The product also showed potent inhibitory activities against human laryngeal carcinoma epithelial cell line (Hep-2), human breast cancer cell line (MDA-MB-231) and hepatocellular liver carcinoma cell line (HepG 2), which was better than the positive control drug 5-fluorouracil (5-FU), implying that newly synthesized compound may be a potential candidate for the development of anti-cancer drugs. Abstract A new strategy to construct polyoxovanadate hybrids incorporating amino acid esters in mild conditions was presented in this paper. These new hybrids were not only structurally determined by Single Crystal X-Ray diffraction, but also exhibited higher antitumor activities against laryngeal carcinoma, rhabdomyoma, and breast adenocarcinoma tumor cells compared with the traditional commercial medicine 5-fluorouracil. These results would provide a promising lead scaffold for further design and synthesis of potential anticancer agents. • Three original hexavanadate hybrids with p -halogenated benzoyl ligands had been synthesized. • The packing manners of the three compounds are disparate depending on the terminal halogen elements. • The influence of different condensing agents on the yield is thoroughly analyzed based on the reaction mechanism. Three original hexavanadate-based materials (DIEA) 2 [V 6 O 13 {(OCH 2) 3 CNHCOC 6 H 4 X− p)} 2 ] (X=Cl, Br, I) have been synthesized via the amidation of H 2 [V 6 O 13 {(OCH 2) 3 CNH 2 } 2 ] and p -halogenated benzoic acid. The components and structures of the products were determined by FT-IR, 1H NMR, ESI-MS, XPS and single-crystal X-ray diffraction. The molecular structures of three compounds are similar: a Lindqvist-type cluster {V 6 O 19 } covalently links to two halogenated benzoyl trialkoxyl ligands. However, they exhibit diverse packing manners because of their difference in hydrogen-bonding interactions. The anions [V 6 O 13 {(OCH 2) 3 CNHCOC 6 H 4 Cl− p)} 2 ]2- are arranged in a 1D chain structure by intermolecular hydrogen-bonding interactions, while the other two compounds show a 2D "sandwich biscuit" layered packing structure without hydrogen bonds between the anions. Moreover, when choosing a condensing agent to increase the yield, PyBOP has an overwhelming advantage over common EDCI/HOBt. This article also discusses the reaction mechanism in detail, and provides experimental guidance for the rational design of POM-based organic-inorganic hybrids. Three novel organic-inorganic hexavanadate-based hybrids with p -halogenated benzoyl ligands had been synthesized by amidation reaction. The components and structures of the products were determined by a series of characterization methods such as FT-IR, 1H NMR, ESI-MS, XPS and single-crystal X-ray diffraction. The three compounds show disparate packing manners depending on the terminal halogen elements. Moreover, the type of the condensing agent significantly affects the yield of the products. We found that PyBOP has an overwhelming advantage over common EDCI/HOBt. This result provides meaningful experimental guidance for the rational design of organic-inorganic POM hybrids.
