Cholesterol has been shown to be present in rat liver chromatin isolated by methods designed to avoid contamination by membrane fragments. Evidence that the cholesterol was actually a component of chromatin includes (a) the constancy of the amount (1.30 +/- 0.14 mug per mg DNA), (b) the striking difference in the ratio of free (i.e. unesterified) to esterified cholesterol between that in chromatin and that in membrane, and (c) the rapid and marked changes which occurred in this ratio during the circadian cycle in chromatin but not in membranes. Although the total amount of chromatin-bound cholesterol did not change throughout the circadian cycle, the concentration of free cholesterol increased sharply a short time before the peak of cholesterol synthetic activity was reached at about midnight; it reached a basal level about 6 h later at approximately the same time the rate of synthesis returned to its basal level. When labelled cholesterol was administered by stomach tube, it was detectable within 2 h in whole nuclei and in chromatin, indicating that chromatin-bound cholesterol is rapidly exchangeable with that in liver cytoplasm and in blood plasma. Removal of basic proteins from chromatin did not result in the loss of any cholesterol, but removal of most of the acidic as well as the basic proteins resulted in loss of most of the chromatin-bound cholesterol. These results indicate that cholesterol is bound either to the acidic proteins or to both the acidic proteins and DNA. The data are compatible with the hypothesis that cholesterol biosynthesis controlled at the nuclear level and suggest that the relative amounts of free and esterified cholesterol associated with chromatin may play a role.

An inhibition in the conversion of mevalonate to cholesterol has been demonstrated in liver of cholesterol-fed rats by both in vitro and in vivo methods. Synthesis decreased to 30% of the control value after 1 week and 20% after 1 month on a 1% cholesterol diet. After a year, synthesis from mevalonate was almost completely inhibited. The rate of conversion of squalene to cholesterol was not consistently decreased but that of farnesyl pyrophosphate to cholesterol was decreased considerably. The rate of conversion of mevalonate to farnesyl pyrophosphate by a soluble liver enzyme preparation was also decreased in cholesterol-fed animals. Sites of inhibition of cholesterol synthesis were detected before mevalonate, between mevalonate and farnesyl pyrophosphate, and after farnesyl pyrophosphate, probably at the conversion of farnesyl pyrophosphate to squalene. The inhibition of mevalonate conversion to cholesterol developed more slowly than that of acetate and appeared to be secondary to it. The maximum capacities of normal liver homogenates and slices to synthesize cholesterol from mevalonate were shown to be far greater than from acetate. Consequently, sites of inhibition after mevalonate probably do not have a significant effect on the over-all rate of cholesterol synthesis in the intact cholesterol-fed animal.

The existence of a circadian rhythm in the rate of hepatic cholesterol synthesis in the rat has been demonstrated in vivo by measuring the conversion of both [1-(14)C]acetate and (3)H(2)O to cholesterol. By both methods there was observed a similar increase in the rate of hepatic cholesterol synthesis between the nadir at noon and the peak at midnight. Circadian changes in the rate of hepatic cholesterol synthesis measured in vivo with [1-(14)C]acetate were very similar to changes in the activity of hepatic microsomal HMG CoA reductase. Cholesterol synthesis in the jejunum and in the distal ileum was also shown to exhibit the same circadian rhythm in vivo but with smaller amplitude (1.6- and 1.3-fold, respectively). Rats trained to eat during a 4-hr period (9 am-1 pm) while housed under normal illumination showed changes in the timing of the circadian rhythm of cholesterol synthesis; in the liver the maximum rate of cholesterol synthesis occurred at 6 pm, 9 hr after the presentation of food, while the two sections of the intestine investigated exhibited a maximum synthetic response between noon and 6 pm. Results obtained in this study support the hypothesis that the major portion of the rise in HMG CoA reductase activity and the increase in overall rate of cholesterol synthesis in liver and intestine during the circadian rhythm are due to the ingestion of food. Under the limited feeding schedule (food access 9 am-1 pm), the rates of hepatic and intestinal synthesis of fatty acids from the injected acetate also showed a circadian rhythm with a peak at about 3 hr after presentation of food.
