The versatility of anthanthrone-based materials in organic electronics is exploited by synthesizing four polymers, PANT, PANT-TBO, PANT-TBT, and PANT-TffBT. PANT represents the self-coupled homopolymer, while the other three are D–A (donor–acceptor) type copolymers. Each of them is successfully applied in three different devices, namely organic field-effect transistors (OFETs), organic photovoltaic cells (OPVs), and chemical sensors. The results indicate that anthanthrone could be a very promising building block for the construction of semiconductors for multi-purpose applications. Specifically, the four polymers exhibit p-type properties in OFETs with hole mobility values in the range of 10−4–10−3 cm2 V−1 s−1. In OPVs, the power conversion efficiencies (PCEs) of non-fullerene cells have a wide range from 0.28% to 5.21%. This indicates that the efficiency may reach a higher value via further structural modification by selecting suitable comonomers to produce new polymers. Additionally, based on the parameters of the OFET sensor devices and the variation in the backbone structures of the four polymers, they show varying response trends for four chemicals: octane, toluene, isopropanol and acetic acid, indicating the important role of the structural composition in generating different chemical sensitivities and fingerprints for sensing. Overall, we clearly disclose the potential of four new anthanthrone-based polymers in various types of electronic devices.

The concentration, nature, and persistence of particulate matter (PM)-bound reactive oxygen species (ROS) are of significant interest in understanding how atmospheric pollution affects health. However, the inherent difficulties in their measurement, particularly regarding the so-called "short-lived" ROS, have limited our understanding of their persistence and concentrations in the atmosphere. This paper aims to address this limitation through the analysis of PM-bound ROS measurements from the Particle Into Nitroxide Quencher (PINQ) system at an atmospheric monitoring site in the city of Heshan, Guangdong Province, China. The measured daily average and standard deviation for the measurement period was 0.050 ± 0.017 nmol·m-3. The averaged measured concentration of ROS per mass of PM and standard deviation was 0.0012 ± nmol·mg. The dataset was also correlated with standard pollutants, and a simplified model was constructed to separate the contributions of short-lived (t1/2 = 5 min) and long-lived (t1/2 ∼ infinity) ROS to total concentration using ozone, carbon monoxide, and PM mass. This showed that the short-lived ROS contribute an average of 33% of the daily PM-bound ROS burden over the measurement period, up to 52% of daily average on elevated days, and up to 71% for hourly averages. These results highlight the need for accurate measurements of short-lived ROS and provide the starting point for a general model to predict PM-bound ROS concentrations using widely available standard pollutants for future epidemiological research.

Diketopyrrolopyrrole (DPP) has been one of the most promising building blocks for constructing organic semiconducting materials used in organic field-effect transistors (OFET). As we have known, extended π-conjugated structure can facilitate intermolecular orbital coupling which determines the magnitude of the charge transfer integral and thus the charge transport in devices. What is more, better intramolecular charge transport and intermolecular charge hopping depend on more superior morphology of the conjugated materials in thin film that is influenced by flexible substitutions. Based on the above principles, it is worthy designing and synthesizing fused ring flanked DPP monomers with alkyl chains to exploit their potential use in OFET. In this work, we first synthesized and characterized a new DPP derivative, naphthalene flanked DPP with butyl-octyl side chain (BO-DPPN). When comparing with its thiophene- and furan-flanked DPP analogues using the same common alkyl chain, BO-DPPN appears as most suitable for OFET applications. For example, DSC analysis demonstrates its high degree of crystallinity, UV–Vis spectra show a strong aggregation in solid state, XRD pattern indicates its tight and long-range ordered lamellar packing. As expected, the OFET devices function well when using BO-DPPN directly as active semiconductor and an impressive hole mobility of 0.0126 cm2 V−1 s−1 was observed with a bottom-contact/top-gate architecture using a solution processable approach indicating its promising potential for use in organic electronics.

Blue-color-emitting organic semiconductors are of significance for organic light-emitting diodes (OLEDs). In this study, through Suzuki coupling polymerization, three 1,4-naphthalene-based copolymers—namely, PNP(1,4)-PT, PNP(1,4)-TF, and PNP(1,4)-ANT—were designed and synthesized. The variation of comonomers, phenothiazine (PT), triphenylamine substituted fluorene (TF), and anthanthrene (ANT), effectively tuned the emitting color and device performance of poly(9-vinyl carbazole) (PVK)-based OLEDs. Especially, the polymer PNP(1,4)-TF, bearing perpendicular aryl side groups, showed a most twisted structural geometry, which enabled an ultra-high thermal stability and a best performance with blue emitting in PVK-host-based OLEDs. Overall, in this work, we demonstrate a promising blue-color-emitting polymer through structural geometry manipulation.

To identify the specific activity sites for both the HER and OER in experimental realized single transition-metal atom decorated graphene sheets, we assume the number of metal–C bonds (coordination) determines the adsorption strength of reaction intermediates on the metal atom sites. We find the strength weakens with the metal coordination number. Low-coordinated Ni sites, i.e. single-coordinated Ni on the zigzag edge, exhibit the highest activity toward HER, while high-coordinated Ni atoms, i.e. quadruple-coordinated Ni, demonstrate the best OER performance. This assumption has been further confirmed by a wide range of transition metals, including Mn, Fe, Co, Cu, and Pd. Our findings highlight a new family of efficient bifunctional catalysts for water splitting and offer a new paradigm for the design of single-atom HER/OER catalysts toward hydrogen production.

The service lifetime of polymer films is controlled by the chemical reactions leading to chain scission and the mediating environmental factors. For application as agricultural cropping film, controlled accelerated degradation is required. For a photo-sensitive linear low density polyethylene (LLDPE) + 1% nano-titania (as the anatase/rutile mixed phase P25), the environmental factors are not only UV dose and temperature but also soil parameters such as moisture and organic material content. This provides a challenge in predicting the useful lifetime from laboratory accelerated ageing studies. To enhance degradation when the (LLDPE + 1% P25) is buried, UV-C pre-irradiation has been shown to accelerate strength loss but the rate of embrittlement is not sufficient for the application as crop propagation film. Biodegradable poly(butylene adipate-co-terephthalate) or PBAT has a higher rate of degradation when buried outdoors in soil than when buried under laboratory conditions: The elongation at break fell from 900% to 70% in one month in the field while similar changes required 6 months in the laboratory. The small changes in M n ¯ for embrittlement in the field suggests that the loss of mechanical properties was not linked to bulk property changes but rather to surface morphology (cracks and holes) as seen by SEM. This suggests that even in thin films, enzyme-mediated hydrolysis of PBAT is surface controlled. DNA analysis of the soil around the buried films after 35 days ageing outdoors showed fungi play a more dominant role in PBAT biodegradation compared to bacteria. UV degradation of PBAT film is controlled by the photochemistry of the terephthalate moiety in the polymer and the development of fluorescence is a useful indicator of the extent of photo-degradation.

Selective oxidation of 5-hydroxymethylfurfural (HMF) to 2,5-furandicarboxylic acid (FDCA) is one of the key reactions for producing chemical commodities from biomass and their derivatives. The challenge for this reaction is to develop an efficient catalytic process that can be conducted under mild conditions (room temperature and atmospheric pressure, using oxygen molecules in air as the oxidant) and a recyclable catalyst. Herein we report a photocatalyst of cobalt thioporphyrazine (CoPz) dispersed on g-C3N4 (abbreviated as CoPz/g-C3N4), which exhibits excellent catalytic activity toward the selective oxidation of HMF into FDCA under simulated sunlight using oxygen molecules in air as a benign oxidant. For example, an FDCA yield of 96.1% in an aqueous solution at pH = 9.18 is achieved at ambient temperature and air pressure. At lower pH (4.01), the product generated is 2,5-diformylfuran. The determination of the safe working life of polymer materials is important for their successful use in engineering, medicine and consumer-goods applications. An understanding of the physical and chemical changes to the structure of widely-used polymers such as the polyolefins, when exposed to aggressive environments, has provided a framework for controlling their ultimate service lifetime by either stabilising the polymer or chemically accelerating the degradation reactions. The recent focus on biodegradable polymers as replacements for more bio-inert materials such as the polyolefins in areas as diverse as packaging and as scaffolds for tissue engineering has highlighted the need for a review of the approaches to being able to predict the lifetime of these materials. In many studies the focus has not been on the embrittlement and fracture of the material (as it would be for a polyolefin) but rather the products of degradation, their toxicity and ultimate fate when in the environment, which may be the human body. These differences are primarily due to time-scale. Different approaches to the problem have arisen in biomedicine, such as the kinetic control of drug delivery by the bio-erosion of polymers, but the similarities in mechanism provide real prospects for the prediction of the safe service lifetime of a biodegradable polymer as a structural material. Common mechanistic themes that emerge include the diffusion-controlled process of water sorption and conditions for surface versus bulk degradation, the role of hydrolysis versus oxidative degradation in controlling the rate of polymer chain scission and strength loss and the specificity of enzyme-mediated reactions.

A range of profluorescent nitroxides (PFNs) were tested as probes to monitor photo-induced radical-mediated damage in polymer materials. The most stable and sensitive probe of the PFNs tested was an alkyne-linked perylenediimide PFN, 6b, with napthalimide and 9,10-bis(phenylethnyl)anthranene-based versions giving lower stability and sensitivity. Results from photo-ageing of poly(1-trimethylsilyl)-1-propyne (PTMSP) and the ethylene norbornene copolymer (TOPAS®) films doped with PFN probes demonstrated that sensors employing these support materials deliver significantly enhanced sensitivity compared to traditional techniques used to monitor photo-oxidative degradation of polymers, such as infrared spectroscopy. This enhanced sensitivity for detecting polymer damage improved methods for the determination of the serviceable application lifetime of polymers.

To estimate the oxidative potential (OP) of particulate matter (PM), two commonly used cell-free, molecular probes were applied: dithiothreitol (DTT) and dichloro-dihydro-fluorescein diacetate (DCFH-DA), and their performance was compared with 9,10-bis (phenylethynyl) anthracene-nitroxide (BPEAnit). To the best of our knowledge, this is the first study in which the performance of the DTT and DCFH has been compared with the BPEAnit probe. The average concentrations of PM, organic carbon (OC) and elemental carbon (EC) for fine (PM2.5) and coarse (PM10) particles were determined. The results were 44.8 ± 13.7, 9.8 ± 5.1 and 9.3 ± 4.8 µg·m−3 for PM2.5 and 75.5 ± 25.1, 16.3 ± 8.7 and 11.8 ± 5.3 µg·m−3 for PM10, respectively, for PM, OC and EC. The water-soluble organic carbon (WSOC) fraction accounted for 42 ± 14% and 28 ± 9% of organic carbon in PM2.5 and PM10, respectively. The average volume normalized OP values for the three assays depended on both the sampling periods and the PM fractions. The OPBPEAnit had its peak at 2 p.m.; in the afternoon, it was three times higher compared to the morning and late afternoon values. The DCFH and BPEAnit results were correlated (r = 0.64), while there was no good agreement between the BPEAnit and the DTT (r = 0.14). The total organic content of PM does not necessarily represent oxidative capacity and it shows varying correlation with the OP. With respect to the two PM fractions studied, the OP was mostly associated with smaller particles.

BODIPY is a highly versatile fluorophore for biological imaging with a tunable fluorescence emission (500 - 800 nm) that overlaps with the optically transparent window for tissue (600 - 1300 nm). Herein, we describe the synthesis of optically distinct BODIPY-based profluorescent probes bearing meso- and β-substituted isoindoline nitroxides and their corresponding methoxyamine derivatives. These profluorescent nitroxide probes possess strongly suppressed fluorescence, which can be revealed upon reduction or reaction with other radicals. Examination of the pentafluorophenylhydrazine reduction of the prepared probes using tandem EPR and fluorescence spectroscopy demonstrated that the asymmetric bis-β-substituted probe 9 (λem = 603 nm) was reduced the fastest, however, the greatest difference in fluorescence emission between the nitroxide and its reduced hydroxylamine analogue was observed for probe 7 (λem = 570 nm). The significant difference in fluorescence output between the nitroxides and their corresponding diamagnetic derivatives makes these probes ideal tools for imaging reactive oxygen species in biological systems.

Two-dimensional (2D) materials are promising for use in lithium (Li) electrodes due to their high surface ratio. By using density functional theory (DFT) calculations, we investigate the adsorption and diffusion of Li on a newly predicted 2D GeP3 material [Nano Lett., 2016, 17, 1833]. The most favourable adsorption sites for Li are identified, and a semiconducting to metallic transition induced by Li adsorption is found, which indicates excellent electrical conductivity. The GeP3 monolayer has an estimated capacity of 648 mA h g−1, which is almost twice that of commercially used graphite (375 mA h g−1). During full Li intercalation, the GeP3 layer undergoes only 1.2% lattice parameter reduction. Moreover, GeP3 possesses the advantages of a small diffusion barrier (∼0.5 eV) and low average open-circuit voltages (∼0.4 V). Our results highlight a new class of promising anode materials, i.e. 2D phosphide, as potential rechargeable lithium batteries with ultrahigh-capacity, superior ionic conductivity, and low average open-circuit voltage.

Profluorescent nitroxides (PFNs) have emerged as an important class of imaging agents for monitoring intracellular redox status and levels of oxidative stress. However, the fast reduction of nitroxides upon incubation within cells limits the window of opportunity for detection. By increasing the concentration of nitroxides per fluorophore, their reduction to the corresponding hydroxylamines and the subsequent switch-on of fluorescence can be delayed. Herein the preparation of nitroxide-containing polymers of different chain length coupled to a fluorophore is reported and their reduction with pentafluorophenylhydrazine is examined. The fluorescence switch-on kinetics and radical concentrations are monitored by fluorescence and electron paramagnetic resonance spectroscopy and compared to a conventional PFN bearing a single nitroxide moiety. The polymeric PFNs display significant delays in reduction and fluorescence switch-on and higher turn-on ratios than their single-nitroxide counterparts. The results of this study indicate that polymeric PFNs are a promising architecture for future imaging agents.

Here we report a class of photocatalysts: mixed-valence vanadium oxide particles grafted onto a variety of oxide supports. In these catalysts V6O13 species with mixed oxidation states (V4+ or V5+) are believed to be catalytically active sites. These catalysts successfully enable alcohol oxidation to selectively produce aldehydes and ketones using O2 as the oxidant. The catalytic process is driven by visible light irradiation at room temperature and, most importantly, progresses with negligible overoxidation. The catalysts can even selectively oxidize aliphatic alcohols, which are much more challenging to control in comparison to aromatic analogues. They can also be applied to the activation and oxidation of the otherwise stable C–H bonds of saturated aromatic hydrocarbons, such as toluene and xylene, under irradiation. The utility of profluorescent nitroxides (PFNs) as sensitive probes to detect early stage photodegradation in a cyclic olefin copolymer, TOPAS®, during both natural aging and accelerated aging under laboratory conditions is reported. PFN additives in TOPAS® capture radicals to form fluorescent adducts as the material degrades. The levels of fluorescence detectable from the polymer reflect the degree of free-radical degradation in the material, with the PFN probes delivering enhanced sensitivity over traditional analytical methods for the detection of photodegradation of TOPAS®. The probes are able to highlight polymer degradation occurring within the oxidation “induction” period, where little change can be observed using infrared spectroscopy; however, their efficacy does not extend far beyond this period. The effective probe lifetime however can be significantly extended through the use of common additives such as the UV absorber (Tinuvin P) and a hindered amine stabiliser analogue (1,1,3,3-tetramethylisoindol-2-yloxyl, TMIO).

Triethylene glycol, a common side chain, is attached to two different dye moieties, diketopyrrolopyrrole (DPP) and isoindigo (II), and their bromo derivative monomers are copolymerized, respectively, with common bisstannyl alkylated bithiophene via Stille coupling. The resulting donor–acceptor low-bandgap copolymers, namely, PTDPP-DT and PTII-DT, are rationally deigned and synthesized conjugated polymeric systems suitable for doping. Both polymers are successfully investigated as single-component and composite-system-based electrochemical transistors and light-emitting electrochemical cells. A PTDPP-DT thin film exhibits relatively high electrical conductivity of up to 80 S cm−1 in the electrochemically doped state, whereas PTII-DT thin film prevents the macroscopic charge transport due to a large-scale crystalline disorientation. Upon evaluating both polymers as active conjugated materials in light-emitting electrochemical cells, they both exhibit emission under efficient electron/hole doping conditions.

The addition of an N–H bond to C C triple bonds in the hydroamination of alkynes is of great importance in synthetic chemistry. We found that visible light irradiation can efficiently drive the direct hydroamination of alkynes using supported gold nanoparticles (AuNPs) due to its Localized Surface Plasmon Resonance (LSPR). Au NPs on nitrogen-doped TiO2 could successfully induce hydroamination with a very high conversion (90%) and excellent product selectivity (91% imine product). Herein, we demonstrate that for such a photocatalytic process, the intensity and wavelength of the incident light, as well as the reaction temperature have a significant impact on the performance of the catalyst. The photocatalytic activity increases with increasing light intensity and reaction temperature. This means that the AuNP photocatalysts couple both the input light energy and thermal energy to catalyze the reaction. The influence of AuNPs and the surface properties of the support materials on the photocatalytic performance are also further discussed

Nitroxides have been exploited as profluorescent probes for the detection of oxidative stress. In addition, they deliver potent antioxidant action and attenuate reactive oxygen species (ROS) in various models of oxidative stress, with these results ascribed to superoxide dismutase or redox and radical-scavenging actions. Our laboratory has developed a range of novel, biostable, isoindoline nitroxide-based antioxidants, DCTEIO and CTMIO. In this study we compared the efficiency of these novel compounds as antioxidant therapies in reducing ROS both in vivo (rat model) and in vitro (661W photoreceptor cells), with the established antioxidant resveratrol. By assessing changes in fluorescence intensity of a unique redox-responsive probe in the rat retina in vivo, we evaluated the ability of antioxidant therapy to (1) ameliorate ROS production and (2) reverse the accumulation of ROS after complete, acute ischemia followed by reperfusion (I/R). I/R injury induced a marked decrease in fluorescence intensity over 60 min of reperfusion, which was successfully ameliorated with each of the antioxidants. DCTEIO and CTMIO reversed the accumulation of ROS when administered intraocularly post ischemic insult, whereas, the effect of resveratrol was not significant. We also investigated our novel agents' capacity to prevent ROS-mediated metabolic dysfunction in the 661W photoreceptor cell line. Cellular stress induced by the oxidant, tert-butyl hydroperoxide, resulted in a loss of spare mitochondrial respiratory capacity (SMRC) and in the extracellular acidification rate in 661W cells. DCTEIO antioxidant administration successfully reduced the loss of SMRC. Together, these findings show we can quantify dynamic changes in cellular oxidative status in vivo and suggest that nitroxide-based antioxidants may provide greater protection against oxidative stress than the current state-of-the-art antioxidant treatments for ROS-mediated diseases.

Novel profluorescent mono- and bis-isoindoline nitroxides linked to napthalimide and perylene diimide structural cores are described. These nitroxide-fluorophore probes display strongly suppressed fluorescence in comparison to their corresponding non-radical diamagnetic methoxyamine derivatives. The perylene-based probe possessing two isoindoline systems tethered through ethynyl linkages was shown to be the most photostable in solution, demonstrating significantly enhanced longevity over the 9,10-bis(phenylethynyl)anthracene fluorophore used in previous profluorescent nitroxide probes.

The impact of commonly used agrichemicals (Paraquat, Mancozeb, Chlorpyrifos and Sulfur) on the environmental stability of polyethylene films has been evaluated under accelerated photo-oxidative conditions. Paraquat and Mancozeb when applied to unstabilized clear polyethylene film, followed by exposure to UV radiation, resulted in direct attack of the polymer, evident by faster rates of oxidation and shorter times to embrittlement compared to a polyethylene control. Reapplication of these chemicals during exposure, as often occurs in agricultural practice, resulted in even faster degradation. In contrast, Sulfur and Chlorpyrifos did not have a significant effect on the photodegradation of exposed unstabilized polyethylene film. When the polyethylene film was pigmented (white top layer containing rutile titanium dioxide and carbon black in the bottom layer) and stabilized with the addition of hindered amine stabilizers (HAS), differences in oxidation susceptibility were seen with repeated exposure to Paraquat showing the most dramatic effect. This was evident by an increased carbonyl index, decrease in elongation at break and an increase in tensile stress at yield compared to a control film and other agrichemical treatments. These studies enable degradation from accelerated consumption of stabilizers by the agrichemicals (and their degradation products) to be differentiated from enhanced photochemical initiation of the polyethylene degradation.

In the prospect of limited energy resources and climate change, effects of alternative biofuels on primary emissions are being extensively studied. Our two recent studies have shown that biodiesel fuel composition has a significant impact on primary particulate matter emissions. It was also shown that particulate matter caused by biodiesels was substantially different from the emissions due to petroleum diesel. Emissions appeared to have higher oxidative potential with the increase in oxygen content and decrease of carbon chain length and unsaturation levels of fuel molecules. Overall, both studies concluded that chemical composition of biodiesel is more important than its physical properties in controlling exhaust particle emissions. This suggests that the atmospheric aging processes, including secondary organic aerosol formation, of emissions from different fuels will be different as well. In this study, measurements were conducted on a modern common-rail diesel engine. To get more information on realistic properties of tested biodiesel particulate matter once they are released into the atmosphere, particulate matter was exposed to atmospheric oxidants, ozone and ultra-violet light; and the change in their properties was monitored for different biodiesel blends. Upon the exposure to oxidative agents, the chemical composition of the exhaust changes. It triggers the cascade of photochemical reactions resulting in the partitioning of semi-volatile compounds between the gas and particulate phase. In most of the cases, aging lead to the increase in volatility and oxidative potential, and the increment of change was mainly dependent on the chemical composition of fuels as the leading cause for the amount and the type of semi-volatile compounds present in the exhaust.

The current study introduces a novel synthetic avenue for the preparation of profluorescent nitroxides via nitrile imine-mediated tetrazole-ene cycloaddition (NITEC). The photoinduced cycloaddition was performed under metal-free, mild conditions allowing the preparation of a library of the nitroxide functionalized pyrazolines and corresponding methoxyamines. High reaction rates and full conversion were observed, with the presence of the nitroxide having no significant impact on the cycloaddition performance. The formed products were investigated with respect to their photophysical properties in order to quantify their "switch on/off" behavior. The fluorescence quenching performance is strongly dependent on the distance between the chromophore and the free radical spin as demonstrated theoretically and experimentally. Highest levels of fluorescence quenching were achieved for pyrazolines with the nitroxide directly fused to the chromophore. Importantly, the pyrazoline profluorescent nitroxides were shown to efficiently act as sensors for redox/radical processes.

Organic electrode materials are a highly promising and environmentally benign class of battery materials with radical polymers being at the forefront of this research. Herein, we report the first example of the 1,1,3,3-tetramethylisoindolin-2-yloxyl class of nitroxides as an organic electrode material and the synthesis and application of a novel styrenic nitroxide polymer, poly(5-vinyl-1,1,3,3-tetramethylisoindolin-2-yloxyl) (PVTMIO). The polymer was synthesized from the precursor monomer, 2-methoxy-5-vinyl-1,1,3,3-tetramethylisoindoline, and subsequent oxidative deprotection yielded the electroactive radical species. Cyclic voltammetry revealed a high oxidation potential of 3.7 V versus Li, placing it among the top of the nitroxide class of electrode materials. The suitability of PVTMIO for utilization in a high-voltage organic radical battery was confirmed with a discharge capacity of 104.7 mAh g–1, high rate performance, and stability under cycling conditions (90% capacity retention after 100 cycles), ...

Diketopyrrolopyrrole (DPP) has been drawing considerable attention for constructing semiconducting materials used in organic optoelectronic applications, mainly for organic field effect transistor (OFET) and organic photovoltaic (OPV) devices. In the present work, we study the effects of varying the number of thiophene units (from four to six) attached to DPP on the physical, chemical, and optoelectronic properties by designing and synthesizing a series of small molecule organic semiconductors. The thermal and optical properties, and electronic energy levels of these molecular semiconductors are studied, and their performance in organic field effect transistor devices (OFETs) compared. These small molecules exhibit promising charge carrier mobility and behave as p-type semiconductors. Hole mobility increases with conjugation length and degree of symmetry of the backbone. By adjusting the number of thiophene units on each side of DPP, the hole mobility is enhanced by almost one order of magnitude, from 1.18 × 10-4 to 1.11 × 10-3 cm2 V-1 s-1. Density functional theory (DFT) calculations indicate that increasing the number of homo-coupled thiophene results in a relatively planar configuration, while the terminal alkoxyl benzene unit causes significant torsional rotation which could hamper electron and hole transport in active layer. Three fullerene isoindoline nitroxides N-methyl-3,4-fulleropyrrolidine-2-spiro-5′- (1′,1′,3′,3′-tetramethylisoindolin-2′-yloxyl), (C60-(TMIO)m, and C70-(TMIO)n) were synthesized by the covalent bonding of 5-formyl-1,1,3,3-tetramethyl isoindolin-2-yloxyl to the fullerenes C60 and C70. Significantly, the X-ray photoelectron spectra indicated the characteristic N 1s signals of NO. at 402 eV. The atomic force microscope morphologies showed that the average particle sizes of C60-(TMIO)m and C70-(TMIO)n were 38 and 15 nm. The electrochemical experiments indicated that fullerene bound isoindoline nitroxides retained similar electrochemical properties and redox reaction mechanisms as the parent nitroxides. The electron paramagnetic resonance spectra of the fullerene isoindoline nitroxides all exhibited the hyperfine splittings and characteristic spectra of tetramethyl isoindoline nitroxides, with typical nitroxide g-values and nitrogen isotropic hyperfine coupling constants. A simple modular strategy for the synthesis of profluorescent nitroxide containing polymers is described. The incorporation of an epoxide as a pendant functionality on a polymer backbone synthesized using ATRP and subsequent nucleophilic ring-opening with sodium azide gave hydroxyl and azide functionality within a 3-bond radius. Orthogonal coupling chemistry then allowed the independent attachment of fluorophore and nitroxide groups in close proximity, giving rise to a profluorescent polymer. Validation of the viability of these materials as fluorescent sensors is demonstrated through efficient fluorescence switch-on observed when the materials are exposed to a model reductant or carbon-centred radical source.

Dual-acting hybrid anti-oxidant/anti-inflammatory agents were developed employing the principle of pharmacophore hybridization. Hybrid agents were synthesized by combining stable anti-oxidant nitroxides with conventional non-steroidal anti-inflammatory drugs (NSAIDs). Several of the hybrid nitroxide-NSAID conjugates displayed promising anti-oxidant and anti-inflammatory effects on two Non-Small Cell Lung Cancer (NSCLC) cells (A549 and NCI-H1299) and in ameliorating oxidative stress induced in 661 W retinal cells. One ester-linked nitroxide-aspirin analogue (27) delivered better anti-inflammatory effects (cyclooxygenase inhibition) than the parent compound (aspirin), and also showed similar reactive oxygen scavenging activity to the anti-oxidant, Tempol. In addition, a nitroxide linked to the anti-inflammatory drug indomethacin (39) significantly ameliorated the effects of oxidative stress on 661 W retinal neurons at efficacies greater or equal to the anti-oxidant Lutein. Other examples of the hybrid conjugates displayed promising anti-cancer activity, as demonstrated by their inhibitory effects on the proliferation of A549 NSCLC cells.

Changes to the redox status of biological systems have been implicated in the pathogenesis of a wide variety of disorders including cancer, Ischemia-reperfusion (I/R) injury and neurodegeneration. In times of metabolic stress e.g. ischaemia/reperfusion, reactive oxygen species (ROS) production overwhelms the intrinsic antioxidant capacity of the cell, damaging vital cellular components. The ability to quantify ROS changes in vivo, is therefore essential to understanding their biological role. Here we evaluate the suitability of a novel reversible profluorescent probe containing a redox-sensitive nitroxide moiety (methyl ester tetraethylrhodamine nitroxide, ME-TRN), as an in vivo, real-time reporter of retinal oxidative status. The reversible nature of the probe's response offers the unique advantage of being able to monitor redox changes in both oxidizing and reducing directions in real time. After intravitreal administration of the ME-TRN probe, we induced ROS production in rat retina using an established model of complete, acute retinal ischaemia followed by reperfusion. After restoration of blood flow, retinas were imaged using a Micron III rodent fundus fluorescence imaging system, to quantify the redox-response of the probe. Fluorescent intensity declined during the first 60 min of reperfusion. The ROS-induced change in probe fluorescence was ameliorated with the retinal antioxidant, lutein. Fluorescence intensity in non-Ischemia eyes did not change significantly. This new probe and imaging technology provide a reversible and real-time response to oxidative changes and may allow the in vivo testing of antioxidant therapies of potential benefit to a range of diseases linked to oxidative stress.

The powerful oxidant HOCl (hypochlorous acid and its corresponding anion, (-)OCl) generated by the myeloperoxidase (MPO)-H2O2-Cl(-) system of activated leukocytes is strongly associated with multiple human inflammatory diseases; consequently there is considerable interest in inhibition of this enzyme. Nitroxides are established antioxidants of low toxicity that can attenuate oxidation in animal models, with this ascribed to superoxide dismutase or radical-scavenging activities. We have shown (M.D. Rees et al., Biochem. J. 421, 79-86, 2009) that nitroxides, including 4-amino-TEMPO (4-amino-2,2,6,6-tetramethylpiperidin-1-yloxyl radical), are potent inhibitors of HOCl formation by isolated MPO and activated neutrophils, with IC50 values of ~1 and ~6 µM respectively. The utility of tetramethyl-substituted nitroxides is, however, limited by their rapid reduction by biological reductants. The corresponding tetraethyl-substituted nitroxides have, however, been reported to be less susceptible to reduction. In this study we show that the tetraethyl species were reduced less rapidly than the tetramethyl species by both human plasma (89-99% decreased rate of reduction) and activated human neutrophils (62-75% decreased rate). The tetraethyl-substituted nitroxides retained their ability to inhibit HOCl production by MPO and activated neutrophils with IC50 values in the low-micromolar range; in some cases inhibition was enhanced compared to tetramethyl substitution. Nitroxides with rigid structures (fused oxaspiro rings) were, however, inactive. Overall, these data indicate that tetraethyl-substituted nitroxides are potent inhibitors of oxidant formation by MPO, with longer plasma and cellular half-lives compared to the tetramethyl species, potentially allowing lower doses to be employed.

Air pollution exposure is associated with a range of adverse health effects, including cardiovascular and respiratory diseases. Particle-bound ROS has been recognised as one of the prevailing parameters to indicate the toxic potential of airborne particulate matter (PM). The temporal variability of particle-bound ROS is a very important metric crucial for the improvement of public health and risk assessment policies. To our knowledge this is the first study aiming to investigate diurnal ROS profiles in both the particle and gas phase and associate them with diurnal variations of important pollutants. For that purpose, we have successfully applied a new instrument to continuously monitor diurnal ROS profiles at two locations in Hong Kong: a busy roadside and an urban background. Data was collected over both the working week and during weekends. We have observed a high correlation between particle-bound ROS and lower concentrations of black carbon (BC) at the roadside during the working week. These associations were less significant over the weekend and at all times with ozone. Our results suggest that most of the particle-bound ROS from both the particle and the gas phase arises from fresh emission sources directly from the traffic. A very interesting observation came out as a result of this study where measured ROS concentration was decreasing with the rise of ozone in conjunction with particle number, suggesting potential role of ROS in particle growth and aging.

Purpose: To evaluate the utility of a novel profluorescent nitroxide (PFN) probe that selectively detects superoxide radicals in live cells, as a reporter of retinal oxidative status in models of retinal metabolic challenge.Method: (i) Cultured RGC-5 cells were transformed with trichostatin A to stimulate a “neuronal-like” morphology. Following treatment with 0.01 μM methylester rhodamine nitroxide probe, cells were exposed to the mitochondrial complex III inhibitor, antimycin (0.01−100 μM) to stimulate superoxide production. Fluorescence (556 nm/590 nm) was quantified via flow cytometry with statistical analysis based on 10,000 cells. (ii) Following intraocular injection of the PFN (2 μM), unilateral acute retinal ischaemia was induced in rats by elevation of intraocular pressure (IOP, 120 mmHg 60 min). The robust and diversely useful isoindoline nitroxide, 5-carboxy-1,1,3,3-tetramethylisoindolin-2-yloxyl (1; CTMIO), has previously been synthesised in low-to-moderate yields from phthalic anhydride (3). Recent interest in its biological potential as a potent antioxidant and in other areas has seen an increased demand for its production. Herein, three new synthetic routes to CTMIO are presented and their efficiencies assessed. Two routes, via the nitrile 9 and the formyl compound 11, derive from 5-bromo-1,1,3,3-tetramethylisoindoline (6). The third approach starts from the readily accessible starting material, 4-methylphthalic anhydride (12), and proceeds by a methylarene oxidation with potassium permanganate. The three new approaches yield CTMIO in comparable overall yields (16–18 %); however, the synthetic efficiency is most improved when employing the nitrile intermediate 9.

Cyclic nitroxide radicals represent promising alternatives to the iodine-based redox mediator commonly used in dye-sensitized solar cells (DSSCs). To date DSSCs with nitroxide-based redox mediators have achieved energy conversion efficiencies of just over 5 % but efficiencies of over 15 % might be achievable, given an appropriate mediator. The efficacy of the mediator depends upon two main factors: it must reversibly undergo one-electron oxidation and it must possess an oxidation potential in a range of 0.600-0.850 V (vs. a standard hydrogen electrode (SHE) in acetonitrile at 25 °C). Herein, we have examined the effect that structural modifications have on the value of the oxidation potential of cyclic nitroxides as well as the reversibility of the oxidation process. These included alterations to the N-containing skeleton (pyrrolidine, piperidine, isoindoline, azaphenalene, etc.), as well as the introduction of different substituents (alkyl-, methoxy-, amino-, carboxy-, etc.) to the ring. Standard oxidation potentials were calculated using high-level ab initio methodology that was demonstrated to be very accurate (with a mean absolute deviation from experimental values of only 16 mV). An optimal value of 1.45 for the electrostatic scaling factor for UAKS radii in acetonitrile solution was obtained. Established trends in the values of oxidation potentials were used to guide molecular design of stable nitroxides with desired E(ox)°, and a number of compounds were suggested for potential use as enhanced redox mediators in DSSCs.

A range of varying chromophore nitroxide free radicals and their nonradical methoxyamine analogues were synthesized and their linear photophysical properties examined. The presence of the proximate free radical masks the chromophore's usual fluorescence emission, and these species are described as profluorescent. Two nitroxides incorporating anthracene and fluorescein chromophores (compounds 7 and 19, respectively) exhibited two-photon absorption (2PA) cross sections of approximately 400 G.M. when excited at wavelengths greater than 800 nm. Both of these profluorescent nitroxides demonstrated low cytotoxicity toward Chinese hamster ovary (CHO) cells. Imaging colocalization experiments with the commercially available CellROX Deep Red oxidative stress monitor demonstrated good cellular uptake of the nitroxide probes. Sensitivity of the nitroxide probes to H(2)O(2)-induced damage was also demonstrated by both one- and two-photon fluorescence microscopy. These profluorescent nitroxide probes are potentially powerful tools for imaging oxidative stress in biological systems, and they essentially "light up" in the presence of certain species generated from oxidative stress. The high ratio of the fluorescence quantum yield between the profluorescent nitroxide species and their nonradical adducts provides the sensitivity required for measuring a range of cellular redox environments. Furthermore, their reasonable 2PA cross sections provide for the option of using two-photon fluorescence microscopy, which circumvents commonly encountered disadvantages associated with one-photon imaging such as photobleaching and poor tissue penetration.

A series of new spin-labeled porphyrin containing isoindoline nitroxide moieties were synthesized and characterized as potential free radical fluorescence sensors. Fluorescence-suppression was observed in the free-base monoradical porphyrins, whilst the free-base biradical porphyrins exhibited highly suppressed fluorescence about three times greater than the monoradical porphyrins. The observed fluorescence-suppression was attributed to enhanced intersystem crossing resulting from electronexchange between the doublet nitroxide and the excited porphyrin fluorophore. Notably, fluorescencesuppression was not as strong in the related metalated porphyrins, possibly due to insufficient spin coupling between the nitroxide and the porphyrin. Continuous wave EPR spectroscopy of the diradical porphyrins in fluid solution suggests that the nitroxyl-nitroxyl interspin distance is long enough and tumbling is fast enough not to detect dipolar coupling.

Degradation of polypropylene (PP) during melt processing was studied using a novel profluorescence technique. The profluorescent nitroxide probe, 1,1,3,3-tetramethyldibenzo[ e,g ]isoindolin-2-yloxyl (TMDBIO) was added to PP during melt processing to act as a sensor for carbon-centred radicals. Trapping of carbon-centred radicals, formed during degradation of PP, led to an increase in fluorescence emission from TMDBIO adducts. Through analysis of viscosity changes during processing cumulative chain scission degradation was estimated. At processing temperatures of 210 °C or below, fluorescence emission from TMDBIO adducts could be correlated with cumulative chain scissions when the number of chain scissions was small. At higher temperatures, a correlation was not observed most probably due to radical-trap instability rather than decomposition of the TMDBIO. Thus, TMDBIO may be used as a profluorescent sensor for degradation of PP during melt processing when the processing temperature is 210 °C or below.

The profluorescent nitroxide, 1,1,3,3-tetramethyldibenzo[e,g]isoindolin-2- yloxyl (TMDBIO) was investigated as a probe for the radical-mediated degradation of stabilised polypropylene. TMDBIO has been previously shown to be a sensitive probe for free-radical degradation during the thermo-oxidation of unstabilised polypropylene. Here we report on the effect that adding hindered phenol or phosphate stabilisers to polypropylene has on the free-radical sensing ability of TMDBIO during thermo-oxidation. In addition, novel dual-functional, hindered phenol containing profluorescent nitroxides, 5-[2-(4-hydroxy-3,5-di-tert- butylphenyl)ethenyl]-1,1,3,3-tetramethylisoindolin-2-yloxyl (HSTMIO) and its derivatives were investigated as probes for the radical-mediated degradation of polypropylene. These dual-functional probes were shown to be efficient stabilisers for polypropylene during thermo-oxidation at 150 °C in oxygen and sensors of thermo-oxidation during its early stages, in the so-called "induction period".

Tissue damage resulting from the extracellular production of HOCl (hypochlorous acid) by the MPO (myeloperoxidase)-hydrogen peroxide-chloride system of activated phagocytes is implicated as a key event in the progression of a number of human inflammatory diseases. Consequently, there is considerable interest in the development of therapeutically useful MPO inhibitors. Nitroxides are well established antioxidant compounds of low toxicity that can attenuate oxidative damage in animal models of inflammatory disease. They are believed to exert protective effects principally by acting as superoxide dismutase mimetics or radical scavengers. However, we show here that nitroxides can also potently inhibit MPO-mediated HOCl production, with the nitroxide 4-aminoTEMPO inhibiting HOCl production by MPO and by neutrophils with IC50 values of approx. 1 and 6 μM respectively. Structure–activity relationships were determined for a range of aliphatic and aromatic nitroxides, and inhibition of oxidative damage to two biologically-important protein targets (albumin and perlecan) are demonstrated. Inhibition was shown to involve one-electron oxidation of the nitroxides by the compound I form of MPO and accumulation of compound II. Haem destruction was also observed with some nitroxides. Inhibition of neutrophil HOCl production by nitroxides was antagonized by neutrophil-derived superoxide, with this attributed to superoxide-mediated reduction of compound II. This effect was marginal with 4-aminoTEMPO, probably due to the efficient superoxide dismutase-mimetic activity of this nitroxide. Overall, these data indicate that nitroxides have considerable promise as therapeutic agents for the inhibition of MPO-mediated damage in inflammatory diseases.

A range of novel tetramethyl- and tetraethylisoindolinenitroxides, possessing aryl-linked carboxylic acids, amines, alcohols and phosphonic acids were prepared. Notably, the chemistry established for the aromatic dibromination of the tetramethylisoindolines was not easily transferred to the corresponding tetraethylisoindoline system. Instead, various tetraethylisoindoline analogues were accessed by the oxidation of methyl groups attached to the aromatic ring to give the carboxylic acids. The increased steric bulk of the tetraethyl structures should limit bio-reduction and these compounds may have potential as antioxidants.

New profluorescent mono- and di-isoindoline nitroxides (5, 11, 16 and 19) containing 9,10-diphenylanthracene and 9,10-bis(phenylethynyl)anthracene structural cores were synthesised by palladium-catalysed Suzuki and Sonogashira couplings. These nitroxide-fluorophore probes possess strongly suppressed fluorescence, even in the presence of only one nitroxide radical. Upon reduction, or reaction with other radicals, normal fluorescence emission is returned. The significant difference in fluorescence output between the nitroxides and their corresponding diamagnetic analogues makes these probes ideal tools for imaging polymer degradation using fluorescence microscopy. Nitric oxide ((*)NO) may act as either a pro-oxidant or an antioxidant in biological systems. Although (*)NO and nitroxide radicals react slowly with most molecules, they react at near diffusion-controlled rates with other radicals and may therefore be efficient protective agents. This study assessed the ability of (*)NO and nitroxides to intercept specific protein-derived radicals and compared the efficacy of these species. Three protein radical systems were investigated as follows: BSA-derived radicals generated via radical transfer from H(2)O(2)-activated horseradish peroxidase, radicals formed on myoglobin via reaction with H(2)O(2), and carbon-centered radicals formed from amino acid hydroperoxides on exposure to Fe(2+)-EDTA. In each case, radicals were generated in the absence or presence of (*)NO or nitroxides of different size and charge. Concentration-dependent loss of the protein radicals was detected by electron paramagnetic resonance with both (*)NO and nitroxides and time-dependent consumption of (*)NO using an (*)NO electrode. The protein oxidation product dityrosine was significantly reduced by (*)NO and nitroxides, and 3,4-dihydroxyphenylalanine levels were reduced by nitroxides but not (*)NO. Overall, these studies demonstrate that (*)NO and nitroxides are efficient near-stoichiometric scavengers of protein radicals and, hence, are potential protective agents against protein oxidation reactions and resulting damage. These reactions show little dependence on nitroxide structure or charge.

In this study, three profluorescent nitroxides 1,1,3,3-tetramethyldibenzo[ e , g ]isoindolin-2-yloxyl (TMDBIO), 1,1,3,3-tetramethyl-2,3-dihydro-2-azaphenalene-2-yloxyl (TMAO) and 5-[2-(4-methoxycarbonyl-phenyl)-ethenyl]1,1,3,3-tetramethylisoindoline-2-yloxyl (MeCSTMIO) were tested as probes for radical-mediated damage in polypropylene arising from both UV and thermally initiated sources. These nitroxides possess a very low fluorescence quantum yield due to quenching by the nitroxide group; however, when the free-radical moiety is removed by reaction with alkyl radicals (to give an alkoxyamine), strong fluorescence is observed. The results obtained from this profluorescent nitroxide trapping technique compare favourably with other methods of monitoring degradation, provided the appropriate probe is chosen for the conditions of oxidation, signalling an indication of damage well before other techniques show any response. The technique was also applied to the monitoring of crosslinked polyester coating resins. Differentiation in the oxidative stability of the resins was evident after as little as 200 min where other monitoring techniques require up to 300 h of accelerated degradation. This highlights the sensitivity of this method as well as demonstrating the scope of this technique to assess polymer stability.

Free-radical processes underpin the thermo-oxidative degradation of polyolefins. Thus, to extend the lifetime of these polymers, stabilizers are generally added during processing to scavenge the free radicals formed as the polymer degrades. Nitroxide radical precursors, such as hindered amine stabilizers (HAS),1,2 are common polypropylene additives as the nitroxide moiety is a potent scavenger of polymer alkyl radicals (R¥). Oxidation of HAS by radicals formed during polypropylene degradation yields nitroxide radicals (RRNO¥), which rapidly trap the polymer degradation species to produce alkoxyamines, thus retarding oxidative polymer degradation. This increase in polymer stability is demonstrated by a lengthening of the “induction period” of the polymer (the time prior to a sharp rise in the oxidation of the polymer). Instrumental techniques such as chemiluminescence or infrared spectroscopy are somewhat limited in detecting changes in the polymer during the initial stages of degradation. Therefore, other methods for observing polymer degradation have been sought as the useful life of a polymer does not extend far beyond its “induction period”

The synthesis of the first example of an azaphenalene-based fused aromatic nitroxide TMAO, [1,1,3,3-tetramethyl-2,3-dihydro-2-azaphenalen-2-yloxyl, (5)], is described. This novel nitroxide possesses some of the structural rigidity of the isoindoline class of nitroxides, as well as some properties akin to TEMPO nitroxides. Additionally, the integral aromatic ring imparts fluorescence that is switched on by radical scavenging reactions of the nitroxide, which makes it a sensitive probe for polymer degradation.(© Wiley-VCH Verlag GmbH & Co. Electron spin lattice relaxation rates were measured for 12 nitroxyls with molecular weights between 144 and 438, and for galvinoxyl, 1,3-bisdiphenylene-2-phenylallyl (BDPA), and 2,2-diphenyl-1-picrylhydrazyl (DPPH) in glassy sucrose octaacetate. Relaxation rates for polar nitroxyls also were measured in glassy sorbitol. Dependence on T where V eff is effective molecular volume and γ is a material-specific parameter, was used to compare processes. Values of V eff were determined based on molecular libration in glassy sucrose octaacetate (γ = 3.5), tumbling in viscous decalin at 233 K (γ = 4.7), or tumbling in heavy mineral oil (γ = 6.0). For nitroxyl relaxation there is a master curve: log(1/T 1) vs log( T) (γ = 0.89). The similarity of the values of γ for the Raman process and for the additional process that contributes at higher temperatures, and the absence of frequency dependence between X- and Q-band, support assignment of this additional process as a local mode. The novel profluorescent nitroxide, 1,1,3,3-tetramethyldibenzo[e,g]isoindolin-2-yloxyl (TMDBIO) was investigated as a probe for the formation of polymer alkyl radicals during the thermo-oxidative degradation of unstabilised polypropylene. TMDBIO possesses a very low fluorescence quantum yield due to quenching by the nitroxide group; however, when the free-radical moiety is removed by reaction with alkyl radicals (to give an alkoxyamine), strong fluorescence is observed. Using spectrofluorimetry, the reaction of the nitroxide with polymer alkyl radicals during oxidation has been monitored. Significantly, the trapping of polymer alkyl radicals during the "induction period" at 120 degrees Celsius is observed, when it is not possible to detect changes in the polymer using either chemiluminescence or infrared spectroscopy. This highlights the sensitivity of this method and represents the direct observation of free-radical generation in polypropylene in the "induction period". TMDBIO also successfully stabilises polypropylene under thermo-oxidative conditions, which is consistent with its action as a radical trap. At elevated temperatures (150 degrees Celsius), at the end of the "induction period" when the polymer is extensively degraded, the fluorescence decreases, due to secondary oxidation of the TMDBIO

A series of dendritic gadolinium complexes based on dendrimers with 1,4,7,10-tetraazacyclododecane as the core were synthesized. Both diethylenetriaminepentaacetic acid and pyridoxamine were incorporated to the amine groups on the surface to give the dendritic ligands. These dendritic ligands were then reacted with gadolinium chloride to yield the corresponding dendritic gadolinium complexes. The dendritic ligands and their gadolinium complexes were characterized by 1HNMR, IR, UV and elemental analysis. Relaxivity studies showed that the dendritic gadolinium complexes have higher relaxation effectiveness than that of the clinically used small molecular gadolinium complexes. Magnetic resonance imaging of the liver in rats indicated that the dendritic gadolinium complex containing pyridoxamine enhances the image contrast of the liver, provides prolonged intravascular duration and produces highly contrasted visualization of blood vessels in the liver.

A series of biodegradable amphiphilic A-B-A type triblock copolymers P(BLA-PEG-BLA), composed of hydrophilic poly(ethylene glycol) (PEG) as a middle block component (B) and hydrophobic poly(β-benzyl-L-aspartate) as outer polypeptide block components (A), were synthesized by copolymerization of β-benzyl-L-aspartate N-carboxy anhydride (BLA-NCA) and the diaminated PEG with the primary amino groups capped at both ends. These P(BLA-PEG-BLA) copolymers were characterized by 1H-NMR, DSC, and GPC. The triblock copolymers were used to prepare three kinds of drug delivery systems including Norfloxacin (INN)-incorporated P(BLA-PEG-BLA) microparticles and tablets. The morphologies of the microparticles were characterized by SEM. The in vitro release properties of the microparticles and tablets in PBS were also evaluated. A mathematical model, which incorporates a linear first-order dissolution term and the transient Fickian diffusion equation, was developed to account for the kinetics of drug release from the INN-incorporated P(BLA-PEG-BLA) microparticles. The methoxyamine group represents an ideal protecting group for the nitroxide moiety. It can be easily and selectively introduced in high yield (typically >90%) to a range of functionalised nitroxides using FeSO4·7H2O and H2O2 in DMSO. Its removal is readily achieved under mild conditions in high yield (70–90%) using mCPBA in a Cope-type elimination process.
