance, the melon plants fed with Pars nutrient solution showed higher values of growth parameters comparing to the other plants, and it could be related to high N content of this solution. The similar results have been reported on the other crops such as cucumber and tomato. 4. Conclusion: The outcomes of the current study, which investigated variations in vegetative growth, yield, and quantitative and qualitative traits of melon cultivars under different nutrient solutions, indicated that the genetic traits of the cultivar, vegetative growth, and quality attributes of melon fruit were influenced in the presence of cocopeat and perlite media. The findings suggest that prior to selecting a melon cultivar for greenhouse cultivation, it is crucial to assess the impact of various nutrients on both vegetative growth and the qualitative characteristics of the fruits. The availability of multiple sequenced genomes from a single species made it possible to explore intra- and inter-specific genomic comparisons at higher resolution and build clade-specific pan-genomes of several crops. The pan-genomes of crops constructed from various cultivars, accessions, landraces, and wild ancestral species represent a compendium of genes and structural variations and allow researchers to search for the novel genes and alleles that were inadvertently lost in domesticated crops during the historical process of crop domestication or in the process of extensive plant breeding. Fortunately, many valuable genes and alleles associated with desirable traits like disease resistance, abiotic stress tolerance, plant architecture, and nutrition qualities exist in landraces, ancestral species, and crop wild relatives. The novel genes from the wild ancestors and landraces can be introduced back to high-yielding varieties of modern crops by implementing classical plant breeding, genomic selection, and transgenic/gene editing approaches. Thus, pan-genomic represents a great leap in plant research and offers new avenues for targeted breeding to mitigate the impact of global climate change. Here, we summarize the tools used for pan-genome assembly and annotations, web-portals hosting plant pan-genomes, etc. Furthermore, we highlight a few discoveries made in crops using the pan-genomic approach and future potential of this emerging field of study. Identifying cultivars of leguminous crops exhibiting drought resistance has become crucial in addressing water scarcity issues. This investigative study aimed to select soybean and cowpea cultivars with enhanced potential to grow under water restriction during the vegetative stage. Two parallel trials were conducted using seven soybean (AS3810IPRO, M8644IPRO, TMG1180RR, NS 8338IPRO, BMX81I81IPRO, M8808IPRO, and BÔNUS8579IPRO) and cowpea cultivars (Aracê, Novaera, Pajeú, Pitiúba, Tumucumaque, TVU, and Xique-xique) under four water levels (75, 60, 45, and 30% field capacity—FC) over 21 days. Growth, water content, membrane damage, photosynthetic pigments, organic compounds, and proline levels were analyzed. Drought stress significantly impacted the growth of both crops, particularly at 45 and 30% FC for soybean and 60 and 45% FC for cowpea plants. The BÔNUS8579IPRO and TMG1180RR soybean cultivars demonstrated the highest performance under drought, a response attributed to increased amino acids and proline contents, which likely help to mitigate membrane damage. For cowpea, the superior performance of the drought-stressed Xique-xique cultivar was associated with the maintenance of water content and elevated photosynthetic pigments, which contributed to the preservation of the photosynthetic efficiency and carbohydrate levels. Persian buttercup (Ranunculus asiaticus L.) and poppy anemone (Anemone coronaria L.) are ornamental, outcrossing, perennial species belonging to the Ranunculaceae family, characterized by large and highly repetitive genomes. We applied K-seq protocol in both species to generate high-throughput sequencing data and produce a large number of genetic polymorphisms. The technique entails the application of Klenow polymerase-based PCR using short primers designed by analyzing k-mer sets in the genome sequence. To date the genome sequence of both species has not been released, thus we designed primer sets based on the reference the genome sequence of the related species Aquilegia oxysepala var. kansuensis (Brühl). A whole of 11,542 SNPs were selected for assessing genetic diversity of eighteen commercial varieties of R. asiaticus, while 1,752 SNPs for assessing genetic diversity in six cultivars of A. coronaria. UPGMA dendrograms were constructed and in R. asiaticus integrated in with PCA analysis. This study reports the first molecular fingerprinting within Persian buttercup, while the results obtained in poppy anemone were compared with a previously published SSR-based fingerprinting, proving K-seq to be an efficient protocol for the genotyping of complex genetic backgrounds. The autotetraploid potato (Solanum tuberosum L.) is an important crop in China, and it is widely cultivated from Northeast China to South China. Thousands of varieties are bred by breeding institutions or companies, and distinguishing the different varieties based on morphological characteristics is difficult. Using DNA fingerprints is an efficient method to identify varieties that plays an increasingly important role in germplasm identification and property rights protection. In this study, the genetic diversity and population structure of 135 autotetraploid potatoes were evaluated using specific-locus amplified fragment sequencing (SLAF-seq) methods. A total of 3,397,137 high-quality single-nucleotide polymorphisms (SNPs), which were distributed across 12 chromosomes, were obtained. Principal component analysis (PCA), neighbour-joining genetic trees, and model-based structure analysis showed that these autotetraploid potato subpopulations, classified by their SNPs, were not consistent with their geographical origins. On the basis of the obtained 3,397,137 SNPs, 160 perfect SNPs were selected, and 71 SNPs were successfully converted to penta-primer amplification refractory mutation (PARMS-SNP) markers. Additionally, 190 autotetraploid potato varieties were analysed using these 71 PARMS-SNP markers. The PCA results show that the accessions were not completely classified on the basis of their geographical origins. The SNP DNA fingerprints of the 190 autotetraploid potato varieties were also constructed. The SNP fingerprint results show that both synonyms and homonyms were present amongst the 190 autotetraploid potatoes. Above all, these novel SNP markers can lay a good foundation for the analysis of potato genetic diversity, DUS (distinctness, uniformity, and stability) testing, and plant variety protection. The consumer demand for an enhanced diet intake of antioxidants and bioactive compounds is continuously rising. This work aims to evaluate the fruit nutritional composition and antioxidant activity of five tomato germplasm varieties, alongside a commercial F1 hybrid. Three varieties bear small-fruit (14–40 g), while two varieties and the commercial cultivar yield large fruit (150–300 g). Genotypes under study were cultivated in a greenhouse under the same environmental conditions. Fat, protein, carbohydrate, total phenol, total flavonoid, lycopene, and ascorbic acid contents were assessed at two fruit maturity stages (breaker, red ripe). For both hydrophilic and lipophilic fractions, antioxidant behavior was also evaluated by employing DPPH and FRAP assays. Small-fruit varieties generally possess higher fat and ascorbic acid content, as well as hydrophilic FRAP values as compared to large-fruit ones. In all varieties, lycopene content and lipophilic fraction radical scavenging capacity was considerably higher at red ripe stage. At red ripe stage, all germplasm varieties were clearly and consistently superior in terms of antioxidant activity at the lipophilic fraction owing to enhanced lycopene content. Phytophthora infestans, the causal agent of late blight (LB) in tomato (Solanum lycopersicum L.), is a devastating disease and a serious concern for plant productivity. The presence of susceptibility (S) genes in plants facilitates pathogen proliferation; thus, disabling these genes may help provide a broad-spectrum and durable type of tolerance/resistance. Previous studies on Arabidopsis and tomato have highlighted that knock-out mutants of the PMR4 susceptibility gene are tolerant to powdery mildew. Moreover, PMR4 knock-down in potato has been shown to confer tolerance to LB. To verify the same effect in tomato in the present study, a CRISPR–Cas9 vector containing four single guide RNAs (sgRNAs: sgRNA1, sgRNA6, sgRNA7, and sgRNA8), targeting as many SlPMR4 regions, was introduced via Agrobacterium-tumefaciens-mediated transformation into two widely grown Italian tomato cultivars: 'San Marzano' (SM) and 'Oxheart' (OX). Thirty-five plants (twenty-six SM and nine OX) were selected and screened to identify the CRISPR/Cas9-induced mutations. The different sgRNAs caused mutation frequencies ranging from 22.1 to 100% and alternatively precise insertions (sgRNA6) or deletions (sgRNA7, sgRNA1, and sgRNA8). Notably, sgRNA7 induced in seven SM genotypes a −7 bp deletion in the homozygous status, whereas sgRNA8 led to the production of fifteen SM genotypes with a biallelic mutation (−7 bp and −2 bp). Selected edited lines were inoculated with P. infestans, and four of them, fully knocked out at the PMR4 locus, showed reduced disease symptoms (reduction in susceptibility from 55 to 80%) compared to control plants. The four SM lines were sequenced using Illumina whole-genome sequencing for deeper characterization without exhibiting any evidence of mutations in the candidate off-target regions. Our results showed, for the first time, a reduced susceptibility to Phytophtora infestans in pmr4 tomato mutants confirming the role of KO PMR4 in providing broad-spectrum protection against pathogens. Availability of standardized morphological and molecular characterization data is essential for the efficient development of breeding programmes in emerging crops. Pepino ( Solanum muricatum) is an increasingly important vegetatively propagated vegetable crop for which concurrent data on morphological descriptors and molecular markers are not available. We evaluated 58 morphological traits, using a collection of 14 accessions of pepinos (including local Andean varieties and modern cultivars) and 8 of wild relatives, using the IPGRI and COMAV descriptors lists coupled with 20 EST-SSRs from tomato. High morphological diversity was found in both cultivated and wild accessions; all morphological traits except three were variable. Cultivated pepino and wild relatives were significantly different for 26 traits. Also, local varieties and modern cultivars of pepino were different from each other for 13 morphological traits and were clearly separated in a principal components analysis. Fourteen of the 20 tomato EST-SSRs were polymorphic, with an average number of alleles per locus of 4.07 and a polymorphic information content value of 0.4132. This revealed a high degree of transferability from tomato to pepino and wide molecular diversity in the collection. Cultivated materials manifest high levels of observed heterozygosity, suggesting that it is related to heterosis for yield associated with heterozygosis. SSR data clearly differentiated cultivated and wild materials. Furthermore, for pepinos, the modern varieties were genetically much less diverse than the traditional local varieties. However, both groups of cultivated material expressed a low degree of genetic differentiation. A strong correlation (r = 0.673) between morphological and molecular distances was found. Pollen grains, the male gametophytes for reproduction in higher plants, are vulnerable to various stresses that lead to loss of viability and eventually crop yield. A conventional method for assessing pollen viability is manual counting after staining, which is laborious and hinders high-throughput screening. We developed an automatic detection tool (PollenDetect) to distinguish viable and nonviable pollen based on the YOLOv5 neural network, which is adjusted to adapt to the small target detection task. Compared with manual work, PollenDetect significantly reduced detection time (from approximately 3 min to 1 s for each image). Meanwhile, PollenDetect can maintain high detection accuracy. When PollenDetect was tested on cotton pollen viability, 99% accuracy was achieved. Furthermore, the results obtained using PollenDetect show that high temperature weakened cotton pollen viability, which is highly similar to the pollen viability results obtained using 2,3,5-triphenyltetrazolium formazan quantification. PollenDetect is an open-source software that can be further trained to count different types of pollen for research purposes. Thus, PollenDetect is a rapid and accurate system for recognizing pollen viability status, and is important for screening stress-resistant crop varieties for the identification of pollen viability and stress resistance genes during genetic breeding research. Background. Solanum melongena is a medicinally important vegetable crop that belongs to the family Solanaceae, which is cultivated worldwide. Methodology. In the present study, 22 eggplant varieties from the different ecogeographical regions were evaluated for nine quantitative and twenty-two qualitative morphological characters. A significant divergence was observed in all characters and wide regional variations for plant characteristics, flower, and fruit characteristics. Principle component analysis (PCA) was performed using PAST3 software to determine the relationship among eggplant accessions. Results. The principal component analysis showed that the first two principal component axes explained 97.17% of the total multivariate variation. Cluster analysis using the Unweighted Pair Group Method of Arithmetic Means (UPGMA) grouped the 22 eggplant accessions into two main clusters based on similarities in morphological characteristics. The study showed that the Solanum melongena accessions belonging to Pakistan and other geographical regions of the world possess marked variation in fruit weight, fruit shape, fruit color, leaf spine, number of locules in fruit, plant height, and flower color. Different studies have reported a narrow genetic base for garden asparagus (Asparagus officinalis L.) due to its common origin, a diploid population ('Purple Dutch'). The present study focused on the development of new diploid plant material that may be useful to widen the genetic base of the crop by using a tetraploid landrace 'Morado de Huétor' (A. officinalis × A. maritimus). With this purpose, a diploid pre-breeding population (n = 1000) carrying introgressions of 'Morado de Huétor' has been obtained. This new population derived from crosses under open pollination of a parental collection (n = 77) that was developed in a previous study. The parental collection derived from the first backcrossing using different diploid cultivated plants as a recurrent parent and 'Morado de Huétor' as a donor. The genetic diversity of the pre-breeding population was assessed using a set of EST-SSR markers (AG7, AG8, TC1, TC3, TC7, TC9) in a collection of plants (n = 57), which was randomly sampled in the pre-breeding population. The results were compared to previous data obtained from the parental collection, a set of current diploid asparagus cultivars and the landrace 'Morado de Huétor'. The average of PICm (Polymorphic Information Content) values obtained in the pre-breeding population (0.75) resulted higher than the value obtained in the diploid cultivars (0.63) but lower than in 'Morado de Huétor' (0.83). Twenty-two alleles (52.4%) detected in the new diploid population were specific from 'Morado de Huétor'. Principal Coordinate Analyses (PCoA) revealed that the new population had a genetic diversity distribution different from the current cultivars. This new population was also evaluated for different morpho-agronomic traits (earliness, stalk number, branching height and stalk thickness) for two years. Significant differences among plants (p < 0.001) were found for these five traits and, therefore, a genotype variation is suggested. As a result, 71 plants were selected to develop a breeding base population. The genetic variability of those selected plants was also analyzed and similar genetic variability to the pre-breeding population was obtained. Leafy vegetables are associated with Listeriosis outbreaks due to contamination with Listeria monocytogenes. To date, contradictory findings were reported on spinach, rocket, and kale, where some studies reported growth of L. monocytogenes, while others did not. Thus, the current study investigated the reason for conflicting findings by producing leafy vegetables, where cultivation factors were known for growth potential studies. Of all polytunnel produce, kale Nero di Toscana demonstrated the highest growth potential (2.56 log cfu g−1), followed by spinach F1 Cello (1.84 log cfu g−1), rocket Buzz (1.41 log cfu g−1), spinach F1 Trumpet (1.37 log cfu g−1), and finally rocket Esmee (1.23 log cfu g−1). Thus, plant species and variety influenced L. monocytogenes growth potentials. Moreover, significantly lower growth potentials of 0.3 log cfu g−1 were identified when rocket Buzz was cultivated in open fields (1.11 log cfu g−1) instead of a polytunnel. The opposite effect was observed for spinach F1 Trumpet, where growth potentials increased significantly by 0.84 log cfu g−1 when cultivated in open fields (2.21 log cfu g−1). Furthermore, a significant seasonality effect between batches was found (p < 0.05). This study revealed that spinach and rocket cultivation conditions are at least co-factors in the reporting of differing growth potentials of L. monocytogenes across literature and should be considered when conducting future growth potential studies. Eggplant is a widely consumed vegetable, with significant nutritional value and high antioxidant content, mainly due to its phenolic constituents. Our goal was to determine the levels of carbohydrates, proteins, total phenolics, anthocyanins, flavonoids, chlorogenic acid, and the antioxidant capacity in thirteen eggplant cultivars cultivated in Greece and to identify sequence polymorphisms in key regulating genes of the phenylpropanoid pathway (C4H, HCT, HQT, C3H, F3H, ANS, MYB1), which might relate to the phytochemical content of those cultivars. The carbohydrates' content differs among and within cultivars, while the rest of the phytochemicals differ only among cultivars. The cultivars 'EMI' and 'Lagkada' scored higher than the rest in phenolics, anthocyanins, ascorbic acid, caffeoylquinic acid, and antioxidant capacity. Moreover, significant correlations were observed between various ingredients and the antioxidant capacity (FRAP and DPPH). Sequence analysis revealed several SNPs in C4H, HQT, F3H, ANS, and MYB1 among the cultivars studied. According to chi-square and logistic regression analyses, the missense mutation C4H4-108 correlates significantly with flavonoids, anthocyanins, and proteins; the synonymous mutation HQT-105 correlates with anthocyanins and ascorbic acid; the missense mutation HQT-438 correlates with flavonoids and chlorogenic acid, while the missense mutation ANS1-65 correlates with anthocyanins and sugars. These polymorphisms can be potentially utilized as molecular markers in eggplant breeding, while our data also contribute to the study of eggplant's secondary metabolism and antioxidant properties. Drought stress, which is the most important abiotic stress factor affecting arable land in the world, causes serious crop losses. These crop losses reach up to 70% in some agricultural plants. Understanding the complex drought stress response is very important to develop a strategy against this form of stress. Although some progress has been achieved with the previous studies, the desired targets have not been reached up to now. Therefore, using resistant varieties in environmental conditions has become a widely used strategy in combating drought stress today. In this study, a total of 23 cultivars (16-landraces and 7 modern wheat cultivars) were used. The aim of this study was to reveal the drought tolerance degrees of 16 landraces by comparing them to 7 modern wheat cultivars. For this purpose, 23 cultivars were exposed to drought stress for seven days by withholding watering. After that, stem length, MDA and proline content of cultivars were determined and compared. According to our results, MDA and proline contents of sensitive modern cultivars were found to be high, while tolerant cultivars were found to be low. It has also been determined that some of the landraces exhibit a similar profile to the cultivars known to be tolerant. Among these cultivars, especially 88, 90 and 108 cultivars have low MDA and proline content under stress, which may indicate that these cultivars are potentially drought tolerant. The cucumber (Cucumis sativus L.) is one of the most important vegetables in Bangladesh as well as across the globe. However, many of the important cucumber landraces have disappeared in Bangladesh due to climate change, particularly erratic rainfall, extreme temperature, salinity, and drought. Therefore, to protect against the extinction of the cucumber landraces, we collected 103 landraces in different geographical regions of Bangladesh, including drought and saline-prone areas, and studied their divergence for the future breeding programme for the development of cultivars suitable for the climate-changing situations. Data on morphological features, yield, and its components, which include 17 qualitative and quantitative traits, were recorded during the observation. Among the cucumber landraces, the Shannon–Weaver diversity index analysis revealed the presence of genetic diversity in these landraces. The biggest diversity appeared in the fruit-related characteristics, i.e., stem end fruit shape, bottom end fruit shape, fruit shape, and fruit skin colour at the table and harvest maturity. The descriptive statistics and analysis of variance expressed a wide range of variability for quantitative traits. A broad phenotypic variation was also observed for traits such as yield plant−1 [CV (%) 31.88, ranges 0.96 to 3.11 kg] and fruits plant−1 (CV (%), 28.71, ranges, 2.58 to 9.75). High heritability (broad sense) coupled with a high genetic gain was observed for yield and yield-contributing characteristics, indicating that these characteristics are controlled by additive gene effects, and they are more reliable for effective selection. The phenotypic correlation studies showed that fruit yield plant−1 exhibited a positive and significant correlation with fruits plant−1, fruit length, fruit weight, fruit width, branches plant−1, and plant height. All landraces were grouped into six clusters, and the maximum number of landraces were accommodated in cluster VI (30), followed by cluster V (22), cluster III (22), cluster IV (14), cluster I (13), and cluster II (2). Comparing cluster means with studied traits revealed that cluster III with landraces AC-14, AC-97, AC-471, AC-451, and RAI-209 were more divergent for improving average fruit weight, fruit length, and fruit width. On the other hand, cluster IV with landraces AC-201, TT-161, RAI- 217, RAI-215, and TRMR-103 were more divergent for improving average vine length, internode length, and the number of primary branches plant−1, the number of fruits plant−1, and yield plant−1. According to the MGIDI index, AC-14 (G1), AC-201 (G7), AC-471 (G24), AC-97 (G30), RAI-215 (G68) and TT-161 (G 94) may be considered to be the best parents based on their qualitative and quantitative characteristics for the future breeding programme. Moreover, crossing between the landraces, which were collected from saline and drought areas, in clusters I, V, and VI with those in other clusters could produce suitable cucumber varieties for the climatic changing situation. Image analysis can provide reproducible, objective and accurate results using a non-destructive, inexpensive procedure. The aim of this study was to evaluate the usefulness of the textures calculated from images of the flesh and skin of tomato fruit for cultivar discrimination. The color images of six tomato cultivars were acquired for whole peeled tomatoes and whole tomatoes with skin using a digital camera. The images were converted to color channels R, G, B, L, a, b, X, Y, and Z. The discriminative models were built based on a set of selected textures from all color channels, each color channel, and color space, separately. For images of tomato flesh and skin, the highest average accuracies equal to 92.67% and 94.33%, respectively, were obtained for models combining textures selected from all channels. For color spaces, the correctness reached 92% and 94% for tomato flesh and skin images, respectively. In the case of color channels, cultivar discrimination reached 86.33% for channel a for tomato flesh and 88.67% for channel b for tomato skin. Recent advances in next generation sequencing (NGS) technologies have led the surge of genomic resources for the improvement legume crops. Advances in high throughput genotyping (HTG) and high throughput phenotyping (HTP) enable legume breeders to improve legume crops more precisely and efficiently. Now, the legume breeder can reshuffle the natural gene combinations of their choice to enhance the genetic potential of crops. These genomic resources are efficiently deployed through molecular breeding approaches for genetic augmentation of important legume crops, such as chickpea, cowpea, pigeonpea, groundnut, common bean, lentil, pea, as well as other underutilized legume crops. In the future, advances in NGS, HTG, and HTP technologies will help in the identification and assembly of superior haplotypes to tailor the legume crop varieties through haplotype-based breeding. Salinity stress is a major constraint to sustainable crop production due to its adverse impact on crop growth, physiology, and productivity. As potato is the fourth most important staple food crop, enhancing its productivity is necessary to ensure food security for the ever-increasing population. Identification and cultivation of salt-tolerant potato genotypes are imperative mitigating strategies to cope with stress conditions. For this purpose, fifty-three varieties of potato were screened under control and salt stress conditions for growth and yield-related traits during 2020. Salt stress caused a mean reduction of 14.49%, 8.88%, and 38.75% in plant height, stem numbers, and tuber yield, respectively in comparison to control. Based on percent yield reduction, the genotypes were classified as salt-tolerant (seven genotypes), moderately tolerant (thirty-seven genotypes), and salt-sensitive genotypes (nine genotypes). Seven salt-tolerant and nine salt-sensitive genotypes were further evaluated to study their responses to salinity on targeted physiological, biochemical, and ionic traits during 2021. Salt stress significantly reduced the relative water content (RWC), membrane stability index (MSI), photosynthesis rate (Pn), transpiration rate (E), stomatal conductance, and K+/Na+ ratio in all the sixteen genotypes; however, this reduction was more pronounced in salt-sensitive genotypes compared to salt-tolerant ones. The better performance of salt-tolerant genotypes under salt stress was due to the strong antioxidant defense system as evidenced by greater activity of super oxide dismutase (SOD), peroxidase (POX), catalase (CAT), and ascorbate peroxidase (APX) and better osmotic adjustment (accumulation of proline). The stepwise regression approach identified plant height, stem numbers, relative water content, proline content, H2O2, POX, tuber K+/Na+, and membrane stability index as predominant traits for tuber yield, suggesting their significant role in alleviating salt stress. The identified salt-tolerant genotypes could be used in hybridization programs for the development of new high-yielding and salt-tolerant breeding lines. The Canary Islands have an enormous richness of crops and varieties, many of them traditional or local, selected for decades by farmers based on the most desirable characteristics. Pear trees were introduced to the Canary Islands presumably in the first years after their Conquest in the 15th century, reaching a high degree of diversification. In this study, to determine the genetic identity of the genus Pyrus in the Canary Islands for conservation purposes, 266 pear accessions from the islands of Tenerife, La Palma and Gran Canaria were characterized with 18 SSRs, in addition to 190 genotypes from Galicia, Asturias, wild and commercial varieties as references to detect possible synonyms, genetic relationships and the possible genetic structure. We identified 310 unique genotypes, both diploid and putative triploid, 120 of them present only in the Canary Islands (39%, with 50% clonality). The population structure of the genotypes was analyzed by STRUCTURE 2.3.4 software (Pritchard Lab, Stanford University, Stanford, CA, USA). The dendrogram, by using the Jaccard coefficient and principal component analysis (PCoA), separated the analyzed genotypes into stable groups. One of these groups was formed only by Canarian varieties present at lower altitudes, showing adaptation to low chilling requirements with a significant positive correlation (0.432, p < 0.01). This first study of the pear germplasm in the Canary Islands reflects the importance of the group of local cultivars and their need for conservation given they are adapted to their peculiar climatic conditions and have a low number of chill units. New pepino (Solanum muricatum Aiton) cultivars with improved flavor were bred in Israel in recent years. Various parental lines with attractive fruit appearance and promising horticultural traits, but not necessarily with very high soluble solid concentration (SSC) levels or with good flavor, were used for crosses. Selection was performed in the segregating populations in search of new genotypes with good horticultural attributes combined with improved flavor. Individual clones with improved flavor were selected among the progeny populations, indicating that flavor is mediated through specific gene combinations.‘Pepo’, the first locally bred cultivar, has been grown in modest quantities during the last seven years, mainly for the local market and occasionally for export to Europe. The crop is grown in greenhouses from September through July, yielding up to five tons of fruits per 1000 m2. ‘Pepo’ has been well received both locally and abroad. The fruit has a pleasant mild flavor, and due to its low calorie content was approved as suitable for diabetic people. It can be used as a dessert as well as in green and fruit salads. Pickled green immature ‘Pepo’ fruits are very flavorful. Additional new locally-bred cultivars with improved sweetness or with a special unique aroma are now being introduced.Irrigation with saline water (3.5 dS/m) was able to enhance the SSC and flavor of pepino but could not amend occasional unpleasant off-flavors. The total phenolic content, anthocyanins, phenolic acids, antioxidant capacity and α-amylase inhibitory activity of black (Aydin Siyahi), purple (Kadife Kemer) and white (Trabzon Kadife) eggplants grown in Turkey were subjected to a comparative investigation. The black cultivar exhibited the highest total phenolic (17,193 and 6552 mg gallic acid equivalent/kg fw), flavonoid (3019 and 1160 quercetin equivalent/kg fw) and anthocyanin (1686 and 6167 g delphinidin-3-O-glucoside equivalent/kg fw) contents in crude extracts of the peel and pulp. The majority of the caffeic acid was identified in the ester (2830 mg/kg fw) and ester-bound (2594 mg/kg fw) forms in the peel of 'Kadife Kemer' and in the glycoside form (611.9 mg/kg fw) in 'Aydin Siyahi', as well as in the pulp of these two eggplants. 'Kadife Kemer' (purple eggplant) contained the majority of the chlorogenic acid in free form (27.55 mg/kg fw), compared to 'Aydin Siyahi' in the ester (7.82 mg/kg fw), glycoside (294.1 mg/kg dw) and ester-bound (2.41 mg/kg fw) forms. The eggplant cultivars (peel and pulp, mg/kg fw) exhibited a relatively high delphinidin-3-O-rutinoside concentration in the peel of 'Aydin Siyahi' (avg. 1162), followed by 'Kadife Kemer' (avg. 336.6), and 'Trabzon Kadife' (avg. 215.1). The crude phenolic extracts of the eggplants exhibited the highest antioxidant capacity values (peel and pulp, µmoL Trolox equivalent/kg fw) of 2,2-diphenyl-1-picrylhydrazyl (DPPH, 8156 and 2335) and oxygen radical absorbance capacity (ORAC, 37,887 and 17,648). The overall results indicate that black and purple eggplants are the cultivars with greater potential benefits in terms of their phenolics and antioxidant values than the white eggplant. Key message: Comparative analysis of herbivory responsive miRNAs between pod borer susceptible C. cajan and its resistant Crop Wild Relative (CWR) C. scarabaeoides revealed miRNA-based regulation of defense genes and plant–insect interactions. Gram pod borer (Helicoverpa armigera) is one of most devastating pests of pigeon pea (Cajanus cajan) worldwide, responsible for huge losses in crop productivity. The lack of genes conferring resistance to pod borer in pigeon pea has proven to be a bottleneck for its improvement. One of its CWR, C. scarabaeoides has demonstrated resistance to this pest and can be exploited for developing pest resistant crop varieties. Differences in expression patterns of herbivory responsive microRNAs in the susceptible C. cajan and resistant C. scarabaeoides after different time duration of pod borer infestation (2 h, 8 h and 18 h) were identified, characterized and functionally validated to understand their role in insect defense response. A total of 462 conserved and 449 novel miRNAs and 273 conserved and 185 novel miRNAs, were identified in C. cajan and C. scarabaeoides, respectively. Among the identified miRNAs, 65, 68 and 65 miRNAs were found to be differentially expressing between the C. scarabaeoides and C. cajan libraries 2 h, 8 h and 18 h post infestation, respectively. These miRNAs were found to target genes involved in a number of pathways contributing to defense and acquired resistance in C. scarabaeoides against pod borer, indicating miRNA-based regulation of defense pathways. Expression patterns of eight of these miRNAs were validated by qRT-PCR. This study provides novel insights into the miRNA-mediated plant–insect interactions and the mechanisms of regulatory pathways involved in insect defense. Crop wild relatives represent a valuable resource for the breeding of new crop varieties suitable for sustainable productivity in conditions of climate change. The aim of the present study was to assess salt tolerance of several wild accessions of T. fragiferum from habitats with different salinity levels in controlled conditions. Decrease of plant biomass and changes in partitioning between different organs was a characteristic response of plants with increasing substrate salinity, but these responses were genotype-specific. In several accessions, salinity stimulated reproductive development. The major differences in salinity responses between various T. fragiferum genotypes were at the level of dry biomass accumulation as well as water accumulation in plant tissues, resulting in relatively more similar effect on fresh mass. Na+ and Cl− accumulation capacity were organ-specific, with leaf petioles accumulating more, followed by leaf blades and stolons. Responses of mineral nutrition clearly were both genotype- and organ-specific, but several elements showed a relatively general pattern, such as increase in Zn concentration in all plant parts, and decrease in Ca and Mg concentration. Alterations in mineralome possibly reflect a reprogramming of the metabolism to adapt to changes in growth, morphology and ion accumulation resulting from effect of NaCl. Marigolds (Tagetes spp.) are multifunctional flowering plants belonging to the Asteraceae family, well-known and widespread for their ornamental value and many other uses. In this study, morphological differences and genetic relationships among 21 cultivars of three species of marigold (Tagetes patula, T. erecta and T. tenuifolia) were analysed. Results have revealed obvious differences among genotypes, starting from the morphological characteristics of the seeds and their capacity to germinate to adult plant morphological characteristics, both between cultivars and species. The genotypic differences were manifested in considerable variation in the development of phenological stages and the main morphological traits of plants and flowers. PCA and hierarchical clustering analyses of morphological traits revealed a homogeneous grouping of cultivars within each species, except for Orion, belonging to T. patula, which was closer to T. erecta cultivars. A subset of 13 cultivars from the three species was subjected to SSR analysis, revealing considerable genetic diversity and good separation between T. patula on the one side and T. erecta and T. tenuifolia on the other. The observed heterozygosity was much lower than the expected heterozygosity, revealing a high degree of fixation. The results reveal that the three species evaluated have considerable morphological and genetic diversity, which has important implications for assessing genetic diversity, conserving germplasm and selecting parents for new breeding works in marigolds. Understanding the evolutionary history of crops, including identifying wild relatives, helps to provide insight for conservation and crop breeding efforts. Cultivated Brassica oleracea has intrigued researchers for centuries due to its wide diversity in forms, which include cabbage, broccoli, cauliflower, kale, kohlrabi, and Brussels sprouts. Yet, the evolutionary history of this species remains understudied. With such different vegetables produced from a single species, B. oleracea is a model organism for understanding the power of artificial selection. Persistent challenges in the study of B. oleracea include conflicting hypotheses regarding domestication and the identity of the closest living wild relative. Using newly generated RNA-seq data for a diversity panel of 224 accessions, which represents 14 different B. oleracea crop types and nine potential wild progenitor species, we integrate phylogenetic and population genetic techniques with ecological niche modeling, archaeological, and literary evidence to examine relationships among cultivars and wild relatives to clarify the origin of this horticulturally important species. Our analyses point to the Aegean endemic B. cretica as the closest living relative of cultivated B. oleracea , supporting an origin of cultivation in the Eastern Mediterranean region. Additionally, we identify several feral lineages, suggesting that cultivated plants of this species can revert to a wild-like state with relative ease. By expanding our understanding of the evolutionary history in B. oleracea , these results contribute to a growing body of knowledge on crop domestication that will facilitate continued breeding efforts including adaptation to changing environmental conditions.
