Phytoferritin was explored as an attractive nanocarrier to encapsulate bioactive compounds due to its excellent stability and biocompatibility. In the present study, a novel phytoferritin derived from alfalfa (Medicago sativa) was successfully expressed, purified and characterized. Results confirmed that alfalfa ferritin, self-assembled by 24 subunits, formed a spherical hollow structure. Baicalein exhibits superior antioxidant properties and nutritious values, but low bioavailability and solubility limit its application. Herein, we fabricated water-soluble chitosan-ferritin-baicalein nanoparticles to overcome its drawbacks. It was calculated that one apoferritin cage could encapsulate 52 molecules of baicalein. Moreover, chitosan-ferritin-baicalein nanoparticles prolonged the release of baicalein in simulated gastrointestinal tract digestion. Caco-2 cell monolayer absorption analysis demonstrated that baicalein encapsulated within ferritin-chitosan double layers was more efficient in cellular transportation. The effect of chitosan decoration on the transport of epigallocatechin (EGC)-encapsulated ferritin cage across the Caco-2 cells was investigated. After the encapsulation of EGC in apo-red bean (adzuki) ferritin (apoRBF), the EGC-loaded apoRBF nanoparticle (ER) was fabricated with an encapsulation ratio of 11.6% (w/w). The results indicated that different chitosan molecules (with molecular weights of 980, 4600, 46 000, and 210 000 Da) could attach onto the apoRBF via electrostatic interactions to form ER-chitosan complexes (ERCs) (ERCs980, ERCs4600, ERCs46000, and ERCs210000). ERCs980 and ERCs4600 retained the typical shell-like morphology of ferritin with a size distribution of 12 nm and showed weak negative zeta-potentials at pH 6.7, while ERCs46000 and ERCs210000 significantly aggregated. Furthermore, the transport of EGC in ERCs980 and ERCs4600 across the Caco-2 cells was enhanced by the transferrin receptor 1 (TfR-1)-mediated absorption pathway, demonstrating that chitosan molecules with low molecular weights of 980 and 4600 Da were beneficial to the absorption of EGC based on the ferritin cage. This study will facilitate the application of ferritin-chitosan materials for fabricating the core-shell platform for encapsulation and bioavailability enhancement of bioactive molecules.

Chitosan oligosaccharides longer than a hexamer have higher bioactivity than polymer or shorter oligosaccharides, such as the monomer or dimer. In our previous work, we generated Paenibacillus fukuinensis chitosanase-displaying yeast using yeast cell surface displaying system and demonstrated the catalytic base. Here we investigated the specific function of putative four amino acid residues Trp159, Trp228, Tyr311, and Phe406 engaged in substrate binding. Using this system, we generated chitosanase mutants in which the four amino acid residues were substituted with Ala and the chitosanase activity assay and HPLC analysis were performed. Based on these results, we demonstrated that Trp159 and Phe406 were critical for hydrolyzing both polymer and oligosaccharide, and Trp228 and Tyr311 were especially important for binding to oligosaccharide, such as the chitosan-hexamer, not to the chitosan polymer. From the results, we suggested the possibility of the effective strategy for designing useful mutants that produce chitosan oligosaccharides holding higher bioactivity.

The DNA-chitosan polyplexes have attracted for some years now the attention of physical-chemists and biologists for their potential use in gene therapy, however, the correlation between the physicochemical properties of these polyplexes with their transfection efficiency remains still unclear. In a recent paper we demonstrated by means of DLS that the DNA-chitosan complexation is favored at acidic conditions considering that fewer amounts of chitosan were required to compact the DNA. As a second study, in the present work we analyze the influence of chitosan valence on the complexation and transfection of DNA. Three chitosans of different molecular weights (three different valences) are characterized as gene carriers at 25°C and pH 5 over a wide range of chitosan-Nitrogen to DNA-Phosphate molar ratios, N/P, by means of conductometry, electrophoretic mobility, isothermal titration calorimetry (ITC), transmission electron microscopy (TEM), atomic force microscopy (AFM), and β-galactosidase and luciferase expression assays., (Copyright © 2010 Elsevier B.V. Skin wound healing is an important clinical challenge, and the main treatment points are accelerating epidermal regeneration and preventing infection. Therefore, it is necessary to develop a wound dressing that can simultaneously cure bacterial infections and accelerate wound healing. Here, we report a multifunctional composite wound dressing loaded with chitosan (CS)-binding bFGF (CSBD-bFGF) and antimicrobial peptides (P5S9K). First, CS was used as the dressing matrix material, and P5S9K was encapsulated in CS. Then, CSBD-bFGF was designed by combining recombinant DNA technology and tyrosinase treatment and modified on the dressing material surface. The results show that the binding ability of CSBD-bFGF and CS was significantly improved compared with that of commercial bFGF, and CSBD-bFGF could be controllably released from the CS dressing. More importantly, the prepared dressing material showed excellent antibacterial activity in vivo and in vitro and could effectively inhibit the growth of E. coli and S. aureus. Using NIH3T3 cells as cellular models, the results showed that the CSBD-bFGF@CS/P5S9K composite dressing was a friendly material for cell growth. After cells were seeded on the composite dressing surface, collagen-1 (COL-1) and vascular endothelial growth factor (VEGF) genes expression in cells were significantly upregulated. Finally, the full-thickness wound of the rat dorsal model was applied to analyse the tissue repair ability of the composite dressing. The results showed that the composite dressing containing CSBD-bFGF and P5S9K had the strongest ability to repair skin wounds. Therefore, the CSBD-bFGF@CS/P5S9K composite dressing has good antibacterial and accelerated wound healing abilities and has good application prospects in the treatment of skin wounds.

In insects, cuticle proteins interact with chitin and chitosan of the exoskeleton forming crystalline, amorphic or composite material structures. The biochemical and mechanical composition of the structure defines the cuticle's physical properties and thus how the insect cuticle behaves under mechanical stress. The tissue-specific ratio between chitin and chitosan and its pattern of deacetylation are recognized and interpreted by cuticle proteins depending on their local position in the body. Despite previous research, the assembly of the cuticle composites in time and space including its functional impact is widely unexplored. This review is devoted to the genetics underlying the temporal and spatial distribution of elastic proteins and the potential function of elastic proteins in insects with a focus on Resilin in the fruit fly Drosophila. The potential impact and function of localized patches of elastic proteins is discussed for movements in leg joints, locomotion and damage resistance of the cuticle. We conclude that an interdisciplinary research approach serves as an integral example for the molecular mechanisms of generation and interpretation of the chitin/chitosan matrix, not only in Drosophila but also in other arthropod species, and might help to synthesize artificial material composites., (Copyright © 2024 The Authors. The formation of electrostatic interactions between polyanionic siRNA and polycations gives an easy access to the formation of colloidal particles capable of delivering siRNA in vitro or in vivo. Among the polycations used for siRNA delivery, chitosan occupies a special place due to its unique physicochemical and biological properties. In this chapter we describe the fundamental and practical aspects of the formation of colloidal complexes between chitosan and siRNA. The basis of the electrostatic complexation between oppositely charged polyelectrolytes is first introduced with a focus on the specific conditions to obtain stable colloid complex particles. Subsequent, the properties that make chitosan so special are described. In a third part, the main parameters influencing the colloidal properties and stability of siRNA/chitosan complexes are reviewed with emphasis on some practical aspects to consider in the preparation of complexes.

To know the mechanism of ammonia assimilation in non-heading Chinese cabbage (Brassica campestrishL. ssp.chinensis(L.) Makino) leaves regulated by chitosan (CTS), a CTS-binding protein was isolated from non-heading Chinese cabbage leaves using the chitosan affinity chromatography approach and this CTS-binding protein was partially characterized. The profile of the 53.1 kDa purified protein on sodium dodecyl sulfate-polyacrylamide gel electrophoresis was compared with the native molecular weight of 106.5 kDa, which indicated that the purified protein was a dimer with identical subunits. After isoelectric focusing, a band was obtained at pH 8.25. The agglutination test and periodic acid-Schiff staining further revealed that the protein was a glycoprotein with lectin activity. Moreover, the purified protein contained 17.4% (w/w) neutral carbohydrate and 82.56% (w/w) protein. The comparison of this protein and the 67 kDa CTS-binding protein isolated previously fromRubusculture tissue exhibited some differences in characterization. According to results of peptide mass fingerprinting analysis, the protein purified in the present study does not show any similarity with any protein in the protein data bank. Chitosan-related enzymes including chitosanases, exo-β-glucosaminidases, and enzymes having chitosan-binding modules recognize ligands through electrostatic interactions between the acidic amino acids in proteins and amino groups of chitosan polysaccharides. However, in GH8 chitosanases, several aromatic residues are also involved in substrate recognition through stacking interactions, and these enzymes consequently hydrolyze β-1,4-glucan as well as chitosan. The binding grooves of these chitosanases are extended and opened at both ends of the grooves, so that the enzymes can clamp a long chitosan polysaccharide. The association/dissociation of positively charged glucosamine residues to/from the binding pocket of a GH2 exo-β-glucosaminidase controls the p K a of the catalytic acid, thereby maintaining the high catalytic potency of the enzyme. In contrast to chitosanases, chitosan-binding modules only accommodate a couple of glucosamine residues, predominantly recognizing the non-reducing end glucosamine residue of chitosan by electrostatic interactions and a hydrogen-bonding network. Chitin deacetylation is required to make the cuticle rigid and compact through chitin chain crosslinking. Thus it is presumed that specialized proteins are required to bind deacetylated chitin chains together. However, deacetylated-chitin binding proteins have not ever been reported. In a previous work, six cuticular proteins analogous to peritrophin 3 (CPAP3s) were found to be abundant in the moulting fluid of Bombyx mori. In this study, these BmCPAP3s (BmCPAP3-A1, BmCPAP3-A2, BmCPAP3-B, BmCPAP3-C, BmCPAP3-D1 and BmCPAP3-D2) were cloned and expressed in Escherichia coli and purified using metal-chelating affinity chromatography. Their binding activities demonstrated that although all of the BmCPAP3s showed similar binding abilities toward crystalline chitin and colloidal chitin, they differed in their affinities toward partially and fully deacetylated chitin. Amongst them, BmCPAP3-D1 exhibited the highest binding activity toward deacetylated chitin. The gene expression pattern of BmCPAP3-D1 was similar to BmCPAP3-A1 and BmCPAP3-C at most stages except that it was dramatically upregulated at the beginning of the pupa to adult transition stage. Background and purpose: Activated textiles have created a new field of application apart from standard textile applications. Novel, functions, for example, antimicrobial properties have been enhanced in textiles. New textile application fields have been recognized for particular areas of healthcare as well as industry, sportswear, and, home textiles. Advances in the field of antibacterial textiles have fascinated enhancing attention because those are a means to solve serious health challenges such as bacterial overgrowth. A lot of research has been done in the field of adding synthetic and toxic materials to fabrics, such as nanosilver triclosan or other metals and organic materials to produce antimicrobial fabrics. These fabrics kill bacteria by binding to intracellular proteins and inactivating these proteins. Also, by using these chemicals, the content of heavy metals in these textiles can be increased. The application of heavy metals in antimicrobial textiles is limited by many environmental tags and standards. Therefore, it is recommended to use natural antimicrobial agents, such as nisin and chitosan to produce environmentally friendly and safe antimicrobial fabrics. Nowadays finishing process with the natural antibacterial agents that protect the environment and human health is gaining importance. Therefore, this study aims to produce fabric coated with chitosan and nisin and to investigate its antimicrobial properties. Materials and methods: In this research, chitosan was used to bind nisin to the surface of polypropylene span band non-woven fabric. The fabric samples were coated with a solution containing chitosan and nisin using a pad-dry-cure method. The appearance, color, and softness of the coated fabrics were compared to the uncoated fabric. The nisin and chitosan binding properties on the fabric were explored by using the Fouriertransform infrared (FTIR( technique. The diffusion method in agar and the standard method (ASTME2149-01) were used to check the antimicrobial properties of fabric samples. The antibacterial effectiveness of fabric samples was tested against common bacteria such as Escherichia coli, Pseudomonas aeruginosa, Staphylococcus aureus, Bacillus cereus, Listeria monocytogenes, and Enterococcus faecalis. After ten washing cycles, the durability of the antimicrobial properties of these fabrics was checked by the method mentioned above. Also, the toxicity of this fabric on fibroblast cells was determined using the MTT colorimetry method after seven days. Results: Fabrics coated with chitosan and nisin do not differ much from the control sample in terms of appearance and softness. The changes observed in the FTIR spectrum, such as the appearance of new absorption peaks and additional peaks associated with peptide bonds, confirm the successful incorporation of nisin and chitosan into the fabric. Antibacterial results have shown that nisin improved the antibacterial effect of coated fabrics versus B. cereus, S. aureus, and P. aeruginosa. The antimicrobial properties of the fabric coated with chitosan-nisin were maintained at some of the efficacy after washing against B. cereus, S. aureus, E. faecalis, L. monocytogenes, E. coli, and P. aeruginosa respectively. Also, the chitosan-nisin coating showed that it did not cause significant toxicity in fibroblast cells even after one week. Conclusion: However, the results showed that the chitosan/nisin coating might be used for medical textiles and antibacterial textiles. Previously, we reported properties of a glycosylase belonging to GH-8 glycosyl hydrolase (GH) and having both chitosanase and glucanase activities. This enzyme (D2), whose molecular mass (86 kDa) was the largest among the GH-8 group, has its catalytic domain at the N-terminal region, and discoidin domain (DD) at the C-terminal region. Although various chitosanases, chitinases and glucanases have been known, DD is unique to the D2 enzyme. Glucanase and chitinase, but not chitosanase, are known to have functional domain such as carbohydrate-binding module, besides catalytic domain. Accordingly, function of the DD of D2 chitosanase was analyzed, using zygomycete cell wall containing chitosan, glucan and chitin as the basic constituents. The DD specifically and tightly bound to chitosan, but did not participate in affinity for glucan and chitin. Deletion of the DD caused marked reduction in absorbability to cell wall and in hydrolytic activity toward chitosan and glucan. These results suggest that the DD is concerned in binding of the enzyme to cell wall and in effective digestion of the insoluble substrate, through hydrolysis of not only chitosan but also coexisting glucan. Chitosan/fucoidan nanoparticles were created using two fucoidans from the Fucus evanescens algae. One of them was a regular fucoidan obtained for the first time from the alga harvested at the reproductive growth stage, using only standard extraction methods, without additional modifications. Its structure was established via NMR spectroscopy to consist of the repeating →3)-α-L-Fucp-(2,4SO 3 - )-(1→ fragment. Such fragment also coustituted 55% of the other fucoidan's structure, however it also included long sequences of α-L-fucopyranose residues sulfated only at C2. The nanoparticles were re-dispersed in water and the influence of fucoidan/chitosan mass ratio on the nanoparticles' size and zeta potential was investigated. 3D models of the regular fucoidan and chitosan's sections were created and their molecular docking was performed, showing that either polymer could occupy the exterior of the complex, depending on their ratio. The functionality of halloysite (Hal) nanotubes as drug carriers can be improved by lumen enlargement and polymer modification. This study investigates the influence of selective acid etching on Hal functionalization with cationic biopolymer chitosan. Hal was subjected to lumen etching under mild conditions, loaded under vacuum with nonsteroidal antiinflammatory drug aceclofenac, and incubated in an acidic solution of chitosan. The functionality of pristine and etched Hal before and upon polymer functionalization was assessed by ζ-potential measurements, structural characterization (FT-IR, DSC and XRPD analysis), cell viability assay, drug loading and drug release studies. Acid etching increased specific surface area, pore volume and pore size of Hal, decreased ζ-potential and facilitated binding of the cationic polymer. XRPD and DSC analysis revealed crystalline structure of etched Hal. Successful chitosan binding and drug entrapment were further confirmed by FT-IR and DSC studies. XRPD showed surface polymer binding. DSC and FT-IR analyses confirmed the presence of the entrapped drug in its crystalline form. Drug loading was increased for ≈81% by selective lumen etching. Slight decrease of drug content occurred during chitosan functionalization due to aceclofenac diffusion in the polymer solution. The drug release was more sustained from etched Hal nanocomposites (up to ≈87% for 12 h) than from pristine Hal (up to ≈97% for 12 h) due to more intensive chitosan binding. High human fibroblast survival rates upon exposure to pristine and etched Hal before and after chitosan functionalization (>90% in the concentration of 1000 μg/mL) confirmed that both lumen etching under mild conditions and polymer functionalization had no significant effect on cytocompatibility. Based on these findings, selective lumen etching in combination with polycation modification appears to be a promising approach for improvement of Hal nanotubes functionality by increasing payload, polymer binding capacity, and sustained release properties with no significant effect on their cytocompatibility., (Copyright © 2021 Elsevier B.V. Context: The main objective of root canal treatment is the removal of bacteria. Established medicaments and their combinations have been compromised in efficacy against Enterococcus faecalis , causing the need to explore novel intracanal medicaments. Aim: The aim of the study was to evaluate the antibacterial efficacy of chitosan, N -acetylcysteine (NAC), and Aegle marmelos as intracanal medicaments against E. faecalis biofilm. Materials and Methodology: Minimum inhibitory concentration and susceptibility of medicaments determined. Two hundred and forty dentin disc specimens were prepared and inoculated with E. faecalis for 21 days. Samples were divided into four groups (A – N -acetylcysteine; B – Aegle marmelos ; C – Chitosan; D –Control) (n = 60), and two subgroups (n = 30) based on the duration of medicament placed (subgroup 1: 24 h, subgroup 2: 7 days). Thereafter, dentinal shavings were retrieved, incubated in agar plate, visible colonies counted, and statistically analyzed. Results: At 24 h Group C1 exhibited the lowest CFUs, followed by Group A1, Group B, and Group D1. On the 7th day, Group B2 showed the lowest CFUs, followed by Group A2, Group C2, and Group D2. Conclusions: Against E. faecalis , NAC has the highest antimicrobial properties closely followed by Aegle marmelos and both provide promising novel possibilities for use as intracanal medicaments. Background/Objectives: Crickets are recognized as an alternative source of chitosan. This study aimed to assess the potential of cricket-derived chitosan as a natural source to develop chitosan nanoparticles (CNPs). Methods: Chitosan were isolated from different cricket species, including Gryllus bimaculatus, Teleogryllus mitratus, and Acheta domesticus. The isolated chitosan were characterized by their functional groups, crystallographic and thermal properties, molecular structure, morphology, water solubility, molecular weight, binding capacity, irritation potential, and cytotoxicity in comparison to commercial shrimp-based chitosan. CNPs were developed through an ionotropic gelation method, followed by the evaluation of particle size, polydispersity index (PDI), and zeta potential. Results: The findings of this study indicate that chitosan can be successfully isolated from the three cricket species, with yields ranging from 4.35% to 5.22% w/w of the dried material. The characteristics of cricket-based chitosan were similar to those of commercial chitosan, except that the cricket-based chitosan displayed a higher crystallinity and a lower molecular weight. Additionally, CPNs were successfully produced from cricket-based chitosan using sodium citrate as a crosslinking agent. All cricket-based chitosan exhibited no irritation or cytotoxicity. Chitosan derived from A. domesticus however was found to be the most suitable to develop CPNs, as it produced the smallest particle size (522.0 ± 12.1 nm) with a comparatively narrow PDI (0.388 ± 0.026) and an acceptable positive zeta potential (34.2 ± 4.4 mV). Conclusions: Cricket-derived chitosan compares favorably with crustacean-derived chitosan and showed potential for a range of applications, including the use as a nanocosmeceutical delivery system in topical and cosmetic formulations. Background and Objectives: A current public health issue is preventing post-surgical complications by designing antibacterial implants. To achieve this goal, in this study we evaluated the antibacterial activity of an animal-free chitosan grafted onto a titanium alloy., Methods: Animal-free chitosan binding on the substrate was performed by covalent link via a two-step process using TriEthoxySilylPropyl Succinic Anhydride (TESPSA) as the coupling agent. All grafting steps were studied and validated by means of X-ray Photoelectron Spectroscopy (XPS), Time-of-Flight secondary ion mass spectrometry (ToF-SIMS) analyses and Dynamic-mode Secondary Ion Mass Spectrometry (DSIMS). The antibacterial activity against Escherichia coli and Staphylococcus aureus strains of the developed coating was assessed using the number of colony forming units (CFU)., Results: XPS showed a significant increase in the C and N atomic percentages assigned to the presence of chitosan. A thick layer of polymer deposit was detected by ToF-SIMS and the results obtained by DSIMS measurements are in agreement with ToF-SIMS and XPS analyses and confirms that the coating synthesis was a success. The developed coating was active against both gram negative and gram positive tested bacteria., Conclusion: The success of the chitosan immobilization was proven using the surface characterization techniques applied in this study. The coating was found to be effective against Escherichia coli and Staphylococcus aureus strains.

Chitosan is a natural biopolymer that is industrially produced from chitin via deacetylation. Due to its unique properties and a plethora of biological activities, chitosan has found application in diverse areas from biomedicine to agriculture and the food sector. Chitosan is regarded as a biosafe, biodegradable, and biocompatible compound that was demonstrated to stimulate plant growth and to induce a general plant defense response, enhancing plant resistance to various pathogens, including bacteria, fungi, nematodes, and viruses. Here, we focus on chitosan application as an antiviral agent for plant protection. We review both the pioneer studies and recent research that report the effect of plant treatment with chitosan and its derivatives on viral infection. Special attention is paid to aspects that affect the biological activity of chitosan: polymer length and, correspondingly, its molecular weight; concentration; deacetylation degree and charge; application protocol; and experimental set-up. Thus, we compare the reported effects of various forms and derivatives of chitosan as well as chitosan-based nanomaterials, focusing on the putative mechanisms underlying chitosan-induced plant resistance to plant viruses. Native mucus is heterogeneous, displays high inter-individual variation and is prone to changes during harvesting and storage. To overcome the lack of reproducibility and availability of native mucus, commercially available purified mucins, porcine gastric mucin (PGM) and mucin from bovine submaxillary gland (BSM), have been widely used. However, the question is to which extent the choice of mucin matters in studies of their interaction with polymers as their composition, structure and hence physicochemical properties differ. Accordingly, the interactions between PGM or BSM with two widely used polymers in drug delivery, polyethylene oxide and chitosan, was studied with orthogonal methods: turbidity, dynamic light scattering, and quartz crystal microbalance with dissipation monitoring. Polymer binding and adsorption to the two commercially available and purified mucins, PGM and BSM, is different depending on the mucin type. PEO, known to interact weakly with mucin, only displayed limited interaction with both mucins as confirmed by all employed methods. In contrast, chitosan was able to bind to both PGM and BSM. Interestingly, the results suggest that chitosan interacts with BSM to a greater extent than with PGM indicating that the choice of mucin, PGM or BSM, can affect the outcome of studies of mucin interactions with polymers. An antifungal chitosanase/glucanase isolated from the soil bacterium Paenibacillus sp. IK-5 has two CBM32 chitosan-binding modules (DD1 and DD2) linked in tandem at the C-terminus. In order to obtain insights into the mechanism of chitosan recognition, the structures of DD1 and DD2 were solved by NMR spectroscopy and crystallography. DD1 and DD2 both adopted a β-sandwich fold with several loops in solution as well as in crystals. On the basis of chemical shift perturbations in(1)H-(15)N-HSQC resonances, the chitosan tetramer (GlcN)4 was found to bind to the loop region extruded from the core β-sandwich of DD1 and DD2. The binding site defined by NMR in solution was consistent with the crystal structure of DD2 in complex with (GlcN)3, in which the bound (GlcN)3 stood upright on its non-reducing end at the binding site. Glu(14)of DD2 appeared to make an electrostatic interaction with the amino group of the non-reducing end GlcN, and Arg(31), Tyr(36)and Glu(61)formed several hydrogen bonds predominantly with the non-reducing end GlcN. No interaction was detected with the reducing end GlcN. Since Tyr(36)of DD2 is replaced by glutamic acid in DD1, the mutation of Tyr(36)to glutamic acid was conducted in DD2 (DD2-Y36E), and the reverse mutation was conducted in DD1 (DD1-E36Y). Ligand-binding experiments using the mutant proteins revealed that this substitution of the 36th amino acid differentiates the binding properties of DD1 and DD2, probably enhancing total affinity of the chitosanase/glucanase toward the fungal cell wall., (© 2016 The Author(s). Background and objectives: A current public health issue is preventing post-surgical complications by designing antibacterial implants. To achieve this goal, in this study we evaluated the antibacterial activity of an animal-free chitosan grafted onto a titanium alloy. Methods: Animal-free chitosan binding on the substrate was performed by covalent link via a two-step process using TriEthoxySilylPropyl Succinic Anhydride (TESPSA) as the coupling agent. All grafting steps were studied and validated by means of X-ray Photoelectron Spectroscopy (XPS), Time-of-Flight secondary ion mass spectrometry (ToF-SIMS) analyses and Dynamic-mode Secondary Ion Mass Spectrometry (DSIMS). The antibacterial activity against Escherichia coli and Staphylococcus aureus strains of the developed coating was assessed using the number of colony forming units (CFU). Results: XPS showed a significant increase in the C and N atomic percentages assigned to the presence of chitosan. A thick layer of polymer deposit was detected by ToF-SIMS and the results obtained by DSIMS measurements are in agreement with ToF-SIMS and XPS analyses and confirms that the coating synthesis was a success. The developed coating was active against both gram negative and gram positive tested bacteria. Conclusion: The success of the chitosan immobilization was proven using the surface characterization techniques applied in this study. Background and purpose: Activated textiles have created a new field of application apart from standard textile applications. Novel, functions, for example, antimicrobial properties have been enhanced in textiles. New textile application fields have been recognized for particular areas of healthcare as well as industry, sportswear, and, home textiles. Advances in the field of antibacterial textiles have fascinated enhancing attention because those are a means to solve serious health challenges such as bacterial overgrowth. A lot of research has been done in the field of adding synthetic and toxic materials to fabrics, such as nanosilver triclosan or other metals and organic materials to produce antimicrobial fabrics. These fabrics kill bacteria by binding to intracellular proteins and inactivating these proteins. Also, by using these chemicals, the content of heavy metals in these textiles can be increased. The application of heavy metals in antimicrobial textiles is limited by many environmental tags and standards. Therefore, it is recommended to use natural antimicrobial agents, such as nisin and chitosan to produce environmentally friendly and safe antimicrobial fabrics. Nowadays finishing process with the natural antibacterial agents that protect the environment and human health is gaining importance. Therefore, this study aims to produce fabric coated with chitosan and nisin and to investigate its antimicrobial properties. Materials and methods: In this research, chitosan was used to bind nisin to the surface of polypropylene span band non-woven fabric. The fabric samples were coated with a solution containing chitosan and nisin using a pad-dry-cure method. The appearance, color, and softness of the coated fabrics were compared to the uncoated fabric. The nisin and chitosan binding properties on the fabric were explored by using the Fourier-transform infrared (FTIR (technique. The diffusion method in agar and the standard method (ASTME2149-01) were used to check the antimicrobial properties of fabric samples. The antibacterial effectiveness of fabric samples was tested against common bacteria such as Escherichia coli, Pseudomonas aeruginosa, Staphylococcus aureus, Bacillus cereus, Listeria monocytogenes, and Enterococcus faecalis. After ten washing cycles, the durability of the antimicrobial properties of these fabrics was checked by the method mentioned above. Also, the toxicity of this fabric on fibroblast cells was determined using the MTT colorimetry method after seven days. Results: Fabrics coated with chitosan and nisin do not differ much from the control sample in terms of appearance and softness. The changes observed in the FTIR spectrum, such as the appearance of new absorption peaks and additional peaks associated with peptide bonds, confirm the successful incorporation of nisin and chitosan into the fabric. Antibacterial results have shown that nisin improved the antibacterial effect of coated fabrics versus B. cereus, S. aureus, and P. aeruginosa. The antimicrobial properties of the fabric coated with chitosan-nisin were maintained at some of the efficacy after washing against B. cereus, S. aureus, E. faecalis, L. monocytogenes, E. coli, and P. aeruginosa respectively. Also, the chitosan-nisin coating showed that it did not cause significant toxicity in fibroblast cells even after one week. Conclusion: However, the results showed that the chitosan/nisin coating might be used for medical textiles and antibacterial textiles. It also offers an innovative solution to protect human health and the environment.

The roughness of woven fabric surface has so far been mainly investigated as a key characteristic of comfort in contact with the skin. The analysis of roughness can be extended to various contexts and applications, becoming an important tool for understanding how textile materials react in interaction with different finishing agents, as well as for gaining insight into the durability and effectiveness of treatments. This research presents a comprehensive study on the impact of alkaline hydrolysis and chitosan coating on the roughness of polyester woven fabric, utilizing both novel and adapted methods. The study employed contact and optical methods to analyze fabric and fiber surface characteristics, highlighting the significance of roughness profile parameters in understanding material changes post-treatment. The investigation revealed that mechanical action, alkaline medium, washing temperature, and detergent residues contribute to fabric erosion and modification during washing, with chitosan coatings creating pronounced surface irregularities. Comparative analysis showed significant fabric roughness changes post-washing, while fiber roughness changes were treatment specific. Despite initial increases in fiber roughness due to treatments, their durability decreased after washing. The study included the extraction of Chitin (Ch) from shrimp shells and preparation of Chitin nanoparticles (NCh) using acid hydrolysis and ultrasound method. In addition, the preparation of chitosan (Chs) and Chitosan nanoparticles (NChs) by Deacetylation method, then we study some of the characteristics, including yield, diameter by Size Analyzer, Field Emission Scanning Electron Microscopy (FESEM), Fourier transform infrared (FTIR) spectroscopy, XRay Diffractometer (X-RD), Viscosity, Water Binding Capacity (WBC) and Fat Binding Capacity (FBC). The results showed, (a) decreased the percentage of yield of NChs to 8.20%, (b) the effective diameter of the natural chitosan using a Size Analyzer and was 28121.5 nm, (c) FESEM analysis revealed that the NChs displayed a nanoscale structure with diameters ranged from 20.93 - 30.20 nm (d) the degree of deacetylation (DD%) using FT-IR spectroscopy device, of NChs was (80.21%) compared to Chs 71.45%, and in addition, from the X-ray spectrum, two peaks in spectra of (Ch, NCh) and (Chs, NChs) were observed at the diffraction angles (2θ) which are (10.70-10.99) and (19.99-20.37). Results show increasing the crystallization coefficient of NCh and NChs, while viscosity, WBC and FBC of NChs decreased compared to Chs. The aim of this study was to analyze binding of chitosan and chitosan nanoparticles onto cellulose via oxidized cellulose. The ability of chitosan and chitosan nanoparticles to be adsorbed onto surfaces was determined by the use of the XPS spectroscopy which provided information about chemical composition of the fiber surface. On the other hand, the gravimetric method was also used by which the amount of chitosan and chitosan nanoparticles bounded onto surface was calculated based on the difference in masses before and after functionalization. The most important was to study the influence of aldehyde groups on the stability of chitosan binding onto cellulose. Thus, desorption of chitosan/chitosan nanoparticles from the fiber surfaces was evaluated by the presence of total nitrogen (TN) in desorption bath as well as by polyelectrolyte titrations. Together with these two methods, desorption was evaluated also by gravimetric method, where the extent of desorption was evaluated on the basis of the differences in the masses of fibers before and after desorption. It is concluded that the chitosan and chitosan nanoparticles are more efficiently bounded onto oxidized cellulose in comparison with the nonoxidized (reference) ones. Despite the binding of the positively-charged amino groups with the negative groups of cellulose and consequently smaller amount of available/residual protonated amino groups that are responsible for bioactivity, such functionalized fibers are still specifically antimicrobial. Nanocomposites based on silver nanoparticles and chitosan present important advantages for medical applications, showing over time their role in antibacterial evaluation. This work presents the comparative study of two chemical synthesis procedures of nanocomposites, based on trisodium citrate dihydrate and sodium hydroxide, using various chitosan concentrations for a complex investigation. The nanocomposites were characterized by AFM and DLS regarding their dimensions, while FT-IR and UV–VIS spectrometry were used for the optical properties and to reveal the binding of silver nanoparticles with chitosan. Their antibacterial effect was determined using a disk diffusion method on two bacteria strains, E. coli and S. aureus. The results indicate that, when using both methods, the nanocomposites obtained were below 100 nm, yet the antibacterial effect proved to be stronger for the nanocomposites obtained using sodium hydroxide. Furthermore, the antibacterial effect can be related to the nanocomposites' sizes, since the smallest dimension nanocomposites exhibited the best bacterial growth inhibition on both bacteria strains we tested and for both types of silver nanocomposites. Tailorable cationic chitosan/PLGA nanoparticles (CPNP) were used for the delivery of an antisense 2'-O-methyl-RNA (2OMR) directed against RNA template of human telomerase. Here, we describe the influence of the chitosan content on binding efficiency, complex stability, uptake in different human lung cell types and finally demonstrate the efficacy of this nanoplex system. CPNPs were prepared by the emulsion-solvent evaporation method using different amounts of chitosan and purified by preparative size exclusion chromatography. The characterization by photon correlation spectroscopy and zeta potential measurements showed a small increase in size and an increase of zeta potential with increasing amounts of chitosan. Binding efficiency and complex stability with 2OMR was high in water and correlated well with the chitosan content of particles but was weak in physiologically relevant media (PBS and RPMI cell culture medium). However, flow cytometry analysis showed that the uptake of 2OMR into A549 lung cancer cells was considerably higher in combination with nanoparticles and dependent on the amount of chitosan when compared to 2OMR alone. Confocal laser scanning microscopy revealed that the uptake into A549 cells is mediated via complexes of 2OMR and chitosan/PLGA nanoparticles despite the weak binding in cell culture medium. The nanoparticles were well tolerated and efficient in inhibiting telomerase activity.

In modern conditions, the increase in the yield of agricultural crops is provided not by expanding the areas of their cultivation, but mainly by introducing advanced technologies. The most effective strategy for this purpose is the development of genetically resistant and productive cultivars in combination with the use of a variety of plant protection products (PPPs). However, traditional, chemical PPPs, despite their effectiveness, have significant drawbacks, namely, pollution of environment, ecological damage, toxicity to humans. Recently, biological PPPs based on natural compounds have attracted more attention, since they do not have these disadvantages, but at the same time they can be no less effective. One of such agents is chitosan, a deacetylation product of chitin, one of the most common polysaccharides in nature. The high biological activity, biocompatibility, and safety of chitosan determine the breadth and effectiveness of its use in medicine, industry, and agrobiology. The review considers various mechanisms of action of chitosan as a biopesticide, including both a direct inhibitory effect on pathogens and the induction of plant internal defense systems as a result of chitosan binding to cell surface receptors. The effect of chitosan on the formation of resistance to the main classes of pathogens: fungi, bacteria, and viruses has been shown on a variety of plant objects. The review also discusses various ways of using chitosan: for the treatment of seeds, leaves, fruits, soil, as well as its specific biological effects corresponding to these ways. A separate chapter is devoted to protection products based on chitosan, obtained by its chemical modifications, or by means of combining of a certain molecular forms of chitosan with various substances that enhance its antipathogenic effect. The data presented in the review generally give an idea of chitosan and its derivatives as very effective and promising plant protection products and biostimulants.

In weak acidic buffer medium, chitosan binding with an anionic surfactant, such as sodium dodecyl benzene sulphonate (SDBS), sodium lauryl sulphate (SLS) or sodium dodecyl sulphonate (SDS), can result in a significant enhancement of resonance Rayleigh scattering (RRS) intensity. The results showed that under optimum conditions the enhanced RRS intensity is proportional to the concentration of chitosan in the range 0.10-20.0 microg/mL for SDBS, 0.27-15.0 microg/mL for SLS and 0.20-15.0 microg/mL for SDS. Among these, the sensitivity of SDBS is the highest and its detection limit for chitosan is 29 ng/mL, while those of SLS and SDS are 83 and 61 ng/mL, respectively. The method has good selectivity and was applied to the determination of trace amounts of chitosan in practical samples with satisfactory results. Chitosan is a cationic biopolymer that has many potential applications in the food industry because of its unique nutritional and physicochemical properties. Many of these properties depend on its ability to interact with anionic surface-active molecules, such as surfactants, phospholipids, and bile acids. The purpose of this study was to examine the influence of pH (3 and 7), ionic strength (0-200 mM NaCl), and temperature (10-50 degrees C) on the interactions between a model anionic surfactant (sodium dodecyl sulfate, SDS) and chitosan using isothermal titration calorimetry, selective surfactant electrode, and turbidity measurements. At pH 3 and 30 degrees C, SDS bound strongly to chitosan to form an insoluble complex that contained about 4-5 mmol of SDS/1 g of chitosan at saturation. When SDS and chitosan were mixed at pH 7 they did not interact strongly, presumably because the biopolymer had lost most of its positive charge at this pH. However, when SDS and chitosan were mixed at pH 3 and then the solution was adjusted to pH 7, the SDS remained bound to the chitosan. The presence of NaCl (0-200 mM) in the solutions decreased the critical micelle concentration (cmc) of SDS (in both the absence and the presence of chitosan) but had little influence on the amount of SDS bound to chitosan at saturation. The cmc of SDS and the amount of SDS bound to the chitosan at saturation were largely independent of the holding temperature (10-40 degrees C). Nevertheless, the enthalpy changes associated with micelle dissociation were highly temperature-dependent, indicating the importance of hydrophobic interactions, whereas the enthalpy changes associated with SDS-chitosan binding were almost temperature-independent, indicating the dominant contribution of electrostatic interactions. This study provides information that may lead to the rational design of chitosan-based ingredients or products with specific nutritional and functional characteristics, for example, cholesterol lowering.

Iron deficiency anemia (IDA) presents a global health challenge, impacting crucial development stages in humans and other mammals. Pigs, having physiological and metabolic similarities with humans, are a valuable model for studying and preventing anemia. Commonly, a commercial iron dextran formulation (CIDF) with iron dextran particles (IDPs) is intramuscularly administered for IDA prevention in pigs, yet its rapid metabolism limits preventive efficacy. This study aimed to develop and evaluate chitosan thermosensitive hydrogels (CTHs) as a novel parenteral iron supplementation strategy, promoting IDPs' prolonged release and mitigating their rapid metabolism. These CTHs, loaded with IDPs (0.1, 0.2, and 0.4 g of theoretical iron/g of chitosan), were characterized for IM iron supplementation. Exhibiting thermosensitivity, these formulations facilitated IM injection at ~4 °C, and its significant increasing viscosity at 25–37 °C physically entrapped the IDPs within the chitosan's hydrophobic gel without chemical bonding. In vitro studies showed CIDF released all the iron in 6 h, while CTH0.4 had a 40% release in 72 h, mainly through Fickian diffusion. The controlled release of CTHs was attributed to the physical entrapment of IDPs within the CTHs' gel, which acts as a diffusion barrier. This study investigates the mechanical properties of α-chitin and chitosan biocomposites using molecular dynamics (MD) and stress–strain analyses under uniaxial tensile loading in an aqueous environment. Our models, validated against experimental data, show that α-chitin has a higher directional elastic modulus of 51.76 GPa in the x and 39.76 GPa in the y directions compared to its chitosan biocomposite, with 31.66 GPa and 26.00 GPa in the same directions, demonstrating distinct mechanical behaviors between α-chitin and the biocomposite. The greater mechanical stiffness of α-chitin can be attributed to its highly crystalline molecular structure, offering potential advantages for applications requiring load-bearing capabilities. Heavy metal pollution in water bodies is a global concern. The prominent source of metal contamination in aqueous streams and groundwater is wastewater containing heavy metal ions. Elevated concentrations of heavy metals in water bodies can have a negative impact on water quality and public health. The most effective way to remove metal contaminants from drinking water is thought to be adsorption. A deacetylated derivative of chitin, chitosan, has a wide range of commercial uses since it is biocompatible, nontoxic, and biodegradable. Due to its exceptional adsorption behavior toward numerous hazardous heavy metals from aqueous solutions, chitosan and its modifications have drawn a lot of interest in recent years. Due to its remarkable adsorption behavior toward a range of dangerous heavy metals, chitosan is a possible agent for eliminating metals from aqueous solutions. The review has focused on the ideas of biosorption, its kinds, architectures, and characteristics, as well as using modified (physically and chemically modified) chitosan, blends, and composites to remove heavy metals from water. The main objective of the review is to describe the most important aspects of chitosan-based adsorbents that might be beneficial for enhancing the adsorption capabilities of modified chitosan and promoting the usage of this material in the removal of heavy metal pollutants.
