Tribolium castaneum and Rhyzopertha dominica are cosmopolitan, destructive postharvest pests. Although research has investigated how high densities of T. castaneum affect attraction to the aggregation pheromone by conspecifics, research into the behavioral response of both species to food cues after high density exposure has been lacking despite its importance to foraging ecology. Our goal was to manipulate and observe the effects of crowding on the behavioral response of both species to common food and pheromonal stimuli and to determine how the headspace emission patterns from grain differed under increasing densities. Densities of colonies for both species was altered (10-500 adults) on a fixed quantity of food (10 g of flour or whole wheat), then the behavioral response to common food and pheromonal cues was evaluated in a wind tunnel and release-recapture experiment, while volatiles were examined through gas chromatography coupled with mass spectrometry. Importantly, at least for T. castaneum, crowded conditions attenuate attraction to food-based stimuli, but not pheromonal stimuli. Crowding seemed to have no effect on R. dominica attraction to food and pheromonal stimuli in the wind tunnel, but exposure to high density cues did elicit 2.1-3.8-fold higher captures in traps. The relative composition and abundance of headspace volatiles emitted varied significantly with different densities of beetles and was also species-specific. Long-term trapping programs of stored product pests provide information for timely and accurate pest management. Tribolium castaneum (Herbst) (Coleoptera: Tenebrionidae) is a highly successful external-infesting grain pest and is frequently monitored using a commercial pitfall trap that combines pheromonal and kairomonal stimuli. However, an often overlooked component of lure-based traps is the potential for the volatile plume to change over time as individuals are captured. These now-dead insects may then release necromones altering the captures of conspecifics. In this study, we evaluated changes in (i) the behavior of T. castaneum and (ii) the relative change in volatiles over time since dead insects were added and among different densities of dead conspecifics in a commercially available kairomone oil. We used multiple behavior assays, including wind tunnel, release-recapture, and 2-way olfactometer, and performed chemical analyses via headspace collection and gas chromatography coupled with mass spectrometry. Tribolium castaneum response to the kairomone lure was not consistent among assays of density of conspecifics between 4 and 40 adults after 24 or 96 h, or time of seeding over 1-96 h or 8-11 days prior. Tested strains collected in 2012 and 2019 ruled out strain-specific differences. Oil batch effects were also ruled out as a factor contributing to the response of T. castaneum. The relative volatile composition was generally stable among the treatments despite using different seeding densities and seeding times. The increasing popularity of low- and no-gluten flours as wheat flour alternatives has driven the need to understand risks of insect pest infestation in these products. Previous research using no-choice bioassays found that the red flour beetle, Tribolium castaneum Herbst can oviposit and develop on a range of alternative flours; here we test T. castaneum preference by assessing attraction in a wind tunnel assay and oviposition preference in both small- and large-scale choice assays between alternative and wheat flour. Some flours such as buckwheat, teff, millet, rice, and rye elicit similar responses as wheat flour for both attraction and oviposition. Other flours such as cassava and oat were not preferred for either oviposition or attraction behaviors. Flours like sorghum and amaranth, had mixed preferences for oviposition, with decreased oviposition in the small arena but not the large arena. Comparisons to published developmental success rates of T. castnaeum on these dietsindicate that females can choose diets on which they have high developmental success, such as buckwheat or teff, and avoid flours like cassava where developmental success is low. However, mismatch of oviposition preference and developmental success also occurs, in flours such as rice and amaranth. These results suggest the red flour beetle has limited ability to make adaptive food selections for egg laying. The lesser grain borer, Rhyzopertha dominica (F.) (Coleoptera: Bostrichidae), is a major global pest of cereal grains. Infestations are difficult to control as larvae feed inside grain kernels, and many populations are resistant to both contact insecticides and fumigants. We sequenced the genome of R. dominica to identify genes responsible for important biological functions and develop more targeted and efficacious management strategies. The genome was assembled from long read sequencing and long-range scaffolding technologies. The genome assembly is 479.1 Mb, close to the predicted genome size of 480.4 Mb by flow cytometry. This assembly is among the most contiguous beetle assemblies published to date, with 139 scaffolds, an N 50 of 53.6 Mb, and L 50 of 4, indicating chromosome-scale scaffolds. Predicted genes from biologically relevant groups were manually annotated using transcriptome data from adults and different larval tissues to guide annotation. The expansion of carbohydrase and serine peptidase genes suggest that they combine to enable efficient digestion of cereal proteins. A reduction in the copy number of several detoxification gene families relative to other coleopterans may reflect the low selective pressure on these genes in an insect that spends most of its life feeding internally. Chemoreceptor genes contain elevated numbers of pseudogenes for odorant receptors that also may be related to the recent ontogenetic shift of R. dominica to a diet consisting primarily of stored grains. Analysis of repetitive sequences will further define the evolution of bostrichid beetles compared to other species. The data overall contribute significantly to coleopteran genetic research.

Tribolium castaneum has a long history as a model species in many distinct subject areas, but improved connections among the genetics, genomics, behavioral, ecological, and pest management fields are needed to fully realize this species' potential as a model. Tribolium castaneum was the first beetle whose genome was sequenced, and a new genome assembly and enhanced annotation, combined with readily available genomic research tools, have facilitated its increased use in a wide range of functional genomics research. Research into T. castaneum 's sensory systems, response to pheromones and kairomones, and patterns of movement and landscape utilization has improved our understanding of behavioral and ecological processes. Tribolium castaneum has also been a model in the development of pest monitoring and management tactics, including evaluation of insecticide resistance mechanisms. Application of functional genomics approaches to behavioral, ecological, and pest management research is in its infancy but offers a powerful tool that can link mechanism with function and facilitate exploitation of these relationships to better manage this important food pest.

Preventing insect infestations is a critical component for establishing a pest management plan for stored-product insects. Long-lasting insecticide-treated netting (LLIN) is a potential tool to reduce insect movement by providing a chemical barrier, where insects may be able to physically pass through but ultimately die after exposure to the netting. Sublethal effects, such as reduced movement immediately after exposure and reduced ability to colonize, have been reported. Here we examine the sublethal effects of exposure to LLIN on two beetle species, Trogoderma variabile Ballion, warehouse beetle, and Tribolium castaneum Herbst, red flour beetle. We found that both female and male T. castaneum exposed to LLIN produced significantly less adult progeny than those exposed to untreated netting. Adult progeny output did not differ for T. variabile, but survivorship increased in T. variabile females exposed to LLIN. Importantly, the overall net reproductive rate was significantly decreased for both T. variabile and T. castaneum. The number of copulation attempts did not differ between males or females exposed to LLIN compared to untreated netting, but males exposed to LLIN showed increased durations of attempted and successful copulation events. Background: Prevention is the first line of defense in mitigating losses of post-harvest crops. Long-lasting insecticide treated (LLIN) could be used in food facilities to expose insects to insecticide at different areas within a facility. Prior research has shown that single short exposures reduce movement and longer exposures increase mortality for stored-product insect pests, but we do not know how multiple short duration exposures and biotic and abiotic conditions affect insects exposed to LLIN. Here, we repeatedly exposed red flour beetles, Tribolium castaneum, to LLIN to assess the cumulative effects. We also examined the effects of beetle age and time of day during exposure, and temperature, humidity and food availability during recovery after a single exposure to LLIN., Results: We found that four repeated 10-min exposures had similar knockdown effects as a single 30-min exposure. We also found that beetles were more affected when aged 1-6 days versus 14-20 days or were exposed at mid- or late in the day versus earlier in the day. Higher recovery levels were observed with food and at higher relative humidity. In addition, older beetles were more active than younger beetles during exposure, which could reduce time in contact with netting and partially explain why older beetles tended to be less affected., Conclusion: Some individuals can recover after exposure to LLIN, dependent on exposure duration and environmental factors, but our study shows that sublethal effects likely persist and future work should consider the physiology of T. castaneum before, during, and after exposure to LLIN. Background: Phosphine-susceptible or resistant populations of Tribolium castaneum (Herbst) (Coleoptera: Tenebrionidae) and Rhyzopertha dominica (F.) (Coleoptera: Bostrychidae) adults were exposed to 0 (control), 1000 and 3000 ppm of phosphine for 15 or 90 min, to estimate behavioral and mobility responses after exposure to phosphine. Knockdown of the exposed individuals after exposure was recorded visually. The total distance moved and velocity of movement were assessed immediately after exposure to phosphine, 2 or 24 h later using a camera coupled with automated video tracking software (i.e. Ethovision®)., Results: For both species tested, the highest percentage of dead adults was noted at the highest concentration (3000 ppm) for both exposure times. For T. castaneum, total distance moved and velocity decreased as the concentration increased for the susceptible population, whereas there was significant variation among individuals in the resistant population. For R. dominica, the distance moved was reduced at the highest concentrations. Individuals of R. dominica moved less than those of T. castaneum and there were significant differences in mobility between susceptible and resistant populations for both species tested. Recovery was much faster in the case of the resistant populations., Conclusions: Changes in movement parameters can be further exploited in assessing the efficacy of different management tactics, such as trapping and sampling. Automated video tracking systems such as Ethovision® can be used to track and record insect behavioral response, providing a more objective measure of insecticide efficacy than visual categorizations. Tribolium castaneum (Herbst) (Coleoptera: Tenebrionidae) and Trogoderma variabile Ballion (Coleoptera: Dermestidae) are two stored-product insects that cause extensive damage to a variety of postharvest commodities. Long-lasting insecticide-incorporated netting (LLIN), commonly used to control vector-borne diseases in tropical regions, has only been recently studied in an agricultural setting. While prior research showed that LLIN was successful against stored-product beetles, little is known about differential susceptibility among stored-product insect life stages. The aim of this study was to evaluate LLIN efficacy against immature T. castaneum and T. variabile compared with adults. Movement and dispersal ability were evaluated after exposure to LLIN or an untreated, control netting. For the movement assay, video-tracking software recorded the postexposure effects of LLIN on distance traveled and velocity of the insects in 2-h trials. For the dispersal assay, insects were exposed to the netting then released into one end of a PVC pipe and allowed 48 h to disperse to a novel food patch located at the opposite end of the pipe. Our study found that movement and dispersal ability of T. variabile and T. castaneum are significantly reduced, often by multiple-fold, after LLIN exposure, with the larval stage of each species more tolerant to the insecticide netting than adults. Aerosol insecticides are one tool that pest management professionals can utilize as a spatial treatment inside food facilities and storage warehouses. Methods of aerosol application can vary significantly and can affect the spatial pattern of efficacy achieved. We investigated how the location from which an aerosol insecticide is applied inside a mill influenced the spatial dispersal of the insecticide. Treatments were performed using two commercial formulations, pyrethrin + pyriproxyfen (insect growth regulator [IGR]) and pyrethrin + methoprene (IGR), applied at one of three static locations or a fourth application comprising of splitting the application among all three locations. Concrete arenas were placed out at different locations within the mill during applications. At 2, 4, and 6 wk post-aerosol application, Tribolium confusum Jacquelin du Val, confused flour beetle, larvae were added to the concrete arenas and monitored for development and efficacy was evaluated based on percent adult emergence and an efficacy index that ranged from 1 (low) to 21 (high). The spatial pattern of aerosol coverage varied between insecticide formulations and the aerosol application location. Areas of the mill near walls, corners, equipment, and farthest away from the application location had larger zones of low efficacy index values among all four application locations. This study illustrated that the aerosol insecticide formulation, application location, and delivery method all significantly influenced residual efficacies of the insecticides. To increase the overall spatial coverage and IGR efficacy, targeting these areas of a mill floor with the aerosol or additional intervention techniques would increase uniform coverages and overall effectiveness., (Published by Oxford University Press on behalf of Entomological Society of America 2020.)

Extreme temperature has been used as an alternative to chemical treatments for stored product pests for years. Resistance to heat or cold treatments has not been documented in stored product insects, but repeated use of ineffective treatments could lead to adaptive tolerance. Trogoderma variabile (Dermestidae) is a common pest of stored products, and the larval stage is highly resistant to cold and destructive. We artificially selected populations by inducing chill coma at four different cold temperature treatments: 3 and 5 h at -10°C and 3 and 5 h at 0°C. Recovery time was highly heritable after selection for seven generations for decreased recovery time (cold tolerance) and increased recovery time (cold susceptibility) at all time and temperature combinations. Three replicate populations for each time and temperature combination varied substantially, suggesting different mutations in each population were probably responsible for selected phenotypes. Body size decreased in populations selected for cold susceptibility compared with those selected for cold tolerance and survivorship to long-term cold stress increased in the cold-tolerant populations compared with the susceptible populations. After the cessation of the selection experiment, cold tolerance dissipated within four generations from the populations at -10°C, but was maintained in populations exposed to 0°C. Our results suggest that warehouse beetles can adapt to cold stress quickly, but in the absence of cold stress, the proportion of cold-tolerant/susceptible individuals is quickly reduced, suggesting that some of the mutations responsible for these phenotypes may be associated with fitness costs under normal conditions., (Published by Oxford University Press on behalf of Entomological Society of America 2020.)

The objectives of the study are to understand how naïve beetles disperse after emerging as an adult in a homogeneous resource patch. We compared the movement of adult male and female Rhyzopertha dominica (F.) (Coleoptera: Bostrychidae) using a laboratory and a field-collected strain during the first 2 d after their emergence from the wheat kernel in which they developed. We first asked if naïve male and female beetles show any innate orientation pattern. Males showed an upward orientation bias during the first day, but not in the second, whereas females had a random pattern of orientation in both days of evaluation. No significant differences were observed between the two strains. Given that males release an aggregation pheromone, we next asked if the upward movement of males improved their ability to be found by a naïve female. The presence of a male, whether above or below a newly emerged female, changed the females' movement direction from random to bias towards the male. In contrast, free-walking males exhibited the same upward movement bias on the first day regardless of the position of the caged male. Only on the second day did male movement change to the opposite direction of the caged male. Here, we report differences between males and females' movement orientation strategies and their response to males producing aggregation pheromone within the grain mass. Filter cake and Triplex are powdered by-products of aluminum sulfate and soap factories in Ethiopia, respectively. This study was aimed at determining contact toxicity of filter cake and Triplex powders against maize weevil, Sitophilus zeamais Motschulsky. Lethal concentrations for 99% mortality (LC99) against S. zeamais were determined by exposing adults for 12 h to filter cake (0.5-8 g/m2) and Triplex (1-9 g/m2) in concrete arenas. Lethal times for 99% mortality (LT99) were determined by exposing adults over time (1-24 h) in concrete arenas to 3 g/m2 of filter cake and 9 g/m2 of Triplex. Exposed adults were transferred to containers with 30 g of organic wheat and held at 28°C and 65% RH for 14 d to determine mortality. LC99 values for S. zeamais adults were 7.54 and 23.46 g/m2 when exposed to filter cake and Triplex, respectively. The corresponding LT99 values were 21.92 and 39.62 h when exposed to filter cake and Triplex, respectively. Effective concentrations and times for the 99% reduction of progeny production were determined from percentage reduction in adult progeny relative to production in control treatments after 42 d. EC99 values for progeny reduction were 2.48 and 18.59 g/m2 for filter cake and Triplex treatments, respectively. The corresponding ET99 values for progeny reduction were 17.49 and 22.31 h for filter cake and Triplex, respectively. Sitophilus zeamais exposed to filter cake produced lower percentage insect-damaged kernels and weight loss than Triplex. Population genetic structure of agricultural pests can be impacted not only by geographic distance and the broader ecological and physical barriers but also by patterns related to where crops are produced and how they are moved after harvest. Stored-product pests, for instance, specialize in exploiting grains such as wheat and rice from on-farm storage through transportation to final processing at often geographically distant locations; therefore human-aided movement may impact their dispersal. Although stored product insects are associated with stored grain, they can also exploit resources in the surrounding environments so different ecological regions where the grain is grown and stored may also influence population structure. Here we used 1,156 SNP markers to investigate how geographic distance, ecological and agricultural variables can impact the genetic structure and gene flow of the stored food pest beetle Rhyzopertha dominica We found a substantial degree of admixture between weakly structured populations in the US. Ecological regions were more important in explaining R. dominica population structure than crop type, suggesting insect movement between wheat and rice grain distribution channels. We have also found a significant correlation between the genetic and geographical distance ( i.e. , isolation by distance). However, our modeling approach combining the ecological and management variables has highlighted the importance of the volume of grain received by a location in the dispersal dynamics of the pest. The first-generation migrant analysis offered additional supported to movement over great distances that are likely associated with grain movement. Our data suggest that a multitude of factors play small but significant parts in the movement dynamics of the pest. The beetles can take advantage of the source-sink dynamic of grain movement in the US, but also engage in a high rate of movement at the local scale. Understanding population structure for R. dominica will provide insights into the potential for local processes of adaptation and broader patterns of movement that will impact management programs and the potential for spread of resistance genes., (Copyright © 2019 Cordeiro et al.)

In recent times, research has focused on integrated pest management approaches using nonchemical alternatives such as mating disruption for the control of stored product insect pests. In this study, we examined the effect of imposed delayed mating on the longevity and reproductive performance of the cigarette beetle, Lasioderma serricorne (F.), a serious insect pest of value added grain-based products, tobacco products, and spices. Delayed mating, progressing 0-14 d, was imposed on both sexes simultaneously, on males only, and on females only. Insects were observed daily for longevity, and F1 progeny was recorded 7-10 wk after mating pairs were placed together. Unmated adults lived significantly longer than mated adults, and mated females lived significantly longer than mated males. However, the length of the period of mating delay did not significantly affect the longevity of mated adults. Progeny production decreased with the age of adults at mating. The number of progeny produced when mating delay was imposed on both sexes simultaneously or on females only was significantly lower than when mating delay was imposed on males only. Hence, females were more adversely affected by delayed mating than males. Findings from this study may provide information for the development of mating disruption techniques that can delay mating and may be effective in keeping populations of L. serricorne below levels that would warrant a control action.

The temporal and spatial patterns in flight activity outside of a rice mill were evaluated for the lesser grain borer [Rhyzopertha dominica (F.)], warehouse beetle [Trogoderma variabile Ballion], cigarette beetle [Lasioderma serricorne (F.)], and Indian meal moth [Plodia interpunctella (Hüϋbner)] to determine critical times of year when the mill would be vulnerable to invasion. Insect activity was monitored using pheromone-baited glue traps (N = 99) from June 2008 to October 2010. Traps were placed along exterior walls of all major buildings and along the fence around the perimeter of the facility. Trogoderma variabile was the most abundant species, with flight activity between mid-March and November. No activity of T. variabile was observed during December through March. Rhyzopertha dominica was also abundant, with activity in mid-April through October. A few adult R. dominica were captured in traps during winter months in the first year of study. Trap captures for all four species increased with an increase in temperature and can be described by linear equations. Knowing seasonal patterns in insect activity allows rice facilities to better understand when facilities are most vulnerable to pest activity. In trapping programs prior capture of individuals of the same or different species may influence subsequent attractiveness of the trap. To evaluate this process with stored-product insects, the effect of the presence of dead or alive adults in traps on the behavioral responses of six stored-product insect species, Tribolium confusum Jacquelin du Val, T. castaneum (Herbst) (Coleoptera: Tenebrionidae), Sitophilus oryzae (L.), S. granarius (L.) (Coleoptera: Curculionidae), Oryzaephilus surinamensis (L.) and O. mercator (Fauvel) (Coleoptera: Silvanidae), was studied in choice tests under laboratory conditions. Two series of tests were carried out. In the first series, the choice was between either alive or dead adults of same species and blank (no adults). Overall, dead adults had the strongest influence, increasing the response of S. oryzae, S. granarius, O. surinamensis, and O. mercator and decreasing the response of the two Tribolium species. Presence of alive adults generally did not result in a response different from blank, except for T. castaneum and O. surinamensis that had a reduced response. In the second series of tests, the choice was between alive or dead individuals of the species vs. alive or dead individuals of its relative cogeneric species. For choices between alive individuals, S. oryzae, S. granarius, and O. surinamensis showed a preference for alive individuals of the same species vs. the cogeneric species, and for all the other species there was no preference. For choices between dead individuals, T. castaneum exhibited a preference for individuals of the same species, while S. oryzae, S. granarius, and O. mercator showed a preference for individuals of the cogeneric species, and all other combinations did exhibit a preference. Grain stored in bins is initially a relatively homogenous resource patch for stored-product insects, but over time, spatial pattern in insect distribution can form, due in part to insect movement patterns. However, the factors that influence stored-product insect movement patterns in grain are not well-understood. This research focused on the movement of the lesser grain borer, Rhyzopertha dominica (F.), within a simulated wheat grain mass (vertical monolayer of wheat) and the identification of factors that contribute to overall and upward movement (age since adult emergence from an infested kernel [1, 7, and 14 d], sex, strain, and different levels of environment quality). We also used the model selection approach to select the most relevant factors and determine the relationships among them. Three-week-old adults tended to stay closer to the surface compared with 1- or 2-wk-old insects. Also, females tended to be more active and to explore a larger area compared with males. Explored area and daily displacement were also significantly strain-dependent, and increasing grain infestation level decreased daily displacement and explored area. Variation in movement pattern is likely to influence the formation of spatial pattern and affect probability to disperse. Cannibalism is a very important factor regulating population dynamics of the red flour beetle. After several days of feeding, the flour becomes conditioned by the beetles, which can affect rates of cannibalism. Flour conditioning is caused by an accumulation of feces, pheromones, and ethylquinone, which is a repellent produced by the beetles. We determined the effect of five different levels of flour conditioning on cannibalism of red flour beetle eggs and pupae by adult and larval stages. Larvae had the highest rates of egg cannibalism (12 eggs eaten over the 4-d period) followed by female adults (seven eggs consumed). Adult males had the lowest rates of cannibalism with only two eggs consumed. Cannibalism of eggs by females was correlated negatively with the level of flour conditioning. There was no effect of flour conditioning on egg or pupal cannibalism by larvae or adult males. Cannibalism by adult females may decrease as the level of flour conditioning increases because females may spend less time tunneling in highly conditioned flour and more time trying to disperse to other areas that are better for oviposition.

Experiments were conducted in tightly sealed pilot scale warehouses to assess the efficacy of common aerosolized insecticides on all life stages of Tribolium castaneum (Herbst) (Coleoptera: Tenebrionidae) when exposed in dishes containing 0 to 2 g of wheat flour either under pallets or out in the open. Petri dishes containing 0, 0.1, 1, or 2 g of flour were prepared with 25 eggs, 3rd instars, pupae, or adults and then immediately treated with aerosolized solvent, Pyrethrins, or esfenvalerate. Twenty-four h after insecticide exposure, the dishes were brought to the laboratory and placed in a growth chamber and held for a 3 day moribund (knockdown) assessment and a 21 day mortality assessment. Mortality in untreated controls was generally less than 10%, with the exception of the 21 day counts of adults and eggs. Solvent-treated replications followed similar trends, except that additional mortality was observed in exposed larvae and pupae. In the insecticide-treated dishes, mortality of T. castaneum provisioned with flour generally showed a linear decrease with increasing flour deposits. Regardless of life stage, mortality did not exceed 60% when individuals were exposed in petri dishes containing 2 g of flour. Exposure location also made a significant difference in observed mortality. While mortality never exceeded 75% in dishes positioned under pallets, there was never less than 80% mortality in dishes exposed in the open. Although there was a perceptible increase in mortality with esfenvalerate compared to Pyrethrins, these differences were considerably less than the variation observed among flour deposits. The study suggests that sanitation and preparation prior to aerosol insecticide treatments were more important than choice of a particular insecticide.

Milling wheat, Triticum aestivum L., infested with low densities of internal feeding insects can result in flour containing insect fragments. The Food and Drug Administration (FDA) enforces a standard or defect action level stating that a maximum of 75 insect fragments per 50 g of flour is allowed. However, the relationship between level of infestation and number of resulting fragments is not well documented, and a more rapid method for enumerating insect fragments is needed. We characterized the number of insect fragments produced from milling small lots of wheat spiked with known densities and life stages of Sitophilus oryzae (L.) (Coleoptera: Curculionidae). Insect fragments were enumerated with near-infrared spectroscopy (NIRS), a quick nondestructive procedure, and with the industry standard flotation method. Results showed that an individual small larva, large larva, pupa, or adult produced 0.4, 0.7, 1.5, and 27.0 fragments, respectively. NIRS-predicted counts of < or =51 (from small larvae), < or =53 (from large larvae), < or =43 (from pupae), or 0 (from adults) indicated that there were <75 actual fragments in that sample, because the upper bound of associated 95% inverse prediction confidence intervals was less than the standard; NIRS-predicted counts of > or =98, > or =117, 108, or > or =225 fragments (same life stages as above) signaled that these flour samples contained >75 actual fragments. These data suggest that NIRS could be adopted for rapid assessment of insect fragments resulting from relatively low levels of infestation with immature life states, but that it was not accurate enough for enumerating insect fragments, relevant to FDA standards, resulting from adults.

Computed tomography, an imaging technique commonly used for diagnosing internal human health ailments, uses multiple x-rays and sophisticated software to recreate a cross-sectional representation of a subject. The use of this technique to image hard red winter wheat, Triticum aestivm L., samples infested with pupae of Sitophilus oryzae (L.) was investigated. A software program was developed to rapidly recognize and quantify the infested kernels. Samples were imaged in a 7.6-cm (o.d.) plastic tube containing 0, 50, or 100 infested kernels per kg of wheat. Interkernel spaces were filled with corn oil so as to increase the contrast between voids inside kernels and voids among kernels. Automated image processing, using a custom C language software program, was conducted separately on each 100 g portion of the prepared samples. The average detection accuracy in the five infested kernels per 100-g samples was 94.4 +/- 7.3% (mean +/- SD, n = 10), whereas the average detection accuracy in the 10 infested kernels per 100-g sample was 87.3 +/- 7.9% (n = 10). Detection accuracy in the 10 infested kernels per 100-g samples was slightly less than the five infested kernels per 100-g samples because of some infested kernels overlapping with each other or air bubbles in the oil. A mean of 1.2 +/- 0.9 (n = 10) bubbles (per tube) was incorrectly classed as infested kernels in replicates containing no infested kernels. In light of these positive results, future studies should be conducted using additional grains, insect species, and life stages.

Spatial analysis of insect counts provides important information about how insect species respond to the heterogeneity of a given sampling space. Contour mapping is widely used to visualize spatial pest distribution patterns in anthropogenic environments, and in this study we outlined recommendations regarding semivariogram analysis of small data sets (N < 50). Second, we examined how contour maps based upon linear kriging were affected by the spatial structure of the given data set, as error estimation of contour maps appears to have received little attention in the entomological domain. We used weekly trap catches of the warehouse beetle, Trogoderma variabile, and the accuracy assessment was based upon data sets that had either a random spatial structure or were characterized by asymptotic spatial dependence. Asymptotic spatial dependence (typically described with a semivariogram analysis) means that trap catches at locations close to each other are more similar than trap catches at locations further apart. Trap catches were poorly predicted for data sets with a random spatial structure, while there was a significant correlation between observed and predicted trap catches for the spatially rearranged data sets. Therefore, for data sets with a random spatial structure we recommend visualization of the insect counts as scale-sized dots rather than as contour maps.

The red flour beetle, Tribolium castaneum ( Herbst) ( Coleoptera: Tenebrionidae), is an important insect pest in food processing facilities. Pheromone trapping is frequently used to monitor red flour beetle populations in structures; however, the optimal trap density and the relationship between trap captures and beetle density is not known. Two experiments were performed concurrently in environmentally controlled 30-m2 walk-in chambers to determine the relationship between aggregation pheromone trap captures of red flour beetles and beetle and trap number. In one experiment, beetle density was kept constant at 200 individuals per chamber while trap number was varied from 1 to 8, and in the other experiment trap number remained constant at one per chamber while beetle density varied from 20 to 800 individuals. Results indicated that approximately one out of 23 red flour beetles were captured in a trap. Number of beetles captured in traps increased significantly as beetle density increased; however, the proportion of beetles captured remained consistent across beetle densities with a mean of 4.7 ± 0.6% of individuals captured. Trap captures varied significantly with trap placement within experimental chambers, indicating that subtle differences in the trapping environment can influence trap captures. Data suggested that trap densities of 0.07-0.10 m−2 (2-3 traps per chamber) would maximize trap capture, whereas a trap density of 0.13 m−2 (four traps per chamber) would maximize the predictive ability of a trapping equation estimating beetle density from trap captures. Results provide information needed to more thoroughly explore how environmental factors might influence red flour beetle trap capture in the absence of changes in beetle density. Further understanding of these relationships will allow for more accurate assessments of absolute beetle density from pheromone trap capture data. [ABSTRACT FROM AUTHOR]

The flight activity of lesser grain borer, Rhyzopertha dominica F. (Coleoptera: Bostrichidae), was monitored at two Foundation seed wheat warehouses during the 2003 and 2004 field seasons, using pheromone-baited Lindgren funnel traps positioned indoors and outdoors. General stored-product insect activity was also monitored using unbaited sticky traps positioned inside the warehouses around overhead doors. Pheromone-baited traps were useful for monitoring R. dominica activity, however insect captures decreased when lures were not changed weekly. Flight peaks were documented in early May and again from September through October, and insect captures inside warehouses correlated with timing of outdoor captures. Multiple regression analyses showed that slightly more than half of the variability in R. dominica captures could be explained by mean ambient air temperature and wind speed during the 2 h preceding sunset. Stored-product Coleoptera captured on unbaited glue boards around overhead doors included Ahasverus advena, Cryptolestes ferrugineus, R. dominica, Sitophilus oryzae, Tribolium castaneum, Trogoderma variabile, and Typhaea stercorea. Door gaskets significantly reduced the number of insect captures on glue boards placed around the overhead doors, and generally restricted their entry to ground level. These studies demonstrated that outdoor pheromone-baited traps are effective monitoring tools for determining when grain-handling facilities are most susceptible to infestation and that exclusion may be an effective component of a pest management program. In a wheat flour mill, seasonal trends in stored-product insect trap capture, relationships between trap captures inside and outside the mill, and between pheromone trap capture and product infestation, and the impact of fumigation on pest populations, were assessed. Mark-recapture was used to evaluate the potential for movement of insects outside the mill into the mill. For Plodia interpunctellu (Hübner) (Lepidoptera: Pyralidae) and Trogoderma variabile Ballion (Coleoptera: Dermestidae), pheromone trap captures outside were higher than inside the mill, and when inside and outside trap captures were correlated, both indoor and outdoor trap captures tended to cycle according to a seasonal pattern; fumigations did not consistently influence pheromone trap captures, and in only one instance were they found in product samples. Mark-recapture data indicated that P.. interpunctella was capable of entering the building from outside. Tribolium castaneum (Herbst) (Coleoptera: Tenebrionidae) trap captures, in contrast, tended to be lower outside compared to inside, followed a pattern of sharp decline after fumigation treatment, and then steadily increased (0.002–0.005 beetles/trap/day) until the next fumigation. This pattern, other than potentially the rate of increase, was not impacted by mason and outside trap capture levels. Tribolium castaneum was the primary species infesting the product. The information generated in this study provides some of the information needed to develop improved monitoring and management programs. [ABSTRACT FROM AUTHOR]
