Laccase is a copper-containing polyphenol oxidase that can oxidize phenolic and non-phenolic organic substrates. In the past decades, laccases had received considerable attention because of the ability to degrade various organic substances. Based on the codon preference of the Pichia pastoris expression system, this study optimized the gene structure of the laccase gene Lcc1 from Coprius cinerea through synthetic biology methods. A new gene Lcc1I was synthesized and heterologously expressed in P. pastoris. After 3 days of cultivation in a shake flask at 30°C, the transformants produced at a yield of 890 mg L -1 protein. The highest production level of the recombinant laccase was 2760 U L -1 . The molecular mass of the recombinant laccase was estimated at 60 kDa. The enzyme showed highest activity at pH 3.4 and 45°C. It possessed better stability at higher pH and lower temperature condition. Using 2,2'-azino-bis-(3-ethylbenzothiazoline)-6-sulphonate (ABTS) as the substrate, the K m and V max values were 0.136 mM and 9778 μM min -1  mg -1 , respectively. The recombinant laccase could directly oxidize some triphenylmethane dyes like leuco-crystal violet (LCV) and leuco-malachite green (LMG). With the help of ABTS mediator, it could oxidize and degrade 77.7% crystal violet (CV) and 79.2% malachite green (MG) within 1 h. Our results indicate that optimization of the laccase gene achieves good expression results in the host system. The purpose of this study is to evaluate the decolorization ability and detoxification effect of LAC-4 laccase on various types of single and mixed dyes, and lay a good foundation for better application of laccase in the efficient treatment of dye pollutants. The reaction system of the LAC-4 decolorizing single dyes (azo, anthraquinone, triphenylmethane, and indigo dyes, 17 dyes in total) were established. To explore the decolorization effect of the dye mixture by LAC-4, two dyes of the same type or different types were mixed at the same concentration (100 mg/L) in the reaction system containing 0.5 U laccase, and time-course decolorization were performed on the dye mixture. The combined dye mixtures consisted of azo + azo, azo + anthraquinone, azo + indigo, azo + triphenylmethane, indigo + triphenylmethane, and triphenylmethane + triphenylmethane. The results obtained in this study were as follows. Under optimal conditions of 30 °C and pH 5.0, LAC-4 (0.5 U) can efficiently decolorize four different types of dyes. The 24-hour decolorization efficiencies of LAC-4 for 800 mg/L Orange G and Acid Orange 7 (azo), Remazol Brilliant Blue R (anthraquinone), Bromophenol Blue and Methyl Green (triphenylmethane), and Indigo Carmine (indigo) were 75.94%, 93.30%, 96.56%, 99.94%, 96.37%, and 37.23%, respectively. LAC-4 could also efficiently decolorize mixed dyes with different structures. LAC-4 can achieve a decolorization efficiency of over 80% for various dye mixtures such as Orange G + Indigo Carmine (100 mg/L+100 mg/L), Reactive Orange 16 + Methyl Green (100 mg/L+100 mg/L), and Remazol Brilliant Blue R + Methyl Green (100 mg/L+100 mg/L). During the decolorization process of the mixed dyes by laccase, four different interaction relationships were observed between the dyes. Decolorization efficiencies and rates of the dyes that were difficult to be degraded by laccase could be greatly improved when mixed with other dyes. Degradable dyes could greatly enhance the ability of LAC-4 to decolorize extremely difficult-to-degrade dyes. It was also found that the decolorization efficiencies of the two dyes significantly increased after mixing. The possible mechanisms underlying the different interaction relationships were further discussed. Free, but not immobilized, LAC-4 showed a strong continuous batch decolorization ability for single dyes, two-dye mixtures, and four-dye mixtures with different structures. LAC-4 exhibited high stability, sustainable degradability, and good reusability in the continuous batch decolorization. The LAC-4-catalyzed decolorization markedly reduced or fully abolished the toxic effects of single dyes (azo, anthraquinone, and indigo dye) and mix dyes (nine dye mixtures containing four structural types of dyes) on plants. Soluble oligomers of the β-amyloid peptide, Aβ, are associated with the progression of Alzheimer's disease. Although many small molecules bind to these assemblies, the details of how these molecules interact with Aβ oligomers remain unknown. This paper reports that crystal violet, and other C3 symmetric triphenylmethane dyes, bind to C3 symmetric trimers derived from Aβ 17-36 . Binding changes the color of the dyes from purple to blue, and causes them to fluoresce red when irradiated with green light. Job plot and analytical ultracentrifugation experiments reveal that two trimers complex with one dye molecule. Studies with several triphenylmethane dyes reveal that three N, N-dialkylamino substituents are required for complexation. Several mutant trimers, in which Phe 19 , Phe 20 , and Ile 31 were mutated to cyclohexylalanine, valine, and cyclohexylglycine, were prepared to probe the triphenylmethane dye binding site. Size exclusion chromatography, SDS-PAGE, and X-ray crystallographic studies demonstrate that these mutations do not impact the structure or assembly of the triangular trimer. Fluorescence spectroscopy and analytical ultracentrifugation experiments reveal that the dye packs against an aromatic surface formed by the Phe 20 side chains and is clasped by the Ile 31 side chains. Docking and molecular modeling provide a working model of the complex in which the triphenylmethane dye is sandwiched between two triangular trimers. Collectively, these findings demonstrate that the X-ray crystallographic structures of triangular trimers derived from Aβ can be used to guide the discovery of ligands that bind to soluble oligomers derived from Aβ.

Laccase CotA from Bacillus subtilis 168 was successfully displayed on the membrane of Escherichia coli cells using poly-γ-glutamate synthetase A protein (PgsA) from B. subtilis as an anchoring matrix. Further analyses demonstrated that the fusion protein PgsA/CotA efficiently translocates to the cell surface of E. coli with an enzymatic activity of 65 U/10 8 cells. Surface-displayed CotA was shown to possess improved enzymatic properties compared with those of the wild-type CotA, including higher thermal stability (above 90% activity at 70 °C and nearly 40% activity at 90 °C after 5-h incubation) and stronger inhibitor tolerance (approximately 80 and 65% activity when incubated with 200 and 400 mM NaCl, respectively). Furthermore, the whole-cell system was demonstrated to have high enzymatic activity against anthraquinone dye, Acid Blue 62, triphenylmethane dye, Malachite Green, and azo dye, Methyl Orange with the decolorization percentages of 91, 45, and 75%, after 5-h incubation, respectively.

Detection of triphenylmethane dyes (TDs), especially the widely used malachite green (MG) and crystal violet (CV), plays an important role in safety control of aquatic products. There are two chromatic forms of TDs: oxidized or reduced. Usually, only one form can be detected by reported ELISA antibodies. In this article, molecular shape superimposing and quantum mechanics calculation were employed to elucidate the differences between MG, CV, and their reduced chromatic forms (leucomalachite green, LMG and leucocrystal violet, LCV). A potential hapten was rationally designed and synthesized. Polyclonal antibodies were raised through immunizing New Zealand white rabbits and BALB/C mice. We tested the cross-reactivity ratios between the hapten and TDs. The cross-reactivity ratios were correlated with the difference in surface electrostatic potential. The determination coefficients ( r ²) of the correlations are 0.901 and 0.813 for the rabbit and mouse antibody, respectively. According to this linear model, the significant difference in the atomic charge seemed to make it impossible to find a hapten that can produce antibodies with good cross-reactivities with both reduced and oxidized TDs., Competing Interests: The authors declare no conflict of interest.

The industrial dye-contaminated wastewater has been considered as the most complex and hazardous in terms of nature and composition of toxicants that can cause severe biotic risk. Reactive azo, anthroquinone and triphenylmethane dyes are mostly used in dyeing industries; thus, the unfixed hydrolysed molecules of these dyes are commonly found in wastewater. In this regard, bacterial species have been proved to be highly effective to treat wastewater containing reactive dyes and heavy metals. The bio-decolourisation of dye occurs either by adsorption or through degradation in bacterial metabolic pathways under optimised environmental conditions. The bacterial dye decolourisation rates vary with the type of bacteria, reactivity of dye and operational parameters such as temperature, pH, co-substrate, electron donor and dissolved oxygen concentration. The present paper reviews the efficiency of bacterial species (individual and consortia) to decolourise wastewater containing reactive azo, anthroquinone and triphenylmethane dyes either individually or mixed or with metal ions. It has been observed that bacteria Pseudomonas spp. are comparatively more effective to treat reactive dyes and metal-contaminated wastewater. In recent studies, either immobilised cell or isolated enzymes are being used to decolourise dye at a large scale of operations. However, it is required to investigate more potent bacterial species or consortia that could be used to treat wastewater containing mixed reactive dyes and heavy metals like chromium ions.

Staphylococcus aureus, showing the greatest decolorization ability, was further investigated for Methyl Red (MR) Congo Red (CR), Crystal Violet (CV) and Malachite Green (MG) decolorization using response surface methodology (RSM). The chemometric methods use, based on statistical design of experiments (DOEs) such as RSM is becoming increasingly widespread in several sciences such as analytical chemistry, engineering and environmental chemistry. Stapphylococcus aureus ATCC 25923, Stapphylococcus aureus (S1) and Stapphylococcus aureus (S2), were isolated from textile wastewater plant located in KsarHellal, Tunisia and were tested for their decolorization capacity. PCR technique was utilized to identify the 3 bacterial strains and to detect the adhesin gene "cna". Biodegradation of MR, CR, CV and MG (750 ppm), were investigated under shaking condition in Mineral Salt Medium (MSM) solution at pH 7.5 and temperature 30 °C, using a 3.7 × 10 5  CFU/ml as inoculum size. Our results showed that Staphylococcus aureus had a high decolorization capacity. Nuclear magnetic resonance (NMR) spectroscopy analysis confirmed the biodegradation of dyes. The four dyes mutagenicity with the S9 metabolizing system decreased significantly after biodegradation and totally disappeared. An anaerobic sludge (AS), capable of decolorizing a variety of synthetic dyes, was acclimated and is reported here. The sludge presented a much better dye decolorizing ability than that of different individual strains. A broad spectrum of dyes could be decolorized by the sludge. Continuous decolorization tests showed that the sludge exhibited the ability to decolorize repeated additions of dye. The chemical oxygen demand (COD) removal rate of the dye wastewater reached 52% after 12 h of incubation. Polymerase chain reaction and denaturing gradient gel electrophoresis (PCR-DGGE) profiles revealed that the microbial community changed as a result of varying initial concentrations of dyes. Phylogenetic analysis indicated that microbial populations in the sludge belonged to the phyla Acidobacteria, Firmicutes, Bacteroidetes, Chloroflexi and Proteobacteria. The degradation products of the three types of dye were identified. For azo dyes, the anaerobic sludge converted Methyl Orange to N , N -dimethylbenzene-1,4-diamine and 4-aminobenzenesulfonic acid; for triphenylmethane dyes, after Malachite Green was decolorized, the analyzed products were found to be a mixture of N , N -dimethylbenzenamine, 3-dimethyl-aminophenol and 4-dimethylaminobenzophenone; for anthraquinone dyes, two products (acetophenone and 2-methylbenzoic acid) were observed after Reactive Blue 19 decolorization. Together, these results suggest that the anaerobic sludge has promising potential for use in the treatment of industrial wastewater containing various types of dyes., Competing Interests: The authors declare no conflict of interest. Because loop-mediated isothermal amplification (LAMP) can amplify substantial amounts of DNA under isothermal conditions, its applications for simple genetic testing have attracted considerable attention. A positive LAMP reaction is indicated by the turbidity caused by by-products or by the color change after adding a metallochromic indicator to the reaction solution, but these methods have certain limitations. Leuco crystal violet (LCV), a colorless dye obtained after sodium sulfite treatment of crystal violet (CV), was used as a new colorimetric method for detecting LAMP. LCV is reconverted into CV through contact with double-stranded DNA (dsDNA). Therefore, the positive reaction of LAMP is indicated by color change from colorless to violet. The assay is sensitive enough to detect LAMP products, with a detection limit of 7.1 ng/μl for dsDNA. It is also highly selective to dsDNA, and interference with single-stranded DNA and deoxynucleotide triphosphates (dNTPs) is not observed. LCV facilitates direct colorimetric detection of the main product rather than a by-product of the LAMP reaction; therefore, this method can be used under various reaction conditions such as those with added pyrophosphatase in solution. To investigate the removal capability of millimetric zero valent iron (mmZVI), sponge iron (s-Fe(0)) particles were characterized with XRD, XPS, TEM, HRSEM and EDS techniques. Moreover, the roles of particle size, catalyst dosage, dye concentration, mixing conditions (e.g. ultrasound (US), stirring or shaking), and regeneration treatment were studied with the removal of cationic triphenylmethane dyes. Notably, the reduction process was also revealed as compared to nanoscale zero valent iron (nZVI), microscale iron power, and iron scurf. Furthermore, the reductive mechanism was exemplified with brilliant green. The results demonstrated that (1) the synergetic effect between US and s-Fe(0) greatly enhanced the removal of dyes, (2) the dosage of preferred s-Fe(0) (1-3mm) particles was optimized as 30.0g/L; (3) reuse cycles of s-Fe(0) catalyst were enhanced with the assistance of diluted HCl solution; (4) the main degradation routes included the cleavage of conjugated structure reactions, N-de-ethylation reactions, hydroxylation reactions, the removal of benzene ring reactions, and opening ring reactions. Accordingly, the pretreatment of aqueous solution over s-Fe(0) was hypothesized to achieve mainly through direct reduction reaction by electron transfer and indirect reductive reactions by the highly activated hydrogen atom. Excited state dynamics of S2 and S1 states for a series of TPM dyes, pyrogallol red (PGR), bromopyrogallol red (Br-PGR) and aurin tricarboxylic acid (ATC), have been monitored by using ultrafast transient absorption and fluorescence up-conversion techniques. Optical absorption studies indicate that all the TPM dyes exist as keto-enol tautomers depending upon the pH of the solution. Interestingly, all the TPM dyes give S2 emission (major emitting state) in addition to weak S1 emission. S2 emission lifetimes as fast as ∼150-300 fs and S1 emission lifetimes of 2-5 ns were observed depending upon the molecular structure of the dyes. Femtosecond transient absorption studies suggest the presence of an ultrafast non-radiative decay channel from the S2 state in addition to S2 luminescence. The vibrational relaxation time from hot S1 state is found to be 2-6 ps. The heavy atom effect has been observed in ultrafast relaxation dynamics of Br-PGR.

Although conjugation is well known as an important contributor to color, there is scant recognition concerning involvement of imine and iminium functions in the physiological effects of this class of dyes and pigments. The group includes the dyes methylene blue, rhodamine, malachite green, fuchsin, crystal violet, auramine and cyanins, in addition to the pigments consisting of pyocyanine, phthalocyanine and pheophytin. The physiological effects consist of both toxicity and beneficial aspects. The unifying theme of electron transfer-reactive oxygen species-oxidative stress is used as the rationale in both cases. Toxicity is frequently prevented or alleviated by antioxidants. The apparent dichotomy of methylene blue action as both oxidant and antioxidant is rationalized based on similar previous cases. This mechanistic approach may have practical benefit. This contribution examines the utility of vibrational spectroscopy by bench and portable Raman/surface enhanced Raman and infrared methods for the investigation of ten early triarlymethane dye powder references and dye solutions applied on paper. The complementary information afforded by the techniques is shown to play a key role in the identification of specific spectral marker ranges to distiguish early synthetic dyes of art-historical interest through the elaboration of an in-house database of modern organic dyes. Chemometric analysis has permitted a separation of data by the discrimination of di-phenyl-naphthalenes and triphenylmethanes (di-amino and tri-amino derivatives). Triphenylmethane (TPM) dye is one of the most prevalent and recalcitrant water contaminants. Magnetic reduced graphene oxide (rGO) is an efficient adsorbent for organic pollutants removal. However, the performance and adsorption kinetics of magnetic rGO towards TPM have not yet been studied. In this study, a magnetic Fe3O4@rGO nano-composite, which could be easily removed from water with a simple magnetic separation step was synthesized and characterized. The magnetic rGO showed fast adsorption rate and high adsorption capacity towards different TPM dyes (the Langmuir monolayer adsorption capacity is 64.93 mg/g for adsorption of crystal violet). The adsorption processes are well-fitted to the pseudo-second-order kinetic model (R(2) > 0.99) and the Langmuir isotherm model (R(2) = 0.9996). Moreover, the magnetic rGO also showed excellent recycling and regeneration capabilities. The results indicated that adsorption with magnetic rGO would be a promising strategy to clean up the TPM contamination.

Femtosecond time-resolved infrared spectroscopy was employed to study intramolecular charge transfer in triphenylmethane dyes, including malachite green (MG), malachite green carbinol base (MGCB), and leucomalachite green (LMG). A local excited state (LE) and a twisted intramolecular charge-transfer (TICT) state have been observed directly in MG. Furthermore, solvent-controlled TICT measurements in a series of linear alcohols indicate that the transition time (4-11 ps) from LE to TICT is strongly dependent on alcohol viscosity, which is due to rotational hindrance of dimethylaniline in high-viscosity solvents. For LMG, no TICT is observed due to steric hindrance caused by the sp(3)-hybridized central carbon atom. However, for MGCB, TICT is rescued by the addition of the electron-donating hydroxyl group to the bridge. These results for MG and its analogues provide new insight regarding the dynamics and mechanism of twisted intramolecular charge transfer (TICT) in triphenylmethane dyes.

The production of porous cross-linked enzyme aggregates (p-CLEAs) is a simple and effective methodology for laccase immobilization. A three-phase partitioning technique was applied to co-precipitate laccase and starch, followed by cross-linking with glutaraldehyde and removal of starch by α-amylase to create pores in the CLEAs. Scanning electron microscopy revealed a very smooth spherical structure with numerous large pores. The half-life of free laccase at 55°C was calculated to be 1.3h, while p-CLEAs did not lose any activity even after 14 h. p-CLEAs also exhibited improved storage stability, catalytic efficiency and could be recycled 15 times with 60% loss of activity. The catalysts decolorized triphenylmethane and reactive dyes by 60-70% at initial dye concentrations of 2 and 0.5 g L(-1), respectively, without any mediators. Decolorization of triphenylmethane dyes by DTT was researched. For malachite green, content of DTT in reaction system was optimized to investigate the quantitative relation between DTT and malachite green and the decolorization capacity of DTT was confirmed. Effect of pH of reaction system on reducibility of DTT was explored. The results indicated 4 mol malachite green could be decolorized by 1 mol DTT averagely within 1 min, when pH of the reaction system was above 5. The decolorization rate was up to 97%. Decolorization product of malachite mreen was corroborated to be its leuco form by HPLC analysis. Some insoluble compounds, which could be the complex products of leuco malachite green with DTT, were formed during the decolorization reaction. Decolorization of crystal violet, brilliant green and basic fuchsin by DTT was tested further, and the decolorization rates were all above 85%, which suggested DTT was a broad-spectrum decolorization agent for triphenylmethane dyes.

A leuco derivatives of triphenylmethane dyes degrading bacterium, strain CM9, was isolated from an aquafarm field. Based on morphology, physiologic tests, 16S rDNA sequence, and phylogenetic characteristics, it was identified as Sphingomonas sp. This strain was capable of degrading leucomalachite green (LMG), leucocrystal violet and leucobasic fuchsin completely. The relationship between bacterium growth and LMG degradation suggested that strain CM9 could use LMG as the sole source of carbon. The most LMG degradation activity of CM9 crude extract was observed at pH 7.0 and at 30°C. Many metal ions had little inhibition effect on the degradation activity of the crude extract. CM9 also showed strong decolorization of triphenylmethane dyes to their leuco derivatives. GC/MS analysis detected two novel metabolic products, methylbenzene and 4-aminophenol, during the LMG degradation by CM9.

Objective: We studied the effects of different organic solvents and inhibitors on the decolorization of recombinant triphenylmethane dyes decolorization enzyme (TpmD), expressed extracellularly from Pichia pastoris., Methods: The recombinant TpmD was purified by ultrafiltration and Ni2+ affinity chromatography. The decolorization activity for malachite green was determined by UV-visible spectrophotometer. We studied the effects of some organic solvents and inhibitors on TpmD activity through the rate of decolorization decreased using malachite green as the substance. We also compared the difference between dithicthreitol (DTT) and NADH as the cofactor in assisting the decolorization by monitoring the dissolved oxygen consumption and the end products., Results: The effect of methanol on the enzymatic activity was weak since TpmD still retained its high activity in the reaction environment containing 10% - 20% methanols. The ethanol and acetone made the enzymatic activity fade away quickly. Dimethylsulfoxide in low concentration was propitious to keep the TpmD activity, although 30% dimethylsulfoxide inhibited the enzymatic activity lost in a half. Ethylenediaminetetraacetic acid (EDTA) of low concentration, L-cysteine and NaN3 all exhibited only weakly inhibitory effects; higher concentrated (25 mmol/L) EDTA could strongly inhibit enzymatic activity and sodium dodecylsulfonate (SDS) inhibited the activity completely. The effect of DTT on TpmD activity was beyond the expectation. It could substitute of coenzyme NADH to assist and accelerate the enzyme in decolorizing reaction. We found that the dissolved oxygen consumption behaviors and reaction end products measuring by UV-Visible full wave-scan were completely different between the reactions assisted with DTT or NADH. This is the first report about DTT which can act as a cofactor for a decolorization enzyme., Conclusion: The effects of different organic solvents and inhibitors on the enzymatic activity of TpmD are very different. By the dissolved oxygen assay and the end products analysis, we concluded that the decolorizing reaction assisted with DTT was a novel reaction process. The mechanism of reaction with DTT as cofactor is completely different from that with NADH as coenzyme.

Triphenylmethane dyes are aromatic xenobiotic compounds that are widely considered to be one of the main culprits of environmental pollution. Triphenylmethane reductase (TMR) from Citrobacter sp. strain KCTC 18061P was initially isolated and biochemically characterized as an enzyme that catalyzes the reduction of triphenylmethane dyes. Information from the primary amino acid sequence suggests that TMR is a dinucleotide-binding motif-containing enzyme; however, no other functional clues can be derived from sequence analysis. We present the crystal structure of TMR in complex with NADP+ at 2.0-angstroms resolution. Despite limited sequence similarity, the enzyme shows remarkable structural similarity to short-chain dehydrogenase/reductase (SDR) family proteins. Functional assignments revealed that TMR has features of both classic and extended SDR family members and does not contain a conserved active site. Thus, it constitutes a novel class of SDR family proteins. On the basis of simulated molecular docking using the substrate malachite green and the TMR/NADP+ crystal structure, together with site-directed mutagenesis, we have elucidated a potential molecular mechanism for triphenylmethane dye reduction.

Fenton's reagent (Fe2+ +H2O2) has been electrogenerated in situ in an undivided electrolytic cell from the effective reduction of Fe3+ and O2 at carbon-felt cathode for the treatment of aqueous solutions of four triphenylmethane dyes (TPMs), namely malachite green (MG), crystal violet (CV), methyl green (MeG) and fast green FCF (FCF), at pH 3.0 and room temperature. MG has been used as a model among them to study the influence of some experimental parameters on the decay kinetics, COD removal and current efficiency. The results in such electro-Fenton system are explained in terms of the many parasitic reactions involving .OH. Higher efficiency values are obtained with rising organic content and decreasing applied current. The first stage of the mineralization process, involving aromatic by-products, leads to fast decoloration as well as quick initial COD removal that fit well to a pseudo-first-order kinetics. At prolonged electrolysis time, the mineralization rate and efficiency decrease due to the formation of hardly oxidizable compounds and the enhancement of wasting reactions. Solutions of all four TPMs are quickly degraded following a pseudo-first-order decay kinetics. The absolute rate constant (kTPM) for their reaction with .OH increases in the order MeG

In pH 4.2-4.8 HAc-NaAc buffer solution, folic acid (FA) could react with uranium (VI) to form a 2:1 anionic chelate which further reacted with some basic triphenylmethane dyes (BTPMD) such as Ethyl Violet (EV), Methyl Violet (MV) and Crystal Violet (CV) to form 1:2 ion-association complexes. As a result, not only the absorption spectra were changed, but also the intensities of resonance Rayleigh scattering (RRS) were enhanced greatly and the new RRS spectra were observed. The maximum RRS wavelengths were located at 328 nm for EV system, 325 nm for MV system and 328 nm for CV system. The fading degree (DeltaA) and RRS intensities (DeltaI) of three systems were different. Under given conditions, the DeltaA and DeltaI were all directly proportional to the concentration of FA. The linear ranges and the detection limits of RRS methods were 0.0039-5.0 microg mL(-1) and 1.2 ng mL(-1) for EV system, 0.0073-4.0 microg mL(-1) and 2.2 ng mL(-1) for MV system, 0.014-3.5 microg mL(-1) and 4.7 ng mL(-1) for CV system. The RRS methods exhibited higher sensitivity, so they are more suitable for the determination of trace FA. The optimum conditions, the influencing factors and the effects of coexisting substances on the reaction were investigated. The method can be applied to the determination of FA in serum and urine samples with satisfactory results. The structure of the ternary ion-association complex and the reaction mechanism were discussed in this work.

The colour change of triphenylmethane (TPM) dyes induced by surfactants at concentrations much greater than their critical micellar concentrations is found to be accompanied by enhanced fluorescence. Thus, the otherwise weak fluorescence of TPM dyes can be detected using supramicellar surfactant concentrations. In this respect, the nonionic polyoxyethylene (POE) chain-containing surfactants are found to be more efficient compared with ionic surfactants. The POE surfactants, Triton X-100, Tween-20 and Tween-60 present a polymer-like surface to the dyes, which can thus easily bind to them. At supramicellar concentrations, the hydrophobic environment formed in these micelles is effective in preventing nonradiative relaxation processes of the dyes. As a result, there is enhanced fluorescence for even micromolar concentrations of the dyes. Among the Tween series, Tween-60 being more hydrophobic leads to greater fluorescence enhancement than Tween-20. From the fluorescence properties, binding constants for dye binding to the surfactants can be determined. Thus the relative efficiency of these surfactants as binding substrates can be assessed. Another interesting observation is that the electrolyte LiCl in presence of the surfactants leads to even larger fluorescence enhancement than the surfactants alone.

Polarographic reduction of some triphenylmethane dyes in strictly anhydrous solutions was studied in the absence and presence of alpha-lipoic acid. The values of the half-wave potentials E1/2 and the parameter of potential carcinogenicity tg alpha were determined for all compounds studied. The current value of the first diffuse polarographic wave was increased in the presence of alpha-lipoic acid for all of the compounds. The increase was linear and depended on the alpha-lipoic acid concentration in anhydrous solutions. The highest determined value of tg alpha belonged to crystal violet (0.420) and to methyl violet 2B (0.440). The values indicate potential carcinogenic activities of the respective triphenylmethane dyes.

(1) Basic triphenylmethane dyes related to pararosanilin inhibit class alpha glutathione S-transferases (GSTs) from rat liver. The inhibitory potency of each dye correlates with its octanol-water partition coefficient. Values of Ki determined at pH 6.5 ranged from about 1 x 10(-7) M for Ethyl violet to 7 x 10(-5) M for Methyl green. GST 3-3, a class mu isoenzyme, was an order of magnitude less sensitive to inhibition by Ethyl violet. (2) All of the dyes tested were bleached to varying degrees by glutathione. The bleaching appears to result from the formation of an adduct between the dye and glutathione. At pH 6.5, adduct formation is significant only for Malachite green and Methyl green. There is kinetic evidence that for these dyes the adduct contributes significantly to the overall inhibition. It is probable that at physiological pH, all of the dyes would exist to a significant extent in the adduct form. (3) The dyes are excreted extensively in the bile, at least partly as the glutathione adduct. The free dye is regenerated on standing, it is assumed as a result of removal of glutathione by oxidation.
