The article examines the viability assay of coralline algae using triohenyltetrazolium chloride. Several techniques for determining the viability assay were hypothesized however none of these was proven reliable. These include counting numbers of stained cells under the microscope and analysis on the color of the algae. In its aimed to determine the viability and proliferation, the authors have optimized conditions of the tetrazolium chloride (TTC) assay, removing phycoerythrin or byproducts, for use in the viability determination of coralline algae. Optimization of conditions for the TTC viability assay was required with respect to pH of the incubation buffer, time of incubation, concentration of TTC, and temperature of incubation.
