Antineoplastic drugs (ADs) pose risks to healthcare staff. Surface disinfectants are used in hospitals to prevent microbial contamination but the efficiency of disinfectants to degrade ADs is not known. We studied nine disinfectants on ten ADs in the standardized laboratory and realistic in situ hospital conditions. A survey in 43 hospitals prioritized nine most commonly used disinfections based on different ingredients. These were tested on inert stainless steel and in situ on contaminated hospital flooring. The effects against ten ADs were studied by LC–MS/MS (Cyclophosphamide CP; Ifosfamide IF; Capecitabine CAP; Sunitinib SUN; Methotrexate MET; Doxorubicin DOX; Irinotecan IRI; Paclitaxel PX; 5-Fluorouracil FU) and ICP-MS (Pt as a marker of platinum-based ADs). Monitoring of the floor contamination in 26 hospitals showed that the most contaminated are the outpatient clinics that suffer from a large turnover of staff and patients and have limited preventive measures. The most frequent ADs were Pt, PX, FU and CP with maxima exceeding the recommended 1 ng/cm2 limit by up to 140 times. IRI, FU, MET, DOX and SUN were efficiently removed by hydrolysis in clean water and present thus lower occupational risk. Disinfectants based on hydrogen peroxide were efficient against PX and FU (> 70% degradation) but less against other ADs, such as carcinogenic CP or IF, IRI and CAP. The most efficient were the active chlorine and peracetic acid-based products, which however release irritating toxic vapors. The innovative in situ testing of ADs previously accumulated in hospital flooring showed highly problematic removal of carcinogenic CP and showed that alcohol-based disinfectants may mobilize persistent ADs contamination from deeper floor layers. Agents based on hydrogen peroxide, peracetic acid, quaternary ammonium salts, glutaraldehyde, glucoprotamine or detergents can be recommended for daily use for both disinfection and AD decontamination. However, they have variable efficiencies and should be supplemented by periodic use of strong chlorine-based disinfectants efficient also against the carcinogenic and persistent CP.

Coastal areas are final recipients of various contaminants including pesticides. The effects of pesticides on non-target organisms are often unclear, especially at environmentally relevant concentrations. This study investigated the impacts of insecticide imidacloprid (IMI) and fungicide propiconazole (PRO), some of the most detected pesticides in the Arcachon Bay in France. This work also included the research of propiconazole nanoformulation (nanoPRO). The effects were assessed studying the development of the early life stages of the Pacific oyster (Magallana gigas). Oyster embryos were exposed for 24, 30, and 42 h (depending on the endpoint) at 24 degrees C to environmentally relevant concentrations of the two pesticides as well as to nanoPRO. The research focused on sublethal endpoints such as the presence of developmental malformations, alterations of locomotion patterns, or changes in the gene expression levels. No developmental abnormalities were observed after exposure to environmental concentrations detected in the Arcachon Bay in recent years (maximal detected concentration of IMI and PRO were 174 ng/L and 29 ng/L, respectively). EC50 of PRO and nanoPRO were comparable. 2.93 7..t 1.35 and 226 +/- 1.36 mg/L, while EC50 of IMI exceeded 200 mg/L IMI did not affect larval behavior. PRO affected larval movement trajectory and decreased average larvae swimming speed (2 mu g/L), while nanoPRO increased the maximal larvae swimming speed (0.02 mu g/L). PRO upregulated especially genes linked to reactive oxygen species (ROS) production and detoxification. NanoPRO effects on gene expression were less pronounced - half of the genes were altered in comparison with PRO. IMI induced a strong dose-response impact on the genes linked to the detoxification, ROS production, cell cycle, and apoptosis regulation. In conclusion, our results suggest that current pesticide concentrations detected in the Arcachon Bay are safe for the Pacific oyster early development. but they might have a small direct effect via altered gene expressions, whose longer-term impacts cannot be ruled out.

Drinking water treatment ultimately aims to provide safe and harmless drinking water. Therefore, the suitability of a treatment process should not only be assessed based on reducing the concentration os a pollutant concentration but, more importantly, on reducing its toxicity. Hence, the main objective of this study was to answer whether the degradation of a highly toxic compound of global concern for drinking water equals its detoxification. We, therefore, investigated the treatment of cylindrospermopsin (CYN) by center dot OH and SO4-center dot produced in Fenton and Fenton-like reactions. Although SO4-center dot radicals removed the toxin more effectively, both radical species substantially degraded CYN. The underlying degradation mechanisms were similar for both radical species and involved hydroxylation, dehydrogenation, decarboxylation, sulfate group removal, ring cleavage, and further fragmentation. The hydroxymethyl uracil and tricyclic guanidine moieties were the primary targets. Furthermore, the residual toxicity, assessed by a 3-dimensional human in vitro liver model, was substantially reduced during the treatment by both radical species. Although the results indicated that some of the formed degradation products might still be toxic, the overall reduction of the toxicity together with the proposed degradation pathways allowed us to conclude: "Yes, degradation of CYN equals its detoxification!".

Although the production of engineered nanoparticles increases our knowledge of toxicity and mechanisms of bioactivity during relevant exposures is lacking. In the present study mice were exposed to PbO nanoparticles (PbONP; 192.5 mu g/m(3); 1.93 x 10(6) particles/cm(3)) for 2, 5 and 13 weeks through continuous inhalation. The analyses addressed Pb and PbONP distribution in organs (lung, liver, kidney, brain) using electrothermal atomic absorption spectrometry and transmission electron microscopy, as well as histopathology and analyses of oxidative stress biomarkers. New LC-MS/MS methods were validated for biomarkers of lipid damage F2-isoprostanes (8-iso-prostaglandins F2-alpha and E-2) and hydroxylated deoxoguanosine (8-OHdG, marker of DNA oxidation). Commonly studied malondialdehyde was also measured as TBARS by HPLC-DAD. The study revealed fast blood transport and distribution of Pb from the lung to the kidney and liver. A different Pb accumulation trend was observed in the brain, suggesting transfer of NP along the nasal nerve to the olfactory bulbs. Long-term inhalation of PbONP caused lipid peroxidation in animal brains (increased levels of TBARS and both isoprostanes). Membrane lipid damage was also detected in the kidney after shorter exposures, but not in the liver or lung. On the contrary, longer exposures to PbONP increased levels of 8-OHdG in the lung and temporarily increased lung weight after 2 and 5 weeks of exposure. The histopathological changes observed mainly in the lung and liver indicated inflammation and general toxicity responses. The present long-term inhalation study indicates risks of PbONP to both human health and the environment.

Rehabilitation of aged drinking water pipes is an extensive renovation and increasingly topical in many European cities. Spray-on-lining of drinking water pipes is an alternative cost-effective rehabilitation technology in which the insides of pipes are relined with organic polymer. A commonly used polymer is epoxy resin consisting of monomer bisphenol A (BPA). Leaching of BPA from epoxy lining to drinking water has been a concern among public and authorities. Currently epoxy lining is not recommended in some countries. BPA leaching has been demonstrated in laboratory studies but the behavior and ageing process of epoxy lining in situ is not well known. In this study 6 locations with different age epoxy linings of drinking water pipes done using two distinct technologies were studied. While bisphenol F, 4-n-nonylphenol, and 4-t-octylphenol were rarely found and in trace concentrations, BPA was detected in majority of samples. Pipes lined with the older technology (LSE) leached more BPA than those with more recent technology (DonPro): maxima in cold water were 0.25 μg/L and 10 ng/L, respectively. Incubation of water in pipes 8–10 h prior to sampling increased BPA concentration in cold water 1.1–43-fold. Hot water temperature caused even more BPA leaching - at maximum 23.5 μg/L. The influence of ageing of epoxy lining on BPA leaching on could be shown in case of LSE technology: locations with 8–9 years old lining leached 4–20-fold more BPA compared to a location with 2-year-old lining. Analysis of metals showed that epoxy lining can reduce especially iron concentration in water. No significant burden to water could be shown by the analyzed 72 volatile organic compounds, including epichlorhydrin, precursor used in epoxy resin. Estrogenicity was detected in water samples with the highest BPA loads. Comparable responses of two yeast bioreporters (estrogen receptor α and BPA-targeted) indicated that bisphenol-like compounds were the main cause of estrogenicity. Compared to the estimated average daily BPA exposure, additional BPA load via cold drinking water in the studied locations was low, maximum 8.7%. However, hot water should also be considered as exposure source due to higher BPA concentrations. Epoxy lined locations should be monitored in future in order to evaluate ageing process and control increasing leaching of potentially harmful chemicals.

The goal of this study was to contribute to understanding of the mechanisms behind sensitivity differences between early and late instar larvae of Chironomus riparius and to address the influence of the differences in standard testing approaches on the toxicity evaluation. A 10-day contact sediment toxicity test was carried out to assess sensitivity to cadmium exposure in relation to different age and laboratory culture line origin of test organisms. Chironomid larvae of early (OECD 218 method) and late instar (US-EPA600/R-99/064 method) differed substantially in sensitivity of traditional endpoints (OECD: LOEC 50 and 10 μg Cd/g dry weight (dw); US-EPA: LOEC > 1000 and 100 μg Cd/g dw for survival and growth, respectively). Bioaccumulated cadmium and metallothioneins (MTs) concentrations were analyzed to investigate the role of MTs in reduced sensitivity to cadmium in late instar larvae. Metallothioneins were induced after treatment to greater Cd concentrations, but their levels in relation to cadmium body burdens did not fully explain low sensitivity of late instars to cadmium, which indicates some other effective way of detoxification in late instars. This study brings new information related to the role of MTs in age-dependent toxicant sensitivity and discusses the implications of divergence in data generated by chironomid sediment toxicity tests by standardized methods using different instars.

Colonization of man-made surface materials by algae and other microorganisms, i.e. biofouling, causes various aesthetical and health problems as well as deterioration of appearance and surface functioning. The present study investigated eight complex surfaces combining various TiO2, alkoxy-siloxane and hydrophobization agents that were combined to prevent growth of phototrophic filamentous alga Klebsormidium sp. Regarding photoactivity, NOx abatement at the studied surfaces has been improved by adding anti-aggregation substances such as bentonite. Surface properties were characterized by confocal microscopy, scanning electron microscopy, SEM, with the energy dispersive X-ray spectroscopy, EDX, and the contact angle measurements. Biological experiments with Klebsormidium alga were performed in a constructed flow-through system. The results indicated that superhydrophobicity, i.e. very low surface energy, was critical for efficient antifouling during the early colonization by algae. This was specifically demonstrated in two novel composite samples made of titanium dioxide and commercial hydrophobization agent UltraEverDry. Low surface energy of these materials (around 1.3 mJ/m2), high water contact angles (>140°) as well as decent photoactivity (NOx conversions by 16.7 % and 1.4 % in UV and VIS, respectively), provide a promising basis for further longer-term anti-fouling experiments. The present study introduces novel composite materials preventing the algal fouling, and it brings new evidences on the interactive effects on hydrophobic and photoactive surfaces.

Herbicides and their metabolites are often detected in water bodies where they may cause adverse effects to non-target organisms. Their effects at environmentally relevant concentrations are often unclear, especially concerning mixtures of pesticides. This study thus investigated the impacts of one of the most used herbicides: S-metolachlor and its two metabolites, metolachlor oxanilic acid (MOA) and metolachlor ethanesulfonic acid (MESA) on the development of zebrafish embryos (Danio rerio). Embryos were exposed to the individual substances and their environmentally relevant mixture until 120 hpf (hours post-fertilization). The focus was set on sublethal endpoints such as malformations, hatching success, length of fish larvae, spontaneous movements, heart rate and locomotion. Moreover, expression levels of eight genes linked to the thyroid system disruption, oxidative stress defense, mitochondrial metabolism, regulation of cell cycle and retinoic acid (RA) signaling pathway were analyzed. Exposure to S-metolachlor (1 mu g/L) and the pesticide mixture (1 mu g/L of each substance) significantly reduced spontaneous tail movements of 21 hpf embryos. Few rare developmental malformations were observed, but only in larvae exposed to more than 100 mu g/L of individual substances (craniofacial deformation, non-inflated gas bladder, yolk sac malabsorption) and to 30 mu g/L of each substance in the pesticide mixture (spine deformation). No effect on hatching success, length of larvae, heart rate or larvae locomotion were found. Strong responses were detected at the molecular level including induction of p53 gene regulating the cell cycle (the pesticide mixture - 1 mu g/L of each substance; MESA 30 mu g/L; and MOA 100 mu g/L), as induction of cyp26a1 gene encoding cytochrome P450 (pesticide mixture - 1 mu g/L of each substance). Genes implicated in the thyroid system regulation (dio2, thra, thrb) were all overexpressed by the environmentally relevant concentrations of the pesticide mixture (1 mu g/L of each substance) and MESA metabolite (1 mu g/L). Zebrafish thyroid system disruption was revealed by the overexpressed genes, as well as by some related developmental malformations (mainly gas bladder and yolk sac abnormalities), and reduced spontaneous tail movements. Thus, the thyroid system disruption represents a likely hypothesis behind the effects caused by the low environmental concentrations of S-metolachlor, its two metabolites and their mixture.

Cyanobacterial toxin cylindrospermopsin (CYN) is an emerging freshwater contaminant, whose expanding environmental occurrence might result into increased human health risks. CYN is potent hepatotoxin, with cytotoxicity and genotoxicity documented in primary hepatocytes or hepatoma cell lines. However, there is only limited information about CYN effects on adult human liver stem cells (LSCs), which play an important role in liver tissue development, regeneration and repair. In our study with human liver cell line HL1-hT1 which expresses characteristics of LSCs, CYN was found to be cytotoxic and increasing cell death after 24–48 h exposure to concentrations >1 μM. Subcytotoxic 1 μM concentration did not induce cell death or membrane damage, but inhibited cellular processes related to energy production, leading to a growth stagnation after >72 h. Interestingly, these effects were not associated with increased DNA damage, reactive oxygen species production, or endoplasmic reticulum stress. However, CYN induced a sustained (24–48 h) activation of mitogen-activated protein kinases ERK1/2 and p38, and increased expression of stress-related transcription factor ATF3. Thus, LSCs were not primarily affected by CYN-induced genotoxicity and oxidative stress, but via activation of signaling and transcriptional pathways critical for regulation of cell proliferation, stress responses, cell survival and inflammation. Alterations of LSCs during CYN-induced liver injury, including the role of nongenotoxic mechanisms, should be therefore considered in mechanistic assessments of chronic CYN hepatotoxicity and hepatocarcinogenicity.

Cyanotoxins microcystin-LR (MC-LR) and cylindrospermopsin (CYN) represent hazardous waterborne contaminants and potent human hepatotoxins. However, in vitro monolayer cultures of hepatic cell lines were found to recapitulate, poorly, major hepatocyte-specific functions and inadequately predict hepatotoxic effects of MC-LR and CYN. We utilized 3-dimensional (3D), scaffold-free spheroid cultures of human telomerase-immortalized adult liver stem cells HL1-hT1 to evaluate hepatotoxic potential of MC-LR and CYN. In monolayer cultures of HL1-hT1 cells, MC-LR did not induce cytotoxic effects (EC50 > 10 micromol/L), while CYN inhibited cell growth and viability (48h-96h EC50 ≈ 5.5-0.6 micromol/L). Growth and viability of small growing spheroids were inhibited by both cyanotoxins (≥0.1 micromol/L) and were associated with blebbing and disintegration at the spheroid surface. Hepatospheroid damage and viability reduction were observed also in large mature spheroids, with viability 96h-EC50 values being 0.04 micromol/L for MC-LR and 0.1 micromol/L for CYN, and No Observed Effect Concentrations

Insight into how environmental change determines the production and distribution of cyanobacterial toxins is necessary for risk assessment. Management guidelines currently focus on hepatotoxins (microcystins). Increasing attention is given to other classes, such as neurotoxins (e.g., anatoxin-a) and cytotoxins (e.g., cylindrospermopsin) due to their potency. Most studies examine the relationship between individual toxin variants and environmental factors, such as nutrients, temperature and light. In summer 2015, we collected samples across Europe to investigate the effect of nutrient and temperature gradients on the variability of toxin production at a continental scale. Direct and indirect effects of temperature were the main drivers of the spatial distribution in the toxins produced by the cyanobacterial community, the toxin concentrations and toxin quota. Generalized linear models showed that a Toxin Diversity Index (TDI) increased with latitude, while it decreased with water stability. Increases in TDI were explained through a significant increase in toxin variants such as MC-YR, anatoxin and cylindrospermopsin, accompanied by a decreasing presence of MC-LR. While global warming continues, the direct and indirect effects of increased lake temperatures will drive changes in the distribution of cyanobacterial toxins in Europe, potentially promoting selection of a few highly toxic species or strains. © 2018 by the authors. Growing concern about the adverse environmental and human health effects of a wide range of micropollutants requires the development of novel tools and approaches to enable holistic monitoring of their occurrence, fate and effects in the aquatic environment. A European-wide demonstration program (EDP) for effect-based monitoring of micropollutants in surface waters was carried out within the Marie Curie Initial Training Network EDA-EMERGE. The main objectives of the EDP were to apply a simplified protocol for effect-directed analysis, to link biological effects to target compounds and to estimate their risk to aquatic biota. Onsite large volume solid phase extraction of 50 L of surface water was performed at 18 sampling sites in four European river basins. Extracts were subjected to effect-based analysis (toxicity to algae, fish embryo toxicity, neurotoxicity, (anti-) estrogenicity, (anti-) androgenicity, glucocorticoid activity and thyroid activity), to target analysis (151 organic micropollutants) and to nontarget screening. The most pronounced effects were estrogenicity, toxicity to algae and fish embryo toxicity. In most bioassays, major portions of the observed effects could not be explained by target compounds, especially in case of androgenicity, glucocorticoid activity and fish embryo toxicity. Estrone and nonylphenoxyacetic acid were identified as the strongest contributors to estrogenicity, while herbicides, with a minor contribution from other micropollutants, were linked to the observed toxicity to algae. Fipronil and nonylphenol were partially responsible for the fish embryo toxicity. Within the EDP, 21 target compounds were prioritized on the basis of their frequency and extent of exceedance of predicted no effect concentrations. The EDP priority list included 6 compounds, which are already addressed by European legislation, and 15 micropollutants that may be important for future monitoring of surface waters. Despite intensive research and management efforts in the past decades, cyanobacterial blooms and their toxins, such as microcystins (MCs), continue to represent a major ecological and health problem in fresh waters throughout the world. Our objective was to compare the efficacy of two commonly used drinking water treatment technologies, chlorination and ozonation, in removing MCs and in reducing tumour promotion-related effects of cyanobacteria, such as inhibition of gap junctional intercellular communication (GJIC) and activation of mitogen activated protein kinases (MAPKs) in a rat liver epithelial stem-like cell line (WB-F344). This combined chemical and bioassay approach demonstrated that ozone effectively removed all MCs from an extract of a globally important bloom-forming cyanobacterium, Microcystis sp. Ozone also significantly reduced the overall tumour promotional potency of the cyanobacterial extract, as indicated by a substantial reduction in the ability of the extract to inhibit GJIC and activate extracellular receptor kinase 1/2 (ERK1/2). Although comparable reduction of total organic carbon was achieved by ozone and chlorine treatment, chlorination was much less effective in removing MCs and reducing the effects on GJIC. Chlorination had a biphasic effect with an observed decrease of extract-induced activation of ERK1/2 at the lower chlorine doses; whereas at high doses of chlorine the by-products of chlorination actually induced the activation of ERK1/2. The extracts induced p38 activation, and chlorination was not effective in reversing this effect, while ozone did reverse this effect, albeit not as much as the activation of ERK1/2. Thus, ozone was effective in reducing the toxicity of cyanobacterial extracts while chlorination was not only lacking efficacy, but at high doses of chlorination further produced by-products that were equally toxic as the untreated samples. Our study indicates the value of using an effect-based approach to assess the efficacy of water treatment systems in removing toxins, and more specifically demonstrates that ozone was more effective at reducing the toxic potential of cyanobacterial-contaminated water.

Advanced LC-MSMS methodology of MC analyses is presented. The chapter addresss both theoretical and practical aspects of cyanobacterial toxic blooms and relevant analyses of natural toxic blooms.

The paper presents the development of an advanced extraction and fast analytical LC MS/MS method for simultaneous analyses of reduced and oxidized glutathione (GSH and GSSG, respectively) in different animal tissues. The simultaneous determination of GSH and GSSG is crucial because the amount and ratio of both GSH and GSSG may be altered in response to oxidative stress, an important mechanism of toxicity. The method uses the derivatization of free thiol groups in GSH. Its performance was demonstrated for less explored tissues (lung, brain, and liver) in mouse. The combined extraction and analytical method has very low variability and good reproducibility, maximum coefficients of variance for within-run and between-run analyses under 8 %, and low limits of quantification; for GSH and GSSG, these were 0.2 nM (0.06 ng/mL) and 10 nM (6 ng/mL), respectively. The performance of the method was further demonstrated in a model experiment addressing changes in GSH and GSSG concentrations in lung of mice exposed to CdO nanoparticles during acute 72 h and chronic 13-week exposures. Inhalation exposure led to increased GSH concentrations in lung. GSSG levels were in general not affected; nonsignificant suppression occurred only after the longer 13-week period of exposure. The developed method for the sensitive detection of both GSH and GSSG in very low tissue mass enables these parameters to be studied in cases where only a little sample is available, i.e. in small organisms or in small amounts of tissue.

In vitro assays are broadly used tools to evaluate the estrogenic activity in Waste Water Treatment Plant (WWTP) effluents and their receiving rivers. Since potencies of individual estrogens to induce in vitro and in vivo responses can differ it is not possible to directly evaluate risks based on in vitro measures of estrogenic activity. Estrone, 17beta-estradiol, 17alfa-ethinylestradiol and to some extent, estriol have been shown to be responsible for the majority of in vitro estrogenic activity of municipal WWTP effluents. Therefore, in the present study safe concentrations of Estrogenic Equivalents (EEQs-SSE) in municipal WWTP effluents were derived based on simplified assumption that the steroid estrogens are responsible for all estrogenicity determined with particular in vitro assays. EEQs-SSEs were derived using the bioassay and testing protocol-specific in vitro potencies of steroid estrogens, in vivo predicted no effect concentration (PNECs) of these compounds, and their relative contributions to the overall estrogenicity detected in municipal WWTP effluents. EEQs-SSEs for 15 individual bioassays varied from 0.1 to 0.4 ng EEQ/L. The EEQs-SSEs are supposed to be increased by use of location-specific dilution factors of WWTP effluents entering receiving rivers. They are applicable to municipal wastewater and rivers close to their discharges, but not to industrial waste waters. Despite intensive research into toxic bloom-forming cyanobacteria, the majority of their metabolites remain unknown. The present study explored in detail a novel bioactivity identified in cyanobacteria, i.e. inhibition of gap junctional intercellular communication (GJIC), a marker of tumor promotion. The extracellular mixture (exudate) of the cyanobacterial strain Cylindrospermopsis raciborskii (SAG 1.97) was fractionated by semi-preparative reversed phase HPLC, and the fractions assessed for their potencies to inhibit GJIC. Two non-polar fractions that significantly inhibited GJIC were further fractionated, tested and analyzed using multiple mass spectrometric methods. Investigations led to the identification of a putative chemical compound (molecular formula C18H34O3, m/z 299.2581 for the [M+H](+) ion) responsible for observed bioactivities. Specific inhibitors of signaling pathways were used to screen for biochemical mechanisms beyond GJIC inhibition, and the results indicate the involvement of ERK1/2 kinases via a mechanism related to the action of epidermal growth factor EGF but clearly distinct from other anthropogenic tumor promoters like polychlorinated biphenyls or polycyclic aromatic hydrocarbons. The chemical and in vitro toxicological characterizations of the newly described metabolite provide important insights into the still poorly understood health impacts of complex toxic cyanobacterial blooms and indicate that currently applied monitoring practices may underestimate actual risks.

Compounds with estrogenic potencies and their adverse effects in surface waters have received much attention. Both anthropogenic and natural compounds contribute to overall estrogenic activity in freshwaters. Recently, estrogenic potencies were also found to be associated with cyanobacteria and their blooms in surface waters. The present study developed and compared the solid phase extraction and LC-MS/MS analytical approaches for determination of phytoestrogens (8 flavonoids - biochanin A, coumestrol, daidzein, equol, formononetin, genistein, naringenin, apigenin and 5 sterols - ergosterol, (beta-sitosterol, stigmasterol, campesterol, brassicasterol) and cholesterol in water. The method was used for analyses of samples collected in stagnant water bodies dominated by different cyanobacterial species. Concentrations of individual flavonoids ranged from below the limit of detection to 3.58 ng/L. Sterols were present in higher amounts up to 2.25 mu g/L. Biological potencies of these phytoestrogens in vitro were characterized using the hER alpha-HeLa-9903 cell line. The relative estrogenic potencies (compared to model estrogen - 17 beta-estradiol) of flavonoids ranged from 2.25E-05 to 1.26E-03 with coumestrol being the most potent. None of the sterols elicited estrogenic response in the used bioassay. Estrogenic activity was detected in collected field water samples (maximum effect corresponding to 2.07 ng/L of 17 beta-estradiol equivalents, transcriptional assay). At maximum phytoestrogens accounted for only 1.56 pg/L of 17 beta-estradiol equivalents, contributing maximally 8.5% of the total estrogenicity of the water samples. Other compounds therefore, most likely of anthropogenic origin such as steroid estrogens, are probably the major drivers of total estrogenic effects in these surface waters.

In recent years, the environmental presence of pharmaceuticals - including anticancer drugs - is an emerging issue. Because of the lack of appropriate critical studies about anticancer drug effects in frogs, the aim of the present study was to investigate lethal and teratogenic effects of five anticancer drugs widely used in large quantities, i.e. 5-flourouracil, capecitabine, cisplatin, etoposide, and imatinib, in the embryos of the South African clawed frog, Xenopus laevis, using FETAX - Frog Embryo Teratogenesis Assay in Xenopus. None of the studied anticancer drugs induced statistically significant mortality within the concentrations tested (0.01–50 mg/L, depending on the studied compound), and no growth inhibition of embryos after a 96-h exposure was observed. Except for cisplatin, the other pharmaceuticals induced an increase of developmental malformations such as abdominal edema, axial flexure, head, eyes, gut and heart malformations with statistically significant effects observed at the highest concentrations tested (50 mg/L for 5-flourouracil; 30 mg/L for etoposide and 20 mg/L for capecitabine and imatinib). The results indicate that anticancer drugs can affect embryogenesis mechanisms.

Isoflavones are natural phytoestrogens with antioxidant and endocrine-disrupting potencies. Monitoring of their levels is important to ensure the high quality and safety of food, milk, and dairy products. The efficiency and accuracy of phytoestrogen analyses in complex matrices such as milk depend on the extraction procedure, which often uses hydrolysis by means of the β-glucuronidase/sulfatase enzyme originating from Helix pomatia. The present study reveals that the commercially available hydrolytic enzyme is contaminated by several phytoestrogen isoflavones (genistein, daidzein, formononetin, and biochanin A) and their metabolite equol, as well as flavones (naringenin and apigenin) and coumestrol. We show that the concentrations of daidzein and genistein in the enzyme could have impaired the results of analyses of the main isoflavones in several previously published studies. Of 8 analyzed compounds, only equol was confirmed in the present study and it serves as a reliable marker of phytoestrogens originating from cow feed. Critical reassessment of phytoestrogen concentrations in milk is needed because several previously published studies might have overestimated the concentrations depending on the extraction procedure used.

Here is presented some of the first information on interactions of compounds produced by cyanobacteria and green algae with estrogen receptor signaling. Estrogenic potency of aqueous extracts and exudates (culture spent media with extracellular products) of seven species of cyanobacteria (10 different laboratory strains) and two algal species were assessed by use of in vitro trans-activation assays. Compounds produced by cyanobacteria and algae, and in particular those excreted from the cells, were estrogenic. Most exudates were estrogenic with potencies expressed at 50% of the maximum response under control of the estrogen receptor ranging from 0.2 to 7.2 ng 17β-estradiol (E2) equivalents (EEQ)/L. The greatest estrogenic potency was observed for exudates of Microcystis aerigunosa, a common species that forms water blooms. Aqueous extracts of both green algae, but only one species of cyanobacteria (Aphanizomenon gracile) elicited significant estrogenicity with EEQ ranging from 15 to 280 ng 17β-estradiol (E2)/g dry weight. Scenedesmus quadricauda exudates and extracts of Aphanizomenon flos-aquae were antagonistic to the ER when coexposed to E2. The EEQ potency was not correlated with concentrations of cyanotoxins, such as microcystin and cylindrospermopsin, which suggests that the EEQ was comprised of other compounds. The study demonstrates some differences between the estrogenic potency of aqueous extracts prepared from the same species, but of different origin, while the effects of exudates were comparable within species. The observed estrogenic potencies are important namely in relation to the possible mass expansion of cyanobacteria and release of the active compounds into surrounding water. Oxidative stress is one of the biochemical mechanisms involved in toxicity of cyanobacterial toxins microcystins (MC), but its role in the effects of complex water blooms is elusive. The aim of this study was to investigate effects of pure MCs and different complex mixtures of cyanobacterial metabolites on the growth and biochemical markers of oxidative stress and detoxification in green alga Pseudokirchneriella subcapitata. Pure MCs at high concentrations (300 μg/L) had no effects on the growth of P. subcapitata (up to 10 day exposures) but stimulated activity of glutathione reductase (GR) after short 3 and 24 h exposures. Other biomarkers (levels of glutathione, GSH, and activities of glutathione-S-transferase, GST, and glutathione peroxidase, GPx) were not affected by pure MCs). Crude extract of the laboratory culture of cyanobacteria Microcystis aeruginosa (containing 300 μg/L of MCs) had no effects on algal growth or any of the biomarkers. Weak growth stimulations after 4-7 days were observed after exposures to the growth-spent medium of the M. aeruginosa culture, which also inhibited activities of GST after prolonged exposures. Other investigated parameters (reduced GSH and activity of GPx) were not affected by any of the cyanobacterial samples. The results were compared with effects of model oxidative stressor herbicide paraquat, which exhibited variable effects on both algal growth and biomarkers (decrease in reduced GSH, stimulations of GR). Taken together, although pure MCs induce oxidative stress in green alga, the effects of cyanobacterial mixtures, which are more relevant to the natural situation, are more complex and they differ from the pure toxin. High variability in the biochemical responses to the oxidative stress makes the interpretation of results complicated, which limits the use of these biomarkers as early warnings of toxicity under natural conditions.

Microcystins (MC) are potent hepatic toxins delivered into the cells by organic anion transporting peptides (OATP) where they target protein phosphatases and mitochondria. We analyzed the effects of MC-LR on primary hepatocytes, HepG2, and Jurkat T cells, and isolated rat liver mitochondria by measuring changes in O(2) consumption by optical oxygen sensing technique. Respiration of fresh primary hepatocytes was inhibited by MC-LR with EC50 = 2.74 +/- 0.65 nM, whereas an uncoupling effect on mitochondrial state 2 and state 3 respiration was observed with glutamate/malate as a substrate. HepG2 and Jurkat T cells lacking OATP showed no sensitivity to MC-LR; however, facilitated delivery of MC-LR resulted in a marked enhancement of HepG2 O(2) consumption and inhibition of Jurkat O(2) consumption at >or=0.1 nM. The respiratory response did not coincide with changes in viability, total cellular ATP, extracellular acidification, ROS formation, or protein phosphorylation, which were detectable at higher MC-LR doses. Such prominent effect on cellular respiration was therefore used for the detection of MC-LR in environmental samples. A simple and sensitive screening assay for MC-LR toxicity was developed, which uses Jurkat cells, facilitated delivery of the toxin(s) and measurement on a fluorescent reader. The assay was applied to a panel of environmental samples suspected to contain MC and benchmarked against the ELISA test. It allowed identification of toxic samples and quantification of both nonspecific and MC-LR type of toxicity.

In spite of substantial research on health and the ecological risks as- sociated with cyanobacterial toxins in the past decades, the understanding of the natural dynamics and variability of toxic cyanobacterial blooms is still li- mited. Herein, the results of long term monitoring 1998-1999/2001-2008 of three reservoirs (Vir, Brno and Nove Mlýny, Chech Republic), where toxic blooms develop annually, are reported. These three reservoirs provide a unique model because they are interconnected by the Svratka River, which allows pos- sible transfer of phytoplankton as well as toxins from one reservoir to another. The frequency of the occurrence and dominance of the major cyanobacterial taxa Microcystis aeruginosa did not change during the investigated period but substantial variability was observed in the composition of other phytoplankton. Although absolute concentrations of the studied toxins (microcystins) differed among the reservoirs, there were apparent parallel trends. For example, during certain years, the microcystin concentrations were systematically elevated in all three studied reservoirs. Furthermore, the concentration profiles in the three sites were also correlated (parallel trends) within individual seasons based on monthly sampling. Microcystin-LR, a variant for which the World Health Or- ganization has recommended a guideline value, formed only about 30-50 % of the total microcystins. This is of importance, especially in the Vir reservoir that serves as a drinking water supply. The maxima in the cell-bound microcystins (intracellular; expressed per dry weight biomass) generally preceded the maxima of total microcystins (expressed per volume of water sample). Overall, the maximum concentration in the biomass (all three reservoirs, period 1993- 2005) was 6.1 mg g -1 dry weight and the median values were in the range 0.065-2.3 mg g -1 dry weight. These are generally high concentrations in comparison with both Czech Republic and worldwide reported data. The present data revealed substantial variability of both toxic cyanobacteria and their peptide toxins that should be reflected by detailed monitoring programs.

Cyanobacterial blooms in freshwaters represent a major ecological and human health problem worldwide. This paper briefly summarizes information on major cyanobacterial toxins (hepatotoxins, neurotoxins etc.) with special attention to microcystins - cyclic heptapeptides with high acute and chronic toxicities. Besides discussion of human health risks, microcystin ecotoxicology and consequent ecological risks are also highlighted. Although significant research attention has been paid to microcystins, cyanobacteria produce a wide range of currently unknown toxins, which will require research attention. Further research should also address possible additive, synergistic or antagonistic effects among different classes of cyanobacterial metabolites, as well as interactions with other toxic stressors such as metals or persistent organic pollutants.

Retinoids are dietary hormones acting through nuclear receptors for retinoic acid, important especially during embryonic development. This study focuses on the disruption of signaling pathways of retinoids by polycyclic aromatic hydrocarbons (PAHs) and their N-heterocyclic analogs (N-PAHs), important environmental contaminants with numerous biological effects. In vitro test with P19/A15 cell line stably transfected with luciferase reporter gene under control of retinoic acid-responsive elements was used to investigate both direct activation of retinoic acid receptors and modulation of response induced by natural ligand all-trans retinoic acid (ATRA) by 26 PAHs and N-PAHs. While none of individual compounds alone activated retinoic acid receptors, many of them modulated ATRA-mediated activity both after 6 h and 24 h exposure. Majority of compounds active after 6 h downregulated ATRA-mediated activity (most effective were two analogs of dibenz[a,h]anthracene with LOECs about 185 nM), while most compounds active after 24 h upregulated the effects of ATRA (most effective benz[a]acridine and dibenz[a,i]acridine caused 400% induction of ATRA response). Quantitative structure-activity relationship analysis identified molecular volume and dipole moment as the most important descriptors of inhibitory effects after 6 h, while length, total molecular energy, gap-HOMO/LUMO and Van der Waals energy are important descriptors for stimulatory effects of PAHs and N-PAHs. This study demonstrates those abundant pollutants such as PAHs and their analogs interfere in vitro with retinoid signaling, which could play role in some in vivo effects of these organic contaminants such as teratogenicity.

Progress in ecotoxicological research documents that crustaceans are highly vulnerable to diverse chemicals and toxicants in the environment. In particular, pollutants affecting endocrine homeostasis in crustaceans (i.e., endocrine disruptors) are intensively studied, and serious reproductive disorders have been documented. In this review, current knowledge about the endocrine regulation of the crustacean reproduction is put together with the published ecotoxicological data with an attempt to summarize the potential of xenobiotics to affect crustacean reproduction. Following gaps and trends were identified: (1) Studies are required in the field of neurohormone (serotonin and dopamine) regulation of the reproduction and possible modulations by environmental toxicants such as antidepressant drugs. (2) Molting-related parameters (regulated by ecdysteroid hormones) are closely coordinated with the development and reproduction cycles in crustaceans (cross-links with methyl farnesoate signalling), and their susceptibility to toxicants should be studied. (3) Other biochemical targets for xenobiotics were recently discovered in crustaceans and these should be explored by further ecotoxicological studies (e.g., new information about ecdysteroid receptor molecular biology). (4) Some sex steroid hormones known from vertebrates (testosterone, progesterone) have been reported in crustaceans but knowledge about their targets (crustacean steroid receptors) and signalling is still limited. (5) Determination of the sex in developing juveniles (affecting the sex ratio in population) is a sensitive parameter to various xenobiotics (including endocrine disruptors) but its modulation by general environmental stress and non-specific toxicity should be further studied.

Massive cyanobacterial water blooms and production of toxins (cyanotoxins) have become a worldwide problem. In this report, we present results of cyanotoxins analyses (peptide microcystins, alkaloid cylindrospermopsin) in the Czech Republic reservoirs using HPLC-PDA and ELISA. Our study suggests the occurrence of cylindrospermopsin in the Czech Republic for the first time (particularly, in water blooms containing Aphanizomenon klebahnii). We also discuss human health risks associated with microcystins in relation to the drinking water guideline value of 1.0 μg/l as recommended by the World Health Organization.

This work evaluated the effects of various cyanobacterial fractions in Frog Embryo Teratogenesis Assay Xenopus (FETAX) with African clawed frog embryos. Fractions were prepared from five biomasses with different dominant genera (Microcystis, Aphanizomenon, Anabaena, Planktothrix) and different microcystin content. Effects of following fractions were investigated: (I) homogenate of complex cyanobacterial biomass, (II) cell debris (pellet) after centrifugation of complex biomass, (III) supernatant after centrifugation of complex biomass (= crude aqueous extract), (IV) permeate after passing of crude extract through C-18 column (fraction devoid of microcystins), and (V) eluate from C-18 column (containing microcystins, if present). Besides classical parameters evaluated in 96 h FETAX (mortality, growth inhibition, malformations), we have also assessed the effects on biochemical markers of oxidative stress and detoxification (glutathione pool, GSH; activity of glutathione peroxidase, GPx; glutathione reductase, GR; activity of glutathione-S-transferase, GST). Complex biomass (I) and aqueous extract (III) were generally the most toxic fractions in terms of mortality and growth inhibition, whereas eluates containing microcystins (V) were generally less toxic. On the other hand, the same fraction (eluates) induced significant malformations in low concentrations but the effects were not related to the content of microcystins. Biomarkers were affected in variable manner but no significant effect or clear relation to microcystin content was observed. Our data support the hypothesis that microcystins are not the only or major toxic compounds in the complex cyanobacterial samples (at least for some species) and that more attention should be paid to other components of complex cyanobacterial biomass including non-specific parameters such as oxygen content or toxic ammonia released during bacterial decay of organic material.

Quaternary benzo[c]phenanthridine alkaloids (QBAs) sanguinarine and chelerythrine exert a plethora of biological activities. Nevertheless, the specific cellular target for these alkaloids within the cell was not identified as far. Several literary data indicate that biological effects of QBAs could be associated with aryl hydrocarbon receptor (AhR) signaling pathway, including cytochrome P450 CYP1A, however, available information are controversial. In this work we analyzed the effects of sanguinarine and chelerythrine on AhR activity in rat hepatoma cells HII4E. luc stably transfected with dioxin responsive element fused to luciferase gene (DRE-LUC). Studied QBAs were tested in submicromolar concentration range (0.0001–1 μM) and in incubation times 6, 24 and 48 h. Transcriptional activity of AhR was monitored by chemiluminiscence measurement of luciferase catalytic activity. Sanguinarine and chelerythrine did not activated AhR in any time or dose tested. Chelerythrine (1 μM) but not sanguinarine caused moderate inhibition of AhR activation by 10 picomolar dioxin (exponential phase of receptor activation). In contrast, AhR activation by 2.5 nM dioxin (saturated receptor) was not affected by either alkaloid tested. In conclusion, the findings presented here favor rather for inactivity or modest inhibitory effect of QBAs on AhR signaling pathways in vitro than for the activation of the receptor. Regarding the concentrations of QBAs occurring in vivo , the use of products containing sanguinarine and/or chelerythrine has low toxicological potential in terms of the interactions with AhR signaling pathways.

Mar‰alek: Sublethal Toxic Effects and Induction of Gutathione S-transferase by Short chain Chlorinated Paraffins (SCCPs) and C-12 alkane (dodecane) in Xenopus laevis Frog Embryos. Acta Vet. Brno 2006: 75: 115-122. Short chain chlorinated paraffins (SCCPs) are important industrial chemicals with high persistence in the environment but poorly characterized ecotoxicological effects. We studied embryotoxic effects of commercial mixture of SCCP (carbon length C-12, 56% of chlorine; CP5612) and non-chlorinated n-alkane (dodecane, C-12) in the 96h Frog Embryo Teratogenesis Assay – Xenopus (FETAX). Only weak lethal effects were observed for both substances (the highest tested concentration 500 mg/L of both chemicals caused up to 11% mortality). On the other hand, we observed developmental malformations and reduced embryo growth at 5 mg/l and higher concentrations. However, the effects were not related to chlorination pattern as both SCCPs and dodecane induced qualitatively similar effects. SCCPs also significantly induced phase II detoxification enzyme glutathione S-transferase (GST) in Xenopus laevis embryos even at 0.5 mg/L, and this biomarker might be used as another early warning of chronic toxic effects. Our results newly indicate significant developmental toxicity of both SCCPs and n-dodecane to aquatic organisms along with inductions of specific biochemical toxicity mechanisms.

Berberine has long been considered a candidate for an antimalarial drug. It exerts a plethora of biological activities and has been used in the treatment of diarrhea and gastro-enteritis for centuries. Here we provide evidence that berberine activates the aryl hydrocarbon receptor (AhR) in human hepatoma (HepG2) and rat hepatoma cells stably transfected with a dioxin responsive element fused to the luciferase gene (H4IIE.luc). AhR was activated by high doses of berberine (10-50 microM) after 6 and 24 h of incubation as revealed by CYP1A1 mRNA expression (HepG2) and AhR-dependent luciferase activity (H4IIE.luc). Toxicity endpoints (nonspecific cytotoxicity, hepatotoxicity, neurotoxicity, immunotoxicity, and mutagenicity) were studied in cyanobacterial mats obtained from a shallow river. Some of the cyanobacterial mats tested were known to be non-geosmin producers, while others were geosmin-producers. No microcystin-like compounds were detected by HPLC in any of the biofilm samples. The mutagenicity and neurotoxicity of biofilm metabolites was negligible, and generally weak adverse effects of biofilm extracts detected in a battery of in-vitro assays indicated relatively low human health risks associated with biofilm toxicity. While the toxicity responses detected in the studied biofilms were weak, effects were not related to production of geosmin. It was therefore concluded that the production of this metabolite cannot be taken as an indication per se of the existence of a health hazard.

Acta Vet. Brno 2004, 73: 259-265. The aim of this study was to examine the influence of a repeated exposure of the carp to the extract of cyanobacteria. We exposed carp embryos and larvae to the extract of cyanobacteria of the cumulative amount of microcystin LR, YR and RR of 1.3 µg⋅l-1 or 13 µg⋅l-1, respectively. One year later we exposed one half of these fish to the second dose of the extract of cyanobacteria of the cumulative amount of microcystins of 13 µg⋅l-1 and the second half to pure water to eliminate the influence of handling stress. The haematological and histological changes were examined. Similar changes of haematological indices were detected both after exposure to the extract and after the handling stress of fish. Especially indices of the red blood cells were characterised by significant increase (p ≤ 0.01 or p ≤ 0.05) of haematocrit value and haemoglobin concentration in groups with long-term exposure (30 days) during the early life stages as well as in control groups. The initial values of white blood cell count were significantly increased (p ≤ 0.01) in the control group and in the group exposed to the low concentration (1.3 µg⋅l -1 ) and after short exposure (8 days) during the early life stages as compare to other groups. The white blood cell count of these groups significantly decreased (p ≤ 0.01) after exposure to both pure water and the extract. Phagocytic activity was significantly decreased (p ≤ 0.01 or p ≤ 0.05) after stress and after exposure to the extract in groups of long term exposure (30 days) during the early life stages in comparison to control and groups of low exposure. Total plasma protein concentration significantly increased (p ≤ 0.01) in all groups after exposure to the extract in comparison to the initial values and only in groups of long term exposure during the early life stages and in the control group after the handling stress. The activities of aspartate aminotransferase (AST) and lactate dehydrogenase (LDH) were decreased in groups of long term exposure during the early life stages after the exposure to the extract. Clinical and histological changes were not observed in any group of fish, except for the fatty infiltration of liver with a tendency to focal big droplet dystrophic steatosis in all groups of fish. A pan-European monitoring campaign of the wastewater treatment plant (WWTP) effluents was conducted to obtain a concise picture on a broad range of pollutants including estrogenic compounds. Snapshot samples from 75 WWTP effluents were collected and analysed for concentrations of 150 polar organic and 20 inorganic compounds as well as estrogenicity using the MVLN reporter gene assay. The effect-based assessment determined estrogenicity in 27 of 75 samples tested with the concentrations ranging from 0.53 to 17.9 ng/L of 17-beta-estradiol equivalents (EEQ). Approximately one third of municipal WWTP effluents contained EEQ greater than 0.5 ng/L EEQ, which confirmed the importance of cities as the major contamination source. Beside municipal WWTPs, some treated industrial wastewaters also exhibited detectable EEQ, indicating the importance to investigate phytoestrogens released from plant processing factories. No steroid estrogens were detected in any of the samples by instrumental methods above their limits of quantification of 10 ng/L, and none of the other analysed classes of chemicals showed correlation with detected EEQs. The study demonstrates the need of effect-based monitoring to assess certain classes of contaminants such as estrogens, which are known to occur at low concentrations being of serious toxicological concern for aquatic biota.

Activation of aryl hydrocarbon receptor (AhR) by 30 polycyclic aromatic hydrocarbons (PAHs) was determined in the chemical-activated luciferase expression (CALUX) assay, using two exposure times (6 and 24 h), in order to reflect the metabolization of PAHs. AhR-inducing potencies of PAHs were expressed as induction equivalency factors (IEFs) relative to benzo[ a ]pyrene and 2,3,7,8-tetrachlorodibenzo- p -dioxin (TCDD). In 24 h exposure assay, the highest IEFs were found for benzo[ k ]fluoranthene, dibenzo[ a , h ]anthracene and dibenzo[ a , k ]fluoranthene (approximately three orders of magnitude lower than TCDD) followed by dibenzo[ a , j ]anthracene, benzo[ j ]fluoranthene, indeno[1,2,3- cd ]pyrene, and naphtho[2,3- a ]pyrene. The 6 h exposure to PAHs led to a significantly higher AhR-mediated activity than the 24 h exposure (generally by two orders of magnitude), probably due to the high rate of PAH metabolism. The strongest AhR inducers showed IEFs approaching that of TCDD. Several PAHs, including some strong mutagens, such as dibenzo[ a , l ]pyrene, cyclopenta[ cd ]pyrene, and benzo[ a ]perylene, elicited only partial agonist activity. Calculation of IEFs based on EC25 values and/or 6 h exposure data is suggested as an alternative approach to estimation of toxic potencies of PAHs with high metabolic rates and/or the weak AhR agonists. The IEFs, together with the recently reported relative mutagenic potencies of PAHs [Mutat. Res. 371 (1996) 123; Mutat. Res. 446 (1999) 1] were combined with data on concentrations of PAHs in extracts of model environmental samples (river sediments) to calculate AhR-mediated induction equivalents and mutagenic equivalents. The highest AhR-mediated induction equivalents were found for benzo[ k ]fluoranthene and benzo[ j ]fluoranthene, followed by indeno[1,2,3- cd ]pyrene, dibenzo[ a , h ]anthracene, benzo[ a ]pyrene, dibenzo[ a , j ]anthracene, chrysene, and benzo[ b ]fluoranthene. High mutagenic equivalents in the river sediments were found for benzo[ a ]pyrene, dibenzo[ a , e ]pyrene, and naphtho[2,3- a ]pyrene and to a lesser extent also for benzo[ a ]anthracene, benzo[ b ]fluoranthene, indeno[1,2,3- cd ]pyrene, benzo[ j ]fluoranthene, dibenzo[ a , e ]fluoranthene and dibenzo[ a , i ]pyrene. These data illustrate that AhR-mediated activity of PAHs, including the highly mutagenic compounds, occurring in the environment but not routinely monitored, could significantly contribute to their adverse effects.

We investigated whether and how could various modulators of arachidonic acid metabolism affect apoptosis induced by tumour necrosis factor-alpha (TNF-alpha) in human myeloid leukaemia HL-60 cells. These included arachinonyltrifluoromethyl ketone (AACOCF3; cytosolic phospholipase A2 inhibitor), indomethacin (cyclooxygenase inhibitor), MK-886 (3-[1-(4-chlorobenzyl)-3-t-butyl-thio-5-isopropylindol-2-yl]-2,2-dimethyl propanoic acid; 5-lipoxygenase-activating protein inhibitor), nordihydroguaiaretic acid (general lipoxygenase inhibitor), and arachidonic acid itself. Incubation of HL-60 cells with nordihydroguaiaretic acid resulted in apoptosis and it was characterised by mitochondria membrane depolarisation, release of cytochrome c from mitochondria into cytosol and activation of caspase-3. Indomethacin and nordihydroguaiaretic acid synergistically potentiated TNF-alpha-induced apoptosis, while arachidonic acid, AACOCF3 and MK-886 did not modulate its effects. Furthermore, indomethacin potentiated apoptosis in cells treated with a differentiating agent, all-trans retinoic acid, which induces resistance to TNF-alpha. However, the observed effects were probably not associated either with the cyclooxygenase- or lipoxygenase-dependent activities of indomethacin and nordihydroguaiaretic acid, respectively. Since indomethacin may reportedly activate peroxisome proliferator-activated receptors (PPARs), the effects of specific ligands of PPARs on apoptosis were studied as well. It was found that selective PPARs ligands had no effects on TNF-alpha-induced apoptosis. The findings suggest that arachidonic acid metabolism does not play a key role in regulation of apoptosis induced by TNF-alpha in the present model. Nevertheless, our data raise the possibility that indomethacin could potentially be used to improve the treatment of human myeloid leukaemia.

Massive toxic blooms of cyanobacteria represent a major threat to water supplies worldwide, yet serious gaps exist in understanding their complex toxic effects, including the role of lipopolysaccharides (LPS). The present comparative study focused on the levels and biological activities of LPS isolated from Microcystis aeruginosa, which is one of the most globally distributed toxic species. Using hot phenol extraction, LPS was isolated from 3 laboratory cultures and 11 natural water blooms. It formed 0.2-0.7% of the original dry biomass of the cyanobacteria, based on gravimetry. Additional analyses by commercial anti-LPS ELISA were correlated with gravimetry but showed concentrations that were about 7-times lower, which indicated either impurities in isolated LPS or the poor cross-reactivity of the antibodies used. LPS isolates from M. aeruginosa were potent pyrogens in the traditional Limulus amebocyte lysate (LAL)-test, but comparison with the PyroGene test demonstrated the limited selectivity of LAL with several interferences. The determined pyrogenicity (endotoxin units, EU) ranged from very low values in laboratory cultures (less than 0.003 up to 0.008-EU per 100 pg LPS) to higher values in complex bloom samples (0.01-0.078 EU per 100 pg of LPS), which suggested the role of bloom-associated bacteria in the overall effects. Potent pro-inflammatory effects of the studied LPS from both cultures and bloom samples were observed in a highly-relevant ex vivo human blood model by studying reactive oxygen species production in phagocytes as well as increased productions of interleukin 8, IL-8, and tumor necrosis factor α, TNF-α. LPS from M. aeruginosa seem to modulate several pathways involved in the regulation of both innate immunity and specific responses. In comparison to the standard pathogenic bacterial LPS (World Health Organization Escherichia coli O113:10 endotoxin; activity 1 EU per 100 pg), the studied cyanobacterial samples had pyrogenicity potencies that were at least 12-times lower. However, the health risks associated with LPS from M. aeruginosa should not be underestimated, especially with respect to diverse biological effects observed ex vivo and in the case of massive blooms in drinking water reservoirs, where the estimated pyrogenicity can reach up to 46,000 EU per mL of water.

Inhibition of gap junctional intercellular communication (GJIC) is affiliated with tumor promotion process and it has been employed as an in vitro biomarker for evaluation of tumor promoting effects of chemicals. In the present study we investigated combined effects of anthropogenic environmental contaminants 2,2′,4,4′,5,5′-hexachlorobiphenyl (PCB 153) and fluoranthene, cyanotoxins microcystin-LR and cylindrospermopsin, and extracts of laboratory cultures of cyanobacteria Aphanizomenon gracile and Cylindrospermopsis raciborskii, on GJIC in the rat liver epithelial cell line WB-F344. Binary mixtures of PCB 153 with fluoranthene and the mixtures of the two cyanobacterial strains elicited simple additive effects on GJIC after 30 min exposure, whereas microcystin-LR and cylindrospermopsin neither inhibited GJIC nor altered effects of PCB 153 or fluoranthene. However, synergistic effects were observed in the cells exposed to binary mixtures of anthropogenic contaminants (PCB 153 or fluoranthene) and cyanobacterial extracts. The synergistic effects were especially pronounced after prolonged (6–24 h) co-exposure to fluoranthene and A. gracile extract, when mixture caused nearly complete GJIC inhibition, while none of the individual components caused any downregulation of GJIC at the same concentration and exposure time. The effects of cyanobacterial extracts were independent of microcystin-LR or cylindrospermopsin, which were not detected in cyanobacterial biomass. It provides further evidence on the presence of unknown tumor promoting metabolites in cyanobacteria. Clear potentiation of the GJIC inhibition observed in the mixtures of two anthropogenic contaminants and cyanobacteria highlight the importance of combined toxic effects of chemicals in complex environmental mixtures.

Cyanobacterial blooms represent a serious threat to the aquatic environment. Among other effects, biochemical markers have been studied in aquatic vertebrates after exposures to toxic cyanobacteria. Some parameters such as protein phosphatases may serve as selective markers of exposure to microcystins, but under natural conditions, fish are exposed to complex mixtures, which affect the overall biomarker response. This review aims to provide a critical summary of biomarker responses in aquatic vertebrates (mostly fish) to toxic cyanobacteria with a special focus on detoxification and oxidative stress. Detoxification biomarkers such as glutathione (GSH) and glutathione-S-transferase (GST) showed very high variability with poor general trends. Often, stimulations and/or inhibitions and/or no effects at GSH or GST have been reported, even within a single study, depending on many variables, including time, dose, tissue, species, etc. Most of the oxidative stress biomarkers (e.g., superoxide dismutase, catalase, glutathione peroxidase, and glutathione reductase) provided more consistent responses, but only lipid peroxidation (LPO) seemed to fulfill the criteria needed for biomarkers, i.e., a sufficiently long half-life and systematic response. Indeed, reviewed papers demonstrated that toxic cyanobacteria systematically elevate levels of LPO, which indicates the important role of oxidative damage in cyanobacterial toxicity. In summary, the measurement of biochemical changes under laboratory conditions may provide information on the mode of toxic action. However, comparison of different studies is very difficult, and the practical use of detoxification or oxidative stress biomarkers as diagnostic tools or early warnings of cyanobacterial toxicity is questionable.

Cyanobacterial extracts have been recently shown to alter two in vitro biomarkers of tumor promotion, namely to cause inhibition of gap-junctional intercellular communication (GJIC) and activation of mitogen-activated protein kinases (Blaha et al., 2010a). In the present study, we investigated GJIC-inhibitory potencies of 10 laboratory strains representing common water bloom-forming cyanobacteria (Anabaena, Aphanizomenon, Cylindrospermopsis, Microcystis and Planktothrix) and six natural water bloom samples (dominated by Aphanizomenon sp. or Microcystis). The most pronounced inhibitions of GJIC in a model rat liver epithelial cell line WB-F344 were caused by methanolic extracts of Anabaena flos-aquae UTEX 1444, Aphanizomenon flos-aquae SAG 31.87, Aphanizomenon gracile RCX 06, Microcystis aeruginosa PCC 7806, Cylindrospermopsis raciborskii SAG 1.97, Planktothrix agardhii CCALA 159 and SAG 32.79, whereas weaker effects were induced by Aphanizomenon klebahnii CCALA 009 and no inhibition was induced by extracts of Aph. flos-aquae PCC 7905 and Aph. gracile SAG 31.79. Exudates of the laboratory cultured strains concentrated by solid phase extraction also induced species-specific inhibitory effects, but they did not necessarily correlate with the inhibitory potencies of extracts from the corresponding species. Interestingly, the GJIC-inhibitory effects may not be restricted to cyanobacteria, since exudates of two green alga species also affected GJIC, although their extracts caused no effects. The extracts from different natural water blooms inhibited GJIC with different potencies without apparent relation to bloom-species composition. Since the observed effects on GJIC did not correlate with the content of cyanotoxins microcystins and cylindrospermopsin in the tested samples, they were most likely induced by unknown compound(s). Our results indicate that putative tumor promoting compound(s) could be associated with different species of bloom-forming cyanobacteria, but their production is probably species- and strain-specific.

Polycyclic aromatic hydrocarbons (PAHs) and their N-heterocyclic analogs (N-PAHs) are important environmental contaminants with negative effects in living organisms, including teratogenicity and embryotoxicity. Though most studies linked their embryotoxicity with aryl hydrocarbon receptor (AhR) and cytochrome P450 activation, the exact mechanism is not known. Other mechanisms such as disruption of retinoid signaling were recently suggested to be of importance. This study investigated PAHs and N-PAHs interference with retinoid signaling in vitro by modulating all-trans retinoic acid (ATRA) mediated response in a reporter gene assay using P19/A15 cell line. Further, effects on pluripotency and differentiation processes were evaluated by measuring octamer-4 (Oct-4), an important pluripotency marker and master differentiation factor. Two of the studied compounds, benz[a]anthracene and benz[c]acridine significantly up-regulated ATRA-mediated response in the co-exposure with a range of ATRA concentrations. Another structural N-PAH variant, 1,7-phenanthroline, downregulated ATRA-mediated response at most of tested ATRA concentrations and exposure times. Interesting concentration-dependent biphasic effects (i.e. downregulation with subsequent up-regulation to control levels) were observed at co-exposures of ATRA and parent PAH phenanthrene. Non significant Oct-4 modulation in co-exposure with ATRA was observed at compounds, which potentiated ATRA-mediated effects in the reporter gene assay. On the other hand, 1,7-phenanthroline and phenanthrene significantly suppressed Oct-4 levels in higher tested concentrations. Our results further extend the knowledge of PAH and N-PAH in vitro effects and indicate that these environmental toxicants may have influence on differentiation process and embryonic development by interfering with ATRA signaling and by modulating levels of Oct-4.

Ecotoxicity and genotoxicity of widely used veterinary antimicrobials oxytetracycline and flumequine was studied with six model organisms (Vibrio fischeri, Pseudomonas putida, Pseudokirchneriella subcapitata, Lemna minor, Daphnia magna, Escherichia coli). Overall median effective concentration (EC50) values ranged from 0.22 mg/L to 86 mg/L. Pseudomonas putida was the most sensitive organism (EC50 values for 16-h growth inhibition were 0.22 and 0.82 mg/L for oxytetracycline and flumequine, respectively), followed by duckweed Lemna minor (7-d growth inhibition, EC50 2.1 and 3.0 mg/L) and green alga Pseudokirchneriella subcapitata (4-d growth inhibition, EC50 3.1 and 2.6 mg/L). The least sensitive organism was Daphnia magna (48-h immobilization, lowest-observed-effect concentration [LOEC] of oxytetracycline of 400 mg/L). Oxytetracycline showed limited genotoxicity (SOS-chromotest with Escherichia coli, minimal genotoxic concentration of 500 mg/L), and flumequine was genotoxic at 0.25 mg/L. Based on the reported measured concentrations (MECs) and predicted no-effect concentrations (PNECs), oxytetracycline may be concluded to be of ecotoxicological concern (calculated risk quotient = 8), whereas flumequine seems to represent lower risk.

Cylindrospermopsin (CYN) is a newly emerging carcinogenic alkaloid originally identified in tropical cyanobacteria Cylindrospermopsis raciborskii . Some recent studies reported production of this toxin also by other cyanobacteria in European countries. Here we report comparison of the immunochemical ELISA determination with the liquid chromatography/mass spectrometry (LC/MS) method for CYN analyses in the samples from the Czech Republic, Central Europe. We have analysed 56 samples of raw lake waters collected from 32 localities. CYN was found at 3 localities with Aphanizomenon sp. sub-dominated water blooms. Results of the ELISA and LC/MS showed good qualitative agreement. However, concentrations determined by ELISA (0.4–4 μg/L) were systematically higher than concentrations determined by LC/MS (0.01–0.3 μg/L). Differences between methods could be attributed to matrix effects (both in ELISA and LC/MS) or ELISA cross-reactivity with other unidentified derivatives of CYN. Our study demonstrates for the first time occurrence of CYN in the Central Europe. With respect to its suspected carcinogenicity, further research on distribution, concentrations and risks of this cyanobacterial toxin is needed.

Toxicity and liver tumor promotion of cyanotoxins microcystins have been extensively studied. However, recent studies document that other metabolites present in the complex cyanobacterial water blooms may also have adverse health effects. In this study we used rat liver epithelial stem-like cells (WB-F344) to examine the effects of cyanobacterial extracts on two established markers of tumor promotion, inhibition of gap-junctional intercellular communication (GJIC) and activation of mitogen-activated protein kinases (MAPKs) – ERK1/2. Extracts of cyanobacteria (laboratory cultures of Microcystis aeruginosa and Aphanizomenon flos-aquae and water blooms dominated by these species) inhibited GJIC and activated MAPKs in a dose-dependent manner (effective concentrations ranging 0.5 - 5 mg d.w./mL). Effects were independent of the microcystin content and the strongest responses were elicited by the extracts of Aphanizomenon sp. Neither pure microcystin-LR nor cylindrospermopsin inhibited GJIC or activated MAPKs. Modulations of GJIC and MAPKs appeared to be specific to cyanobacterial extracts since extracts from green alga Chlamydomonas reinhardtii, heterotrophic bacterium Klebsiella terrigena, and isolated bacterial lipopolysaccharides had no comparable effects. Our study provides the first evidence on the existence of unknown cyanobacterial toxic metabolites that affect in vitro biomarkers of tumor promotion, i.e. inhibition of GJIC and activation of MAPKs.

N-heterocyclic polycyclic aromatic hydrocarbons (N-PAHs) belong among newly identified classes of environmental pollutants with relatively high toxic potential. N-PAHs have been detected in air, soil, marine environments, and freshwater sediments. The N-PAHs are present at lower concentrations than their nonsubstituted analogues but their greater solubility would lead to greater bioavailibity and potential for toxic effects. Here we present results of acute and chronic toxicity in traditional aquatic invertebrate ecotoxicological model (Daphnia magna) along with assessment of biochemical responses. Studied biomarkers in D. magna exposed to N-heterocyclic derivatives included glutathione levels and activities of detoxication and antioxidative enzymes glutathione S-transferase and glutathione peroxidase. Phenanthrene and 1,10-phenathroline were the most toxic of all tested compounds (EC50 < 6 microM after 48 h exposure) and all tested N-PAHs suppressed reproduction of Daphnia magna. The data suggest that N-PAHs can induce oxidative stress in D. magna. The significant decline of glutathione content was found in animals treated with acridine, 1,10-phenanthroline, benzo(h)quinoline, phenantridine, and phenazine. Significant decrease of GPx activities relative to controls was found for all tested compounds except of phenanthrene and phenazine. Activities of GST increased after exposure to phenanthridine, phenazine, and benzo(h)quinoline, and declined in D. magna treated with phenanthrene (significant at one concentration) or anthracene (not significant). Our results confirmed significant acute as well as chronic toxicities of N-PAHs as well as potential of biochemical parameters to be used as early warning signals of toxicity in Daphnia magna.

Humic substances (HS) are ubiquitous in the environment. However, some studies indicate that HS could induce direct adverse effects on human health and hormone-like effects in fish, amphibians, and invertebrates. In this study we investigated a possible biochemical mechanism of HS toxicity via acti- vation of the aryl hydrocarbon receptor (AhR). AhR mediates the toxic and biological effects of environ- mental contaminants such as 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD), but a number of structurally diverse compounds has also been found to activate AhR. Alkali solutions of humic acids (HA) were pre- pared, and subsequently, lipophilic compounds (including parts of HA) were extracted by liquid-liquid extraction with hexane/dichloromethane. Organic extract of HA was further treated with sulfuric acid to study the role of possible trace persistent contaminants. In vitro dioxin-like activities of obtained extract and HA solutions have been evaluated using H4IIE.luc cells by determining the ethoxyresorufin-O-deethy- lase (EROD) activity and induction of AhR-dependent reporter luciferase. Traces of nonpersistent residues in HA with known AhR activity were identified and quantified by GC-MS. Our results show that an alkali solution as well as organic extract of HA were active in both EROD and luciferase assays, while H2SO4- treated extract activity was negligible. Only nonsignificant levels of AhR-inducing contaminants (PAHs and PCBs) were found in the HA samples. Our results indicate that HA or their fragments can elicit significant inductions of AhR-mediated effects in vitro. To our best knowledge, this study is the first in providing direct evidence of dioxin-like effects of HA. Humic substances (HS) were for decades regarded as inert in the ecosystems with respect to their possible toxicity. However, HS have been recently shown to elicit various adverse effects generally attributed to xenobiotics. In our study, we used MVLN and H4IIE-luc cell lines stably transfected with luciferase gene under control of estrogen receptor (ER) and Ah receptor (AhR; receptor connected with so-called dioxin-like toxicity) for assessment of anti/estrogenic and AhR-mediated effects of 12 commercially available humic substances. Out of those, five humic acids were shown to induce AhR-mediated activity with relative potencies related to TCDD 2.6 x 10(-8)-7.4 x 10(-8). Organic extracts of HS solutions also elicited high activities what means that lipophilic molecules are responsible for a great part of effect. However, relatively high activity remaining in extracted solution suggests also presence of polar AhR-agonists. Contribution of persistent organic compounds to the observed effects was ruled out by H(2)SO(4) treatment. Eight out of twelve HS elicited significant antiestrogenic effects with IC(50) ranging from 40 to 164 mg l(-1). The possible explanations of the antiestrogenic effect include sorption of 17-beta-estradiol (E2) on HS, changes in membrane permeability for E2 or another specific mechanism.

Contribution on microcystin variant analysis by capillary electrochromatography (CEC) with easily affordable spectrophotometric detection is presented. Two types of reversed-phase capillary columns formed by inorganic or organic polymer monoliths were prepared for this purpose. The analyses were performed isocratically by means of tris(hydroxymethyl) aminomethane (TRIS) buffers of mildly alkaline pH containing 30% (v/v) acetonitrile as the mobile phases. The samples were injected electrokinetically and the analyses were done at the same separation field strength of 500 V/cm. Microcystins were detected at 238 nm. Although both column types differ not only in monolith quality (inorganic versus organic) but also in the length of the aliphatic moiety (C8 versus C12) similar results were achieved. The on-column preconcentration as the encouraging prospect of electrochromatographic technique was also tested. Consequently 5% of column volume was injected in contrast with 0.5% at standard injection scheme resulting in the six times enrichment of the low concentrated cyanobacterial extract at the top of the separation column. From these preliminary results can be seen that the CEC method is fully applicable for rapid microcystin screening.

Polycyclic aromatic hydrocarbons (PAHs) are a broad class of ubiquitous environmental pollutants with known or suspected carcinogenic properties. Tumor promotion is a cell-proliferative step of cancer that requires the removal of cells from growth suppression via the inhibition of gap-junctional intercellular communication (GJIC). Inhibition of GJIC measured with an in vitro WB-F344 rat liver epithelial cell system was used to assess the relative potencies of 13 PAHs suggested by the U.S. Environmental Protection Agency (EPA) as the principal contaminants and 22 other PAHs, most of them identified in environmental samples. Maximal inhibition of GJIC was detected after 30 min of exposure, followed by a recovery in intercellular communication after an additional 30 min of exposure, suggesting a transient character of inhibition. Although microM concentrations of PAHs were required to reach the inhibition level equal to the model tumor promoter phorbol 12-myristate 13-acetate (IC50 = 8 nM), 12 of the PAHs under study were found to be strong inhibitors of GJIC (strongest effects were observed with fluoranthene, picene, 5-methylchrysene and nine additional PAHs). The other nine PAHs, including benzo[a]pyrene, inhibited GJIC only up to 50-75% of the control level. Interestingly, several high molecular weight PAHs with known strong carcinogenic properties possessed only weak (dibenzopyrenes) or no inhibition potency (dibenzofluoranthenes, naphtho[2,3-a]pyrene and benzo[a]perylene). Based on the IC50 values related to the reference PAH benzo[a]pyrene, we suggested arbitrary values of inhibition equivalency factors (GJIC-IEFs) ranging from 0 (noninhibiting PAHs) to 10.0 (strongest inhibitors), suitable for the purposes of environmental risk assessment.
