In this work, a modification method of H3PO4 plus H2O2 (PHP) was introduced to targetedly form abundant oxygenated functional groups (OFGs) on biochar, and methylene blue (MB) was employed as a model pollutant for adsorption to reflect the modification performance. Results indicated that parent biochars, especially derived from lower temperatures, substantially underwent oxidative modification by PHP, and OFGs were targetedly produced. Correspondingly, approximately 21.5-fold MB adsorption capacity was achieved by PHP-modified biochar comparing with its parent biochar. To evaluate the compatibility of PHP-modification, coefficient of variation (CV) based on MB adsorption capacity by the biochar from various precursors was calculated, in which the CV of PHP-modified biochars was 0.0038 comparing to 0.64 of the corresponding parent biochars. These results suggested that the PHP method displayed the excellent feedstock compatibility on biochar modification. The maximum MB adsorption capacity was 454.1 mg/g when the H3PO4 and H2O2 fraction in PHP were 65.2% and 7.0%; the modification was further intensified by promoting temperature and duration. Besides, average 94.5% H3PO4 was recovered after 10-batch modification, implying 1.0 kg H3PO4 (85%) in PHP can maximally modify 2.37 kg biochar. Overall, this work offered a novel method to tailor biochar towards OFGs-rich surface for efficient adsorption.

Triphenylamine (TPA) derivatives have been widely used as useful building blocks for diverse functional materials because of their excellent redox activity. Most of the molecular structures of TPA-based organic functional materials contain 4-anisyl groups, which on one hand could reduce their oxidation potential and on the other hand significantly delocalize the spin density of the resultant TPA radical cation species and enhance their stability. However, molecular-level investigation of the redox behavior of triphenylamines consisting of 4-anisyl group and the electronic structures of their radical cation species has not been reported in the literature. Herein, we design a series of triphenylamines consisting of one, two, or three 3,5-di-tert-butyl-4-anisyl groups and investigate their redox behaviors and corresponding radical cation species. We disclose that the resonance hybrid and steric protection could both contribute to the stability of triphenylamine radical cations. Moreover, further oxidation leads to an unexpected oxidative demethylation. The findings in this work may reveal new insights for the understanding of the unique redox properties of 4-anisyl substituted triphenylamines.

BACKGROUND Curcumin has become increasingly popular in functional foods and beverages field as a result of its high biological activity. Nevertheless, the application of curcumin is usually limited by its poor water solubility, low absorption, rapid metabolism and instability. Accordingly, the development of an appropriate wall material is crucial for its effective use. In the present study, curcumin-octenyl succinic anhydride modified pullulan (Cur-OSAP) micelles were successfully prepared by an anti-solvent co-precipitation method. RESULTS Octenyl succinic anhydride modified pullulan (OSAP) micelles exhibited the highest encapsulation efficiency (57.31%) and loading capacity (5.73%) of curcumin when the mass ratio of OSAP to curcumin was 10:1 and the degree of substitution of OSAP was 0.0469, at which point Cur-OSAP micelles formed via hydrogen binding and hydrophobic interactions, as confirmed by Fourier transform infrared and fluorescence techniques. The transmission electron microscopy results showed that the Cur-OSAP micelles were roughly spherical in shape with diameters in the approximate range 30-60 nm. CONCLUSION The encapsulation of OSAP greatly improved photostability and sustained release properties of curcumin in Cur-OSAP micelles. These findings suggest that OSAP can be used as a carrier to encapsulate and protect hydrophobic food ingredients. © 2021 Society of Chemical Industry.

SnO2 films as a promising electron transport layer (ETL) have been widely used in planar-type perovskite solar cells to achieve an impressive improvement in the conversion efficiency. However, compared with a mesoporous ETL, the interfacial charge carrier transfer of the SnO2 ETL is severely limited due to the issues of oxygen vacancy defects and crystal lattice mismatch between SnO2 and the perovskite, which generally leads to the growth of randomly stacked and porous perovskite layers and subsequently impacts the charge transport and transfer properties. In this work, we developed a facile approach by inducing a bifunctional molecule, β-alanine, into the SnO2 ETL, which can serve as a bridge to modulate the interfacial charge transfer and the perovskite crystallization kinetics. Benefited by the interfacial β-alanine, we grew a highly orientational perovskite layer that exhibited superior charge transport properties. Meanwhile, the β-alanine caused an intimate connection between the perovskite and SnO2 to enhance the interfacial charge transfer. As a result, the power conversion efficiency (PCE) of the β-alanine-modified device achieved a much-improved value of 19.67% and showed high reproducibility. This work provides a way for developing a high-performance ETL toward the scalable fabrication of highly efficient PSCs.

Hyperectumine (1), the first C19 benzylisoquinoline alkaloid with a complicated ring system, was isolated from Hypecoum erectum and structurally characterized. Its biosynthetic origin should involve a hybrid pattern of C8 + C8 + C1 + C2, from which a C17 benzylisoquinoline alkaloid might be further attacked by a malonamic acid and undergo decarboxylation and cyclization to produce 1. Compound (-)-1 exhibited moderate anti-inflammatory activity via suppression of LPS-activated inflammatory mediators in RAW 264.7 macrophage cells.

Purpose Endothelial dysfunction and the subsequent decrease in endothelium-dependent vascular relaxation of small arteries are major features of hypertension. Artemisinin, a well-known antimalarial drug, has been shown to exert protecting roles against endothelial cell injury in cardiac and pulmonary vascular diseases. The current study aimed to investigate the effects of artemisinin on endothelium-dependent vascular relaxation and arterial blood pressure, as well as the potential signalling pathways in spontaneously hypertensive rats (SHRs). Methods In this study, acetylcholine (ACh)-induced dose-dependent relaxation assays were performed to evaluate vascular endothelial function after treatment with artemisinin. Artemisinin was administered to the rats by intravenous injection or to arteries by incubation for the acute exposure experiments, and it was administered to rats by intraperitoneal injection for 28 days for the chronic experiments. Results Both acute and chronic administration of artemisinin decreased the heart rate and improved ACh-induced endothelium-dependent relaxation but negligibly affected the arterial blood pressure in SHRs. Incubation with artemisinin decreased basal vascular tension, NAD(P)H oxidase activity and reactive oxygen species (ROS) levels, but it also increased endothelial nitric oxide (NO) synthase (eNOS) activity and NO levels in the mesenteric artery, coronary artery, and pulmonary artery of SHRs. Artemisinin chronic administration to SHRs increased the protein expression of eNOS and decreased the protein expression of the NAD(P)H oxidase subunits NOX-2 and NOX-4 in the mesenteric artery. Conclusion These results indicate that treatment with artemisinin has beneficial effects on reducing the heart rate and basal vascular tension and improving endothelium-dependent vascular relaxation in hypertension, which might occur by increasing eNOS activation and NO release and inhibiting NAD(P)H oxidase derived ROS production., Graphical Abstract

The hydrogel wound dressings are mostly sheet-like and do not possess adhesive properties, so it is often difficult for them to adhere well to the skin tissues and fit completely with the irregularly shaped wounds or deep wounds, thereby affecting their therapeutic effects. Herein, the chitosan derivative-based mussel-inspired hydrogels have been developed to overcome these obstacles. Specifically, the principle of adhesion of marine mussel was imitated to synthesize catechol-modified chitosan (CMC), which was potentially used as the raw material to prepare the mussel-inspired hydrogels, that can contribute to the excellent adhesive property and make the hydrogels adhere well to the wound sites. The o-diphenyl groups in CMC could be easily oxidized to form o-diquinone to further form a covalent binding with amino groups (–NH2), or can directly form coordination binding with Fe3+.This strong covalent and coordination binding causes the hydrogels to possess a complex spatial structure, which facilitates the hydrogels to display good swelling properties, drug loading and release properties. Furthermore, the total saponins of Panax notoginseng were loaded in the hydrogels and the therapeutic effects of drug-loaded hydrogel were investigated in the model of full-thickness skin injury in mice. The wound healing rate was observed to be as high as 92.5% without exhibiting any scarring. Overall, our findings provided a novel strategy for the preparation of hydrogels for therapeutic applications.

A pair of two-dimensional (2D) isostructural coordination polymers (CPs), {[Co(2,3-qldc)(H2O)]} n (1) and {[Mn(2,3-qldc)(H2O)]} n (2), where 2,3-H2qldc = quinoline-2,3-dicarboxylic acid, were hydrothermally synthesized and characterized through IR spectroscopy, elemental and thermal analysis, power X-ray diffraction, and single-crystal X-ray diffraction. The results have revealed that the fully deprotonated 2,3-H2qldc ligand connects the Co(II)/Mn(II) atoms with a μ 3-bridge to form a square-wave 2D network, which is further extended into 3D stacks through O–H···O, C–H···O hydrogen bonds and π···π stacking interactions. Topologically, 1 or 2 can be simplified as a 4-connected sql type with a Schläfli symbol {44·62} and a Shubnikov tetragonal plane net, or as a 3-connected fes type with a Schläfli symbol {4·82} and a Shubnikov plane net. The thermal stability and the solid state fluorescence properties of 1 and 2 were investigated.

In this work, novel antibacterial cellulose diacetate/poly(ethylenimine)/hyaluronic acid (CDA/PEI/HA) composite three-dimension (3D) scaffolds were prepared through electrospinning and post-processing. Firstly, CDA/PEI 2D composite nanofiber membranes were prepared by electrospinning, and then the CDA/PEI/HA 3D composite scaffolds were fabricated by post-processing and freeze-drying of CDA/PEI membranes. In particular, HA was added to improve the biocompatibility of composite scaffolds. Compared with CDA/PEI scaffolds, CDA/PEI/HA composite scaffolds showed higher water absorbing rate, higher water retention rate and higher mechanical strength. Moreover, CDA/PEI/HA composite scaffolds with abundant positive charge were benefit for improving antibacterial activity. CDA/PEI/HA scaffolds with biology function of HA were benefit for improving hemocompatibility and cell proliferation of CDA/PEI scaffolds. In summary, antibacterial CDA/PEI/HA scaffolds could protect wound from bacterial infection, improve cellular behavior and accelerate wound healing. Thus, CDA/PEI/HA scaffolds could be potential application for wound dressing.

A new six coordinated tetra-nuclear macrocyclic Zn(II) complex, ZnL4(Phen)2 (1) (HL= 3-bromo-2-hydroxybenzaldehyde-pyridine-2-carbohydrazone, Phen = 1,10-phenanthroline) has been synthesized by the self-assembly of 3-bromo-2-hydroxybenzaldehyde-pyridine-2-carbohydrazone, Zn(CH3COO)2•2H2O, NaOH and 1,10-phenanthroline in water/ethanol (v:v = 1:3) solution. Complex 1 was characterized by elemental analysis, infra red (IR), and single-crystal X-ray diffraction (XRD) analysis. The results show that Zn1 and Zn1b ions are six-coordinated with a distorted octahedral geometric configuration by four O atoms of two different L ligands and two N atoms of two different L ligands, Zn1a and Zn1c ions are also six-coordinated with a distorted octahedral geometric configuration by two N atoms of two different L ligands, two N atoms of Phen ligands and two O atoms of two different L ligands. Complex (1) forms 3D network structure by the - interaction. The selective oxidation reactions of benzyl alcohols catalyzed by complex (1) was investigated. The highest benzyl alcohol conversion and benzaldehyde selectivity were obtained at 100 °C for 4 h under 5 bar of O2. Copyright © 2021 by Authors, Published by BCREC Group. Background Previous studies have demonstrated the preclinical pharmacological and toxicological consistency, and clinical pharmacokinetic equivalence of bevacizumab biosimilar LY01008 with reference bevacizumab (Avastin). This randomized controlled trial aimed to compare the efficacy and safety of LY01008 with Avastin in first‐line treatment of Chinese patients with advanced or recurrent non‐squamous non‐small cell lung cancer (NSCLC). Methods Stage IIIB‐IV NSCLC patients with evaluable lesions, good physical status, and adequate organ functions from 67 centers across China were randomized in a ratio of 1:1 to receive LY01008 or Avastin 15 mg/kg intravenously in combination with paclitaxel/carboplatin (combined treatment) for 4‐6 cycles, followed by maintenance monotherapy with LY01008 until disease progression, intolerable toxicity, or death. The primary endpoint was objective response rate (ORR) in accordance with Response Evaluation Criteria in Solid Tumors (RECIST) version 1.1 confirmed by independent radiological review committees (IRRC). Secondary endpoints included disease control rate (DCR), duration of response (DoR), progression‐free survival (PFS), overall survival (OS), and safety. This study was registered in ClinicalTrials.gov (NCT03533127). Results Between December 15th, 2017, and May 15th, 2019, a total of 649 patients were randomized to the LY01008 (n = 324) or Avastin (n = 325) group. As of September 25th, 2019 for primary endpoint analysis, 589 patients received ORR evaluation, with a median number of combined treatment cycles of 5 (range 1‐6) and median duration of treatment of 3.0 (range 0.0‐5.1) months. ORR of response‐evaluable patients in the LY01008 and Avastin groups were 48.5% and 53.0%, respectively. The stratified ORR ratio was 0.91 (90% CI 0.80‐1.04, within the prespecified equivalence margin of 0.75‐1.33). Up to May 15th, 2020, with a median follow‐up of 13.6 (range 0.8‐28.4) months, no notable differences in DCR, median DoR, median PFS, median OS, and 1‐year OS rate were observed between the LY01008 and Avastin groups. There were no clinically meaningful differences in safety and immunogenicity across treatment groups. Conclusions LY01008 demonstrated similarity to Avastin in terms of efficacy and safety in Chinese patients with advanced or recurrent non‐squamous NSCLC. LY01008 combined with paclitaxel/carboplatin is expected to become a new treatment option for unresectable, metastatic, or recurrent non‐squamous NSCLC patients in the first‐line setting., This study demonstrated similarity between LY01008 and reference bevacizumab (Avastin) in terms of efficacy, safety, and immunogenicity in combination with paclitaxel and carboplatin as first‐line treatment in Chinese patients with advanced non‐squamous NSCLC.

Temperature-responsive resistance transition behaviors of the melamine sponges wrapped with different graphene oxide derivatives (i.e. nanoribbon, wide-ribbon and sheet) were investigated. Melamine sponge composites coated by three types of GO derivatives were prepared by a simple dip-coating approach. All these composites show good mechanical flexibility and reliability (almost unchanged compressive stress at 70 % strain after 100 cycles), high hydrophobicity (water contact angle >120°), excellent flame resistance (self-extinguishing) and structural stability even after burning, which was used to construct the resistance-based fire alarm/warning sensor. Notably, the different resistance response behaviors of such sensors are strongly dependent on the GO size and network formed on the MF skeleton surface. Typically, at a fixed high temperature of ∼350 ℃, the three fire alarm sensors show different response time (to trigger the alarm light) of 6.3, 8.4 and 11.1 s for nanoribbon, wide-ribbon and sheet at the same concentration, respectively. The structural observation and chemical analysis demonstrated that the discrepancy of temperature-responsive resistance transition behaviors of various GO derivatives was strongly determined by their different thermal reduction degrees during the high-temperature or flame treating process. This work offers a design and development for construction of smart fire alarm device for potential fire prevention and safety applications.

Background:. With the ongoing worldwide coronavirus disease 2019 (COVID-19) pandemic, an increasing number of viral variants are being identified, which poses a challenge for nucleic acid-based diagnostic tests. Rapid tests, such as real-time reverse transcription-polymerase chain reaction (rRT-PCR), play an important role in monitoring COVID-19 infection and controlling its spread. However, the changes in the genotypes of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) variants may result in decreased sensitivity of the rRT-PCR assay and it is necessary to monitor the mutations in primers and probes of SARS-CoV-2 detection over time. Methods:. We developed two rRT-PCR assays to detect the RNA-dependent RNA polymerase (RdRp) and nucleocapsid (N) genes of SARS-CoV-2. We evaluated these assays together with our previously published assays targeting the ORF1ab and N genes for the detection and confirmation of SARS-CoV-2 and its variants of concern (VOCs). In addition, we also developed two rRT-PCR assays (S484K and S501Y) targeting the spike gene, which when combined with the open reading frames (ORF)1ab assay, respectively, to form duplex rRT-PCR assays, were able to detect SARS-CoV-2 VOCs (lineages B.1.351 and B.1.1.7). Results:. Using a SARS-CoV-2 stock with predetermined genomic copies as a standard, the detection limit of both assays targeting RdRp and N was five copies/reaction. Furthermore, no cross-reactions with six others human CoVs (229E, OC43, NL63, HKU1, severe acute respiratory syndrome coronavirus and Middle East respiratory syndrome coronavirus) were observed using these assays. In addition, the S484K and S501Y assays were combined with the ORF1ab assay, respectively. Conclusions:. Four rRT-PCR assays (RdRp, N, S484K, and S501Y) were used to detect SARS-CoV-2 variants, and these assays were shown to be effective in screening for multiple virus strains.

Chlorin e6 (Ce6) is a promising photosensitizer for tumor photodynamic therapy (PDT). However, the efficacy of Ce6 PDT is limited by Ce6's poor water solubility, rapid blood clearance, and inadequate accumulation in the tumor tissue. This problem is tackled in this work, wherein functionalized superparamagnetic iron oxide nanoparticles (IO-NPs) were used as carriers to deliver Ce6 to melanoma. The IO-NPs were coated with polyglycerol (PG) to afford good aqueous solubility. The chemotherapeutic agent doxorubicin (DOX) was attached to the PG coating via the hydrazone bond to afford affinity to the cell membrane and thereby promote the cell uptake. The hydrophobic nature of DOX also induced the aggregation of IO-NPs to form nanoclusters. Ce6 was then loaded onto the IO nanoclusters through physical adsorption and coordination with surface iron atoms, yielding the final composites IO-PG-DOX-Ce6. In vitro experiments showed that IO-PG-DOX-Ce6 markedly increased Ce6 uptake in mouse melanoma cells, leading to much-enhanced photocytotoxicity characterized by intensified reactive oxygen species production, loss of viability, DNA damage, and stimulation of tumor cell immunogenicity. In vivo experiments corroborated the in vitro findings and demonstrated prolonged blood clearance of IO-PG-DOX-Ce6. Importantly, IO-PG-DOX-Ce6 markedly increased the Ce6 distribution and retention in mouse subcutaneous melanoma grafts and significantly improved the efficacy of Ce6-mediated PDT. No apparent vital organ damage was observed at the same time. In conclusion, the IO-PG-DOX NPs provide a simple and safe delivery platform for efficient tumor enrichment of Ce6, thereby enhancing antimelanoma PDT.

Carbon nitride (CN) polymer is considered as a potential material for photocatalysis, but surfers from the low photoconversion efficiency due to its high defect density. Although the defects can be limited through the crystallization of CN, the photocatalytic activity of single-component photocatalyst is still restricted by the recombination of photoexcited carriers. In our work, the ordered Schottky heterojunction of heptazine-based crystalline carbon nitride (HCN) and Ti3C2 MXene was successfully prepared through ionothermal method. The HCN/Ti3C2 composites exhibit higher photocatalytic performance than pristine HCN. Particularly, the HCNT20 sample exhibits the highest photocatalytic H2 evolution activity, which is about 8 and 2 times higher than that of bulk CN and pristine HCN, respectively. The results of charge transfer dynamics and DFT calculation reveal that the photocatalytic enhancement mechanism is primarily attributed to the synergistic effects of crystallization of CN, excellent conductivity of Ti3C2 and well-constructed Schottky heterojunction.

Bronsted acid-catalyzed direct C(sp2)-H alkylation of N-heteroaromatics with cyclic ethers via a cross-dehydrogenative coupling reaction in the presence of benzoyl peroxide was developed. This methodology successfully provided an easy access to a variety of alkyl-substituted quinoxaline, benzoimidazole, pyrazine, pyrimidine, quinoline, isoquinoline, and pyridine derivatives in up to 94% yield under metal-free conditions.

Background. Ulcerative colitis (UC) is a chronic nonspecific inflammatory disease of the colon and rectum. Recent studies found that berberine had effects on inflammatory diseases and immune diseases. Methods. The PharmMapper database was used to predict the berberine potential target and GeneCards database and OMIM database were utilized to collect UC genes. The Cytoscape software was used to construct and analyze the networks and DAVID was utilized to perform enrichment analysis. Then, animal experiments were performed to validate the prediction results. The experimental rats were randomly divided into normal group (control group), model group, and berberine group. The general condition, body weight, gross morphology of colon tissue, and colonic mucosal damage index (CMDI) score were observed. The pathological changes of colon tissue were observed by H&E staining. The levels of serum interleukin-1β (IL-1β), tumor necrosis factor-α (TNF-α), and IL-4 were detected by ELISA. The expressions of IL-1β, TNF-α, and IL-4 protein in colon tissue were detected by immunohistochemistry. Results. A total of 211 Berberine’s potential targets and 210 UC genes were obtained. The enrichment analysis showed that berberine may regulate inflammation, inflammatory cytokines, and their mediated inflammation signal pathways such as inflammatory bowel disease (IBD), rheumatoid arthritis, cytokine-cytokine receptor interaction, TNF, T cell receptor, Toll-like receptor, and JAK/STAT signaling pathway. Yttrium borides have attracted much attention for their superconducting and high chemical stability. However, the fundamental knowledge of the mechanical properties and hardness of yttrium borides is rather lack. Here, we performed a systematic investigation on yttrium borides with various stoichiometries based on an unbiased CALYPSO structure search method and first-principles calculations. A new YB6 compound with R3m hexagonal structure is observed, which is more stable than the experimental synthesized Pm3m phase. The calculated formation enthalpy, elastic constants, and phonon dispersions distinctly show that the R3m-YB6 is energetically, mechanically, and dynamically stable. The density of states and electronic band structure reveal that all the stable yttrium borides are metallic. Based on a semiempirical method, the Vicker hardness of R3m-YB6 is expectant to be 37.0 GPa, indicating it is a potential ultrahard metal. Our results provide insights for future synthesis and design of new type functional materials.

To convert lignin into high-valued carbon materials and understand the lignin structure function, oxidized lignin, a by-product from lignocellulose PHP-pretreatment (phosphoric acid plus hydrogen peroxide), was carbonized by one-step KOH-activation; the physico-chemical characteristics and electrochemical performances of the harvested carbons were also investigated. Results indicated the resultant carbons displayed 3-dimensional hierarchical porous morphology with maximum specific surface area of 3094 m2 g−1 and pore volume of 1.72 cm3 g−1 using 3:1 KOH/lignin ratio for carbonization. Three-electrode determination achieved a specific capacitance of 352.9 F g−1 at a current of 0.5 A g−1, suggesting a superior rate performance of this carbon. Two-electrode determination obtained an excellent energy density of 9.5 W h kg−1 at power density of 25.0 W kg−1. Moreover, 5000 cycles of charge/discharge reached 88.46% retention at 5 A g−1, implying an outstanding cycle stability. Basically, low molecular weight and abundant oxygen-containing functional groups of employed lignin mainly related to the excellent porous morphology and the outstanding electrochemical performances, suggesting the oxidized lignin was an ideal precursor to facilely prepare activated carbon for high-performance supercapacitor. Overall, this work provides a new path to valorize lignin by-product derived from oxidative pretreatment techniques, which can further promote the integrality of lignocellulose biorefinery.

We report herein the asymmetric total synthesis of norzoanthamine using radical reactions as key steps for rapid access to the congested carbocyclic core, which is the major synthetic challenge for most zoanthamine alkaloids. (1) The Ueno-Stork radical cyclization was applied to construct the adjacent quaternary centers at the C-9 and C-22 positions; (2) a Co-catalyzed HAT radical reaction was successfully applied to construct the quaternary center at C-12 via Csp3 -Csp2 bond formation; (3) a Mn-catalyzed HAT radical reaction was used to stereospecifically reduce the tetra-substituted olefin (C13=C18) and install the contiguous stereocenters in proximity to the quaternary center. A one-pot bio-inspired cyclization step was finally applied to forge the unstable bis-amino acetal skeleton. Our approach can precisely control the stereochemistry of seven vicinal stereocenters and effectively construct the highly congested heptacyclic skeleton.

Previous studies have shown that single-frequency microwave radiation can lead to cognitive decline in rats. However, few studies have focused on the combined effects of irradiation with different frequencies of microwaves. Our research aimed to investigate the effects of 1.5 GHz and 4.3 GHz microwave radiation, singly and in combination, on cognitive function and hippocampal tissue structure in rats. A total of 140 male Wistar rats were randomly divided into 4 groups: the S group (sham radiation group), L10 group (10 mW/cm2 1.5 GHz group), C10 group (10 mW/cm2 4.3 GHz band group) and LC10 group (10 mW/cm2 1.5 and 4.3 GHz multi-frequency radiation group). For 1–28 days after microwave radiation, we analyzed the average escape latency for the Morris water maze task, electroencephalograms, change in hippocampal tissue structure and ultrastructure, content of the Nissl body in the hippocampus, and activities of lactate dehydrogenase and succinate dehydrogenase. Compared to the S group, all exposure groups showed varying degrees of learning and memory decline and hippocampal structural damage. The results showed that 1.5 GHz and 4.3 GHz microwave radiation was able to induce cognitive impairment and hippocampal tissue damage in rats and combined radiation with both frequencies caused more serious injuries, but none of these damaging effects varied with microwave frequency.

Chaenomeles speciosa (Sweet) Nakai is a dual-purpose Chinese herbal medicine and functional food favored by minorities in Southwest China, and its fruits are used for the treatment of dyspepsia, dysentery, enteritis, and rheumatism inflammation. Some diseases may be related to microbial infection; however, it is not known how the fruits possess antimicrobial activity. We evaluated the antimicrobial bioctivity of different evaluation extracts of C. speciosa fruits by in vitro and in vivo with colony-forming unit assays, and the strongest bioactive-guided fraction was selected for column chromatography (CC), UHPLC-QTOF-MS/MS, and NMR spectroscopy to confirm the chemical constituents. The most possible antimicrobial mechanism of C. speciosa fruits was explored by metabolomics approach, fluorescence microscopy imaging, and scanning electron microscopy (SEM). Thirty compounds, which were major characteristic ions of the bioactive fraction, were determined precisely. The bioactive fraction could inhibit 18 pathogenic microorganisms, significantly reduced, especially drug-resistant bacteria, compared to ampicillin sodium salt, fluconazole, and berberine chloride form; and the minimum inhibitory concentration (MIC) or minimum fungicidal concentration (MFC) values were in the range of 0.1-1 mg/mL. The compounds 2'-methoxyaucuparin (1) and oleanolic acid (20) not only have antibacterial activity but also may have synergistic effects. Further, the bioactive fraction might inhibit the biofilm formation, enhance immunity, and restore bacterial infection damage in vitro and in vivo to kill microorganisms. The data indicated that C. speciosa fruits' major bioactive fraction enriched with triterpenes, flavonoids, and phenolics could be developed as a functional supplement for individuals to prevent and treat microbial infection.

A series of triphenylamine (TPA) derivatives with various substituent groups were prepared and showed different absorption and fluorescence characteristics due to the substituent effect. On account of the existence of pyridine units, these TPA derivatives exhibited acid-induced tunable multicolor fluorescence emission including white light emission. In addition, acid-induced fluorescence regulation of these compounds has been also realized in the solid state, which enable them to be successfully constructed the stimuli-responsive fluorescent films and fluorescent inks for inkjet printing.

Aggregation of antibodies, especially multimers, is a problem that puzzles scientific and industrial fields, and it is more serious in high concentration antibodies. We found that the high concentration antibody solution with rich multimers has a deeper color, which was confirmed by SEC–HPLC, DLS, and SEC–DLS methods. After that, the reasons for formation of multimers were analyzed. LC–MS was used for complete molecular weight analysis, which showed that the multimer sample could not be ionized, indicating a complex structure of multimers. ICP–MS was carried out to analyze the elements in the multimers, and revealed an important message that iron ion in the multimers was significantly higher. On further analysis of the interaction between iron ions and antibody molecules, the results showed that iron ions (Fe(III)) and antibody monomers irreversibly formed multimers under acidic conditions. This study provided useful insights for optimizing the development of antibody drug substances.

The coordinated utilization of nitrogen (N) and phosphorus (P) is vital for plants to maintain nutrient balance and achieve optimal growth. Previously, we revealed a mechanism by which nitrate induces genes for phosphate utilization; this mechanism depends on NRT1.1B-facilitated degradation of cytoplasmic SPX4, which in turn promotes cytoplasmic-nuclear shuttling of PHR2, the central transcription factor of phosphate signaling, and triggers the nitrate-induced phosphate response (NIPR) and N-P coordinated utilization in rice. In this study, we unveiled a fine-tuning mechanism of NIPR in the nucleus regulated by Highly Induced by Nitrate Gene 1 (HINGE1, also known as RLI1), a MYB-transcription factor closely related to PHR2. RLI1/HINGE1, which is transcriptionally activated by PHR2 under nitrate induction, can directly activate the expression of phosphate starvation-induced genes. More importantly, RLI1/HINGE1 competes with PHR2 for binding to its repressor proteins in the nucleus (SPX proteins), and consequently releases PHR2 to further enhance phosphate response. Therefore, RLI1/HINGE1 amplifies the phosphate response in the nucleus downstream of the cytoplasmic SPX4-PHR2 cascade, thereby enabling fine-tuning of N-P balance when nitrate supply is sufficient.

Hydrogel-based wearable devices have attracted tremendous interest due to their potential applications in electronic skins, soft robotics, and sensors. However, it is still a challenge for hydrogel-based wearable devices to be integrated with high conductivity, a self-healing ability, remoldability, self-adhesiveness, good mechanical strength and high stretchability, good biocompatibility, and stimulus-responsiveness. Herein, multifunctional conductive composite hydrogels were fabricated by a simple one-pot method based on poly(vinyl alcohol) (PVA), sodium alginate (SA), and tannic acid (TA) using borax as a cross-linker. The composite hydrogel network was built by borate ester bonds and hydrogen bonds. The obtained hydrogel exhibited pH- and sugar-responsiveness, high stretchability (780% strain), and fast self-healing performance with healing efficiency (HE) as high as 93.56% without any external stimulus. Additionally, the hydrogel displayed considerable conductive behavior and stable changes of resistance with high sensitivity (gauge factor (GF) = 15.98 at a strain of 780%). The hydrogel was further applied as a strain sensor for monitoring large and tiny human motions with durable stability. Significantly, the healed hydrogel also showed good sensing behavior. This work broadens the avenue for the design and preparation of biocompatible polymer-based hydrogels to promote the application of hydrogel sensors with comfortable wearing feel and high sensitivity.

The design and synthesis of low-cost and efficient bifunctional electrocatalysts for water splitting are critical and challenging. Hereby, a bimetallic phosphide embedded in a N and P co-doped porous carbon (FeCoP2@NPPC) material was synthesized by using sustainable biomass-derived N- and P-containing carbohydrates and non-noble metal salts as precursors. The obtained material exhibits good catalytic activities in hydrogen evolution reaction (HER), oxygen evolution reaction (OER), and overall water splitting. The bimetallic alloy phosphide (FeCoP2) is the active site for electrocatalysis. Theoretical calculation indicates that the sub-layer Fe atoms and top-layer Co atoms in FeCoP2 exhibit a synergistic effect for enhanced electrocatalytic performance. The carbon matrix around the FeCoP2 can prevent the corrosion during the catalytic reactions. The hierarchically porous structure of the FeCoP2@NPPC material can promote the transfer of electrons and electrolyte, and increase the contact area of the active sites and electrolytes. N- and P-containing functionalities improve the wetting and conductivity properties of the porous carbon. Due to the synergistic effects, FeCoP2@NPPC requires a low overpotential of 114 and 150 mV at the current density of 10 mA cm-2 for HER in 0.5 M H2SO4 and 1.0 M KOH, and an overpotential of 236 mV for OER in 1.0 M KOH solution, which are much lower than those of FeP@NPPC and CoP@NPPC. Based on the density functional theory calculation, FeCoP2 yields the smallest Gibbs free energy change of rate-determining step among the samples, which leads to better electrochemical performances. In addition, when FeCoP2@NPPC was used as a bifunctional catalyst in water splitting, the electrolyzer needed a low voltage of 1.60 V to deliver the current density of 10 mA cm-2. Furthermore, this work provides a strategy for preparing sustainable, stable, and highly active electrocatalysts for water splitting.

Alzheimer's disease (AD) is a neurodegenerative disorder disease, disturbing people's normal life. Syringin was mentioned to antagonize Amyloid-β (Aβ)-induced neurotoxicity. However, the action mechanism is still not fully elucidated. This study aimed to explore a molecular mechanism of syringin in defending Aβ-induced neurotoxicity. SK-N-SH and SK-N-BE cells were treated with amyloid β-protein fragment 25-35 (Aβ25-35) to induce cell neurotoxicity. The injury effects were distinguished by assessing cell viability and cell apoptosis using cell counting kit-8 (CCK-8) assay and flow cytometry assay, respectively. The expression of Cleaved-caspase3 (Cleaved-casp3), B cell lymphoma/leukemia-2 (Bcl-2), Bcl-2 associated X protein (Bax) and BH3 interacting domain death agonist (BID) at the protein level was determined by western blot. The expression of miR-124-3p and BID was detected using quantitative real-time polymerase chain reaction (qRT-PCR). The interaction between miR-124-3p and BID was predicted by the online database starBase and confirmed by dual-luciferase reporter assay plus RNA pull-down assay. Aβ25-35 treatment inhibited cell viability and induced cell apoptosis, while the addition of syringin recovered cell viability and suppressed cell apoptosis. MiR-124-3p was significantly downregulated in Aβ25-35-treated SK-N-SH and SK-N-BE cells, and BID was upregulated. Nevertheless, the addition of syringin reversed their expression. BID was a target of miR-124-3p, and its downregulation partly prevented Aβ25-35-induced injuries. Syringin protected against Aβ25-35-induced neurotoxicity by enhancing miR-124-3p expression and weakening BID expression, and syringin strengthened the expression of miR-124-3p to diminish BID level. Syringin ameliorated Aβ25-35-induced neurotoxicity in SK-N-SH and SK-N-BE cells by regulating miR-124-3p/BID pathway, which could be a novel theoretical basis for syringin to treat AD.

The development of novel luminescent materials with multi-stimuli responsive and multi-functional properties is required. In this work, we designed and synthesized a novel AIE-based fluorenone derivative, MBPF. The donor–acceptor–donor (D–A–D) structure promoted the intermolecular hydrogen bonding capacity of the exposed oxygen atom in fluorenone. MBPF displayed three different aggregation morphologies: an amorphous floccule Y-MBPF with yellow-green luminescence, a block crystal O-MBPF with orange luminescence, and a needlelike crystal R-MBPF with red emission. They all exhibited reversible stimuli-response phase transitions to temperature and shear, accompanied by fluorescence switching. The orange crystal O-MBPF was also sensitive to isotropic hydrostatic pressure with prominent and contrasting luminescent conversion. Furthermore, we prepared MBPF nanocrystallites and encapsulated them with DSPE–PEG and the monoclonal antibody cetuximab (C225) to obtain MBPF-C225 NPs, which were biocompatible and photostable. They could be used for specific cancer cell imaging through receptor-mediated endocytosis.

With the increasing interest in polyoxometalates (POMs) materials over the past few decades, scientific researchers have focused their attention on the fruitful utilization of POMs hybrids as efficient catalysts in many organic reactions. In this paper, dipyridine amine (dpa) organic ligand hybrid polyoxomolybdate material was solvothermally synthesized and structurally identified by elemental analysis, X-ray diffraction and infrared spectrum. Structural refinement of the hybrid polyoxomolybdate compound 1 with general formula [Hdpa]4Mo8O26 · 2DMF (DMF = N,N-dimethylformamide) shows that compound 1 presents in the space group P-1 with triclinic system, and is built up from a well-known β-[Mo8O26]4– anionic cluster, four free protonated dpa cations and two free DMF molecules. Remarkably, the β-Mo8O26-based hybrid material was further applied as the catalyst in the conversion of sulfides utilizing hydrogen peroxide (30%) as green terminal oxidant and exhibited efficient catalytic activities for selective oxidation of sulfides.

Diabetic nephropathy (DN), a frequent diabetes complication, has complex pathogenesis. Circular RNAs (circRNAs) circ_0000712 has been reported to be upregulated in kidney tissues and high glucose (HG)-inducted Mesangial cells (MCs). This study is designed to explore the role and mechanism of circ_0000712 in the HG-inducted MCs injury in DN. Circ_0000712, microRNA-879-5p (miR-879-5p), and SRY-Box Transcription Factor 6 (SOX6) levels were detected by real-time quantitative polymerase chain reaction (RT-qPCR). Cell apoptosis was examined by flow cytometry assay. Protein levels of B-cell lymphoma-2 (Bcl-2), Bcl-2 related X protein (Bax), fibronectin (FN), collagen type I (Col. I), collagen type IV (Col. IV), and SOX6 were assessed by western blot assay. Levels of interleukin-1β (IL-1β), interleukin-6 (IL-6), and tumor necrosis factor α (TNF-α) were measured by enzyme-linked immunosorbent assay (ELISA). Reactive oxygen species (ROS) generation, Lactate Dehydrogenase (LDH) activity, and Superoxide Dismutase (SOD) activity were detected by the corresponding kits. The binding relationship between miR-879-5p and circ_0000712 or SOX6 was predicted by starBase and Targetscan, and then verified by a dual-luciferase reporter and RNA Immunoprecipitation (RIP) assays. Circ_0000712 and SOX6 were highly expressed, and miR-879-5p was decreased in db/db DN mice and HG-inducted SV40-MES13 cells. Furthermore, circ_0000712 deficiency repressed HG-caused apoptosis, inflammation, oxidative stress, and fibrosis in SV40-MES13 cells. Mechanically, circ_0000712 could regulate SOX6 expression by sponging miR-879-5p. Circ_0000712 knockdown could hinder HG-inducted SV40-MES13 cell injury through targeting the miR-879-5p/SOX6 axis, implying a possible circRNA-targeted therapy for DN.

Occurrence and flow of hydrocarbons in nanopores are two important issues in the effective exploitation of shale oil reservoirs. In this study, molecular dynamics simulations are employed to investigate the mechanisms about occurrence and flow of octane in slit-shaped quartz nanopores. We show that the occurrence state of octane and, therefore, its flow behavior are profoundly affected by the potential field from quartz walls and adsorption layers if the nanopore width w becomes less than 50 Å. Two main adsorption layers are always formed, adjacent to the walls and independent of w, due to two potential wells generated by the attractive potentials of the walls. Each pair of symmetrical adsorption layers, each of which can be considered as a solid-like surface, forms a confined environment similar to a nano-slit. The attractive potentials from them are found to be the cause for the formation of the adsorption layers between them. The obvious bulk phase of octane is formed in the pore of w = 50 Å due to the wide zero potential barrier induced by the innermost two adsorption layers. The nonlinear dependence of flow rate on pressure gradient shows that Darcy's law fails to describe the flow in the nanopore. The non-Darcy behavior mainly arises from adsorption effects from the walls and the adsorption layers, slippage between octane and walls and between adjacent two adsorption layers, and the molecular exchange between adsorption layers. A modified microscopic model is established to predict the dependence of flow rate on potential field, pressure gradient and w, which is in a good agreement with our simulation results and verified by the dodecane flow through the nanopore. Our work can be of great importance for revealing the mechanisms of occurrence and transport and guiding the estimation and exploitation of shale oil resources.

Zirconia ceramics and carbon-based materials are widely adopted in medical and dental applications due to their excellent biocompatibility and aesthetics. However, fracture toughness of ceramic materials limits their application in clinical dentistry because of the existence of residual stress. In this study, zirconia/graphene oxide (ZrO2-GO) composite ceramics were fabricated by hot-press sintering. Residual stresses developed on the surface of ZrO2-GO composite ceramics were evaluated by X-ray residual stress analysis and indentation techniques. The variation of surface residual stress with GO content was evaluated, and found to be consistent with that of fracture toughness. The generation of residual stress was found to be directly related to fracture toughness. Residual stress calculated by theoretical formula of indentation method was consistent with that measured by X-ray diffraction in line with the content of GO. Based on above results, it is concluded that 0.1–0.15 wt% GO composite ceramics possessed better mechanical properties.

A series of fully conjugated macrocycles Mn (n = 4–7) consisting of N-(3,5-di-tert-butyl-4-methoxyphenyl) substituted carbazole (Cz-Ar) were successfully synthesized. The aryl carbazole and macrocycle M4 can be readily oxidized and the corresponding radical cation species were found to be highly stable. Moreover, macrocycle M5 was found to form 1 : 1 stoichiometric complexes with fullerenes C60 and C70 with association constants as high as (8.38 ± 0.33) × 104 M−1 and (7.64 ± 0.26) × 104 M−1, respectively.

In this study, Cu(OH)F microspheres suppported on a carbon cloth (Cu(OH)F MS/CC) were rapidly synthesized (at 90 V with 20 min) using an atmospheric dielectric barrier discharge microplasma (DBD). As a multifunctional electrochemical sensor for the detection of glucose (Glu), formaldehyde and hydrogen peroxide, it can accurately detect blood glucose levels in actual serum samples and can determine the contents of formaldehyde and hydrogen peroxide in water samples. Furthermore, it shows good sensitivity and selectivity, which confirmed the feasibility of the Cu(OH)F microsphere electrode for electrochemical sensing. This method was not only rapid and mild (at room temperature and atmospheric pressure) but also provided a promising route for the preparation of nanomaterials for electrochemical sensors.

The intestinal barrier is vital for preventing inflammatory bowel disease (IBD). This study aimed to investigate the potential mechanism behind the protective effects of B. dentium N8 on the intestinal barrier using the lipopolysaccharide (LPS)-induced Caco-2 cells model. Our probiotic validation results showed that B. dentium N8 had a higher adhesion ability and a more substantial inhibition effect on Escherichia coli ATCC 25922 adhesion to HT-29 cells. Regarding the epithelial integrity, B. dentium N8 significantly increased the trans-epithelial electrical resistance (TEER) value and decreased the paracellular permeability of Caco-2 cells stimulated by lipopolysaccharide (LPS). In addition, B. dentium N8 significantly increased ZO-1, occludin, and claudin-1 mRNA expression. B. dentium N8 downregulated the mRNA expression level of TLR4 and pro-inflammatory cytokines (TNF-α, IL-1β, IL-6). Furthermore, B. dentium N8 had a better protective effect on the intestinal barrier than that of E7. Comparative genomics of B. dentium N8 and E7 showed B. dentium N8 had the specific genes encoding for adhesion ability and immune system regulation. The findings provide the theoretical basis for B. dentium N8 possessing a protective effect on the intestinal barrier, which indicate that it could be used as a novel therapy for IBD.

A systematic theoretical study of the regulating effect of the substituent position on the photoinduced deactivation process of the benzyluracil systems has been performed based on the high-level static electronic structure calculations and on-the-fly full-dimensional excited-state dynamics simulations. Similarities and differences coexist for the two systems by comparative studies on the photoinduced deactivation process of the 5-benzyluracil (5-BU) and 6-benzyluracil (6-BU) systems. They both obey an S2 → S1 → S0 two-step decay pattern, and the decay coordinates of the S2 → S1 and S1 → S0 processes are mainly driven by the elongation of the bridging bond and the out-of-plane ring deformation motion, respectively. However, the puckering motion occurring at the C2 atom in the uracil fragment dominates the decay pathway of the 5-BU system. On the contrary, the puckering motion at the C5 atom in the benzene fragment mainly drives the decay coordinate of the 6-BU system. Therefore, the substituent position could play significant roles in the deactivation process of the benzyluracil systems. Moreover, the S1 → S0 decay process of the 6-BU system consists of five pathways, possessing a more complex deactivation picture than the 5-BU system. The fitted time scale of the puckering motion is compatible with the experimentally observed lifetimes. This work provides a fundamental understanding of the photophysical and photochemical properties of the benzyluracil systems and can give rational suggestions to further design or regulate the bionic molecular systems.

Health and wellbeing are significantly impaired by alcoholic liver disease (ALD), and although some lactic acid bacteria strains have been shown previously to relieve ALD symptoms, the mechanisms behind these effects are still unclear. Here, the Lieber-DeCarli liquid diet containing alcohol was fed to C57BL/6J mice for 6 weeks to build a chronic alcoholic liver lesion model to study the protective effects and possible mechanisms of Lactobacillus mixture (Lactobacillus plantarum KLDS1.0344 and Lactobacillus acidophilus KLDS1.0901). The results showed that Lactobacillus mixture improved intestinal epithelial permeability and reduced the serum lipopolysaccharide (LPS) levels. Furthermore, Lactobacillus mixture inhibited liver lipid accumulation, oxidative stress, and inflammation by regulating AMPK, Nrf-2, and TLR4/NF-κB pathways. Importantly, the Lactobacillus mixture modulated the gut microbiota, resulting in increased short-chain fatty acid (SCFA) producers and decreased Gram-negative bacteria. Taken together, these findings indicated that the Lactobacillus mixture could positively regulate the gut microbiota, causing increased levels of SCFAs, which inhibited alcohol-induced liver lipid accumulation and oxidative stress through the gut-liver axis. Moreover, following administration of the Lactobacillus mixture, the improvement of intestinal epithelial permeability and the reduction of Gram-negative bacteria led to the decrease of LPS entering the portal vein, thereby inhibiting alcohol-induced liver inflammation.

Chrysene, one of the basic polycyclic aromatic hydrocarbons (PAHs), has been reported to make damages to human health and living environment. Chronic obstructive pulmonary disease (COPD) is a progressive disorder with high morbidity and mortality. To investigate the role of chrysene in the development of COPD, male C57BL/6 mice were exposed to the cigarette smoke (CS) followed with the administration of chrysene. Morphological analyses indicated that chrysene caused earlier and severer pathological changes in CS-exposed mice. Besides, CS-exposed mice with chrysene treatment showed obvious collagen deposition, elevated α-smooth muscle actin (α-SMA) expression and reduced E-cadherin abundance at earlier stage, which suggested the acceleration and aggravation of pulmonary fibrosis. Moreover, quantification of leukocytes and pro-inflammatory cytokines in bronchoalveolar lavage fluid (BALF) and lung tissues implied that chrysene significantly exacerbated the proceeding of inflammation in CS-exposed mice. Furthermore, significantly increased apoptotic rates, augmented expressions of apoptotic related proteins and highly expressed TRPV1 were determined in CS-exposed mice with chrysene treatment, which indicated the association between COPD pathogenesis and TRPV1 channel. In summary, our findings elucidate that chrysene accelerates the development of COPD in a murine model with new molecular mechanisms.

Chronic obstructive pulmonary disease (COPD) is becoming a leading cause of morbidity and mortality in China. In the IMPACT trial, fluticasone furoate[FF]/umeclidinium[UMEC]/vilanterol[VI] single-inhaler triple therapy demonstrated lower rates of moderate/severe exacerbations than dual therapy with FF/VI or UMEC/VI in patients with symptomatic COPD and a history of exacerbations. This analysis investigates the China cohort and its consistency with the overall ITT population. 10,355 patients were randomized 2:2:1 to once-daily FF/UMEC/VI 100/62.5/25 µg, FF/VI 100/25 µg, or UMEC/VI 62.5/25 µg for 52 weeks. Endpoints included: annual rates of exacerbations, time-to-first on-treatment moderate/severe exacerbation and change from baseline in trough forced expiratory volume in 1 s (FEV1) at Week-52. Clinical trial registration is NCT02164513 (CTT116855). 535 patients (5.2%) were from China. Annual on-treatment moderate/severe exacerbation rate was 0.81 with FF/UMEC/VI versus 0.96 with FF/VI (rate ratio: 0.84; 95% confidence interval [CI]: 0.64, 1.11; p = .227) and 0.80 with UMEC/VI (rate ratio: 1.02; 95% CI: 0.72, 1.44; p = .929). Hazard ratio for time-to-first moderate/severe exacerbation was 0.84 (95% CI: 0.63, 1.11; p = .218) for FF/UMEC/VI versus FF/VI and 0.89 (95% CI: 0.62, 1.27; p = .516) versus UMEC/VI. Significant improvements in mean change from baseline in trough FEV1 were observed for FF/UMEC/VI versus FF/VI (treatment difference 137 mL; 95% CI: 86, 188; p p = .050). Health status was improved with FF/UMEC/VI versus both dual therapies. Results were similar to the overall ITT population. No new safety signals were identified. Single-inhaler triple therapy with FF/UMEC/VI versus FF/VI or UMEC/VI reduced the rate and risk of exacerbations, and improved lung function and health status in the China cohort similar to the overall ITT population. Pollution of N-methyl carbamate (NMC) pesticides is threatening the non-target organisms’ survival. Thus, broad-specific antibodies and class-selective immunoassay are demanding for multiple NMCs determination. In this study, we employed a molecular docking-based virtual screening strategy to fast profile antibody spectrum, based on a designed chemical pool containing 17 compounds. A monoclonal antibody (mAb)-6G against carbofuran was used as the objective. The recombinant full-length IgG was successfully expressed to validate the antibody sequences for homology modeling. After docking, we manually categorized the antibody-chemical binding strength into three groups. Non-competitive surface plasmon resonance (SPR) demonstrated the mAb-6G affinitive binding toward five NMCs (carbofuran, isoprocarb, propoxur, carbaryl and carbosulfan), which were classified into strong and moderate binding categories. Antibody binding properties were confirmed again by ic-ELISA and lateral flow immunochromatographic strip. Subsequently, an ultrasensitive indirect competitive fluoromicrosphere-based immunoassay (ic-FMIA) was established with the IC50 (half-maximal inhibitory concentration) values of 0.08-3.37 ng/mL. This portable assay presented a 30~230-fold improved sensitivity than traditional ic-ELISA and was applied in European surface water analysis. Overall, our work provides an efficient platform integrating in-silico and experimental methodologies to accelerate the characterization of hapten-specific antibody binding properties and the development of high-sensitive immunoassays for multi-pollutants monitoring.

Mammalian carboxylesterases (CES) are key enzymes that participate in the hydrolytic metabolism of various endogenous and exogenous substrates. Human carboxylesterase 2A (hCES2A), mainly distributed in the small intestine and colon, plays a significant role in the hydrolysis of many drugs. In this study, 3-arylisoquinolones 3 h [3-(4-(benzyloxy)-3-methoxyphenyl)-7,8-dimethoxyisoquinolin-1(2H)-one] and 4 a [3-(4-(benzyloxy)-3-methoxyphenyl)-4-bromo-7,8-dimethoxyisoquinolin-1(2H)-one] were found to have potent inhibitory effects on hCES2A (IC50 =0.68 μΜ, Ki =0.36 μΜ) and excellent specificity (more than 147.05-fold over hCES1 A). Moreover, 4 a exhibited threefold improved inhibition on intracellular hCES2A in living HepG2 cells relative to 3 h, with an IC50 value of 0.41 μΜ. Results of inhibition kinetics studies and molecular docking simulations demonstrate that both 3 h and 4 a can bind to multiple sites on hCES2A, functioning as mixed inhibitors. Structure-activity relationship analysis revealed that the lactam moiety on the B ring is crucial for specificity towards hCES2A, while a benzyloxy group is optimal for hCES2A inhibitory potency; the introduction of a bromine atom may enhance cell permeability, thereby increasing the intracellular hCES2A inhibitory activity.

QF-036 is an HIV-1 maturation inhibitor in pre-clinical development, and its antiviral activity against a laboratory HIV-1 strain and two drug-resistant strains was determined in the C8166 line. QF-036 was also subjected to absorption, distribution and metabolism (ADM) assessment in vitro, and pharmacokinetic profiles were evaluated in rats and monkeys. The 50% effective concentrations (EC50 ) of QF-036 against the three strains were 20.36 nM, 0.39 μM and 2.11 nM, respectively, demonstrating better antiviral potential than the first-generation antiviral maturation inhibitor bevirimat. QF-036 demonstrated moderate cell permeability, high plasma protein binding ability and good metabolic stability in vitro. After oral QF-036 administration to rats and monkeys, both species exhibited moderate bioavailability, and the plasma drug exposure increased in an approximately dose-proportional manner. When administered orally (30 mg/kg) to monkeys, the QF-036 plasma concentration (Cmax ) peaked at 3671 ng/mL (4.82 μM), 12 to 2410 times higher than the EC50 of laboratory or resistant HIV-1 strains. Moreover, the plasma concentration of QF-036 at 12 hours after administration was 263 ng/mL (0.35 μM), which approximately matched the highest EC50 value of the three test strains. The favourable viral inhibitory activity and pharmacokinetic properties provide critical support for QF-036 as a promising anti-HIV therapeutic candidate.
