Orychophragmus violaceus (OV) and chicory ( Cichorium intybus L., CC) can be used as fresh or dry forage for animals. To determine whether OV and/or CC have beneficial effects on performance and egg quality, a total of 1212 28-wk-old Beijing You Chicken (BYC) laying hens with similar performance were randomly allocated to 4 groups with 3 replicate pens per group, and 101 birds per pen. The birds were fed a basal diet (control), the basal diet + OV (3.507 kg/d/pen), the basal diet + CC (2.525 kg/d/pen), and the basal diet + OV + CC (OVC, 1.7535 kg/d/pen OV + 1.2625 kg/d/pen CC) for 3 wks after one wk of adaptation. The results showed that egg-laying rate was not affected by OV, CC and OVC ( p  > 0.05), but weekly average egg mass was significantly increased by OV and CC ( p  < 0.05). The feed egg ratio in the CC group (2.82) was significantly lower than that in the other three groups ( p  < 0.05). The eggshell thickness (EST), albumen height (AH) and Haugh unit (HU) were decreased by OV and CC ( p  < 0.05); while yolk color (YC) was increased in the CC and OVC groups ( p  < 0.05). Egg grade was decreased by OV ( p  < 0.05). Sensory evaluation showed that there was a trend for increased YC in OV, CC and OVC ( p  = 0.089). Serum total protein was significantly lower in OV group than those in the control and CC group ( p  < 0.05); serum albumin content was significantly decreased in OV, CC and OVC groups ( p  = 0.006). Serum glutathione peroxidase activity in CC and OVC groups was significantly higher than that in the control group ( p  < 0.05). In conclusion, the present study suggests that CC had a better effect on the performance of the native laying hens than OV. The OV and CC affected egg quality, while YC was increased in CC and OVC groups. The OVC improved YC and serum antioxidative properties of native laying hens without affecting the performance.

Chicory (Cichorium intybus L.; witloof) is a crisp bitter leafy vegetable, popularly used in western cuisine in salads and soups (leaves) and as an alternative to coffee (roasted roots). In this study, we explored the effect of heat processing under various temperatures and for different durations on the nutritional composition of chicory leaves using gas chromatography-mass spectrometry (GC/MS) and principal component analysis (PCA). "Vintor" chicory leaves were processed and homogenized to obtain lyophilized samples, and their moisture content and pH were measured. Heat processing was conducted at 4, 30, 60, and 100°C. Metabolites were extracted and analyzed using GC/MS. The results were statistically analyzed using multiple t-tests and Tukey-Kramer method. A PCA was conducted using standardized data. A lower temperature (≤60°C) positively influenced the concentrations of nutritional components (sugars, free amino acids, and organic acids), branched-chain amino acids (which reportedly improve exercise performance), and γ-aminobutyric acid (which exerts antihypertensive effects). Whereas, a higher temperature (100°C) and microwave processing induced the generation of low-molecular-weight sugars from polysaccharides and glycosides, decreased free amino acid concentrations, and caused heat-induced aminocarbonyl reactions. This study provides valuable information for enhancing the flavor profiles and potential health benefits of chicory leaves by identifying the optimal heat processing parameters for preserving the desired nutritional value. PRACTICAL APPLICATION: The palatability, nutritional content, and health benefits of chicory have been evaluated based on its inherent constituents, but changes in these parameters during food processing remain unclear. Heating at 30 and 60°C activated secondary metabolism in chicory, increasing the amino acid and organic acid concentrations, whereas heating at 100°C and microwave processing increased the sugar concentrations in chicory. Purslane (P), chard (CHA), and chicory (CHI) leaf extracts are individually and traditionally used in the treatment of diabetes mellitus. Polyphenols, flavonoids, the polyphenolic profile of the extracts, and their antioxidant activity were determined. This study evaluated the antidiabetic activity of combinations of these extracts in streptozotocin-induced diabetic rats. Diabetic groups were administered orally and daily for 40 days with the investigated extracts at 250 mg/kg body weight (b.w.) or metformin (100 mg/kg b.w.) as a drug. Fasting blood glucose, oral glucose tolerance, insulin, and fructosamine were assessed. The combined extracts with high levels of P or CHI exerted potent hypoglycemic activity compared with metformin in addition to the restoration of the histopathological changes in the liver and pancreas of diabetic rats to a near-normal state. Therefore, these combined extracts could be developed as natural drugs for diabetes.

Cichorium intybus L. is the most economically important species of its genus and among the most important of the Asteraceae family. In chicory, many linkage maps have been produced, several sets of mapped and unmapped markers have been developed, and dozens of genes linked to traits of agronomic interest have been investigated. This treasure trove of information, properly cataloged and organized, is of pivotal importance for the development of superior commercial products with valuable agronomic potential in terms of yield and quality, including reduced bitter taste and increased inulin production, as well as resistance or tolerance to pathogens and resilience to environmental stresses. For this reason, a systematic review was conducted based on the scientific literature published in chicory during 1980-2023. Based on the results obtained from the meta-analysis, we created two consensus maps capable of supporting marker-assisted breeding (MAB) and marker-assisted selection (MAS) programs. By taking advantage of the recently released genome of C. intybus , we built a 639 molecular marker-based consensus map collecting all the available mapped and unmapped SNP and SSR loci available for this species. In the following section, after summarizing and discussing all the genes investigated in chicory and related to traits of interest such as reproductive barriers, sesquiterpene lactone biosynthesis, inulin metabolism and stress response, we produced a second map encompassing 64 loci that could be useful for MAS purposes. With the advent of omics technologies, molecular data chaos (namely, the situation where the amount of molecular data is so complex and unmanageable that their use becomes challenging) is becoming far from a negligible issue. In this review, we have therefore tried to contribute by standardizing and organizing the molecular data produced thus far in chicory to facilitate the work of breeders.

Identifying suitable plants for phytoremediation of Cd (cadmium) contaminated agricultural soil is critical. In this study, whether chicory (Cichorium intybus L.) qualified as an ideal accumulator for phytoremediation was investigated. The hydroponic and pot experiments showed that Cd concentration in chicory leaves exceeded 100 mg kg -1 (BCF >1, TF >1) with 40 mg kg -1 Cd in pot; No significant effects on chicory growth, leaf protein and physiological and biochemical aspects when treated with ≤ 20 μM or 40 mg kg -1 Cd, because chicory could relieve Cd toxicity by increasing activities of photoprotection mechanisms, the reactive oxygen species scavenging system and concentrations of functional groups in plant tissues. In field experiment, 16.2 and 26.6 t ha -1 of chicory leaves was harvested in winter and summer, respectively. The highest Cd concentration in leaves was close to 25.0 mg kg -1 (BCF >1, TF >1) from the acid soil with 0.980 mg kg -1 Cd. Over 320 g ha -1 Cd was extracted from soil by harvesting chicory leaves both in winter and summer, with 9.24% and 12.9% of theoretical phytoremediation efficiency. Background & Aims: The beneficial effects of Cichorium intybus L., chicory, in patients with non-alcoholic fatty liver disease (NAFLD) are controversial. This review aimed to systematically summarize the evidence on the effects of chicory on liver function and lipid profile in patients with NAFLD., Methods: Online databases of Scopus, Web of Science, PubMed, EMBASE, Cochrane Library, and grey literature were searched for relevant randomized clinical trials. Weighted mean differences (WMD) with 95% confidence intervals (CIs) were used as effect sizes and a random-effects model was used to pool the data. Besides, sensitivity analyses and publication bias analysis were performed., Results: In total, five articles containing 197 patients with NAFLD were included. The study showed that chicory significantly decreased the levels of both aspartate transaminase (WMD: -7.07 U/L, 95%CI: -13.82 to -0.32) and alanine transaminase (WMD: -17.53 U/L, 95%CI: -32.64 to -2.42). However, no significant effects on alkaline phosphatase and gamma-glutamyl transferase levels and the components of the lipid profile were observed with the use of chicory., Conclusions: This meta-analysis showed that chicory supplementation may exert potential hepatoprotective effects in patients with NAFLD. The aim of the study was to explain the effects of sesquiterpene lactones (SLs) from chicory ( Cichorium intybus L.) root extracts as inhibitors of acetylcholinesterase (AChE) at the molecular level and to determine the inhibition of AChE activity by specific SLs (lactucin and lactucopicrin) and different chicory extracts. The obtained SLs-rich extracts were purified by the countercurrent partition chromatography (CPC) technique. AChE inhibitors were analyzed using two models: isothermal titration calorimetry (ITC) and docking simulation. The results of ITC analysis of the enzyme and the ligands' complexation showed strong interactions of SLs as well as extracts from chicory with AChE. In a test of enzyme activity inhibition after introducing acetylcholine into the model system with SL, a stronger ability to inhibit the hydrolysis of the neurotransmitter was observed for lactucopicrin, which is one of the dominant SLs in chicory. The inhibition of enzyme activity was more efficient in the case of extracts, containing different enzyme ligands, exhibiting complementary patterns of binding the AChE active site. The study showed the high potential of using chicory to decrease the symptoms of Alzheimer's disease.

Cadmium (Cd) contamination seriously threatens the soil health and food safety. Combination of amendment and accumulator plant is a green and effective technique to improve phytoremediation of Cd-contaminated alkaline soil. In this study, a potting experiment was conducted to investigate the effect of sulfur on Cd phytoextraction by Cichorium intybus (chicory). Soil chemical and microbial properties were determined to reveal the mechanism of sulfur-assisting Cd phytoremediation by chicory. Soil pH decreased from 7.77 to the lowest 7.30 with sulfur addition (0.6, 0.9 and 1.2 g kg -1 , LS, MS and HS treatment); Electric conductivity, sulfate anion and available cadmium concentration increased gradually with increasing sulfur doses. Cd concentration of shoot and root significantly increased from 1.47 to 4.43 mg kg -1 , 6.15 to 20.16 mg kg -1 by sulfur treatment relative to CK, which were attributed to increased available Cd concentration induced by decreased pH. Sulfur treatments significantly increased the Cd bioconcentration factor by 64.1%, 118.6%, 201.0% for shoot, 76.3%, 145.6% and 227.7% for root under LS, MS and HS relative to CK treatment, respectively (P < 0.05). However, only MS treatment significantly improved the Cd removal efficiency by 82.9% in comparison of CK treatment (P < 0.05). Microbial community diversity measured by 16SrRNA showed that Thiobacillus and Actinobacteria were the key and dominant strains of soil microbial communities after sulfur addition, which played a pivotal role in the process of sulfur oxidation involved in decrease of soil pH and the transformation of Cd forms. Correlation analysis and path analysis by structural equation model indicated that soil sulfate anion and Thiobacillus directly affected Cd removal efficiency by chicory in Cd-contaminated alkaline soil. Production advantages, environmental benefits and increasing parasite resistance are changing the composition of New Zealand pastures. Traditional ryegrass/clover pasture mixes are being replaced by forage herb crops such as lucerne, chicory and plantain that accumulate a higher concentration of contaminants such as cadmium (Cd). To explore the relationship between Cd in forage crops and the Cd concentration accumulated by animals, four-month-old lambs at four farms across the central North Island of New Zealand were grazed on different forage crops (ryegrass, chicory, lucerne and plantain) between weaning and slaughter. Soil and pasture samples, and sequential liver biopsies, were collected and analysed for total Cd. There were significant differences in Cd concentration between the forage crops (chicory > plantain > lucerne > ryegrass) and this ordering was repeated for Cd in liver. There was no exceedance of maximum limits (ML) for Cd in offal set by the EU and NZ/Australia food safety standards authorities for animals of this study, although the highest concentration of Cd in chicory (0.85 mg/kg DW) was considerably lower than has been recorded elsewhere in New Zealand (4.5 mg/kg DW). Provisional Soil Management Values (SMVs) were developed to explore compliance of liver with EU food standards as a function of grazing chicory. For a soil pH of 5, exceedance might occur at a soil cadmium concentration of 0.34 mg/kg. This concentration falls within Tier 0 of the New Zealand Tiered Fertiliser Management System which seeks to ensure soil Cd remains within acceptable limits over the next 100 years and beyond. Increased Cd uptake by fodder crops and its management in these Tier 0 pastoral soils is therefore an emerging issue for pastoral agriculture. Fructan metabolism in bacteria and plants relies on fructosyltransferases and fructanases. Plant fructanases (fructan exohydrolase, FEH) only hydrolyse terminal fructose residues. Levan (β-2,6 linkages) is the most abundant fructan type in bacteria. Dicot fructan accumulators, such as chicory (Cichorium intybus), accumulate inulin (β-2,1 linkages), harbouring several 1-FEH isoforms for their degradation. Here, a novel chicory fructanase with high affinity for levan was characterized, providing evidence that such enzymes widely occur in higher plants. It is adapted to common microbial fructan profiles, but has low affinity towards chicory inulin, in line with a function in trimming of microbial fructans in the extracellular environment. Docking experiments indicate the importance of an N-glycosylation site close to the active site for substrate specificity. Optimal pH and temperature for levan hydrolysis are 5.0 and 43.7 °C, respectively. Docking experiments suggested multiple substrate binding sites and levan-mediated enzyme dimerization, explaining the observed positive cooperativity. Alignments show a single amino acid shift in the position of a conserved DXX(R/K) couple, typical for sucrose binding in cell wall invertases. A possible involvement of plant fructanases in levan trimming is discussed, in line with the emerging 'fructan detour' concepts, suggesting that levan oligosaccharides act as signalling entities during plant-microbial interactions., (© The Author(s) 2021. Industrial chicory has been the subject of numerous studies, most of which provide clinical observations on its health effects. Whether it is the roasted root, the flour obtained from the roots or the different classes of molecules that enter into the composition of this plant, understanding the molecular mechanisms of action on the human organism remains incomplete. In this study, we were interested in three molecules or classes of molecules present in chicory root: fructose, chlorogenic acids, and sesquiterpene lactones. We conducted experiments on the murine model and performed a nutrigenomic analysis, a metabolic hormone assay and a gut microbiota analysis, associated with in vitro observations for different responses. We have highlighted a large number of effects of all these classes of molecules that suggest a pro-apoptotic activity, an anti-inflammatory, antimicrobial, antioxidant, hypolipidemic and hypoglycemic effect and also an important role in appetite regulation. A significant prebiotic activity was also identified. Fructose seems to be the most involved in these activities, contributing to approximately 83% of recorded responses, but the other classes of tested molecules have shown a specific role for these different effects, with an estimated contribution of 23-24%.

This study is focused on the evaluation of the accumulation of Cd, Pb and Zn in five cultivars of Cichorium intybus L. (chicory) which were produced on contaminated agricultural soil. Over a growth period of 211 days, the roots and leaves were collected in four stages and then analyzed by flame atomic absorption spectrometry to measure the concentration and distribution of the target metals in these two chicory organs considering the weather and the nature of the cultivar. For all cultivars, sharp decreases of Pb (from 165 mg kg -1 to 3 mg kg -1 ), Cd (from 11 mg kg -1 to 5 mg kg -1 ) and Zn concentrations (from 157 mg kg -1 to 40 mg kg -1 ) in the roots were highlighted over time. The data collected enabled the calculation of the variation of the bioconcentration factor, the biological absorption coefficient and the translocation factor for Cd, Pb and Zn. These parameters were then correlated with the distribution of the fresh biomass of leaves and roots and several indicators such as chlorophyll content, flavonols, anthocyanin and nitrogen balance index were measured. The study concludes with the discussion on the ability of chicory to clean up contaminated agricultural soil. The current investigation has shown: i) a translocation of Cd (and Zn to a lesser extent) from the roots to the leaves; ii) an increase in the level of anthocyanins with the increase of the metal trace elements concentration in the leaf, while the content of chlorophyll and the nitrogen balance index decrease, which could be linked to the phenomenon of senescence; iii) an ability of the chicory to reduce the bioavailable pool of the three metal trace elements studied, in particular for Cd., (Copyright © 2021 Elsevier Ltd. Background: Hepatocellular carcinoma (HCC) is one of the most common cancers, associated with a high rate of mortality. A disturbance between cell proliferation and cell death is one of the cancer hallmarks including HCC. Cell proliferation is mainly controlled by the cell cycle. The arrest of the cell cycle is one of the important targets of anticancer agents., Objectives: The present study tries to clarify the exact role of some natural products such as daidzein (DAZ) and alcoholic chicory leaf extract (CE), as possible regulators of cell cycle and apoptosis., Methods: HCC in rats was induced using diethylnitrosamine (DENA). Ninety rats were allocated and divided equally into nine groups, treated with CE, DAZ, a combination of both, and sorafenib with non-treated control groups., Results: Treatment with CE, DAZ, and their combination significantly downregulated hepatic tissue expression of cyclin D1/CDK4 axis as well as cyclin A/CDK2 axis. The suggested therapeutic protocol inhibited the proliferation and dampened Bcl-2 expression. Chicory (Cichorium intybus var. sativum) is an industrial crop species cultivated for the production of a fructose polymer inulin, which is used as a low-calorie sweetener and prebiotic. Besides, inulin chicory taproots also accumulate sesquiterpene lactones (STLs). These are bitter tasting compounds, which need to be removed during inulin extraction, resulting in additional costs. In this work, we describe chicory lines where STL accumulation is almost completely eliminated. Genome editing using the CRISPR/Cas9 system was used to inactivate four genes that encode the enzyme that performs the first dedicated step in STL synthesis, germacrene A synthase (CiGAS). Chicory lines were obtained that carried null mutations in all four CiGAS genes. Lines lacking functional CiGAS alleles showed a normal phenotype upon greenhouse cultivation and show nearly complete elimination of the STL synthesis in the roots. It was shown that the reduction in STLs could be attributed to mutations in genetically linked copies of the CiGAS-short gene and not the CiGAS-long gene, which is relevant for breeding the trait into other cultivars. The inactivation of the STL biosynthesis pathway led to increase in phenolic compounds as well as accumulation of squalene in the chicory taproot, presumably due to increased availability of farnesyl pyrophosphate (FFP). These results demonstrate that STLs are not essential for chicory growth and that the inhibition of the STL biosynthesis pathway reduced the STL levels chicory which will facilitate inulin extraction., (© 2021 The Authors. Increasing resistance towards anthelmintic drugs has necessitated the search for alternative treatments for the control of gastrointestinal nematode parasites. Animals fed on chicory (Cichorium intybus L.), a temperate (pasture) crop, have reduced parasite burdens, hence making C. intybus a potentially useful source for novel anthelmintic compounds or a diet-based preventive/therapeutic option. Here, we utilized in vitro bioassays with the parasitic nematode Ascaris suum and molecular networking techniques with five chicory cultivars to identify putative active compounds. Network analysis predicted sesquiterpene lactones (SL) as the most likely group of anthelmintic compounds. Further bioassay-guided fractionation supported these predictions, and isolation of pure compounds demonstrated that the SL 8-deoxylactucin (8-DOL) is the compound most strongly associated with anti-parasitic activity. Furthermore, we showed that 8-DOL acts in a synergistic combination with other SL to exert the anti-parasitic effects. Finally, we established that chicory-derived extracts also showed activity against two ruminant nematodes (Teladorsagia circumcincta and Cooperia oncophora) in in vitro assays. Collectively, our results confirm the anti-parasitic activity of chicory against a range of nematodes, and pave the way for targeted extraction of active compounds or selective breeding of specific cultivars to optimize its future use in human and veterinary medicine., (Copyright © 2021 The Authors. The role of oxidative stress in female fertility is a compelling area for research. According to traditional medicine, Cichorium intybus, known as Kasni, is believed to improve fertility. For this purpose, the effects of C. intybus distillate (CI) on blood antioxidant status were assessed in rats with carbon tetrachloride (CCl4)-induced toxicity. The rats were assigned to four experimental groups of Control, CI, CCl4, and CI+CCl410 (n=10 in each group). The level of antioxidant enzymes, such as glutathione peroxidase (GPx), glutathione reductase (GR), and catalase (CAT), as well as lipid peroxidation and reduced glutathione (GSH) level, were measured in serum samples. In the second part of the study, the antioxidant activity and phytochemical composition of the hydrodistillate of C. intybus aerial parts were determined by DPPH radical scavenging and gas chromatography-mass spectrometry analysis, respectively. The administration of CCl4 decreased the enzyme activities of GPx, GR, and CAT which were significantly ameliorated after CI administration. The decreased level of serum GSH following CCl4 administration was not considerably elevated in the CI+CCl4 group. Furthermore, the level of malondialdehyde in the serum of CI+CCl4 rats was decreased, compared to the CCl4 group. The main compositions of the essential oil from the C. intybus distillate were the antioxidants of Pulegone (8.10%), Piperitenone (7.68%), dihydroactinidiolide (5.0%), and carvone (4.18%). The antioxidant activity of the distillate was obtained at 75&micro;g/l using the DPPH (2,2-diphenyl-1-picryl-hydrazyl-hydrate) test. Chicory (Cichorium intybus L.) is a perennial herb from the Cichorium genus, Asteraceae family, and is worldwide cultivated. So far, chicory has been used mainly in animal feed, but also in several cases in the food industry: as salad, for teas and tea blends, for coffee supplementation, and as a source for the inulin production. Nowadays there is an increasing interest in chicory utilization for food production and supplementation. Some compounds present in chicory, such as polyphenols, inulin, oligofructose and sesquiterpene lactones may be considered as potential carriers of food functionality. This review describes nutritional, mineral and bioactive composition of the chicory plant and summarized the main biological activities associated with the presence of bioactive compounds in the different plant parts. This experiment was conducted to provide a better insight into the plant responses to nitric oxide (NO) and selenium nanoparticle (nSe). Chicory seedlings were sprayed with nSe (0, 4, and 40 mg l -1 ), and/or NO (0 and 25 μM). NO and/or nSe4 improved shoot and root biomass by an average of 32%. The nSe40 adversely influenced shoot and root biomass (mean = 26%), exhibiting moderate toxicity partly relieved by NO. The nSe and NO treatments transcriptionally stimulated the dehydration response element B1A (DREB1A) gene (mean = 29.6-fold). At the transcriptional level, nSe4 or NO moderately upregulated phenylalanine ammonia-lyase (PAL) and hydroxycinnamoyl-CoA quinate transferase (HCT1) genes (mean = sevenfold). The nSe4 + NO, nSe40, and nSe40 + NO groups drastically induced the expression of PAL and HCT1 genes (mean = 30-fold). With a similar trend, hydroxycinnamoyl-CoA Quinate/shikimate hydroxycinnamoyl transferase (HQT1) gene was also upregulated in response to nSe and/or NO (mean = 25-fold). The activities of nitrate reductase and catalase enzymes were also induced in the nSe- and/or NO-treated seedlings. Likewise, the application of these supplements associated with an increase in ascorbate concentration (mean = 31.5%) reduced glutathione (mean = 35%). NO and/or nSe enhanced the PAL activity (mean = 36.4%) and soluble phenols (mean = 40%). The flowering was also influenced by the supplements in dose and compound dependent manner exhibiting the long-time responses. It appears that the nSe-triggered signaling can associate with a plethora of developmental, physiological, and molecular responses at least in part via the fundamental regulatory roles of transcription factors, like DREB1A as one the most significant genes for conferring tolerance in crops.

Background: High-purine diet can cause gut microbiota disorder, which is closely related to the occurrence of hyperuricemia (HUA). At the same time, the development of HUA is often accompanied by renal impairment. Chicory, a natural medicine, has a significant effect on lowering uric acid. However, whether its concrete mechanism is associated with the regulation of gut microbiota and renal damage is still unclear., Methods: Hyperuricemic quails induced by high-purine diet were used, and quails were divided into control (CON), model (MOD), and model plus high, middle, low doses of chicory. The uricosuric effect was evaluated by detecting the uric acid levels in serum and feces. Meanwhile, the morphology of intestine and kidney were observed by hematoxylin and eosin (HE) staining, and the expression of intestinal barrier junction proteins Occludin, Claudin-1 were detected by quantitative real-time polymerase chain reaction (qPCR) and western blotting. Furthermore, the latent mechanism was clarified by analyzing 16S rRNA amplicon of gut microbiota and measuring the changes of LPS/TLR4 axis inflammatory response of the kidney by western blotting and enzyme-linked immunosorbent assay (ELISA)., Results: The results showed that serum uric acid levels were significantly decreased, and the feces uric acid levels were noticeably increased after the intervention of chicory. In addition, chicory could repair the damage of intestinal mucosa and improve the permeability of intestinal barrier. Moreover, the 16S rRNA sequencing analysis uncovered that chicory restored gut microbiota by increasing the probiotics flora (Bifidobacterium, Erysipelotrichaceae) and reducing the pathogenic bacteria group (Helicobacteraceae). Furthermore, it was found that chicory reduced the LPS/TLR4 axis inflammatory response by down regulating the serum LPS and TLR4/NF-κB inflammatory pathway in kidney, thus promoting the excretion of uric acid in kidney., Conclusion: Chicory intervention ameliorated HUA via modulating the imbalance of gut microbiota and suppressing LPS/TLR4 axis inflammatory reaction in quail model, which may be a promising candidate for hyperuricemia-relieving properties., (Copyright © 2020 The Author(s). Background: A frequent problem associated with the tissue culture of Compositae species such as chicory (Cichorium intybus L.) and lettuce (Lactuca sativa L.) is the premature bolting to in vitro flowering of regenerated plants. Plants exhibiting such phase changes have poor survival and poor seed set upon transfer from tissue culture to greenhouse conditions. This can result in the loss of valuable plant lines following applications of cell and tissue culture for genetic manipulation., Results: This study demonstrates that chicory and lettuce plants exhibiting stable in vitro flowering can be rejuvenated by a further cycle of adventitious shoot regeneration from cauline leaves. The resulting rejuvenated plants exhibit substantially improved performance following transfer to greenhouse conditions, with increased frequency of plant survival, a doubling of the frequency of plants that flowered, and substantially increased seed production., Conclusion: As soon as in vitro flowering is observed in unique highly-valued chicory and lettuce lines, a further cycle of adventitious shoot regeneration from cauline leaves should be implemented to induce rejuvenation. This re-establishes a juvenile phase accompanied by in vitro rosette formation, resulting in substantially improved survival, flowering and seed set in a greenhouse, thereby ensuring the recovery of future generations from lines genetically manipulated in cell and tissue culture.

There is a lot of evidence that chicory could be a highly palatable and nutritious source of forage for ruminants, well adapted to climate change and dry conditions in summer, thanks to its resistance to drought and high water content. This study aimed to describe the effect of incorporating chicory to ryegrass or to a ryegrass-white clover mixture on feeding behaviour, digestive parameters, nitrogen (N) balance and methane (CH4) emissions in sheep. In total, three swards of ryegrass, white clover and chicory were established and managed in a manner ensuring the forage use at a constant vegetative stage throughout the experiment. In all, four dietary treatments (pure ryegrass; binary mixture: 50% ryegrass-50% chicory; ternary mixture: 50% ryegrass-25% white clover-25% chicory; and pure chicory) were evaluated in a 4×4 replicated Latin square design with eight young castrated Texel sheep. Each experimental period consisted of an 8-day diet adaptation phase, followed by a 6-day measuring phase during which intake dynamics, chewing activity, digestibility, rumen liquid passage rate, fermentation end-products, N balance and CH4 emissions were determined. Data were analysed using a mixed model and orthogonal contrasts were used to detect the potential associative effects between ryegrass and chicory. The daily voluntary dry matter intake was lower for pure ryegrass than for diets containing chicory (P&lt;0.001) and increased quadratically from 1.39 to 1.74 kg/day with increasing proportion of chicory. Huge positive quadratic effects (P&lt;0.001) between ryegrass and chicory were detected on eating time and eating rate just after feeding indicating an increase of the motivation to eat with mixtures, whereas rumination activity decreased linearly with the proportion of chicory (P&lt;0.001). The organic matter digestibility was similar among treatments (around 80%), but a strong positive quadratic P&lt;0.001) effect was observed on liquid passage rate suggesting that chicory allowed fast particle breakdown in the rumen. Animals fed with the ryegrass-white clover-chicory mixture had the higher urinary N losses (P&lt;0.001), whereas retained N per day or per g N intake was greater when the proportion of chicory was at least 50% (P&lt;0.001) being ~40% greater than for the other treatments. The CH4 yield was lower with pure chicory than with the other treatments (P&lt;0.001) for which emissions were similar. In conclusion, mixing ryegrass and chicory in equal proportions produces a synergy on voluntary intake and an improved N use efficiency likely due to complementarity in chemical composition, increased motivation to eat and faster ruminal particle breakdown.

Increasing drug resistance in gastrointestinal (GI) parasites of livestock and concerns about chemical residues in animal products and the environment are driving the development of alternative control strategies that are less reliant on the use of synthetic drugs. An increasingly investigated approach is the use of bioactive forages with antiparasitic properties as part of the animal's diet (nutraceuticals) or as potential sources of novel, natural parasiticides. Chicory (Cichorium intybus) is a multi-purpose crop and one of the most promising bioactive forages in temperate regions, and numerous in vivo trials have explored its potential against parasitic nematodes in livestock. However, it is unclear whether chicory can induce a direct and broad activity against various GI parasites in different livestock species, and the levels of chicory in the diet that are required to exert an efficient antiparasitic effect. Moreover, the mechanisms leading to the reported parasiticidal activity of chicory are still largely unknown, and its bioactive phytochemicals have only recently been investigated. In this review, we summarise the progress in the study of the antiparasitic activity of chicory and its natural bioactive compounds against GI parasites in livestock, through examination of the published literature. The available evidence indicates that feeding chicory can reduce faecal egg counts and/or worm burdens of abomasal nematodes, but not infections with intestinal worms, in ruminants. Highly chicory-rich diets (≥ 70% of chicory dry matter in the diet) may be necessary to directly affect abomasal parasitism. Chicory is known to synthesise several bioactive compounds with potential antiparasitic activity, but most research has been devoted to the role of sesquiterpene lactones (SL). Recent in vitro studies have confirmed direct and potent activity of SL-rich extracts from chicory against different GI helminths of livestock. Chicory SL have also been reported to exhibit antimalarial properties and its potential antiprotozoal activity in livestock remains to be evaluated. Furthermore, the detailed identification of the main antiparasitic metabolites of chicory and their pharmacokinetics need further confirmation. Research gaps and perspectives on the potential use of chicory as a nutraceutical forage and a source of bioactive compounds for parasite control in livestock are discussed.

Biochar is an organic amendment used for soil remediation, there are only a few studies documenting the effects of nitrogen on the role of biochar in contaminated soils. A pot experiment was conducted to investigate the impacts of biochar (0%, 1%, and 2.5%, w/w) and nitrogen (0, 100, and 200 mg N kg -1 ) on plant growth, nutrient and cadmium (Cd) uptake of Cichorium intybus. N, P, Ca, Mg, and Cd concentrations increased with N level in 0% and 1% biochar treatments. In plants treated with 2.5% biochar, 200 mg N kg -1 addition caused significant reductions of N, P, Ca, Mg, and Cd concentrations in comparison to 100 mg N kg -1 treatments. Nitrogen promoted shoot biomass at all biochar treatments, while biochar had no effect on shoot biomass in 0 and 200 mg N kg -1 addition treatments. Nitrogen also significantly increased N, P, K, Ca, Mg, and Cd contents in the 0% and 1.5% biochar addition treatments. Although soil DTPA-extractable Cd concentration showed the lowest values in 1% biochar in combination with 100 and 200 mg N kg -1 addition treatments, lowest shoot Cd concentration, and relatively high shoot biomass occurred in the 2.5% biochar + 200 mg N kg -1 treatment. Based on these results, biochar application at its highest rate (2.5%) in combination with high N supply (200 mg N kg -1 ) contributed to both crop yield and agricultural product safety. N input alone might increase the risk of human health, and the optimum N dose should be determined during phytostabilization process.

Blood and urine biochemistry screening tests are important for initial detection of diabetes, determination of severity of its complications, and monitoring of therapy. We evaluated the effects of aqueous chicory seed extract (CSE), on renal biochemical parameters, histology, and Na+/glucose cotansporters, SGLT1 and SGLT2 expression levels using metformin, and aspirin as controls. Late stage type 2 diabetes (LT2D; FBS, >300 mg/dl) and early stage type 2 diabetes (ET2D; FBS, 140-220 mg/dl) were induced in rats by streptozotocin (STZ group) and a combination of STZ and niacinamide (NIA/STZ group), respectively. A non-diabetic group was included as control. Treatment included daily intraperitoneal injections of either CSE (125 mg/kg b.w.) or metformin (100 mg/kg b.w.) and oral aspirin (120 mg/kg b.w.) for 21 days. At the end, blood and 24 h urine samples were collected; and kidneys were saved at -80 ˚C. CSE reduced urinary α1-microgobulin excretion in ET2D (p = .043), and serum uric acid (p = .045), and glomerular diameter (p < .01) in LT2D. Metformin appeared to be more effective in LT2D with respect to serum uric acid, urea, and BUN (< .05). Both CSE and metformin improved histology. Aspirin improved several blood and urine variables, but appeared to aggravate morphological damages to the kidney tissue. The absolute values of albumin, α1-microglobulin or total protein in urine rather than their creatinine ratios seemed more useful in the detection of early kidney damage; CSE was able to repair the kidney damage and α1-microglobulin was sensitive enough to allow monitoring of the improvements caused by the treatment.

Sixty SD male rats were randomly divided into normal group, model group, benzbromarone group(20 mg•kg⁻¹•d⁻¹), chicory extract high dose, middle dose and low dose groups (5, 7.5, 10 g•kg⁻¹•d⁻¹). The rats in normal group were given with water, and the rats in other groups were given with 10% fructose solution to establish hyperuricemia models. All the rats were sacrificed on the 42th day. Then their serum uric acid(SUA), serum creatinine(CRE), urea nitrogen(BUN) and urinary uric acid(UUA) levels were detected to calculate the clearance rate of uric acid in kidney(CUA). Meanwhile, the protein and gene expression levels of renal glucose transporter family member 9(Glut9) were detected by immunohistochemical and Real-time quantitative reverse transcription-polymerase chain reaction(RT-qPCR) methods. In this study, the changes in phenolics, anthocyanin, antioxidant capacity, and bioaccessibility of chicory varieties (Cichorium spp.) in Turkey were investigated. A total of 19 phenolic standards were screened in the chicory varieties studied and the most abundant compounds in the samples, extracted with methanol, were phenolic acids, syringic (2.54mg/kg) and trans-ferulic acid (1.85mg/kg), whilst (+)-catechin was the major flavanol. The highest flavanol content using either methanol or ethanol was determined in the green chicory samples (0.62mg/kg). The red chicory variety had higher anthocyanin (12.80mg/kg), and contained more phenolics, extractable (8855.50mg GAE/100g) and hydrolysable (7005.51mg GAE/100g), than the other varieties. Also, the antioxidant capacities in this variety, as measured using the CUPRAC assay (570.54 and 425.14μmol Trolox/g dw, respectively), had a wider range of difference than was found in the other assays used. Total phenolics were more bioaccessible from the white chicory variety (61.48%). Compared with 7,8,3'-trihydroxyflavone, a CNT2 inhibitors, 23 score higher chicory compounds were hit.Meanwhile, the ten top compounds have been revealed that play important role in decrease the uric level. Scope: Chicory (Cichorium intybus L. var. foliosum, Belgian endive), a typical Mediterranean vegetable, and its constituent protocatechuic acid (PCA) can inhibit established atherosclerosis progression. We thus investigated whether chicory can improve vascular relaxation, a critical pathway for combating atherosclerosis, and whether PCA is a contributor to a chicory-induced effect., Methods and Results: Apolipoprotein E-deficient (ApoE -/- ) mice with established atherosclerosis and C57BL/6J mice without atherosclerosis were fed an AIN-93G diet, or AIN-93G plus 0.5% freeze-dried chicory or 0.003% PCA for 1 wk. In ApoE -/- mice, both chicory and PCA consumption increased endothelium-dependent vasodilation and endothelial nitric oxide synthase (eNOS) activity independent of eNOS and phospho-eNOS Ser1177 and Thr495 protein expression. Chicory- or PCA-induced eNOS activities were associated with increased vascular tetrahydrobiopterin (BH 4 ) levels that result from reduced BH 4 oxidation partially through preventing eNOS uncoupling. In C57BL/6J mice, neither chicory nor PCA consumption affected endothelium-dependent vasodilation and eNOS activity. Notably, in vitro studies showed that PCA increases eNOS activity in mouse aortic endothelial cells in co-culture with macrophage foam cells, but not in aortic endothelial cells alone., Conclusions: Chicory improves eNOS-mediated endothelium-dependent vasodilation by increasing BH 4 levels in mice with established atherosclerosis, which might be partially ascribed to its constituent PCA., (© 2016 WILEY-VCH Verlag GmbH & Co. Some medications may change trace element levels in the body. Extracts of various plants, due to having the several elements, can have beneficial effects. Consumption of herbal extracts with chemical drugs may reduce adverse effects of medication. The goal of this study was to evaluate copper (Cu), zinc (Zn), and manganese (Mn) concentrations in serum of rats treated with tamoxifen, chicory, and/or milk thistle extracts. Therefore, 36 adult female Wistar rats were divided into six groups: normal control, chicory control, milk thistle control, tamoxifen, tamoxifen-chicory, and tamoxifen-milk thistle. At the end of the study, the blood samples were collected and sera isolated by centrifugation and analyzed by the atomic absorption spectrophotometry for Cu, Zn, and Mn levels. The Zn concentration increased in milk thistle-supplemented groups. The Cu level increased in the chicory control group only. Tamoxifen had no affect on Cu, Zn, and Mn levels, but seed extract of milk thistle increased Zn concentration, and chicory root extract increased Cu concentration. Although elevated levels of Cu in rats receiving tamoxifen-chicory were milder than rats treated only with chicory, it seems that the extract and tamoxifen impact on the Cu are in conflict with each other.

Backgrounds and Aims: Type 2 diabetic mellitus (T2DM) as one of the main causes of morbidity and mortality is associated with immune system disturbances and metabolic abnormalities. In the current study we aimed to evaluate the effects of enriched chicory inulin supplementation on liver enzymes, serum calcium and phosphorous concentrations and hematological parameters in patients with T2DM., Methods: Forty-six diabetic females patients were randomly allocated into intervention (n=27) and control (n=22) groups. Subjects in the intervention group received a daily dose of 10g of chicory and subjects in control group received a placebo for two months. Anthropometric variables, glucose homeostasis, hematological parameters and metabolic indices including serum alanine aminotransfersae (ALT), aspartate aminotransferase (AST), alkaline phosphatase (ALP), calcium and phosphorous as well as creatinine concentrations, glomerular filtration rate (GFR) and blood pressure were assessed at the beginning and end of the trial., Results: Significant reductions in fasting serum glucose (FSG), Hb A1C, AST and ALP concentrations were observed in chicory-treated group. Systolic and diastolic blood pressures were also reduced in chicory-treated group. Serum calcium significantly increased after chicory supplementation but no change in placebo treated group has been occurred (P=0.014). Supplementation with enriched chicory for two months significantly reduced hematocrit and mean corpuscular volume (MCV) values (P<0.05). Changes in serum insulin, creatinine and GFR were not significant., Conclusion: The present study showed beneficial effects of oligofructose-enriched chicory on the improvement of the glucose and calcium homeostasis, liver function tests, blood pressure and reduction in hematologic risk factors of diabetes in female patients with T2DM. Chicory is a perennial crop that has been investigated as a forage source for outdoor-reared ruminants and pigs, and has been reported to have anthelmintic properties. Here, we investigated in vitro anthelmintic effects of forage chicory-extracts against the highly prevalent swine parasites Ascaris suum and Oesophagostomum dentatum. Methanol extracts were prepared and purified from two different cultivars of chicory (Spadona and Puna II). Marked differences were observed between the anthelmintic activity of extracts from the two cultivars. Spadona extracts had potent activity against A. suum third (L3) and fourth (L4) - stage larvae, as well as O. dentatum L4 and adults, whereas Puna II extracts had less activity against A. suum and no activity towards O. dentatum L4. Transmission-electron microscopy of A. suum L4 exposed to Spadona extracts revealed only subtle changes, perhaps indicative of a specific anthelmintic effect rather than generalized toxicity. Ultra-high liquid chromatography-mass spectrometry analysis revealed that the purified extracts were rich in sesquiterpene lactones (SL), and that the SL profile differed significantly between cultivars. This is the first report of anthelmintic activity of forage chicory towards swine nematodes. Our results indicate a significant anthelmintic effect, which may possibly be related to SL composition.

During the heat treatment of coffee and its substitutes some compounds potentially deleterious to health are synthesized by the Maillard reaction. Among these, N(ε)-carboxymethyl-lysine (CML) was detected at high levels in coffee substitutes. The objective of this study was to evaluate the impact of changes in agricultural practice on the lysine content present in chicory roots and try to limit CML formation during roasting. Of the 24 varieties analyzed, small variations in lysine content were observed, 213 ± 8 mg/100 g dry matter (DM). The formation of lysine tested in five commercial varieties was affected by the nitrogen treatment with mean levels of 176 ± 2 mg/100 g DM when no fertilizer was added and 217 ± 7 mg/100 g DM with a nitrogen supply of 120 kg/ha. The lysine content of fresh roots was significantly correlated to the concentration of CML formed in roasted roots (r = 0.51; p < 0.0001; n = 76).

Chicory (Cichorum intybus L.) is a summer-active forage herb which has been proposed as an option to increase summer feed supply, increase dry matter intake, nutrient intake, and milk yield from nonirrigated dairy production systems in southern Australia. Dry matter intake, nutrient intake, milk yield, and yield of milk fat and protein of predominantly Holstein-Friesian dairy cows in late lactation consuming 3 herbage-based diets (4 replicates per treatment) were measured. The 3 grazed herbages were second-year chicory (CHIC) and perennial ryegrass (Lolium perenne L.; PRG) monocultures and a mixed sward (~50:50) of chicory and perennial ryegrass (MIX). All diets (CHIC, PRG, and MIX) were supplemented with alfalfa (Medicago sativa L.) hay (5.5kg of DM/cow per day) and an energy-based concentrate pellet (4.0kg of DM/cow per day). There were no significant differences in milk yield (12.0 to 12.6kg/d across the treatments) or the yield of milk fat (539 to 585g/d) and milk protein (433 to 447g/d) between the 3 herbage-based diets. No differences in DMI (17.9 to 19.2kg/d) or estimated metabolizable energy intake (173 to 185MJ/d) were noted between treatments. Estimated metabolizable energy concentrations in the forages on offer were lower in CHIC than PRG (7.6 vs. 8.2MJ/kg of dry matter), but the concentration in consumed herbage was not different (9.1 vs. 9.2MJ/kg of dry matter); as such, potential for increased milk yield in cows offered CHIC was limited. Increased concentration of polyunsaturated fatty acids was observed in chicory herbage compared with perennial ryegrass. This was associated with increased milk conjugated linoleic acid and milk polyunsaturated fatty acids when chicory formed part of the diet (CHIC compared to PRG and MIX). Chicory could be used as an alternative to perennial ryegrass in summer; however, the developmental stage of chicory will influence concentrations of metabolizable energy and neutral detergent fiber and, therefore, intake and milk production responses compared with perennial ryegrass., (Copyright © 2015 American Dairy Science Association. Bioactive forages contain compounds, such as tannins, that are active against pathogens. They have been successfully used in ruminants to control parasite infections. Because cattle may find bioactive forages unpalatable, it is of interest to know if an afternoon harvest time, which has been shown to increase the percentage of nonstructural carbohydrates (NSC), hence palatability, may mitigate this. The objectives of this study were to quantify voluntary intake and preference of dairy cows for 2 bioactive forages, harvested in the morning and evening, in addition to determining their time spent grazing on each forage species. The forage species evaluated were fresh chicory harvested at 0700 h (FCAM) and 1800 h (FCPM), fresh birdsfoot trefoil harvested at 0700 h (FBAM) and 1800 h (FBPM), birdsfoot trefoil baleage harvested the previous summer at 0700 h (BBAM) and at 1800 h (BBPM), and third-cut alfalfa baleage harvested the previous summer and used as control (CON). Single forages were offered ad libitum in 30-min tests to 14 dairy cows to determine intake in a 7 × 7 Latin square (experiment 1). Every possible pair of forages (21 pairs) was then presented for a 30-min test to 8 different dairy cows, and feed intake was measured (experiment 2). Finally, time spent grazing on chicory and birdsfoot trefoil was measured on 12 dairy cows (experiment 3). The tests consisted of 2 d of restriction on 1 of the 2 fields for 1h, and 2 d of free-choice sessions (1h) between the 2 fields adjacent to each other. Grazing time and location of the animals on the field was assessed through 2-min scan sampling. In experiment 1, the highest voluntary intakes were for CON, BBPM, and BBAM. In experiment 2, BBPM was preferentially consumed over all other forages followed by CON and BBAM. Multidimensional scaling showed that preference for BBPM, CON, and BBAM in dimension 1 was positively associated with dry matter and nitrogen content, and negatively associated with hemicellulose and soluble N/total N. No relationships between dimension coordinates and any of the measured chemical composition variables could be found for the other 2 dimensions. In experiment 3, cows spent 71% of their time grazing in the birdsfoot trefoil field and 23% in the chicory field during the free-choice sessions. In conclusion, cows in the present experiments showed an overall preference toward baled forages compared with fresh forages, most notably toward birdsfoot trefoil baleage. Scope: Since protocatechuic acid exerts an atheroprotective role, we investigated how chicory (Cichorium intybus L. var. foliosum, Belgian endive) rich in protocatechuic acid, a typical vegetable in Mediterranean diet, affects preestablished atherosclerosis progression., Methods and Results: Apolipoprotein E-deficient mice fed AIN diets containing 0.5% freeze-dried chicory for 10 weeks displayed a reduction in lesion size with a concomitant improvement in lesion stability indicated by fewer macrophages and more collagen content. Chicory consumption suppressed aortic cholesterol accumulation and intercellular adhesion molecule-1, vascular cell adhesion molecule-1, and monocyte chemoattractant protein-1 expression, whereas it increased aortic ATP-binding cassette transporter A1 (ABCA1) and G1 (ABCG1) expression. Furthermore, chicory consumption improved peritoneal macrophage phenotype with less cellular cholesterol associated with an enhancement of cholesterol efflux capacity through upregulation of ABCA1 and ABCG1, less cellular oxidative stress associated with an inhibition of nicotinamide adenine dinucleotide phosphate oxidase activity, and weaker inflammatory responses associated with an inhibition of nuclear factor-κB activation. The objective of this study was to explore the antioxidant levels and anticancer properties of chicory cultivated using three different kinds of fertilizers (i.e., developed, organic, and chemical) in the presence and absence of pesticides. Phenolic phytochemicals, including total polyphenols and flavonoids, and antioxidant activities, including reducing power, ABTS+ and DPPH radical scavenging activity, were analyzed using several antioxidant assays. HepG2 cell viability was analyzed using the MTT assay. The antioxidant properties of chicory were found to increase when cultivated with chemical fertilizer in the absence of pesticides. On the other hand, antioxidant capacity was higher in chicory cultivated with eco-developed fertilizer even in the presence of pesticides. Chicory grown using eco-developed or organic fertilizer was more effective in suppressing the proliferation of HepG2 cells when compared to chicory grown with chemical fertilizer. This effect was time dependent, regardless of treatment with or without pesticides. In conclusion, the antioxidant activity of chicory were affected by the presence or absence of pesticides. However, developed and organic fertilizers showed a strong anti-proliferative effect against HepG2 cells, regardless of the presence or absence of pesticides.

This study was conducted during the 2009 and 2010 grazing seasons to assess carcass parameters and chevon (goat meat) quality when meat-goat kids (n=72) were finished on pastures of red clover (Trifolium pratense L.; RCL), birdsfoot trefoil (Lotus corniculatus L.; BFT), or chicory (Cichorium intybus L.; CHIC). Final live weight (P<0.05) and carcass weight (P=0.10) were greater when goats were finished on RCL compared to CHIC with BFT being intermediate. Ribeye area, backfat thickness, body wall thickness, internal fat score, and leg score were not different (P>0.10) among treatments when adjusted for the covariate of carcass weight. Finishing meat-goat kids on RCL, BFT, or CHIC impacted concentrations of fatty acids (FAs) 18:1 trans-10, 18:1 cis-11, 18:2, 18:3 polyunsaturated fatty acids (PUFAs), omega-6, omega-3, and PUFA:saturated fatty acid ratio in longissimus lumborum samples. This study aimed to explore the spectrum-effect relationships between high-performance liquid chromatography fingerprints and the uric acid-lowering activities of chicory. Chemical fingerprints of chicory samples from ten different sources were determined by high-performance liquid chromatography, and then investigated by similarity analysis and hierarchical clustering analysis. Pharmacodynamics experiments were conducted in animals to obtain the uric acid-lowering activity information of each chicory sample. The spectrum-effect relationships between chemical fingerprints and the uric acid-lowering activities of chicory were established by canonical correlation analysis. The structures of potential effective peaks were identified by liquid chromatography with tandem mass spectrometry. The results showed that a close correlation existed between the spectrum and effect of chicory. Aesculin, chlorogenic acid, chicoric acid, isochlorogenic acid A/B/C and 13,14-seco-stigma5(6),14(15)-diene-3α-ol might be the main effective constituents. This work provides a general model of the combination of high-performance liquid chromatography and uric acid-lowering activities to study the spectrum-effect relationships of chicory, which can be used to discover the principle components responsible for the bioactivity.

Dry matter intakes (DMI), nutrient selection, and milk production responses of dairy cows grazing 3 herbage-based diets offered at 2 allowances were measured. The 2 allowances were 20 (low) and 30 (high) kg of dry matter (DM)/cow per day and these were applied to 3 herbage types: perennial ryegrass (PRG) and chicory (CHIC+) monocultures and a mixed sward of chicory and perennial ryegrass (MIX). The CHIC+ diet was supplemented with alfalfa hay (approximately 2 kg of DM/cow per day) to maintain dietary neutral detergent fiber (NDF) concentration and all diets were supplemented with energy-based pellets (6 kg of DM/cow per day). Holstein-Friesian dairy cows averaging 136 ± 30 d in milk were allocated to 4 replicates of the 6 treatments using stratified randomization procedures. Cows were adapted to their experimental diets over a 14-d period, with measurements of DMI, milk yield, and composition conducted over the following 10 d. Herbage DMI was lowest (12.8 vs. 14.0 kg of DM/d) for CHIC+ compared with the MIX and PRG, although total forage intake (grazed herbage plus hay) was similar (14.0 to 15.0 kg of DM/d) across the 3 treatments. Milk production, milk protein, and milk fat concentrations were not different between herbage types. Grazed herbage DMI increased with increasing herbage allowance and this was associated with increased milk protein concentration (3.23 to 3.34%) and total casein production (41.7 to 43.6 mg/g). Concentrations of polyunsaturated fatty acids in milk fat, particularly linoleic acid, were increased in milk from cows offered the CHIC+ or the MIX diets, indicating potential benefits of chicory herbage on milk fatty acid concentrations. We evaluated the effect of chicory (Cichorium intybus L.) seed extract (CI) on hepatic steatosis caused by early and late stage diabetes in rats (in vivo), and induced in HepG2 cells (in vitro) by BSA-oleic acid complex (OA). Different dosages of CI (1.25, 2.5 and 5 mg/ml) were applied along with OA (1 mM) to HepG2 cells, simultaneously and non-simultaneously; and without OA to ordinary non-steatotic cells. Cellular lipid accumulation and glycerol release, and hepatic triglyceride (TG) content were measured. The expression levels of sterol regulatory element-binding protein-1c (SREBP-1c) and peroxisome proliferator-activated receptor alpha (PPARα) were determined. Liver samples were stained with hematoxylin and eosin (H&E). Significant histological damage (steatosis-inflammation-fibrosis) to the cells and tissues and down-regulation of SREBP-1c and PPARα genes that followed steatosis induction were prevented by CI in simultaneous treatment. In non-simultaneous treatment, CI up-regulated the expression of both genes and restored the normal levels of the corresponding proteins; with a greater stimulating effect on PPARα, CI acted as a PPARα agonist. CI released glycerol from HepG2 cells, and targeted the first and the second hit phases of hepatic steatosis. Although the nutritive value of chicory (Cichorium intybus L.) and plantain (Plantago lanceolata L.) has been thoroughly studied, little is known about the grazing behavior of cattle feeding on chicory and plantain swards. The objective of the present study was to assess and describe the grazing behavior of dairy cows as affected by dietary proportions of chicory and plantain fed as monocultures for part of the day. Ninety Holstein-Friesian cows (489±42 kg of body weight; 4.1±0.3 body condition score, and 216±15 d in milk) were randomly assigned to 15 groups (6 cows per group) and grazed according to 7 treatments: control (CTL, 3 groups), perennial ryegrass (Lolium perenne L.) dominant sward (24-h pasture strip); 3 chicory treatments comprising 20, 40, and 60% of the diet, strip-grazing a monoculture of chicory to a fixed postgrazing residual before strip-grazing a perennial ryegrass dominant sward (2 groups of cows per treatment); and 3 plantain treatments comprising 20, 40, and 60% of the diet, strip-grazing a monoculture of plantain to a fixed postgrazing residual before strip-grazing a perennial ryegrass dominant sward (2 groups of cows per treatment). Four focal animals per group were equipped with 3-dimensional motion sensors, which provided the number of steps taken at each minute of the day. These cows were also fitted with automatic jaw-movement recorders that identified bites, mastication during ingestion, chewing during rumination, and determined grazing, rumination and idling times and bouts. Daily grazing time and bouts were not affected by treatments but rumination time differed and was reduced by up to 90 min when cows were allocated to chicory and plantain as 60% of their diet. Ruminative chewing was reduced in cows grazing chicory and plantain by up to 20% in cows allocated to the 60% treatments. Compared with perennial ryegrass, as the dietary proportion of chicory and plantain increased, cows spent more time idling and less time ruminating, and increased ingestive mastications 5 and 3 times for chicory and plantain, respectively. Cows allocated to chicory and plantain reduced bite rate and bites per grazing step linearly, and increased the number of mastications per bite of pasture dry matter intake while grazing pasture after having grazed chicory and plantain. These results indicate that cows grazing chicory and plantain masticate more during ingestion and reduce rumination time and chewing. They also suggest that chicory presents greater constraints to ingestion than does plantain. Care for human well-being and health requires the consumption of healthy foods, and therefore, the “babyleaf” gains increased space in the market as they have higher concentrations of bioactive compounds. The present study aimed to evaluate the growth and bioactive compounds of baby-leaf chicory (Cichorium intybus L.) according to the color of cultivation benches with different wavelengths. The experiment was conducted in a non-acclimatized greenhouse, covered with low density polyethylene film with an aluminized screen of 50% shading under the film and 50% shading screens on the sides. In a completely randomized design, five types of benches were evaluated without and with bright reflective colored materials: Control bench without reflective material, a bench with reflective bright white laminate, reflective bright dark red laminate, reflective bright dark blue laminate, and reflective bright yellow laminate. The bright white and yellow laminates amplified the most photosynthetically active radiation. All the laminates enhanced yield of baby-leaf chicory plants, where white, yellow, red, and dark blue enhanced the fresh matter by 102.6%, 50.2%, 60.6%, and 37.4% compared to the control, respectively. The bright dark blue laminates enhanced the bioactive compounds of baby-leaf chicory plants. The bright dark blue laminate enhanced the carotenoid production of baby-leaf chicory plants by 11.0%. The use of colored laminates was effective in the cultivation of baby-leaf chicory, both in terms of yield and quality, with significant increases in fresh matter and bioactive compounds. In this study, an easy, efficient, economical, and eco-friendly green bio-synthesis method was utilized to synthesize silver nanoparticles (AgNPs) using the extracts of four plants: Ginkgo biloba, Cichorium Intybus, Adiantum Capillus-Veneris, and Rosmarinus Officinalis. The synthesis of AgNPs was confirmed by using a uv-vis spectrometer, which showed distinct surface plasmon resonance (SPR) bands. The surface of AgNPs was characterized using scanning electron microscopy and Fourier-transform infrared spectroscopy. The anti-inflammatory activity of Tenoxicam/Meloxicam-loaded AgNPs has been studied using the inhibition of albumin denaturation method. Tenoxicam-loaded AgNPs showed higher % Inhibition, but Meloxicam-loaded AgNPs showed lower % Inhibition. Cichorium intybus, commonly known as chicory or coffee weed has been utilized since ancient times to treat an array of conditions relating to the kidney, heart, and liver. The aim of present study was to demonstrate Cichorium intybus's potential for antioxidant activity. This study was designed to examine the antioxidant capacity of Cichorium intybus leaf samples that were collected from Uttarakhand. To accomplish this purpose, the 2, 2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging capability assay were used to determine the free radical potential for each extract (methanol, and ethyl acetate) of leaf sample including the positive control of ascorbic acid. The plant leaf extract showed strong radical scavenging activities for both methanol as well as ethyl acetate extracts. Although the IC50 value for methanol is 155.85 μg/ml for DPPH inhibition, that is comparatively observed as lower than the IC50 value for ethyl acetate extract and control ascorbic acid. Therefore, the results show that methanol extract can be considered more potent at scavenging free radicles and implies a higher antioxidant activity. The findings of the present study suggested Cichorium intybus is a powerful natural antioxidant that can protect against damage caused by free radicals. Cichorium intybus leaves have an excellent antioxidant composition, which makes them a useful choice in pharmaceutical formulations that improve the health of humans, animals, and poultry. Using resilient, self-sustaining plants in urban green spaces enhances environmental and cultural benefits and reduces management costs. We assessed two spontaneous plant species, Linaria vulgaris Mill. and Cichorium intybus L., in four sites from the surrounding urban areas, ranging in altitude from 78 to 1040 m. Protection against UV-B radiation is crucial for plants at higher altitudes, guiding our focus on UV-visible absorption spectra, fluorometric emission spectra, secondary metabolite accumulation, and pigment dynamics in leaves. Our findings revealed a slight increase in UV-absorbing compounds with altitude and species-specific changes in visible spectra. The UV-emission of fluorochromes decreased, while red emission increased with altitude but only in chicory. Polyphenols and flavonoids showed a slight upward trend with altitude. Divergent trends were observed in condensed tannin accumulation, with L. vulgaris decreasing and C. intybus increasing with altitude. Additionally, chicory leaves from higher altitudes (792 and 1040 m) contained significantly lower triterpene concentrations. In L. vulgaris, chlorophyll pigments and carotenoids varied with sites, contrasting with UV absorbance variations. For C. intybus, pigment variation was similar to absorbance changes in the UV and VIS range, except at the highest altitude. The study was conducted to optimize the germination test and assess seed quality parameters in Kasani during 2022 in the laboratories of the Department of Seed Science and Technology, aimed. The results revealed maximum germination was recorded in Top of Paper (66.67%) method at 25°C and lowest germination (38.67%) was recorded at 30oC in sand substrate. Maximum hard seeds (17.33%) were recorded in between paper at 30°C and lowest hard seeds (7.33%) at 20°C in top of the paper. The other seed quality parameters viz., seedling length, seedling dry weight, vigour Indices and radicle emergence were also observed maximum at 20°C. The radicle emergence was started on 1st day in all temperatures except at 15°C temperature. From 2nd day radicle emergence was observed in all temperatures and till the final day the maximum radicle emergence (74.67%) was observed at 20°C. Thus, the study concludes that for conducting the germination test in Kasani crop, the optimum temperature is 20°C, and both the top of the paper (TP) and between paper methods are suitable.
