Diese Arbeit beschreibt die Entwicklung und Charakterisierung polymerer Implantatmaterialien mit einem photochemisch aktivierbaren Wirkstoffdepot, welches mit dem Zytostatikum 5-Fluoruracil ausgerüstet ist. Das Einsatzgebiet dieser Implantatmaterialien (bzw. der Implantate, die aus diesen hergestellt werden) wird hauptsächlich in der Ophthalmologie als Ersatz für die natürliche Augenlinse im Zuge einer Kataraktoperation gesehen. Das Wirkstoffdepot soll eine alternative, medikamentöse Behandlung des so genannten Sekundärkatarakts, einer häufigen postoperativen Komplikation, ermöglichen. Es wurden zwei unterschiedliche Polymertypen untersucht, die zur Herstellung moderner Faltlinsen geeignet sind: Hydrophobe und hydrophile Polyacrylate. Die Wirkstoffimmobilisierung basiert in beiden Fällen auf der photochemisch reversiblen [2+2]-Cycloaddition des Wirkstoffs 5-Fluoruracil bzw. einer entsprechenden Wirkstoffvorstufe an polymergebundene Cumaringruppen. Die Cycloreversion, welche im Experiment Ein- und Zwei-Photonen-induziert untersucht wurde, stellt somit die Wirkstofffreisetzung dar., The development and characterization of polymeric implant materials for the photo-induced release of the cytostatic drug 5-fluorouracil is described in this work. The materials (respectively the implants made from these materials) are designed to work as replacement for the natural lens of the human eye during cataract surgery. The incorporated drug depot provides an alternative method for the treatment of secondary cataract, a common post-operative complication. Two different types of polymers were investigated, both suitable for the fabrication of modern foldable intraocular lenses: hydrophobic and hydrophilic polyacrylates. The drug immobilization is based on the photochemical reversible [2+2]-cycloaddition of 5-fluorouracil respectively a 5-fluorouracil pro-drug to polymer bound coumarin groups. The cycloreversion which can be triggered by a single- or two-photon-absorption process is therefore equivalent to the drug release mechanism.
