Several factors can affect lamb body weight (BW) and immune status during the first days of life, including colostrum source and timing of the first colostrum feeding. The aim of this study was to evaluate the effects of colostrum source (goat or sheep) and timing of the first colostrum feeding (2 or 14h after birth) on lamb BW and immune status. In this study, 40 lambs were removed from their dams at birth and randomly assigned into 4 groups of 10 lambs each. Lambs were subsequently fed at 2 or 14h after birth with goat or sheep colostrum. Blood samples and BW recording were performed before feeding. Blood plasma was used to measure the immunoglobulin concentration (IgG and IgM), chitotriosidase activity, and complement system activity (total and alternative pathways). In general, no differences in any of the measured variables were observed among the 4 groups, indicating that neither colostrum source nor timing of the first colostrum feeding had an effect on these variables. These findings may improve management on lamb farms that raise animals under artificial conditions, because our results indicate that it is not necessary to feed colostrum to lambs immediately after birth and that goat colostrum may be used to feed newborn lambs., (Copyright © 2015 American Dairy Science Association. The effect of induction of parturition with a PGF(2)α analog on plasma concentration of prolactin (PRL) and its effects on colostrum concentration of IgG and chitotriosidase (ChT) activity were studied in 16 pregnant Majorera goats. Treated goats, those in which parturition was induced, had greater concentrations of PRL than control goats 24 h before parturition (P < 0.05) and 48 h after parturition (P < 0.05). Control goats had greater concentrations of PRL than treated goats 96 h after parturition (P < 0.05). Plasma concentration of IgG did not differ between groups during the experimental period, but colostrum concentrations of IgG were greater in control goats than in treated goats at parturition (P < 0.05). Plasma ChT activity decreased during the period 72 h before parturition to 24 h after parturition in control and treated goats. Time evolution after partum affected the colostrum ChT activity, being greater at parturition than after parturition in both groups (P < 0.05). In summary, concentration of IgG in colostrum is slightly diminished if parturition is induced. Induction of parturition causes an early increase in PRL, which is most likely responsible for preterm suppression of IgG transport into mammary secretions., (Copyright © 2011 Elsevier Inc. To investigate the use of sodium dodecyl sulfate (SDS) as a biocide on goat colostrum, 2 experiments were performed. In the first, 20 goat colostrum samples were divided into 3 aliquots. A different treatment was performed on each aliquot: pasteurization (56°C, 30 min) or addition of SDS to a final concentration of either 0.1 or 1% (36°C, 10 min). Immunoglobulin G and colony-forming units were evaluated before and after treatment. Both pasteurization and treatment with 1% SDS significantly reduced the colony-forming units in colostrum. Treatment with 0.1% SDS was not effective at reducing the colony-forming units in colostrum. The IgG concentration of pasteurized colostrum was significantly lower than that of untreated colostrum, whereas treatment with 1% SDS did not affect the colostrum IgG concentration. In the second experiment, the effects of SDS colostrum treatment on immune passive transfer were evaluated. Forty goat kids were fed either refrigerated colostrum or colostrum treated with 1% SDS twice daily for 2 d. Blood samples were obtained at birth and every day for 5 d. IgG, IgM, and IgA were measured in blood serum to monitor the passive immune transfer process. Creatinine, glucose, total cholesterol, blood urea nitrogen, bilirubin, and aspartate transaminase were also monitored to evaluate the health of kids. No differences in serum IgG, IgM, IgA, creatinine, glucose, total cholesterol, blood urea nitrogen, bilirubin, or aspartate transaminase levels were observed between groups. Our findings indicate that SDS is an efficient colostrum biocide that, unlike pasteurization, does not affect immune passive transfer or goat kid health., (Copyright © 2011 American Dairy Science Association. The aim of the research was to evaluate the effects of immunoglobulin G (IgG) colostrum concentration on goat kid immune status when the total amount of IgG fed was constant. Majorera goat kids (n = 56) were randomly assigned to 1 of 4 groups, and kids received 4 g of IgG per kg of body weight of atomized colostrum at 4 different IgG concentrations: 20 (AC-20), 40 (AC-40), 60 (AC-60), and 80 (AC-80) mg/mL. Blood samples were obtained on d 0, 1, 2, 3, 4, and 5 postpartum. Immunoglobulin G, IgA, and IgM plasma concentrations, apparent efficiency of absorption of IgG, plasma chitotriosidase activity, plasma complement activity, and plasma proteinogram were measured. Plasma IgG and IgM concentrations were highest on d 1 in AC-80 animals, and IgA plasma concentration was lower in AC-20 than in AC-80. The apparent efficiency of absorption was higher in AC-80 (24.4%) than in the other treatment groups (by an average of 13.8%). Chitotriosidase plasma activity on d 5 (1,488 nmol/mL per hour) was higher than on d 0 and 1 (average of 1,183 nmol/mL per hour). There were no effects of colostrum IgG concentration on complement activity and plasma protein distribution, but gamma-globulin and alpha-globulin were lower on d 0 than on d 1, 2, 3, 4, and 5. Increasing the immunoglobulin concentration in colostrum using atomized colostrum improves the immunoglobulin absorption at the same amount of immunoglobulin fed.

Majorera goat kids (n = 200) were used to evaluate the effects of litter size, birth body weight, sex, and suckling duration on serum IgG concentrations. Kids were assigned to 1 of 3 experimental groups: litter size and sex were equally distributed in each group. In the first group, kids (n = 67) stayed with their dams for 24 h; in the second group, kids (n = 66) stayed with their dams for 48 h; and in the third group, kids (n = 67) stayed with their dams for 120 h. Blood samples were obtained every 24 h for 5 d, and serum IgG concentration was measured using radial immunodiffusion. In litter sizes of 1 to 2 kids, IgG blood serum concentration was significantly higher (18.30 +/- 5.40 mg/mL) than in litters of 3 kids (9.85 +/- 4.23 mg/mL). Kid sex did not affect IgG blood serum concentrations. Suckling duration did not affect kid serum IgG concentrations. In conclusion, kids with low birth body weight (<2.8 kg) or from litters of 3 may need special attention. If newborn goat kids are allowed to suckle colostrum for at least 24 h from their dams, this seems to be sufficient time to ingest enough IgG from colostrum to achieve an adequate serum IgG concentration and passive immune protection to avoid failure of passive immune transfer.

Chitotriosidase (ChT) activity has not been investigated in ruminants, and therefore, we studied this activity in blood and colostrum of 25 pregnant goats and 60 goat kids. Blood samples were taken from pregnant goats at 3, 2, and 1 d prepartum; at partum; and at 1, 2, 3, and 4 d postpartum. Colostrum samples were obtained by machine-milking at partum and 1, 2, 3, and 4 d postpartum. Goat kid blood was collected at birth and every 7 d thereafter until goats kids were 56 d old. The ChT activity ranged from 2,368 to 3,350 nmol/ mL per hour in goat blood serum, and no statistical differences were detected through time. However, activity tended to decrease from 3 d prepartum to 2 d post-partum. Colostrum ChT activity was 3,912 nmol/mL per hour and 465 nmol/mL per hour on the day of delivery and 4 d postpartum, respectively. Colostrum ChT activity was significantly higher at partum than at any other time. The ChT activity in colostrum was significantly greater at 1 d postpartum than at 2, 3, and 4 d postpartum. Chitotriosidase activity did not differ in colostrum collected on d 2, 3, and 4 postpartum. Chitotriosidase activity in goat kid blood serum ranged from 2,664 to 9,231 nmol/mL per hour at birth and 49 d of life, respectively. Chitotriosidase activity in the blood serum increased with age: at birth, activity was significantly less than at 28, 35, 42, 49, and 56 d postpartum. The maximum ChT activity in blood serum was observed at 49 d postpartum. Activity in 49-d-old kids was significantly greater than that observed in kids at 0, 7, and 14 d postpartum.

Forty-two Majorera kids (21 males and 21 females) were assigned to 3 groups, a colostrum group (C), a colostrum whey group (CW), and a colostrum whey plus milk replacer group (CWMR). All kids were fed twice on the first day and received 4 g of IgG/kg of body weight. No differences were found in serum IgG among the different treatments. Kid serum IgG concentrations on d 2 were 14.57, 17.25, and 13.32 mg/mL in the C, CW, and CWMR group, respectively. Labor time per animal was higher in the C and CW treatments than in the CWMR group (24.2 +/- 2.3, 20.9 +/- 3.4, and 16.1 +/- 1.5 min, respectively). This new management system may decrease labor costs during the colostrum feeding period.

Caprine colostrums (6 batches) were subjected to heat (56 degrees C for 60 min and 63 degrees C for 30 min) and high-pressure (400 and 500 MPa for 10 min at 20 degrees C) treatments at laboratory scale, and analyses of the main microbial groups and the extent of IgG denaturation (determined by immunodiffusion) were performed. Overall mean microbial values in raw colostrums were: total count, 5.55 log cfu/mL; Enterobacteriaceae, 2.64 log cfu/mL; lactococci, 5.41 log cfu/mL; lactobacilli, 2.34 log cfu/mL; and enterococci, 4.06 log cfu/mL. Neither Salmonella spp. nor Listeria monocytogenes were detected, whereas coagulase-positive staphylococci were found in various colostrum samples with an overall mean of 1.02 log cfu/mL. Heat and high-pressure treatments significantly reduced total count (1.47 log), lactococci (1.45 log), enterococci (2.47 log), and Enterobacteriaceae, whereas lactobacilli and coagulase-positive staphylococci counts were reduced to undetectable levels, but differences between technological treatments were not statistically significant. High-pressure treatments were as efficient in reducing the bacterial population as were heat pasteurization treatments: 95.50 and 96.93% for pressure treatments of 400 and 500 MPa, and 91.61 and 97.59% for heat treatments of 56 degrees C for 60 min and 63 degrees C for 30 min, respectively. All treatments assayed produced a reduction in colostrum IgG concentration (27.53, 23.58, 23.33, 22.09, and 17.06 mg/mL for raw, heat-treated at 56 degrees C for 60 min or 63 degrees C for 30 min, and pressure-treated at 400 and 500 MPa, respectively), but differences were only observed between raw colostrums and those pressure-treated at 500 MPa. This laboratory-scale study indicated that 20- to 30-mL volumes of goat colostrum could be heated and pressure-treated (400 MPa) to produce hygienic colostrum without affecting IgG concentration.

The effects on blood serum and colostrum immunoglobulin G (IgG) levels from including conjugated linoleic acid (CLA) in the diet were evaluated in goats. In experiment 1, 20 goats were allotted into two groups; the CLA group received 20 g/kg of dry matter of CLA 60 and the control group received 0 g/kg of CLA 60, from the third month of gestation until partum. Blood samples were taken every 2 weeks from 1 day before CLA inclusion to partum. From partum until 96 hours, blood and colostrum samples were taken every 24 h. Before partum, IgG serum blood levels were 15.6 and 9.4 mg/ml (CLA and control, respectively); the time effect was statistically significant. After partum (96 h), the effect of CLA dietary inclusion was statistically significant.

Three experiments were conducted including 180 Majorera kids. In the first experiment, the effect of use of lyophilized colostrum vs. frozen colostrum on immunoglobulin G (IgG) blood serum concentration was evaluated. Kids (n = 40) received the same management and IgG mass [3368 mg/kg of body weight (BW)] during the colostrum feeding period. The IgG in blood serum of kids from the lyophilized colostrum group was greater than that for kids that received frozen colostrum. The second experiment evaluated the effect of total IgG ingested by kids (n = 60) on IgG in blood serum during the colostrum feeding period. Three groups of animals received 3368, 1684, and 842 mg of IgG/kg of BW in 4 feedings for 2 d [high IgG concentration (H-IgG), medium IgG concentration (M-IgG), and low-IgG concentration (L-IgG), respectively]. The IgG blood serum in the kids that received H-IgG was greater than in the other 2 treatment groups, and no statistical differences were found for IgG in blood serum of kids that received either M-IgG or L-IgG. The third experiment evaluated the effect of timing of lyophilized colostrum meals on IgG blood serum concentration. Four groups of kids (n = 80) were used. Two groups received 1684 mg of IgG/kg of BW (higher level-1 d and higher level-2 d) and the other 2 groups received 842 mg of IgG/kg of BW (lower level-1 d and lower level-2 d). Two groups received 2 feedings in 1 d, and the other 2 groups received 4 feedings over a 2-d period, as denoted. Higher level-1 d kids had greater IgG blood serum concentration than the higher level-2 d kids, and no statistical differences were found between lower level-1 d and lower level-2 d kids.

Colostrum samples (n = 1084) of first and second milking from Majorera goats were taken. The immunoglobulin (Ig) G concentrations estimated by measurement of the color of goat colostrum and by the radial immunodiffusion technique were compared. Least squares analysis of the relationship between the color measurement method and IgG concentration resulted in a significant linear relationship. Using 20 mg of IgG/ mL of colostrum as the cut-off point for colostrum selection, the sensitivity, specificity, and negative predictive value of the color method as a test of IgG concentration in goat colostrum were 93.03, 71.43, and 78.12%, respectively.

Sixty Canary Caprine kids (30 males and 30 females) were assigned to three colostrum feeding managements, natural suckling (NS), hand-fed ad libitum colostrum (HALC) and restricted hand-fed colostrum (RHC). IgG concentrations were recorded in colostrum and kids serum at 12, 24, 36, 48, 60, 70, and 84 h of life. No significant differences in serum IgG concentrations were observed between kids receiving NS, HALC and RHC at any of the sampling times. Birth weight, prolificacy and sex were not significantly associated with serum IgG concentration. Kids that survived to completion of the experiment had significantly higher serum IgG concentrations than kids, which died. Higher correlations were founded between IgG feed and IgG serum levels. The aim of this paper was to evaluate the effects of refrigeration, several different methods of thawing, and pasteurization on the concentration of IgG in goat colostrum. Three different experiments were designed to analyse these effects. In the first of these, 50 samples of goat colostrum were stored in a cold-storage room at a temperature of 4 °C for a 3-month period. No statistically significant effects were observed within this time, although there was a reduction in IgG concentrations (32.98 and 25.11 mg/ml IgG at day 0 and 91, respectively). In the second experiment, 20 samples of goat colostrum were frozen and subsequently thawed using four different methods: hot water (60 °C), refrigeration (4 °C), room temperature (27 °C) and microwave (55 °C). The process was carried out seven times for each of the four methods. The method of thawing did not affect the colostrum IgG concentration. However, the repetition of freezing and thawing tended to reduce IgG concentrations, albeit to no significant degree (15.50 and 10.73 mg/ml IgG at cycle 0 and 7, respectively). In the third experiment, 30 goat colostrum samples were used and a reduction of approximately 35% of IgG concentration after pasteurization was observed. Abstract: The objective of this study was to examine the chemical and immunological quality of goat colostrum following delivery. Twenty dairy goats of the Majorera breed were milked 1h postpartum and then every hour for 10h after the first milking. Residual colostrum (RC) was also obtained at the time of the first milking after i.v. injection of 2I.U. of oxytocin. Colostrum yield, approximate composition, SCC, IgG, IgM, IgA and chitotriosidase activity were measured in milking colostrum (MC), RC, and colostrums in each hour sample. A PROC MIXED procedure was performed and a Tukey''s test was done to determine the statistical significance of differences in the composition of the colostrum fractions and the colostrum obtained over time. At the first milking, MC and RC weighed 2506 and 237g, respectively. At 1h and 10h after the first milking, the colostrum yield was 174 and 120g, respectively, with a continuous drop in yield over the course of the experiment. A drop in protein production was also seen over time. Colostrum protein percentages were 10.4 and 10.2 in MC and RC. Colostrum protein percentages at 1 and 10h after first milking were 9.7% and 4.5%, respectively. The percentage of colostrum fat increased 1h after the first milking and then decreased to 6.1% at 10h after the first milking. The lactose colostrum percentage displayed an increase during the experimental period. Colostrum SCC was not affected by colostrum fractioning or time, with a range of 4.2–5.8×106 cells/mL. IgG, IgM, IgA levels and chitotriosidase activity did not present differences between colostrum fractions at the first milking but displayed a drastic drop in subsequent milkings. In sum, the chemical and immunological quality of colostrum dropped quickly after the first milking, and thus goat keepers need only recover the first milked colostrum when they rear goat kids separately from dams. Abstract: Forty newborn Majorera goats were randomly assigned to one of two groups. On the first day of life, the kids were fed atomized colostrum. The kids were subsequently fed a milk replacer (16% w/v) twice a day. The experimental group received two intramuscular doses of a reputed immunostimulant composed of a Propionibacterium acnes extract, Ochrobactrum intermedium lipopolysaccharides, and Proclin® at 10 and 40 days of age, whereas, the control group received two doses of saline solution. Blood samples were collected weekly from kids age 10 to 66 days postpartum to measure the total and differential white blood cell counts, the plasma IgG, IgA, and IgM levels, and the activities of the total and the alternative complement systems. The white blood cell and the differential cell counts were the same for both groups. No statistical differences were found for the plasma IgG, IgA, and IgM concentrations. Complement system activity was undetectable in both groups. Summarizing, the reputed immunostimulant did not affect the immune system of goats. Thus, veterinarians should carefully consider whether it is prudent to recommended this kind of product, because while it increases farmers’ expenses it apparently does not improve the state of the innate immune system of the neonatal goat. Abstract: Thirty new born male kids were allotted into three groups to evaluate the effects of diet and age on complement system activity in serum. After the colostrum feeding period, the control group (C) received a commercial milk replacer; the CLA group received 20 g/kg milk replacer DM of conjugated linoleic acid; and the GMK group was fed with goat milk. The kids were fed colostrum for 2 days and then either milk replacer or goat milk from days 3 to 60. Blood samples were taken at 1, 10, 20, 30, 40, 50 and 60 days of age. The complement system activity was higher (P <0.05) in the GMK group throughout the whole experiment through alternative pathway. C kids did not present complement activity at any time. CLA group complement activity was statistically higher than that of C kids from 30 to 60 days, through alternative pathways. In conclusion, the milk replacer formula should be reformulated because it did not induce any complement system activity in the first two months of life. [Copyright &y& Elsevier]

Abstract: Sixty Majorera goats were allotted into groups according to whether they were in their first or second lactation, or whether they had more than two (n =20, respectively). In each group, 10 goats were of single litter size and 10 were of multiple litter size. After giving birth, twelve 300ml colostrum samples were obtained every 12h by manual milking from both udders. The contents of fat, protein, total IgG and lactose were determined and pH, density and electric conductivity were measured. The levels of protein and total IgG contents dropped quickly from the time of birth to 132h postpartum, whereas fat content increased from birth to 24h postpartum and subsequently decreased until 132h postpartum. The lactose content increased slightly from birth to 132h postpartum and pH and electric conductivity increased slightly, whilst density values dropped quickly after partum. [Copyright &y& Elsevier]

The aim of the present work was to evaluate the effectiveness of the use of refrigerated and frozen goat colostrum and commercial sheep colostrum in kids. Forty-five Canary Caprine Group kids were distributed in three groups (n=15) according to the type of colostrum fed. The refrigerated and frozen colostrum were administered twice daily for 2 days, each kid receiving 5% of the body weight per feed. Both the refrigerated and frozen colostrum had the same IgG concentration before refrigerating or freezing. The commercial colostrum was administered according to the recommendations of the manufacturer. Blood samples were obtained from the kids (by jugular extraction) every 12 h from birth to the third day post-partum and two additional samples were taken at 15 and 30 days of life. The IgG concentration in the blood was measured by radial immunodifusion on agarose gel. At birth the kids were agammaglobulinemic.The peak IgG was at 24 h with frozen goat colostrum fed kids (25.47±19.89 mg/ml), 36 h for kids fed with refrigerated goat colostrum (15.84±5.91 mg/ml) while those fed commercial sheep colostrum peaked at 30 days (0.84±0.65 mg/ml). The birth weight is of great significance with regard to these results. In conclusion, kids fed only with commercial colostrum do not acquire the necessary immunity to protect them during the first month of life. [Copyright &y& Elsevier]
