In this paper, the mechanical properties of polyvinyl chloride (PVC)-coated woven fabric at high-temperature and after exposure to high-temperature were studied using a uniaxial tensile test. The test temperature range is 20 °C to 170 °C. The high-temperature test shows that the maximum stress at 170 °C reduced to 57%−59% of that at 20 °C. The after exposure to high-temperature test of prestressing shows that the combined effect of high-temperature and constant external force enhanced the stiffness and reduced the material’s impact toughness. The experimental phenomena can be explained by polymer physics.

Loop-extrusion and phase-separation have been proposed as mechanisms that shape chromosome large-scale spatial organization. It is unclear, however, how they perform relative to each other in explaining chromatin architecture data and whether they compete or co-exist at the single-molecule level. Here, we compare models of polymer physics based on loop-extrusion and phase-separation, as well as models where both mechanisms act simultaneously in a single molecule, against multiplexed FISH data available in human loci in IMR90 and HCT116 cells. We find that the different models recapitulate bulk Hi-C and average microscopy data. Single-molecule chromatin conformations are also well captured, especially by phase-separation based models that better reflect the experimentally reported segregation in globules of the considered genomic loci and their cell-to-cell structural variability. Such a variability is consistent with two main concurrent causes: single-cell epigenetic heterogeneity and an intrinsic thermodynamic conformational degeneracy of folding. Overall, the model combining loop-extrusion and polymer phase-separation provides a very good description of the data, particularly higher-order contacts, showing that the two mechanisms can co-exist in shaping chromatin architecture in single cells.

Mechanophores are molecules that are incorporated into a host material and react to the local mechanical condition—the state of stress or strain—of that host material. Among their many purposes is that of a reporter: Mechanophores whose optical activity changes in response to mechanical cues can reveal bulk material processes that are ordinarily hidden, such as fatigue and fracture. Moreover, they may do so well before a material is fully fractured. To extract quantitative information from the optical signals from embedded mechanophores it is important that the mechanophores, which are generally a minority component of the material, report proportionally and unambiguously on the mechanical condition of the bulk. This is particularly important for early reporting of damage and wear, for which the optical signal from the mechanophore should accurately reflect bulk bond scission. In this article, we develop and analyze a general theory for the quality of optical mechanoreporting by mechanophores in soft materials, based on the Bell-Evans theory of bond breaking. We find, that at the typical low fractions in which mechanophores are incorporated the overall change in strength is limited, but that the proportionality of the reporting can be off by significant amounts, particularly at short times after loading but, for non-scissile bonds, at long times as well.

Plants have evolved complex nanofibril-based cell walls to meet diverse biological and physical constraints. How strength and extensibility emerge from the nanoscale-to-mesoscale organization of growing cell walls has long been unresolved. We sought to clarify the mechanical roles of cellulose and matrix polysaccharides by developing a coarse-grained model based on polymer physics that recapitulates aspects of assembly and tensile mechanics of epidermal cell walls. Simple noncovalent binding interactions in the model generate bundled cellulose networks resembling that of primary cell walls and possessing stress-dependent elasticity, stiffening, and plasticity beyond a yield threshold. Plasticity originates from fibril-fibril sliding in aligned cellulose networks. This physical model provides quantitative insight into fundamental questions of plant mechanobiology and reveals design principles of biomaterials that combine stiffness with yielding and extensibility.

The development of controlled/living polymerization has contributed significantly to the exploration of new phenomena and applications in polymer physics and materials science. This method has recently been translated into the supramolecular domain, where it is likewise expected to open the door to fruitful fields of research; however, the principles of monomer design remain elusive. In this focus review, I present a case study of a series of porphyrin-based monomers that we have investigated over the past 10 years. By systematically designing monomers, we have been able to gain deep insights into the mechanisms of controlled/living supramolecular polymerization, and we have even been able to apply the developed method to synthesize two-dimensional supramolecular nanosheets with controlled areas and aspect ratios. This case study provides evidence for how nanostructures with precisely defined dimensions and optimized properties can be created using appropriate monomer design. Recently, controlled/living supramolecular polymerization has been evolved by many research groups; however, the principles of monomer design remain elusive. Despite their wide potential utility, the manufacture of polymer-nanoparticle (NP) composites with high filler fractions presents significant challenges because of difficulties associated with dispersing and mixing high volume fractions of NPs in polymer matrices. Polymer-infiltrated nanoparticle films (PINFs) circumvent these issues, allowing fabrication of functional composites with extremely high filler fractions (50 vol %). In this work, we present a one-step, room-temperature method for porous PINF fabrication through solvent-driven infiltration of polymer (SIP) into NP packings from a bilayer film composed of a densely packed layer of NPs atop a polymer film. Upon exposure to solvent vapor, capillary condensation occurs in the NP packing, leading to plasticization of the polymer layer and subsequent infiltration of polymer into the NP layer. This process results in a porous PINF without the need for energy-intensive processes. We show that the extent of polymer infiltration depends on the quality of solvent and the duration of solvent annealing as well as the molecular weight of the polymer. SIP can also be induced using a slightly poor solvent, which offers a great advantage of inducing SIP via liquid solvent annealing, eliminating potential hazards associated with solvent vapor annealing. The SIP process circumvents challenges associated with dispersing high concentrations of nanoparticles in a polymer matrix to prepare a nanocomposite film with high filler fraction. Thus, SIP is a potentially scalable method that can be used for the manufacturing of porous PINFs of a wide range of compositions, structures, and functionalities for applications in structural and barrier coatings as well as electrodes for energy storage and conversion devices.

Solvents have an essential association with polymer solution behavior. However, few researches have been deeply done on this respect. In recent years, our research group focus on the study on effect of solvent properties on solution behavior and film condensed state structure for semi-rigid conjugated polymer up till to apply for optoelectronic device. Herein, influence of solvent properties including solubility of solvent, aromaticity, polarity and hydrogen bonds on semi-rigid polymer chain solution behavior, i.e., single chain conformation, chain shape, size and chains aggregated density were studied by means of static/dynamic laser light scattering (DLS/SLS) and exponential law etc. Effect of solvent properties on condensed state structure of the semi-rigid conjugated polymer film was studied by UV absorption spectroscopy, PL spectroscopy and electron microscopy etc. The essential reasons for the influence were discovered and the mechanism was revealed. It was found that solution behavior with different solvent properties had an essential physical relationship with chains condensed state structure of the semi-rigid conjugated polymers. More importantly, there was a quantitative structure-activity relationship between solution and film. The key to this relationship depended on the interaction between solvent molecules and the semi-rigid conjugated polymer chains. This interaction could also affect optoelectronic devices performance. This study is of great significance to effectively control the condensed state structure of the semirigid conjugated polymers in the process of dynamic evolution from solutions to films. It not only enriches the knowledge and understanding of both semi-rigid conjugated polymer solution behaviors and film condensed state physics based on polymer physics, but also is meaningful to practical application for conjugated polymer and other traditional polymer systems.

Heat propagation in quasi-one-dimensional materials (Q1DMs) often appears puzzling. For example, while an isolated Q1DM, such as a nanowire, a carbon nanotube, or a polymer, can exhibit a high thermal conductivity κ, forests of the same materials can show a reduction in κ. Until now, the complex structures of these assemblies have hindered the emergence of a clear molecular picture for this intriguing phenomenon. We combine coarse-grained simulations with concepts known from polymer physics and thermal transport to unveil a generic microscopic picture of κ reduction in molecular forests. We show that a delicate balance among the persistence length of the Q1DM, the segment orientations, and the flexural vibrations governs the reduction in κ.

The exploration of macromolecular topology provides common ground amongst chemists, material scientists, mathematicians, and artists. Fascinating polymer architectures, ranging from linear to branched to cyclic to star, can be constructed from myriad building blocks; their topological-dependent properties merge the beauty of symmetry with the intricacies of polymer physics. Specifically in cyclic macromolecules, the lack of chain ends result in unique physical characteristics (e.g., smaller hydrodynamics volume, lower viscosity, and lack of chain entanglement) compared to their linear counterparts. In this review, developments in the synthesis of cyclic polymers will be addressed, with an emphasis of recent advances in the past five years.

Cellular membranes are densely covered by proteins. Steric pressure generated by protein collisions plays a significant role in shaping and curving biological membranes. However, no method currently exists for measuring steric pressure at membrane surfaces. Here, we developed a sensor based on Förster resonance energy transfer (FRET), which uses the principles of polymer physics to precisely detect changes in steric pressure. The sensor consists of a polyethylene glycol chain tethered to the membrane surface. The polymer has a donor fluorophore at its free end, such that FRET with acceptor fluorophores in the membrane provides a real-time readout of polymer extension. As a demonstration of the sensor, we measured the steric pressure generated by a model protein involved in membrane bending, the N-terminal homology domain (ENTH) of Epsin1. As the membrane becomes crowded by ENTH proteins, the polymer chain extends, increasing the fluorescence lifetime of the donor. Drawing on polymer theory, we use this change in lifetime to calculate steric pressure as a function of membrane coverage by ENTH, validating theoretical equations of state. Further, we find that ENTH's ability to break up larger vesicles into smaller ones correlates with steric pressure rather than the chemistry used to attach ENTH to the membrane surface. This result addresses a long-standing question about the molecular mechanisms of membrane remodeling. More broadly, this sensor makes it possible to measure steric pressure in situ during diverse biochemical events that occur on membrane surfaces, such as membrane remodeling, ligand-receptor binding, assembly of protein complexes, and changes in membrane organization.

The use of mechanical forces to chemically transform polymers dates back decades. In recent years, the use of mechanochemistry to direct constructive transformations in polymers has resulted in a range of engineered molecular responses that span optical, mechanical, electronic and thermal properties. The chemistry that has been developed is now well positioned for use in materials science, polymer physics, mechanics and additive manufacturing. Here, we review the historical backdrop of polymer mechanochemistry, give an overview of the existing toolbox of mechanophores and associated theoretical methods, and speculate as to emerging opportunities in materials science for which current capabilities are seemingly well suited. Non-linear mechanical responses and internal, amplifying stimulus–response feedback loops, including those enabled by, or coupled to, microstructured metamaterial architectures, are seen as particularly promising. Polymer mechanochemistry converts mechanical forces in materials to chemical reactions through the response of functional groups known as mechanophores. This Review discusses the colorimetric, mechanical, chemical and electronic responses of mechanophores that may be useful in materials for strain sensing and strengthening, soft devices and additive manufacturing.

In this chapter, we outline a simple physical description of polymers, first as a single chain, explaining the concepts of an ideal chain and a real chain, and then in a solution with many chains where the enthalpic interactions with solvent molecules corroborate with the configurational entropy of the polymer chain to give rise to the rich properties we observe on a macroscopic level. The Flory-Huggins theory will be introduced in describing polymers in solution, and a discussion on some of its limitations is presented. We conclude with a brief introduction to polymer brushes and some examples of application of polymers in nanoscience.

Recent advances in microscopy of living cells have established membraneless organelles as critical elements of diverse biological processes. The body of experimental work suggests that formation of such organelles is driven by liquid-liquid phase separation, a physical process that has been studied extensively for both simple liquids and mixtures of polymers. Here, we combine molecular dynamics simulations with polymer theory to show that the thermodynamic behavior of one particular biomolecular condensate-fused in sarcoma (FUS)-can be quantitatively accounted for at the level of the chain collapse theory. First, we show that a particle-based molecular dynamics model can reproduce known phase separation properties of a FUS condensate, including its critical concentration and susceptibility to mutations. Next, we obtain a polymer physics representation of a FUS condensate by examining the behavior of a single FUS protein as a function of temperature. We use the chain collapse theory to determine the thermodynamic properties of the condensate and to characterize changes in the single-chain conformation at the onset of phase separation. Altogether, our findings suggest that the phase behavior of FUS condensates can be explained by the properties of individual FUS proteins and that the change in the FUS conformation is the main force driving for the phase separation.

We have quantified the structure of the colloidal gas-liquid interface using synchrotron X-ray reflectivity measurements on a model colloid-polymer mixture. The interfacial width shows mean-field scaling with the colloid density difference, and the density profiles appear to be monotonic. Furthermore, our measurements allow us to distinguish between different theoretical polymer descriptions commonly used to model colloid-polymer mixtures. Our results highlight the importance of capturing the correct polymer physics in obtaining a quantitative theoretical description of the colloidal gas-liquid interface.

Herein, sequence-controlled polymerization and sequential Ugi-4CR/thiol–yne click reactions inspired a novel synthetic strategy for preparing a well-defined multigraft polymer in which the sequence-controlled amino-functionalized backbones were converted to mono- or di-modular alkynyl-functionalized main chains via an Ugi-4CR and clicked with well-defined thiol end-functionalized polystyrene branches through the “graft-onto” method. The Ugi-4CR showed high conversions (above 91%), while the efficiencies for the thiol–yne click reaction were above 97%. Meanwhile, an innovative concept of “polymeric topological isomers” that had similar molecular weights but different grafted structures was proposed for the first time. These four well-defined multigraft isomeric polymers (mono-brush polymer, double-brush polymer, mono-comb polymer and double-comb polymer) provide a high-level platform for polymer physics studies. Through preliminary measurements, the dilute solution properties were found to be significantly different and to change regularly with variations in the grafted structures.

Lignin solubilization is key to a viable biorefinery because its removal leads to facile deconstruction of biomass and because the isolated lignin can serve to derive precursors of novel high-value materials. The mixing of organic solvents with water has been shown to improve biomass fractionation and lignin conversion reactions. However, generally-applicable solubilization strategies are lacking because of the remarkable variability of lignin across plant feedstocks. Here, to obtain a predictive understanding of lignin solvation, we perform molecular dynamics simulations of model lignin polymers in two mixtures of water with polar aprotic solvents: tetrahydrofuran (THF) : water and γ-valerolactone (GVL) : water. The model lignins include H-, G- and S-only homopolymers and lignins with an S : G 1 : 1 ratio. We find that a well-established theory of self-avoiding polymers in a "good"solvent describes accurately the physical conformations of all types of lignin in both solvents. As the degree of methoxy substitution increases in the homopolymers, the distributions of the lignin radius of gyration and the Flory exponent ν, which describes the lignin-solvent interactions, do not change in THF : water, while ν shifts to slightly higher values in GVL : water. We attribute this increase to the interaction between the methyl group of GVL with the lignin methoxy groups. We also find that the reduction in the lignin radius of gyration due to branching is accurately described by the Zimm-Stockmayer theory for both THF : water and GVL : water. The above findings validate the applicability of polymer physics concepts to lignin and suggest that GVL : water may have the most favorable interaction with S-lignin, whereas the interactions of THF : water with lignin are independent of lignin monomeric content. Tailoring interfaces with polymer brushes is a commonly used strategy to create functional materials for numerous applications. Existing methods are limited in brush thickness, the ability to generate high-density brushes of biopolymers, and the potential for regeneration. Here we introduce a scheme to synthesize ultra-thick regenerating hyaluronan polymer brushes using hyaluronan synthase. The platform provides a dynamic interface with tunable brush heights that extend up to 20 microns – two orders of magnitude thicker than standard brushes. The brushes are easily sculpted into micropatterned landscapes by photo-deactivation of the enzyme. Further, they provide a continuous source of megadalton hyaluronan or they can be covalently-stabilized to the surface. Stabilized brushes exhibit superb resistance to biofilms, yet are locally digested by fibroblasts. This brush technology provides opportunities in a range of arenas including regenerating tailorable biointerfaces for implants, wound healing or lubrication as well as fundamental studies of the glycocalyx and polymer physics., Polymer brushes are often used to functionalise surfaces. Here, the authors report on the creation of high-density, regenerating hyaluronan brushes using hyaluronan synthase enzymes which can be patterned by photo-deactivation and demonstrate biocompatibility and resistance to biofilm formation.

This study aims to use molecular dynamics (MD) simulations of Kremer–Grest (KG) chains to inform future developments of models of entangled polymer dynamics. We perform nonequilibrium MD simulations, under shear flow, for well‐entangled KG chains. We study chains of 512 and 1000 KG beads, corresponding to 8 and 15 entanglements, respectively. We compute the linear rheological properties from equilibrium simulations of the stress autocorrelation and obtain from these data the tube model parameters. Under nonlinear shear flow, we compute the shear viscosity, the first and second normal stress differences, and chain contour length. For chains of 512 monomers, we obtain agreement with the results of Cao and Likhtman (ACS Macro. Lett. 2015, 4, 1376). We also compare our nonlinear results with the Graham, Likhtman and Milner‐McLeish (GLaMM) model. We identify some systematic disagreement that becomes larger for the longer chains. We made a comparison of the transient shear stress maximum from our simulations, two nonlinear models and experiments on a wide range of melts and solutions, including polystyrene (PS), polybutadiene, and styrene–butadiene rubber. This comparison establishes that the PS melt data show markedly different behavior to all other melts and solutions and KG simulations reproduce the PS data more closely than either the GLaMM or Xie and Schweizer models. We discuss the performance of these models against the data and simulations. Finally, by imposing a rapid reversing flow, we produce a method to extract the recoverable strain from MD simulations, valid for sufficiently entangled monodisperse polymers. The next materials challenge in organic stretchable electronics is the development of a fully degradable semiconductor that maintains stable electrical performance under strain. Herein, we decouple the design of stretchability and transience by harmonizing polymer physics principles and molecular design in order to demonstrate for the first time a material that simultaneously possesses three disparate attributes: semiconductivity, intrinsic stretchability, and full degradability. We show that we can design acid-labile semiconducting polymers to appropriately phase segregate within a biodegradable elastomer, yielding semiconducting nanofibers that concurrently enable controlled transience and strain-independent transistor mobilities. Along with the future development of suitable conductors and device integration advances, we anticipate that these materials could be used to build fully biodegradable diagnostic or therapeutic devices that reside inside the body temporarily, or environmental monitors that are placed in the field and break down when they are no longer needed. This fully degradable semiconductor represents a promising advance toward developing multifunctional materials for skin-inspired electronic devices that can address previously inaccessible challenges and in turn create new technologies., A concurrently stretchable and degradable semiconducting polymeric system with strain-independent electrical performance is achieved by harnessing both polymer chemistry and polymer physics.

Since the discovery of conductive poly(acetylene), the study of conjugated polymers has remained an active and interdisciplinary frontier between polymer chemistry, polymer physics, computation, and device engineering. One of the ultimate goals of polymer science is to reliably synthesize structures, similar to small molecule synthesis. Kumada catalyst-transfer polymerization (KCTP) is a powerful tool for synthesizing conjugated polymers with predictable molecular weights, narrow dispersities, specific end groups, and complex backbone architectures. However, expanding the monomer scope beyond the well-studied 3-alkylthiophenes to include electron-deficient and complex heterocycles has been difficult. Revisiting the successful applications of KCTP can help us gain new insight into the CTP mechanisms and thus inspire breakthroughs in the controlled polymerization of challenging π-conjugated monomers.In this Account, we highlight our efforts over the past decade to achieve controlled synthesis of homopolymers (p-type and n-type), copolymers (diblock and statistical), and monodisperse high oligomers. We first give a brief introduction of the mechanism and state-of-the-art of KCTP. Since the extent of polymerization control is determined by steric and electronic effects of both the catalyst and monomer, the polymerization can be optimized by modifying monomer and catalyst structures, as well as finding a well-matched monomer-catalyst system. We discuss the effects of side-chain steric hindrance and halogens in the context of heavy atom substituted monomers. By moving the side-chain branch point one carbon atom away from the heterocycle to alleviate steric crowding and stabilize the catalyst resting state, we were able to successfully control the polymerization of new tellurophene monomers. Inspired by innocent role of the sterically encumbered 2-transmetalated 3-alkylthiophene monomer, we introduce the treatment of hygroscopic monomers with a bulky Grignard compound as a water-scavenger for the improved synthesis of water-soluble conjugated polymers. For challenging electron-deficient monomers, we discuss the design of new Ni(II)diimine catalysts with electron-donating character which enhance the stability of the association complex between the catalyst and the growing polymer chain, resulting in the quasi-living synthesis of n-type polymers. Beyond n-type homopolymers, the Ni(II)diimine catalysts are also capable of producing electron-rich and electron-deficient diblock and statistical copolymers. We discuss how density functional theory (DFT) calculations elucidate the role of catalyst steric and electronic effects in controlling the synthesis of π-conjugated polymers. Moreover, we demonstrate the synthesis of monodisperse high oligomers by temperature cycling, which takes full advantage of the unique character of KCTP in that it proceeds through distinct intermediates that are not reactive. The insight we gained thus far leads to the first example of isolated living conjugated polymer chains prepared by a standard KCTP procedure, with general applicability to different monomers and catalytic systems. In summarizing a decade of innovation in KCTP, we hope this Account will inspire future development in the field to overcome key challenges including the controlled synthesis of electron-deficient heterocycles, complex and high-performance systems, and degradable and recyclable materials as well as cutting-edge catalyst design.

We present computer simulations of a dynamic Monte Carlo algorithm for polymer chains on the FCC lattice which takes explicitly into account the possibility to overcome topological constraints by controlling the rate at which nearby polymer strands may cross through each other. By applying the method to systems of interacting ring polymers at melt conditions, we characterize their structure and dynamics by measuring, in particular, the amounts of knots and links which are formed during the relaxation process. In comparison to standard melts of unknotted and unconcatenated rings, our simulations demonstrate that the mechanism of strand crossing is responsible for fluidizing the melt provided the time scale of the process is faster than the internal relaxation of the chain, in agreement with recent experiments employing solutions of DNA rings in the presence of the type II topoisomerase enzyme. In the opposite case of slow rates the melt is shown to become slower, and this prediction may be easily validated experimentally., 12 pages, 9 figures in the main text, 8 supplementary figures. Polymers with precisely defined monomeric sequences present an exquisite tool for controlling material properties by harnessing both the robustness of synthetic polymers and the ability to tailor the inter- and intramolecular interactions so crucial to many biological materials. While polymer scientists traditionally synthesized and studied the physics of long molecules best described by their statistical nature, many biological polymers derive their highly tailored functions from precisely controlled sequences. Therefore, significant effort has been applied toward developing new methods of synthesizing, characterizing, and understanding the physics of non-natural sequence-defined polymers. This perspective considers the synergistic advantages that can be achieved via tailoring both precise sequence control and attributes of traditional polymers in a single system. Here, we focus on the potential of sequence-defined polymers in highly associating systems, with a focus on the unique properties, such as enhanced proton conductivity, that can be attained by incorporating sequence. In particular, we examine these materials as key model systems for studying previously unresolvable questions in polymer physics including the role of chain shape near interfaces and how to tailor compatibilization between dissimilar polymer blocks. Finally, we discuss the critical challenges-in particular, truly scalable synthetic approaches, characterization and modeling tools, and robust control and understanding of assembly pathways-that must be overcome for sequence-defined polymers to attain their potential and achieve ubiquity.

A fundamental paradigm in polymer physics is that macromolecular conformations in equilibrium can be described by universal scaling laws, being key for structure, dynamics, and function of soft (biological) matter and in the materials sciences. Here, we reveal that during diffusion-influenced, nonequilibrium chain-growth polymerization, scaling laws change qualitatively, in particular, the growing polymers exhibit a surprising self-avoiding walk (SAW) behavior in poor and theta-solvents. Our analysis, based on monomer-resolved reaction-diffusion computer simulations, demonstrates that this phenomenon is a result of i) nonequilibrium monomer density depletion correlations around the active polymerization site, leading to a locally directed and self-avoiding growth, in conjunction with ii) chain (Rouse) relaxation times larger than the competing polymerization reaction time. These intrinsic nonequilibrium mechanisms are facilitated by fast and persistent reaction-driven diffusion (sprints) of the active site, with analogies to pseudo-chemotactic active Brownian particles. α1-Antitrypsin is a protease inhibitor belonging to the serpin family. Serpin polymerisation is at the core of a class of genetic conformational diseases called serpinopathies. These polymers are known to be unbranched, flexible, and heterogeneous in size with a beads-on-a-string appearance viewed by negative stain electron microscopy. Here, we use atomic force microscopy and time-lapse dynamic light scattering to measure polymer size and shape for wild-type (M) and Glu342→Lys (Z) α1-antitrypsin, the most common variant that leads to severe pathological deficiency. Our data for small polymers deposited onto mica and in solution reveal a power law relation between the polymer size, namely the end-to-end distance or the hydrodynamic radius, and the polymer mass, proportional to the contour length. We use the scaling concepts of polymer physics to assess that α1-antitrypsin polymers are random linear chains with a low persistence length.

The dynamics of polymer-nanoparticle (NP) mixtures, which involves multiple scales and system-specific variables, has posed a long-standing challenge on its theoretical description. In this paper, we construct a microscopic theory for polymer diffusion in the mixtures based on a combination of generalized Langevin equation, mode-coupling approach, and polymer physics ideas. The parameter-free theory has an explicit expression and remains tractable on pair correlation level with system-specific equilibrium structures as input. Taking a minimal polymer-NP mixture as an example, our theory correctly captures the dependence of polymer diffusion on NP concentration and average interparticle distance. Importantly, the polymer diffusion exhibits a power law decay as the polymer length increases at dense NPs and/or long chain, which marks the emergence of entanglement-like motion. Novel technologies are revealing that chromosomes have a complex three-dimensional organization within the cell nucleus that serves functional purposes. Models from polymer physics have been developed to quantitively understand the molecular principles controlling their structure and folding mechanisms. Here, by using massive molecular-dynamics simulations we show that classical scaling laws combined with finite-size effects of a simple polymer model can effectively explain the scaling behavior that chromatin exhibits at the topologically associating domains level, as revealed by experimental observations. Model results are then validated against recently published high-resolution in situ Hi-C data.

Polyelectrolyte brushes are important stimuli-responsive materials in a variety of technological applications as well as in biological systems. Their small size, however, introduces characterization challenges, particularly in studying 3D structure and time-dependent behavior. In this Letter, we report on the polyelectrolyte brush behavior of extra-large hyaluronan brushes (∼15 μm) recently developed using an enzyme-mediated growth process. In response to increasing ionic strength, the brush displays the osmotic brush regime and the salted brush regime. We also show a collapse of 96% when the brush is placed in a poor solvent. This collapse is rapid when changing from a good to poor solvent, but re-expansion is slow when changing back to a good solvent. The observed brush behavior described in this Letter is similar to that seen for smaller polyelectrolyte brushes, indicating that these larger brushes may serve as model systems to study more complex phenomena through confocal microscopy.

Internal dynamics of flexible polymer chains is one of the most important fundamental problems in polymer physics. Although numerous efforts have been devoted to the understanding of internal dynamics for linear polymers, little attention was paid to branched systems. This work aims to elucidate how the branching effect quantitatively influences the internal motions of long-subchain hyperbranched polymers in dilute solution. By light scattering study of four hyperbranched polystyrene samples with controlled branching patterns and low polydispersities (Mw/Mn < 1.40 and Mw ∼ 107 g/mol), we quantified and analyzed the asymptotic behavior of the reduced first cumulant [Γ* = Γ(q)/(q3kBT/η0)], the value of Γ* at high q-regime [the approximate values of Γ*(∞)], the scattering vector (q) dependence of linewidth [Γ(q)], the number of relaxation modes in Γ(q)-distribution curves, the characteristic relaxation time (τ1) of the first internal mode, and the q-dependent relative strength of internal motions. Theoretical models of the conformational behavior of flexible polymer chains in mixed solvents enunciated in the world literature during the last decade are critically reviewed. Models describing different mechanisms of coil-to-globule transitions in a good solvent induced by cosolvent addition are highlighted. Special attention is given to the analysis of theoretical approaches to describing the collapse of polymer chains in binary mixtures of good solvents. The review addresses researchers engaged in polymer physics and chemistry and materials scientists involved in the design of smart polymers.

2D nanoslit devices, where two crystals with atomically flat surfaces are separated by only a few nanometers, have attracted considerable attention because their tunable control over the confinement allows for the discovery of unusual transport behavior of gas, water, and ions. Here, the passage of double-stranded DNA molecules is studied through nanoslits fabricated from exfoliated 2D materials, such as graphene or hexagonal boron nitride, and the DNA polymer behavior is examined in this tight confinement. Two types of events are observed in the ionic current: long current blockades that signal DNA translocation and short spikes where DNA enters the slits but withdraws. DNA translocation events exhibit three distinct phases in their current-blockade traces—loading, translation, and exit. Coarse-grained molecular dynamics simulation allows the different polymer configurations of these phases to be identified. DNA molecules, including folds and knots in their polymer structure, are observed to slide through the slits with near-uniform velocity without noticeable frictional interactions of DNA with the confining graphene surfaces. It is anticipated that this new class of 2D-nanoslit devices will provide unique ways to study polymer physics and enable lab-on-a-chip biotechnology.

Knots in long DNA molecules are prevalent in biological systems and serve as a model system for investigating static and dynamic properties of biopolymers. We explore the dynamics of knots in double-stranded DNA in a new regime of nanometer-scale confinement, large forces, and short time scales, using solid-state nanopores. We show that DNA knots undergo isomorphic translocation through a nanopore, retaining their equilibrium morphology by swiftly compressing in a lateral direction to fit the constriction. We observe no evidence of knot tightening or jamming, even for single-digit nanopores. We explain the observations as the malleability of DNA, characterized by sharp buckling of the DNA in nanopores, driven by the transient disruption of base pairing. Our molecular dynamics simulations support the model. These results are relevant not only for the understanding of DNA packing and manipulation in living cells but also for the polymer physics of DNA and the development of nanopore-based sequencing technologies.

The random motion of a flexible molecular chain floating in a solvent or a melt can itself be quite complex. The discussion in this chapter starts with some basic concepts of an ideal polymer chain. It is then followed by classical models describing the dynamics and diffusion of polymer chains applicable to polymer melts. This chapter’s main objective is to contrast the differences between the mobility of polymer chains in melts (or solvents) and the kinetically trapped glassy state.

The static light scattering (SLS) method is introduced with key equations used to extract quantitative information such as weight-average molecular weight, osmotic second virial coefficient, and radius of gyration from the experimental data. The use of Zimm plots to obtain visual cues of the polymer's characteristics is presented. A treatise of the thermodynamic process (modified by structural parameters) sensed by light scattering from a single atom, multiple atoms, fluids, small and large polymers is presented with reliance on a plethora of equations. The design of the SLS instrument as well as a few case studies of SLS applications are summarized to help the user understand the experiment better and to discern compromises in commercial instruments. The opportunity in combining DLS and SLS to obtain a complete picture of polymer characterization and polymer physics closes the loop on these light scattering techniques.

Ring polymers exhibit unique flow properties due to their closed chain topology. Despite recent progress, we have not yet achieved a full understanding of the nonequilibrium flow behavior of rings in nondilute solutions where intermolecular interactions greatly influence chain dynamics. In this work, we directly observe the dynamics of DNA rings in semidilute ring-linear polymer blends using single molecule techniques. We systematically investigate ring polymer relaxation dynamics from high extension and transient and steady-state stretching dynamics in planar extensional flow for a series of ring-linear blends with varying ring fraction. Our results show multiple molecular sub-populations for ring relaxation in ring-linear blends, as well as large conformational fluctuations for rings in steady extensional flow, even long after the initial transient stretching process has subsided. We further quantify the magnitude and characteristic timescales of ring conformational fluctuations as a function of blend composition. Interestingly, we find that the magnitude of ring conformational fluctuations follows a non-monotonic response with increasing ring fraction, first increasing at low ring fraction and then substantially decreasing at large ring fraction in ring-linear blends. A unique set of ring polymer conformations are observed during the transient stretching process, which highlights the prevalence of molecular individualism and supports the notion of complex intermolecular interactions in ring-linear polymer blends. Together with results from molecular simulations, our results suggest that ring conformational fluctuations arise due to ring-linear threading and intermolecular hydrodynamic interactions (HI). Simulating soft matter systems such as the cytoskeleton can enable deep understanding of experimentally observed phenomena. One challenge of modeling such systems is realistic description of the steric repulsion between nearby polymers. Previous models of the polymeric excluded volume interaction have the deficit of being non-analytic, being computationally expensive, or allowing polymers to erroneously cross each other. A recent solution to these issues, implemented in the MEDYAN simulation platform, uses analytical expressions obtained from integrating an interaction kernel along the lengths of two polymer segments to describe their repulsion. Here, we extend this model by re-deriving it for lower-dimensional geometrical configurations, deriving similar expressions using a steeper interaction kernel, comparing it to other commonly used potentials, and showing how to parameterize these models. We also generalize this new integrated style of potential by introducing a segmental Lennard-Jones potential, which enables modelling both attractive and repulsive interactions in semi-flexible polymer networks. Since chemical shift interaction was discovered in 1953, nuclear magnetic resonance (NMR) spectroscopy has emerged as one of the most promising tools in polymer chemistry as well as polymer physics. In this chapter, basic principles of solution- and solid-state NMR spectroscopy will be introduced and recent NMR applications to polymer structure and dynamics will be reviewed.

Many emerging materials, such as ultrastable glasses1,2 of interest for phone displays and OLED television screens, owe their properties to a gradient of enhanced mobility at the surface of glass-forming liquids. The discovery of this surface mobility enhancement3–5 has reshaped our understanding of the behaviour of glass formers and of how to fashion them into improved materials. In polymeric glasses, these interfacial modifications are complicated by the existence of a second length scale—the size of the polymer chain—as well as the length scale of the interfacial mobility gradient6–9. Here we present simulations, theory and time-resolved surface nano-creep experiments to reveal that this two-scale nature of glassy polymer surfaces drives the emergence of a transient rubbery, entangled-like surface behaviour even in polymers comprised of short, subentangled chains. We find that this effect emerges from superposed gradients in segmental dynamics and chain conformational statistics. The lifetime of this rubbery behaviour, which will have broad implications in constraining surface relaxations central to applications including tribology, adhesion, and surface healing of polymeric glasses, extends as the material is cooled. The surface layers suffer a general breakdown in time−temperature superposition (TTS), a fundamental tenet of polymer physics and rheology. This finding may require a reevaluation of strategies for the prediction of long-time properties in polymeric glasses with high interfacial areas. We expect that this interfacial transient elastomer effect and TTS breakdown should normally occur in macromolecular systems ranging from nanocomposites to thin films, where interfaces dominate material properties5,10. Surface enhancements in glass mobility are complicated in polymers by the interplay of the surface mobile layer thickness with a second length scale (the size of the polymer chains), giving rise to a transient rubbery surface even in polymers with short chains.

Water-mediated interactions (WMIs) play diverse roles in molecular biology. They are particularly relevant in geometrically confined spaces such as the interior of the chaperonin, at the interface between ligands and their binding partners, and in the ribosome tunnel. Inspired in part by the geometry of the ribosome tunnel, we consider confinement effects on the stability of peptides. We describe results from replica exchange molecular dynamics simulations of a system containing a 23-alanine or 23-serine polypeptide confined to non-polar and polar nanotubes in the gas phase and when open to a water reservoir. We quantify the effect of water in determining the preferred conformational states of these polypeptides by calculating the difference in the solvation free energy for the helix and coil states in the open nanotube in the two phases. Our simulations reveal several possibilities. We find that nanoscopic confinement preferentially stabilizes the helical state of polypeptides with hydrophobic side chains, which is explained by the entropic stabilization mechanism proposed on the basis of polymer physics. Polypeptide chains with hydrophilic side chains can adopt helical structures within nanotubes, but helix formation is sensitive to the nature of the nanotube due to WMIs. We elaborate on the potential implications of our findings to the stability of peptides in the ribosome tunnel.

The native state structures of globular proteins are stable and well-packed indicating that self-interactions are favored over protein-solvent interactions under folding conditions. We use this as a guiding principle to derive the geometry of the building blocks of protein structures, alpha-helices and strands assembled into beta-sheets, with no adjustable parameters, no amino acid sequence information, and no chemistry. There is an almost perfect fit between the dictates of mathematics and physics and the rules of quantum chemistry. Our theory establishes an energy landscape that channels protein evolution by providing sequence-independent platforms for elaborating sequence-dependent functional diversity. We introduce a powerful, widely applicable approach to characterizing polymer conformational distributions, specifically the end-to-end distance distributions, P(Ree), accessed through double electron-electron resonance (DEER) spectroscopy in conjunction with molecular dynamics (MD) simulations. The technique is demonstrated on one of the most widely used synthetic, disordered, water-soluble polymers: poly(ethylene oxide) (PEO). Despite its widespread importance, no systematic experimental characterization of PEO's Ree conformational landscape exists. The evaluation of P(Ree) is particularly important for short polymers or (bio)polymers with sequence complexities that deviate from simple polymer physics scaling laws valid for long chains. In this study, we characterize the Ree landscape by measuring P(Ree) for low molecular weight (MW: 0.22-2.6 kDa) dilute PEO chains. We use DEER with end-conjugated spin probes to resolve Ree populations from ∼2-9 nm and compare them with full distributions from MD. The P( Ree)'s from DEER and MD show remarkably good agreement, particularly at longer chain lengths where populations in the DEER-unresolvable range ( 10 kDa) PEO and the P(Ree)'s crossover to the theoretical distribution for an excluded volume chain.

We study the structural ensembles of human chromosomes across different cell types. Using computer simulations, we generate cell-specific 3D chromosomal structures and compare them to recently published chromatin structures obtained through microscopy. We demonstrate using a combination of machine learning and polymer physics simulations that epigenetic information can be used to predict the structural ensembles of multiple human cell lines. The chromosomal structures obtained in silico are quantitatively consistent with those obtained through microscopy as well as DNA-DNA proximity ligation assays. Theory predicts that chromosome structures are fluid and can only be described by an ensemble, which is consistent with the observation that chromosomes exhibit no unique fold. Nevertheless, our analysis of both structures from simulation and microscopy reveals that short segments of chromatin make transitions between a closed conformation and an open dumbbell conformation. This conformational transition appears to be consistent with a two-state process with an effective free energy cost of about four times the effective information theoretic temperature. Finally, we study the conformational changes associated with the switching of genomic compartments observed in human cell lines. Genetically identical but epigenetically distinct cell types appear to rearrange their respective structural ensembles to expose segments of transcriptionally active chromatin, belonging to the A genomic compartment, towards the surface of the chromosome, while inactive segments, belonging to the B compartment, move to the interior. The formation of genomic compartments resembles hydrophobic collapse in protein folding, with the aggregation of denser and predominantly inactive chromatin driving the positioning of active chromatin toward the surface of individual chromosomal territories.
