Penicillium expansum is the causal agent of blue mold in harvested fruits and vegetables during storage and distribution, causing serious economic loss. In this study we seek the action modes of bifonazole against this pathogen. Bifonazole exhibited strong antifungal activity against P. expansum by inhibiting ergosterol synthesis. The ergosterol depletion caused damage to the cell structure and especially cell membrane integrity as observed by SEM and TEM. With increased unsaturated fatty acids contents, the cell membrane viscosity decreases and can no longer effectively maintain the cytoplasm, which ultimately decreases extracellular conductivity, changes intracellular pH and ion homeostasis. Exposure of hyphal cells to bifonazole shows that mitochondrial respiration is inhibited and reactive oxygen species (ROS) levels–including H2O2 and malondialdehyde (MDA) – are significantly increased. The emergence of severe acute syndrome coronavirus 2 (SARS-CoV-2) in December 2019 has led to the global COVID-19 pandemic. Although the symptoms of most COVID-19 patients are mild or self-curable, most of severe patients have sepsis caused by cytokine storms, which greatly increases the case fatality rate. Moreover, there is no effective drug that can limit the novel coronavirus thus far, so it is more needed to develop antiviral drugs for the SARS-CoV-2. In our research, we employed the techniques of molecular docking to screen 35 flavonoid compounds among which 29 compounds have Z-scores lower than − 6. Then, ( −)-gallocatechin gallate, ( +)-gallocatechin and baicalein were identified to have potent inhibitory activity against SARS-CoV-2 Mpro with IC50 values of 5.774 ± 0.805 μM, 13.14 ± 2.081 μM and 5.158 ± 0.928 μM respectively by FRET assay. Molecular docking results also showed that ( −)-gallocatechin gallate, ( +)-gallocatechin and baicalein can non-covalently bind to Mpro through π-π stacking and hydrogen bonds in the Cys145 catalytic site. We further evaluated the effect of ( −)-gallocatechin gallate and baicalein on cytokine storms using a mouse model of sepsis. ( −)-Gallocatechin gallate and baicalein significantly reduced sepsis of mouse models on weight, murine sepsis score, and survival rate and reduced the inflammatory factor levels, such as TNF-α, IL-1α, IL-4, and IL-10. Follicular helper T (TFH) cells are a specialized subset of CD4+ T cells that essentially support germinal center responses where high-affinity and long-lived humoral immunity is generated. The regulation of TFH cell survival remains unclear. Here we report that TFH cells show intensified lipid peroxidation and altered mitochondrial morphology, resembling the features of ferroptosis, a form of programmed cell death that is driven by iron-dependent accumulation of lipid peroxidation. Glutathione peroxidase 4 (GPX4) is the major lipid peroxidation scavenger and is necessary for TFH cell survival. The deletion of GPX4 in T cells selectively abrogated TFH cells and germinal center responses in immunized mice. Selenium supplementation enhanced GPX4 expression in T cells, increased TFH cell numbers and promoted antibody responses in immunized mice and young adults after influenza vaccination. Our findings reveal the central role of the selenium–GPX4–ferroptosis axis in regulating TFH homeostasis, which can be targeted to enhance TFH cell function in infection and following vaccination. We aim to explore the effects of emodin and its mechanisms on renal fibrosis (RF). We firstly modeled adriamycin-induced rat RF with unilateral nephrectomy. In vivo and in vitro pharmacological experiments were performed in this study. The presence of collagen deposition was detected by Masson staining. To verify whether emodin attenuates RF by monitoring autophagy, the immunohistochemistry staining for autophagy protein LC3B was performed. We conducted western blot to detect the expression of the autophagy-related proteins in EMT in vitro model after treating with emotin and BMP-7. In vivo, we demonstrated that emodin could improve renal dysfunction and decrease pathological damage of the kidney by activation of autophagy and inhibition of EMT. Upregulation of BMP-7 was recorded in the RF rats subjected to emodin treatment. In vitro studies, emodin has the capacity of reversing EMT and activating autophagy, and emodin could regulate the expression of BMP-7. The results revealed that the attenuation of EMT by emodin could be blocked after the inhibition of BMP-7 and suppression of autophagy. Our findings demonstrated that emodin alleviates EMT during RF by actuating autophagy through BMP-7, suggesting a role of BMP-7 in RF treatment and prevention. more...

Besides widely used surface passivation, engineering the film crystallization is an important and more fundamental route to improve the performance of all-inorganic perovskite solar cells. Herein, we have developed a urea-ammonium thiocyanate (UAT) molten salt modification strategy to fully release and exploit coordination activities of SCN- to deposit high-quality CsPbI3 film for efficient and stable all-inorganic solar cells. The UAT is derived by the hydrogen bond interactions between urea and NH4 + from NH4 SCN. With the UAT, the crystal quality of the CsPbI3 film has been significantly improved and a long single-exponential charge recombination lifetime of over 30 ns has been achieved. With these benefits, the cell efficiency has been promoted to over 20 % (steady-state efficiency of 19.2 %) with excellent operational stability over 1000 h. TFE3 gene fusions often place TFE3 under the control of a more active promoter and cause overexpression of the TFE3 proteins in renal cell carcinoma associated with Xp11.2 translocations (Xp11.2 tRCC). The purpose of this study was to investigate the transcriptional regulation and aggregation mechanism of NONO-TFE3 in NONO-TFE3 tRCC. In this study, we found that the nuclear aggregation of NONO-TFE3 fusion was significantly more than that of intact TFE3 or PRCC-TFE3 fusion. We observed that NONO fragment mediated-phase separation promoted stabilization and aggregation of NONO-TFE3 fusion. Meantime, we revealed that the positive regulation loop between NONO-TFE3 and NRF1 increased mitochondrial biosynthesis and metabolism in NONO-TFE3 tRCC. Therefore, the present study raises the possibility that mitochondrial metabolism is potentially a fruitful arena for NONO-TFE3 tRCC therapy. more...

Aspergillus niger prolyl endopeptidase (An-PEP) has become a research focus because of its advantages in specifically cleaving the C-terminal peptide bond of proline residues, especially it was an industrial food-grade acidic PEP. Aqueous two-phase system (ATPS) was first applied for separating An-PEP from fermentation broth. Via response surface method (RSM) experiment, an effectively separation of An-PEP was achieved by ATPS containing27% (w/w) ethanol and 14.5% (w/w) (NH4)2SO4 at pH 6.0 with the recovery of 90.29 ± 0.23% and purification coefficient of 15.35 ± 0.30. The purified An-PEP was characterized by sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS–PAGE), fourier transform infrared (FTIR) and fluorescence spectrometry. The optimum temperature and pH of An-PEP were 40 °C and 4.5–5.0, respectively. An-PEP was activated and stabilized by Ca2+ but inhibited by Fe3+. The enzymatic application of purified An-PEP was evaluated by hydrolyzing egg white protein (EWP) to prepare bioactive peptides. The obtained hydrolysates had good scavenging ability of OH and 2,2′-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) free radicals, angiotensin converting enzyme (ACE) inhibitory activity and anti-gout activity. The utilization of functional nano-additives provides us a novel research idea to deal with the in situ temperature sensing/monitoring of lubricating oil. In this work, monodispersed and luminescent calcium fluoride (CaF2) nanoparticles with surface modification were synthesized by a hydrothermal approach, which were employed as nano-additives for polyalphaolefin (PAO6) lubricating oil. Because of the surface modification of hydrophobic oleic acid molecule, the as-prepared CaF2 nanoparticles exhibited excellent oil solubility. The tribological results further suggested that after dispersing in PAO6 lubricating oil, the CaF2 nanoparticles could maintain its original friction and wear properties. In addition to the above basic characteristics for lubricating nano-additives, it was attractive to find that the as-synthesized CaF2 nanoparticles exhibited a doublet luminescence characteristic. The intensity ratio of the two emission peaks presented a nearly linear relationship to the surrounding temperature, which was also effective for CaF2 nanoparticles dispersed in PAO6 base oil. By comprehensive consideration of the oil dispersibility, tribological properties and temperature responsiveness, the as-synthesized CaF2 nanoparticles should be promising nano-additives for temperature monitoring of lubricating oil. more...

The promising synergistic electrocatalytic system of phosphate (PO43−) with the mediator cobalt(II) (for short E-Co(II)-PO43−) was employed to degrade cationic dye methylene blue (MB). The exploration in the electrocatalytic process revealed that the main intermediate active oxidation products were Co(III), accompanied with hydroxyl radicals and peroxodiphosphates (P2O84−). Their synergistic electrocatalytic degradation rate to MB and total organic carbon (TOC) was up to 100 and 60% in 40 min, respectively, which was 5 times and 2.6 times that in a direct electrocatalytic system, correspondingly. The degradation process of the E-Co(II)-PO43− system on MB started with the bond being broken at the N-C junction of the MB molecule and intermediate active oxidation substances being generated, such as phenothiazine, 2-amino-5-(N-methylformamide) benzene sulfonic acid and N1,N1-dimethyl-1,4 diaminobenzene. Then, the intermediates were degraded into aniline, phenol and benzene sulfonic acid, and eventually decomposed into inorganic substances like CO2 and water. The electrocatalytic degradation mechanism of E-Co(II)-PO43− system on MB was the combination of indirect oxidation of the intermediate oxidants like Co(III), P2O84− and the hydroxyl radical with direct electrocatalysis on the platinum titanium electrode, where the electrocatalytic oxidation of Co(III) was dominant. more...

Kesterite (CZTS(e)) solar cell has attracted tremendous interest for its feature: Earth-abundant composition, convenient fabrication process, environmental friendliness and low cost. However, the severe large voltage deficient in CZTS(e) solar cell remains a challenge for further enhancement of the power conversion efficiency (PCE). In this prospect, three mechanisms are proposed: Cu/Zn disorder, potential interfacial losses and Sn-related deep-level defects. Sn-related deep-level defects are the most detrimental and hardest factor to manipulate due to the low energy of the formation of atomic antisite and multivalency nature of Sn. Adding extrinsic elements into the absorber layer is of an important method to passivate Sn-related defects. Among them, incorporation of Ge (CZTS:Ge) shows great potential. Comparing to Sn, Ge is more likely to be stabilized at +4 oxidation state. DFT calculation suggests that the deep-level defect density of CZTS(e):Ge is much smaller than that of CZTS(e). The maximum trap-limited conversion efficiency can be promoted to 24.1% with Ge incorporation compared with 20.9% in intrinsic CZTS(e). Early works show that CZTS(e):Ge solar cells present relative higher V oc and has reached 12.3% power conversion efficiency (PCE), promising to champion the efficiency of CZTS(e) solar cell. Up to date, the benefits from Ge incorporation can be summarized in three aspects. First, Ge incorporation can induce the formation of Ge-Se low-temperature liquid phase as crystallization flux during annealing, which finally enlarges grain size and promote morphology. In the meantime, Sn-Se phases formation can be minimized through a modified reaction pathway brought by Ge doping. Second, Sn-related deep-level defects experimentally observed will be passivated when Ge is incorporated. Third, the energy difference between kesterite, stannite and primitive mixed CuAu is enlarged in Cu2ZnGeS4, indicating that Ge may stabilize kesterite in absorber and suppress band tailing caused by atomic disorder (or phase instability). To break through current efficiency bottleneck of CZTS(e) solar cells, we prospect the futural directions in constructing CZTS(e):Ge. First, Zn composition in absorber should be handled with great care to suppress the formation of GeZn-CuZn further. Next, modulation of the gradient band gap, achieved by vertical Ge-Sn compositional gradient in absorber, is also an important aspect. How to avoid the fast diffusion of Ge to form the vertical Ge-Sn gradient will be of the most essential question. Vertical control of independent Sn composition or Ge incorporation in the post-selenization process may be two potential solutions. Furthermore, the nature of the wide band gap of pure sulfide CZTS(e):Ge makes itself suitable to fabricate the top cell in tandem solar cell. Finally, interface modifications and band engineering in CZTS(e) solar cells will also benefit in constructing CZTS(e):Ge solar cells. more...

A series of modified poly(ether ether ketone)s containing different content of benzene pendant group in the molecular chain have been successfully synthesized with 1,1-bis(4-hydroxyphenyl)-1-phenylethane, bisphenol A and 4,4′-difluorobenzophenone by nucleophilic substitution. The inherent viscosities of obtained polymers were in the range of 0.261–0.889 dLg−1. They were found to have high glass transition temperatures (Tg) of 150–168°C and 148–169°C (examined by DSC and DMA, respectively), good thermal stability with 5% weight-loss temperature (T5%) of 439–469°C. The tensile strengths of the obtained polymers were 67.7–86.6 MPa, and the storage modulus was over 0.9 GPa at about 140°C. The modified poly(ether ether ketone)s showed the excellent solubility property, they could be processed by solution method. Additionally, the obtained polymers had good optical transmittance (>70%) at 400 nm, suggesting that they have potential to be applied to the heat-resistant optical films. more...

Six benzimidazole–naphthalimide naked-eye probes containing nitrobenzoquinone have been designed and synthesized to study the positional effect of –NO2 group on their anion recognition abilities in DMSO. The push–pull character of –NO2 rendered the anion-binding site hydrazinium –NH more acidic and thus easier for hydrogen bonds or deprotonation. So disubstituted isomer responded to F−, CN−, AcO− and H2PO4−; para-substituted isomer responded toward F− and CN−; and ortho-substituted isomer responded to F− due to the forming of intramolecular hydrogen bond between oxygen in nitro and hydrogen in hydrazine. The ortho-substituted isomer demonstrated highly sensitive and selective detection toward F− with a distinct color change from orange to blue-green accompanied by UV–Vis absorption redshift and fluorescence turn-on; the detection limits were in the range of 7 × 10−8 mol/L. Previous studies demonstrated that maternal exposure to di-n-butyl phthalate (DBP) resulted in developmental disorder of the male reproductive organ; however, the underlying mechanism has not been thoroughly elucidated to date. The present study was aimed to investigate the effects of maternal exposure to DBP on the formation of the Sertoli cell (SC)-based tight junctions (TJs) in the testes of male offspring mice and the underlying molecular mechanism. By observing the pathological structure and ultrastructure, permeability analysis of the testis of 22 day male offspring in vivo, and transepithelial electrical resistance measurement of inter-SCs in vitro, we found that the formation of TJs between SCs in offspring mice was accelerated, which was paralleled by the accumulation of TJ protein occludin at 50 mg/kg/day DBP exposure in utero and 0.1 mM monobutyl phthalate (MBP, the active metabolite of DBP) in vitro. Our in vitro results demonstrated that 0.1 mM MBP downregulated the expression of matrix metalloproteinase-2 (MMP-2) by inhibiting the activation of nuclear factor-κB (NF-κB)/cyclooxygenase-2 (COX-2)/prostaglandin E2 (PGE2) cascades via attenuated binding of NF-κB to both the MMP-2 promoter and COX-2 promoter. Taken together, the data confirmed that maternal exposure to a relatively low dose of DBP promoted the formation of testicular TJs through downregulation of NF-κB/COX-2/PGE2/MMP-2, which might promote the development of the testis during puberty. The purpose of this work was to conjugate phosvitin (Pv) with gallic acid (GA) to explore a new emulsifier that had both good emulsifying properties and antioxidant activity. The Pv-GA complex was prepared at a GA concentration of 1.5 mg/mL with pH 9.0. The Pv-GA complex obtained was identified by matrix-assisted laser desorption/ionization time-of-flight mass spectrometry and characterized with infrared, ultraviolet, and fluorescence spectra. The emulsifying activity and stability of the Pv-GA complex were slightly improved, and antioxidant activities was significantly enhanced. Furthermore, the Pv-GA complex was used to load conjugated linoleic acid (CLA) for microemulsion preparation. Results showed that the Pv-GA complex could increase the viscosity and lipid antioxidant capacity of Pv-GA/CLA microemulsion. The Pv-GA/CLA microemulsion had remarkable emulsifying activity, emulsifying stability, pH, and thermal stability and poor salt stability. more...

Thiocyanate could effectively inhibit bacteria in milk and extend the shelf life of milk. However, excessive addition will lead to health risks. Therefore, the determination of thiocyanate in raw milk has received a lot of attention, but the determination could be interfered with by other components in raw milk and the pre-treatment of raw milk is complex. In this study, a new pretreatment method combined with ion chromatography (IC) for rapid and sensitive determination of thiocyanate is proposed. An acetonitrile/(NH4)2SO4 aqueous two-phase system (ATPS) was developed for the separation and enrichment of thiocyanate in raw milk. Response surface methodology was performed to optimize the extraction conditions and an efficient pretreatment were obtained using ATPS composed of 42% acetonitrile (w/w) and 16% (NH4)2SO4 (w/w), with the pH 4.7, and the recovery of thiocyanate reached 107.24 ± 0.5%, and the enrichment ratio was 10.74 ± 0.03. IC was used to establish a thiocyanate enrichment method. The linear range was from 0.05 to 15 mg/L and R2 = 0.998, the limit of detection (LOD) was 0.2 μg/L, the limit of quantification (LQD) was 0.6 μg/L. Background Xp11.2 translocation renal cell carcinoma (tRCC) is defined by translocation of the transcription factor E3 (TFE3) gene located on chromosome Xp11.2. Due to the high incidence in women, 17β-estradiol (E2) may be a factor influencing TFE3 breaks, and the topoisomerase II (TOP2) poison is considered one of the important risk factors in mediating DNA breaks. In this study, we investigated the potential pathogenesis of Xp11.2 tRCC using the renal epithelial cell line HK-2. Methods Immunofluorescence assay was performed to analyze DNA breaks by quantifying phosphorylation of H2AX (γH2AX), and the micronuclei (MN) assay was designed for monitoring chromosome breaks. The chromatin immunoprecipitation (CHIP) was used to detect whether proteins bound to specific DNA site, and the co-immunoprecipitation (Co-IP) was used to confirm whether proteins bound to other proteins. In some experiments, siRNA and shRNA were transfected to knockdown target genes. Results Our results demonstrated that DNA double-strand breaks were mediated by TOP2β in HK-2 cells, and this process could be amplified through estrogen receptor α (ERα)-dependent pathway induced by E2. After performing translocation site analysis using target region sequencing data in two Xp11.2 tRCC cell lines and ten Xp11.2 tRCC patients, we confirmed that TOP2β and ERα could both bind to TFE3 translocation sites directly to mediate DNA breaks in a E2-dependent manner. However, TOP2β and ERα were not observed to have direct interaction, indicating that their collaborative may be implemented in other ways. Besides, TFE3 was found to be upregulated through NRF1 with increasing E2 concentration, which could increase the risk of TFE3 breaks. Conclusion Our results indicate that E2 amplifies TOP2β-mediated TFE3 breaks by ERα-dependent pathway, and E2 upregulates TFE3 by NRF1 to increase the risk of TFE3 breaks. Abscisic acid (ABA) plays a key role in fruit development and ripening in non-climacteric fruit. A variety of metabolites such as sugars, anthocyanins, fatty acids, and several antioxidants, which are regulated by various phytohormones, are important components of fruit quality in grape. Here, grape cultivar “Ruiduhongyu” was used to investigate the relationship between endogenous phytohormones and metabolites associated to grape berry quality under exogenous ABA treatment. 500 mg/L ABA significantly improved the appearance parameters and the content of many metabolites including sugar, anthocyanin, and other compounds. Exogenous ABA also increased the contents of ABA, auxin (IAA), and cytokinins (CTKs), and transcription level of ABA biosynthesis and signaling related genes in fruit. Furthermore, a series of genes involved in biosynthesis and the metabolite pathway of sugars, anthocyanins, and fatty acids were shown to be significantly up-regulated under 500 mg/L ABA treatment. In addition, Pearson correlation analysis demonstrated that there existed relatively strong cooperativities in the ABA/kinetin (KT)-appearance parameters, ABA/IAA/KT-sugars, ABA/indolepopionic acid (IPA)/zeatin riboside (ZR)-anthocyanins, and gibberellin 3 (GA3)/methyl jasmonate (MeJA)-fatty acids, indicating that 13 kinds of endogenous phytohormones induced by ABA had different contributions to the accumulation of quality-related metabolites, while all of them were involved in regulating the overall improvement of grape fruit quality. In this study, a new p-type π-conjugated ladder-like polymer, poly(3,3′-(((2-(4,8-bis(5-(2-ethylhexyl)thiophen-2-yl)-6-methylbenzol[1,2-b:4,5-b′]dithiophen-2-yl)-5-methyl-1,4-phenylene)bis(oxy))bis(hexane-6,1-diyl))bis(1,1,1,3,5,5,5-heptamethyltrisiloxane)) (P-Si), has been designed for SnO2-based perovskite solar cells. Our investigation reveals that this π-conjugated ladder-like polymer system decorated with hydrophobic alkyl chains and siloxane chains is likely the reason for the improvement in the surface morphology and crystallinity of the perovskite films, leading to reduced defect states and enhanced device stability. In addition, P-Si with a suitable highest occupied molecular orbital energy level (−5.41 eV) can act as a hole transport medium between the perovskite and spiro-OMeTAD to enhance the excellent hole transportation. As a result, power conversion efficiency (PCE) as high as 21.5% with 21.3% steady-state PCE was achieved. Typically, the device with P-Si can retain 98% of the original average PCE after being stored in an ambient environment with 20% relative humidity for 120 days, indicating its good long-term stability. Furthermore, after aging for 100 h under illumination at the maximum power point in a nitrogen atmosphere, the P-Si modified device retains over 98% of its initial performance. more...

The dormancy-associated MADS-box (DAM) genes PpDAM5 and PpDAM6 have been shown to play important roles in bud endodormancy; however, their molecular regulatory mechanism in peach is unclear. In this study, by use of yeast one-hybrid screening, we isolated a TEOSINTE BRANCHED1/CYCLOIDEA/PROLIFERATING CELL FACTOR transcription factor, PpTCP20, in the peach cultivar ‘Zhongyou 4’ (Prunus persica var. nectarina). The protein was localized in the nucleus and was capable of forming a homodimer. Electrophoretic mobility shift assays demonstrated that PpTCP20 binds to a GCCCR element in the promoters of PpDAM5 and PpDAM6, and transient dual luciferase experiments showed that PpTCP20 inhibited the expression of PpDAM5 and PpDAM6 as the period of the release of flower bud endodormancy approached. In addition, PpTCP20 interacted with PpABF2 to form heterodimers to regulate bud endodormancy, and the content of abscisic acid decreased with the release of endodormancy. PpTCP20 also inhibited expression of PpABF2 to regulate endodormancy. Taken together, our results suggest that PpTCP20 regulates peach flower bud endodormancy by negatively regulating the expression of PpDAM5 and PpDAM6, and by interacting with PpABF2, thus revealing a novel regulatory mechanism in a perennial deciduous tree., PpTCP20 controls the release of endodormancy in peach flower buds by negatively regulating the expression of PpDAM5 and PpDAM6, and by interacting with PpABF2. more...

Ionic liquids (ILs) are solvents with negligible volatility and corrosiveness that are non-flammable and thermally stable. These environmentally friendly properties allow ILs to be widely used in various applications. However, the high cost of ILs has been the major constraint that limits their widespread use in industry. In this paper, we report the application of a two-dimensional nanofiltration membrane, MoS2, to recover ILs from their aqueous solution since ILs are often diluted with water in many applications. MoS2 membranes from the common octahedral coordinated (1T) phase possess a laminar structure that allows water molecules to permeate through the interlayer nanochannels between nanosheets. Often the interlayer spacing is smaller than many ILs, enabling separation of water from ILs including 1-butyl-3-methylimidazolium tetrafluoroborate (BmimBF4) and 1-hexyl-3-methylimidazolium chloride (HmimCl). In this paper we first demonstrated that the rejection rate of BmimBF4 and HmimCl from IL/water mixtures reached 40.8% and 86.2% respectively. More importantly, we found that IL rejection rate can be further increased by 25% for BmimBF4, 8% for HmimCl under higher operating pressures, indicating that the applied transmembrane pressure reduces the interlayer spacing of MoS2 membranes. This hypothesis was proved by our XRD characterization. Finally, the effects of hydrophilicity and charge of dopamine-modified 2D MoS2 membranes on the IL rejection were also investigated, revealing that increased surface hydrophilicity and more positively charged membrane surfaces enabled further increase in IL rejection. more...

In the last two decades, with the wide use of azoles, antifungal resistance among Candida parapsilosis has considered a matter of concern worldwide. The aim of this study is to evaluate the antifungal potentials of tetrandrine (TET) alone and in combination with fluconazole (FLC)/voriconazole (VRC) against C. parapsilosis. Susceptibility tests were performed by microdilution method, checkerboard assay, time-kill test, spot assay. Subsequently, rhodamine 6G efflux test and the expressions of transporter related genes, namely CDR1 and MDR1 for C. parapsilosis were analyzed by qRT-PCR. The susceptibility test showed that TET presented strong synergism with FLC and VRC with fractional inhibitory concentration index (FICI) in a range of 0.094-0.562. The susceptibility results were also confirmed by spot assay and time-kill studies. With TET treatment, a vast quantity of rhodamine 6G could not be pumped out from the cells as considerably intracellular red fluorescence was accumulated. Meanwhile, the expressions of efflux-associated genes presented varying degrees of inhibition. These results indicated that TET was a decent antifungal synergist to promote the antifungal efficacy of FLC/VRC, and the underlying antifungal mechanism might be associated with the inhibition of efflux pump and the elevation of intracellular drug content. more...

Foodborne nanoparticles produced from food components during thermal processing have received attention due to their particular properties. Herein, the presence of fluorescent carbon dots (CDs) was reported from mackerel flesh roasted at 230 °C in an electronic oven, and the CD capability for free radical scavenging was investigated. The average size of the CDs was about 2.2 nm with a size distribution in the range of 0.9–3.5 nm as characterized with transmission electron microscopy (TEM). X-ray photoelectron spectroscopy (XPS) showed that the mackerel CDs mainly contain C, N and O. Fourier transform infrared (FTIR) analysis showed the existence of hydroxyl, amino, and carboxyl groups on the mackerel CD surface. The fluorescence lifetime was about 9.1 ns. Two absorption peaks at 277 and 328 nm were observed in UV–vis spectra, which are assigned to the π-π* and n-π* charge-transfer, respectively. The mackerel CDs showed excellent photostability in various metal ion solutions and pH environment. In addition, these CDs exhibited strong dose-dependent scavenging capability for hydroxyl radicals (·OH) and methyl radicals (·CH3) produced from methylene blue (MB)/visible-light photosensitization system. BACKGROUND Sea cucumber (Stichopus japonicus) is easy to autolysis in response to a variety of environmental and mechanical factors. In the current study, collagen fibres were extracted from fresh sea cucumber body wall and then incubated with endogenous matrix metalloprotease (MMP) of sea cucumber. Scanning electron microscopy (SEM), differential scanning calorimetry (DSC), chemical analysis and sodium dodecyl sulphate polyacrylamide gel electrophoresis (SDS-PAGE) analysis were utilized to demonstrate the changes in collagen fibres, collagen fibrils and collagen proteins. Moreover, a verification experiment was also carried out to confirm the contribution of MMP to the autolysis of sea cucumber. RESULTS Endogenous MMP caused complete depolymerization of collagen fibres into smaller collagen fibril bundles and collagen fibrils due to the fracture of proteoglycan interfibrillar bridges. Meanwhile, endogenous MMP also caused partial degradation of collagen fibrils by releasing soluble hydroxyproline and pyridinium cross-links. Furthermore, the treatment with MMP inhibitor (1,10-phenanthroline) prevented the autolysis of tissue blocks from S. japonicus dermis. CONCLUSION Endogenous MMP was the key enzyme in the autolysis of sea cucumber, while its action still focused on high-level structures of collagens especially collagen fibres. © 2019 Society of Chemical Industry. more...

The present study reports a novel method for hydrogen generation and simultaneous removal of Cr(VI) from synthetic wastewater using NaBH4 as the reducing agent and Fe-Al-Si composite as accelerator. The results showed that the hydrogen generation yields of NaBH4 occurred at low pH, high temperature and stirring speed. The presence of Cr(VI) was found to inhibit hydrogen generation at the initial pH 3.0, especially at low temperature conditions. Increasing temperature resulted in the increase of hydrogen generation, whereas a higher reduction rate of Cr(VI) was obtained at low temperature. Therefore, to alleviate the contradiction between hydrogen generation and Cr(VI) removal with respect to temperature, we introduced reduction accelerator by preparing Fe-Al-Si composite using wasted fly ash as raw materials. The resultant hydrogen generation yield could be enhanced from 32.04 to 80.70%, and total Cr removal was increased from 46.72 to 98.96% at 30 °C. First, H+ produced by hydrolysis of Fe-Al-Si composite improves the hydrolysis ability of NaBH4, thus promoting its ability to reduce Cr(VI) and to produce hydrogen by itself. The Cr(VI) reduction and hydrogen generation process are competitive for H+, which is particularly evident at insufficient H+. Second, at low doses, Fe3+ exhibited a lower ability to promote hydrogen generation and simultaneous Cr(VI) removal than Al3+. One of the reasons for the low promotion ability of Fe3+ in the whole pH range was the formation of Fe0 at pH less than 3.0, and the other was the weaker hydrolysis ability of Fe3+ itself at pH greater than 3.0. more...

Tb(acac)3 (TAH) and Eu(TTA)3Phen (ETP) doped polyacrylonitriles (PAN) have been electrospun into heterogeneous hybrid TAH/ETP/PAN and core-shell structured [(TAH/PAN)core + (ETP/PAN)shell] wire-shaped flexible nanofibers with diameter of ∼350 nm. Differential photon releasing behaviors reveal the double-color-emitting potential of Tb3+/Eu3+ combination systems and verify the effectiveness of coaxial configuration in avoiding the cross-relaxation between different rare earth ions. Luminous fluxes of the hybrid and the coaxial systems exceed 283.5 and 326.3 μlm under the excitation of 308 nm UVB-LED pump source, respectively. By adopting coaxial electrospun configuration, the color coordinates are tuned from (0.561, 0.332) in TAH/ETP/PAN to (0.435, 0.366) in [(TAH/PAN)core + (ETP/PAN)shell] fibers, which demonstrates that the core-shell structure is promising to extend the dimension of color tuning. The fungal pathogen Botrytis cinerea is the causal agent of devastating gray mold diseases in many economically important fruits, vegetables, and flowers, leading to serious economic losses worldwide. In this study, a novel actinomycete NEAU-LD23T exhibiting antifungal activity against B. cinerea was isolated, and its taxonomic position was evaluated using a polyphasic approach. Based on the genotypic, phenotypic and chemotaxonomic data, it is concluded that the strain represents a novel species within the genus Streptomyces , for which the name Streptomyces botrytidirepellens sp. nov. is proposed. The type strain is NEAU-LD23T (=CCTCC AA 2019029T=DSM 109824T). In addition, strain NEAU-LD23T showed a strong antagonistic effect against B. cinerea (82.6±2.5%) and varying degrees of inhibition on nine other phytopathogenic fungi. Both cell-free filtrate and methanol extract of mycelia of strain NEAU-LD23T significantly inhibited mycelial growth of B. cinerea. To preliminarily explore the antifungal mechanisms, the genome of strain NEAU-LD23T was sequenced and analyzed. AntiSMASH analysis led to the identification of several gene clusters responsible for the biosynthesis of bioactive secondary metabolites with antifungal activity, including 9-methylstreptimidone, echosides, anisomycin, coelichelin and desferrioxamine B. The OVATE family protein (OFP) genes (OFPs) have been shown to respond to salt stress in plants. However, the regulatory mechanism for salt tolerance of the peach (Prunus persica) OFP gene PpOFP1 has not been elucidated. In this study, using yeast two-hybrid screening, we isolated a nucleus-localized ZF-HD_dimer domain protein PpZFHD1, which interacts with the PpOFP1 protein in the peach cultivar “Zhongnongpan No.10”. A segmentation experiment further suggested that the interaction happens more specifically between the N-terminal, contains ZF-HD_dimer domain, of PpZFHD1 and the C-terminal, consists of OVATE domain, of PpOFP1. Additionally, quantitative real-time polymerase chain reaction (qRT-PCR) experiments indicate that transcription of these two genes are induced by 200 mmol/L (mM) NaCl treatment. Heterogeneous transformation experiments suggested that the growth status of transformed yeast strain over-expressing each of these two genes was more robust than that of control (CK). Furthermore, transgenic tomato plants over-expressing PpOFP1 were also more robust. They had a higher content of chlorophyll, soluble proteins, soluble sugars, and proline. Activities of the superoxide dismutase (SOD), peroxidase (POD), and catalase (CAT) in these plants were higher, and tissues from these plants exhibited a lower relative conductivity and malondialdehyde (MDA) content. These results suggest that PpOFP1 physically interacts with PpZFHD1 and confers salt tolerance to tomato and yeast, thus revealing a novel mechanism for regulating salt tolerance in peach and other perennial deciduous trees. more...

The coronavirus disease 2019 (COVID-19) has spread widely around the world and has seriously affected the human health of tens of millions of people. In view of lacking anti-virus drugs target to SARS-CoV-2, there is an urgent need to develop effective new drugs. In this study, we reported our discovery of SARS-CoV-2 Mpro inhibitors. We selected 15 natural compounds, including 7 flavonoids, 3 coumarins, 2 terpenoids, one henolic, one aldehyde and one steroid compound for molecular docking and enzymatic screening. Myricetin were identified to have potent inhibit activity with IC50 3.684 ± 0.076 μM in the enzyme assay. The binding pose of Myricetin with SARS-CoV-2 Mpro was identified using molecular docking method. In the binding pocket of SARS-CoV-2 Mpro, the chromone ring of Myricetin interacts with His41 through π-π stacking, and the 3’-, 4’- and 7-hydroxyl of Myricetin interact with Phe140, Glu166and Asp187 through hydrogen bonds. Significantly, our results showed that Myricetin has potent effect on bleomycin-induced pulmonary inflammation by inhibiting the infiltration of inflammatory cells and the secretion of inflammatory cytokines IL-6, IL-1α, TNF-α and IFN-γ. Overall, Myricetin may be a potential drug for anti-virus and symptomatic treatment of COVID-19. more...

Photoperiod strictly controls vegetative and reproductive growth stages in soybean (Glycine max). A soybean GmRAV (Related to ABI3/VP1) transcription factor containing both AP2 and B3 domains was shown to be a key component of this process. We identified six polymorphisms in the GmRAV promoter that showed significant association with flowering time and maturity of soybean in one or multiple environments. Soybean varieties with minor polymorphism exhibited a longer growth period contributing to soybean adaptation to lower latitudes. The cis-acting element GT1CONSENSUS motif of the GmRAV promoter controlled the growth period, and the major allele in this motif shortened duration of late reproductive stages by reducing GmRAV expression levels. Three GmRAV-overexpressing (GmRAV-ox) transgenic lines displayed later flowering time and maturity, shorter height and fewer numbers of leaves compared with control plants, whereas transgenic inhibition of GmRAV expression resulted in earlier flowering time and maturity and increased plant height. Combining DNA affinity purification sequencing and RNA sequencing analyses revealed 154 putative target genes directly bound and transcriptionally regulated by GmRAV. Two GmRAV binding motifs [C(A/G)AACAA(G/T)A(C/T)A(G/T)] and [C(T/A)A(C)C(T/G)CTG] were identified, and acting downstream of E3E4, GmRAV repressed GmFT5a transcriptional activity through binding a CAACA motif, thereby delaying soybean growth and extending both vegetative and reproductive phases. more...

Maize (Zea mays L.) is a chilling-sensitive plant. Chilling stress in the early seedling stage seriously limits the growth, development, productivity and geographic distribution of maize. Salicylic acid (SA) is a plant growth regulator involved in the defenses against abiotic and biotic stresses as well as in plant development. However, the physiological mechanisms underlying the effects of foliar applied SA on different maize inbred lines under chilling stress are unclear. Two inbred lines, cold-sensitive cv. C546 and cold-tolerant cv. B125, were used to study the effects of SA on the growth and physiology of maize seedlings. The results showed that the application of SA at 50 mg/L on the leaves of maize seedlings under 4 °C decreased the relative electrolyte conductivity (REC) and the malondialdehyde (MDA) and reactive oxygen species (ROS) (H2O2 and O2−) content due to increased superoxide dismutase (SOD), peroxidase (POD), catalase (CAT) and ascorbate peroxidase (APX) activity, SA also improved photosynthesis in the seedlings through increased chlorophyll content, enhanced Pn and Gs, and decreased Ci. SA application also increased the proline content and the relative water content (RWC) in the maize seedlings, thereby improving their osmotic adjustment capacity. The increase rate caused by SA of plant height and dry weight in C546 were 10.5% and 5.4% higher than that in B125 under 4 °C. In conclusion, SA promotes maize seedling growth and physiological characteristics, thus enhancing chilling resistance and the effect of SA on the chilling resistance of cold-sensitive cv. was stronger than that on cold-tolerant cv. at the low temperature. more...

The dormancy-associated MADS-box (DAM) gene DAM5 has crucial roles in bud endodormancy; however, the molecular regulatory mechanism of PpDAM5 in peach (Prunus persica) has not been elucidated. In this study, using yeast two-hybrid screening, we isolated a BTB-TAZ Domain Protein PpBT3, which interacts with PpDAM5 protein, in the peach cultivar 'Chun xue'. As expected, we found that abscisic acid (ABA) maintained bud endodormancy and induced expression of the PpDAM5 gene, and that over-expressing PpDAM5 in Arabidopsis thaliana repressed seed germination. In contrast, over-expressing PpBT3 in A. thaliana promoted seed germination, and conferred resistance to ABA-mediated germination inhibition. Additionally, a qRT-PCR (quantitative real-time polymerase chain reaction) experiment suggested that the transcript level of PpBT3 gradually increased towards the endodormancy release period, which is the opposite trend of the expression pattern of PpDAM5. Our results suggest that PpBT3 modulates peach bud endodormancy by interacting with PpDAM5, thus revealing a new mechanism for regulating bud dormancy of perennial deciduous trees. more...

Background NONO-TFE3 translocation renal cell carcinoma (NONO-TFE3 tRCC) is one subtype of RCCs associated with Xp11.2 translocation/TFE3 gene fusions RCC (Xp11.2 tRCCs). Long non-coding RNA (lncRNA) has attracted great attention in cancer research. The function and mechanisms of TRAF3IP2 antisense RNA 1 (TRAF3IP2-AS1), a natural antisense lncRNA, in NONO-TFE3 tRCC remain poorly understood. Methods FISH and qRT-PCR were undertaken to study the expression, localization and clinical significance of TRAF3IP2-AS1 in Xp11.2 tRCC tissues and cells. The functions of TRAF3IP2-AS1 in tRCC were investigated by proliferation analysis, EdU staining, colony and sphere formation assay, Transwell assay and apoptosis analysis. The regulatory mechanisms among TRAF3IP2-AS1, PARP1, PTEN and miR-200a-3p/153-3p/141-3p were investigated by luciferase assay, RNA immunoprecipitation, Western blot and immunohistochemistry. Results The expression of TRAF3IP2-AS1 was suppressed by NONO-TFE3 fusion in NONO-TFE3 tRCC tissues and cells. Overexpression of TRAF3IP2-AS1 inhibited the proliferation, migration and invasion of UOK109 cells which were derived from cancer tissue of patient with NONO-TFE3 tRCC. Mechanistic studies revealed that TRAF3IP2-AS1 accelerated the decay of PARP1 mRNA by direct binding and recruitment of N6-methyladenosie methyltransferase complex. Meanwhile, TRAF3IP2-AS1 competitively bound to miR-200a-3p/153-3p/141-3p and prevented those from decreasing the level of PTEN. Conclusions TRAF3IP2-AS1 functions as a tumor suppressor in NONO-TFE3 tRCC progression and may serve as a novel target for NONO-TFE3 tRCC therapy. Coal-derived carbon nanofibers (CCNFs) have been recently found to be a promising and low-cost electrode material for high-performance supercapacitors. However, the knowledge gap still exists between holistic understanding of coal precursors derived from different solvents and resulting CCNFs’ properties, prohibiting further optimization of their electrochemical performance. In this paper, assisted by laser desorption/ionization (LDI) and gas chromatography–mass spectrometry (GC–MS) technologies, a systematic study was performed to holistically characterize mass distribution and chemical composition of coal precursors derived from various ionic liquids (ILs) as extractants. Sequentially, X-ray photoelectron spectroscopy (XPS) revealed that the differences in chemical properties of various coal products significantly affected the surface oxygen concentrations and certain species distributions on the CCNFs, which, in turn, determined the electrochemical performances of CCNFs as electrode materials. We report that the CCNF that was produced by an oxygen-rich coal fragment from C6mimCl ionic liquid extraction showed the highest concentrations of quinone and ester groups on the surface. Consequentially, C6mimCl-CCNF achieved the highest specific capacitance and lowest ion diffusion resistance. Finally, a symmetric carbon/carbon supercapacitor fabricated with such CCNF as electrode delivered an energy density of 21.1 Wh/kg at the power density of 0.6 kW/kg, which is comparable to commercial active carbon supercapacitors. more...

A chiral sensor with optical rotation detection based on weak measurement for the kinetic study of sucrose hydrolysis is presented. Based on the polarization modulation to the pre-selection state, the optical rotation of chiral sample was accurately determined through the central wavelength shift of the output spectrum. With this approach, the concentration response curves of sucrose and its hydrolysis products, i.e., fructose and glucose, were experimentally obtained for the hydrolysis analysis. By collecting the output spectrum with a frequency of 100 Hz and fitting the central wavelength shift synchronously during the measurement, the sucrose hydrolysis process was monitored in real time. Different hydrolysis conditions with varied concentration of invertase enzyme and citrate were implemented in this work. As a consequence, the real-time hydrolysis curves of the hydrolysis process with distinct velocities was achieved and analyzed. Such a kinetic monitoring about sucrose hydrolysis with optical rotation detection technology played a critical role in the researches involving sucrose, and also revealed the great potential of weak measurement in intersections, such as food safety inspection and chemical analysis. more...

In China, more than 3.5 million tons of Camellia oleifera discarded shells are produced every year. This work first prepared phosphorus-containing biochar (PBC) from C. oleifera shells and was successfully applied to the efficient removal of tetracycline (TC) from solutions. The prepared PBC exhibits superior TC adsorption capacity of 451.5 mg/g, and TC uptake rapidly reached 315.5 mg/g at the first 5 min (C0 = 50 mg/L). Furthermore, PBC also shows excellent applicability to the broad range pH value (1–9) and superior selective removal in the presence of various high concentration coexisting ions (1 mM). Mechanisms underlying TC adsorption were also put forward, and analysis suggested that pyrophosphate-like surface functional groups (C-O-P bond) played a critical role in this process. Notably, treating pharmaceutical wastewater with PBC can efficiently reduce chemical oxygen demand (COD) and total organic carbon (TOC) concentration below the discharge standard of China (GB21904-2008). more...

We report a unique synthesis of a nanoporous two-dimensional covalent organic framework (COF) from a single self-assembling monomer. The novel 2D-COF (DPCOF) contains two chemically distinct nanopores, one that can coordinate transition metals and a second more unreactive pore that is approximately the size of small gas molecules. We then demonstrate the synthetic flexibility of the material by coordination of various transition metals into the framework and finally its application as a membrane for degasification and gas separation. Density functional theory (DFT) and molecular mechanics (MM) calculations were used to inform the computational construction of material models, allowing for the interpretation of pXRD data to elucidate the crystallographic structure of the novel materials, including the revelation that disordered monomer vacancies are likely present in the material. more...

Optoelectronic biomaterials have recently emerged as a potential treatment option for neurodegenerative diseases, such as optic macular degeneration. Though initial works in the field have involved bulk heterojunctions mimicking solar panels with photovoltaics (PVs) and conductive polymers (CPs), recent developments have considered abandoning CPs in such systems. Here, we developed a simple antioxidant, biocompatible, and fibrous membrane heterojunction composed of photoactive polymer poly(3-hexylthiophene) (P3HT), polycaprolactone (PCL) and polypyrrole (PPY), to facilitate neurogenesis of PC-12 cells when photo-stimulated in vitro. The photoactive prototype, referred to as PCL-P3HT/PPY, was fabricated via polymerization of pyrrole on electro-spun PCL-P3HT nanofibers to form a membrane. Four experimental groups, namely PCL alone, PCL/PPY, PCL-P3HT and PCL-P3HT/PPY, were tested. In the absence of the CP, PCL-P3HT demonstrated lower cell survival due to increased intracellular reactive oxygen/nitrogen species production. PCL-P3HT/PPY rescued these cells by virtue of scavenging radicals, where the CP, PPY, acted as an antioxidant. Apart from having lower impedance, the material also enhanced neurogenesis of PC-12 cells when photo-stimulated, compared to the traditional PCL-P3HT. Lastly, the in vitro system with PC-12 was used to demonstrate the practicality of the material for potential use as a cellular patch in optic and nerve regeneration. This work demonstrated the importance of maintaining PV-CP heterojunctions while simultaneously providing an optoelectrical platform for neural and optical tissue engineering. Statement of significance Regeneration and repair of injured nervous systems have always been a major clinical challenge. Stem cell therapy is a promising approach for nerve regeneration, and opto-electrical stimulation, which converts light into an electrical signal, has been shown to efficiently regulate stem cell behaviors with enhanced neurogenesis. We developed a micro-fibrous membrane, composed of photoactive polymer, P3HT, scaffold material PCL and conductive polymer PPY. Our heterojunction system improved cell survival via PPY quenching PCL-P3HT-generated cell-damaging reactive oxygen species. PPY also conducted electrons produced from light-stimulated P3HT to promote neurogenesis. This photoactive microfiber biomaterial has great potential as a highly biocompatible and efficient platform to wirelessly promote neurogenesis and survival. BACKGROUND α-lactalbumin (α-La) is of great interest to the industry as a result of its excellent functional properties and nutritional value. Aqueous two-phase flotation (ATPF) of thermo-sensitive polymer poly (ethylene glycol-ran-propylene glycol) monobutyl ether (UCON) and KH2 PO4 was applied to directly separate and purify α-La from milk whey, which was purposed to simplify the production process and reduced cost of production. RESULTS The effect of ATPF composition and operating parameters on the flotation efficiency (E) and purity of α-La were investigated. The optimal conditions included 2 min of premixing time, 30 mL min-1 flow velocity and 20 min of flotation time, whereas the composition conditions comprised 35.0 mL 0.18 g mL-1 phosphate solution (containing 10% (cow milk whey/salt solution, v/v) cow milk whey, 50 ppm defoamer and 2 g NaCl) and 5.0 mL of 40% (w/w) UCON solution. Under the optimal conditions, E of α-La was 95.67 ± 1.04% and purity of α-La was 98.78 ± 1.19%. UCON was recovered by a thermally-induced phase separation and reused in next ATPF process without reducing E of α-La. Purified α-La was characterized by several key technologies. The results indicated that α-La in cow milk whey could be directly separated and purified by the ATPF and the purity was satisfactory. Moreover, it was suggested there was no obvious structure difference between the α-La separated by ATPF and the α-La standard. CONCLUSION The present study enabled the recycling of UCON, providing an effective, economically viable and environmentally friendly approach for the separation and purification of protein. © 2021 Society of Chemical Industry. more...

This study aimed to explore the synergistic action of pentapeptides Gln-Met-Asp-Asp-Gln (QMDDQ) and Ala-Gly-Leu-Pro-Met (AGLPM) on memory improvement against scopolamine-induced impairment in mice compared to those of either peptide alone. In behavioral tests, the codelivery of QMDDQ and AGLPM was superior to the individual supplements of either peptide alone not only in enhancing the memory ability at training trials but also in recovering the memory impairment in scopolamine-induced amnesiac mice in test trials. Furthermore, combination treatment with QMDDQ and AGLPM could significantly reduce the acetylcholinesterase (AChE) level and increase the acetylcholine (ACh) level in the hippocampus, and noticeably improve the pathological morphology of the neuron cells in hippocampal regions CA1 and CA2 and dentate gyrus (DG). The cyclic adenosine 3′,5′-monophosphate (cAMP)/protein kinase A (PKA) pathway ofCandida albicansresponds to nutrient availability to coordinate a series of cellular processes for its replication and survival. The elevation of cAMP for PKA signaling must be both transitory and tightly regulated. Otherwise, any abnormal cAMP/PKA pathway would disrupt metabolic potential and ergosterol synthesis and promote a stress response. One possible mechanism for controlling cAMP levels is direct induction of the phosphodiesterasePDE2gene by cAMP itself. Our earlier studies have shown that most single-gene-deletion mutants of the mitochondrial electron transport chain (ETC) complex I (CI) are hypersensitive to fluconazole. To understand the fluconazole hypersensitivity observed in these mutants, we focused upon the cAMP/PKA-mediated ergosterol synthesis in CI mutants. Two groups of the ETC mutants were used in this study. Group I includes CI mutants. Group II is composed of CIII and CIV mutants; group II mutants are known to have greater respiratory loss. All mutants are not identical in cAMP/PKA-mediated ergosterol response. We found that ergosterol levels are decreased by 47.3% in thendh51Δ (CI core subunit mutant) and by 23.5% ingoa1Δ (CI regulator mutant). Both mutants exhibited a greater reduction of cAMP and excessive trehalose production compared with other mutants. Despite the normal cAMP level, ergosterol content decreased by 33.0% in the CIII mutantqce1Δ as well, thereby displaying a cAMP/PKA-independent ergosterol response. While the two CI mutants have some unique cAMP/PKA-mediated ergosterol responses, we found that the degree of cAMP reduction correlates linearly with a decrease in total nicotinamide adenine dinucleotide (NAD) levels in all mutants, particularly in the seven CI mutants. A mechanism study demonstrates that overactivePDE2and cPDE activity must be the cause of the suppressive cAMP-mediated ergosterol response in thendh51Δ andgoa1Δ. While the purpose of this study is to understand the impact of ETC proteins on pathogenesis-associated cellular events, our results reveal the importance of Ndh51p in the regulation of the cAMP/PKA pathway through Pde2p inhibition in normal physiological environments. As a direct link between Ndh51p and Pde2p remains elusive, we suggest that Ndh51p participates in NAD homeostasis that might regulate Pde2p activity for the optimal cAMP pathway state. more...

Background A previous study showed that dibutyl phthalate (DBP) exposure disrupted the growth of testicular Sertoli cells (SCs). In the present study, we aimed to investigate the potential mechanism by which DBP promotes juvenile SC proliferation in vivo and in vitro. Methods Timed pregnant BALB/c mice were exposed to vehicle, or DBP (50, 250, and 500 mg/kg/day) from 12.5 days of gestation until delivery. In vitro, CCK-8 and EdU incorporation assays were performed to determine the effect of monobutyl phthalate (MBP), the active metabolite of DBP, on the proliferation of TM4 cells, which are a juvenile testicular SC cell line. Western blotting analysis, quantitative PCR (q-PCR), and flow cytometry were performed to analyse the expression of genes and proteins related to the proliferation and apoptosis of TM4 cells. Coimmunoprecipitation was used to determine the relationship between the ubiquitination of interleukin 1 receptor-associated kinase 1 (IRAK1) and the effect of MBP on promoting the proliferation of TM4 cells. Results In the 50 mg/kg/day DBP-exposed male mice offspring, the number of SCs was significantly increased. Consistent with the in vivo results, in vitro experiments revealed that 0.1 mM MBP treatment promoted the proliferation of TM4 cells. Furthermore, the data showed that 0.1 mM MBP-mediated downregulation of the E3 ubiquitin ligase Pellino 2 (Peli2) increased ubiquitination of IRAK1 by K63, which activated MAPK/JNK signalling, leading to the proliferation of TM4 cells. A new carboxyl-functionalized covalent organic framework (referred to as COF hereafter, unless otherwise noted) contains aromatic backbones and carboxyl-functionalized nanopores has been synthesized and used as a two-dimensional (2D) nanofiller in polyacrylonitrile (PAN) polymer matrix of ultrafiltration (UF) membranes. A series of mixed-matrix UF membranes with various concentrations COF ranging from 0 to 0.8 wt% have been investigated using non-solvent induced phase separation (NIPS) method. The effect of COF concentration has been evaluated on the membrane morphology, intrinsic and mechanical properties, separation performance, and fouling propensity. Increasing COF concentration from 0 to 0.8 wt% could significantly enhance the rejection rate from 3.5 to 81.9 wt% for bovine serum albumin (BSA) and 65.0–99.4 wt% for γ-globulin. Moreover, membrane hydrophilicity greatly increased, with the decrease of water contact angle from 64.4° to 45.3°. Overall, compared to the pristine PAN membrane, the mixed-matrix PAN membrane with 0.8 wt% COF demonstrated high water flux of 940 L m−2 h−1 bar−1, better protein retention rate, thermal stability, mechanical properties, and fouling resistance. more...

To investigate the mechanisms of phospholipase C (PLC)-mediated calcium (Ca2+) signaling in Alternaria alternata, the regulatory roles of PLC were elucidated using neomycin, a specific inhibitor of PLC activity. Three isotypes of PLC designated AaPLC1, AaPLC2, and AaPLC3 were identified in A. alternata through genome sequencing. qRT-PCR analysis showed that fruit wax extracts significantly upregulated the expression of all three PLC genes in vitro. Pharmacological experiments showed that neomycin treatment led to a dose-dependent reduction in spore germination and appressorium formation in A. alternata. Appressorium formation was stimulated on hydrophobic and pear wax-coated surfaces but was significantly inhibited by neomycin treatment. The appressorium formation rates of neomycin treated A. alternata on hydrophobic and wax-coated surfaces decreased by 86.6 and 47.4%, respectively. After 4 h of treatment, exogenous CaCl2 could partially reverse the effects of neomycin treatment. Neomycin also affected mycotoxin production in alternariol (AOH), alternariol monomethyl ether (AME), altenuene (ALT), and tentoxin (TEN), with exogenous Ca2+ partially reversing these effects. Drought stress is a serious threat to agriculture and the environment. Brassinosteroids (BRs) increase tolerance to drought stress of plant. Autophagy plays important roles in plant responses to drought stress; however, there are few reports on autophagy in peach (Prunus persica). In total, 23 putative autophagy-related genes (ATGs) in peach were identified using ATGs from the Arabidopsis thaliana genome as query in BLASTx algorithm-based searches. Under drought stress, the photosynthetic abilities of peach leaves decreased, while antioxidant enzyme activities, autophagy and ATG expression increased. A correlation analysis showed that antioxidant enzyme activities are inversely correlated to the expression levels of the PpATGs. During drought, the PpATG8s and some PpATG18s had the strongest responses. To investigate enhanced drought-stress tolerance, peach was treated with water, 100 nM 24-epibrassinolide (EBR), 1 μM EBR, 10 μM EBR and 1 μM voriconazole. Exogenous EBR at 1 μM decreased the malondialdehyde (MDA) content under drought stress when compared with water-, 1 μM voriconazole-, 100 nM EBR- and 10 μM EBR-treated peach leaf. The 1-μM EBR application increased superoxide dismutase (SOD), catalase (CAT), peroxidase (POD), ascorbate peroxidase (APX) and glutathione peroxidase (GR) activities during drought stress. In addition, the expression levels of PpATGs were inhibited by EBR. Nitrogen is an essential macronutrient for plant growth and development and plays an important role in the whole life process of plants. Nitrogen is an important component of amino acids, chlorophyll, plant hormones and secondary metabolites. Nitrogen deficiency leads to early senescence in plants, which is accompanied by changes in gene expression, metabolism, growth, development, and physiological and biochemical traits, which ensures efficient nitrogen recycling and enhances the plant's tolerance to low nitrogen. Therefore, it is very important to understand the adaptation mechanisms of plants under nitrogen deficiency for the efficient utilization of nitrogen and gene regulation. With the popularization of molecular biology, bioinformatics and transgenic technology, the metabolic pathways of nitrogen-deficient plants have been verified, and important progress has been made. However, how the responses of plants to nitrogen deficiency affect the biological processes of the plants is not well understood. The current research also cannot completely explain how the metabolic pathways identified show other reactions or phenotypes through interactions or cascades after nitrogen inhibition. Nitrate is the main form of nitrogen absorption. In this review, we discuss the role of nitrate in plant senescence. Soybean (Glycine max (L.) Merr.) is an important crop that serves as a source of edible oil and protein. However, little is known about its molecular mechanism of adaptation to extreme environmental conditions. Based on the Arabidopsis thaliana sequence database and Phytozome, a soybean gene that had a highly similar sequence to the reduced induction of the non-photochemical quenching2 (AtRIQ2) gene, GmRIQ2-like (accession NO.: Glyma.04G174400), was identified in this study. The gene structure analysis revealed that GmRIQ2-like encoded a transmembrane protein. Elements of the promoter analysis indicated that GmRIQ2-like participated in the photosynthesis and abiotic stress pathways. The subcellular localization results revealed that the protein encoded by GmRIQ2-like was located in chloroplasts. The quantitative real-time (qRT)-PCR results revealed that GmRIQ2-like-overexpression (OE) and -knock-out (KO) transgenic soybean seedlings were cultivated successfully. The relative chlorophyll (Chl) and zeaxanthin contents and Chl fluorescence kinetic parameters demonstrated that GmRIQ2-like dissipated excess light energy by enhancing the non-photochemical quenching (NPQ) and reduced plant photoinhibition. These results suggested that GmRIQ2-like was induced in response to strong light and depressed Chl production involved in soybean stress tolerance. These findings indicate that the transgenic seedlings of GmRIQ2-like could be used to enhance strong light stress tolerance and protect soybean plants from photoinhibition damage. We describe herein the first enantioselective organocatalytic Michael addition reaction of α-amido sulfones to ynones to access axially chiral styrenes. Sulfone-containing axially chiral styrene compounds were constructed with excellent enantioselectivity and almost complete E/Z-selectivity (up to 97% ee, >20 : 1 E/Z) in the presence of an easily accessible cinchona-alkaloid-derived multiple hydrogen-bonding catalyst. The enantioenriched sulfone-containing products were used as precursors for further transformations without any loss of the ee value. Salted duck egg yolk (SDEY) is one of the traditional pickled egg products in Asian countries, which suffers from the weight loss and deterioration of texture characteristics during storage. To better maintain the texture of SDEY, an edible coating based on whey protein isolate nanofibers (WPNFs) with glycerol (Gly) as a plasticizer and incorporating carvacrol (CA) as an antimicrobial agent was developed. Whey protein isolate (WPI, 5%) was used to self-assemble into WPNFs at 80 &deg, C for 10 h. The particle size, zeta-potential and microstructure of WPNFs&ndash, CA emulsion were investigated to evaluate the distribution. Results proved that WPNFs&ndash, CA emulsion had smaller particle size and better distribution than WPI&ndash, CA emulsion. WPNFs&ndash, CA/Gly edible coating was then prepared based on WPNFs&ndash, CA emulsion. The WPNFs&ndash, CA/Gly edible coating exhibited higher antibacterial activity while the WPNFs&ndash, CA/Gly film had smooth and continuous surfaces and better transmittance compared with other samples. Furthermore, weight losses and textural properties changes of SDEYs with WPNFs&ndash, CA/Gly coating were evaluated. Results proved that salted duck egg yolks with WPNFs&ndash, CA/Gly coating exhibited lower weight losses. Textural properties were significantly improved by the WPNFs&ndash, CA/Gly coating on SDEYs than those uncoated samples. It was noted that the egg yolks coated with the WPNFs&ndash, CA/Gly coating had the lowest hardness increase rate (18.22%). An organocatalytic enantioselective method for the synthesis of multiple stereoisomers bearing E, Z configurations, stereogenic carbon centers, and axially chiral styrenes is reported. The method enabled the rapid construction of a series of stereochemical complexity products with excellent E, Z selectivity, diastereoselectivity (>20:1 dr), and enantioselectivity (up to 96% ee).
