Summary Background and aim Cardiovascular diseases (CVDs) are the number one cause of mortality worldwide. Apolipoprotein B (ApoB), apolipoprotein A1 (ApoA1), and ApoB/ApoA1 ratio are considered as predictors of CVD alongside with lipid profile. Evidence suggest that nutrients with antioxidant properties, especially vitamin E, are essential for a healthy cardiovascular system. The aim of present meta-analysis was to determine the effect alpha-tocopherol on ApoA1 and ApoB levels. Methods PubMed-Medline and SCOPUS databases and Google Scholar were searched up to July 2021. Random-effects model was employed to perform meta-analysis. In order to find heterogeneity sources, subgroup analysis was performed. Trim and fill analysis was performed in case of presence of publication bias. Quality assessment was performed using Cochrane Collaboration's tool. Results Seven eligible studies, involving 1284 individuals were included. Mean age of participants ranged between 25.4 and 59 years. There was no significant effect of vitamin E supplementation on Apo A1 (SMD = 0.22 IU/d; 95% CI: −0.38, 0.28; P = 0.481) and Apo B levels (SMD = −0.62 IU/d; 95% CI: −1.94, 0.70; P = 0.360). Conclusion No remarkable effect of vitamin E supplementation was observed on ApoA1 and ApoB levels in adults. Additional studies investigating the influence of vitamin E on apolipoproteins as primary outcome with larger sample size are suggested.

How retinol as a clinical indicator of vitamin A status is related to long-term mortality is unknown. Here we report the results of a prospective analysis examining associations between serum retinol and risk of overall and cause-specific mortality. During a 30-year cohort follow-up, 23,797 deaths were identified among 29,104 men. Participants with higher serum retinol experienced significantly lower overall, CVD, heart disease, and respiratory disease mortality compared to men with the lowest retinol concentrations, reflecting 17–32% lower mortality risk (Ptrend, Vitamin A, of which retinol is the major form in the circulation, is a determinant of human health but whether vitamin A status is associated with mortality is not well understood. 24(S)-Hydroxycholesterol (24S-OHC) and 25-hydroxycholesterol (25-OHC) are produced by cholesterol 24-hydroxylase and cholesterol 25-hydroxylase, respectively. The purpose of the present study was to determine the type of cell death induced by these oxysterols in neuronal cells, hepatic cells, and keratinocytes, and to elucidate the inhibitory effect of vitamin E homologues on various types of cell death. In human neuronal cells (SH-SY5Y cells), 24S-OHC and 25-OHC caused a cell death that was independent of caspase activation. We reported previously that the esterification of 24S-OHC by acyl-CoA:cholesterol acyltransferase 1 (ACAT1) and the resulting formation of a lipid droplet (LD)-like structure are responsible for the 24S-OHC-induced neuronal cell death. Here, we found that 25-OHC also induced ACAT1-mediated 25-OHC esterification and LD formation in neuronal cells. 25-OHC-induced cell death was inhibited by α-tocopherol (α-Toc) but not by α-tocotrienol (α-Toc3), as observed for 24S-OHC-induced cell death in SH-SY5Y cells. In human hepatic cells (HepG2 cells), these oxysterols caused a cell death that was caspase- and oxysterol-esterification-independent. This cell death was suppressed by both α-Toc and α-Toc3, suggesting the involvement of free-radical-mediated lipid peroxidation in the cell death induced by these oxysterols in hepatic cells. In human keratinocytes (HaCaT cells), these oxysterols caused a caspase-dependent but oxysterol-esterification-independent cell death that was inhibited by α-Toc but not by α-Toc3. These results suggest that α-Toc and α-Toc3 act as radical-scavenging antioxidants against oxysterol-induced cell death in the same way in hepatic cells, whereas their behavior is different in inhibition of cell death in neuronal cells and keratinocytes. Collectively, these results demonstrated that 24S-OHC and 25-OHC induced the same type of cell death in each of the cell types examined, and that α-Toc and α-Toc3 exerted different effects, depending on the type of cell death.

OBJECTIVES: To estimate the prevalence of deficiency or insufficiency of vitamins C and E and associated factors among women of childbearing age, in Brazil. DESIGN AND SETTING: Systematic review and meta-analysis conducted at a Brazilian public university. METHODS: A search from index inception until May 2020 was conducted. Meta-analyses were performed using inverse variance for fixed models, with summary proportions calculation using Freeman-Tukey dou-ble arcsine (base case). Reporting and methodological quality were assessed using the Joanna Briggs Institute tool for prevalence studies. RESULTS: Our review identified 12 studies, comprising 1,316 participants, especially breastfeeding wom-en. There was at least one quality weakness in all studies, mainly regarding sampling method (i.e. conve-nience sampling) and small sample size. The prevalence of vitamin C deficiency ranged from 0% to 40%. Objectives Cancer monotherapy is associated with various limitations; therefore, combination chemotherapy is widely explored for optimum drug efficacy. In this study, 4-(N-Phenyl-N′-substituted benzenesulfonyl)-6-(4-hydroxyphenyl) quinoline-based mammalian target of rapamycin (mTOR) inhibitor (IIIM-4Q) was investigated in combination with tocopherol succinate (TOS), and the mechanism of cytotoxicity was elucidated. Methods The cytotoxic potential of IIIM-4Q and TOS was evaluated in five cell lines. Further, to understand the mechanism of cytotoxicity of IIIM-4Q, TOS and their combination, various studies including morphological analysis using scanning electron microscopy and 6-diamidino-2-phenylindole (DAPI) staining, estimation of reactive oxygen species (ROS) level, measurement of mitochondrial membrane potential (MMP), in-vitro cell migration assay, Western blotting and staining with acridine orange (AO) for autophagy detection were performed. Key findings Investigated combination was synergistic in nature and exhibited greater oxidative stress and mitochondrial dysfunction in pancreatic cancer cells. The migration potential of MIA PaCa-2 cells was significantly mitigated under the influence of this combination, and morphological changes such as chromatin condensation and nuclear blebbing were observed. Also, poly (adenosine diphosphate-ribose) polymerase cleavage and caspase-3 activation were observed in IIIM-4Q and TOS combination-treated cells. Conclusions The investigated combination synergistically inhibited proliferation of MIA PaCa-2 cells through simultaneous induction of autophagy followed by apoptosis, and this combination demonstrated potential for further translational studies.

Objectives Fat-soluble vitamins are measured to identify deficiencies that may lead to rickets, osteomalacia, night blindness, and reversible motor and sensory neuropathies. We present a rapid liquid chromatography–mass spectrometry (LC-MS/MS) method that simultaneously measures 25-hydroxyvitamin D3 (25[OH]D3), epi-25(OH)D3, 25(OH)D2, vitamin A, α-tocopherol, and γ-tocopherol. Methods We mixed 100 µL serum with internal standard and extracted it by using supported liquid extraction plates. Reconstituted specimens were analyzed by LC-MS/MS with a 10-minute gradient. Results The method was linear, covering physiological levels with r2 > 0.99, and the total precision was less than 15% at all quality control levels. The lower limit of the measuring intervals for 25(OH)D3, epi-25(OH)D3, 25(OH)D2, vitamin A, α-tocopherol, and γ-tocopherol were 4 ng/mL, 4 ng/mL, 4 ng/mL, 1 µg/dL, 0.2 µg/mL, and 0.2 µg/mL, respectively, with coefficient of variation of 20% or less. Recoveries were between 92% and 111% for National Institute of Standards and Technology reference materials and 81% and 122% for spike-recovery studies. Comparison studies for vitamin D total, vitamin A, and α-tocopherol demonstrated slopes between 1.04 and 1.11 and r2 between 0.94 and 0.96. Minimal matrix effect was observed for all analytes. Conclusions We developed and validated a rapid LC-MS/MS method for the simultaneous measurement of 25(OH)D3, epi-25(OH)D3, 25(OH)D2, vitamin A, α-tocopherol, and γ-tocopherol.

Tocotrienol mixtures have been shown to exert anabolic actions on the skeletal system in animal studies, but it is unclear which tocotrienol isomer shows the most prominent effects. This study aims to investigate the most active tocotrienol isomers using hFOB 1.19 human osteoblasts cultured on a bovine bone scaffold. The bovine trabecular bone was sectioned, demineralised and freeze-dried to form the scaffold. hFOB 1.19 osteoblasts were cultured on the bone scaffolds in humidified condition at 37 °C and 5% carbon dioxide with vitamin E isomers (alpha-, beta-, gamma-, delta-tocotrienol and alpha-tocopherol). The cell differentiation capacity of tocotrienol isomers was investigated through morphological observation, alkaline phosphatase (ALP) activity and osteocalcin expression. Changes in the bone scaffolds were determined using histomorphometry methods. Osteoblast culture treated with gamma- and delta-tocotrienols showed a significant increase in ALP activity and osteocalcin expression. Bone structural histomorphometry analysis showed that bone scaffolds treated with gamma- and delta-tocotrienol showed significant increases in bone volume and trabecular thickness. In conclusion, gamma- and delta-tocotrienol show the most prominent bone anabolic effects by increasing osteoblast differentiation and enhancing bone microstructure.

The seminiferous tubules where spermatogenesis occurs are enveloped and protected by the Sertoli cells to support germ cells undergoing meiosis to produce haploid gametes. Clearly, induction of apoptosis in seminiferous tubules leads to abnormalities in spermatogenesis and male infertility. Studies demonstrated that increased hyperlipidemia impairs male infertility and spermatogenesis by enhancing seminiferous tubules apoptosis. However, molecular mechanisms underlying high-cholesterol-mediated testicular damage remain poorly elucidated. In this scope, we established a rabbit model and investigated the role of endoplasmic reticulum (ER) stress on high cholesterol diet induced seminiferous tubule apoptosis. Histopatological examinations revealed increased seminifer tubule apoptosis in testes of rabbits fed high cholesterol diet. In addition, phosphorylated forms of IRE1 and PERK, two well-identified markers of ER stress, were significantly induced in accordance with high cholesterol diet. High cholesterol diet also exhibited CHOP induction in testes, indicating increased ER stress related apoptosis. Supplementation of α-tocopherol significantly attenuated cholesterol mediated ER stress, and restored seminiferous tubules apoptosis. Taken together, our findings suggest that α-tocopherol might be capable to reduce testicular damage via ameliorating histopatological features and inhibiting seminiferous tubules apoptosis in hypercholesterolemic rabbits.

This research was undertaken in order to determine the impact of nutritional vitamin E on genetic expression of Interferon-gamma (IFN-γ) and Interleukin-1β (IL-1β) as pro-inflammatory and Interleukin-4 (IL-4) and Interleukin-10 (IL-10) as anti-inflammatory cytokines in spleen and liver, heamagglutination inhibition antibody titers (HI titers) against Newcastle Disease Virus (NDV), total protein, albumin, globulin and growth efficiency of broiler chickens. Seventy two one-day-old Cobb broiler chicks had been allocated randomly into two groups. Each group has 3 replicates, each with 12 chicks. The first group was given basal diet. The second group was fed the basal diet with vitamin E (100 mg/kg diet). In this study, bioadhesive nanostructured lipid carriers (NLC) were developed for the co-encapsulation of compounds with antioxidant (α-tocopherol and quercetin) and antimicrobial (tea tree oil) activity for management of wounds. The NLC was produced with shea butter and argan oil, and modified with sodium alginate or chitosan to confer bioadhesive properties. Spherical nanoparticles of ~307–330 nm and zeta potential varying from −21.2 to +11.8 mV were obtained. Thermal analysis demonstrated that the lipid matrix reduced tea tree oil thermal loss (~1.8-fold). Regardless of the type of polysaccharide employed, the NLCs promoted cutaneous localization of antioxidants in damaged (subjected to incision) skin, with a ~74 to 180-fold higher delivery into the skin compared to percutaneous delivery. This result is consistent with the similar bioadhesive properties of chitosan or sodium alginate-modified NLC. Nanoencapsulation of tea tree oil did not preclude its antimicrobial effects against susceptible and resistant strains of S. aureus and P. aeruginosa, while co-encapsulation of antioxidants increased the NLC-induced fibroblasts migration, supporting their potential usefulness for management of wounds. Background and aims: The accumulation of fat increases the formation of lipid perox-ides, which are partly scavenged by alpha-tocopherol (a-TOH). Here, we aimed to investigate the associations between different measures of (abdominal) fat and levels of urinary a-TOH metab-olites in middle-aged individuals. Methods and results: In this cross-sectional analysis in the Netherlands Epidemiology of Obesity study (N Z 511, 53% women; mean [SD] age of 55 [6.1] years), serum a-TOH and a-TOH metab-olites from 24-h urine were measured as alpha-tocopheronolactone hydroquinone (a-TLHQ, oxidized) and alpha-carboxymethyl-hydroxychroman (a-CEHC, enzymatically converted) using liquid-chromatography-tandem mass spectrometry. Body mass index and total body fat were measured, and abdominal subcutaneous and visceral adipose tissue (aSAT and VAT) were as-sessed using magnetic resonance imaging. Using multivariable-adjusted linear regression ana -lyses, we analysed the associations of BMI, TBF, aSAT and VAT with levels of urinary a-TOH metabolites, adjusted for confounders. We observed no evidence for associations between body fat measures and serum a-TOH. Higher BMI and TBF were associated with lower urinary levels of TLHQ (0.95 [95%CI: 0.90, 1.00] and 0.94 [0.88, 1.01] times per SD, respectively) and with lower TLHQ relative to CEHC (0.93 [0.90, 0.98] and 0.93 [0.87, 0.98] times per SD, respectively). We observed similar associations for VAT (TLHQ: 0.94 [0.89, 0.99] times per SD), but not for aSAT. Conclusions: Opposite to our research hypothesis, higher abdominal adiposity was moderately associated with lower levels of oxidized a-TOH metabolites, which might reflect lower vitamin E antioxidative activity in individuals with higher abdominal fat instead. Eighteen brands of vegetable oils available in the local market were extracted with n-hexane before analysis for alpha-tocopherol by RP-HPLC method. The chromatographic separation occurred isocratically with methanol-water [96:4%v/v] at 0.9 ml/min flow rate. Tocopheryl acetate was the internal standard and alpha-tocopherol was eluted at 7.87 min. Free fatty acids value [FFAVs], peroxide value [PV], iodine value [IV] and saponification values [SV] were determined as quality parameters. Calibration curve was linear [r2 0.9969] and the method was precise with relative standard deviation of 0.35% and mean recovery, 87.39%. Alpha-tocopherol concentration ranged from 0-9.22 mg/100g with the highest in Tropical sunflower oil [9.22 mg/100g] and the lowest [1.16 mg/100g] in Laziz oil. Gas Chromatography-Mass Spectrometry (GC-MS) is normally used for direct analysis of chemical components existing in herbal medicines. The medicinal plants are having numerous bioactive components which are identified even at less than 1ng by using GC-MS or LC-MS analysis. The aim of this study is to identify the secondary metabolites present in the leaves of B. tomentosa using gas chromatography-mass spectrometry (GC-MS) analysis. In the present study the ethanol extract of the leaves of Bauhinia tomentosa has been subjected to GC-MS analysis, while the mass spectra of the compounds found in the extract was matched with the National Institute of Standards and Technology (NIST) library. GC-MS analysis revealed the presence of 14 secondary metabolites. These compounds were identified by comparing their retention times and peak areas with those from the literature and by interpretation of the mass spectra. The major secondary metabolites were DL-.alpha.-tocopherol (14.84%), 2-[(trimethylsilyl oxy]-, methyl ester, 1-alpha,2-alpha.-epoxy-1-beta-methylcholesta-4,6-dien-3-one (12.93%), pentacosenoic acid (12.71%), phytol (10.28%), Ethyl Isoallocholate (8.197%), Spirost-8-en-11-one-3-hydroxy-,(3-beta,5 alpha,14 beta,20 beta, 22 beta,25R)-(8.162%), Urs-12-en-28-ol (6.675%), 1-Octadecyne (5.702%) and Cholest-8-en-3-beta-ol,Acetate (5.426%). The compounds having area less that 5% were considered of no significance. These findings suggest that the presence of these secondary metabolites may be the cause for the properties exhibited by Bauhinia tomentosa. Thus, presence of various bioactive compounds justifies the use of the leaf for various ailments by traditional practitioners.

Oxidative stability of prototypical groundnut oil (GO) and mustard oil (MO) blended with a significant characteristic addition of antioxidants, α-tocopherol (α-T) and TBHQ (tert-butyl hydroquinone), at different concentrations was analysed. α-T (natural) and TBHQ (synthetic) antioxidants at a different concentrations from 0 to 20 mg/L were blended with GO and MO to prepare 18 samples. Compositional analysis of groundnut oil and mustard oil was carried out using gas chromatography and mass spectra (GCMS). Electrical properties like dielectric constant, tanδ, specific resistance etc. were investigated at frequencies from 1 Hz to 10 MHz for the modified samples. Thermal properties like breakdown voltage, flash and fire points and viscosity of all samples were also premeditated. It was observed that dielectric constant (ε') of GO reduces from 3.58 to 2.82 with the addition of α-T but increases to 3.29 with the addition of TBHQ, whereas in MO, dielectric constant increases from 2.93 to 3.38 with the addition of α-T and rises to 3.27 with the addition of TBHQ at 15 mg/L. The synergistic effect of antioxidants in regulating the insulation nature of the oil is found to be more effective with α-tocopherol compared to TBHQ in GO. Higher breakdown voltage and more stability were observed in oil with the addition of antioxidant (TBHQ) in low concentration. The study would be useful in the selection of eco-friendly coolants in engineering and industry.

Alpha-tocopherol, a highly active form of the antioxidant vitamin E in humans, effectively scavenges free radicals and protects the cell membranes. The aim of our study was to enhance alpha-tocopherol production in sunflower cell suspension using elicitation, precursor addition and culture condition optimization. Among various elicitors and precursors studied, sodium chloride (50 mg/L) added as elicitor and phytol (50 mg/L) added as the precursor to an untransformed sunflower cell suspension, resulted in maximum productivity enhancement of alpha-tocopherol up to 2.3- and 1.9-fold, respectively compared to a control untransformed sunflower cell suspension (0.02 mg/L/d). In separate experiments, the volumetric productivity of alpha-tocopherol in the untransformed suspension was enhanced up to 2.1- fold (0.09 mg/L/d) under optimized conditions of light intensity (3500 lx), temperature (30 °C), initial pH (6.5) and inoculum size (6 g/L). We then extrapolated these strategies (elicitor and precursor addition, culture condition optimization) for implementation on a high alpha-tocopherol yielding metabolically engineered cell suspension overexpressing 4-hydroxyphenylpyruvate dioxygenase (TS4), which resulted in enhancement (1.9-, 1.45- and 2.72-fold respectively) in the volumetric productivity of alpha-tocopherol compared to the transformed cell suspension grown under control conditions (0.18 mg/L/d). Thus, integration of bioprocess optimization and metabolic engineering resulted in a multi-fold (up to 12-fold) enhancement in alpha-tocopherol productivity in comparison to the untransformed cells. Therefore, our study demonstrates that productivity enhancement strategies like bioprocess optimisation and metabolic engineering (developed separately in parallel) for a given system can be combined to achieve maximum productivity in lesser time and cost than a relay system of optimization in long-duration plant cell cultivations.

Objective: To study the ameliorative effect of Alpha-Tocopherol (Vitamin E) against E-cigarette induced histomorphalogical changes in adrenal cortex of male Albino rats. Study Design: Randomized Animal Control Trial. Setting: Department of Anatomy, Islamic International Medical College Rawalpindi in collaboration with National Institute of Health (NIH), Islamabad. Period: September 2018 to September 2019. Material & Methods: 30 adult male albino rats were divided in three groups having 10 rats each. Normal diet and distilled water was given to control group (Group I) including 10 rats, for 4 weeks. The Rats of Group II were injected E-cigarette liquid 0.5mg per kg body weight per day diluted in 500ul normal saline intraperitoneally for 4 weeks. The experimental rats in Group III, in addition to E-cigarette liquid also received Vitamin E, 100mg per kg body weight per day mixed in daily diet. Dissection of the animals was done after completion of 4 weeks and adrenal gland was removed for histological analysis. Data was recorded and analyzed in SPSS™ version 21. Results: Alpha-Tocopherol restored the quantitative changes induced by E-Cigarette liquid in adrenal cortex in Group III, which was increased in the rats of Group II who were injected E-Cigarette liquid. Among microscopic parameters Orientation of cells, sinusoidal dilatation was significantly reduced in all adrenal cortex zones by Alpha-Tocopherol in Group III animals. Conclusion: The current study demonstrated that Alpha-Tocopherol had an ameliorative effect on histological changes caused by E-cigarette liquid on adrenal cortex.

This is the first study evaluated oil characteristic of 23 olive cultivar candidates, which obtained by cross breeding and later selection among 492 olive genotypes. In this research, oil characteristics such as alfa tocopherol, total phenol, antioxidant activity, oxidative stability and sensory analysis were determined. Olive oils of standard ‘Gemlik’ and ‘Ayvalik’ cultivars were also included analysis to compare candidates. Significant genetic influence was determined on nutritional, oxidative stability and sensory characteristics of oils. Oils of GT009, LE003 and LT019 genotypes had highest both total phenol content and antioxidant activity so that they had remarkable characters from nutritional physiology perspective. GE363 showed longest induction time and LE003, GE417 and GE363 showed longest shelf life. Highest alpha tocopherol content was determined in oils of BK013 and ‘Gemlik’ olives. According to sensory evaluation, oil of GU429 was classified as well balanced oil and oils of GE067, GE110, GE366, GU247, GU320, GU323 and GU434 classified as mild oil. These results will be used to give decision for their final selection and register the selected best cultivar candidates.

Multidrug resistance (MDR) remains a significant impediment to chemotherapy during cancer therapy. In this study, the amphiphilic biomaterials PEI-TOS and HA-QU were synthesized to self-assemble into PEI-TOS/HA-QU core–shell micelles for the targeted codelivery of paclitaxel (PTX) and quercetin (QU) to alleviate multidrug drug resistance and enhance therapeutic efficacy. The PTX-loaded micelles possessed a uniform particle size (167.60 ± 8.185 nm), stable negative charge (–19.13 ± 0.321 mV), and pH-responsive drug release with good compatibility. The drug-loaded micelles increased the chemosensitivity of MDR tumor cells (MDA-MB-231/MDR1) to PTX and activated mitochondria-dependent apoptotic pathways (the IC50 was 2.22-fold lower than that of PTX alone). Moreover, PEI-TOS/HA-QU micelles increased the cellular uptake of lipophilic antitumor drugs by downregulating P-gp expression in MDA-MB-231/MDR1 cells. Compared with Taxol, PTX-loaded PEI-TOS/HA-QU micelles presented excellent antitumor efficacy in tumor-bearing mice, with an average tumor size that was 3.7-fold lower than that of the control group. The drug-loaded formulation showed low in vitro / in vivo toxicity and better tumor accumulation than the free drug, which led to a high tumor inhibition rate of 80.56% and considerable biocompatibility. This work describes a new platform for the codelivery of lipophilic anticancer drugs and natural active ingredients such as PTX and QU for the treatment of MDR cancer cells.

In cyanobacteria, the PII protein (the glnB gene product) regulates a number of proteins involved in nitrogen assimilation including PipX, the coactivator of the global nitrogen regulator protein NtcA. In Synechococcus elongatus PCC 7942, construction of a PII-less mutant retaining the wild-type pipX gene is difficult because of the toxicity of uncontrolled action of PipX and the other defect(s) resulting from the loss of PIIper se, but the nature of the PipX toxicity and the PipX-independent defect(s) remains unclear. Characterization of a PipX-less glnB mutant (PD4) in this study showed that the loss of PII increases the sensitivity of PSII to ammonium. Ammonium was shown to stimulate the formation of reactive oxygen species in the mutant cells. The ammonium-sensitive growth phenotype of PD4 was rescued by the addition of an antioxidant α-tocopherol, confirming that photo-oxidative damage was the major cause of the growth defect. A targeted PII mutant retaining wild-type pipX was successfully constructed from the wild-type S. elongatus strain (SPc) in the presence of α-tocopherol. The resulting mutant (PD1X) showed an unusual chlorophyll fluorescence profile, indicating extremely slow reduction and re-oxidation of QA, which was not observed in mutants defective in both glnB and pipX. These results showed that the aberrant action of uncontrolled PipX resulted in an impairment of the electron transport reactions in both the reducing and oxidizing sides of QA.

We recently reported the antisense properties of a DNA/RNA heteroduplex oligonucleotide consisting of a phosphorothioate DNA-gapmer antisense oligonucleotide (ASO) strand and its complementary phosphodiester RNA/phosphorothioate 2′-O-methyl RNA strand. When α-tocopherol was conjugated with the complementary strand, the heteroduplex oligonucleotide silenced the target RNA more efficiently in vivo than did the parent single-stranded ASO. In this study, we designed a new type of the heteroduplex oligonucleotide, in which the RNA portion of the complementary strand was replaced with phosphodiester DNA, yielding an ASO/DNA double-stranded structure. The ASO/DNA heteroduplex oligonucleotide showed similar activity and liver accumulation as did the original ASO/RNA design. Structure-activity relationship studies of the complementary DNA showed that optimal increases in the potency and the accumulation were seen when the flanks of the phosphodiester DNA complement were protected using 2′-O-methyl RNA and phosphorothioate modifications. Furthermore, evaluation of the degradation kinetics of the complementary strands revealed that the DNA-complementary strand as well as the RNA strand were completely cleaved in vivo. Background: Monosodium Glutamate (MSG) is one of the most commonly used flavor-enhancing substances that may lead to neurological disorders. Objectives: The present work aimed to evaluate the role of L-ascorbic acid (AA) and -tocopherol (T) on the MSG-induced memory and neurobehavioral changes in rats. Methods: Thirty male Wistar albino rats were randomized into five equal groups: (1) control group, (2) MSG group received MSG (2mg/g BW) daily, (3) MSG+A group received MSG as in MSG group, and AA (100 mg/kg BW) daily, (4) MSG+T group received MSG as in MSG group, and T (600 mg/kg BW) twice weekly, and (5) MSG+AT group received MSG as in MSG group, AA as in MSG+A group, and T as in MSG+T group. After 3 weeks, neurobehavioral changes were assessed by open field test and Y maze. Oxidative stress markers were estimated, and immunohistochemistry was studied in hippocampal region. Results: MSG resulted in impairment of memory and induction of anxiety, with increased hippocampal malondialdehyde and decreased superoxide dismutase and glutathione peroxidase. Treatment with AA or T improved all the measured biochemical parameters, and the MSG-induced hippocampal degenerative changes, with decreased glial fibrillary acidic protein (GFAP) and synaptophysin expression. Combined administration of both vitamins was more effective in amelioration of MSG-induced impairments rather than taking AA or T alone. Conclusion: Both AA and T exhibit protective effects against neurobehavioral changes, oxidative stress and hippocampal degenerative changes induced by MSG toxicity, with more potent efficacy of their combination.

Soybean seeds contain high-quality proteins that are used as a source of food, and also abundant lipids that are used as a vegetable oil. Many functional ingredients are also included in soybean seeds. We have identified qualitative and/or quantitative genetic variations in several components, such as protein, alpha-tocopherol, chlorophyll, and isoflavone, among soybean genetic resources. These genetic variations are available to improve the processing properties and to add some values for soy foods. We also demonstrated that a high and low temperature during seed filling enhanced the biosynthetic pathways of alpha-tocopherol and isoflavone, respectively. These suggest that it is possible to increase the amounts of functional components, such as alpha-tocopherol and isoflavones, by improving the cultivation environment of soybean in addition to the development of new soybean varieties by using genetic variations. The successful use of genetic variations and physiological properties related to soybean quality will not only enable differentiation from conventional soy products, but also create new uses for soybean.

Abstract. Background: Contrast–induced nephropathy (CIN) is a relevant cause of acute renal dysfunction and is associated with an increased morbidity and mortality. Purpose: Verify the effect of α-tocopherol pre-treatment on CIN prevention in subjects with chronic kidney disease. Methods: A Medline/Embase and clinicaltrials.gov were searched up to May 1st, 2017. Randomized controlled trials recruiting patients undergoing diagnostic or therapeutic radiocontrast infusion comparing the effect of either oral or i.v. multiple administration of pharmacological dose of α-tocopherol in preventing CIN versus placebo were included. A random-effects model, calculating Mantel-Haenszel odds ratio with 95% confidence interval, was applied to study the effect of α-tocopherol on CIN occurrence. Funnel plot analysis was used to assess publication bias, while agreement within studies was measured by the I2 index and tested with the Q-Cochran test. Results: Out of 242 studies, 4 trials were selected. CIN incidence resulted significantly lower in α-tocopherol compared to placebo group (5.8% vs. 15.4%, MH-OR [95% C.I.] 0.34 [0.19 – 0.59]). Alpha-tocopherol treatment was associated with both a tendential higher eGFR (mean difference 2.19 [95% C.I. -0.41; 4.79] mL/min) and lower creatinine level (mean difference −0.06 [95% C.I. −0.21; 0.09] mg/dl) compared to placebo. No relevant publication bias (p = 0.48) and heterogeneity (I2 = 0%; χ2 = 1.01, df = 3 [p = 0.80], I2 = 0%) were evident. Conclusions: Alpha-tocopherol pre-treatment is associated with reduction of incidence of CIN. Its administration deserves to be further explored as a simple and inexpensive tool for CIN prevention.

Objective: To observe the effects of vitamin E on post-vitrification preimplantation development, gross morphology as well as mitochondrial distribution and ultrastructure. Methods: Twenty-four female C57BL/6NTac mice, aged 12-16 weeks, were randomly divided into four groups. Group A did not receive any treatment and served as the control group. Group B was treated with corn oil stripped of tocopherols and served as the vehicle group. Group C was treated with 60 mg/kg body weight of tocotrienol-rich-fraction with corn oil stripped of tocopherols. Group D was treated with 60 mg/kg body weight of alpha-tocopherol with corn oil stripped of tocopherols. All treatments were administered orally for 7 consecutive days. After superovulation and mating with fertile males, 2-cell stage embryos were harvested for vitrification. Post vitrification development in vitro, gross morphology and ultrastructure were compared between groups. Results: The number of 2 and 8-cell embryo, and blastocysts in the treatment and control groups were not significantly different (P>0.05). Following vitrification, all 2-cell embryos had equal-sized blastomeres and intact zona pellucida. Mitochondrial aggregation toward the perinuclear region was seen in all of the treatment groups. Both groups C and D had vacuolated mitochondria, which was reflected in the trend of preimplantation development reduction. Conclusions: Vitamin E supplementation of 60 mg/kg body weight does not improve the viability of healthy embryos according to this study. As a result, the most effective dose of vitamin E supplementation may be determined by the initial quality of the embryos.

Obesity is a pathological state related to various lifestyle-related diseases, such as diabetes and dyslipidemia, that may be prevented through the development of anti-obesity treatments. Lipid accumulation in cells could be affected by vitamin E ester α-tocopheryl succinate (TS), which has various biological activities, such as anti-cancer effect, via activation of cell signaling pathways, although the antioxidative activity of TS is lost due to esterification of the phenolic OH group. In this study, we found for the first time that TS significantly suppressed lipid accumulation in mouse 3T3-L1 adipocytes. TS treatment reduced the amount of triglycerides in the culture medium, and inhibited activity of glycerol-3-phosphate dehydrogenase, a marker of lipid synthesis. Furthermore, TS accelerated lipolysis. Treatment of adipocytes with TS for 24 h induced no significant cytotoxicity. In TS-treated cells, phosphorylation of Akt, which is involved in fatty acid synthesis via sterol regulatory element-binding proteins (SREBP), was prevented, while levels of phosphorylated protein kinase A (PKA) did not change. Taken together, these results suggest that vitamin E ester TS can suppress lipid accumulation in adipocytes by regulating lipid metabolic cell signaling.

Background In order to improve the stability of vitamin E and broaden its application scope, it is an effective solution to convert vitamin E into its derivatives. This work developed a new candida rugosa lipase nanogel (CRL nanogel) based on modification of ionic liquid with vinyl functional groups. This novel CRL nanogel was used in the preparation process of vitamin E succinate based on the principle of non-aqueous enzymology. At the same time, various factors including enzyme concentration, substrate molar ratio, reaction temperature and reaction time, that affect the yield of vitamin E succinate were optimized and analyzed. Results Different solvent with various hydrophobicity parameters (Log P values) from -1.3 to 3.5 was studied, it was found that dimethyl sulfoxide (DMSO) had the lowest LogP value among organic solvents but vitamin E succinate had the highest yield in DMSO. Effect of different operating conditions, such as molar ratios of substrate, enzyme concentration, reaction temperature and reaction time was studied. Under the optimal process conditions (enzyme concentration 6 mg mL-1 , substrate molar ratio 4: 1, reaction temperature 55 ° C and reaction time 15 h), the product yield was 62.58±1.16%. CRL and CRL nanogel were characterized using Fourier transform infrared (FT-IR) and scanning electron microscopy (SEM). It was shown that CRL nanogel influenced the catalytic behavior of lipase significantly by changing the structure and surface properties of CRL. Our objective was to supplement colostrum with n-3 fatty acids (FA) to provide anti-inflammatory mediators that may improve the immune response of neonatal calves. Elevated markers of inflammation have been associated with increased occurrence of calf disease in early life, thus decreasing animal productivity. We hypothesized that a colostrum supplement containing 60-mL of a 1:1 ratio fish:flaxseed oil blend with or without 200 mg of α-tocopherol might provide an advantageous start to early life by decreasing oxidative stress and regulating the inflammatory response. Calves were blocked by birth order and randomly assigned to 1 of 3 treatments: no supplement added to colostrum (control), 60 mL of 1:1 fish:flaxseed oil blend, and 60 mL of 1:1 fish:flaxseed oil blend with 200 mg of α-tocopherol. In total, 180 heifer calves (n = 60 per treatment) were enrolled on a commercial farm. After colostrum feeding, all calves were housed in individual hutches and fed milk replacer 3 times per day. Health was scored 3 times per week until weaning at 8 wk of age. Weight, wither height, and heart girth were measured after birth, 3 wk, and 8 wk of age to assess preweaning growth. A subgroup of 54 calves (18 blocks or 18 calves per treatment) were sampled 2 d (± 8 h) after birth to evaluate oxidant status, serum total protein, and inflammatory gene and cytokine protein expression in blood after an in vitro lipopolysaccharide (LPS) challenge as indicators of health and immunity. At 9 wk, calves were transported 18 h to another farm, and medical records were kept as an indicator of disease incidence up to 13 wk of age. Calf mortality was 1.8%, which is below industry average, and exceptional health scores were observed throughout the study. Health scores and growth were similar throughout the preweaning period regardless of treatment. Serum total protein indicated successful passive transfer in all calves, and oxidant status index was not affected by treatments on d 2 of age. Concentrations of tumor necrosis factor α increased with LPS stimulation, but the increase was not altered by treatment. Likewise, leukocyte gene expression of tumor necrosis factor α, IL-8 and IL-10, and cyclooxygenase-2 increased upon LPS stimulation, but the fold change did not differ with treatment. In conclusion, 60 mL of 1:1 ratio fish:flaxseed oil colostrum supplement did not enhance preweaning calf performance. Supplementing n-3 FA in a one-time meal may not provide the anti-inflammatory benefits observed with continuous feeding.

The effects of fish oil (40 ml/day) supplementation, with or without synthetic all-rac-alpha-tocopherol-acetate (2,500 IU/day), during the last 65 days before expected parturition were investigated in 15 adult mares (553 ± 24 kg BW) and their foals. Mares were assigned to one of three diets: control (n = 5), control plus fish oil and alpha-tocopherol (n = 4; FO + AT) or control with just fish oil (n = 6; FO). Blood samples were obtained from the mares before a 15-day dietary adaptation period (T1) and from mares and foals the first (T2) and fifth (T3) days post-partum. Colostrum was collected at T2 and milk at T3. Routine haematological, biochemical and alpha-tocopherol analyses were undertaken on all blood samples. Fatty acid concentrations were determined in the foal serum and alpha-tocopherol concentrations measured in the milk and colostrum. Diet had no effect on haematology or biochemistry in the mares. Alpha-tocopherol concentrations were significantly higher at T2 & T3 in the FO + AT mares. Background: Our previous study of the lipid peroxidation and antioxidant defense system (LPO-AOS) state in women with chronic viral hepatitis (CVH) showed the content of fat-soluble vitamin alpha tocopherol significant reduction. Reproductive health of women with CVH needs special attention. The objectives of this study was to determine the frequency of menstrual disorders in the group of women with CVH with marked alpha tocopherol insufficiency. Study Design: A case-control study. Place and Duration of Study: A study was conducted at Scientific Centre for Family Health and Human Reproduction Problems and in the hepatological department of the Irkutsk clinical hospital of infectious diseases from 2010 to 2014. This work was performed with the use of equipment of Collective research centre “Center for the development of progressive personalized technologies for health” SC FHHRP, Irkutsk. Methods and Results: Our study included 44 reproductive age women with CVH and comparable age 28 healthy women. All women underwent clinical examination and questionary survey. We found alpha tocopherol insufficiency in 95.5% cases of patients with CVH and menstrual disorders. Conclusion: Our results suggest that menstrual disorders in patients with CVH may be associated with alpha tocopherol insufficiency.

For simultaneous analysis of four fat-soluble tocopherols (α-, β-, γ-, and δ-) in edible oils, an efficient and green method using deep eutectic solvent-based liquid-phase microextraction (DES-LPME) coupled with reversed-phase high-performance liquid chromatography (RP-HPLC) was developed. The DESs formed by different quaternary ammonium salts and ethanol were used as the extractants. Tetrabutylammonium chloride (TBAC)-ethanol DES at a molar ratio of 1:2 achieved the best extraction efficiency. Under the optimized conditions, the detection limits were in the range of 2.1–3.0 ng mL−1. The intra-day and inter-day repeatability were in the ranges of 3.9–5.3% and 4.8–7.1%, respectively, and the recoveries for the real samples varied from 80.7% to 105.4%. The developed method was successfully employed for the determination of all four tocopherol homologues with an RP-HPLC system containing a COSMOSIL π-NAP column in five edible oils collected locally.

The objective:to compare the results of follow-up over patients with a mild form of pulmonary sarcoidosis who received alpha-tocopherol and those receiving no treatment.Subjects and methods.The results of follow-up over 80 sarcoidosis patients were retrospectively analyzed – two groups of 40 patients enrolled according to the copy – pair principle (gender, age, initial X-ray stage, Löfgren syndrome, and follow-up duration). The only sign that statistically distinguished the groups was a high frequency of complaints about weakness in those who received vitamin E – 55% versus 30% (p < 0.05). The follow-up period for patients in both groups was a year or more (a control point). The database was maintained and processed using SPSS-18, Windows 10. A clear advantage of vitamin E administration was revealed when comparing the data of computed tomography, which resulted in improvement in 87.5%, and the disease progressed in 2.5%, while in the group receiving no treatment – 60 and 12.5%, respectively. The OR of improvement in computed tomography with alpha-tocopherol versus the group receiving no treatment made 4.67 (95% CI, 1.51-14.46).Conclusion.The use of vitamin E in mild forms of thoracic sarcoidosis gives a better result compared to the group receiving no treatment.

Background Job's tears possess an exceptionally high amount of fat and the unsaponifiable fraction is a potential source of bioactive compounds. The aim of this study was to determine the effect of hydrothermal processing on squalene, α-tocopherol, and fatty acids in the whole-grain, milled fractions, and their storage stability. Results The highest level of squalene content was found in the bran fractions of native and processed Job's tears (11.54-12.75 mg 100 g-1 ). A remarkably high amount of α-tocopherol was also found in the bran (59.75 ± 0.47 mg 100 g-1 ) and germ (67.05 ± 0.94 mg 100 g-1 ) of the processed grain fractions. The storage stability evaluation of these bioactive compounds in the polished grains revealed no significant difference between 0 and 15 days of storage under elevated temprature of 37 °C and 92% relative humidity (accelarated storage conidition) and retained the bioactive compounds longer in the processed grains. Conclusion These results suggest that the processed Job's tears milled fractions with high bioactive compounds and improved shelf life can be used as food ingredients in product development. © 2020 Society of Chemical Industry.

PurposeFamilial hypercholesterolemia (FH) requires early treatment. However, statins, which are regarded the first-line therapy, have an influence on redox balance. Antioxidant vitamins are important for many metabolic processes in the developing body. There are few data available on the long-term safety of statin use in children. The aim of this study was to evaluate the influence of statin treatment in children with FH on plasma concentrations of antioxidant vitamins: retinol, alpha-tocopherol and coenzyme Q10.MethodsThe first study group consisted of 13 children aged 10–18 years treated with simvastatin for at least 6 months, and the second group comprised 13 age- and sex-matched children with hypercholesterolemia, in whom pharmacological treatment had not been applied yet. Analyses were performed using a high-performance liquid chromatograph coupled with a MS detector.ResultsThe analysis did not reveal significant differences in the concentration of retinol, alpha-tocopherol or coenzyme Q10 between the studied groups. The adjustment of the concentrations of the vitamins to the cholesterol level also indicated no significant differences. We found no deficits in antioxidant vitamins in patients treated with statins, or any risk of adverse effects associated with an increase in their concentration.ConclusionThere is no rationale for additional supplementation using antioxidant vitamins or modification of low-fat and low-cholesterol diet in pediatric patients treated with statins.

Here, we assessed the relationship between IL-6 variants and serum α-tocopherol levels on periodontal condition by considering effect modification. Background: Alpha-tocopherol is a potent antioxidant involved in sperm protection particularly during cryopreservation. However, its poor solubility limits the optimal protection in aqueous solutions. Objective: The aim of this study was to enhance the solubility of α-tocopherol by the use of liposomes. Methods: The experimental approach consisted to load vitamin E in liposomes prepared by ethanol injection method and the optimization carried out by an experimental design. The optimum solution was characterized by high performance liquid chromatography and scanning electron microscope. Finely, the impact on sperm motility protection was studied by the freezing technic of bovine sperm. Results: The optimum solution was obtained when using 10.9 mg/ml of phospholipids, 1.7 mg/ml of cholesterol and 2 mg/ml of vitamin E. The liposome size was 99.86 nm, providing 78.47% of loaded efficiency. The results showed also a significant positive impact on sperm motility after hours of preservation. Conclusion: In conclusion, the current results showed the interest of liposome preparation as an alternative to enhance vitamin E solubility and to protect spermatozoa during cryopreservation.

Background and Aim: Alpha-tocopherol supplementation influences milk yield, milk quality, and udder health of dairy cows, which needs to be investigated for Jersey crossbred cows at hot-humid climate. Therefore, the present study was framed with an objective to study the effect of pre and postpartum Alpha-tocopherol supplementation on milk yield, milk quality, and udder health status of Jersey crossbred cows at tropical climate. Materials and Methods: For this study, 19 similar parity, body condition score, and production level dairy animals were separated randomly into three groups, namely, Control, T1 and T2. Control group (no supplementation) was compared with two treatment groups, namely, T1 and T2. Supplementation of Alpha-tocopherol was done in concentrate fed to the animals (at 1 g/cow/day) 30 days prepartum to 30 days postpartum in T1 and 30 days prepartum to 60 days postpartum in T2 groups. Observations were taken for different parameters up to 5 months of lactation. Results: Statistically analyzed data revealed that overall significantly (p0.05). Conclusion: Supplementation of Alpha-tocopherol during prepartum to initial lactation period may enhance milk yield, milk quality, and udder health status of Jersey crossbred cows at the tropical lower Gangetic region.

No. of bitstreams: 0 Previous issue date: 2020-09-01 Aim: The aim of this study to evaluate the effects of 10% alpha-tocopherol (10AT) or 20% alpha-tocopherol (20AT) incorporation on biological compatibility, pH, and calcium release of calcium hydroxide (CH) paste associated with 2% chlorhexidine digluconate (CHX). Materials and Methods: Four groups were evaluated: CH, CH-CHX, CH-CHX-10AT, and CH-CHX-20AT. For biological compatibility test, polyethylene tubes containing several pastes were implanted in Wistar rats' subcutaneous tissue (n = 28). After 48 h and 7, 14, and 21 days postimplantation, the specimens were removed and subjected to histologic and histomorphometric analysis. The number of inflammatory cells was evaluated. For pH and calcium release analysis, the pastes were placed individually (n = 10) in plastic tubes and immersed in deionized water. The calcium release and pH changes were evaluated in 24 and 48 h and 7, 14, and 21 days. All data were submitted to Kruskal-Wallis test (α = 0.05). Results: Concerning biological compatibility, all materials shown a similar decidual response (P > 0.05). In the first hours, there was as increase in the number of inflammatory cells, inducing an expressive inflammatory response. After 14 days, inflammation reaction decreased and collagen fiber was organized for the tested pastes (P = 0.05). The pH analysis of the groups maintained the same relationship during the different periods evaluated: the CH and CH-CHX groups showed higher values and were similar to each other (P > 0.05), followed by the CH-CHX-10AT and CH-CHX-20 AT groups. Regarding the amount of calcium ions, in the initial (24 hours) and final (21 days) periods, the groups did not present differences between them (P > 0.05). Conclusion: The 10AT or 20AT, as an antioxidant agent, incorporation to CH and 2% CHX paste negatively affected biological and physicochemical properties. A high intake of green leafy vegetables rich in antioxidative nutrients such as vitamin C and β-carotene may protect against the risk of type 2 diabetes. Measurement of the circulating nutrient concentrations can indicate the nutrient status more directly, and vitamin C and carotenoids are recognized as good biomarkers for the intake of fruits and vegetables. The aim of this study was to investigate the relationships between serum antioxidative vitamin concentrations and type 2 diabetes in Japanese subjects. The study subjects comprised 506 men and 493 women who first underwent anti-aging health checks at Tokai University Tokyo Hospital. Serum concentration of vitamin (V) A, VC, α-tocoferol, β-carotene, VB12, folate, ferritin and homocysteine, and fasting plasma glucose and HbA1c were used for analysis. Low levels of β-carotene and VC were significantly associated with dysglycemia. Diabetic subjects showed significantly decreased β-carotene and VC levels, and multivariate analyses suggested that low levels of β-carotene and VC were factors related to diabetes. Low levels of β-carotene and VC are significantly related to dysglycemia/type 2 diabetes, and encouraging people at a higher risk of diabetes to take more green vegetables may be useful as a dietary intervention to improve the antioxidative vitamin status and dysglycemia.

In several diseased conditions like asthma, allergy, ageing, arthritis, cancer etc., there is a production of reactive nitrogen and oxygen species, as well as our immune system also gets harmed. In order to stop or quench these harmful free radicals, antioxidants, which are the first line of defense, are used. And the immune system can be modulated to work properly by using natural agents like medicinal plants, having antioxidant property. In this context, the present study was undertaken to investigate the immunomodulatory activity and free radical quenching ability of the Boswellia serrata Roxb. (Bs) in murine model. This herb is known to afford protection to human physiological system against diverse stressors. It was tested at three dose levels of 50mg /kg of mice body weight, 100mg /kg of mice body weight, and 150 mg /kg of mice body weight for five consecutive days. Alpha tocopherol (25 mg/ kg, of mice body weight) and Di-methyl Sulphoxide (35mg /kg of mice body weight) were used as the reference standards. The extent of protection against immunosuppression was evaluated after 9 days of respective drug administration, by measuring macrophage phagocytic activity. A statistically significant antioxidant and immunomodulatory activity were shown by the extract of Boswellia serrata as evident by its effect on the activity of non-enzyme antioxidant reduced glutathione (GSH), antioxidant enzyme glutathione peroxidase (GPx), the degree of lipid peroxidation, and increase in the number of peritoneal macrophages.

Background Selenium or alpha‐tocopherol deficiency can cause neuromuscular disease. Beta‐carotene has limited documentation in horses. Since vitamin E is one of the most potent antioxidant and anti-inflammatory agents, vitamin E can play a role against arteriosclerosis through various actions. Then, we have studied the relationship between serum vitamin E status and risk factors for arteriosclerosis in Japanese postmenopausal women. One hundred and seven subjects (70.0±7.7 y) were evaluated for vitamin E status by measuring serum α- and γ-tocopherol (αT and γT) levels. The number of arteriosclerosis risk factors was defined by the existence of high blood pressure, hyperglycemia, and dyslipidemia. Prostate cancer (PC) is one of the the most commonly diagnosed cancer type being the second major reason of cancer-associated death in male particularly over the age of 50. Accumulating scientific evidences suggest the role oxidative stress and Reactive oxygen species (ROS) in prostate cancer. ROS are produced by carcinogenic molecules, infection, toxic compounds all of which can contribute to disturbed homeostasis and genetic mutation. Antioxidants can decrease the negative effects of ROS in vitro. The vitamins C (Ascorbic acid, Asc), A (beta carotenoids and retinoids, β-Crt) and E (alpha tocopherol, α-Toc) play important role in inhibiting oxidation and reducing the concentration of free radicals in the body. The aim of this study was to determine the anticancer effect of α-Toc, β-Crt and Asc on PC-3 prostate cancer cells in vitro. This was carried out by cell proliferation, ROS and Lipid Peroxidation assay, caspase-3 and propidium iodide staining experiments. The findings suggest that these agents behave as prooxidant by lowering cell viability and increasing the production of ROS and LPO in prostate cancer. These oxidants induce apoptosis as supported by propidium iodide and caspase-3 staining.

Vitamin E is essential for neuromuscular function. The primary treatment, oral supplementation with natural (‘RRR’) α-tocopherol, is not effective in all horses. The objectives of this pilot study were to evaluate the safety and efficacy of a subcutaneously administered RRR-α-tocopherol preparation. Horses were randomly assigned in a cross-over design to initially receive RRR-α-tocopherol (5000 IU/450 kg of 600 IU/mL) subcutaneously (n = 3) or orally (n = 3) or were untreated sentinels (n = 2). Tissue reactions following injection in Phase I of the study necessitated adjustment of the preparation with reduction of the RRR-α-tocopherol concentration to 500 IU/mL in Phase 2. Following an 8-week washout period, horses received the reciprocal treatment route with the new preparation (5000 IU/450 kg of 500 IU/mL). Serum, CSF and muscle α-tocopherol concentrations were determined by high-performance liquid chromatography over a 14-day period during each phase. Cell senescence is due to the permanent cell cycle arrest that occurs as a result of the inherent limited replicative capacity toward the Hayflick limit (replicative senescence), or in response to various stressors (stress-induced premature senescence, SIPS). With the acquisition of the senescence-associated secretory phenotype (SASP), cells release several molecules (cytokines, proteases, lipids), and express the senescence-associated beta-galactosidase (SA-β-Gal). Here we tested whether vitamin E affects SA-β-Gal in an in vitro model of cell ageing. Skin fibroblasts from human subjects of different age (1, 13, 29, 59, and 88 years old) were cultured until they reached replicative senescence. At different passages (Passages 2, 9, 13, and 16), these cells were treated with vitamin E for 24 hr. Vitamin E reduced SA-β-Gal in all cells at passage 16, but at earlier passage numbers it reduced SA-β-Gal only in cells isolated from the oldest subjects. Therefore, short time treatment with vitamin E decreases SA-β-Gal in cells both from young and old subjects when reaching replicative senescence; but in cells isolated from older subjects, a decrease in SA-β-Gal by vitamin E occurs also at earlier passage numbers. The possible role of downregulation of CD36 by vitamin E, a scavenger receptor essential for initiation of senescence and SASP, is discussed.

BACKGROUND The aim of this study was to investigate the metabolism of α-tocopherol derived from vitamin E-enriched transgenic maize (VER) and its effects on antioxidant and immune functions in broilers aged 1-42 days. A total of 360 1-day-old male broilers were randomly divided into three groups containing six replicates with 20 broilers per replicate. The negative control (NC) group and the positive control (PC) group were given non-GM maize and non-GM maize plus exogenous vitamin E (VE), respectively, and the VER group was given VER, replacing the non-GM maize given to the NC group. Between days 1 and 21 and days 22 and 42, VE levels were 4.38 and 4.63 mg kg-1 in the NC group, and 14.11 and 14.91 mg kg-1 in the PC and VER group, respectively. RESULTS The results showed that α-tocopherol from both VER and additives increased α-tocopherol transfer protein and cytochrome P450 concentrations. Norflurazon 100 µM alone or in combination with α-tocopherol (0.25 mM) was applied in pre-emergence of peanut seedlings (Arachis hypogaea L.). Norflurazon treatment allowed to partially or totally photobleach plants which were noticeably smaller than the control. Norflurazon impaired the photosynthetic activity by decreasing photosynthetic pigments (carotenoids and chlorophylls) and by reducing quantities of soluble sugar. The determination of malondialdehyde (MDA) showed that its content was higher in treated plants in relation with enhancement of reactive oxygen species by the herbicide and decreased the endogenous α-tocopherol. The addition of exogenous α-tocopherol reduced the damage done by the herbicide at the membrane level because of the MDA content was less important than in norflurazon treated seedlings. Furthermore, the norflurazon decreased the glutathione S-transferase (GST) activity in the leaves and the roots of peanut seedlings, while it increased the level of reduced glutathione. This activity decreased even more with the application of exogenous α-tocopherol in combination with the herbicide. The herbicide alone or in association with the antioxidant α-tocopherol increased ascorbic acid content. The supplementation of α-tocopherol did not decrease the phytotoxicity of norflurazon although we observed a decrease in MDA content.
