In this study, an overall survey regarding the determination of several bioactive compounds in olive fruit is presented. Two methodologies were developed, one UPLC-Q-TOF-MS method for the determination of olive fruit phenolic compounds and one HPLC-DAD methodology targeting the determination of pigments (chlorophylls and carotenoids), tocopherols (α-, β, -γ, δ-) and squalene. Target and suspect screening workflows were developed for the thorough fingerprinting of the phenolic fraction of olives. Both methods were validated, presenting excellent performance characteristics, and can be used as reliable tools for the monitoring of bioactive compounds in olive fruit samples. The developed methodologies were utilized to chemical characterize the fruits of the Kolovi olive variety, originating from the island of Lesvos, North Aegean Region, Greece. Twenty-five phenolic compounds were identified and quantified in Kolovi olives with verbascoside, hydroxytyrosol, oleacein and oleomissional found in significantly high concentrations. Moreover, 12 new bioactive compounds were identified in the samples using an in-house suspect database. The results of pigments analysis suggested that Kolovi variety should be characterized as low pigmentation, while the tocopherol and squalene content was relatively high compared to other olive varieties. The characterization of Kolovi olive bioactive content highlighted the high nutritional and possible economic value of the Kolovi olive fruit.

There is an increasing need for comparable and harmonized retention times (tR) in liquid chromatography (LC) among different laboratories, to provide supplementary evidence for the identity of compounds in high-resolution mass spectrometry (HRMS)-based suspect and nontarget screening investigations. In this study, a rigorously tested, flexible, and less system-dependent unified retention time index (RTI) approach for LC is presented, based on the calibration of the elution pattern. Two sets of 18 calibrants were selected for each of ESI+ and ESI-based on the maximum overlap with the retention times and chemical similarity indices from a total set of 2123 compounds. The resulting calibration set, with RTI set to range between 1 and 1000, was proposed as the most appropriate RTI system after rigorous evaluation, coordinated by the NORMAN network. The validation of the proposed RTI system was done externally on different instrumentation and LC conditions. The RTI can also be used to check the reproducibility and quality of LC conditions. Two quantitative structure-retention relationship (QSRR)-based models were built based on the developed RTI systems, which assist in the removal of false-positive annotations. The applicability domains of the QSRR models allowed completing the identification process with higher confidence for substances within the domain, while indicating those substances for which results should be treated with caution. The proposed RTI system was used to improve confidence in suspect and nontarget screening and increase the comparability between laboratories as demonstrated for two examples. A platform for archiving liquid chromatography high-resolution mass spectrometry (LC-HRMS) data was developed for the retrospective suspect screening of thousands of environmental pollutants with the ambition of becoming a European and possibly global standard. It was termed Digital Sample Freezing Platform (DSFP) and incorporates all the recent developments in the HRMS screening methods within the NORMAN Network. In the workflow, raw mass spectral data are converted into mzML, then mass spectral and chromatographic information on thousands of peaks of each sample is extracted into Data Collection Templates. The ‘digitally frozen’ samples can be retrospectively screened for the presence of virtually any compound amenable to LCeMS using a combination of information on its (i) exact mass, (ii) predicted retention time window in the chromatogram, (iii) isotopic fit and (iv) qualifier fragment ions. Its potential was demonstrated on monitoring of 670 antibiotics and 777 REACH chemicals from the Joint Black Sea Surveys (JBSS).

A simple, sensitive and efficient confirmatory method was developed and validated for the determination of 16 coccidiostats in animal tissues and eggs using hydrophilic interaction liquid chromatography–tandem mass spectrometry (HILIC-MS/MS). The sample preparation consisted of a solid-liquid extraction with ACN and dispersive SPE cleanup with MgSO4 and C18. Analysis was realized in an Acquity BEH HILIC silica column, in SRM mode. Both positive and negative ionization was performed, using polarity switching. Isocratic elution was used with a mobile phase of ACN: aqueous ammonium formate 1 mM with 0.1% formic acid (80:20, v/v). Method validation was performed in eggs, poultry, bovine, ovine, porcine and rabbit tissue and exceptionally low LODs were achieved, varying from 0.004 μg kg−1 (decoquinate in porcine tissue) to 0.560 μg kg−1 (halofuginone in eggs). The developed methodology was applied in 82 muscle and egg samples through the Greek National Residue Control Plan for coccidiostats.

INTRODUCTION Pelargonium sidoides is a member of the Geraniaceae family and it originates from the coastal regions of South Africa. In the last decades, Pelargonium sidoides root has been subjected to several surveys due to the assertion of its health benefits, such as the relief of symptoms of acute bronchitis, common cold and acute rhinosinusitis. Many studies have been conducted to reveal its naturally occurring bioactive chemicals, yet no wide-scope chemical characterisation strategies have been done using mass spectrometry. OBJECTIVE This research aimed to comprehensively characterise the chemical profile of Pelargonium sidoides root via high-resolution mass spectrometry. METHODOLOGY The Pelargonium sidoides root was extracted by a mixture of methanol: water in the proportion of 80:20. The extraction procedure included vortexing, shaking as well as the use of an ultrasound sonication bath under 40°C. After centrifugation, the supernatant was evaporated to dryness. The dry residue was reconstituted with a mixture of methanol/water (50:50, v/v), filtered and injected into an ultra-high-pressure liquid chromatography-quadruple time-of-flight mass spectrometer. RESULTS Overall, 33 compounds were identified in the root using suspect and non-target screening. These compounds were originated from different classes of compounds such as amino acids, phenolic acids, α-hydroxy-acids, vitamins, polyphenols, flavonoids, coumarins, coumarins glucosides, coumarin sulphates and nucleotides. Quantitative results were provided for the identified compounds, where their reference standards were available. CONCLUSION Some important compounds were elucidated, belonging to different classes of compounds such as antioxidants (coumarins and phenolic compounds), amino acids, nucleotides and vitamins revealing the importance of the bioactive content of this root.

During ozonation in wastewater treatment plants, ozone reacts with emerging pollutants, which are partially removed through the secondary treatment, as long as, with their biotransformation products, triggering the formation of ozonation transformation products (TPs). Although the transformation of parent compounds (PCs) and their metabolites has been reported in the literature, the probable transformation of biotransformation products has not been investigated so far. This study evaluates the fate of citalopram (CTR) and four of its biotransformation products (DESCTR, CTRAM, CTRAC and CTROXO) during ozonation experiments. A Gaussian curve-based trend analysis was performed for the first time for the automated detection of TPs in ozone concentrations ranging from 0.06 to 12 mg/L. In total 46 ozonation TPs were detected; 7 TPs of CTR, 10 of DESCTR, 9 of CTRAM, 12 of CTRAC and 8 of CTROXO and were structurally elucidated based on their high resolution tandem mass spectra interpretation and tandem mass spectra similarity with the respective PC. Results have demonstrated that the examined compounds follow common transformation pathways in reaction with ozone and that common TPs were formed through the ozonation of different structurally-alike compounds. Moreover, the toxicity of the identified TPs was predicted with an in-house risk assessment program.

Little information exists on the effects of shipping and handling on per- and polyfluoroalkyl substances (PFASs) in environmental samples. Thus, we evaluated the integrity of dried wastewater extracts and the sensitivity of our high-resolution mass spectrometry (HRMS) instrument to perform such analyses by monitoring 13 representative PFASs in samples extracted, evaporated, and stored at room temperature up to one month. Relative to zero-day recoveries of six detected PFASs ranged between 94 and 124% (RSD 38%) for influents, between 88 and 126% (RSD 18%) for effluents after 28 days. Larger variabilities are tentatively associated with the lack of specific mass-labeled standards and the interactions between analytes and remaining matrix components over time. In a second stage, a mix of local and international dry-shipped wastewater samples were analyzed and the same PFASs were quantified. Up to six PFASs were identified, with median concentrations ranging from 1.3 (perfluoro butyl sulfonate (PFBS)) to 7.7 ng/L (perfluoro hexanoic acid (PFHxA)) and from 1.5 (PFBS) to 13.8 ng/L (PFHxA) in local influents and effluents respectively; and from 0.7 (perfluoro hexyl sulfonate (PFHxS)) to 52.8 ng/L (PFHxA) and from 0.5 (PFHxS) to 21.4 ng/L (PFHxA) in Greek influents and effluents, respectively. The importance of this study lies on the need to consider the wider recovery shifts and expanded variability ranges of PFASs derived from the transport and storage times of dried extracts, particularly when applied to HRMS and wide-scope screening approaches.

Monitoring of biogenic amines in food is important for quality control, in terms of freshness evaluation and even more for food safety. A novel and cost-effective method was developed and validated for the determination of the main biogenic amines: histamine, putrescine, cadaverine, spermidine and spermine in fish tissues. The method includes extraction of amines with perchloric acid, pre-column derivatization with Pyrene Sulfonyl Chloride (PSCl), extraction of derivatives with toluene, back-dissolution in ACN after evaporation and determination by reversed phase high performance liquid chromatography with UV and intramolecular excimer fluorescence detection. The structure of the pyrene-derivatives was confirmed by liquid chromatography&ndash, mass spectrometry with electrospray ionization. The standard addition technique was applied for the quantitation due to significant matrix effect, while the use of 1,7-diaminoheptane as internal standard offered an additional confirmation tool for the identification of the analytes. Method repeatability expressed as %RSD ranged between 7.4&ndash, 14% for the different amines and recovery ranged from 67% for histamine up to 114% for spermine. The limits of detection ranged between 0.1&ndash, 1.4 mg kg&minus, 1 and the limits of quantification between 0.3&ndash, 4.2 mg kg&minus, 1. The method was applied to canned fish samples and the concentrations of the individual biogenic amines were below the detection limit up to 40.1 mg kg&minus, 1, while their sum was within the range 4.1&ndash, 49.6 mg kg&minus, 1.

This study presents the development and validation of a comprehensive quantitative target methodology for the analysis of 2316 emerging pollutants in water based on Ultra-Performance Liquid Chromatography Quadrupole-Time-Of-Flight Mass Spectrometry (UPLC-Q-ToF-HRMS/MS). Target compounds include pesticides, pharmaceuticals, drugs of abuse, industrial chemicals, doping compounds, surfactants and transformation products, among others. The method was validated for 195 analytes, chosen to be representative of the chemical space of the target list, enabling the assessment of the performance of the method. The method involves a generic sample preparation based on mixed mode solid phase extraction, a UPLC-QTOF-MS/MS screening method using Data Independent Acquisition (DIA) mode, which provides MS and MS/MS spectra simultaneously and an elaborate strong post-acquisition evaluation of the data. The processing method was optimized to provide a successful identification rate >95 % and to minimize the number of false positive results (< 5 %). Decision limit (CCα) and detection capability (CCβ) were also introduced in the validation scheme to provide more realistic metrics on the performance of a HRMS-based wide-scope screening method. A new system of identification points (IPs) based on the one described in the Commission Decision 2002/657/EC was applied to communicate the confidence level in the identification of the analytes. This system considers retention time, mass accuracy, isotopic fit and fragmentation; taking full advantage of the capacities of the HRMS instruments. Screening of a large number of chemicals of emerging concern is highly desirable for the control of crops irrigated with reclaimed water since it is considered an alternative water source of great value. This study describes a high resolution mass spectrometry approach for developing methods for quantification in lettuce leaves of 48 different wastewater-borne pollutants (including analgesics and anti-inflammatories, anti-hypertensives, antifungal agents, lipid regulators, psychiatric drugs and stimulants, β-blockers, antibiotics, antimycotics, and sweeteners) frequently found in water resources. In this respect, a simple and fast QuEChERS-based method for the determination of contaminants in lettuce has been developed. During extraction, the use of formic acid was adopted to further improve the results of some problematic compounds (e.g., fenofibrate, furosemide, metronidazole, oxcarbazepine, sulfanilamide). High resolution multiple reaction monitoring (MRM) and SWATH acquisition were compared in term of accuracy, repeatability, sensitivity, linearity and matrix effect. Both methods provided similar recoveries between 80 and 120% in lettuce leaves, although sulfanilamide, ciprofloxacin, and sulfamethazine presenting values of 26.8, 27.8, and 28.4% in MRM and 25, 33.9, and 35% in SWATH, respectively. The effectiveness of a two-step cleanup on analyte recovery was also assessed and matrix effects were also taken into consideration during the method validation. The developed method allows the simultaneous quantitative analysis of 48 compounds (drug residues and metabolites) in lettuce leaves irrigated with treated wastewater for human consumption. Application of the present method to lettuce crops growth in controlled conditions showed the presence of 14 out 48 studied compounds with similar concentrations in both acquisition modes ranging from 3.3 and 1.3 ng g for climbazole (for MRM and SWATH, respectively) to 33.2 and 17.7 ng g for sulfamethazine. Drug residues such as carbamazepine (6.0 and 8.5 ng g ), and its metabolite carbamazepine epoxide (18.1 and 16.5 ng g ), frequently found in wastewater effluents, were also detected., This study has been financially supported by the EU through the WaterJPI-2015 AWARE project (PCIN-2017–067). A robust and sensitive methodology, utilizing GC-MS, for the identification and quantitation of seven β-agonist residues (mabuterol, clenbuterol, brombuterol, mapenterol, clenpenterol, clenproperol, and clencyclohexerol), in liver and meat samples, is described. Different extraction procedures, followed by GC/MS measurement, were tested and compared in order to achieve optimum extraction conditions and eliminate matrix effects. The optimized method consisted of consecutive extractions with hydrochloric tris(hydroxymethyl)aminomethane (TRIS-HCl) and tert-butyl-methyl ether (TBME), defatting with hexane and solid-phase extraction (SPE) with C18 cartridges. Ν,O-bis(trimethylsilyl) trifluoroacetamide (BSTFA) with 1% trimethylchlorosilane (TMCS) was used as a derivatization agent and the GC-EI-MS determination was performed using a MDN-5S capillary column in single-ion monitoring (SIM) acquisition mode. The method was validated according to the Commission Decision 2002/657/EC, fulfilling all the EU criteria. Quantification was performed via internal standard calibration using deuterated analogs of the compounds. Recoveries ranged from 83 to 118% (mabuterol at concentration levels of 2.0 and 1.5 μg kg−1, respectively) and precision, expressed as %relative standard deviation (% RSD), was in every case lower than the % RSDs obtained from Horwitz equation. The obtained decision limit (CCα) and detection capability (CCβ) values varied from 0.21 ng g−1 (clenbuterol) to 0.49 ng g−1 (clenproperol, clencyclohexerol) and from 0.60 (mapenterol) to 0.69 ng g−1 (clenpenterol), respectively.

The aim of this work was to study the chromatographic behavior of polar artificial sweeteners on three hydrophilic interaction liquid chromatography (HILIC) columns: Kinetex HILIC (unbounded silica, core shell), ZIC HILIC (sulfoalkylbetain) and TSKgel Amide-80 (amide). The studied analytes were dipeptides (aspartame, neotame, advantame, alitame), sulfamates (acesulfame, saccharin, cyclamate), and sugars (sucralose, neohesperidin dihydrochalcone). Critical parameters such as the mobile phase pH, buffer concentration, column temperature, and organic modifier were studied for the investigation of the retention mechanism. Partition and adsorption models were also used. The retention mechanism of dipeptides seems to be partition for the aqueous layer and weak electrostatic interactions or hydrogen bonds. This study investigated the use of liquid chromatography–atmospheric pressure photoionization tandem mass spectrometry (LC-APPI-MS/MS) for the determination of six polybrominated diphenyl ethers (PBDEs) coded as brominated diphenyl ether (BDE) 28, 47, 99, 100, 153, and 154 that are used as flame retardants. It was shown that diphenyl ethers with four to six bromine atoms can be ionized in negative polarity with parent ions corresponding to [M-Br +O]−. On the contrary, the ionization of the diphenyl ether with three bromine atoms (BDE 28), required the use of positive polarity in order to generate precursor ions [M]+. PBDEs were separated by a C18 column (50 × 4.6 mm, 1.8 µm) with a mixture of methanol–water as the mobile phase following gradient elution. The discrimination of organic and conventional production has been a critical topic of public discussion and constitutes a scientific issue. It remains a challenge to establish a correlation between the agronomical practices and their effects on the composition of olive oils, especially the phenolic composition, since it defines their organoleptic and nutritional value. Thus, a liquid chromatography-electrospray ionization-quadrupole time of flight tandem mass spectrometric method was developed, using target and suspect screening workflows, coupled with advanced chemometrics for the identification of phenolic compounds and the discrimination between organic and conventional extra virgin olive oils. The method was optimized by one-factor design and response surface methodology to derive the optimal conditions of extraction (methanol/water (80:20, v/v), pure methanol, or acetonitrile) and to select the most appropriate internal standard (caffeic acid or syringaldehyde). The results revealed that extraction with methanol/water (80:20, v/v) was the optimum solvent system and syringaldehyde 1.30 mg L-1 was the appropriate internal standard. The proposed method demonstrated low limits of detection in the range of 0.002 (luteolin) to 0.028 (tyrosol) mg kg-1. Then, it was successfully applied in 52 olive oils of Kolovi variety. In total, 13 target and 24 suspect phenolic compounds were identified. Target compounds were quantified with commercially available standards. A novel semi-quantitation strategy, based on chemical similarity, was introduced for the semi-quantification of the identified suspects. Finally, ant colony optimization-random forest model selected luteolin as the only marker responsible for the discrimination, during a 2-year study. The fate of four antimicrobials (cefadroxil, CFD; metronidazole, METRO; trimethoprim, TRI; sulfamethoxazole, SMX) was studied in Lemna minor systems and the role of different mechanisms on their removal was evaluated. All micropollutants were significantly removed in batch experiments with active Lemna minor; the highest removal was observed for CFD (100% in 14 d), followed by METRO (96%), SMX (73%) and TRI (59%) during 24 d of the experiment. Calculation of kinetic constants for hydrolysis, photodegradation, sorption to biomass and plant uptake revealed significant differences depending on the compound and the studied mechanism. For METRO, TRI and SMX the kinetic constants of plant uptake were by far higher comparing to those of the other mechanisms. The transformation products of antimicrobials were identified using UHPLC-QToF-MS. Two were the main degradation pathways for TRI; hydroxylation takes place during both phyto- and photodegradation, while demethylation occurs only in absence of Lemna minor. The operation of a continuous-flow duckweed system showed METRO and TRI removal equal to 71 ± 11% and 61 ± 8%, respectively. The application of mass balance and the use of published biodegradation constants showed that plant uptake and biodegradation were the major mechanisms governing METRO removal; the most important mechanism for TRI was plant uptake.

Untargeted analysis of a composite house dust sample has been performed as part of a collaborative effort to evaluate the progress in the field of suspect and nontarget screening and build an extensive database of organic indoor environment contaminants. Twenty-one participants reported results that were curated by the organizers of the collaborative trial. In total, nearly 2350 compounds were identified (18%) or tentatively identified (25% at confidence level 2 and 58% at confidence level 3), making the collaborative trial a success. However, a relatively small share (37%) of all compounds were reported by more than one participant, which shows that there is plenty of room for improvement in the field of suspect and nontarget screening. An even a smaller share (5%) of the total number of compounds were detected using both liquid chromatography–mass spectrometry (LC-MS) and gas chromatography–mass spectrometry (GC-MS). Thus, the two MS techniques are highly complementary. Most of the compounds were detected using LC with electrospray ionization (ESI) MS and comprehensive 2D GC (GC×GC) with atmospheric pressure chemical ionization (APCI) and electron ionization (EI), respectively. Collectively, the three techniques accounted for more than 75% of the reported compounds. Glycols, pharmaceuticals, pesticides, and various biogenic compounds dominated among the compounds reported by LC-MS participants, while hydrocarbons, hydrocarbon derivatives, and chlorinated paraffins and chlorinated biphenyls were primarily reported by GC-MS participants. Plastics additives, flavor and fragrances, and personal care products were reported by both LC-MS and GC-MS participants. It was concluded that the use of multiple analytical techniques was required for a comprehensive characterization of house dust contaminants. Further, several recommendations are given for improved suspect and nontarget screening of house dust and other indoor environment samples, including the use of open-source data processing tools. One of the tools allowed provisional identification of almost 500 compounds that had not been reported by participants. [Figure not available: see fulltext.].

Two liquid chromatographic systems, one coupled to atmospheric pressure chemical ionization and tandem mass spectrometric methods (UHPLC-APCI-MS/MS) and the other a high-performance liquid chromatographic coupled to diode array detector (HPLC-DAD) were used to develop and validate methods for the simultaneous determination of fat-soluble vitamins A, D3 and E in rice cereal baby foods. The chromatographic separation was performed on C18 columns with a mixture of methanol-acetonitrile as mobile phase for all methods. The extraction of fat-soluble vitamins included enzymatic hydrolysis with α-amylase, saponification, extraction with petroleum ether or n-hexane and purification with silica cartridge (only for vitamin D3). Quantification of vitamin D3 and E through UHPLC-APCI-MS/MS was performed by the use of internal standards (IS) D3-d3 and E-d6, respectively, while IS was not used for vitamin A. The methods were optimized and validated in terms of linearity, precision, trueness, limits of detection and quantification. The recoveries were in the range of 85.0–107% for retinol, 92.0–105% for α-tocopherol and 95.2–106% for cholecalciferol and the %RSD (Relative Standard Deviation) values ranged from 6.4% to 15%. Citalopram (CTR) is a worldwide highly consumed antidepressant which has demonstrated incomplete removal by conventional wastewater treatment. Despite its global ubiquitous presence in different environmental compartments, little is known about its behaviour and transformation processes during wastewater treatment. The present study aims to expand the knowledge on fate and transformation of CTR during the biological treatment process. For this purpose, batch reactors were set up to assess biotic, abiotic and sorption losses of this compound. One of the main objectives of the study was the identification of the formed transformation products (TPs) by applying suspect and non-target strategies based on liquid chromatography quadrupole-time-of-flight mass spectrometry (LC-QTOF-MS). The complementary use of reversed phase liquid chromatography (RPLC) and hydrophilic interaction liquid chromatography (HILIC) for the identification of polar TPs, and the application of in-house developed quantitative structure-retention relationship (QSRR) prediction models, in addition to the comprehensive evaluation of the obtained MS/MS spectra, provided valuable information to support identification. In total, fourteen TPs were detected and thirteen of them were tentatively identified. Four compounds were confirmed (N-desmethylCTR, CTR amide, CTR carboxylic acid and 3-oxo-CTR) through the purchase of the corresponding reference standard. Probable structures based on diagnostic evidence were proposed for the additional nine TPs. Eleven TPs are reported for the first time. A transformation pathway for the biotransformation of CTR was proposed. The presence of the identified TPs was assessed in real wastewater samples through retrospective analysis, resulting in the detection of five compounds. Finally, the potential ecotoxicological risk posed by CTR and its TPs to different trophic levels of aquatic organisms was evaluated by means of risk quotients.

Bisphenol A (BPA), a known potential endocrine disrupting compound (EDC) is expected to be present in low quantities in canned food due to its migration from the inner surface coating of cans made of epoxy resins. A selective and confirmatory analytical method, based on microwave assisted extraction (MAE), molecularly imprinted solid phase extraction (MISPE) using a polymer prepared by a non-covalent molecular imprinting technique and liquid chromatography coupled with electrospray ionization mass spectrometry (LC-ESI/MS) was developed for the determination of BPA in canned pineapple, tuna and mushrooms. First, the effect of the loading medium of hydro- organic solutions on the binding of BPA and its deuterated analogue on the MISPE sorbent was investigated. Subsequently, the effects of the experimental conditions of the microwave assisted extraction (solvent, sample mass/solvent volume, time and temperature) on the obtained recovery of BPA from canned food were assessed and the parameters were optimized to provide maximum recovery and selectivity. It was demonstrated that the combination of MAE with MISPE permits the use of a selective extraction solvent (methanol/water, 4/6, v/v), simplifying the sample preparation steps and enhancing sample clean-up of complex food matrices. The method was validated in different food matrices, using BPA-d16 as internal standard and quantitative relative recoveries were determined. The precision (RSD %) of the method ranged from 7% to 10% and the limit of detection was at low ng/g level for all food matrices. The determined concentration of BPA in commercial canned samples ranged between 7.3 and 42.3 ng/g.

Hydrophilic interaction liquid chromatography (HILIC) and reversed phase LC (RPLC) coupled to high resolution mass spectrometry (HRMS) are widely used for the identification of suspects and unknown compounds in the environment. For the identification of unknowns, apart from mass accuracy and isotopic fitting, retention time (tR) and MS/MS spectra evaluation is required. In this context, a novel comprehensive workflow was developed to study the tR behavior of large groups of emerging contaminants using Quantitative Structure-Retention Relationships (QSRR). 682 compounds were analyzed by HILIC-HRMS in positive Electrospray Ionization mode (ESI). Moreover, an extensive dataset was built for RPLC-HRMS including 1830 and 308 compounds for positive and negative ESI, respectively. Support Vector Machines (SVM) was used to model the tR data. The applicability domains of the models were studied by Monte Carlo Sampling (MCS) methods. The MCS method was also used to calculate the acceptable error windows for the predicted tR from various LC conditions. This paper provides validated models for predicting tR in HILIC/RPLC-HRMS platforms to facilitate identification of new emerging contaminants by suspect and non-target HRMS screening, and were applied for the identification of transformation products (TPs) of emerging contaminants and biocides in wastewater and sludge.

In this study, the simultaneous determination of avermectins (emamectin, eprinomectin, abamectin, doramectin and ivermectin) and milbemycines (moxidectin) in fish tissue with LC-ESI-MS/MS, was studied. Optimum chromatographic separation of target analytes was achieved using a Waters Acquity UPLC BEH C18 (100 mm × 2.1 mm, 1.7 mm) analytical column, operated at 40 °C and the composition of the mobile phase used was (A): ACN-MeOH (0.1% HCOOH) (1:1) and (B): 1 mM HCOONH4 (0.1% HCOOH). Various mobile phases were tested and the effect of the mobile phase composition on the analytes ionization was thoroughly examined in an extensive ionization study, aiming to increase the analytes' sensitivity. Deuterated ivermectin (IVR-d2) was used as an internal standard (IS). Avermectin's and milbemycine's extraction from the fish matrix was conducted with acidified ACN (0.1% HCOOH), followed by QuEChERS methodology. The method developed herein was validated according to the European Legislation requirements (Commission Decision 657/2002/EC) and recoveries ranged from 86 to 106% for all target analytes, with relative standard deviations

The current study reports on the toxicity, uptake, and biotransformation potential of zebrafish (embryos and larvae) exposed to benzotriazoles (BTs). Acute toxicity assays were conducted. Cardiac function abnormalities (pericardial edema and poor blood circulation) were observed from the phenotypic analysis of early life zebrafish embryos after BTs exposure. For the uptake and biotransformation experiment, extracts of whole body larvae were analyzed using liquid chromatography-high-resolution tandem mass spectrometry (UPLC-Q-TOF-HRMS/MS). The utility of hydrophilic interaction liquid chromatography (HILIC) as complementary technique to reversed phase liquid chromatography (RPLC) in the identification process was investigated. Through HILIC analyses, additional biotransformation products (bio-TPs) were detected, because of the enhanced sensitivity and better separation efficiency of isomers. Therefore, reduction of false negative results was accomplished. Both oxidative (hydroxylation) and conjugative (glucuronidation, sulfation) metabolic reactions were observed, while direct sulfation proved the dominant biotransformation pathway. Overall, 26 bio-TPs were identified through suspect and nontarget screening workflows, 22 of them reported for the first time. 4-Methyl-1- H-benzotriazole (4-MeBT) demonstrated the highest toxicity potential and was more extensively biotransformed, compared to 1- H-benzotriazole (BT) and 5-methyl-1- H-benzotriazole (5-MeBT). The extent of biotransformation proved particularly informative in the current study, to explain and better understand the different toxicity potentials of BTs.

In this study, the removal of niflumic acid (NA) and the identification of its transformation products (TPs) during ozonation was investigated. The influence of initial ozone concentration and pH value on NA's removal was tested, while suspect and non-target screening approaches were followed for the identification of ozonation TPs. The structure elucidation was based on accurate mass and isotopic pattern criteria and interpretation of the acquired MS/MS spectra. Moreover, an in-house retention time prediction model was used as a supporting tool for their identification. Results indicated the highly reactivity of NA with the molecular ozone, since the reaction was extremely fast and was completed within the first minute of the reaction. A total of thirteen TPs of NA were identified, and their proposed structures show that the oxidation occurred in the heterocyclic ring of the molecule, while the aniline-like part remained intact by ozone attack due to the presence of the three fluoride atoms, which act as electron withdrawing groups. Ozonation experiments were conducted also using the most abundant TP of NA as parent compound. Three second generation TPs of NA were detected and possible structures were proposed.

New psychoactive substances (NPSs) have become increasingly popular in recent years. The analysis of these substances in influent wastewater (IWW) can be used to track their use in communities. In addition, an evaluation of the amount of NPSs released to the aquatic environment can be performed through the analysis of effluent wastewater (EWW). This study presents the development, validation and application of an analytical methodology, based on solid phase extraction (SPE) and liquid chromatography-tandem mass spectrometry (LC-MS/MS), for the determination of 10 NPSs in IWW and EWW. Synthetic cannabinoids, cathinones, piperazines and pyrrolidophenones are included among the target analytes. To the authors' knowledge, it is the first time that eight out of these substances (4'-methylpyrrolidinobutyrophenone (MPPP), a-pyrrolidinopentiophenone (a-PVP), 2-[(1S,3R)-3-hydroxycyclohexyl]-5-(2-methyl-2-octanyl) phenol (CP47,497), (1-naphthyl(1-pentyl-1H-indol-3-yl) methanone (JWH-018), (1-butyl-1H-indol-3-yl)(1-naphthyl) methanone (JWH-073), (4-ethyl-1-naphthyl)(1-pentyl-1H-indol-3-yl) methanone (JWH-210), (4-methyl-1-naphthyl) (1-pentyl-1H-indol-3-yl) methanone (JWH-122) and 2-(2-methoxyphenyl)-1-(1-pentyl-1H-indol-3-yl) ethanone (JWH-250)) are investigated in wastewater. The optimized conditions for the analysis of this set of compounds included a SPE clean-up step using a polymeric sorbent and the use of a pentafluorophenyl (PFP) chromatographic column. Despite the broad range of physicochemical properties of the analytes the method allowed acceptable absolute recoveries (40-109%) for all the studied compounds at different levels of concentration. Low method limits of detection (MLODs) were achieved, ranging between 0.3 and 10 ng/L except for BZP and CP47,497 (20 and 23 ng/L, respectively), allowing a reliable and accurate quantification of the analytes. The method was successfully applied to the analysis of IWW and EWW samples from five wastewater treatment plants (WWTPs) located in Santorini Island (a highly touristic resort in Greece). Four out of 10 compounds (a-PVP, CP47,497, JWH-122 and JWH-210) were detected at least in one sample, being the first evidence of their presence in wastewater. CP47,497 was the most ubiquitous and abundant compound, showing concentrations up to 634 ng/L in some cases.

Biodegradation of benzotriazole (BTR), 5-chlorobenzotriazole (CBTR), xylytriazole (XTR), 4-methyl-1H-benzotriazole (4TTR), 5-methy-1H-lbenzotriazole (5TTR) and 2-hydroxybenzothiazole (OHBTH) was studied in activated sludge batch experiments under aerobic and anoxic conditions, presence of organic substrate and different sludge residence times (SRTs). Their sludge-water distribution coefficients were also calculated in sorption experiments and ranged between 87 and 220 L kg(-1). Significant biodegradation of BTR, CBTR, XTR and OHBTH was observed in all biotic experiments. Half-life values ranged between 23 and 45 h (BTR), 18 and 47 h (CBTR), 14 and 26 h (XTR), 6.5 and 24 h (OHBTH). The addition of substrate did not suppress biodegradation kinetics; whereas in some cases accelerated biodegradation of microcontaminants. Except for CBTR, no effect of SRT on biodegradation constants was observed. Prediction of micropollutants removal in Sewage Treatment Plants (STPs) indicated that they will be partially removed, mainly due to aerobic biodegradation. Higher removal is expected at STPs operating at higher SRT and higher suspended solids concentrations.

A fast and sensitive multianalyte/multiclass high-performance reversed-phase liquid chromatography–tandem mass spectrometry (LC–MS/MS) method was developed and validated for the simultaneous analysis of 89 pharmaceuticals in influent and effluent wastewater samples. The method developed consists of solid-phase extraction (SPE) using a hydrophilic–lipophilic-balanced polymer followed by LC–MS/MS with electrospray ionization in both positive mode and negative mode. The selected pharmaceuticals belong to different classes—analgesic/anti-inflammatory drugs, antibiotics, antiepileptics, β-adrenoceptor-blocking drugs, lipid-regulating agents, statins, and many others. The influence of the mobile phase composition on the sensitivity of the method, and the optimum conditions for SPE in terms of analyte recovery were extensively studied. Chromatographic separation was performed on an Atlantis T3 (100 mm × 2.1 mm, 3-μm) column with a gradient elution using methanol–0.01 % v/v formic acid as the mobile phase in positive ionization mode determination and methanol–acetonitrile–1 mM ammonium formate as the mobile phase in negative ionization mode determination. Recoveries for most of the compounds ranged from 50 to 120 %. Precision, expressed as relative standard deviations, was always below 15 %, and the method detection limits ranged from 1.06 ng/L (4-hydroxyomeprazole) to 211 ng/L (metformin). Finally, the method developed was applied to the determination of target analytes in wastewater samples obtained from the Psyttalia wastewater treatment plant, Athens, Greece. Although SPE of pharmaceuticals from wastewater samples and their determination by LC–MS/MS is a well-established technique, the uniqueness of this study lies in the simultaneous determination of a remarkable number of compounds belonging to more than 20 drug classes. Moreover, the LC–MS/MS method has been thoroughly optimized so that maximum sensitivity is achieved for most of the compounds, making the proposed method a valuable tool for pharmaceutical analysis in influent and effluent wastewater at the sub-nanogram per liter level.

A wide-scope screening methodology has been developed for the identification of veterinary drugs and pharmaceuticals in fish tissue and milk using ultrahigh-performance liquid chromatography quadrupole time-of-flight mass spectrometry (UHPLC-QTOF MS). The method was validated using a qualitative approach at two concentration levels. The detection of the residues was accomplished by retention time, accurate mass, and the isotopic fit using an in-house database. Product-ion spectra were used for unequivocal identification of the compounds. Generic sample treatment was applied. The majority of the compounds were successfully detected and identified at concentration levels of 150 ng mL(-1) in milk and 200 μg kg(-1) in fish (80% of the compounds in both matrices), whereas satisfactory results were also obtained at concentration levels of 15 ng mL(-1) in milk and 20 μg kg(-1) in fish (60% of the compounds detected and identified).

Identification of transformation products (TPs) of emerging pollutants is challenging, due to the vast number of compounds, mostly unknown, the complexity of the matrices and their often low concentrations, requiring highly selective, highly sensitive techniques. We compile background information on biotic and abiotic formation of TPs and analytical developments over the past five years. We present a database of biotic or abiotic TPs compiled from those identified in recent years. We discuss mass spectrometric (MS) techniques and workflows for target, suspect and non-target screening of TPs with emphasis on liquid chromatography coupled to MS (LC-MS). Both low- and high-resolution (HR) mass analyzers have been applied, but HR-MS is the technique of choice, due to its high confirmatory capabilities, derived from the high resolving power and the mass accuracy in MS and MS/MS modes, and the sophisticated software developed.

The aim of this work was the development of a simple, novel and accurate method for the determination of adenosine triphosphate (ATP) and its first five catabolites: adenosine diphosphate (ADP), adenosine monophosphate (AMP), inosine monophosphate (IMP), inosine (Ino) and hypoxanthine (Hx), in fish tissue, based on hydrophilic interaction liquid chromatography (HILIC). For this purpose, a stationary phase for polar and hydrophilic compounds (ZIC-pHILIC) was used. The effect of different chromatographic parameters and the molecular mechanism based on the van’t Hoff plot were examined. The t-test and Dixon’s Q-test were applied in order to examine statistical differences and outlier values. The recovery of the method ranged between 82.7% and 127% and the %RSD values were lower than 10% for all analytes determined. The method was applied in frozen sea bream samples stored at 0–4 °C. The Ki-, G-, H- and F values were calculated for the estimation of the level of fish freshness.

A laboratory scale Hybrid Moving Bed Biofilm Reactor (HMBBR) was used to study the removal of five benzotriazoles and one benzothiazole from municipal wastewater. The HMBBR system consisted of two serially connected fully aerated bioreactors that contained activated sludge (AS) and K3-biocarriers and a settling tank. The average removal of target compounds ranged between 41% (4-methyl-1H-benzotriazole; 4TTR) and 88% (2-hydroxybenzothiazole; OHBTH). Except for 4TTR, degradation mainly occurred in the first bioreactor. Calculation of biodegradation constants in batch experiments and application of a model for describing micropollutants removal in the examined system showed that AS is mainly involved in biodegradation of OHBTH, 1H-benzotriazole (BTR) and xylytriazole (XTR), carriers contribute significantly on 4TTR biodegradation, while both types of biomass participate on elimination of 5-chlorobenzotriazole (CBTR) and 5-methyl-1H-benzotriazole (5TTR). Comparison of the HMBBR system with MBBR or AS systems from literature showed that the HMBBR system was more efficient for the biodegradation of the investigated chemicals. Biotransformation products of target compounds were identified using ultra high-performance liquid chromatography, coupled with a quadrupole-time-of-flight high-resolution mass spectrometer (UHPLC-QToF-MS). Twenty two biotransformation products were tentatively identified, while retention time denoted the formation of more polar transformation products than the parent compounds.

This study presents the atmospheric pressure photoionization (APPI) of high-chlorinated (five or more chlorine atoms) polychlorinated biphenyls (PCBs) using toluene as dopant, after liquid chromatographic separation. Mass spectra of PCB 101, 118, 138, 153, 180, 199, 206 and 209 were recorded by using liquid chromatography-APPI-tandem mass spectrometry (LC-APPI-MS/MS) in negative ion full scan mode. Intense peaks appeared at m/z that correspond to (MCl+O) � ions, where M is the analyte molecule. Furthermore, a detailed strategy, which includes designs of experiments, for the development and optimization of LC-APPI-MS/MS methods is described. Following this strategy, a sensitive and accurate method with low instrumental limits of detection, ranging from 0.29pg for PCB 209 to 8.3pg for PCB 101 on column, was developed. For the separation of the analytes, a Waters XSELECT HSS T3 (100mm×2.1mm, 2.5 μm) column was used with methanol/water as elution system. This method was applied for the determination of the above PCBs in water samples (surface water, tap water and treated wastewater). For the extraction of PCBs from water samples, a simple liquid-liquid extraction with dichloromethane was used. Method limits of quantification, ranged from 4.8ngl � 1 , for PCB 199, to 9.4ngl � 1 , for PCB 180, and the recoveries ranged from 73%, for PCB 101, to 96%, for PCB 199. A simple, rapid, and sensitive method was developed for the simultaneous determination of 21 pesticides from different chemical classes (organophosphorus, triazines, carbamates, and anilinopyrimidine pesticides) in Chios mastic gum (Pistacia lentiscus L. var. Chia). Quick, easy, cheap, effective, rugged, and safe (QuEChERS) technique was used for the extraction of the target analytes; the mastic gum samples were initially extracted with acetonitrile (ACN), and then, the extracts were purified by dispersive solid-phase extraction using primary secondary amine (PSA) and C18 as sorbents. The determination of pesticides in the final extract was carried out by liquid chromatography coupled with tandem mass spectrometry with electrospray ionization (LC-ESI-MS/MS). The method was validated for linearity, accuracy, precision (repeatability and inter-laboratory reproducibility), limits of detection and quantification, and the matrix effect factor. Recoveries of the pesticides fortified at 5, 50, and 500 ng g−1 ranged from 49.7 % (quinalphos) to 127 % (pirimiphos-ethyl) and reproducibility (RSDR, n = 2 × 3) ranged from 4.4 % (propham) to 20 % (pirimiphos-methyl, propham, and triazophos). The detection limits of the method were 0.8 (mecarbam, pyrazophos) to 23 ng g−1 (parathion-ethyl). The uncertainty of the method was also calculated based on the Eurachem/Citac Guidelines, and the contribution of each independent parameter was investigated.

The removal of endocrine disrupting compounds (EDCs) and non-steroidal anti-inflammatory drugs (NSAIDs) was studied in three lab-scale anaerobic digestion (AD) systems; a single-stage mesophilic, a single-stage thermophilic and a two-stage thermophilic/mesophilic. All micropollutants underwent microbial degradation. High removal efficiency (80%) was calculated for diclofenac, ibuprofen, naproxen and ketoprofen; whereas triclosan, bisphenol A and the sum of nonylphenol (NP), nonylphenol monoethoxylate (NP1EO) and nonylphenol diethoxylate were moderately removed (40-80%). NSAIDs removal was not affected by the type of AD system used; whereas slightly higher EDCs removal was observed in two-stage system. In this system, most microcontaminants were removed in thermophilic digester. Biotransformation of NP1EO and NP was affected by the temperature applied to bioreactors. Under mesophilic conditions, higher removal of NP1EO and accumulation of NP was noticed; whereas the opposite was observed under thermophilic conditions. For most analytes, higher specific removal rates were calculated under thermophilic conditions and 20 days SRT.

Perfluorinated compounds (PFCs) are known chemicals that are used in a wide variety of industrial and consumer products, and have been reported to exist in the environment as contaminants. In this study, a liquid chromatography-electrospray-tandem mass spectrometry (LC-ESI-MS/MS) method was developed for simultaneous determination of 10 perfluoroalkyl carboxylate acids (PFCAs), 5 perfluoroalkyl sulfonates (PFASs), and 3 perfluoroalkyl sulfonamides (PFSAs) (18 PFCs in total), in dissolved and particulate phases of wastewater (raw and treated), and in dewatered sewage sludge. The target PFCAs were perfluoropentanoic acid (PFPeA; C5), perfluorohexanoic acid (PFHxA; C6), perfluoroheptanoic acid (PFHpA; C7), perfluorooctanoic acid (PFOA; C8), perfluorononanoic acid (PFNA; C9), perfluorodecanoic acid (PFDA; C10), perfluoroundecanoic acid (PFUdA; C11), perfluorododecanoic acid (PFDoA; C12), perfluorotridecanoic acid (PFTrDA; C13), and perfluorotetradecanoic acid (PFTeDA; C14). The target PFASs were potassium perfluorobutanesulfonate (PFBS; C4), sodium perfluorohexanesulfonate (PFHxS; C6), sodium perfluoroheptanesulfonate (PFHpS; C7), sodium perfluorooctanesulfonate (PFOS; C8), and sodium perfluorodecanesulfonate (PFDS; C10), and the target PFSAs were perfluorooctane sulfonamide (PFOSA), N-methylperfluorooctane sulfonamide (N-MeFOSA), and N-ethylperfluorooctane sulfonamide (N-EtFOSA). Wastewater samples were filtered after collection and extracted/purified/pre-concentrated by a solid-phase extraction (SPE) procedure. Particulate matter and sludge samples were extracted by a liquid–solid extraction (LSE) and ultra-sonication procedure, and thereafter purified/preconcentrated by the same SPE procedure that was followed for the dissolved phase of wastewater. The internal standards, perfluoro-n-[1,2,3,4-13C4]octanoic acid (13C4-PFOA), sodium perfluoro-1-[1,2,3,4-13C4]octanesulfonate (13C4-PFOS), and N-methyl-d3-perfluoro-1-octanesulfonamide (2D3-N-MeFOSA) provided adequate compensation for variations in the extraction percentages and instrumental response. The limits of quantification (LOQs) ranged from 0.29 (PFHpS) to 3.0 ng L−1 (PFDoA) for dissolved phase samples, and from 0.15 (PFHpS) to 1.5 ng g−1 dry weight (dw) (PFDoA) for particulate matter and sludge samples. The developed methods were applied successfully to wastewater and sludge samples originated from Athens WWTP. PFC concentrations up to 113 ng L−1 (PFUdA), 33 ng L−1 (PFOA) and 1042 ng g−1 (PFUdA) were determined in influent wastewater, treated wastewater and dewatered sludge, respectively. Analysis of PFCs in the particulate matter of wastewater is needed to avoid underestimation of their concentrations.

Embargo until 20 April 2019. LC-HR-QTOF-MS recently has become a commonly used approach for the analysis of complex samples. However, identification of small organic molecules in complex samples with the highest level of confidence is a challenging task. Here we report on the implementation of a two stage algorithm for LC-HR-QTOF-MS datasets. We compared the performances of the two stage algorithm, implemented via NIVA MZ AnalyzerTM, with two commonly used approaches (i.e. feature detection and XIC peak picking, implemented via UNIFI by Waters and TASQ by Bruker, respectively) for the suspect analysis of four influent wastewater samples. We first evaluated the cross platform compatibility of LC-HR-QTOF-MS datasets generated via instruments from two di erent manufacturers (i.e. Waters and Bruker). Our data showed that with an appropriate spectral weighting function the spectra recorded by the two tested instruments are comparable for our analytes. As a consequence, we were able to perform full spectral comparison between the data generated via the two studied instruments. Four extracts of wastewater influent were analyzed for 89 analytes, thus 356 detection cases. The analytes were divided into 158 detection cases of artificial suspect analytes (i.e. verified by target analysis) and 198 true suspects. The two stage algorithm resulted in a zero rate of false positive detection, based on the artificial suspect analytes while producing a rate of false negative detection of 0.12. For the conventional approaches, the rates of false positive detection varied between 0.06 for UNIFI and 0.15 for TASQ. The rates of false negative detection for these methods ranged between 0.07 for TASQ and 0.09 for UNIFI. The effect of background signal complexity on the two stage algorithm was evaluated through the generation of a synthetic signal. We further discuss the boundaries of applicability of the two stage algorithm. The importance of background knowledge and experience in evaluating the reliability of results during the suspect screening was evaluated.

Food analysis is continuously requiring the development of more robust, efficient, and cost-effective food authentication analytical methods to guarantee the safety, quality, and traceability of food commodities with respect to legislation and consumer demands. Hence, a novel reversed-phase ultra high performance liquid chromatography–electrospray ionization quadrupole time of flight tandem mass spectrometry analytical method was developed that uses target, suspect, and nontarget screening strategies coupled with advanced chemometric tools for the investigation of the authenticity of extra virgin olive oil. The proposed method was successfully applied in real olive oil samples for the identification of markers responsible for the sensory profile. The proposed target analytical method includes the determination of 14 phenolic compounds and demonstrated low limits of detection ranging from 0.015 μg mL-1 (apigenin) to 0.039 μg mL-1 (vanillin) and adequate recoveries (96–107 %). A suspect list of 60 relevant compounds was compiled, and suspect screening was then applied to all the samples. Semiquantitation of the suspect compounds was performed with the calibration curves of target compounds having similar structures. Then, a nontarget screening workflow was applied with the aim to identify additional compounds so as to differentiate extra virgin olive oils from defective olive oils. Robust classification-based models were built with the use of supervised discrimination techniques, partial least squares–discriminant analysis and counterpropagation artificial neural networks, for the classification of olive oils into extra virgin olive oils or defective olive oils. Variable importance in projection scores were calculated to select the most significant features that affect the discrimination. Overall, 51 compounds were identified and suggested as markers, among which 14, 26, and 11 compounds were identified by target, suspect, and nontarget screening respectively. Retrospective analysis was also performed and identified 19 free fatty acids.

Two liquid chromatography-tandem mass spectrometric methods were developed and validated to determine spinosyn A and D, thiacloprid and pyridalyl in spring onions cultivated under Egyptian field conditions. The degradation rates, the pre-harvest interval (PHI) values and the half-life values of the three pesticides were estimated. QuEChERS was used for sample preparation and the separation was performed on an X-Bridge C18 column with ACN-formic acid 0.1% as the mobile phase. Linear range, method detection limits (MDLs), precision, recovery and matrix effects were estimated. The multi-residue MDLs ranged from 0.02μg/kg (spinosyn A & D) to 0.05μg/kg for pyridalyl. All the investigated pesticides showed high degradation rates. For spinosad the half-life value was 1.2days, for thiacloprid it reached 2.2days and for pyridalyl 4.4days. Furthermore, the calculated PHI values, according to the maximum residue levels set by the EU, were 0days for spinosad, 9.8days for thiacloprid and 39.4days for pyridalyl.

A multiresidue/multiclass method for the simultaneous determination of 76 veterinary drugs and pharmaceuticals in bovine muscle tissue has been developed and validated according to the requirements of European Commission Decision 2002/657/EC. The analytes belong in 13 different classes, including aminoglycoside antibiotics, whose different physicochemical properties (extremely polar character) render their simultaneous determination with other veterinary drugs quite problematic. The method combines a two-step extraction procedure (extraction with acetonitrile followed by an acidic aqueous buffer extraction) with hydrophilic interaction liquid chromatography-tandem mass spectrometry (HILIC-MS/MS) determination, allowing confirmation and quantification in a single chromatographic run. Further cleanup with solid phase extraction was performed using polymeric SPE cartridges. A thorough ionization study of aminoglycosides was performed in order to increase their sensitivity and significant differences in the abundance of the precursor ions of the analytes were revealed, depending on the composition of the mobile phase tested. Further gradient elution optimization and injection solvent optimization were performed for all target analytes.The method was validated according to the European Commission Decision 2002/657. Quantitative analysis was performed by means of standard addition calibration. Recoveries varied from 37.4% (bromhexine) to 106% (kanamycin) in the lowest validation level and 82% of the compounds showed recovery >70%. Detection capability (CCβ) varied from 2.4 (salinomycin) to 1302 (apramycin) μgkg(-1).

After consumption, pharmaceuticals are excreted as parent compounds and/or metabolites in urine and faeces. Some are not completely removed during wastewater treatments, forcing sewage treatment plants (STPs) to apply alternative technologies to guarantee quality of treated water. To monitor the removal efficiency of STPs, not only unchanged compounds and metabolites have to be taken into account, but also formation of possible transformation products (TPs). In this work, QTOF MS has been used for screening metabolites/TPs of pharmaceuticals in effluent wastewater from Athens. A customised database was built with the exact masses of metabolites reported in literature for the parent drugs found in an initial screening. Additionally, TPs identified in previous degradation experiments performed at our laboratory were included. Up to 34 metabolites/TPs were detected for omeprazole, venlafaxine, clindamycin, clarithromycin, clopidogrel or dipyrone, among others. Seven corresponded to TPs whose reference standards were available at our lab, seven were TPs previously identified in laboratory degradation experiments, eight were TPs tentatively identified by QTOF MS without reference standards, and twelve TPs were discovered after using the common fragmentation pathway approach. Tentative identification of TPs was supported by prediction of their chromatographic retention time based on the use of advanced chemometric QSRR models. The retention behavior of melamine (MEL), ammeline (AMN), ammelide, and cyanuric acid on various hydrophilic interaction liquid chromatography (HILIC) stationary phases was studied. Using fully optimized electrospray conditions and ammonium formate buffer pH 4/acetonitrile (20:80 v/v) as a mobile phase, precursor and product ions for each compound were identified. For the LC separation study, the effect of the following parameters was investigated: the type of buffer, the effect of the pH of ammonium formate buffer, the dilution percentage of the standard solutions in the vial, the stability of the standard solutions, and the column equilibration time. The retention and separation of melamine and its hydrolysis products on several HILIC columns was also investigated and retention models were proposed. Retention mechanisms were discussed for all the compounds. When the percentage of acetonitrile in the mobile phase was increased, the retention times of ammelide, ammeline and melamine were shifted to higher values, while the retention time of cyanuric acid did not change significantly. The separation of compounds with isobaric transitions (MEL, AMN) was achieved on four HILIC columns, namely TSKgel Amide-80, Luna HILIC, XBridge HILIC, and ZIC-HILIC (at either 10/90 or 15/85 ammonium formate buffer pH 4/ACN).

A rapid and sensitive method for the direct determination of hexavalent chromium in natural waters has been developed. Anion exchange chromatography was used for the fast separation of chromates from matrix anions, within 15 minutes, using a 35 mM KOH eluant. Mobile phase suppression was employed prior to direct conductivity (CD) detection, comparing two different electrolytic suppressor models and achieving a 10 times lower detection limit. Post column derivatization of Cr(VI) with diphenylcarbazide was used for further selective diode array (PDA) detection. Possible Cr(III) interference was evaluated with the addition of concentrations up to 10 mg/L of Cr(III) in a Cr(VI) positive natural water sample resulting in negligible interference. Both detection techniques gave instrumental LOD of 0.05 µg/L and method LOD of 0.08 µg/L for CD and 0.05 µg/L for PDA detection in underground water. The fate of four benzotriazoles [1-H-benzotriazole (1-H-BTRi), tolyltriazole (TTRi), xylyltriazole (XTRi) and 1-hydroxy-benzotriazole (1-OH-BTRi)] and three benzothiazoles [benzothiazole (BTH), 2-hydroxy-benzothiazole (2-OH-BTH) and 2-amino-benzothiazole (2-amino-BTH)], during chlorination batch experiments was investigated. In the first step, their degradation under different experimental conditions (applied molar ratio of NaOCl and the target contaminant (m.r.), reaction’s contact time, pH value of the reaction’s solution and the influence of total suspended solids (TSS) presence) was investigated and their removal kinetics parameters (kobs and t1/2) were determined. In the second step, LC-QTOFMS/MS was used for the detection and identification of transformation products (TPs) formed during chlorination, through the application of suspect and non-target screening approaches. Four and five TPs of XTRi and 2-amino-BTH, respectively, were detected and tentatively identified, while 1-H-BTRi was proven to be formed by the chlorination of 1-OH-BTRi. Moreover, since the identified TPs were also detected in spiked wastewater samples, after lab-scale chlorination experiments, toxicity assessment was carried out by ECOSAR calculations for the environmental relevance of their occurrence. The proposed chlorinated TPs were proven to be more toxic than their parent compounds.

This study focuses on the effect of experimental parameters on the removal of ranitidine (RAN) during ozonation and the identification of the formed transformation products (TPs). The influence of pH value, the initial concentrations, the inorganic and the organic matter on RAN's removal were evaluated. Results indicated high reactivity of RAN with molecular aqueous ozone. Initial ozone concentration and pH were proven the major process parameters. Alkaline pH values promoted degradation and overall mineralization. Dissolved organic matter reacts competitively to RAN with the oxidants (ozone and/or radicals), influencing the target compound's removal. The presence of inorganic ions in the matrix did not seem to affect RAN ozonation. A total of eleven TPs were identified and structurally elucidated, with the complementary use of both Reversed Phase (RP) and Hydrophilic Interaction Liquid Chromatography (HILIC) quadrupole time of flight tandem mass spectrometry (Q-ToF-MS/MS). Most of the TPs (TP-304, TP-315b, TP-299b, TP-333, TP-283) were generated by the attack of ozone at the double bond or the adjacent secondary amine, with the abstraction of NO2 moiety, forming TPs with an aldehyde group and an imine bond. Oxidized derivatives with a carboxylic group (TP-315a, TP-331a, TP-331b, TP-299a) were also formed. RAN S-oxide was identified as an ozonation TP (TP-330) and its structure was confirmed through the analysis of a reference standard. TP-214 was also produced during ozonation, through the CN bond rupture adjacent to the NO2 moiety. HILIC was used complementary to RP, either for the separation and identification of TPs with isomeric structures that may have been co-eluted in RPLC or for the detection of new TPs that were not eluted in the RP chromatographic system. Retention time prediction was used as a supporting tool for the identification of TPs and results were in accordance with the experimental ones in both RP and HILIC.

The determination of tin in canned food became very important in the last few years since it gives information about the contamination process helping to increase canned food quality and safety. The present work describes the development of a method for the determination of tin in canned tomato paste by electrothermal atomic absorption spectrometry in order to investigate the influents of (1) the industrial sample pretreatment procedure of canned tomato paste and (2) the type of materials used in packaging, on the total concentration of tin in canned tomato paste samples. For method validation, precision (repeatability and reproducibility) and accuracy (by recovery tests) have been assessed as performance criteria. The calculated recoveries ranged between 87.9% and 91.9%. The calculated expanded uncertainties based on the EURACHEM/CITAC Guidelines achieved from three different concentration levels were also calculated. The obtained results indicated that the method is efficiently and properly implemented. The validated method was applied for tin concentration determination in canned tomato paste purchased from the local market. In all analyzed samples, tin concentrations levels were lower than the maximum permissible value recommended by the European Regulation.
