The efficacy of needle-shaped nano-hydroxyapatite (nHA; Ca10(PO4)6(OH)2) as a phosphate (Pi) fertilizer was evaluated as well as its impact on soil and soybean (Glycine max) bacterial and fungal communities. Microbial communities were evaluated in soy fertilized with nHA using ITS (internal transcribed spacer) and 16S rRNA high-throughput gene sequencing. Separate greenhouse growth experiments using agriculturally relevant nHA concentrations and application methods were used to assess plant growth and yield compared with no Pi (−P), soluble Pi (+P), and bulk HA controls. Overall, nHA treatments did not show significantly increased growth, biomass, total plant phosphorus concentrations, or yield compared with no Pi controls. Soil and rhizosphere community structures in controls and nHA treatment groups were similar, with minor shifts in the nHA-containing pots comparable to bulk HA controls at equal concentrations. The implementation of nHA in an agriculturally realistic manner and the resulting poor soy growth advises that contrary to some reports under specialized conditions, this nano-fertilizer may not be a viable alternative to traditional Pi fertilizers. If nano-phosphate fertilizers are to achieve their conjectured agricultural potential, alternative nHAs, with differing morphologies, physicochemical properties, and interactions with the soil matrix could be investigated using the evaluative procedures described.

Long-term space missions will expose crew members, their cells as well as their microbiomes to prolonged periods of microgravity and ionizing radiation, environmental stressors for which almost no earth-based organisms have evolved to survive. Despite the importance of maintaining genomic integrity, the impact of these stresses on DNA polymerase-mediated replication and repair has not been fully explored. DNA polymerase fidelity and replication rates were assayed under conditions of microgravity generated by parabolic flight and compared to earth-like gravity. Upon commencement of a parabolic arc, primed synthetic single-stranded DNA was used as a template for one of two enzymes (Klenow fragment exonuclease+/−; with and without proofreading exonuclease activity, respectively) and were quenched immediately following the 20 s microgravitational period. DNA polymerase error rates were determined with an algorithm developed to identify experimental mutations. In microgravity Klenow exonuclease+ showed a median 1.1-fold per-base decrease in polymerization fidelity for base substitutions when compared to earth-like gravity (p = 0.02), but in the absence of proofreading activity, a 2.4-fold decrease was observed (p = 1.98 × 10−11). Similarly, 1.1-fold and 1.5-fold increases in deletion frequencies in the presence or absence of exonuclease activity (p = 1.51 × 10−7 and p = 8.74 × 10−13), respectively, were observed in microgravity compared to controls. The development of this flexible semi-autonomous payload system coupled with genetic and bioinformatic approaches serves as a proof-of-concept for future space health research.

The action of three distinct recombinant antifreeze proteins (AFPs) as methane hydrate inhibitors was examined using a recently-developed reactor. Compared with traditional approaches, this reactor uses minimal reactant volumes and short experimentation times to assess phase equilibria, kinetics and morphology of a hydrate system in a single experiment (3-in-1). Two of the recombinant AFPs are considered highly active with respect to the inhibition of ice: ‘Maxi’, a fish AFP, and a beetle AFP (TmAFP). The third protein from a grass, is classified as a low activity AFP (LpAFP). ‘Maxi’, an AFP that has not been tested previously as a hydrate inhibitor, slowed hydrate growth rates up to an order of magnitude compared to pure water. TmAFP and LpAFP also exhibited kinetic inhibition, but were less effective than ‘Maxi’. In the presence of AFPs, hydrate films were thinner and showed a single growth mechanism compared to multiple crystal growth mechanisms observed in control experiments. The addition of TmAFP generated large irregular hydrate halos that propagated outside the original water boundary. Halo propagation was somewhat less prominent with LpAFP, and was not observed with ‘Maxi’. Although, none of the AFP’s showed thermodynamic inhibition properties, ‘Maxi’ appeared to form clusters of hydrate which remained metastable in the liquid–vapour region.

Titanium dioxide (TiO2) nanoparticles (NPs) are used as an additive (E171 or INS171) in foods such as gum, candy and puddings. To address concerns about the potential hazardous effects of ingested NPs, the toxicity of these food-grade NPs was investigated with a defined model intestinal bacterial community. Each titania preparation (food-grade TiO2 formulations, E171-1 and E171-6a) was tested at concentrations equivalent to those found in the human intestine after sampling 1-2 pieces of gum or candy (100 250 ppm). At the low concentrations used, neither the TiO2 food additives nor control TiO2 NPs had an impact on gas production and only a minor effect on fatty acids profiles (C16:00, C18:00, 15:1 w5c, 18:1 w9c and 18:1 w9c, p < 0.05). DNA profiles and phylogenetic distributions confirmed limited effects on the bacterial community, with a modest decrease in the relative abundance of the dominant Bacteroides ovatus in favor of Clostridium cocleatum (-13% and +14% respectively, p < 0.05). Such minor shifts in the treated consortia suggest that food grade and nano-TiO2 particles do not have a major effect on human gut microbiota when tested in vitro at relevant low concentrations. However, the cumulative effects of chronic TiO2 NP ingestion remain to be tested.

Fruit flies, Drosophila melanogaster, have been traditionally valued as a simple model system due to their easy and inexpensive culture, their relatively compact genome, and the variety of available genetic tools. However, due to similarities of their neurological and developmental pathways with those of vertebrates, Drosophila also offers advantages for developmental toxicity assays. The ability to distinguish the effects of a toxicant on adult females, males, and the developing offspring adds to the usefulness of this model. Here we describe key techniques to screen chemicals and other potential emerging toxicants such as nanoparticles on adult Drosophila female and male reproductive success. In addition, assessments of relative toxicity can be revealed by viability assays at each developmental stage from the embryo to the pharate, or preemergent, adult.

Life can persist under severe osmotic stress and low water activity in hypersaline environments. On Mars, evidence for the past presence of saline bodies of water is prevalent and resulted in the widespread deposition of sulfate and chloride salts. Here we investigate Spotted Lake (British Columbia, Canada), a hypersaline lake with extreme (>3 M) levels of sulfate salts as an exemplar of the conditions thought to be associated with ancient Mars. We provide the first characterization of microbial structure in Spotted Lake sediments through metagenomic sequencing, and report a bacteria-dominated community with abundant Proteobacteria, Firmicutes, and Bacteroidetes, as well as diverse extremophiles. Microbial abundance and functional comparisons reveal similarities to Ace Lake, a meromictic Antarctic lake with anoxic and sulfidic bottom waters. Our analysis suggests that hypersaline-associated species occupy niches characterized foremost by differential abundance of Archaea, uncharacterized Bacteria, and Cyanobacteria. Potential biosignatures in this environment are discussed, specifically the likelihood of a strong sulfur isotopic fractionation record within the sediments due to the presence of sulfate reducing bacteria. With its high sulfate levels and seasonal freeze-thaw cycles, Spotted Lake is an analog for ancient paleolakes on Mars in which sulfate salt deposits may have offered periodically habitable environments, and could have concentrated and preserved organic materials or their biomarkers over geologic time.

The performance of polyvinylpyrrolidone (PVP) and polyvinylcaprolactam (PVCap) as kinetic gas hydrate inhibitors in saline solutions and with heptane was evaluated using high pressure microdifferential scanning calorimetry, as well as with a new apparatus, consisting of two high pressure stainless steel crystallizers. Although PVP and PVCap were found to prolong natural gas hydrate induction time in saline solutions, nucleation was followed by catastrophic hydrate crystal growth. PVP was found to be more effective in this case, since this hydrate growth was modestly slower. The addition of n-heptane to the natural gas in the system created a 4th phase. This resulted in increased induction time and a slowing of hydrate growth relative to the gas mixture. Unexpectedly, in the presence of n-heptane, addition of kinetic hydrate inhibitors (KHIs) decreased induction time, but catastrophic growth did not occur. Here PVCap was more effective than PVP in both prolonging the induction time and decreasing the rate of hydrate crystal growth. Once formed, however, hydrate decomposition took longer and proceeded in two steps in the presence of n-heptane. This observation has profound applications on the use these KHIs under ocean field conditions. In the case of hydrate blockages, our observations that hydrate dissociation started later with the KHIs and complete dissociation took longer could have far reaching economic implications for industry. © 2013 Elsevier Ltd. A Lolium perenne ice-binding protein (LpIBP) demonstrates superior ice recrystallization inhibition (IRI) activity and has proposed applications in cryopreservation, food texturing, as well as in being a "green" gas hydrate inhibitor. Recombinant production of LpIBP has been previously conducted in bacterial and yeast systems for studies of protein characterization, but large-scale applications have been hitherto limited due to high production costs. In this work, a codon-optimized LpIBP was recombinantly expressed and secreted in a novel one-step vector system from the nuclear genome of the green microalga Chlamydomonas reinhardtii. Both mixotrophic and photoautotrophic growth regimes supported LpIBP expression, indicating the feasibility of low-cost production using minimal medium, carbon dioxide, and light energy as input. In addition, multiple growth and bioproduct extraction cycles were performed by repetitive batch cultivation trials, demonstrating the potential for semi-continuous production and biomass harvesting. Concentrations of recombinant protein reached in this proof of concept approach were sufficient to demonstrate IRI activity in culture media without additional purification or concentration, with activity further verified by thermal hysteresis and morphology assays. The incorporation of the recombinant LpIBP into a model gas hydrate offers the promise that algal production may eventually find application as a "green" hydrate inhibitor.

The potential for hydrate inhibition in a stirred autoclave at a subcooling of 2.3 K has been tested on two bacterial isolates. Chryseobacterium sp. C14 survives multiple freeze–thaw events and inhibits ice recrystallization, and Escherichia coli has neither of these properties but is a biofilm producer. Both strains showed methane hydrate inhibition, with a significant reduction in total hydrate formed. Chryseobacterium delayed hydrate nucleating time, to a similar level found for the commercial kinetic inhibitor, polyvinylpyrrolidone (PVP; 0.2 wt %). However, in the presence of E. coli, in comparison to PVP, the time to nucleation was almost tripled and the hydrate growth rate was reduced by half. Because of the variation inherent with the microbial samples, likely a result of the complexities associated with varying cell numbers and metabolic activity, numerous experiments were required. Analysis of the distinct hydrate growth profiles shown by the two bacterial strains indicated that different molecular sources were likely responsible for the observed inhibition. As a working hypothesis, it is suggested that the inhibition observed by Chryseobacterium cultures was partially due to ice recrystallization inhibition properties, while that of E. coli may be due to secreted macromolecules and the effect of biofilm.

Antifolates, such as methotrexate (MTX), are the treatment of choice for numerous cancers. MTX inhibits dihydrofolate reductase (DHFR), which is essential for cell growth and proliferation. Mammalian cells can acquire resistance to antifolate treatment through a variety of mechanisms but decreased antifolate titers due to changes in drug efflux or influx, or alternatively, the amplification of the DHFR gene are the most commonly acquired resistance mechanisms. In Drosophila, however, a resistant phenotype has only been observed to occur by mutation resulting in a MTX-resistant DHFR. It is unclear if differences in gene structure and/or genome organization between Drosophila and mammals contribute to the observed differences in acquired drug resistance. To investigate if gene structure is involved, Drosophila Dhfr cDNA was transfected into a line of CHO cells that do not express endogenous DHFR. These transgenic cells, together with wild-type CHO cells, were selected for 19 months for resistance to increasing concentrations of MTX, from 50- to 200-fold over the initial concentration. Since Drosophila Dhfr appears to have been amplified several fold in the selected transgenic mammalian cells, a difference in genome organization may contribute to the mechanism of MTX resistance.

The bactericidal and fungicidal properties of silver (Ag) and Ag salt colloids make them popular choices for a variety of applications including water purification, bio-medical therapies for burns, epilepsy, mental illness, gastroenteritis, syphilis and gonorrhea, amongst other diseases. In addition, individuals deliberately drink colloidal Ag and Ag salts in the belief that this self- therapy will be beneficial. Despite this widespread usage, very little is known about the impact of this metal on human gut consortia. Here we investigated the impact of Ag chloride colloids, at several concentrations, on a defined anaerobic and facultative anaerobic gut bacterial community developed from the collected stool of a healthy donor. Defined microbial ecosystem therapeutic- 1 (MET-1) consortia were exposed to different concentrations of colloidal Ag (25-200 mg/L) for 48 hrs and compared to unamended cultures. Carbon dioxide and nitrogen gases recovered from the headspace of anaerobic cultures were analyzed by an Agilent Technologies 7890B Gas Chromatograph. Fatty acid methyl ester profiles were extracted following the MIDI Sherlock Microbial Identification System protocol and detected by gas chromatography. Colloidal AgCl toxicity was also monitored by a suite of DNA analyses including polymerase chain reaction (PCR), denaturing gradient gel electrophoresis (DGGE) and 16S ribosomal RNA gene fragment 454-pyrosequencing. The ecdysone receptor(1), which is a heterodimer of EcR and the retinoic acid receptor (RXR) homolog, Ultraspiracle (USP), has been well studied in the evolutionarily advanced and derived insects, the flies and moths. It is less well characterized in more primitive insect orders such as the Orthoptera, which include the grasshoppers and locusts. Following our previous isolation from Locusta migratoria (Lm) of a shorter RXR isoform (now called LmRXR-S), the isolation of a second, longer isoform (LmRXR-L) that appears to have characteristics of a ligand-modulated nuclear receptor is reported here. Transcripts for both isoforms, as well as LmEcR, were detected in embryos and in females during oocyte maturation. After expression in E. coli, both LmRXR-S and LmRXR-L form heterodimers with recombinant LmEcR in vitro which bind the active ecdysteroid, ponasterone A. Binding was only weakly competed for by ecdysone agonists that are known to be toxic to more advanced insects, suggesting functionally significant divergence in EcR ligand binding domains. In contrast, the DNA binding domain of LmEcR is less divergent and a protein complex, presumably LmEcR/LmRXR, that bound the ecdysone response element, IR-1, was detected in locust nuclear extracts. Because of reports of juvenile hormone (JH III) binding to Drosophila USP and the observed in silico RXR-like ligand-binding site in LmRXR-L, the recombinant proteins were also tested for binding to JH III. Neither LmRXR isoform, alone or in combination with LmEcR, bound JH III at nanomolar concentrations.

The effect of Type I fish antifreeze protein (AFP) from the winter flounder, Pleuronectes americanus (Walbaum), (WfAFP) on the formation of tetrahydrofuran (THF) clathrate hydrate was studied by observing changes in THF crystal morphology and determining the induction time for nucleation. AFP retarded THF clathrate-hydrate growth at the tested temperatures and modified the THF clathrate-hydrate crystal morphology from octahedral to plate-like. AFP appears to be even more effective than the kinetic inhibitor, polyvinylpyrrolidone (PVP). Recombinant AFP from an insect, a spruce budworm, Choristoneura fumiferana (Clem.), moth, (Cf) was also tested for inhibition activity by observation of the THF-hydrate-crystal-growth habit. Like WfAFP, CfAFP appeared to show adsorption on multiple THF-hydrate-crystal faces. A protein with no antifreeze activity, cytochrome C, was used as a control and it neither changed the morphology of the THF clathrate-hydrate crystals, nor retarded the formation of the hydrate. Preliminary experiments on the inhibition activity of WfAFP on a natural gas hydrate assessed induction time and the amount of propane gas consumed. Similar to the observations for THF, the data indicated that WfAFP inhibited propane-hydrate growth. Taken together, these results support our hypothesis that AFPs can inhibit clathrate-hydrate growth and as well, offer promise for the understanding of the inhibition mechanism. PACS No.: 87.90ty

Although juvenile hormone (JH) has essential roles in insect development and reproduction, the molecular mechanisms of gene regulation by JH remain an enigma. In Locusta migratoria, the partially palindromic 15-nt sequence, GAGGTTCGAGA/TCCTT/C, found upstream of a JH-induced gene, jhp21, was designated as a putative juvenile hormone response element (JHRE). When JH-deprived adult female locusts were treated with the active JH analog, methoprene, a fat body nuclear factor that bound specifically to JHRE appeared after 24 h. Binding exhibited a preference for an inverted repeat with GAGGTTC in the left half-site, a single nucleotide spacer, and a right half-site in which some variation is acceptable. Binding to JHRE was abolished by phosphorylation catalyzed by a C-type protein kinase present in the nuclear extracts. The DNA-binding protein is thus believed to be a transcription factor, which is brought to an active state through the action of JH and then participates in the regulation of certain JH-dependent genes.

The widespread use of silver nanoparticles (AgNPs) has prompted studies on different species and at various trophic levels, however, little is known about possible toxicological impacts on their corresponding microbiota. Here the effects of AgNPs on a defined bacterial community established from the collected stool of a healthy donor have been investigated. β-Helix structure and ice-binding properties of a hyperactive antifreeze protein from an insect

We have previously identified a Thr- and Cys-rich thermal hysteresis (antifreeze) protein (THP) in the beetle Tenebrio molitor that has 10-100 times the freezing point depression activity of fish antifreeze proteins. Because this 8.4 kDa protein is significantly different in its properties from THP preparations previously reported from this insect, a thorough search was undertaken for other antifreeze types. Many active proteins were observed, but all appeared to be isoforms of the THP that differed in their number of 12-amino acid repeats (consensus sequence CTxSxxCxxAxT), amino acid substitutions, and N-linked glycosylation. Mass spectral analysis has matched most of these isoforms with cDNA sequences of 17 different clones from a larval fat body library that encode eight different mature THPs containing 84, 96, or 120 amino acids. Genomic Southern blots suggest there may be 30-50 tightly linked copies of the gene, which is a signature consistently seen with unrelated fish antifreeze protein genes, and one that has been associated with the need to rapidly increase gene product in response to climate change. A three-dimensional model is proposed for the fully disulfide-bonded structure of T. molitor THP, which can accommodate addition or deletion of 12-amino acid repeats. The structure is a beta-helix that places most of the Thr in a regular array on one side of the protein to form a putative ice-binding surface.

A malathion-resistant strain of Culex tarsalis has a malathion carboxylesterase which rapidly hydrolyzes the insecticide. This is in contrast to organophosphate-resistant strains of C. quinquefasciatus and C. pipiens, which have elevated levels of general B esterases due to amplification of the corresponding genes, producing increased amounts of enzyme which appear to protect the insects by sequestering the insecticide. The contribution to resistance of the homologous esterase B3 (Est beta 3) gene (est beta 3) in C. tarsalis was investigated by cloning and characterizing sequences from resistant and susceptible strains. est beta 3 is similar to est beta 1, both structurally and in sequence. The first intron of est beta 3, however, has a region of extensive repeats which may be responsible for the inefficient processing of the transcript. Southern blots indicate that the gene is single copy in both strains, and northern blots show that it is not greatly overexpressed in the resistant insects. est beta 3 cDNAs from resistant and susceptible strains have 98% amino acid identity. It appears that, in contrast to other studies, est beta 3 does not play a significant role in insecticide resistance in our strains of C. tarsalis, and the molecular responses of pest insects to organophosphates may be more diverse than has been suggested.

The formation kinetics of methane/ethane/propane hydrate in the presence of kinetic inhibitors was investigated using 1 H nuclear magnetic resonance imaging (MRI) as well as a more-traditional method using a stirred autoclave. These studies were facilitated by fabricating a multi-drop insert for 1 H NMR micro-imaging, which allowed the comparison of the performance of microliter quantities of several inhibitors simultaneously and under the same conditions. Both methods showed that hydrate nucleation and growth were delayed significantly in the presence of inhibitors, which included two biological inhibitors (antifreeze proteins) and a commercial inhibitor. The results demonstrate that MRI is a useful tool for the visualization and evaluation of the performance of kinetic inhibitors on mixed gas hydrate formation. The MRI technique should prove especially valuable in the case of analysis of potential inhibitors, pre-commercialization, which are available in only limited quantities, such as biological inhibitors. This technique may also find utility in the exploration of differences in inhibitor performance, which may suggest distinct mechanisms of inhibitor action.

Levels of a 28 kDa hemolymph protein, desiccation stress protein (dsp28), in the mealworm beetle, Tenebrio molitor, increase in response to desiccation and cold stress and are also developmentally regulated under nonstress conditions. Dsp28 mRNA is produced in the fat body, and its abundance changes dramatically throughout development. Transcript abundance increases throughout larval development, drops at pupation, and increases again in adults, the highest levels being found in females. The juvenile hormone (JH) analogue, methoprene, increased dsp28 transcript accumulation in pupae, suggesting that changing JH titres have a role in developmental expression and also perhaps contribute to dsp28 regulation during conditions of environmental stress. Genomic DNA, containing the entire dsp28 coding region plus 1.3 kb of upstream sequence, was isolated and potential regulatory sequences, including putative JH response elements, were identified. © 1996 Wiley-Liss, Inc.

Three malathion carboxylesterase (MCE) phenotypes have been described previously in Lucilia cuprina adults: low, intermediate, and high. The MCE specific activities of adults with the intermediate and high phenotypes are 21- and 33-fold higher than those with the low phenotype, but the high phenotype is almost 1000-fold more resistant to malathion than either of the other phenotypes. Here we show that MCE activity also peaks in Day 4 larvae from three lines representative of these phenotypes. The MCE activity of the low-line larvae is only 2.7- and 4-fold lower than those of the intermediate and high-line larvae, respectively. The relatively high MCE activity of the low-line larvae is largely explained by their crop activity. Assays in the presence of esterase inhibitors reveal three distinct types of MCE activity across the three lines and two developmental stages. Activity in the low-line adults is not completely inhibited by paraoxon and is classified as a subclass II carboxylesterase. The MCE activities in the larvae of the low and intermediate lines and adults from the intermediate line are inhibited by low concentrations of paraoxon, classifying them as subclass I carboxylesterases. The MCE activities in the high-line larvae and adults are also classified as subclass I carboxylesterases, but they are more sensitive to inhibition by triphenyl phosphate than those in the other two lines. These data suggest that MCE in the malathion-resistant high line may be structurally different from the MCEs in the malathion-susceptible intermediate and low lines.

Resistance to the organophosphate insecticide, malathion, in a strain of Culex tarsalis mosquitoes is due to increased activity of a malathion carboxylesterase (MCE). To determine whether resistance was due to a qualitative or quantitative change in the MCE, the enzyme was purified from both malathion-resistant and -susceptible mosquitoes. Enzyme kinetic measurements revealed that the two strains have one MCE in common, but resistant mosquitoes also have a unique MCE which hydrolyses malathion 18 times faster. Interestingly, this MCE does not hydrolyse alpha-naphthyl acetate, a substrate commonly used to detect increased levels of esterases in other organophosphate-resistant insects. Unlike the over-produced esterase of some related mosquito species, each MCE in C. tarsalis accounts for only a small fraction (0.015%) of the total extractable protein in either strain. Therefore, resistance in these insects is due to the presence of a qualitatively different enzyme, and not to a quantitative increase of a non-specific esterase. This study therefore demonstrates that the underlying biochemical mechanisms of insecticide resistance in one insect cannot necessarily be predicted from those of another, even closely related species.

The formation of hydrate plugs in oil and gas pipelines is a serious industrial problem and recently there has been an increased interest in the use of alternative hydrate inhibitors as substitutes for thermodynamic inhibitors like methanol. We show here that antifreeze proteins (AFPs) possess the ability to modify structure II (sII) tetrahydrofuran (THF) hydrate crystal morphologies by adhering to the hydrate surface and inhibiting growth in a similar fashion to the kinetic inhibitor poly-N-vinylpyrrolidone (PVP). The effects of AFPs on the formation and growth rate of high-pressure sII gas mix hydrate demonstrated that AFPs are superior hydrate inhibitors compared to PVP. These results indicate that AFPs may be suitable for the study of new inhibitor systems and represent an important step towards the development of biologically-based hydrate inhibitors.

Resistance to malathion in a strain of the Australian sheep blowfly is due to a 10-fold increase in malathion carboxylesterase (MCE) activity relative to a more susceptible strain. MCE was purified to apparent homogeneity from these two strains and was shown to be a monomer of 60,500, with a pI of 5.5 in both strains. Purified MCE from both populations had identical Km and Vmax values for the hydrolysis of malathion as well as for three other esterase substrates. Similarly, the kinetics of inhibition by several inhibitors were the same for the MCE from each strain. These data therefore suggest that resistance to malathion is due to a quantitative rather than a qualitative change in the MCE of the two strains. Estimation of the total MCE content in each strain showed that the resistant blowflies had nine times more MCE than the more susceptible insects. Although blowfly MCE showed greater specificity for naphthyl esters over malathion, it nevertheless hydrolyzes malathion faster than any other esterase yet isolated from an insect. This is in sharp contrast to previously studied insect strains in which organophosphate resistance has been attributed to large increases in nonspecific esterases that show very slow or no hydrolysis of the insecticides.

Adult flies of Drosophila melanogaster were examined for their tolerance to direct (cold-shock) and indirect chilling (chilling) injury. An exponential increase in mortality with low temperatures occurs. Chilling tolerance (T ch ) at 4 or 0°C was 6.3 days or 37.91 h for 50% survival (LT 50 ), respectively, while the cold-shock tolerance (LT 50 of T cs ) at −7°C was only 1.5 h. The increase in cold-shock tolerance (T cs ) at −7°C by acclimation at 4, or 0°C was rapid: the LT 50 increased about 50% with only 2 h of treatment in comparison with the highest LT 50 obtained by acclimation. An equilibrium point (T eq ), the optimal acclimation time for maximal survival of chilling and/or cold-shock, was calculated from the survival data of experiments designed to assess T ch and T cs . The T eq was found to be the half time of the acclimation period which resulted in 90% mortality at the chilling temperature (LT 90 of T ch ). Selection for tolerance to cold-shock or chilling over several generations resulted in a significant increase in T cs or T ch , respectively. Higher contents of glycogen and total proteins were observed in the cold selected lines than in the control line. Exposure to chilling temperatures eventually depleted the energy reserves, but the highest concentrations of triacylglycerols and proteins were observed at the acclimation time with the highest T cs . This suggests that systemic regulation in these energetic reserves may be an important part of the cold-hardening mechanisms to cope with the fluctuations of low temperatures causing cold-shock or chilling injury.

A synthetic natural gas mixture composed of methane, ethane, and propane in a batch reactor was used to form gas hydrates in the presence of two commercial, chemical kinetic inhibitors, polyvinylpyrrolidone (PVP) and H1W85281, and one biological inhibitor, antifreeze protein type III (AFP-III). Powder X-ray diffraction and nuclear magnetic resonance spectroscopy showed that structure II hydrates dominated, as expected, but in the presence of the chemical inhibitors, structure I was also present. Raman spectroscopy confirmed the complexity and the heterogeneity of the guest composition within these hydrates, which was also consistent with the gas analysis obtained using gas chromatography. However, in the presence of AFP-III, hydrates appeared to be relatively homogeneous structure II hydrates, with weaker evidence of structure I. When individual gas cage occupancies were calculated, both classes of inhibitors reduced large cage methane occupancy by 25%. With the chemical inhibitors, these large cage methane guests appeared to be substituted by ethane, likely resulting in a decreased driving force for hydrate production. In contrast to the near full occupancy of large cages with these inhibitors, almost 10% of the large cages were not filled when hydrates were formed in the presence of AFP-III, likely contributing to the easy decomposition of such hydrates seen in other studies. Therefore, hydrates formed in the presence of these two classes of inhibitors appear to be distinct, and as a consequence, their inhibition mechanisms, as well their practical utility in the field, are likely to be marked by important differences.

The effect of kinetic inhibitors, both chemical (PVP and H1W85281) and biological (type III antifreeze protein), on natural gas hydrate formation was investigated using high pressure differential scanning calorimetry (HP-DSC). The presence of inhibitors decreased the overall formation of methane/ethane/propane hydrate compared to systems without added inhibitors. As well, all of the inhibitors significantly delayed hydrate nucleation as compared to water controls. However, the two classes of inhibitors were distinguished by the formation of hydrates with different stabilities. A single hydrate melting peak was seen with the antifreeze protein (AFP), and this was consistent after recrystallization. In contrast, multiple hydrate melting events, some indicating the formation of hydrate structures with high stability, were observed in the presence of the chemical inhibitors, and these varied depending on the crystallization cycle. This heterogeneity suggests that the use of these chemical inhibitors (PVP and H1W85281) may present a special challenge to operators depending upon the gas mixture and environmental conditions and that AFPs may offer a more predictable, efficacious solution in these cases.

Gas hydrates represent both a bane and a potential boon to the oil and gas industry, and considerable research into hydrate formation has been undertaken. We have recently developed a multi-threaded version of a Monte Carlo crystal growth algorithm and applied it to simulate the growth of large structure II gas hydrates. This algorithm allows for the introduction of non-crystal molecules during simulations to study their effects on crystal growth rates and morphologies. Here, we report on our initial simulations of hydrate growth in the presence of poly(N-vinylpyrrolidone) (PVP). We have surveyed the PVP-hydrate interaction space by performing numerous simulations, each with a unique PVP-hydrate docking orientation. This survey produced a broad range of hydrate growth rates, ranging from almost complete suppression through to significant growth enhancement. The effect that PVP has on gas incorporation at crystal surfaces appears to be critical for differentiating between crystal growth inhibition and enhancement.

A novel hemolymph protein from Tenebrio molitor associated with desiccation stress has been purified and characterized. The protein contains cysteine; it was labeled in vivo using [35S]-cysteine, and the amino acid analysis showed a 4% cysteine content. In the native state dsp28 is a dimer with a single subunit Mr of 28,000. The synthesis of dsp28 and its concentration in larval hemolymph increase dramatically after desiccation. Dsp28 is a major component of total larval hemolymph protein representing 5% of total protein in control insects and up to 17% in desiccated larvae. The hemolymph of cold-acclimated larvae also contains elevated levels of dsp28.

The retinoid X receptor (RXR) is activated by its often elusive cognate ligand, 9- cis -retinoic acid (9- cis -RA). In flies and moths, molting is mediated by a heterodimer ecdysone receptor consisting of the ecdysone monomer (EcR) and an RXR homolog, ultraspiracle (USP); the latter is believed to have diverged from its RXR origin. In the more primitive insect, Locusta migratoria (Lm), RXR is more similar to human RXRs than to USPs. LmRXR was detected in early embryos when EcR transcripts were absent, suggesting another role apart from ecdysone signaling. Recombinant LmRXRs bound 9- cis -RA and all- trans -RA with high affinity (IC 50 = 61.2–107.7 nM; K d = 3 nM), similar to human RXR. To determine whether specific binding had functional significance, the presence of endogenous retinoids was assessed. Embryos were extracted by using modified Bligh and Dyer and solid-phase protocols to avoid the oily precipitate that makes this material unsuitable for assay. These extracts contained retinoids (5.4 nM) as assessed by RA-inducible Cyp26A1-promoter luciferase reporter cell lines. Furthermore, the use of HPLC and MS confirmed the presence of retinoids and identified in any embryo, 9- cis -RA, in addition to all- trans -RA. We estimate that whole embryos contain 3 nM RA, including 9- cis -RA at a concentration of 1.6 nM. These findings strongly argue for a functional role for retinoids in primitive insects and favor a model where signaling through the binding of 9- cis -RA to its RXR is established relatively early in evolution and embryonic development.

Drosophila research has been and continues to be an essential tool for many aspects of biological scientific research and has provided insight into numerous genetic, biochemical, and behavioral processes. As well, due to the remarkable conservation of gene function between Drosophila and humans, and the easy ability to manipulate these genes in a whole organism, Drosophila research has proven critical for studying human disease and the physiological response to chemical reagents. Methotrexate, a widely prescribed pharmaceutical which inhibits dihydrofolate reductase and therefore folate metabolism, is known to cause teratogenic effects in human fetuses. Recently, there has been resurgence in the use of methotrexate for inflammatory diseases and ectopic or unwanted pregnancies thus, increasing the need to fully understand the cytotoxicity of this pharmaceutical. Concerns have been raised over the ethics of studying teratogenic drugs like methotrexate in mammalian systems and thus, we have proposed a Drosophila model. We have shown that exposure of female Drosophila to methotrexate results in progeny with developmental abnormalities. We have also shown that methotrexate exposure changes the abundance of many fundamental cellular transcripts. Expression of a dihydrofolate reductase with a reduced affinity for methotrexate can not only prevent much of the abnormal transcript profile but the teratogenesis seen after drug treatment. In the future, such studies may generate useful tools for mammalian antifolate “rescue” therapies.

The kinetic characteristics of a purified insect dihydrofolate reductase (DHFR) have been described. The Km values for the substrate dihydrofolate and the cofactor NADPH have been estimated by primary and secondary Hanes plots to be 0.3 and 5.2 μM, respectively. Drosophila melanogaster DHFR can use folate and NADH at acidic pH values, but at a much lower rate than the preferred substrate and cofactor. Folic acid is a partial competitive inhibitor of Drosophila DHFR (Ki = 0.4 μM) and trimethoprim is a complete competitive inhibitor (Ki = 5.4 μM). Methotrexate binds less tightly to the Drosophila enzyme than to many other DHFRs (Kd = 0.9 nM). Drosophila DHFR is inhibited by KCl and organic mercurials and is slightly activated by urea. These data indicate that Drosophila DHFR has some characteristics which are typical of vertebrate DHFRs and others which are typical of prokaryotic DHFRs. The study of this enzyme, therefore, should aid in the definition of the structural features that are responsible for the kinetic characteristics in different DHFRs.Key words: dihydrofolate reductase, Drosophila melanogaster, methotrexate.

The inhibition activities of two antifreeze proteins (AFPs) on the formation of tetrahydrofuran (THF) clathrate hydrate have been tested. AFPs from fish (wfAFP) and insect (CfAFP) changed the morphology of growing THF hydrate crystals. Also, both AFPs showed higher activities in inhibiting the formation THF hydrate than a commercial kinetic inhibitor, poly(vinylpyrrolidone) (PVP). Strikingly, both AFPs also showed the ability to eliminate the "memory effect" in which the crystallization of hydrate occurs more quickly after the initial formation. This is the first report of molecules that can inhibit the memory effect. Since the homogeneous nucleation temperature for THF hydrate was measured to be 237 K, close to that observed for ice itself, the action of kinetic inhibitors must involve heterogeneous nucleation. On the basis of our results, we postulate a mechanism for heterogeneous nucleation, the memory effect and its elimination by antifreeze proteins.

The effect of Type I antifreeze protein (AFP) from winter flounder on the formation of propane hydrate and methane hydrate was studied. We show that the formation of both hydrates is inhibited significantly, with both nucleation and crystal growth being affected. Also, AFP showed the so-far unique ability to eliminate the ldquomemory effectrdquo in the reformation of gas hydrate. We have proposed a mechanism involving the interference of AFP with heterogeneous nucleation and subsequent growth of the hydrates. It is also shown that a number of samples must be studied in order to obtain meaningful statistics, and that magnetic resonance imaging provides a novel way of studying the nucleation and growth of hydrate in multiple droplets.

Certain freeze-intolerant insects produce antifreeze proteins (AFPs) during overwintering including the spruce budworm (Choristoneura fumiferana) and yellow mealworm (Tenebrio molitor) AFP gene families. However, only a few of the isoforms, encoded by their multiple-copy gene families, have been characterized. When expressed in bacterial systems the insect AFPs have to be denatured and refolded in vitro, a procedure that is not uniformly successful, presumably due to the beta-helix structure and the requirement for disulfide bonds. In an attempt to overcome these difficulties, bacterial vectors and hosts that have been developed to produce soluble, folded proteins, as well as a yeast expression system (Pichia pastoris) were employed. Bacterial expression resulted in low quantities of active recombinant protein for certain isoforms. In contrast, both small and large-scale fermentation of recombinant AFP in Pichia yielded substantial protein production (100 mg/L) but functional ice binding activity of protein produced in three different transformed yeast strains (KM71, X33 or GS115) was low. Inappropriate O-linked glycosylation of the Thr-rich AFPs appeared to be partially reversed by mild chemical deglycosylation, but activity remained low. Substantial quantities, as well as activity were recovered when a fish AFP, with disulfide bonds, but without potential Thr glycosylation sites was expressed in the yeast system.

Antifolates, such as methotrexate, are used to inhibit dihydrofolate reductase (DHFR), an enzyme essential for the biosynthesis of thymidylate, purines, and several amino acids. DHFR sequences corresponding to mutations found in a methotrexate resistant Drosophila S3 cell line (L30Q), a methotrexate resistant fly population (K31P, Q134K), as well as predicted in silico (L22R) were expressed in Chinese Hamster Ovary (CHO) cells. The L30Q and L22R DHFRs both conferred resistance to methotrexate. L22R DHFR provided approximately 200-fold resistance to methotrexate when compared to wild-type Drosophila DHFR allowing CHO(L22R) cells to divide in 10 microM methotrexate, a level of resistance not previously observed in any mammalian system. Constructs using this substitution in combination with other Drosophila DHFR specific residues would make excellent candidates for gene therapy and genetic markers in the treatment of certain human disorders.

There is increasing evidence that heat shock (HS) has long-term effects on electrophysio- logical properties of neurons and synapses. Prior HS protects neural circuitry from a subsequent heat stress but little is known about the mechanisms that mediate this plasticity and induce thermotolerance. Exposure of Locusta migratoria to HS conditions of 45°C for 3 h results in thermotolerance to hitherto lethal tempera- tures. Locust flight motor patterns were recorded during tethered flight at room temperature, before and after HS. In addition, intracellular action potentials (APs) were recorded from control and HS motoneurons in a semi-intact preparation during a heat stress. HS did not alter the timing of representative depressor or elevator muscle activity, nor did it affect the ability of the locust to generate a steering motor pattern in response to a stimulus. However, HS did increase the duration of APs recorded from neuropil segments of depressor motoneu- rons. Increases in AP duration were associated with protection of AP generation against failure at subse- quent elevated temperatures. Failure of AP generation at high temperatures was preceded by a concomitant burst of APs and depolarization of the membrane. The protective effects of HS were mimicked by pharmaco- logical blockade of IK 1 with tetraethylammonium (TEA). Taken together, these findings are consistent with a hypothesis that HS protects neuronal survival and function via K 1 channel modulation. © 2001 John

To facilitate studies on the hormonal control of development in the migratory locust, Locusta migratoria, we have undertaken the cloning of cDNAs for nuclear hormone receptors. Sequences obtained by polymerase chain reaction (PCR) showed homology with receptor family members including the ecdysteroid receptor (EcR). A cDNA clone corresponding to the EcR fragment includes an open reading frame of 1622 nucleotides, predicting a 59 kDa protein showing clear homology with EcRs and distinct from other classes of nuclear receptors. Northern analysis revealed a major transcript of 9.2 kb. In fifth instar fat body, the transcript was most abundant at the end of the instar when ecdysone titres are highest. There was no obvious evidence of EcR regulation by a juvenile hormone analog. Although its role in development may be similar, the locust ecdysone receptor (LmEcR) is divergent from EcRs characterized from insects belonging to the dipteran and lepidopteran orders, presumably reflecting the more ancestral sequence in the relatively primitive locust.

Malathion resistance in a strain of Culex tarsalis mosquitoes is due primarily to the activity of a malathion carboxylesterase (MCE). The resistant strain was 150 times more resistant to malathion than the susceptible strain and was weakly resistant to malaoxon and carbaryl, but not to any other insecticide tested. The phenotype could be reversed with the carboxylesterase inhibitor triphenylphosphate, but no synergism was observed with either the phosphatase or polysubstrate monooxygenase inhibitors, NaF and piperonyl butoxide. MCE is expressed throughout development and is most concentrated in the gut tissues of the larvae. Subcellular fractionation indicated that MCE was localized primarily in the mitochondria of resistant insects and the cytoplasm of susceptible insects. The enzyme was purified to homogeneity from both strains, and has a molecular weight of 59,000. However, chromatofocusing indicated that resistant insects have two MCEs with pIs of 6.8 and 6.2, while susceptible insects possessed only one MCE with a pI of 6.8. The MCE unique to the resistant strain hydrolysed malathion 18 times faster than the MCE common to both strains, suggesting that malathion resistance in C. tarsalis is due to the presence of a qualitatively different esterase in the resistant strain.

Resistance to the organophosphorus insecticide malathion in genetically related strains of the Australian sheep blowflyLucilia curprina was examined. Separate lines of blowflies were established by homozygosis of the fourth chromosome of the parental RM strain. Both the RM and the derived resistant (der-R) strains are approximately 100 times more resistant to malathion than the related susceptible der-S strain, resistance being correlated with a 45- to 50-fold increase in a malathion carboxylesterase (MCE) activity. MCE has a pH optimum ranging between 6.6 and 8.0 and is strongly inhibited by the carboxylesterase inhibitors triphenyl phosphate, paraoxon, and diiospropylfluorophosphate. Subcellular fractionation revealed that MCE was localized predominantly to the cytosol and mitochondria in both resistant and susceptible blowflies. A single MCE was purified to homogeneity from RM blowflies. It has a pI of 5.5, is a monomer of 60.5 kDa, and hydrolyzes malathion with aV max of 755 nmol/min/mg protein and aK m of 11.0 µM. L. cuprina have thus evolved a remarkable MCE which is faster and more efficient at hydrolyzing a specific insecticide than any other insect esterase yet described.

After 5 days at the restrictive temperature (29.5°C) adult Drosophila females heterozygous for the dominant temperature-sensitive mutation, L(3)3DTS, have an ecdysteroid level of about half that in mutant females at 22°C and subsequently become completely sterile due to the inviability of progeny embryos. The lethal phase of progeny from mutant females varies depending upon the length of time DTS-3 females are kept at a sublethal temperature of 27°C. Thus, the DTS-3 mutation shows a maternal effect, and a deficiency of ecdysteroids or ecdysteroid-induced gene products may be responsible for progeny lethality. This lethality cannot be attributed to a deficit in the products of the hormonally-regulated yolk polypeptide genes however, since yolk polypeptide mRNA and protein levels are not reduced in DTS-3 females at the restrictive temperature.

The dominant temperature-sensitive mutation L(3)3DTS (DTS-3) in Drosophila melanogaster causes lethality of heterozygotes during the third larval instar at the restrictive temperature (29°C). Temperature-shift experiments revealed two distinct temperature-sensitive periods, with lethal phases during the third larval instar (which may persist for 4 weeks) and during the late pupal stage. At 29°C mutant imaginal discs are unable to evert in situ, but did evert normally if cultured in the presence of exogenous ecdysterone or when implanted into wild-type larval hosts. The only morphologically abnormal tissue present in the lethal larvae is the ring gland, the prothoracic gland being greatly hypertrophied in third instar DTS-3 larvae. Injection of a single wild-type ring gland rescued these mutant larvae, indicating that the mutant gland is functionally, as well as morphologically, abnormal. Finally, the mutant larvae were shown to have less than 10% of the wild-type ecdysteroid levels. These results are all consistent with a proposed lesion in ecdysteroid hormone production in DTS-3 larvae. A comparison with the phenotypes of other “ecdysone-less” mutants is presented.

The role of esterases in malathion resistance in Culex tarsalis has been investigated. When larvae of a resistant and a sensitive strain were placed in water containing [ 14 C]malathion, malathion penetrated to give initially similar internal levels. With resistant mosquitoes, after 15 min the internal malathion concentration decreased to low levels while the monoacid degradation products accumulated in the larvae and were excreted into the surrounding water, whereas in susceptible larvae the internal malathion level stayed high and was lethal. It is suggested that the decrease in internal malathion and the resulting resistance were caused by an active malathion carboxylesterase in the resistant strain. A specific assay for malathion carboxylesterase with [ 14 C]malathion showed 55 times more activity in resistant than in susceptible larvae, whereas when general esterase activity was assayed with α-naphthyl acetate only 1.7 times the activity was found. Analyses by starch gel electrophoresis showed a peak of malathion carboxylesterase, 60-fold higher from resistant than from susceptible larvae, in a gel zone which did not stain for general esterase activity. General esterases that did not hydrolyze malathion showed different electrophoretic patterns in the two populations, which are likely due to the nonisogenic character of the strains. These results show that use of a specific assay and the demonstration of degradation of malathion in vivo are essential for assessment of the contribution of esterase activity to the malathion-resistant phenotype in mosquito populations.
