Successful pregnancy in the female rat depends upon two sets of physiological events: (1) transport of gametes (sperm and egg) through the reproductive tract so that fertilization can be effected and (2) establishment of an appropriate hormonal environment (progestational state) so that the fertilized egg can implant in the uterus and be maintained during subsequent gestation. This study highlights the independence of the mechanisms controlling the gametic and hormonal aspects of pregnancy by temporally separating the introduction of sperm into the female from the stimulation that triggers the progestational hormonal response. The progestational state was initiated by electrical stimulation of the cervix, and sperm was introduced directly into the uterus by artificial insemination. Although these two events were separated by up to 3 days, pregnancy could ensue. Cervical stimulation, normally a consequence of male intromission behavior, establishes a condition in the central nervous system, a "memory" that signals the probable induction of pregnancy. Without this "memory," animals with a short estrous cycle would continue to cycle after mating, thereby producing a hormonal environment incompatible with implantation. The "memory" is manifested by daily surges of prolactin irrespective of fertilization. This is the first physiological demonstration that a "memory" of cervical stimulation can be called upon to support a viable pregnancy.

The number of pre-ejaculatory intromissions a female rat receives affects the probability of becoming progestational and transporting sperm into the uterus. We now report that the number of intromissions a male rat delivers affects his ejaculatory performance. Males and females were given different copulatory experience by switching females during mating. When either the male or female had few intromissions, few sperm were recovered and the copulatory plugs fit less well. Intromission-dependent sperm recovery was also observed in a review of 94 copulatory tests drawn from archival data. Several possible mechanisms are discussed, including poor placement of the copulatory plug and reduced sperm output. The potential significance for reproductive success in social mating situations is discussed.

Two temporal variables have been implicated in the timing of postpartum heat in Norway rats: time of day and time since parturition. The nature of the interaction between these variables is not clearly understood. We extend previous observations on this problem to freely behaving animals continuously videotaped during parturition and postpartum mating in a seminatural habitat. The timing of postpartum mating is best described by a model in which an interval timer is coupled to a circadian gating mechanism. Interactions between circadian and interval timing mechanisms may be widespread in the neuroendocrine control of behavior.

Vaginocervical stimulation affects progesterone secretion, sperm transport, sexual receptivity, locomotion, and perception of pain in female rats. In this experiment, vaginocervical stimulation produced statistically significant increases in the metabolic uptake of carbon-14-labeled 2-deoxy-D-glucose in the following brain areas (ordered by magnitude of uptake): medial preoptic, mesencephalic reticular formation, bed nucleus of stria terminalis, dorsal raphe, and globus pallidus. The results provide information about the concurrent processing of sensory stimulation by several neural areas and indicate that the medial preoptic area is a receiving area for copulatory stimulation.

Twenty female rats were implanted with myometrial electrodes and intrauterine balloons to study the effects of mating on uterine contractile activity. Mating produced both immediate and delayed effects. Uterine activity typically doubled during the period of active mating (the immediate effect), then briefly returned to the premating activity level after ejaculation. Beginning 5 min after the ejaculation, the delayed effect of mating was observed: contractile activity increased when the baseline activity had been low, but decreased when baseline was fast. This delayed effect lasted for 30 min to several hours. Vaginocervical stimulation seemed to play a critical role in these responses: artificial vaginocervical stimulation mimicked the response while vigorous mounting by males that were prevented from intromitting did not evoke these responses. Vaginocervical stimulation, whether mating-derived or experimenter-delivered, produced immediate and delayed effects on uterine contraction rate.

If female rats received genital stimulation soon enough after their male partners had ejaculated, sperm transport and subsequent pregnancy were inhibited. Manual stimulation by the experimenter or five intromissions by a male rat were sufficient stimuli to reduce the number of sperm found in the uterus and to reduce the number of uterine implantation sites.
