The study was conducted to evaluate the effect of Moringa olifera on the growth and gut health of Tilapia (Oreochromis niloticus). The feed having 30% crude protein was prepared as an experimental diet with 4%, 8% and 10% M. olifera leaf supplementation, respectively. The control diet was devoid of M. olifera leaves. The 10 weeks feeding trial was carried out on 60 fish in aquaria. Fish was fed @ 3% of body weight twice a day. Diet with the high level of inclusion of M. olifera leaves significantly increased the growth rate, Survival Rate (SR), Specific Growth Rate (SGR) and Feed Conversion Efficiency (FCE) in all treatment groups compared to the control group. Similarly, Feed Conversion Ratio (FCR) gradually decreased and found highly-significant. To check the gut health of the Tilapia, random samples were selected and dissected. Nutrient agar was used as culture media to check the growth of bacteria. Pour Plate Method was used for viable colonies count by colony counter. Through staining method, the different bacteria such as Escherichia coli, Salmonella, Shigella and Pseudomonas aeruginosa were identify abundantly in the intestine of control diet fish but less number present in treatment diets groups. These results showed that M. olifera leaves up to 10% of dietary protein can be used for Nile tilapia for significant growth and healthy gut microbiota of fish. Resumo O estudo foi conduzido para avaliar o efeito da Moringa olifera no crescimento e saúde intestinal da tilápia (Oreochromis niloticus). A ração com 30% de proteína bruta foi preparada como dieta experimental com 4%, 8% e 10% de suplementação de folhas de M. olifera, respectivamente. A dieta controle foi desprovida de folhas de M. olifera. O ensaio de alimentação de 10 semanas foi realizado em 60 peixes em aquários. O peixe pesava 3% do peso corporal duas vezes ao dia. A dieta com alto nível de inclusão de folhas de M. olifera aumentou significativamente a taxa de crescimento, taxa de sobrevivência (SR), taxa de crescimento de sobrevivência (SGR) e eficiência de conversão alimentar (FCE) em todos os grupos de tratamento em comparação com o grupo de controle. Da mesma forma, a taxa de conversão de alimentação (FCR) diminuiu gradualmente e foi considerada altamente significativa. Para verificar a saúde intestinal da tilápia, amostras aleatórias foram selecionadas e dissecadas. O ágar nutriente foi usado como meio de cultura para verificar o crescimento das bactérias. O método da placa de Verter foi usado para a contagem de colônias viáveis por contador de colônias. Através do método de coloração, diferentes como Escherichia coli, Salmonella, Shigella e Pseudomonas aeruginosa foram identificados abundantemente no intestino de peixes da dieta controle, mas em menor número nos grupos de dieta de tratamento. Esses resultados mostraram que M. olifera deixa até 10% da proteína dietética e pode ser usado para tilápia do Nilo para um crescimento significativo e microbiota intestinal saudável de peixes.

An 8-week feeding trial investigated the optimum dietary vitamin K3 requirements of largemouth bass. A total of 600 healthy fish (12.96 ± 0.07 g) were fed diets containing varying levels of vitamin K3 at 0.78 (K0), 5.80 (K5), 10.82 (K10), 15.84 (K15), and 20.85 (K20) mg/kg with four replicates per level. Results showed that dietary vitamin K3 enhanced growth, with weight gain rate and specific growth rate in K15 trials significantly higher than K0 trials (P 0.05). However, the supplementation of vitamin K3 significantly contributed to calcium accumulation in muscle tissues and vertebrae (P 0.05). The antioxidant capacity was improved with the addition of vitamin K3, which was inconsistent with the changes in malondialdehyde and MK-4 (a form of menaquinone) within liver and serum, suggesting that the toxicity of excessive artificial vitamin K3 may account for this difference. Using regression analysis, the appropriate dietary vitamin K3 levels within a largemouth bass diet was determined to be in the range of 9.93–15.22 mg/kg, which will provide a reference for the preparation of vitamin premix and artificial diet for largemouth bass.

The objective of this work to review about interaction of different food additives with Haemoglobin (Hb) and Myoglobin (Mb) proteins. Food additives are the important components which are added in food to preserve its flavour, color, appearance along with its taste. Mb and Hb interacts with numerous food dyes at varying conditions. In this study we will use various spectroscopic techniques like fluorescence spectroscopy, UV–Visible spectroscopy, Rayleigh light scattering (RLS), fluorescence resonance energy transfer (FRET), circular dichroism (CD)alongwithtransmissionelectronmicroscopy (TEM)andmoleculardockingtocharacterize the binding consequences of Mb and Hb. India is an agricultural country. Nearly 70% of the populations thrive in rural areas, engaged in agriculture making the backbone of our economy. The Trichoderma genus is widely distributed and provides an alternative to chemical pesticides in crops and plants growth. Trichoderma species are free living and commonly found in soil and root ecosystems. Root colonization by Trichoderma also regularly promotes growth and development of plants. They also perform beneficial and significant role in enhancing crop productivity, fight against pathogens, resistant to abiotic stress and increase of nutrients uptake from soil. The present study was undertaken to investigate quantitative and qualitative estimation of phosphate solubilizer by using nineteen isolates of Trichoderma grown on Pikovskaya’s Agar and NBRIP media. It was found that all isolates were capable of differentially utilizing 5 g/L TCP in NBRIP broth. This was indicated by the soluble phosphate concentrations and increase in acidity of growth medium. Out of 19 isolates, all isolates were capable in phosphate solubilization. However, the P- solubilizing efficiency was found higher in case of the isolate number-59 and 89. While isolate number-102 was the strongest and isolate number-23 was the weakest P-solubilizer. Minimum Phosphate solubilization concentration was observed in 3rd day (0%) that simultaneously increased on 7th day (52.63%) followed by 9th day (100%) and remaining all days 13th, 17th and 21st days also observed 100% efficiency.

This work demonstrated the promise of using industrial-grade sugar syrups derived from corn and wood, i.e., high fructose corn syrup (HFCS), glucose corn syrup (GCS) and wood-based sugar (TMP-Bio Sugar), as cheaper and competitive feedstocks for 5-HMF production using niobium phosphate as a heterogeneous solid acid catalyst in a biphasic continuous-flow tubular reactor. 5-HMF yield as high as 53.1% with 100% sugar (glucose and fructose) conversion was obtained from catalytic dehydration of HFCS-90 (containing 90 wt% fructose) at 150 °C, with feed concentration of 200 mg/ml (glucose and fructose) and aqueous to organic phase ratio of 1:5 (v/v). Catalyst stability with time was tested over 20 h of continuous-time on stream, and the reusability of the catalyst was studied after in-situ regeneration of the used catalyst by calcination in the air for removing the deposited humins and coke on the surface of the catalyst particles. The regenerated catalyst showed good activity with almost constant selectivity, although at lower glucose conversion and reduced 5-HMF yield compared to the fresh catalyst, indicating that the in-situ regeneration process could recover a part of the acid sites on the catalyst surface. The produced humins during the reaction were characterized by Fourier transform infrared spectrometry (FT-IR), thermogravimetric analysis (TGA) and elemental analysis (CHNS). The results showed high aromaticity and the presence of a high degree of unsaturated compounds in the structure of humins.

Increasing fuel prices and depletion of conventional sources of energy has promoted research in non-conventional sources like biodiesel from non-edible seeds and industrial wastes. The present work focuses on the elicitation of biodiesel from the seeds of capparis spinosa L. The crude bio-oil extracted from the seeds of capparis spinosa L transesterified into biodiesel using calcium oxide as a heterogeneous catalyst derived from oyster shells. The oyster shell powder is characterized before and after calcination using XRD and the calcinated oyster shell is observed through SEM to analyze the size of the particle and morphology. The produced biodiesel was characterized by GC analysis and the fuel properties of biodiesel such as density, flash point, cloud point, pour point and kinematic viscosity, ash content, carbon residue was analyzed and compared with commercial diesel.

Introduction The incidence of infections caused by aerobic actinomycetes is increasing. Recent changes in taxonomy and the variability in susceptibility patterns among species make necessary a proper identification and antibiotic susceptibility testing. Material and methods Fifty-three strains of aerobic actinomycetes were identified by MALDI-TOF MS using the VITEK MS Mycobacterium/Nocardia kit (bioMerieux, France) in a tertiary hospital in Spain during a six-year period. Antimicrobial susceptibility testing of the isolates was performed using the Sensititre Rapmycoi microdilution panel (Thermo Fisher Scientific, Massachusetts, USA). Results Forty strains of Nocardia spp. were identified in the study, being N. farcinica and N. cyriacigeorgica the most prevalent ones. All isolates were susceptible to linezolid and the resistance to amikacin was only observed in one isolate of Gordonia sputi. Resistance to cotrimoxazole was only found in five isolates. Conclusions Routine identification and antimicrobial susceptibility testing of aerobic actinomycetes is advisable for an efficient identification of species and effective treatment.

Canola meal is a low-value agricultural residue obtained after oil extraction from canola, the utilization of which requires further attention. On the other hand, plastic waste disposal is also another leading issue that creates severe environmental challenges. Supercritical water gasification is considered an environmentally friendly technology to produce hydrogen from plastic residues and organic wastes. This study deals with hydrothermal co-gasification of canola meal and plastic wastes (i.e., low-density polyethylene) while exploring the influence of temperature (375–525°C), residence time (15–60 min) and plastic-to-biomass ratio (0:100, 20:80, 50:50, 80:20 and 100:0) on hydrogen yield. Maximum hydrogen yield (8.1 mmol/g) and total gas yield (17.9 mmol/g) were obtained at optimal temperature and residence time of 525°C and 60 min, respectively. A change in the gas yield with variable plastic-to-biomass ratio showed synergistic effects between both feedstocks. The trend of catalytic performance towards improving hydrogen yield was in the following order: WO3–TiO2 (18.5 mmol/g) > KOH (16.9 mmol/g) > TiO2 (9.5 mmol/g) > ZrO2 (7.8 mmol/g) > WO3–ZrO2 (7.4 mmol/g).

Clove basil (Ocimum gratissimum L.) is an aromatic, perennial herb belonging to the family Lamiaceae. The plant is indigenous to tropical areas especially India and West Africa. In Nigeria, it is found in the Savannah and coastal areas. The whole herbs of the plant contain essential oils and it is cultivated for various purposes. The plant is a rich source of eugenol in its essential oil. It is also used in the preparation of tea and infusions. It is also used in the treatment of fungal infections, fever, cold, and cough. Eugenol (4-allyl-2-methoxy phenol) is a phenolic compound from the class of phenylpropanoids. It is used in the food industry as a preservative, mainly due to its antioxidant property, and flavoring agent. The fresh herb × winter season/environment produced the highest essential oil 2.07 mL/plot with essential oil content 0.41% followed by fresh herb × summer season/environment (M1S2) = 1.68 mL. For the eugenol content, the autumn season/environment was found highly favorable = 74.52% (leaves = 69.024, stem = 74.531, and in inflorescence = 80.012) followed winter season/environment 72.30% (leaves = 71.841, stem 69.389, inflorescence 76.042, mean = 72.42%). These seasons are recommended for harvesting to obtain the optimum benefit for the quality essential oil yield.

Rosemary (Rosmarinus officinalis) originated in the Mediterranean region, but, due to its adaptive nature, can be readily grown across the world. It contains many bioactive phytochemical moieties, especially polyphenols. Rosemary is used as a flavoring agent, spice condiment, and preservative. Recent studies have shown that rosemary natural extracts have bioactive properties such as antifungal, antibacterial, antioxidant, and anti-inflammatory. Rosemary can withstand dry spells, warm conditions and thrive across many soil textures. Carnosic acid is now a well-known compound that is the main component of rosemary. The key components that were extracted from rosemary consisted of cirsimaritin, diosmin, and genkwanin. Rosmarinic acid in the form of carnosol and carnosic acid possesses significant antioxidant properties. Oil analysis revealed the presence of six monodominant and six intermediate chemotypes. Monoterpenes were dominant in rosemary oil composition across all seasons. Bioclimatic factors, including the temperature of the habitat, were found to be a biogenetic factors leading to differentiation in populations. The phenolic contents are reported to vary depending upon processing and agronomical properties. The major active ingredients are lipophilic compounds (carnosic acid and carnosol). Rosemary extracts are also reported to have volatile compounds that confer a unique flavor, color and aroma. Oleanolic acid is endowed with antiviral, antioxidative, and antiproliferative properties vis-a-vis protection against oxidative apoptosis. Ursolic acid helps to prevent cancer. This review provides a comprehensive overview of rosemary species cultivation, bioactive compounds, health benefits, and applications in the food industry. Rosemary was reported to enhance the levels of antioxidant enzymes. Although the current demand in the food industry is to reduce or eliminate the use of synthetic food additives, rosemary oil and extracts can help to meet that demand. This review provides a comprehensive overview of rosemary species cultivation, bioactive compounds, health benefits, and applications in the food industry.

The present study was conducted to estimate the optimum requirement of phosphorus in practical diets for largemouth bass (Micropterus salmoides). Six iso-energetic and iso-nitrogenous diets were formulated with the supplementation of monocalcium phosphate (MCP) of 0, 5, 10, 15, 20 and 25 g/kg in basal diet containing 350 g/kg fish meal, respectively. The digestible phosphorus of the six diets was measured as 5.7, 6.7, 7.8, 8.7, 9.3 and 10.0 g/kg, respectively. Each diet was randomly fed to triplicate groups of 25 juvenile fish (initial body weight, 16.0 ± 0.2 g) for 60 days. The results showed that weight gain increased, and feed conversion ratio decreased with the increase of dietary phosphorus. When dietary MCP exceeded 15 g/kg (digestible phosphorus of 8.7 g/kg), the WG and FCR were maintained at the similar values. The crude ash, phosphorus and calcium contents in whole body, apparent digestibility of phosphorus and protein retention also increased with the increasing phosphorus level in diets. Vertebrae phosphorus and calcium levels and plasma phosphorus showed no further increase when dietary MCP reached 20, 20 and 10 g/kg with digestible phosphorus of 9.3, 9.3 and 7.8 g/kg, respectively. Broken-line analysis indicated that the digestible phosphorus requirement for largemouth bass were 8.9, 8.7 and 9.6 g/kg based on weight gain, feed conversion ratio and vertebral phosphorus content. In conclusion, the supplementation of MCP in a diet containing 350 g/kg fish meal improved the growth and feed utilization of largemouth bass, and the digestible phosphorus requirement was suggested to be 8.7–8.9 g/kg with MCP supplementation of 15.0–15.8 g/kg.

BackgroundEnvironmental water samples are increasingly recognised as an important reservoir of antimicrobial resistance (AMR) genes. Polymerase chain reaction (PCR) and next generation sequencing (NGS) offer a potentially inclusive surveillance platform for a wide range of AMR genes. However, molecular methods are dependent upon the extraction of DNA of high yield and quality. Current options for DNA extraction from complex environmental matrices for downstream molecular applications are either expensive or low yielding. We present here a novel magnetic bead-based DNA extraction method, for the detection of antimicrobial resistance genes (ARGs) from river water in Malawi, named MagnaExtract. MethodsMagnaExtract involves initial filtration of 250ml freshwater, followed by an overnight incubation of the filter in 15ml buffered peptone water (BPW), common procedure in microbiology laboratories. 200{micro}l is then taken for a boil (95{degrees}C) and spin step and mixed with magnetic beads to bind DNA. Following washes with ethanol, the DNA is eluted in nuclease-free water. To determine the effectiveness of this method, 98 freshwater samples were collected from two rivers in Southern Malawi, and DNA was isolated using the MagnaExtract method, two commercial Qiagen (Germany) kits; PowerWater and DNeasy Blood and tissue, alongside a boil and spin of BPW, and a boil and spin from bacterial isolate grown on agar media. All samples were screened with a high-resolution melt (HRM) PCR panel previously validated for the detection of third generation cephalosporin and carbapenem ARGs. We compared the DNA yield obtained using all extraction methods, as well as the identification of each ARG. ResultsDNA yield using MagnaExtract was statistically greater than both boil and spin methods and DNeasy Blood & Tissue (Qiagen, Germany). DNA yield was slightly lower than using PowerWater (Qiagen) but the difference was not statistically significant. MagnaExtract was the only method to identify ARGs in all 98 water samples compared with PowerWater (n=82), DNeasy (n=95) boilate of BPW (n=75) and boilate of bacterial isolate (n=87). The most commonly detected ARG was OXA-48 (n=93). In addition, we found overnight incubation in non-selective enrichment broth (BPW) to promote the growth of bacteria harbouring extended spectrum beta lactamase (ESBL) genes and reduction in the detection of carbapenemase genes. ConclusionThe MagnaExtract approach offers a simple, affordable, high yielding DNA extraction method for the detection of ARGs isolated from river water samples.

In the present study, biocompatible nanofibers containing aqueous extracts from Muscari neglectum (M. neglectum) plants (produced nanofiber) were prepared and their antifungal and cytotoxicity effects were investigated. For this purpose, the extracts obtained from flowers, stem leaves, and fresh onion from M. neglectum were lyophilized at various concentrations. Produced nanofibers were prepared using electrospinning techniques. During the electrospinning process, two auxiliary natural polymers including gelatin and chitosan were used. After synthesis, the physicochemical properties of the nanofibers were confirmed by Scanning Electron Microscopy (SEM), Fourier-transform infrared spectroscopy (FTIR), and X-ray energy diffraction spectroscopy (EDS or EDX), and Differential Scanning Calorimetry (DSC). The electrospun produced nanofibers have continuous and uniform structures. The cytotoxicity assay of these electrospun nanofibers were done on Human dermal fibroblast cell (HDF) and HUVEC cell (Human Umbilical Endothelial Cells) lines and results showed that nanofiber doesn't have any toxicity to normal cell lines. For anti-fungal activity tests, the appropriate amounts of nanofibers containing M. neglectum were placed in media with five different fungal species utilizing two methods: disc diffusion and well diffusion. In vitro results showed that all electrospun nanofibers containing M. neglectum had strong antifungal activity against Candida albicans, Glabrata, Parapacillus, Guillermoides, Crocus fungi species. Our findings also showed that nanofibers containing 86.88% polyvinyl alcohol/ gelatin/ chitosan/ M. neglectum root extract (produced nanofibers) were had better swelling and physicochemical properties and stronger antifungal activity than others (fiber formed with plant root). In a nutshell, natural nanofibers can be used as a beneficial drug delivery system.

This work reports for the first time the anticandidal activity of the dichloromethane (DCM) extract from the Algerian invasive red marine algae A. armata Harvey. In a preliminary assessment, the DCM extract was screened for its antifungal activity against two strains of Candida albicans, namely (IP 444) and (ATCC 10231). As a well-established approach, the anticandidal activity was expressed as zone of inhibition by agar well and disk diffusion methods. The minimum inhibitory concentration (MIC) was also determined for the two fungal strains. Within this framework, the DCM extract of the red algae A. armata has a strong inhibition effect against the considered Candida albicans strains. In addition, Candida albicans IP444 (MIC = 0.58 mg/mL) was more sensitive to DCM algal extract when compared to Candida albicans ATCC 10231 (MIC = 2.34 mg/mL). GC–MS analysis suggested that brominated compounds and fatty acids are responsible for anticandidal activity of dichloromethane extract of A. armata. These preliminary results may constitute a future applied of lipophilic extract of the red algae A. armata as a promising source of natural compounds with antifungal properties.

Background: Edible oils have gained the interest of several industrial sectors for the different health benefits they offer, such as the supply of bioactive compounds and essential fatty acids. Microencapsulation is one of the techniques that has been adopted by industries to minimize the degradation of oils, facilitating their processing. Objective: To evaluate the intellectual property related to patent documents referring to microencapsulated oils used in foods. Methods and Results: This prospective study investigated the dynamics of patents filed in the Espacenet and National Institute of Industrial Property (INPI) databases, and it mapped technological developments in microencapsulation in comparison with scientific literature. The years 2015 and 2018 showed the greatest growth in the number of patents filed in the Espacenet and INPI databases, respectively, with China leading the domains of origin, inventors, and owners of microencapsulation technology. The largest number of applications of microcapsules were observed in the food industry, and the foods containing microencapsulated oils were powdered seasonings, dairy products, rice flour, nutritional formulae, pasta, nutritional supplements, and bread. The increase in oxidative stabilities of oils was the most cited objective to microencapsulate oils. Spray drying was the most widely used microencapsulation technique, and maltodextrin, gum arabic, and modified starch were the most widely used wall materials. Conclusion: Microencapsulation of oils has been expanding over the years and increasing the possibilities of the use of microcapsules, but further investments and development of policies and incentive programs to boost this technology need to be made in less developed countries. For future perspectives, the microencapsulation technique is already a worldwide trend in the food industry, enabling the development of new products to facilitate their insertion in the consumer market.

Recently, 3D bioprinting has been explored as a promising technology for biomedical applications with the potential to create complex structures with precise features. Cell encapsulated hydrogels composed of materials such as gelatin, collagen, hyaluronic acid, alginate and polyethylene glycol have been widely used as bioinks for 3D bioprinting. However, since most hydrogel-based bioinks may not allow rapid stabilization immediately after 3D bioprinting, achieving high resolution and fidelity to the intended architecture is a common challenge in 3D bioprinting of hydrogels. In this study, we have utilized shear-thinning and self-healing ionically crosslinked oxidized and methacrylated alginates (OMAs) as a bioink, which can be rapidly gelled by its self-healing property after bioprinting and further stabilized via secondary crosslinking. It was successfully demonstrated that stem cell-laden calcium-crosslinked OMA hydrogels can be bioprinted into complicated 3D tissue structures with both high resolution and fidelity. Additional photocrosslinking enables long-term culture of 3D bioprinted constructs for formation of functional tissue by differentiation of encapsulated human mesenchymal stem cells.

The chemical analysis of three Algerian plants was conducted by using Headspace Solid Phase Micro Extraction coupled with Gas Chromatogr aphy Mass Spectrometry (HS SPME GC/ MS). This analysis was performed directly on the dried aerial parts of the plants. The results revealed that the major compounds are 1,8 cineol (27.3%) in Ajuga iva, cis-muurola-3,5 diene (14.6%) in Salvia verbenaca and 2,3-dehydro 1,4-cineol (36%) in Thymus algeriensis . The oxygenated monoterpenes are the major class representing 62.1% and 78.4% in Ajuga iva and Thymus a lgeriensis , respectively, whereas the sesquiterpene hydrocarbons are the major class in Salvia verbena ca representing 50.1%. Identification of naturally rare monoterpene ether, 2,3-dehydro 1,4-cineol was the first time in the Ajuga iva Salvia verbenaca and Thymus algeriensis.

Anthracnose disease caused by Colletotrichum lindemuthianum (Sacc. and Magnus) Lams- -Scrib is one of the most devastating seed-borne diseases of common bean (Phaseolus vulgaris L.). In the present study, we evaluated the antifungal activity of Bunium persicum essential oil (EO) and its main constituents on mycelial growth, sporulation and spore germination inhibition of C. lindemuthianum. The main objective of this study was to investigate the effect of EO and its main constituents on decreasing the activity of cell wall degrading enzymes (CWDEs) produced by C. lindemuthianum, which are associated with disease progress. Also, the effects of seed treatment and foliar application of EO and its main constituent, cuminaldehyde, on anthracnose disease severity was investigated. The essential oil of B. persicum, was obtained by using a clevenger apparatus and its major constituents were identified by gas chromatography-mass spectrometry (GC-MS). The EO was characterized by the presence of major compounds such as cuminaldehyde (37.7%), γ-terpinene (17.1%) and β-pinene (15.4%), which indicated antifungal effects against C. lindemuthianum. This pathogen did not grow in the presence of EO, cuminaldehyde and γ-terpinene, β-pinene at 1,500; 1,010 and 1,835 ppm concentrations, respectively. Also, sporulation and spore germination of C. lindemuthianum was completely inhibited by EO and cuminaldehyde. Synergistic effects of the main constituents showed that combing γ-terpinene with cuminaldehyde induced a synergistic activity against C. lindemuthianum and in combination with β-pinene caused an additive effect. Activities of pectinase, cellulase and xylanase, as main CWDEs, were decreased by EO and its main constituents at low concentration without affecting mycelial growth. Seed treatment and foliar application of peppermint EO and/or cuminaldehyde significantly reduced the development of bean anthracnose. We introduced B. persicum EO and constituents, cuminaldehyde and γ-terpinene, as possible control agents for bean anthracnose.

Background: Streptococcus sanguinis can contribute to tooth demineralization, which can lead to dental caries. Antibiotics used indefinitely to treat dental caries can lead to bacterial resistance. Discovering new antibacterial agents from natural products, like Ocimum basilicum, will help combat antibiotic resistance. In silico analysis (molecular docking) can help determine the lead compound by studying the molecular interaction between the drug and the target receptor (MurA enzyme and DNA gyrase). It is a potential candidate for antibacterial drug development. Objective: The research objective is to isolate the secondary metabolite of O. basilicum extract that exhibits activity against S. sanguinis through in vitro and in silico analysis. Methods: n-Hexane extract of O. basilicum was purified by combining column chromatography with bioactivity-guided fractionation. The in vitro antibacterial activity against S. sanguinis was determined using the disc diffusion and microdilution method, while molecular docking simulation of nevadensin (1) with MurA enzyme and DNA gyrase was performed by using PyRx 0.8 program. Results: Nevadensin from O. basilicum was successfully isolated and characterized by spectroscopic methods. This compound showed antibacterial activity against S. sanguinis with MIC and MBC values of 3750 and 15000 μg/mL, respectively. In silico analysis showed that the binding affinity to MurA was -8.5 Kcal/mol, and the binding affinity to DNA gyrase was -6.7 Kcal/mol. The binding of nevadensin-MurA is greater than fosfomycin-MurA. Otherwise, Nevadensin-DNA gyrase has a weaker binding affinity than fluoroquinolone-DNA gyrase and chlorhexidine-DNA gyrase. Conclusion: Nevadensin showed potential as a new natural antibacterial agent by inhibiting the MurA enzyme rather than DNA gyrase.

Aims The aim of this study was to investigate the effectiveness of bread as substrate for γ-aminobutyric acid (GABA) biosynthesis, establishing a valorization strategy for surplus bread, repurposing it within the food chain. Methods and Results Surplus bread was fermented by lactic acid bacteria (LAB) to produce GABA. Pediococcus pentosaceus F01, Levilactobacillus brevis MRS4, Lactiplantibacillus plantarum H64 and C48 were selected among 33 LAB strains for the ability to synthesize GABA. Four fermentation experiments were set up using surplus bread as such, added of amylolytic and proteolytic enzymes, modifying the pH or mixed with wheat bran. Enzyme-treated slurries led to the release of glucose (up to 20 mg g−1) and free amino acid, whereas the addition of wheat bran (30% of bread weight) yielded the highest GABA content (circa 800 mg kg−1 of dry weight) and was the most suitable substrate for LAB growth. The selected slurry was ultimately used as an ingredient in bread making causing an increase in free amino acids. Conclusions Besides the high GABA concentration (148 mg kg−1 dough), the experimental bread developed in this study was characterized by good nutritional properties, highlighting the efficacy of tailored bioprocessing technologies as means to mitigate food wastage. Significance and Impact of Study Our results represent a proof of concept of effective strategies to repurpose food industry side streams.

The objective of the study was to explore the effects of pomegranate juice (PJ) as a water supplement on performance, egg quality, and blood parameters in laying hens. For this purpose, a total of 72 Babcock laying hens, were divided into three groups, one control and two experiments (n = 24). Each main group consists of 4 subgroups, and each subgroup consists of 6 chickens. PJ was administered in drinking water at 0%, 5%, and 10% to the experimental groups for 4 weeks. Feed was offered to all groups ad libitum. The addition of PJ had no significant effect on performance parameters, Haugh unit, and egg breaking strength. Urbanization and industrialization have led to heavy metals in aquatic environments. These metals are toxic, nondegradable, and persistent. Lead (Pb) is an immunotoxicant that can adversely affect animal health, even at low levels. Chitosan (CHI) is an eco-friendly compound with exceptional antibacterial and immunostimulant characteristics. This study investigated the effect of dietary CHI on the antioxidant defense system and innate immune responses of tilapia (Oreochromis niloticus) against the cytotoxic effect of prolonged Pb exposure. The results indicated dietary CHI could nullify the toxicity of prolonged Pb exposure in fish by substantially activating the antioxidant defense system capacity and fish innate immune responses. In addition, CHI minimized the alterations in the histology of fish organs. The findings suggest that chitosan can be a commercial, eco-friendly immunostimulant and bioactive compound for mitigating the effects of heavy metal toxicity in aquaculture.

In this paper, effects of pectin varying in overall charge (OCH) and local charge density (LCD) on the structure and thermal stability of Lysozyme (lys) are studied. The secondary and local tertiary structures of lys undergo at pH 5.1 a two-step change upon binding with increasing amounts of pectin, in which the peak of fluorescence first increases and shows a blue shift and the amount of helix conformation drops at a pectin/lys mass ratio (q) below or equal to that corresponding to maximal complexation qmax. With increasing amount of pectin, the native like state is recovered at q > qmax. The decrease in helix content depends mainly on the OCH of pectin. Analysis of the thermodenaturation behavior of lys reveals that affinity of pectin with denatured lys is higher than for the native state of lys. At the same OCH of pectin, unfolding of lys is more significant in case of a high LCD (blockwise distribution of charges) of the former. At q = qmax (first stage of binding) the key factors stabilizing lys within its complexes with pectin against thermo-aggregation are a high LCH of pectin and, to a lesser degree, a low OCH. At q > > qmax (second stage of binding), a high LCH of pectin is a key factor inhibiting thermo-aggregation of lys.

Microfluidics is characterized by the manipulation of fluids in submillimeter channels and has great application potential in encapsulation. To further extend the application of microfluidics in food industries, a 3D printed microfluidic device is used to encapsulate vitamin A and improve its stability. Two natural macromolecules, sodium alginate and gelatin, are added to water as the continuous phase to generate monodisperse emulsion. Under different flow rate ratios, the diameter of droplets decreases with the increase of continuous flow rate. However, at the same flow rate ratio, varying the dispersed and continuous flow rates does not significantly change the diameter and size distribution of emulsion collected. The prepared O/W (oil/water) single emulsion can form microgel particles and avoid degradation of vitamin A by simulated gastric acid; the encapsulated vitamin A will not be released until particles reach simulated intestinal tract. In the simulated digestion in vitro, no vitamin A is released for 2 h in the acidic environment; under an alkaline or neutral environment such as those in intestinal fluids, vitamin A can be released from the microgel particles within 2.5 h. Using the presented approach, emulsions encapsulating vitamin A have been prepared and can potentially be applied to encapsulate other oil-soluble substances in the food industry.

Dracocephalum L. is a well-known aromatic and medicinal plant genus in the Mint family. Some species of the genus such as D. kotschyi (DK) are widely used in Iranian folk medicine as tonic and carminative, as well as for relieves of headache, stomachache, and liver ailments. The present study was aimed to: isolate and determine some bioactive flavonoids from DK, evaluate their content in some Iranian Dracocephalum species, and assess their inhibitory activity and molecular dynamics against Human mutT homolog 1 (MTH1) enzyme. Aerial flowering parts of eight Iranian native Dracocephalum species were collected from different regions of Iran. The flavonoids of DK were comprehensively isolated by column chromatography, and then characterized by NMR and MS techniques. For simultaneous determination of flavonoids in methanol extract of the Dracocephalum studied species, a reversed-phase HPLC-DAD method was developed. Luteolin, apigenin, xanthomicrol, calycopterin, ermanin and three newly isolated flavonoids including circilineol, 5-demethylsinenstin, cirsimaritin and 5,7,4´-trihydroxy 3,3´-dimethoxyflavone were characterized in DK. Simultaneous determination of nine flavonoids in eight Iranian Dracocephalum species exerted the highest amount of xanthomicrol (4430±1.8 µg/g DW) and 5-demethylsinensetin (766.2±0.4 µg/g DW) in D. polychaetum and D. aucheri, respectively. Our study showed, different species with similar morphology have different flavonoid profile. To have a molecular insight into their effectiveness in cancer inhibition, Gaussian 03, GROMACS 5.2, and Pymol software were exploited to optimize, simulate, and dock them against MTH1 enzyme, respectively. Molecular docking study revealed that xanthomicrol and 5-demethylsinenstin could tightly bind to MTH1 enzyme via different hydrogen bonds in the active site of the protein. Xanthomicrol and 5-demethylsinenstin, the two flavonoids with stronger bonds with MTH1 enzyme, could be considered as reliable pharmacologically active ingredients against the cancers involving MTH1 activity.

Crop infestation species have acquired resistance to herbicides, stimulating the search for new control methods. Plants secondary metabolites can hold more sustainable alternatives, as they can be used for development of natural herbicides. This study evaluated phytotoxic effects of aqueous extracts of Ilex paraguariensis leaves on Conyza bonariensis, an infesting species of Brazilian crops. Different concentrations of decoction and infusion extracts were tested in germination and growth bioassays. Adult plants were evaluated for leaf anatomy effects caused by the yerba mate extract. HPLC analysis was used to quantify the three major chemical constituents. The most effective aqueous extract (decoction 1.0%) decreased the percentage of seed germination, germination speed index, and percentage of germination-derived seedling formation, and increased the time for seedling formation. Shoot and root growth were not affected by this extract. In adult plants, the same extract damaged epidermis and palisade/lacunous parenchyma six hours after exposure. Caffeine was found as the major compound of this extract, followed by rutin and quercetin. Therefore, aqueous extract by decoction 1.0 % of I. paraguariensis leaves showed potential for control of C. bonariensis infestation and rather promising for the development of a natural herbicide.

The highest score of color hedonic is 3,97, and the highest score of taste hedonic is 3,67. The highest score of color hedonic quality is 3,11 with dark brown color criteria. Growth plate cartilage has limited self-repair ability, leading to poor bone bridge formation post-injury and ultimately limb growth defects in children. The current corrective surgeries are highly invasive, and outcomes can be unpredictable. Following growth plate injury, the direct loss of extracellular matrix (ECM) coupled with further ECM depletion due to the inhibitory effects of inflammation on the cartilage matrix protein greatly hinder chondrocyte regeneration. We designed an exosome (Exo) derived from bone marrow mesenchymal stem cells (BMSCs) loaded ECM-mimic hydrogel to promote cartilage repair by directly supplementing ECM and anti-inflammatory properties. Aldehyde-functionalized chondroitin sulfate (OCS) was introduced into gelatin methacryloyl (GM) to form GMOCS hydrogel. Our results uncovered that GMOCS hydrogel could significantly promote the synthesis of ECM due to the doping of OCS. Herbal cosmetics are a valuable gift of nature and their demand is increasing in the world market. They have good activity with no side effects as compared to synthetic cosmetics. Herbal cosmetics are beauty products and because of natural ingredients, they possess desirable physiological activities. The present research work aimed to formulate and characterize cost-effective herbal nourishment cream containing Aloe vera gel and tomato powder. Four formulations i.e. F1, F2, F3, and F4 were prepared with varying concentrations of Aloe vera gel and tomato powder, while the composition of the cream base was kept the same. The oil phase of the cream base consisted of liquid paraffin (5.0 ml), stearic acid (4.8 g) and coconut oil (3.0 ml). The aqueous phase consisted of triethanolamine (1.5 ml), glycerin (5.0 ml), citric acid (0.4 g), methylparaben (0.1 g), water (q.s.) and key ingredients. The concentration of Aloe vera gel was 2.0 ml, 4.0 ml, 6.0 ml, 8.0 ml and the concentration of tomato powder was 0.2 g, 0.4 g, 0.6 g, 0.8 g in F1, F2, F3, and F4 respectively. The characterization of the formulated creams was carried out by standard methods, such as homogeneity test, smear type, irritancy test, pH test, spreadability test and acid value test. The stability test was carried out at different temperature conditions at 40 °C, below 5 °C and room temperature. All creams were homogeneous, non-irritant and easily removable. The pH of creams was in the range of 7.3–7.6 which is safe for human skin. Spreadability was in the range of 9–13. Acid values of the prepared creams were 5.6, 5.6, 6.4 and 7.2 respectively. Stability was examined by two parameters, homogeneity and pH change. The economical herbal creams prepared by using fewer chemicals are good for skin with moisturizing properties. The efficiency of creams can be improved by increasing the concentration of Aloe vera gel and tomato powder.

Cucurbita moschata D. seed oil contains approximately 75% unsaturated fatty acids, with high levels of monounsaturated fatty acids and antioxidant compounds such as vitamin E and carotenoid, constituting a promising food in nutritional terms. Associated to this, the Brazilian germplasm of C. moschata exhibits remarkable variability, representing an important source for the genetic breeding of this vegetable and other cucurbits. In this context, the present study evaluated the productivity and profile of the seed oil of 91 C. moschata accessions from different regions of Brazil and maintained in the Vegetable Germplasm Bank of the Federal University of Viçosa (BGH-UFV). A field experiment was conducted between January and July 2016. The tested C. moschata accessions showed high genetic variability in terms of characteristics related to seed oil productivity (SOP), such as the mass of seeds per fruit and productivity of seeds, providing predicted selection gains of 29.39 g and 0.26 t ha-1, respectively. Based on the phenotypic and genotypic correlations, greater SOP can be achieved while maintaining high oleic acid content and low linoleic acid content, providing oil of better nutritional and chemical quality. In variability analysis, the accessions were clustered into five groups, which presented different averages for SOP and fatty acid content of seed oil; approach that will guide the use of appropriate germplasm in programs aimed at genetic breeding for SOP and seed oil profile. Per se analysis identified BGH-4610, BGH-5485A, BGH-6590, BGH-5556A, BGH-5472A, and BGH-5544A as the most promising accessions in terms of SOP, with average (μ+g) of approximately 0.20 t ha-1. The most promising accessions for higher oleic acid content of seed oil were BGH-5456A, BGH-3333A, BGH-5361A, BGH-5472A, BGH-5544A, BGH-5453A, and BGH-1749, with average (μ+g) of approximately 30%, and almost all of these accessions were also the most promising in terms of lower linoleic acid content of seed oil, with average (μ+g) of approximately 45%. Overall, part of the C. moschata accessions evaluated in the present study can serve as a promising resource in genetic breeding programs for SOP and fatty acid profile, aiming at the production of oil with better nutritional and physicochemical quality.

Developing an environmental-friendly and highly active catalyst in transesterification for biodiesel production is of great importance for a more economic biodisel process. Herein, we reported that waste eggshells were used to adsorb Au(III) in water and convert the Au(III)-polluted eggshells into the functional nanocatalyst-CaO/Au for the transesterification reaction between soybean oil and methanol to the preparation of biodiesel. By coupling of CaO and Au nanoparticles, CaO/Au nanoparticles showed superior catalytic activity for the transesterification reaction between soybean oil and methanol. An optimum performance was observed over CaO/Au nanocomposites in a methanol-oil molar ratio at 12: 1 with catalyst content of 1.0 wt% at 70 °C for 3 h. Besides, the catalytic activity of CaO/Au nanocatalyst was almost unchanged after recycling for 5 times and the yield of biodiesel still kept at 88.9%. The proof-of concept study provided us a sustainable method for utilization of waste eggshells to remedy the metal ions-polluted wastewater and the synthesis of functional nanocomposite for biodiesel production, show great potential application of waste eggshell in adsorption and catalytic reactions.

India is an emerging basil essential oil producer in South-east Asia. Two high essential oil yielding hybrids, namely one inter specific hybrid between of O. basilicum and O. kilimandscharicum Gurke (HYBL-1) and another intraspecific hybrid of O. basilicum × O. basilicum (OBL-1) of basil were analyzed using GC, enantiomeric GC, NMR, enantio-GC–MS and GC–MS methods. Inter specific hybrid HYBL-1 contained high essential oil-rich in linalool (68.5%), camphor (8%), and 1,8-cineole (4.6%) as characteristic constituents among monoterpenoids, whereas β-caryophyllene (1.9%), germacrene D (1.0%), and epi-α-cadinol (1.9%) were the sesquiterpenoids at the Lucknow (North Indian conditions) and linalool (71.8%), camphor (9.4%) and 1,8-cineole (4.3%) at Hyderabad (South Indian conditions) locations. Intraspecific hybrid (OBL-1) possessed linalool (66.1%), 1,8-cineole (5.4%) and geraniol (8.6%) with sesquiterpenoids in low proportions. Inter specific hybrid HYBL-1 showed superiority over OBL-1 in the multi-location trials conducted at Lucknow and Hyderabad. Average mean performance of inter specific hybrid over locations was: herb yield 44.80 t/ha, oil content 0.63%, oil yield 188.50 kg/ha, linalool content 67.65%, camphor content 8.90% v/s OBL-1 herb yield 21.32 t/ha, oil content 0.53%, oil yield 97.50 kg/ha, linalool content 65.55%, camphor content 0.00%, respectively. The essential oil of these two hybrids subjected to enantiomer differentiation revealed a high enantiomeric excess for (3R) -(−)-linalool, whereas (1R)- (+)-camphor was recorded exclusively in inter specific hybrid. The extensive NMR experiments were performed to confirm constituents in these hybrids and found that NMR spectroscopy could also be an ideal tool for the differentiation of essential oils from commercial samples declared as natural.

Biodiesel can serve as an alternative to petroleum-based fuels. It has been shown that standard biodiesel cannot be mixed to improve efficiency and emissions characteristics. It has a major effect on emissions but has a limited capacity to mix with co-feed at varying temperatures and different ratios. In this study, 63 tertiary blends and three binary blends were mixed to create a stable fuel. Biodiesel was stable after having blended with amphipathioic liquor of a broad range of compatibility for a period of 168 h is recorded. Four samples were chosen for reproducibility, and 2 were selected for higher than average ethanol content in ethanol. These can be defined as one-observed, inferred, and derived in the three-layer, double-blurred, double-layer, two-blurred, one-blurred ring formation, and a single-blurred double-layer format. Even at 27.2 °C, biodiesel micelles could not shape micelles were used. The fuel blend, which has a wider range of miscibility, suitable for transportation and stability, causes more aromatic emissions when used in a ternary formulation than a micro formulation. The blend properties were found to be within tolerable limits.

Aim This study aims to use fermentation waste of ethanol production (solid and liquid) for riboflavin and recycling of bacterial biomass as biofertilizers to enhance the growth of some oily crop plants. Methods and Results Out of 10 yeast isolates from fresh milk, Clavispora lusitaniae ASU 33 (MN583181) was able to ferment different concentrations of glucose (2.5%, 5%, 7.5%, 10%, 15% and 20%) into ethanol with high efficiency at 10%. Among seven non-Lactobacillus bacterial isolates recovered from cheese samples, two bacterial isolates Bacillus subtlis-SR2 (MT002768) and Novosphingobium panipatense-SR3 (MT002778) were selected for their high riboflavin production. Different media (control medium, fermentation waste medium and a mixture of the fermentation waste medium and control medium [1:1]) were used for riboflavin production. These media were inoculated by a single or mixture of B. subtlis-SR2, N. panipatense-SR3. The addition of the waste medium of ethanol production to the control medium (1:1) had a stimulatory effect on riboflavin production whether inoculated with either a single strain or a mixture of B. subtlis-SR2 and N. panipatense-SR3. A mixture of fermentation waste and control media inoculated with N. panipatense produced a high riboflavin yield in comparison with other media. Inoculation of Zea mays and Ocimum basilicum plants with either the bacterial biomass waste of riboflavin production (B. subtlis or N. panipatense) or a mixture of B. subtlis and N. panipatense) shows a stimulatory effect on the plant growth in comparison with control (uninoculated plants). Conclusions These results demonstrate the possibility of minimizing the cost of riboflavin and biofertilizer manufacturing via interlinking ethanol and riboflavin with the biofertilizer production technology. Significance and Impact of Study This study outlines the methods of evaluating the strength of spent media by applying procedures developed in the vitamin production industries. Furthermore, bacterial biomass waste can act as an environmentally friendly alternative for agrochemicals.

In the present study, 30 potential germplasm of oat (Avena sativa L.) were subjected to proximate, elemental, and HPLC analysis to provide a scientific basis to genetic diversity present among them. The extracts of the selected germplasms were also evaluated for their antioxidant potentials through DPPH and ABTS assays. Proximate analysis showed protein contents to be in the range 8.35 to 17.72% with the highest protein contents in the accession line 22365 (17.72±0.38%). The genotype-725 showed the highest carbohydrate, and dry matter (53.35±0.01 and 93.50±0.07% respectively) contents whereas, the germplasm-830 contained the highest fat (7.88±0.12%) contents while the highest moisture contents were there in germplasm-22348 (11.95±0.06%). The crude fiber contents (19.67±0.19%) were found high in germplasm-832. The mentioned contents were also correlated to each other where a negative (-0.431*) correlation was noted for crude protein and carbohydrate while ash content to crude protein has a positive (0.38*) correlation. A positive and a negative correlation were there in Crude fats/crude protein (0.30*) and crude fats/moisture contents (-0.39*) respectively. Principal component analysis showed an Eigenvalue of 0.76 with a total variation of 85.01% when applied to proximate components. Based on cluster analysis to proximate composition all the oat germplasms were divided into 5 sub-clusters, where accession numbers 769 and 817 were found to be the most diverse genotypes. The elemental analysis confirmed the presence of magnesium (2.89-7.62 mg/L), sodium (3.71-8.03 mg/L), manganese (0.93-3.71 mg/L), copper (0.35-3.36 mg/L), iron (2.15-6.82 mg/L), zinc (1.30-3.37 mg/L), chromium (0.37-3.34 mg/L), and potassium (50.70-59.60 mg/L) in the selected germplasms. Principal component analysis for elemental composition showed the total variation of 73.75% with the Eigenvalue of 0.97. Cluster analysis on an elemental basis divided all the oat germplasms into 7 sub-clusters where accession numbers 769 and 22350 were found to be the most diverse germplasm. Phytochemical analysis performed through HPLC resulted in the identification of nine possible compounds (malic acid, epigallocatechin gallate, quercetin, morin, ellagic acid, catechin hydrate, rutin, pyrogallol, and mandelic acid) in various germplasm of oat. A concentration-dependent antioxidant response was recorded when extracts were tested as an inhibitor of DPPH and ABTS free radicals. The results revealed that oat grains are a good source of nutrients, minerals, and phytochemicals that can be used as nutraceuticals and as food. The genetic differences revealed that this plant can be grown under varied environmental conditions.

Introduction: Aminoglycoside-induced acute kidney injury (AKI) is a pathology closely linked to oxidative and inflammatory reactions. Taking into account the previous reported antioxidant and anti-inflammatory effects of D-005, a lipid extract obtained from Cuban palm Acrocomia crispa (Arecaceae) fruits, this work aimed to evaluate the effects of D-005 on kanamycin-induced AKI. Methods: Male Wistar rats were divided into 7 groups: negative control (vehicle, Tween 65/H2O) and six groups treated with kanamycin to induce AKI: positive control (vehicle), D-005 (25, 100, 200, and 400 mg/kg) and grape seed extract (GSE, 200 mg/kg). D-005, vehicle, and GSE oral treatments were administered once daily for seven days, 1 h before kanamycin (500 mg/kg, i.p.). Serum uric acid and urea concentrations, renal histopathology, and oxidative markers (malondialdehyde (MDA), sulfhydryl (SH) groups, and catalase (CAT) activity) were assessed. Results: D-005 significantly reduced uric acid and urea levels, starting from D-005 100 mg/kg. Histopathologically, D-005, at all the tested doses, protected renal parenchyma structures (glomeruli, proximal tubules, and interstitium). These findings were accompanied by a significant reduction of MDA and SH group concentrations as well as restoration of CAT activity. The highest percentages of inhibition were obtained with the dose of 400 mg/kg. GSE, the reference substance, also prevented kanamycin-induced biochemical and histopathological changes, as well as reduced MDA and SH groups and restored CAT activity. Conclusion: The administration of repeated oral doses of D-005 significantly protected against kanamycin-induced AKI, which could be associated with the antioxidant and anti-inflammatory effects of this extract.

The fruits of Coccinia grandis (L.) Voigt have been used to treat several aliments in traditional medicine whereas Coccinia rehmannii Cogn. fruits are consumed as food due to their high starch content. Coccinia rehmannii are also known to contain vast amounts of indispensable bioactive compounds that are used in various cosmetics, foods, and medicines. Among these compounds are hydroxycinnamic acids (HCAs) which are usually conjugated/esterified to quinic acid (QA), tartaric acid, citric acid, shikimic acid and sugars and are collectively referred to as chlorogenic acids (CGAs). Over the years, metabolite profiling has been used in plant taxonomy and species classification. Herein, UPLC-qTOF-MS/MS semi-targeted approached was use to profile HCA conjugates from two closely related plant species i.e. C. grandis and C. rehmannii leaves in order to distinguish them from each other. Examination of their relative metabolite contents showed that the two closely related Coccinia species utilise contrasting means of HCA conjugations as means of metabolite diversification strategy. C. grandis was found to conjugate its HCA derivatives to a QA, whilst C. rehmannii conjugates its HCA derivatives to mainly flavonoid glycosides and to QA, even though present in low concentration in this plant. These results shows that plants from the same genus can utilise different chemical diversification means for storage of their metabolites and that processes such as conjugation and isomerization can be exploited as chemotaxonomic markers. Future studies can also be designed to look at the pharmacological potency of these plants as a function of their contrasting chemical differences.

Aims Escherichia albertii is an emerging diarrheagenic pathogen causing food- and water-borne infection in humans. However, no selective enrichment broths for E. albertii have ever been reported. In this study, we tested several basal media, selective supplements and culture conditions which enabled selective enrichment of E. albertii. Methods and Results We developed a selective enrichment broth, novobiocin–cefixime–tellurite supplemented modified tryptic soy broth (NCT-mTSB). NCT-mTSB supported the growth of 22 E. albertii strains, while inhibited growth of other Enterobacteriaceae at 37°C, except for Escherichia coli and Shigella spp. Enrichment of E. albertii was improved further by growth at 44°C, a temperature that suppresses growth of several strains of E. coli/Shigella. Combined use of NCT-mTSB with XR-DH-agar, xylose–rhamnose supplemented deoxycholate hydrogen sulphide agar, enabled isolation of E. albertii when at least 1 CFU of the bacterium was present per gram of chicken meat. This level of enrichment was superior to those obtained using buffered peptone water, modified-EC broth, or mTSB (with novobiocin). Conclusions Novobiocin–cefixime–tellurite supplemented modified tryptic soy broth enabled effective enrichment of E. albertii from poultry samples and was helpful for isolation of this bacterium. Significance and Impact of Study To our knowledge, this is the first report of selective enrichment of E. albertii from poultry samples.

A monitoring survey was conducted in snake gourd agro ecosystem during the fruiting season at Gangnapur, West Bengal, India to generate extensive data on fruit fly diversity, population dynamics, infestation rates and evaluate the effect of trapping on fruit fly infestation rates vis-a-vis benefit cost ratio of snake gourd cultivation. Methyl Eugenol and Cue Lure traps were used to assess the fruit fly species diversity. The number of adult male flies trapped was used to calculate fruit fly per trap per day and assess the population dynamics. Bactrocera dorsalis, Zeugodacus cucurbitae and Zeugodacus tau were the predominant fruit fly species during the fruiting season. Maximum species diversity and evenness estimates were registered by Methyl Eugenol traps. Species richness estimates for Cue Lure and Methyl Eugenol traps during the entire experiment are analogous due to the trapping of two different types of fruit fly species. Zeugodacus cucurbitae is the most abundant species in the snake gourd ecosystem and dominated the community of fruit flies throughout the study. Fruit fly per trap per day values indicated that the Gangnapur area is at infestation level, for Zeugodacus cucurbitae and Bactrocera dorsalis, while, for Zeugodacus tau the area is at suppression level. The rate of infestation varied from 6.93 % to 44.09 %. Farmers with traps in their fields reported a benefit cost ratio of 0.73 to 4.63, while the no-trap group reported 0.34 to 1.56. This study at Gangnapur has proven the beneficial effect of trapping on the benefit cost ratio of snake gourd cultivation.

Aims The present study aimed to specifically transform stevioside (ST) into rubusoside (RS) through bioconversion with high efficiency, seeking to endow steviol glycosides (SGs) with subtle flavours for commercial acceptability. Methods and Results An endophytic bacterium named Lelliottia LST-1 was screened and confirmed to specifically convert ST into RS, reaching a conversion rate of 75.4% after response surface optimization. Phylogenetic analysis combined with complete genome sequencing demonstrated that LST-1 was also presumed to be a new species. To further explore the principle and process of biological transformation, the potential beta-glucosidases GH3-1, GH3-2, GH3-3 and GH3-4 were expressed, purified and reacted with SGs. High-performance liquid chromatography revealed that all enzymes hydrolysed ST and generated RS, but substrate specificity analysis indicated that GH3-2 had the highest substrate specificity towards STs and the highest enzyme activity. Conclusion The potential β-glucosidase GH3-2 in Lelliottia sp. LST-1 was found to specifically and efficiently convert ST to RS. Significance and Impact of Study The efficient biotransformation of ST into RS will be beneficial to its large-scale production and extensive application in the food and pharmaceutical industries.

Biodiesel has emerged as a promising alternative fuel to replace dwindling fossil-based resources, particularly in view of its added environmental merit of reducing additional air pollution. Its specific attraction stems from the similarity of its physical properties to fossil fuel-derived diesel. Although the production of biodiesel is a relatively straightforward process, reaction progress monitoring and product analysis require costly specialist equipment, such as gas chromatography and mass spectrome-try. In this study, we investigate the use of pH in monitoring the progress of carbonate-catalyzed transesterification reactions. Specifically, we focus on potassium and sodium carbonates and sunflower oil. Our results are consistent with the results obtained by other studies using different methods of monitoring. To test the generality of the method, pH measurements were also used to monitor the progress of the potassium carbonate transesterification reaction in the presence of added water, glycerol and gamma-valerolactone (GVL). The obtained results are as expected, with a limited amount of water increasing the trans-esterification rate; glycerol slowing the reaction slightly in accord with Le Chatellier's principles; and GVL increasing the rate due to co-solvent effects. Atomic-level insights into the adsorption mechanism of methanol and water on the (001) surfaces of Na2CO3 and K2CO3 catalysts are provided by first-principles DFT calculations, which explain the increase in transesterification reaction rate upon the addition of water. Flaxseed oil contains lignans, which exhibit anti-inflammatory and antiatherogenic activities. A 70-year-old male patient presented to our office due to hyperlipidaemia and started to take a tablespoon of flaxseed oil daily. Three months later, he reported left breast swelling and pain. Although the echogram revealed a tumour in the left mammary gland, the breast biopsy was compatible with gynecomastia, showing ductal hyperplasia without evidence of malignancy. His breast epithelia were oestrogen receptor-positive. Potential role of phytoestrogens was discussed.

Background: Low-molecular citrus pectin (LCP) is a pectin polysaccharide with low molec-ular weight, low degree of crux, and no branching. It is obtained by degrading natural citrus pectin (CP) through physical, chemical and enzymatic methods. LCP has received considerable attention in recent years due to its potential applications in the medical and biological fields. Methods: In our previous study, LCP was prepared from CP by using recombinant Bacillus subtilis pectate lyase B. Monosaccharide comparative analysis revealed that the galacturonic acid content of LCP was higher than that of CP. The cell viability effect of LCP was elucidated by using HepG2 cells and the Cell Counting Kit-8 (CCK-8) assay. Terminal deoxynucleotidyl transferase dUTP nick-end labeling (TUNEL) staining, Annexin V-FITC/PI staining, and flow cytometer propidium iodide stain-ing were performed to detect the effects of LCP on apoptosis and cell cycle arrest in HepG2 cells. Mi-tochondrial membrane potential (MMP) was observed through 5,5′,6,6′-tetrachloro-1,1′,3,3′-tetraethyl-imidacarbocyanine assay. Results & discussion: The Mw of the prepared LCP was 7.6 kDa, which was significantly lower than that of CP (140 kDa). Cell viability decreased with the increase in the concentration of LCP. The half-inhibitory concentration of 1.46 ± 0.02 mg/mL was determined. Treatment with 1.6 mg/mL LCP in-duced the apoptosis of HepG2 cells with the inhibition rate of 83.10% ± 4.72%, and the cell cycle was arrested in the S phase. Furthermore, the MMP of HepG2 cells decreased with the increase in LCP concentration. Conclusion: The enzymatically prepared LCP could inhibit the proliferation of HepG2 cells. This study provided a partial experimental basis and reference for LCP to become a potential functional food for anti-liver cancer.

Sperm fertility decreases significantly after freezing. Providing a suitable and useful diluent compound for freezing ram sperm can increase the efficiency of artificial insemination and consequently, the reproductive performance of sheep. Various biological properties such as antibacterial, anti-cancer, immunosuppressive, antioxidant and reproductive properties of royal jelly (RJ) are well known. The aim of this study was to investigate the possible synergistic effect of royal jelly in combination with glycerol and dimethyl sulfoxide (DMSO) in sperm cryopreservation extender of Romanov ram. The pooled semen samples from 5 Romanov rams were allocated into 3 experiments. The effect of 6% DMSO, 6% glycerol and a combination of 3% DMSO +3% glycerol co-supplemented with 1, 2 and 3% RJ was evaluated in 3 experiments. Samples were frozen by conventional slow freezing method and post-thaw parameters of total motility, progressive motility, plasma membrane integrity, DNA damage, apoptosis, enzyme activity of superoxide dismutase (SOD) and glutathione peroxidase (GPx), total antioxidant capacity (TAC) and malondialdehyde (MDA) were evaluated. The results showed that the percentage of motility, progressive motility, TAC, GPx, SOD and all sperm kinematic parameters except LIN in the group containing 2% RJ + 6% DMSO was higher than the control group (p 0.05). Some parameters such as progressive motility, sperm membrane integrity, TAC, GPx, VAP, VCL, STR and SRT in the group containing 2% RJ + 6% DMSO were more than (p 0.05) in the sperm group containing 1% RJ + 6% DMSO. MDA values in sperm groups containing 2% RJ + 6% DMSO were significantly (p 0.05) lower than the sperm containing 1% royal jelly and the control group. In the sperm group containing 2% RJ + 6% glycerol, sperm membrane integrity, TAC, GPx, SOD, progressive motility and all sperm kinematic parameters except VAP were higher and MDA values and sperm abnormalities were lower than the control group (p 0.05). The sperm group containing 1% RJ and 3% DMSO +3% glycerol had higher motility, progressive motility, membrane integrity, and all sperm kinematic parameters except VSL; and lower sperm abnormalities, DNA damage, apoptosis and MDA than the control group (p 0.05). As a general conclusion of this study, the addition of 2% RJ + 3% DMSO and 3% glycerol to the freezing extender improved microscopic and biochemical ram sperm parameters after the freeze-thaw process. Hence, moderate concentrations of royal jelly (2%) are sufficient to protect sperm from freezing damage, and high (3%) and low (1%) concentrations do not have a good cryoprotective effect.

The hydrogel sponges have prepared successfully from gelatin/chitosan (GEL/CTS) matrix with different polyols basing on scanning electron microscopy (SEM) and Fourier-transform infrared spectroscopy (FTIR) analysis. The results indicate that the compressibility and thermal properties (thermal gravimetric analysis, TGA) were improved significantly with adding the different polyols [poly(vinyl alcohol) – PVA, and poly(ethylene glycol)-poly(propylene glycol)- poly(ethylene glycol) – PEPEG] into GEL/CTS matrix. Specially, the as-prepared hydrogel sponges with superabsorbent behavior were also investigated with the attained water absorption capacity to 605 ​g ​g−1 and 585 ​g ​g−1 for GCV_1/1/0.3 and GCE_1/1/0.3 sponges, respectively, as well as the excellent water absorption capacity in a wide range of pH and salt concentration. Hence, these studies demonstrated that the hydrogel sponges are concerned as potential and promising candidates to apply especially in the fields of biomedicine and hygiene.

Recent confirmations of glyphosate-resistant Russian thistle (Salsola tragus L.) in Montana, Washington, and Oregon, warrant greater surveillance of herbicide-resistant Russian thistle in western Canada. A randomized-stratified survey of 315 sites in Manitoba was conducted in 2018 to determine the incidence of herbicide resistance in Russian thistle and other weeds sampled post-harvest. Russian thistle populations were collected from 14 of the 315 sites surveyed. None of these populations exhibited resistance to acetolactate synthase inhibitors (tribenuron/thifensulfuron), synthetic auxins (2,4-D ester or fluroxypyr), or glyphosate. This Manitoba survey of herbicide-resistant Russian thistle serves as a baseline for future surveillance efforts.

A microbial community structure of granules harvested from an anaerobic sludge blanket reactor treating phenolic wastewater was investigated using fluorescence in situ hybridization (FISH) and clone library construction. Clones of Syntrophorhabdaceae and Cryptanaerobacter were observed to be responsible for phenol degradation. For accurate taxonomic assignment of Cryptanaerobacter clones, phylogenetic analysis using nearly full-length 16S ribosomal RNA (rRNA) gene sequences was necessary. Three oligonucleotide probes were designed to detect the following three taxonomic groups: Syntrophorhabdaceae, Cryptanaerobacter, and Syntrophus. FISH analysis of thin sections of anaerobic granules showed a random distribution of Bacteria and Archaea. However, a well-defined distribution of Syntrophorhabdaceae, Cryptanaerobacter, and Syntrophus was observed. Cryptanaerobacter and Syntrophus were found on the outer layer of the granules and were closely associated with each other, while Syntrophorhabdaceae was located in the deeper part of the granules. Such specific distribution of the bacteria is most likely due to their metabolic association and affinity for the substrate. Phenol degradation in the granular sludge was observed to be carried out in the following way. First, Cryptanaerobacter converts phenol to benzoate, which is then degraded by Syntrophus into acetate. This syntrophic degradation of phenol occurs near the surface of the granule, where the phenol concentration is high. In the deeper part of the granule, where the phenol concentration is lower, Syntrophorhabdaceae degrades phenol into acetate. We observed that Syntrophorhabdaceae is less likely to produce benzoate as an intermediate to feed the neighboring organisms, which contradicts the theories presented by previous studies.
