Turmeric (Curcuma longa L.) is of great importance due to its medicinal properties and industrial uses. However, as in most plants, physiological and biochemical changes occur in turmeric plants subjected to saline conditions. This study aimed to evaluate the growth and antioxidant response of turmeric plants, and the yield and composition of turmeric essential oil under saline stress. The study was greenhouse-based, and 180 days old turmeric plants were treated with 50, 100, and 150 mM NaCl solutions for 130 days. Plants grown under high NaCl concentrations showed significantly reduced shoot length and shoot and rhizome biomass accumulation. Large amounts of proline accumulated in the leaves at 150 mM NaCl. Lipid peroxidation and malondialdehyde content increased with salinity in both leaves and rhizomes. Antioxidant enzymes showed greater activity in the leaves of plants exposed to high NaCl concentrations. Salinity altered the yield and chemical composition of the essential oil in the rhizomes. A total of 114 compounds were identified in the essential oil, with the main compounds from the sesquiterpenes class. Sesquiterpenes can constitute a secondary antioxidant system which can assist the primary antioxidant defense system to maintain the levels of hydrogen peroxide in cells at concentrations sub-lethal to turmeric plants. In addition, the presence of rhizomes has a mitigating effect on salt stress in this species due to the antioxidant activity of the essential oil present in the rhizomes.

Introduction: Liver or hepatic disease refers to different conditions that affect the liver. Chronic alcohol consumption is one of the most frequent causes of liver disease and accounts for about 55% of liver cirrhosis deaths recorded in Cameroon in 2020. Standard accessible treatments focus on end-stage liver disease with safety and efficacy obstacles. We have a research gap in Cameroon to understand the alternative use of natural products as treatment with a long traditional history of safe use. Curcuma longa has long been a source of traditional and modern medicine. It is commonly used in Cameroon as a spice and herbal product with some level of activity against various forms of liver disease. Objective: To phytochemically screen for bioactive metabolites and evaluate the hepatoprotective activity of the aqueous extract of Curcuma longaon alcohol-induced toxicity in Wistar rats. Methods: Phytochemical screening was carried out on the aqueous extract obtainedfrom maceration of plant rhizomes. Three doses (125, 250 and 500mg/Kg) of the plant extract and the reference (Silymarin 50mg/Kg) were administered daily (p.o) to rats 30 min before administration of 40% alcohol (2mL/100g p.o) for 21 days. Biochemical parameters such as ALAT, ASAT, GGT, Bilirubin and Lipid profile were quantified and histological studies of the liverwas carried out using standard procedures. Results: Phytochemical screening of the aqueous extract of C. longa revealed polyphenols such as flavonoids, tannins, quinones, saponins and phlobatanins. The plant showed hepatoprotective activity by decreasing liver toxicity markers such as ASAT, ALAT, GGT and Bilirubin. Histology revealed dose-dependent protection with 500 mg/Kg showing the most cellular integrity, no central vein occlusion and minimal fibrosis. Conclusion: This study indicated the presence of polyphenols like flavonoids and tannins in the aqueous extract of C. longa. The presence of these secondary metabolites in the studied extract justifies its antioxidant and anti-inflammatory properties confirmed by its hepatoprotective effects on alcohol-induced toxicity. This was clearly shown by biochemical and histological parameters. More sensitive and specific methods are required to test for these secondary metabolites in serum.

Polyploidization can be a way to produce new varieties in vegetatively propagated species where options on increasing genetic variability are limited compared with sexual reproduction. While there are hundreds of publications with in vitro methods in somatic doubling, it is cardinal to custom-test for a target species of interest on choosing specific reagents and optimizing conditions. This research was performed to provide a reference process for Zingiberaceae species of which the majority is reproduced with vegetative propagation. Ginger lilies, Hedychium gardnerianum Shepard ex Ker Gawl. and Hedychium coronarium J. Koenig, were employed to optimize chromosome doubling for tetraploid production as they are typically used by vegetative propagation. However, they have the popularity as ornamentals globally due to their horticultural aspects. Tetraploid induction was optimized by in vitro somatic chromosome doubling on H. coronarium and H. gardnerianum through callogenesis. The explant segments of young leaf blades or leaf sheaths were treated with different concentrations of either with colchicine or with oryzalin. The regenerated shoots from callus cultures were transferred to basal MS medium for 2mo to confirm somatic stability. The ploidy was assessed by flow cytometry, measuring the size and density of stomata, counting chromosomes, and chloroplasts in guard cells. The highest percentage of tetraploid regenerated plants was observed with 1250 µM colchicine treatment for Hedychium gardnerianum when compared to the other treatments tested, while no tetraploid plants were obtained from the oryzalin treatment. In H. coronarium, four mixoploid regenerated plants treated with 15 µM oryzalin were confirmed and no shoots were obtained from the colchicine treatment. The results indicated that the in vitro polyploid induction is optimized with the two Hedychium species, which could reference other Zingiberaceae species.

Introduction: Kaempferia galanga is a medicinal plant belonging to the family Zingiberaceae: ginger family. It is treated as a folk traditional herb. Anti Inflammatory property refers to the ability of a substance to reduce inflammation or any of its 5 cardinal signs. Aim: To assess and compare in vitro the anti-inflammatory properties of various extracts (ethanol, chloroform and aqueous) of Kaempferia galanga L Rhizome. Materials and Methods: Protein Denaturation Inhibition was carried out in vitro and statistical analysis was done using ONE WAY -ANOVA and Duncan Multiple Range Tests. The test was done in triplicates. Results: Chloroform extract of Kaempferia galanga rhizome has the best anti-inflammatory potential followed by Ethanol and Aqueous extracts of the rhizome. Conclusion: With further in vivo and clinical research, the chloroform, ethanolic and aqueous extracts of Kaempferia galanga can be recommended as a novel, innovative and potent anti-inflammatory drug in the market as it’s natural and doesn’t have side effects.

Amomum compactum Soland Ex Maton is one of the Zingiberaceae family plants which is the endemic plants from West Java, Indonesia. This study was aimed to determine the chemical structure of flavonoid compounds from n-hexane extract of A.compactum Sol. Ex Maton roots. Dried powder of the roots was extracted consecutively with n -hexane, ethyl acetate, and methanol solvents. Three flavonoids, 5-hydroxy-3,7,4`-trimethoxy kaempferol ( 1 ), 5-hydroxy-3,7,3',4'-tetra methoxy kaempferol ( 2) and 4'-hydroxy-3,5,7-trimethoxy kaempferol ( 3 ), have been isolated from the roots of A. compactum Sol. Ex Maton. The chemical structures of compounds 1 - 3 were identified by spectroscopy data including infrared 1D-NMR, 2D-NMR and HRTOF-MS as well as by comparison with previously reported spectral data. Compounds 1-3 were isolated from this plant for the first time and showed free radical DPPH scavenging activity.

Curcuma longa (Turmeric) is a tropical herbaceous perennial plant of the family Zingiberaceae and contains curcuminoids, sesquiterpenoids and monoterpenoids as its major components. Given the broad range of activities that Curcuma longa possesses and also its use as a traditional epilepsy remedy, this review attempts to systematically review the experimentally proven activities of Curcuma longa and its bioactive components, which are related to the management of epileptic seizures. Using the PRISMA model, five databases (Google Scholar, PubMed, ScienceDirect, SCOPUS and SpringerLink) were searched using the keywords [“Curcuma longa” AND “Epilepsy”] and [“Curcuma longa” AND “Seizures”], leaving 34 articles that met the inclusion criteria. The present systematic review elaborated on the experimentally proven potential of Curcuma longa components, such as an aqueous extract of Curcuma longa itself, Curcuma longa oil and active constituents like curcuminoids and bisabolene sesquiterpenoids found in Curcuma longa with anti-seizure potential. Using human equivalent dose calculations, human treatment parameters were suggested for each component by analysing the various studies in this review. This review also determined that the principal components possibly exert their anti-seizure effect via the reduction of corticosterone, modulation of neurotransmitters signalling, modulation of sodium ion channels, reduction of oxidative DNA damage, reduction of lipid peroxidation, upgregulation of brain-derived neurotrophic factor (BDNF) and γ-aminobutyric acid (GABA) mediated inhibition. It is anticipated that this review will help pave the way for future research into the development of Curcuma longa and its neuroactive constituents as potential drug candidates for the management of epilepsy.

Curcuma aeruginosa Roxb. is one of the plants from the Zingiberaceae family which the rhizome has been used for medicinal purposes. However, the biological properties of the leaves have not been fully explored. Therefore, this study was conducted to evaluate the effects of different drying methods and solvents on total phenolic content, antioxidant and anti-hyperglycemic activities of C. aeruginosa leaf extract. Samples were dried by oven drying (OD) and freeze drying (FD), and then extracted using different ratios of ethanol:water (100:0, 50:50 and 0:100). The amount of phenolic content (TPC) was determined using a spectrophotometer. Antioxidant activity was tested using the Free Radical Scavenging (DPPH) test and the Ferric Reducing Antioxidant Power assay (FRAP), while the anti-hyperglycemic test was evaluated by determining the percentage of α-glucosidase inhibition. The results showed that FD at 100:0 and 50:50 concentrations had the highest phenolic content (30.88 and 33.06 mg GAE/g extract, respectively) and highest antioxidant activity value (38.24 and 42.46 mg TEAC/g extract, respectively). For DPPH, FD at 50:50 showed the highest inhibition of 71.48% compared to other extracts. Whereas FD at 100:0, 50:50 and 0:100 showed the highest α-glucosidase inhibition of 25.65, 30.78 and 27.65%, respectively. However, compared to Quercetin, the extract showed mild anti-hyperglycemic activity. The results indicated that FD is the best method of drying while 50:50 showed as the best solvents. Positive correlation between TPC with antioxidant and anti-hyperglycemic activities showed that C. aeruginosa leaf has potential as a source of natural antioxidant with the presence of phenolic compounds.

The rhizome of the temu mangga is a member of the Zingiberaceae family that has been used since ancient times for treatment of diabetes. The aim of this study was to examine the activity of reducing blood glucose levels in diabetic mice induced by alloxan after administration of the ethyl acetate fraction of temu mangga rhizome at a dose of 100; 200 and 300 mg/KgBW were compared with negative control (Na CMC) and positive control (Glibenclamide 0.65 mg/Kg BW). The research data were statistically analyzed using two-way ANOVA for the parameters of the percentage decrease in blood glucose and the percentage change in body weight followed by the Tukey multiple range test. Background: Curcumin is a curcuminoid which is an active constituent of turmeric and is obtained from the rhizomes of Curcuma longa, family Zingiberaceae. Curcumin modulates the activity of various transcription factors and regulates the expression of inflammatory enzymes, cell survival proteins, adhesion molecules and cytokines by binding to a variety of proteins and inhibiting the activity of various kinases. Curcumin falls in the BCS class IV drug, with poor solubility and poor permeability which makes it very challenging in utilizing the maximum therapeutic potential of this moiety. Objective: The major aim of the study was to enhance transdermal penetration of curcumin via ethosomal gel and to overcome the barriers of poor permeability of transdermal drug delivery. Methods: Curcumin loaded ethosomes were prepared with varying quantities of ethanol and soya lecithin by cold method and were optimised based on entrapment efficiency, vesicular size and Ex-vivo studies. Optimised ethosomal formulation was further incorporated into a gel and was evaluated. Ex-vivo studies were performed with the ethosomal gel of curcumin and was compared with simple drug solution. Result: Prepared ethosomal system showed a vesicle size ranging from 211 to 320 nm with spherical, smooth surface and entrapment efficiency of 87 to 91%. Optimised ethosomal system (ET3) was incorporated into gel and was further evaluated. Conclusion: The findings of the research work suggested that the ethosomal gel holds an excellent potential for transdermal delivery of curcumin.

Turmeric, the rhizome of Curcuma longa plant belonging to the ginger family Zingiberaceae, has a history in Ayurvedic and traditional Chinese medicine for treatment of chronic diseases, including metabolic and cardiovascular diseases (CVD). This parallels a prevalence of age- and lifestyle-related diseases, especially CVD and type 2 diabetes (T2D), and associated mortality which has occurred in recent decades. While the chemical composition of turmeric is complex, curcuminoids and essential oils are known as two major groups that display bioactive properties. Curcumin, the most predominant curcuminoid, can modulate several cell signaling pathways involved in the etiology and pathogenesis of CVD, T2D, and related morbidities. Lesser bioactivities have been reported from other curcuminoids and essential oils. This review examines the chemical compositions of turmeric, and related bioactive constituents. A focus was placed on the cellular and molecular mechanisms that underlie the protective effects of turmeric and turmeric-derived compounds against diabetes and CVD, compiled from the findings obtained with cell-based and animal models. Evidence from clinical trials is also presented to identify potential preventative and therapeutic efficacies. Clinical studies with longer intervention durations and specific endpoints for assessing health outcomes are warranted in order to fully evaluate the long-term protective efficacy of turmeric.

Globba leucantha var. bicolor Holttum is a part of Zingiberaceae plant that is not yet widely known but can be used as a medicinal material as well as an ornamental plant. So far Globba plant propagation has not been widely done and has relied solely on from its habitat. Research aims at acquiring Naphthalene Acetic Acid (NAA) concentrations and optimal kinetin for the multiplication of ginger plant shoots (Globba leucantha var. bicolor Holttum) on in vitro culture. The research method uses experimental methods with non-parametric statistics and 24 repeated treatments 3 times as well as being analyzed descriptively using the Kruskal Wallis Test. The treatment used NAA with concentrations of 0 mg/l, 0.1 mg/l, 0.5 mg/l, 1 mg/l, 1.5 mg/l, and 2 mg/l combined with Kinetin with concentrations of 0 mg/l, 0.1 mg/l, 1 mg/l and 10 mg/l. The results showed that the growth of Globba plant's best shoots at N3K3 treatment (NAA 1 mg/l + Kinetin 10 mg/l) with a shoot emergent time of 21.58 HSI, a percentage of shoot forming 66.67%, a shot count of 0.89 buds per explant, and a shoot length of 0.18 cm. Additionally at the same time also formed Globba plant roots with the best treatment of N5K2 (NAA 1 mg/l + Kinetin 10 mg/l) with a time of appearing rooted 25.67 HSI and a root count of 7.78 roots per explant.

Rhynchanthus beesianus (Zingiberaceae) has been an important food spice and vegetable in southern China. Fifteen phenolic compounds (1-15) including three new diarylheptanoids, rhynchanines A-C (1-3) and one new phenylpropanoid, 4-O-methylstroside B (9), were isolated from R. beesianus rhizomes. The structures of new compounds were elucidated by comprehensive analyses through NMR, HRMS technique, acid hydrolysis, and Mosher's reaction. Among them, compound 5 is the first isolated natural product and its NMR data are reported. Most of the isolated compounds, especially 3-6 and 8, showed significant antioxidant activities on DPPH, ABTS+ radical scavenging, and FRAP assays. Furthermore, the antioxidant phenolic compounds were evaluated for their cytoprotective capacity against H2O2-induced oxidative stress in HepG-2 cells. Compounds 3 and 5 could significantly inhibit reactive oxygen species production, and compounds 3, 5, and 6 could remarkably prevent the cell apoptosis. Then, the R. beesianus rhizome, which contained phenolic compounds, might serve as a functional food for potential application on preventing oxidative stress-connected diseases.

Ginger (Zingiber officinale) is a common kitchen spice that belongs to the family Zingiberaceae. It is rich in phytochemistry that is promoting health benefits. It is used as a home remedy to support the common cold, headaches, and pharmacological properties such as anti-inflammatory, antioxidant, antiemetic, antiulcer, and anti-cancer, antiplatelet, anti-diabetic and lipid-lowering activities. Gingerols are key ingredients found in ginger that convert into zingerone, shogaol, and parasols, giving flavor and odor. Zingerone and shogaol are present in limited quantities in fresh ginger and more in dried or extracted goods. Especially 6-gingerol and 6-shogaol are pharmacological properties that are effective in antipyretic, analgesic, and hypotensive. The present review is about different therapeutic properties of ginger, including antioxidant properties, anti-diabetic properties, anti-cancer properties etc.

New copaene-type and nerolidol-type sesquiterpenoids, 7-hydroxymustakone (1) and 15-hydroxynerolidol (2), and a 15-norlabdane diterpenoid, kaempcandiol (3), together with four known compounds (4-7) were isolated from the chloroform extract of Kaempferia candida roots and rhizomes. The structures of the new compounds 1-3 were elucidated based on 1D and 2D NMR and HRESIMS spectroscopic analyses. The extract of the K. candida roots and rhizomes and all isolated compounds 1-7 possessed HIV-1 viral protein R (Vpr) inhibitory activities on the TREx-HeLa-Vpr cell line at a 5 μM concentration, without detectable cytotoxicity.

Daikenchuto (DKT) is one of the most widely used “Kampo” in Japan as a representative of herbal medicine. Because DKT is made from a natural product like food, it requires the management of pesticides; therefore, an analysis of residual pesticides in Kampo is required. The World Health Organization (WHO) indicates that pesticide residue analysis by the U.S. Pharmacopeia (USP) is required. USP defines 107 compounds containing organochlorine pesticides and organophosphorus pesticides and their metabolites, which have a high residual risk. Accordingly, to guarantee the safety of herbal medicines according to global standards is a very important issue. In this study, we developed an analytical method for 91 compounds, which are listed in USP, using DKT as the subject. The method could extract pesticides from DKT with acetone, elute pesticides with acetonitrile using a SepPak C18 column (5 g) and with ethyl acetate using a DSC-NH2 column (2 g), and perform simultaneous analyses by gas chromatography–tandem mass spectrometry (GC–MS/MS). This method, which could quantify 88 compounds, was validated according to USP. A pesticide residue analysis method that meets USP requirements enables the analysis of pesticide residues with a high residue risk and contributes to improving the safety of “Kampo” and other herbal medicines.

The study aimed to investigate the chemical composition and the anti-inflammatory activity of the hydroethanolic rhizomes, stems, and leaf extracts of Renealmia petasites using in vitro and in vivo assays. The chemical composition of the extracts was characterized in a linear iron trap mass spectrometer. Total phenolic, flavonoid, and tannin content were determined by spectrophotometry analyses. In vitro anti-inflammatory activity was investigated in lipopolysaccharide-stimulated macrophages evaluating the influence on the production of superoxide anion (O2−), nitric oxide (NO), and the pro-inflammatory cytokines tumor necrosis factor (TNF-α) and interleukin-6 (IL-6). In vivo effects were determined using the air pouch model in which were inoculated carrageenan and thereafter treated with 50 mg/kg of the hydroethanolic extracts of R. petasites. After 4 and 24 h, the cellular influx, protein exudation, cytokines, and nitric oxide were evaluated. Eight compounds were tentatively identified in the R. petasites extracts, suggesting five diarylheptanoids, one flavonoid, and two fatty alcohols. The in vitro results showed that the extracts were capable of blocking free radicals and/or inhibiting their intracellular actions by inhibiting the production of important mediators of the inflammatory process, such as NO, O2−, TNF-α, and IL-6. In vivo, R. petasites significantly decrease the influx of leukocytes, mainly neutrophils, protein exudation, NO, TNF-α, and IL-6 concentration in the air pouch model. The results evidenced that R. petasites can be considered a promising alternative therapy for the treatment and management of osteoarthritis and other inflammatory diseases.

Objective: To compare the antioxidant and anti-genotoxic properties of Alpinia (A.) galanga, Curcuma (C.) amada, and C. caesia. Methods: Cytotoxicity of ethanolic extracts of A. galanga, C. amada, and C. caesia at selected doses was evaluated by trypan blue, MTT, and flow cytometry-based assays. Genotoxicity and anti-genotoxicity (against methyl methanesulfonate, 35 μM and H2O2, 250 μM) of these plants were studied by comet assay in human lymphocytes in vitro. Furthermore, DPPH, ABTS, FRAP, lipid peroxidation, and hydroxyl radical scavenging assays were performed to study the antioxidant potentials of the plants. Finally, anti-genotoxic potential of C. amada was validated in Swiss albino mice using comet assay. Phytochemical composition of C. amada was determined by GC/MS and HPLC. Results: The selected doses (2.5, 5, and 10 μg/mL) of A. galanga, C. amada, and C. caesia were non-toxic by cytotoxicity tests. All three ethanolic extracts of plant rhizomes demonstrated antioxidant and anti-genotoxic properties against methyl methanesulfonate-and H2O2-induced oxidative stress in human peripheral blood lymphocytes in vitro. Multivariate analysis revealed that various antioxidant properties of these extracts in DPPH, ABTS, and FRAP assays were strongly correlated with their total phenolic constituents. C. amada extract conferred protection against cyclophosphamide-induced DNA damage in the bone marrow cells of mice and DNA damage was significantly inhibited by 2.5 mg/kg C. amada extract. Conclusions: C. amada is rich in potentially bioactive molecules and exhibits potent antioxidant activities. Its anti-genotoxicity against cyclophosphamide-induced oxidative stress is also confirmed in this study.

Alpinia Roxb. is the largest genus of the Zingiberaceae family. A large number of Alpinia species has been used as food and traditional medicines. Alpinia essential oils have been studied for their chemical profiles, in which 1,8-cineole, β-pinene, α-pinene, β-myrcene, camphor, γ-terpinene, p-cymene, geraniol, α-fenchyl acetate, ocimene, methyl cinnamate, and β-caryophyllene have been found to be the major compounds. Essential oils isolated from Alpinia plants have been reported to have antimicrobial, cytotoxic, antioxidant, anti-inflammatory, anti-asthmatic, tyrosinase inhibitory, insecticidal, and larvicidal activities and slimming aromatherapy. In this review, the comprehensive information regarding the volatile components of various Alpinia plants, the bioactivities of Alpinia essential oils and their major compounds are provided.

From the evolution of the mankind, Turmeric has been used in conventional medication. India is in lead for producing, marketing and exporting the Turmeric and its value added products. Curcuma longa (Turmeric) is an Indian rhizomatous medicinal herb from the Zingiberaceae family that is common and widely available across the globe. The components of Turmeric are curcumin, demethoxycurcumin and bisdemethoxycurcumin and these are collectively known as curcuminoids. Curcumin, the active ingredient of Turmeric is generally investigated by the scientific community for its wide range of antioxidant activity, anti-Inflammatory properties and anti-cancer activity, anti-metabolic syndrome activities, neuroprotective activity, antimicrobial effects, anti-arthritis effects, anti-viral effects, anti-asthma and anti-diabetic effects, anti-obesity, cardio and liver toxicity protection activity, anti-depression and anxiety activities. Turmeric has been widely used as a typical household treatment for cough, sore throat, respiratory ailments and could be an effective immunity booster against SARS-CoV-2 therapy during the ongoing pandemic situation. Safety evaluation studies indicate that both turmeric and curcumin are well tolerated at a very high dose without any toxic effects. Thus, turmeric and its constituents have the potential for the development of modern medicine for the treatment of various diseases. So in this review, we describe the various metabolic roles of curcumin and activities for the benefit of human health.

Twenty-seven constituents were identified by using GC/MS, representing 99.57% of the rhizome oil of Distichochlamys benenica. The major constituents of the essential oil are 1,8-cineole (54.39%), β-pinene (7.50%), (E)-citral (7.26%), and (Z)-citral (6.79%). The rhizome essential oil has anti-acetylcholinesterase activity with an IC50 value of 136.63 ± 2.70 mg/mL.

The development of intelligent packaging based on natural and biodegradable resources is getting more attention by researchers in recent years. The aim of this study was to develop and characterize a pH-sensitive films based on sago starch and incorporated with anthocyanin from torch ginger. The pH-sensitive films were fabricated by casting method with incorporation of different torch ginger extract (TGE) concentration. The surface morphology, physicochemical, barrier, and mechanical properties as well as the pH-sensitivity of films were investigated. The film with the highest concentration of TGE showed the lowest tensile strength (4.26 N/m2), toughness (2.54 MJ/m3), Young's modulus (73.96 MPa) and water vapour permeability (2.6 × 10−4 g·m/day·kPa·m2). However, its elongation at break (85.14%), moisture content (0.27%) and water solubility (37.92%) were the highest compared to other films. pH sensitivity analysis showed that the films containing TGE extract, changes in colour by changing the pH. The colour of films changed from pink to slightly green as the pH increased from pH 4 to 9. Thus, the developed pH-sensitive film with torch ginger extract has potential as intelligent packaging for detection of food freshness or spoilage to ensure their quality and safe consumption.

A new sesquiterpenoid derivative, 13-hydroxy-1-oxocurzerenone (1), has been isolated from the rhizomes of Curcuma zedoaria, together with seven known compounds, 1-oxocurzerenone (2), curcolone (3), curcumenone (4), procurcumenol (5), curcumenol (6), zerumin A (7), and labda-8(17), 12-diene-15,16-dial (8). The structure of the new compound 1 was determined through spectroscopic and MS analyses. Among the isolates, 13-hydroxy-1-oxocurzerenone (1), procurcumenol (5), and zerumin A (7) exhibited cytotoxicities, with IC50 values of 3.48, 12.36, and 14.08 μM, respectively, against the DLD-1 cell line. Curcumenone (4) and curcumenol (6) showed cytotoxic effects, with IC50 values of 8.97 and 10.26 μM, respectively, against the MCF-7 cell line. In addition, 13-hydroxy-1-oxocurzerenone (1), 1-oxocurzerenone (2), zerumin A (7), and labda-8(17),12-diene-15,16-dial (8) showed potent inhibition, with IC50 values of 4.63, 6.48, 16.57, and 18.45 μM, respectively, against LPS-induced NO generation.

Fever is a condition where the body temperature rises above normal or more than 37 o C and also tend to be an initial clinical manifestation of the use of antipyretic drugs that cause toxicity such as on the liver due to prolonged usage . Particularly, the b angle ( Zingiber purpureum Roxb.) is one of the Zingiberaceae plants that contain essential oils used for the treatment of fever. Therefore, this research aimed to identify active compounds which have antipyretics potential with the in silico approach. The simulation of molecular docking showed 1,4-naphthalenedione-2-ethyl-3-hydroxy was able to attach to the binding site of cyclooxygenase-2 (COX-2) and interact with main residues that constituted the active cavity of COX-2. While the simulation of molecular dynamics suggested that the bound compound was stable at 4 ns, that is the time taken. The binding free energies expected by the MM-PBSA method indicated the 1,4-naphthalenedione-2-ethyl-3-hydroxy had a higher affinity than a native ligand (2-[(2,6-dichloro-3-methyl-phenyl)-amino] benzoic acid, JMS) and paracetamol. This suggested its capacity for advancing as a new COX-2 inhibitor.

The modern medicine gets up a massive hike in the treatment of various ailments but scarcely any drugs having to stimulate liver activity, offering tutelage to the liver from any harm or promote the palingenesis of hepatic cells. However, a variety of herbal drugs applied in the conventional system of medicine for liver efficiency. Therefore, rhizome of Curcuma amada (Family: Zingiberaceae) are selected to allocate the hepatoprotective activity in scientifically approbate experimental models. This study is an effort to explore the 50% ethanolic extract of rhizome of Curcuma amada (CAE) in a different experimental model for hepatoprotective activities and in vitro antioxidant. 50% ethanolic extract of Curcuma amada (CAE) (100 and 200mg/kg) hepatoprotective efficiency was examined against paracetamol (1000 mg/kg) induced hepatotoxicity, elevated hepatic enzymes alanine aminotransferase (ALT), aspartate aminotransferase (AST), and alkaline phosphatase (ALP), total bilirubin direct bilirubin content in the serum. Moreover, CAE induced antioxidant protection against hepatotoxic disservice of paracetamol was estimated by evaluating several antioxidative biomarkers such as reduced glutathione (GSH), superoxide dismutase (SOD), and catalase (CAT) in the blood serum by using spectrophotometric analysis. It could be concluded that the rhizome of C. amada possesses an intense antioxidant property. Its active principle wipeout free radicals and strive a tutelary impact against oxidative harm induced to cellular macromolecules. 50% ethanolic extract of C. amada (CAE) is tenable to amplify and maintain the working of hepatic enzymes implicated in detention of ROS.

The importance of biogenic silver/silver chloride nanoparticles has become increasing day by day. In the present study, silver/silver chloride nanoparticles (Ag/AgCl‐NPs) were synthesized from Kaempferia rotunda tuberous rhizome extract to evaluate the antiproliferative activity against human glioblastoma stem cells (GSCs) in vitro and Ehrlich ascites carcinoma (EAC) cells in vivo in mice. Synthesis of nanoparticles was confirmed by colour change and UV‐visible spectrum and characterized by TEM, XRD, TGA, AFM and FTIR. K rotunda and recently synthesized Zizyphus mauritiana fruit extract‐mediated Ag/AgCl‐NPs inhibited 77.2% and 71% of GSCs growth at 32 µg/mL concentration with the IC50 values of 6.8 and 10.4 µg/mL, respectively. Cell morphological studies and caspase‐3 immunofluorescence assay revealed that both biogenic nanoparticles induced apoptosis in GSCs. Expression levels of several genes were checked by real‐time PCR after treatment with K rotunda tuberous rhizome‐mediated Ag/AgCl‐NPs. PARP, EGFR, NOTCH2 and STAT3 gene expression were decreased with the increase of NFκB, TLR9, IL1, TNFα, IKK and p21 gene that would be the cause of induction of apoptosis in GSCs. The cell cycle arrest at G2/M phase was confirmed by flow cytometric assay. Both nanoparticles were injected intraperitoneally to rapidly growing EAC cells for 5 consecutive days. Approximately, 32.3% and 55% EAC cells growth were inhibited by K rotunda tuberous rhizome‐mediated Ag/AgCl‐NPs at 6 and 12 mg/kg/day doses, respectively while only 20% cell growth inhibition was monitored at 12 mg/kg/day dose of Z mauritiana ‐mediated Ag/AgCl‐NPs. From the above results, it can be concluded that presently synthesized nanoparticles would be a potent anticancer agent.

Amomum nilgiricum is one of the plant species reported from Western Ghats of India, belonging to the family Zingiberaceae, with ethno-botanical values, and is well-known for their ethno medicinal applications. In the present investigation, ethyl acetate and methanol extracts of A. nilgiricum were analyzed by Fourier transform infrared spectrometer (FTIR) and gas chromatography-mass spectrometry (GC–MS) to identify the important functional groups and phytochemical constituents. The FTIR spectra revealed the occurrence of functional characteristic peaks of aromatic amines, carboxylic acids, ketones, phenols and alkyl halides group from leaf and rhizome extracts. The GC–MS analysis of ethyl acetate and methanol extracts from leaves, and methanol extract from rhizomes of A. nilgiricum detected the presence of 25 phytochemical compounds. Further, the leaf and rhizome extracts of A. nilgiricum showed remarkable antibacterial and antifungal activities at 100 mg/mL. The results of DPPH and ferric reducing antioxidant power assay recorded maximum antioxidant activity in A. nilgiricum methanolic leaf extract. While, ethyl acetate leaf extract exhibited maximum α-amylase inhibition activity, followed by methanolic leaf extract exhibiting aldose reductase inhibition. Subsequently, these 25 identified compounds were analyzed for their bioactivity through in silico molecular docking studies. Results revealed that among the phytochemical compounds identified, serverogenin acetate might have maximum antibacterial, antifungal, antiviral, antioxidant and antidiabetic properties followed by 2,4-dimethyl-1,3-dioxane and (1,3-13C2)propanedioic acid. To our best knowledge, this is the first description on the phytochemical constituents of the leaves and rhizomes of A. nilgiricum, which show pharmacological significance, as there has been no literature available yet on GC–MS and phytochemical studies of this plant species. The in silico molecular docking of serverogenin acetate was also performed to confirm its broad spectrum activities based on the binding interactions with the antibacterial, antifungal, antiviral, antioxidant and antidiabetic target proteins. The results of the present study will create a way for the invention of herbal medicines for several ailments by using A. nilgiricum plants, which may lead to the development of novel drugs.

In our previous published article ( https://doi.org/10.1007/s10989-019-09883-7 ), caP4—a cationic peptide of 2.97 kDa from Curcuma pseudomontana L. (Zingiberaceae) with sequences ASSCKPS and ASSKWVAPSEW showed significant antibacterial activity against Escherichia coli and Staphylococcus aureus. In the present study, circular dichroism (CD) data of caP4 showed 0% α-helix, 21.6% β-sheet, 31.2% β-turns, and 47.2% random coils. Further, the study was focused on understanding the mode of action of caP4 against E. coli. At 8 µg/ml (MIC), caP4 permeabilized the cell membrane with maximum recorded at 50 min. Scanning electron microscopy analysis of E. coli cells with caP4 at 16 µg/ml showed disorientation of the membrane surface. Cetyl pyridinium chloride (CPC, 10 µg/ml-IC50), Mitomycin C (10 µg/ml-IC50), Colchicine (25 µg/ml-IC50), Cytochalasin B (30 µg/ml-IC50) and Cibacron blue (25 µg/ml-IC50) were used as cell growth inhibitors with or without caP4 (8 µg/ml) to check the action of peptide intracellularly. E. coli cells with caP4 showed 100% cell death at 100 min. E. coli cells pre-treated with caP4 for 30 min showed 100% and 96% cell death at 60 min and 56 min with CPC and Mitomycin C respectively, suggesting that caP4 is acting by entering into the cell membrane and thus inhibit protein synthesis similar to CPC and Mitomycin C, whereas Colchicine, Cytochalasin B, Cibacron blue did not show significant cell death with pre and post-treatment of caP4. Thus, the study showed the synergistic effect of caP4 with cell growth inhibitors CPC and Mitomycin C in enhancing the antibacterial activity. Thus the data provide a new insight to understand the mechanism of antibiotic activity of caP4 and could lay the foundation for developing plant based antibacterial drugs in future to combat drug resistant microbes.

Crude plant extracts have been used as insecticides from ancient times. Keeping in view the environmental issues related to the excessive use of insecticides, plant derived products have been intensively investigated for their insecticidal potential. Alpinia galanga, a medicinal plant belonging to family Zingiberaceae exhibits a wide range of biologicial activities. Thus, the present study aims to evaluate the anti-insect potential of A. galanga against Spodoptera litura (Fab.), a polyphagous pest causing economic losses to many agricultural crops in India. Addition of A. galanga extracts to the larval diet adversely affected the survival and development of S. litura. Ethyl acetate extract was found to be the most effective causing 67.50% larval mortality at 2500 ppm and extending the development period by 12.73 days as compared to control. Purified compounds isolated from A. galanga viz. 1′-acetoxychavicol acetate and galangin exhibited more larvicidal activity in comparison to crude extracts. Both of the compounds also retarded the development of S. litura. The present study implies that A. galanga extracts have the potential to manage insect pests, although there is need to evaluate the field efficacy.

Background and objective Etlingera elatior (Jack) R.M. Smith rhizome, which has been traditionally used to reduce stomach discomfort, was reported to possess anti-inflammatory activity, however, there is a lack of such a study on the flower. Materials and methods To investigate the anti-inflammatory activity of the E. elatior flower extract on gastric ulceration-induced Wistar rats. The Wistar rats were divided into 6 groups. Group 1 was the normal control, group 2 was the negative control (Arabic gum suspension 2%), group 3 was the positive control (quercetin), group 4-6 were treated with E. elatior flower extract dose of 500, 1000 and 2000 mg kg-1 of b.wt., respectively. The rats were conditioned to gastric ulceration. The stomach weight, microscopic and macroscopic evaluation of gastric mucosal damage was examined. Subsequently, the nuclear factor-kappaB-p65 (NF-kappaB-p65) expression in the fundus was Western-blotted by employing β-actin and GAPDH as the loading controls. Results Etlingera elatior flower extract dose of 1000 mg kg-1 b.wt., reduces the ulceration index and the infiltration of inflammatory cells. Western blot analysis showed inhibition of NF-kappaB-p65 expression by E. elatior flower extract dose of 1000 mg kg-1 of b.wt. Conclusion Etlingera elatior flower might possess anti-inflammatory activity by downregulating the expression of NF-kappaB-p65 in the fundus of gastric ulceration-induced Wistar rats.

Spodoptera exigua Hubner is one of the most important agricultural pests in Thailand. The present study investigated the toxicity of crude extracts and phenylpropanoid compounds of A. galanga (Zingiberaceae) against S. exigua. After 24 h of a topical application bioassay on secondary instar larvae, the ethyl acetate extract estimation of the LD50 of A. galanga (Zingiberaceae) showed toxicity toward secondary instar S. exigua larvae (LD50 ~ 2.44 µg/larva). Phenylpropanoid compounds were isolated from the ethyl acetate A. galanga crude extract which has [1’S]-1’-acetoxychavicol acetate as highest toxicity (LD50 of 1.93 µg/larvae). [1’S]-1’-acetoxychavicol acetate also showed carboxylesterase activity inhibition. Thus, the ethyl acetate extract of A. galanga may have [1’S]-1’-acetoxychavicol acetate as an active ingredient for controlling this pest, which could be used as an alternative to application of environmentally insecticides for controlling S. exigua.

Turmeric (Curcuma longa) is one of the oldest medicinal plants, which has been used from ancient times. Turmeric derived from the Zingiberaceae family and curcumin is the bioactive component in the turmeric. Extensive researches have proven that most of the turmeric activities are due to curcumin. The purpose of this review was to provide a brief summary of the medicinal and nutritional value of curcumin. Turmeric is good source of macro and micronutrients such as protein, energy, vitamin and minerals, and it is known for various medicinal properties with antioxidant activities and is useful in conditions such as inflammation, ulcer and cancer. It has also antifungal, antimicrobial, renal and hepatoprotective activities. Turmeric is being used since past era to modern era; the major application of turmeric is due to antifungal and anti-bacterial property in skin and hair. The recently published papers in international cite as PubMed/ Medline, Science Citation Index and Google Scholar about turmeric were searched. Therefore, turmeric is widely used in treatment of various diseases such as diabetes, osteoarthritis etc. The nutrient content present in the curumin longa is in significant amount, so it can be helpful in combating the nutrients deficiency. Turmeric is also used for Ayurvedic and various cosmetic purposes like blood purification and different types of skin products.

Keeping in view the increasing importance of botanical insecticides, the various metabolites from different plants are being investigated for insecticidal activity. Alpinia galanga, a medicinal plant belonging to family Zingiberaceae exhibits a wide range of biological activities. In the present study, purified compounds of A. galanga i.e. 1′-acetoxychavicol acetate and galangin were evaluated for their effect on detoxifying enzymes viz. esterases, acid phosphatases, alkaline phosphatses and glutathione-S-transferases as well as DNA damage of the Spodoptera litura (Fab.) larvae. DNA damaging potential of both the compounds was ascertained using comet assay. The purified compounds significantly changed the enzymatic profile of S. litura as well as induced genotoxic effects. Change in enzyme activity and damage to DNA may lead to genetic and physiological changes in insect and ultimately affect its fitness and survival.

A new glucoside, 1R,3S-dihydroxy-1-(4′-hydroxyphenylethyl)cyclohexane-5-O-β-D-glucopyranoside, was isolated from the fruit along with retusin, 3,5-diacetoxy-1,7-bis(3,4-dihydroxyphenyl)heptane, 5-hydroxy-3,7,4′-trimethoxyflavone, and kumatakenin from the rhizomes of Amomum celsum Lamxay & M.F. Newman, Zingiberaceae. Their structures were determined by means of high resolution-mass spectrometry and nuclear magnetic resonance spectroscopy.

Alpinia conchigera Griff. is a plant species from the family Zingiberaceae. Coloquially known as wild ginger, Alpinia conchigera Griff. is used as food condiment and for traditional treatment of skin diseases. Isolation studies to identify bioactive compounds of rhizomes of Alpinia conchigera yielded seven compounds; 1'S-1'-acetoxychavicol acetate (1), trans-p-coumaryl diacetate (2), p-hydroxycinnamyl acetate (3), 1'S-1'-hydroxychavicol acetate (4) p-hydroxybenzaldehyde (5), stigmasterol (6) and β-sitosterol (7). Compounds 1, 2 and 5 were evaluated for antimicrobial activity against methicillin-resistant Staphylococcus aureus (MRSA). Among the compounds tested, Compound 1 showed good antimicrobial activity against the strain of MRSA with minimum inhibition concentration (MIC) value of 0.5 mg/mL. Meanwhile, Compounds 2 and 5 exhibited moderate activity with MIC value between 1.0 and 2.0 mg/mL. These findings indicate antimicrobial potential of 1'S-1'-acetoxychavicol acetate (1), compound derived from rhizome of Alpinia conchigera Griff. against MRSA, which warrant further investigation.

Zingiber montanum rhizomes are traditionally used for the treatment of numerous human ailments. The present study was carried out to investigate the inhibitory activity of the crude extract, chromatographic fractions, and purified compounds from Z. montanum rhizomes on the migration of MDA-MB-231 cells. The effect of the extract on cell migration was investigated by a scratch assay, which showed significant inhibition in a concentration-dependent manner. Vacuum liquid chromatography on silica gel afforded four fractions (Frs. 1 – 4), which were tested on cell migration in the scratch assay. Frs. 1 and 2 showed the most significant inhibition of MDA-MB-231 cell migration. The effect of the most potent fraction (Fr. 2) was further confirmed in a transwell migration assay. The study of Frs. 1 and 2 by gelatin zymography showed significant inhibition of MMP-9 enzyme activity. Chromatographic separation of Frs. 1 and 2 afforded buddledone A (1), zerumbone (2), (2E,9E)-6-methoxy-2,9-humuradien-8-one (3), zerumbone epoxide (4), stigmasterol (5), and daucosterol (6). In a cell viability assay, compounds 1 – 4 inhibited the viability of MDA-MB-231 cells in a concentration-dependent manner. The study of buddledone A (1) and zerumbone epoxide (4) on cell migration revealed that 4 significantly inhibited the migration of MDA-MB-231 cells in both scratch and transwell migration assays. The results of the present study may lead to further molecular studies behind the inhibitory activity of zerumbone epoxide (4) on cell migration and support the traditional use of Z. montanum rhizomes for the treatment of cancer.

Context: Curcumin, the active component of Curcuma longa L. (Zingiberaceae), exhibits a wide variety of biological activities including vasodilation and anti-inflammation. Objective: The relaxant effect of curcumin in tracheal smooth muscle (TSM) was not examined so far, thus, this study was designed to assess the relaxant effect of curcumin on rat TSM and examine the underlying mechanism(s) responsible for this effect. Materials and methods: TSM was contracted by KCl (60 mM) or methacholine (10 μM), and cumulative concentrations of curcumin (12.5, 25, 50, and 100 mg/mL) or theophylline (0.2, 0.4, 0.6, and 0.8 mM, as positive control) were added to organ bath. The relaxant effect of curcumin was examined in non-incubated or incubated tissues with atropine (1 μM), chlorpheniramine (1 μM), indomethacin (1 μM), and papaverine (100 μM). Sterculia setigera, Aframomum melegueta and Citrus aurantifolia are plants commonly used in African traditional medicine and especially in Benin to treat cough and asthma. The Phytochemical analysis of aqueous extracts of these plants, based on methods of precipitation and differential staining, revealed the presence of alkaloids, tannins, flavonoids, anthocyanins, leuco-anthocyanins, mucilages and reducing compounds. The presence of essential oils is found only in the mixture. The orally administration of aqueous extracts before induction of cough by inhalation of vapor 10 mL of 20% citric acid in Cavia porcellus (Guinea Pigs) inhibits the cough significantly with dose compared to the control and reference : Neo-codion ®. The extract of mixture Aframomum melegueta - Citrus aurantifolia (AMCA) was more active than Sterculia setigera’s (SS). These results confirm that aqueous extracts of Sterculia setigera and mixture Aframomum melegueta - Citrus aurantifolia, used in traditional medicine in Benin, have antitussive properties.

Methylated flavonols form a special group with modulating biological activities in comparison with kaempferol and quercetin. The present study isolated ten compounds including two kaempferol methyl ethers: 5-hydroxy-3,7,4′-trimethoxyflavone (1), 3-hydroxy-5,7,4′-trimethoxyflavone (6); four quercetin methyl ethers: retusin (5-hydroxy-3,7,3′,4′-tetramethoxyflavone) (4), 3,5-dihydroxy-7,3′,4′-trimethoxyflavone (5), 3,4′-dihydroxy-5,7,3′-trimethoxyflavone (7), and 3,5,7,3′,4′-pentamethoxyflavone (9); β-sitosterol (2); 5-hydroxy-1-(4′-hydroxyphenyl)eicosan-3-one (3); p-hydroquinone (8); and vanillic acid (10) from the rhizomes and fruit of Amomum koenigii J.F.Gmel. (Zingiberaceae). Their structures were determined by MS, NMR, and X-ray spectroscopic techniques. Among the methylated flavonols, 1, 4–7, and 9 were isolated for the first time from the rhizomes, while 1, 4, and 5 were isolated from the fruit. Compounds 2, 3, 7, 8, and 10 were reported for the first time from the species. Three main methylated flavonols 1, 4, and 5 were quantitatively analyzed in the rhizomes of A. koenigii by RP-HPLC-DAD; their contents were determined to be 1.81% (1), 1.38% (4), and 1.76% (5). The antimicrobial assay against Escherichia coli, Pseudomonas aeruginosa, Bacillus subtilis, Staphylococcus aureus, Aspergillus niger, Fusarium oxysporum, Candida albicans, and Saccharomyces cerevisiae and antioxidant DPPH scavenging test were performed for the isolated methylated flavonols.

Curcuma mangga Val. is one of Indonesian herbs from Zingiberaceae family that is under explored and could contain potentially active substances to serve as an antimalarial. This research intends not only to examine the antimalarial activity by means of heme polymerization inhibitor mechanism by using the ethanolic extract and fraction of Curcuma mangga Val. but also to identify its compound classification. The extract of temu manga was obtained by Soxhlet extraction method using ethanol solvent followed by fractionation using Vacuum Liquid Chromatography with solvent sequence n-hexane, n-hexane: ethyl acetate (2:1), ethyl acetate and ethanol. The extract and fraction were analyzed by using LC-MS and GC-MS. Activities of hem polymerization inhibition showed by IC 50 values which were obtained from analysis of relationship between concentration sample and the percentage of inhibition using the PROBIT on statistical software. The result of HPIA assay shows that the IC 50 value of ethanolic extract and ethanolic fraction of Curcuma mangga Val. rhizome are 2.273 and 1.479 mg/mL, respectively. It clearly shows that the heme polymerization inhibition activity of ethanolic fraction relatively better than that of ethanolic extract. Phytochemical screening determines the ethanolic extract contains saponin, terpenoid, and phenol while the ethanolic fraction contains terpenoid. Thus, terpenoid compound is presumed to be the inhibitor of heme polymerization. The results of analysis with LC-MS and GC-MS showed that the active compounds suspected to inhibit heme polymerization in ethanolic extract and fraction were (E) -labda-8 (17), 12-dien-15,16-dial and di-n-octyl phthalate, respectively. Background and objective Flavonols in plants are catalyzed by flavonol synthase (FLS) enzyme. FLS was reported expressed in flowers and fruits, i.e., Dianthus caryophyllus L. (Caryophyllaceae), Petunia hybrida Hort. (Solanaceae), Arabidopsis thaliana L. (Brassicaceae), Citrus unshiu Marc. (Rutaceae). However, none reported about FLS in medicinal plants, particularly those which possess anti-inflammatory activity. This study was aimed to extract and identify FLS in the rhizome of Boesenbergia rotunda (Zingiberaceae) and to determine quercetin in the ethanol extract of the rhizome. Materials and methods The protein extraction of the rhizome was carried out by employing Laing and Christeller's (2004) and Wang's (2014) methods. The extracted-proteins were separated by using SDS-PAGE, followed by the measurement of FLS intensity by using Gel Analyzer. The FLS-1 of recombinant A. thaliana was employed as the standard. The determination of quercetin in the rhizome was carried out using LC-MS. Results The FLS occurred as a thick band at 38 kDa with intensity 116-158. The LC chromatogram of the extract indicated a small peak at 7.94 min similar to that of quercetin standard. The MS spectra at 7.94 min indicated that quercetin is present in the B. rotunda rhizome (m/z = 303.0549). The concentration of quercetin in the extract is 0.022% w/v. Conclusion The FLS, an enzyme which plays an important role in producing quercetin, was detected in B. rotunda rhizome planted in Indonesia. As a consequence, quercetin in a small amount, was also quantified in the rhizome of this plant. This report will add a scientific insight of B. rotunda for biological sciences.

Zingiberofficinale Roscoe and Curcuma longa L. (Zingiberaceae) essential oils were obtained by the hydrodistillation method. The oils offered yields of 0.120% and 0.044% respectively. GC-MS analyzes allowed the identification of 11 constituents in the Curcuma longa essential oil, with major components: ar -turmerone (36.8%), β-tumerone (32.3%) and curlone (19,2%) For the Zingiberofficinale essential oil 19 constituents were identified, being: neral (22.9%), zingiberene (15.5%) and geranial (14.9%) the main components. The result of the toxicity test showed significant activity against Artemiasalina for both oils. The dose values required to eliminate 50% (LC 50 ) of the larvae were 100 µg / mL for Z. officinale and 25 µg / mL for C. longa . The antioxidant activity was performed against DPPH free radical. The most significant activity was exhibited by Z. officinale with an IC 50 value of 215.8 µg / mL. The antibacterial assay with Z. officinale oil showed efficacy against S. aureus and E. coli , with MIC values of ≥ 213.3 μg / mL and 85.3 μg / mL respectively. For C. longa , MIC values ≥ 1024 μg / mL were observed for both bacteria tested. Modulation with aminoglycoside antibiotics showed synergism of C. longa oil combined with amikacin against S. aureus , with a decrease in MIC from 1024 μg / mL to 14.7 μg / mL. Meanwhile, the Zingiberofficinale oil obtained a MIC reduction to 6.7 μg / mL, demonstrating greater efficacy. The results observed here demonstrate the biological potential of the species as cytotoxic and antimicrobial.

This article presents the results of studying the component composition of essential oils of leaves and rhizomes of C. longa grown in Uzbekistan. The chemical composition of essential oils of vegetative organs of the Curcuma longa (Zingiberaceae) plant introduced in Uzbekistan was studied for the first time. Essential oils of the plant were isolated by hydrodistillation, their chemical composition was studied by chromatography-mass spectrometry. In the composition of essential oils of leaves and rhizomes C. longa L., introduced in the Tashkent oasis, found 27 and 16 components, respectively, among which the leaf is dominated by α-fellandren (33.79%), α-terpinolene (24.28%), n-cymene (12.27%), eucalyptol (9.77%), and in the rhizomes-6-(1,3-dimethyl-buta-1,3-dienyl)-1,5,5-trimethyl-7-oxa-bicyclo [4.1.0] hept-2-EN (22.77%), α-curcumene (4.11%), 2,6,6,9-tetramethyl-tricyclo [5.4.0.0(2.8)] undec-9-EN (3.81%), α-zedren (1.14%), as well as 3 components that were not identified (the mass spectrum did not match the library data) their mass fraction was 47.47%, 10.41% and 3.46%. The composition of leaves and rhizomes of C. longa differ quantitatively and in quality. For example, α-curcumene in percentage content is more contained in rhizomes, and α-fellandren, which is the dominant content in leaves, is not found in the underground part.

Context Alpinia officinarum Hance (Zingiberaceae) is traditionally used to treat inflammation, pain, colds and digestive diseases. Objective To investigate the potential protective mechanism of total flavonoids from the rhizomes of A. officinarum (F-AOH) in ethanol-induced acute gastric in vivo and in vitro. Materials and methods In vivo: Gastric damage was induced in BALB/c mice by administering ethanol (10 mL/kg) after oral treatment with F-AOH at 126.8, 63.4 and 31.7 mg/kg or ranitidine (Ran) at 100 mg/kg (1 week of continuous gavage). In vitro: Gastric mucosal epithelial cells (GES-1) were incubated with F-AOH (8, 4 and 2 μg/mL) for 16 h and treated with 7% ethanol for 4 h. The extent of gastric damage was assessed histopathologically, and the expression of NF-κB, COX-2, TNF-α, iNOS and IL-1β was quantified by Western blot analysis. In addition, proinflammatory mediators and concentrations of motilin (MTL) and gastrin (GAS) were measured by ELISA test. Results F-AOH effectively reduced the ulcer index (from 23.4 ± 4.28 to 8.32 ± 1.5) and reduced release of inflammatory mediators (IL-1β, IL-6, TNF-α and PGE2), increased the content of nitric oxide and improved GAS and MTL secretion. The 50% inhibitory concentration (IC50) of F-AOH on cell damage was 17 μg/mL. F-AOH increased ethanol-induced cell survival (from 47 to 85%) and inhibited the expression of NF-κB, COX-2, TNF-α, IL-1β and iNOS proteins. Conclusions F-AOH inhibits ethanol-induced gastric mucosal damage, provides a theoretical basis for galangal in the treatment of other causes of GU, and promotes the application of galanga in the treatment of GU.

The chemical composition and larvicidal activity of essential oils from the leaves and rhizomes of Zingiber collinsii Mood & Theilade (Zingiberaceae) were reported. The main compounds in the leaf oil were α-pinene (25.6%), β-caryophyllene (16.8%), β-pinene (16.1%) and bicyclogermacrene (6.9%) while the rhizome oil consist mainly of camphene (22.5%), β-pinene (16.3%), α-pinene (9.0%) and humulene oxide II (9.0%). The rhizome oil demonstrated larvicidal effects towards fourth instant larvae of mosquito vectors. The highest mortality (100%) was observed at 24 h exposure against Aedes albopictus (concentration 100 μg/mL) and 48 h (concentration of 50 and 100 μg/mL), while the highest mortality (100%) was observed for Culex quinquefasciatus at 24 h and 48 h at concentration of 100 μg/mL. The 24 h mosquito larvicidal activity of the rhizome oil against Ae. albopictus were LC50 = 25.51 μg/mL; LC90 = 40.22 μg/mL and towards Cx. quinquefasciatus with LC50 = 50.11 μg/mL and LC90 = 71.53 μg/mL). However, the 48 h larvicidal activity were LC50 = 20.03 μg/mL and LC90 = 24.51 μg/mL (Ae. albopictus), as well as LC50 = 36.18 μg/mL and LC90 = 55.11 μg/mL (Cx. quinquefasciatus). On the other hand, no appreciable mortality and larvicidal activity was observed for the leaf oil. The larvicidal activity of the essential oils of Z. collinsii was being reported for the first time.

In this study, the effects of 6-paradol (6P) and 6-paradol-β-glucoside (6PG) on neuritogenesis were investigated using PC12 cells. Treatment with 200 μM 6P or 6PG and nerve growth factor (NGF) (5 ng mL-1) increased the number of elongated dendritic cells 8.7 and 5.4 times, respectively, compared to that with NGF (5 ng mL-1) treatment alone. 6P and 6PG did not stimulate the phosphorylation of extracellular regulated protein kinases (ERK)1/2 and cAMP response element-binding protein (CREB) in the tropomyosin receptor kinase A (TrkA) pathway as their activities were suppressed by the pathway inhibitor, k252a. 6P enhanced Ca2+ influx into the cells, whereas 6PG had no effect on Ca2+ influx, although it stimulated PC12 cell differentiation. High-performance liquid chromatography (HPLC) analysis of 6PG in PC12 culture medium suggested that 6PG was deglycosylated to generate 6P, which exhibited the effect. Furthermore, the bioactivities of 6P and 6PG were investigated in mice, and the results revealed that they ameliorated short-term memory loss in animals during behavioral testing.

Curcuma aromatica, the herbal medicine belongs to Zingiberaceae family, is well known for anti-tumor activity through multiple pathways and a potential candidate for complementary medicine in cancer treatment. The aims of this study were to distinguish between Curcuma species based on polymorphisms of the nucleotide sequence of chloroplast DNA (cpDNA) and preliminarily analyze their potential-anticancer compounds. Totally six samples supposed C. aromatica growing in An Giang province, Vietnam were collected. The contents of curcumin, curdione, and germacrone in the six samples were analyzed and compared by using the high-performance liquid chromatography (HPLC) method. All specimens were identified according to their trnSfMintergenic spacer sequences by Sanger sequencing. Among the six samples, three were determined as C. aromatica, two were C. longa, and one was C. zedoaria. Curcumin, curdione, and germacrone, known as anticancer compounds, were simultaneously found in sample NT3 that identified as C. aromatica by Sanger sequencing. The obtained results revealed a potential herbal candidate for complementary and alternative medicine.

BackgroundZingiber zerumbetrhizome and its bioactive metabolites have previously been reported to exhibit innumerable pharmacological properties particularly anti-inflammatory activities. In the present study, the 80% ethanol extract, essential oil and zerumbone ofZ. zerumbetrhizomes were explored for their in vitro immunosuppressive properties on chemotaxis, CD11b/CD18 expression, phagocytosis and chemiluminescence of isolated human polymorphonuclear neutrophils (PMNs).MethodsThe extract was analyzed quantitatively by performing a validated reversed phase high performance liquid chromatography (RP-HPLC). Zerumbone was isolated by chromatographic technique while the essential oil was acquired through hydro-distillation of the rhizomes and further analyzed by gas chromatography (GC) and GC-MS. Chemotaxis assay was assessed by using a 24-well cell migration assay kit, while CD18 integrin expression and phagocytic engulfment were measured using flow cytometry. The reactive oxygen species (ROS) production was evaluated by applying lucigenin- and luminol-enhanced chemiluminescence assays.ResultsZerumbone was found to be the most abundant compound in the extract (242.73 mg/g) and the oil (58.44%). Among the samples tested, the oil revealed the highest inhibition on cell migration with an IC50value of 3.24 μg/mL. The extract, oil and zerumbone showed moderate inhibition of CD18 integrin expression in a dose-dependent trend.Z. zerumbetextract showed the highest inhibitory effect on phagocytic engulfment with percentage of phagocytizing cells of 55.43% for PMN. Zerumbone exhibited strong inhibitory activity on oxidative burst of zymosan- and PMA-stimulated neutrophils. Zerumbone remarkably inhibited extracellular ROS production in PMNs with an IC50value of 17.36 μM which was comparable to that of aspirin.ConclusionThe strong inhibition on the phagocytosis of neutrophils byZ. zerumbetextract and its essential oil might be due the presence of its chemical components particularly zerumbone which was capable of impeding phagocytosis at different stages.

The essential oils of Curcuma longa L. (Zingiberaceae) were extracted from senescenced leaves by hydrodistillation and analyzed by GC/MS. Leaf essential oils of turmeric were found to be rich in monoterpenes. The GCMS analysis of leaf essential oil of 29 accessions showed presence of total 41 compounds mainly α-phellandrene, p-cymene, α-terpinolene and p-cymen-8-ol as the four major compounds. Based on identification of three major compounds from each oil, 13 compounds viz. β-Myrcene, α-Phellandrene, α-Terpinene, Limonene, 1,8-Cineole, p-cymene, τ-terpenine, α-Terpinolene, 1,3,8-p-Menthatriene, p-menth-1-en-4-ol, Borneol, p-menth-1-en-8-ol and p-cymen-8-ol were found to be the major constituents from each accession. The number of compounds in accessions varied from five in NBH-20 to 22 in Prabha. Out of 41 compounds, δ-Elemene was reported only in R. Sonia while β-Bisabolene was detected in Roma and P. Peetabh only. Based on the major compound(s) of turmeric leaf oil, industrial applications can be explored.

Wualae (Etlingera elatior (Jack) R.M Smith) is one of the plants in the Zingiberaceae family, widely used in medicine ranging from rhizomes, fruits, and flowers. Empirically Wualae are usually used by people for treating skin diseases, flavoring food, a natural soap, and in Southeast Sulawesi, it is used to treat typhoid fever. This study aims to examine the antioxidant activity of Wualae (Etlingera elatior) plant. The plant extracts were extracted by maceration method, antioxidant activity test was performed by 2,2-Diphenyl-1-picrylhydrazyl (DPPH) method. The results showed that ethanol extracts of fruits, leaves, stems and rhizomes of Wualae (Etlingera elatior) has the potential antioxidant activity with the IC50 values of 72.518 mg/L, 99.890 mg/L, 52.345 mg/L and 58.638 mg/L, respectively. and Vitamin C has IC50 value of 3,787 mg/L.
