Cell permeability and epithelial-mesenchymal transition (EMT) were found to be enhanced in diabetic retinopathy, and the aim of this study was to investigate the underlying mechanism. ARPE-19 cell line or primary retinal pigment epithelial (RPE) cells were cultured under high or normal glucose conditions. Specific shRNAs were employed to knock down ADP-ribosylation factor 6 (ARF6), GEP100, or VEGF receptor 2 (VEGFR2) in ARPE-19 or primary RPE cells. Cell migration ability was measured using Transwell assay. Western blotting was used to measure indicated protein levels. RPE cells treated with high glucose showed increased cell migration, paracellular permeability, EMT, and expression of VEGF. Knockdown of VEGFR2 inhibited the high-glucose-induced effects on RPE cells via inactivation of ARF6 and MAPK pathways. Knockdown ARF6 or GEP100 led to inhibition of high-glucose-induced effects via inactivation of VEGFR2 pathway. Knockdown of ARF6, but not GEP100, decreased high-glucose-induced internalization of VEGFR2. High-glucose enhances EMT and cell permeability of RPE cells through activation of VEGFR2 and ARF6/GEP100 pathways, which form a positive feedback loop to maximize the activation of VEGF/VEGFR2 signaling.

Legacy organic contaminants, such as polychlorinated biphenyls (PCBs) and polybrominated diphenyl ethers (PBDEs), and several currently used organic contaminants, such as novel brominated retardants (NBFRs), organophosphate flame retardants (OPFRs), and Dechlorane Plus (DPs), were measured in the segmented hair and hand wipes of e-waste dismantling workers and in the dust from their workplace in an e-waste recycling site in South China to assess the accumulation and sources of organic contaminants in the hair shaft. The median concentrations of DPs, PBDEs, NBFRs, PCBs and OPFRs in hair samples were 23.5, 154, 156, 349, and 371 ng/g, respectively. A linear increase in organic contaminant concentrations was found along the hair shaft, with significant differences among each segment, while the age-related patterns were similar among the 7 hair segments. A linear relationship was found between the accumulation rates and the log KOW, indicating that the accumulation rates of organic contaminants along the hair shaft decreased with increasing log KOW values. Enantiomer fraction (EF) values of PCB-132 in distal segments were close to those in dust and air, while EFs in proximal segments were close to those in serum. The contributions of PCBs from external sources to hair increased with the distance from the scalp of hair segments, from 67.8% in 0-3 cm segments to 95.9% in 18-21 cm segments.

To explore the possible roles of metallothioneins (MTs) played in cadmium (Cd) accumulation of water spinach, three IaMT genes, IaMT1, IaMT2 and IaMT3 in a high-shoot-Cd (T308) and a low-shoot-Cd accumulation cultivar (QLQ) were cloned, characterized, and quantitated. Gene expression analysis suggested that the expression of the IaMTs was differentially regulated by Cd stress in different cultivars, and T308 showed higher MTs expression overall. Furthermore, only shoot IaMT3 expression was cultivar dependent among the three IaMTs. Antioxidant analysis showed that the high production of IaMTs in T308 should be associated with its high oxidation resistance. The role of IaMTs in protecting against Cd toxicity was demonstrated in vitro via recombinant E. coli strains. The results showed that IaMT1 correlated with neither Cd tolerance nor Cd accumulation of E. coli, while IaMT2 conferred Cd tolerance in E. coli, IaMT2 and IaMT3 increased Cd accumulation in E. coli. These findings help to clarify the roles of IaMTs in Cd accumulation, and increase our understanding of the cultivar-dependent Cd accumulation in water spinach.

Taxus yunnanensis (Yew) is known for natural anticancer metabolite paclitaxel (Taxol) and its biosynthesis pathway in yew species still needs to be completely elucidated. In the current study, productions of paclitaxel and 10-DAB III from three different tissues (needle, branch, and root) of T. yunnanensis wild type (WT) and two new cultivars Zhongda-1 (Zd1) and Zhongda-2 (Zd2) were determined, and significant tissue differences in contents of the taxanes were observed among the three experimental lines. The much higher 10-DAB III and lower paclitaxel contents in needle of Zd2 when compared with that of Zd1 indicated the low conversion from 10-DAB III to paclitaxel in the needle of Zd2. In order to uncover the mechanisms of the tissue-specific biosynthesis of the taxanes, transcriptome analysis of cultivar Zd2 was conducted, and the previously reported transcriptome data of Zd1 and WT were used to perform a comparison. The enhancement of TDAT and T10βH side biosynthetic pathway in roots of Zd2 in early taxane synthesis might lead to the biosynthesis of other toxoids, while the preference of T13αH route in the needle and branch of Zd2 was mainly responsible for the tissue-specific reinforced biosynthesis of 10-DAB III and paclitaxel in Zd2. Different from Zd1, the tissue-specific pattern of paclitaxel biosynthesis genes in Zd2 was similar to WT. However, the lower transcript abundance of final steps genes (TBT, DBAT, BAPT, and DBTNBT) of the paclitaxel biosynthesis pathway in Zd2 than in Zd1 might further promote 10-DAB III accumulation in Zd2.

Breeding for pollution-safe cultivars (PSCs) can reduce pollutant accumulation in crops. However, the PSC breeding would face the risk of nutritional quality reduction, which is usually ignored in conventional breeding programs targeting to increase crop yield or nutritional quality. Thus, the doubt whether the risk would exist has to be clarified for supporting the PSC breeding. In the present study, a newly bred Cd/Pb-PSC of water spinach (Ipomoea aquatic Forsk.) and its parents (QLQ with low-Cd/Pb accumulation ability and T308 with high yield) of water spinach were employed to clarify the above-mentioned issue. Yields, and concentrations of Cd, Pb, nitrite, and organic and inorganic nutrients in shoots of the three experimental lines were determined. There were no significant differences in Cd/Pb concentration between the new PSC and QLQ, in nitrite content between the new PSC and its two parents and in yield between the new PSC and T308. It is decisively significant that shoot concentrations of organic and inorganic nutrients in the Cd/Pb-PSC were as high as those in one of its parents. It is affirmed that the breeding operations (crossing and consequently continuous selfing) for lowering Cd/Pb accumulation capacity of water spinach would not lower the nutritional values of the obtained Cd/Pb-PSCs from the breeding, which should be a pillar that supports the feasibility to minimize Cd/Pb pollution in vegetables using PSC-breeding method.

Brominated flame retardants (BFRs) and Dechlorane Plus (DP) were measured in both human hair and paired serum samples from a cohort of university students in South China. Segmental analysis was conducted to explore gender difference and the relationships between the hair and serum. The concentrations of total PBDEs in the hair and serum samples were in a range of 0.28-34.1ng/g dry weight (dw) and 0.16-156ng/g lipid weight (lw), respectively. Concentrations of ∑DPs (sum of the syn-DP and anti-DP isomers) in all hair samples ranged from nd-5.45ng/g dry weight. Concentrations of most PBDEs and decabromodiphenylethane (DBDPE) in distal segments (5-10cm from the scalp) were higher than those in the proximal segments (0-5cm from the scalp) (t-test, p < 0.05), which could be due to the longer exposure time of distal segments. The proximal segments exhibited a unique congener profile, more close to that in the serum rather than the distal segments of hair. An obvious gender difference was found in the levels of ∑PBDEs using integrated hair samples, while the difference disappeared when considering alone the proximal segments of hair (0-5cm from scalp) for both genders. This paper provides supplement to the current knowledge on sources of BFRs and DPs in hair and declares the importance of segmental analysis.

The difference in contents of paclitaxel and 10-deacetylbaccatin III (10-DABIII) in needles between wildtype (WT) and a new cultivar (Zhongdayihao, ZD1) of Taxus yunnanensis was examined. Transcriptome profiling was conducted for different tissues of the ZD1 and WT to illustrate the regulation mechanism of paclitaxel biosynthesis. It was observed that average contents of paclitaxel and 10-DABIII in ZD1 were 4 folds and 32 folds higher than those in WT, respectively. More significant elevations of differential expressed genes (DEGs) from paclitaxel biosynthesis pathway were revealed in ZD1 rather than WT, which should be responsible for the higher contents of paclitaxel and 10-DABIII in the ZD1. Special tissues-dependent expression patterns of paclitaxel biosynthesis DEGs in ZD1 compared to WT were unraveled. The relative higher expressions of paclitaxel biosynthesis genes in needles than other tissues supported the higher content of paclitaxel and 10-DABIII content in needles of ZD1. Attenuation of plant hormone signal transduction pathway led to the lower expression of TFs in ZD1 rather than WT. Besides, the significant negative correlations between differential expressed TFs and DEGs from paclitaxel biosynthesis pathway displayed a possibly negative regulation pattern of these TFs on paclitaxel biosynthesis pathway genes. These results provided new insights into the molecular process of paclitaxel synthesis in Taxus.

Airway surface liquid (ASL) is one of the key factors affecting the respiratory system's physiological function. Abnormal ASL secretion can increase the incidence of various respiratory diseases. Lipopolysaccharide (LPS) stimulation can damage the airway epithelial barrier, affect the concentration of ASL contents, and down-regulate ion channel expression, which in turn causes abnormal ASL secretion. Naringenin, which exists in many Citrus foods, has the ability to promote airway surface liquid secretion. This work is designed to investigate the regulatory mechanism of naringenin on LPS-induced abnormal ASL secretion. The effects of naringenin and LPS on the viability of Calu-3 cells were measured by CellTiter 96® AQueous One Solution Cell Proliferation Assay (MTS). ASL secretion volume was measured by a micropipette on air–liquid interface cultured cells. The concentration of Cl−, Na+, lysozyme, and total protein in ASL were respectively measured by assay kits. The mRNA expressions were determined by quantitative real-time polymerase chain reaction, and proteins were measured by enzyme-linked immunosorbent assay. The results indicated that LPS could affect ASL secretion and regulate cystic fibrosis transmembrane conductance regulator (CFTR), aquaporin 1 (AQP1) and aquaporin 5 (AQP5) expression. Naringenin had the ability to regulate the ASL secretion by increasing secretion volume, and Cl− and Na+ concentrations, reducing lysozyme and total protein content, and regulating CFTR, AQP1, and AQP5 expression. This study indicated that naringenin had regulating effects to attenuate LPS-induced abnormal ASL secretion.

Cadmium (Cd) contamination in arable soils presents a serious threat to agricultural crops and public health. Cd pollution-safe cultivars (Cd-PSCs) can effectively restrict human intake of Cd through food chain. However, not much is known on the genetic basis of low-Cd-accumulating trait. Here, we aim to investigate the microRNA (miRNA)-mediated post-transcriptional regulation underlying different capacities of Cd accumulation in Brassica parachinensis L. Using small RNA sequencing, miRNAs were identified and characterized in two previously identified cultivars, a low- (SJ19) and a high-Cd-accumulating cultivar (CX4) of B. parachinensis. 18 known and 148 novel miRNAs were identified to be differentially expressed in response to Cd (Cd-responsive miRNAs) in the two cultivars. Distinct Cd responsive pathways were employed by the two cultivars. In CX4, the overexpression of miR395 was indicated to engage in Cd tolerance by regulating sulfur assimilation and associated with its high Cd accumulation. In SJ19, miRNA-mediated oxidative resistance was enhanced in response to Cd exposure. Furthermore, miR397, miR393 and miR160 were involved in the Cd-induced growth improvement in both roots and shoots of the low-Cd-accumulating cultivar. Our findings provide new insights into miRNAs-mediated Cd response in B. parachinensis, and shed light on molecular-assisted Cd-PSC screening and breeding for agricultural crops and vegetables.

The concentrations of several organic contaminants (OCs) and heavy metals were measured in indoor dust from e-waste recycling, rural, and urban areas in South China to illustrate the spatial characteristics of these pollutants and to further evaluate human exposure risks. The median concentrations of polychlorinated biphenyls (PCBs), polybrominated diphenyl ethers (PBDEs), decabromodiphenyl ethane (DBDPE), and dechlorane plus (DPs) were 38.6-3560, 2360-30,100, 665-2720, and 19.5-1860ng/g, while the median concentrations of Cd, Pb, Cu, Cr, and Zn were 2.46-40.4, 206-1380, 217- 1200, 25.3-134, and 176-212μg/g in indoor dust. The levels of all pollutants, except Zn, in dust from the e-waste recycling area were significantly higher than those from the other areas. Cd, Pb, and most OCs exhibited similar pollution patterns in the three areas, indicating that e-waste recycling activities are the major pollution source. In contrast, Cu, Cr, Zn, and penta-BDE are likely derived from household products in the rural and urban areas. The highest estimated daily intakes (EDIs) of PCBs, PBDEs, DBDPE, and DPs were 0.15-163, 3.97-1470, 1.26-169, and 0.11-134ng/kg bw/day for toddlers and adults. The highest EDIs of BDE 209 and Pb in toddlers in the e-waste recycling area were 16% and 18 times higher than the reference doses, indicating the high exposure risk of these pollutants in the e-waste recycling area.

Heavy metals have serious health consequences and ecosystem impacts. A pot experiment was conducted to investigate the variation of cadmium (Cd) uptake and accumulation among 40 cultivars of radish (Raphanus sativus L.) at three Cd levels, including 0.31 (T1), 0.83 (T2), and 1.13 (T3) mg kg−1. Most of the tested cultivars had higher taproot biomass in the T3 treatment when compared to those in the T1 treatment, indicating a Cd stress-induced growth in radish. Taproot Cd concentrations in 95 and 5% of the tested cultivars were lower than 0.1 mg kg−1 (fresh weight, FW) in the T1 and T2 treatments, respectively; however, there was no cultivar suitable for safe consumption in the T3 treatment. Radish production showed potential risk of Cd pollution as high as some leafy vegetables when grown in the soils where Cd concentration exceeded 0.8 mg kg−1. When compared with Chinese heat-resisting or imported cultivars, Chinese common cultivars had significantly higher taproot Cd concentrations. Three low-Cd cultivars and five high-Cd cultivars were identified. Taproot Cd concentrations showed significant correlations between any two of the three treatments (p < 0.01), suggesting that Cd accumulation in taproot of radish was genotype-dependent.

Polybrominated diphenyl ethers (PBDEs) and polychlorinated biphenyls (PCBs) are the primary toxicants released by electronic waste (e-waste) recycling, but their adverse effects on people working in e-waste recycling or living near e-waste sites have not been studied well. In the present study, the serum concentrations of PBDEs, PCBs, and hydroxylated PCBs, the circulating levels of thyroid hormones (THs), and the mRNA levels of seven TH-regulated genes in peripheral blood leukocytes of e-waste recycling workers were analyzed. The associations of the hormone levels and gene expression with the exposure to these contaminants were examined using multiple linear regression models. There were nearly no associations of the TH levels with PCBs and hydroxylated PCBs, whereas elevated hormone (T4 and T3) levels were associated with certain lower-brominated BDEs. While not statistically significant, we did observe a negative association between highly brominated PBDE congeners and thyroid-stimulating hormone (TSH) levels in the e-waste workers. The TH-regulated gene expression was more significantly associated with the organohalogen compounds (OHCs) than the TH levels in these workers. The TH-regulated gene expression was significantly associated with certain PCB and hydroxylated PCB congeners. However, the expression of most target genes was suppressed by PBDEs (mostly highly brominated congeners). This is the first evidence of alterations in TH-regulated gene expression in humans exposed to OHCs. Our findings indicated that OHCs may interfere with TH signaling and/or exert TH-like effects, leading to alterations in related gene expression in humans. Further research is needed to investigate the mechanisms of action and associated biological consequences of the gene expression disruption by OHCs. Cadmium (Cd) is a highly toxic heavy metal and has spread widely in the environment in recent decades. This review summarizes current knowledge about Cd contamination of leafy vegetables, its toxicity, exposure, health risks, and approaches to reducing its toxicity in humans. Leafy vegetable consumption has been identified as a dominant exposure pathway of Cd in the human body. An overview of Cd pollution in leafy vegetables as well as the main sources of Cd is given. Notable estimated daily intakes and health risks of Cd exposure through vegetable consumption for humans are revealed in occupational exposure areas and even in some reference areas. Vegetable consumption is one of the most significant sources of exposure to Cd, particularly in occupational exposure regions. Therefore, numerous approaches have been developed to minimize the accumulation of Cd in leafy vegetables, among which the breeding of Cd pollution-safe cultivars is one of the most effective tools. Furthermore, dietary supplements from leafy vegetables perform positive roles in alleviating Cd toxicity in humans with regard to the effects of essential mineral elements, vitamins and phytochemicals taken into the human body via leafy vegetable consumption.

Cultivar-dependent cadmium (Cd) accumulation was principal in developing Cd-pollution safe cultivars (PSCs). Proteins related to different Cd accumulations of the low-Cd-accumulating (SJ19) and high-Cd-accumulating (CX4) cultivars were investigated by iTRAQ analysis. Higher Cd bioaccumulation factors and translocation factor in CX4 than in SJ19 were consistent with the cultivar-dependent Cd accumulations. The Cd uptake was promoted in CX4 due to its higher expression of Cd-binding proteins and the lower expression of Cd-efflux proteins in roots. What's more, significantly elevated thiol groups (PC2 and PC3) in CX4 under Cd stress might contribute to the high Cd accumulation in roots and the root-to-shoot translocation of Cd-PC complex. Up-regulated proteins involved in cellulose biosynthesis and pectin de-esterification in SJ19 enhanced the Cd sequestration of root cell walls, which was considered as the predominant strategy for reducing Cd accumulation in shoots. The present study provided novel insights in the cultivar-dependent Cd accumulation in shoots of B. parachinensis.

Brassica parachinensis L., a daily consumed leaf vegetable, is a high-Cd accumulator that substantially threatens human health. Screening and breeding Cd pollution-safe cultivars (Cd-PSCs) of crops is a low-cost strategy to restrict human Cd intake from contaminated soils via the food chain. However, little is known about the molecular mechanisms underlying the low-Cd-accumulating traits of B. parachinensis Cd-PSCs. In the current study, we analyzed the transcriptomes of the Cd-treated (5 μM) roots and shoots of a low-Cd-accumulating cultivar (SJ19) and a high-Cd-accumulating cultivar (CX4) of B. parachinensis to reveal the molecular mechanisms in response to Cd stress. Compared to CX4, many pathways involved in carbohydrate and amino acid metabolisms were exclusively up-regulated in SJ19 roots upon exposure to low Cd concentrations, which may produce more energy and metabolites for Cd detoxification. Antioxidant enzymes in the peroxisome were up-regulated in both SJ19 and CX4 roots in response to Cd, while glutathione biosynthesis was only activated in SJ19 roots. In SJ19 shoots, pathways of photosynthesis and cell growth were activated to mitigate Cd-induced damages. Furthermore, Cd transport genes, such as MTP1, HMA3 and CAX family genes, were highly induced by Cd stress in SJ19 roots in accordance with the high Cd concentration in roots, while genes involved in root-to-shoot Cd translocation such as FRD3 and CESA3 were suppressed, which may contribute to the low Cd concertation in edible part of SJ19. Our study provides a genetic basis for further Cd-PSCs screening and breeding.

Hair is a promising, non-invasive, human biomonitoring matrix that can provide insight into retrospective and integral exposure to organic pollutants. In the present study, we measured the concentrations of organophosphate flame retardants (PFRs) in hair and serum samples from university students in Guangzhou, China, and compared the PFR concentrations in the female hair segments using paired distal (5~10cm from the root) and proximal (0~5cm from the root) samples. PFRs were not detected in the serum samples. All PFRs except tricresyl phosphate (TMPP) and tri-n-propyl phosphate (TPP) were detected in more than half of all hair samples. The concentrations of total PFRs varied from 10.1 to 604ng/g, with a median of 148ng/g. Tris(chloroisopropyl) phosphate (TCIPP) and tri(2-ethylexyl) phosphate (TEHP) were the predominant PFRs in hair. A low-shoot-Cd (QLQ) and a high-shoot-Cd cultivar (T308) of water spinach (Ipomoea aquatica Forsk.) were used to investigate molecular mechanism of the genotype difference in cadmium (Cd) accumulation. RNA-Seq under 9 and 72 h cadmium exposures (5 mg L(-1)) were undertaken to explore Cd induced genotype differences in molecular processes. In total, 253 747 540 clean reads were assembled into 57 524 unigenes. Among them, 6136 and 10 064 unigenes were differentially expressed in QLQ and T308, respectively. Cell wall biosynthesis genes, such as GAUT and laccase, and three Cd efflux genes (Nramp5, MATE9, and YSL7) had higher expression levels in QLQ, while the genes in sulfur and glutathione metabolism pathway, e.g., sulfate transporter and cysteine synthase, showed higher expression levels in T308. These findings would be useful for further understanding of the mechanisms related to genotype-dependent Cd accumulation and developing the molecular assisted screening and breeding of low-shoot-Cd cultivars for water spinach.

To reduce cadmium (Cd) pollution of food chains, screening and breeding of low-Cd-accumulating cultivars are the focus of much study. Two previously identified genotypes, a low-Cd-accumulating genotype (LAJK) and a high-Cd-accumulating genotype (HAJS) of pakchoi (Brassica chinesis L.), were stressed by Cd (12.5 μM) for 0 h (T0), 3 h (T3) and 24 h (T24). By comparative transcriptome analysis for root tissue, 3005 and 4343 differentially expressed genes (DEGs) were identified in LAJK at T3 (vs T0) and T24 (vs T3), respectively, whereas 8677 and 5081 DEGs were detected in HAJS. Gene expression pattern analysis suggested a delay of Cd responded transcriptional changes in LAJK compared to HAJS. DEG functional enrichments proposed genotype-specific biological processes coped with Cd stress. Cell wall biosynthesis and glutathione (GSH) metabolism were found to involve in Cd resistance in HAJS, whereas DNA repair and abscisic acid (ABA) signal transduction pathways played important roles in LAJK. Furthermore, the genes participating in Cd efflux such as PDR8 were overexpressed in LAJK, whereas those responsible for Cd transport such as YSL1 were more enhanced in HAJS, exhibiting different Cd transport processes between two genotypes. These novel findings should be useful for molecular assisted screening and breeding of low-Cd-accumulating genotypes for pakchoi.

Hair is increasingly used as a biomarker for human exposure to persistent organic pollutants (POPs). However, the internal and external sources of hair POPs remain a controversial issue. This study analyzed polychlorinated biphenyls (PCBs) in human hair and serum from electronic waste recycling workers. The median concentrations were 894 ng/g and 2868 ng/g lipid in hair and serum, respectively. The PCB concentrations in male and female serum were similar, while concentrations in male hair were significantly lower than in female hair. Significant correlations between the hair and serum PCB levels and congener profiles suggest that air is the predominant PCB source in hair and that hair and blood PCB levels are largely dependent on recent accumulation. The PCB95, 132, and 183 chiral signatures in serum were significantly nonracemic, with mean enantiomer fractions (EFs) of 0.440-0.693. Nevertheless, the hair EFs were essentially racemic (mean EFs = 0.495-0.503). Source apportionment using the Chemical Mass Balance model also indicated primary external PCB sources in human hair from the study area. Air, blood, and indoor dust are responsible for, on average, 64.2%, 27.2%, and 8.79% of the hair PCBs, respectively. This study evidenced that hair is a reliable matrix for monitoring human POP exposure.

To investigate the effect of lignin in the cultivar-dependent Cd detoxification of Brassica chinensis L., Cd and lignin contents, lignin composition and laccase genes expressions in low-Cd-accumulating (LAJK) and high-Cd-accumulating (HAJS) cultivars grown under control (CK) and 25 μM Cd-treatment were determined. The results showed that lignin combined about 14 % of total Cd in both LAJK and HAJS. LAC genes were more up-regulated in HAJS than in LAJK, indicating that the LAC genes were involved in the cultivar-dependent lignin functions. Higher β-aryl ether (A) proportion in the lignin side chain region in LAJK than in HAJS were observed, whereas resinol (B) and phenylcoumaran (C) constitute much higher proportions in HAJS than in LAJK. Chemical calculation to estimate Cd affinity associating with lignin side chain region displayed that i) β-aryl ether (A) exhibited major coupling with lignin aromatic region; ii) resinol (B) and phenylcoumaran (C) displayed major participation in complexation with Cd. We therefore conclude that Cd compartmentalization in the secondary cell wall (SCW) by coupling with lignin side chain region is responsible for Cd detoxification related to cultivar-dependent Cd accumulation of Brassica chinensis. This is the first study on lignin composition in relation to Cd retention mechanisms in SCW.

Two pot experiments were conducted to compare and verify Cd accumulation capacities of different cultivars under Cd exposures (0.215, 0.543, and 0.925 mg kg–1 in Exp-1 and 0.143, 0.619, and 1.407 mg kg–1 in Exp-2) and Cd subcellular distributions between low- and high-Cd cultivars. Shoot Cd concentrations between the selected low- and high-Cd cultivars were 1.4-fold different and the results were reproducible. The proportions of Cd-in-cell-wall of shoots and roots were all higher in a typical low-Cd cultivar (DX102) than in a typical high-Cd cultivar (HJK), while those of Cd-in-chloroplast or Cd-in-trophoplast and Cd-in-membrane-and-organelle were opposite. The proportions of Cd-in-vacuoles-and-cytoplasm of roots in DX102 were always higher than in HJK under Cd stresses, while there was no clear pattern in those of shoots. These findings may help to reduce health risk of Cd from Chinese kale consumption and explained biochemical mechanisms of cultivar-dependent Cd accumulation among the species.

To deal with the Cd contaminant of agricultural soil, pollution-safe cultivar (PSC) is developed to minimize the Cd accumulation risk in crops. The present study aimed to investigate the different proteomic responses related to Cd accumulation in different tissues between two Amaranthus gangeticus cultivars, Pen and Nan. A significantly higher Cd accumulation in Pen than in Nan was unraveled, especially in shoot. The proportions of soluble Cd in root and stem of Nan were significantly lower than those of Pen, implying lower Cd transportation from root to shoot in Nan. Higher contents of NaCl-extracted Cd in Pen than in Nan were probably attributed to the enhancement of GSH related metabolism in Pen, which activated the transportation of Cd from root to shoot. Alteration of other proteins involved in Cd detoxification and energy production also demonstrated that Pen had exhibited a stronger tolerance than Nan in dealing with Cd stress. Thus, differences in the proteomic processes associated with biochemical differences between the two typical cultivars suggested a cultivar-dependent capacity of Cd tolerance and accumulation in amaranth for the first time.

Mechanisms of low cadmium (Cd) accumulations in genotypes of watercress are poorly investigated. In order to improve the understanding, genotype differences in subcellular and biochemical properties of Cd were studied. Two hydroponic experiments were conducted to compare the subcellular distribution and chemical forms of Cd in shoots and roots between low-Cd genotypes (G18 and G20) and high-Cd genotypes (G4 and G5) of watercress under Cd exposures of T1 (8.93 μmol L−1), T2 (44.64 μmol L−1) and T3 (89.29 μmol L−1). High shoot Cd concentration exceeding 100 mg kg−1DW even under T1 treatment indicated a high risk of Cd pollution in watercress. Differences in subcellular distributions and chemical forms between low-Cd and high-Cd genotypes were significant under T1 and T2 treatments. Proportions of Cd in cell wall fraction were the highest in both shoots (48–62 %) and roots (40.1–83.3 %) and were higher in high-Cd group than in low-Cd group under T1 treatment, but reversed under T2 and T3 treatments. It was found that to convert Cd into insoluble phosphate precipitation and pectate- or protein-bound forms may be the primary means for reducing Cd mobility and toxicity in watercress. Genotype differences in Cd subcellular distributions responding to the Cd stresses were more significant than those in Cd chemical forms. The intense Cd accumulation of watercress demonstrated the necessity of adopting and breeding low-Cd cultivars to reduce Cd contamination in the crop. Cd subcellular distribution and chemical forms were related to the genotype-dependent differences in Cd accumulation of watercress.

An industrial one-pot, two-step bioprocess catalyzed by nitrile hydratase (NHase) and amidase was developed for ( S )-2,2-dimethylcyclopropanecarboxamide ( 1 ), the key chiral intermediate of cilastatin. The key unit operations of the whole process including fermentative production of enzymes, biotransformation, isolation of product, and recycling of by-product were reported for the first time. The volumetric enzyme activities of NHase and amidase in 1000-L fermentor were enhanced to 351,000 and 5880 U/L, respectively. The two-step, one-pot biotransformation of rac -2,2-dimethylcyclopropanecarbonitrile ( 2 ) took full advantage of both enzymes, leading to accumulation of ( S )- 1 in 47% yield and 99.6% ee . ( S )- 1 and the by-product ( R )-2,2-dimethylcyclopropanecarboxylic acid ( 3 ) were obtained with yields of 38% and 45%, respectively, by a novel macroporous resin adsorption chromatography. Moreover, the total yield of ( S )- 1 was further increased to 53% after one recycling of ( R )-3. The new bioprocess dramatically improved process efficiency compared with the chemical route by elimination of six synthetic steps and proved to be a superior and more cost-effective approach towards ( S )- 1 .

Background PM2.5 is closely related to the incidence and mortality of respiratory diseases. Diesel particulate matter (DPM) is the main component of particulate air pollution and an important source of PM2.5. Hypothesis/purpose This study mainly explored the effect of DPM on airway surface liquid (ASL) secretion and the regulation of naringin in this process, to evaluate therapeutic potentials of naringin for the treatment of abnormal secretion of the respiratory tract caused by PM2.5. Methods The concentration of lysozyme was measured by Lysozyme Assay Kit. Total protein content was determined by the BCA Protein Assay Kit. The concentration of cAMP and MUC5AC, expressions of CFTR, AQP1, and AQP5 proteins were measured by ELISA. Expressions of CFTR, AQP1 and AQP5 mRNA were determined by qPCR. Amount of CFTR on the cell membrane was determined by immunofluorescence. Results The in vitro and in vivo studies had indicated that DPM could inhibit ASL secretion and increased the viscosity of the liquid. Naringin had the functions to attenuate DPM-induced injury, reduce liquid viscosity by reducing MUC5AC and total protein secretion, increase DPM-induced CFTR, AQP1, and AQP5 mRNA and protein expression, positively regulate apical CFTR insertion and promote CFTR activation by increasing intracellular cAMP. Conclusion These results demonstrated that naringin had regulating effects on the DPM-induced abnormal secretion of the respiratory tract.

Subcellular distributions and chemical forms of cadmium (Cd) in the leaves, stems and roots were investigated in low-Cd accumulation cultivars and high-Cd accumulation cultivars of pakchoi (Brassica chinensis L.). Root cell wall played a key role in limiting soil Cd from entering the protoplast, especially in the low-Cd cultivars. The high-Cd cultivars had significantly higher leaf and stem Cd concentrations than the low-Cd cultivars in cell wall fraction, chloroplast/trophoplast fraction, organelle fraction and soluble fraction. In low-Cd cultivars, which were more sensitive and thus had greater physiological needs of Cd detoxification than high-Cd cultivars, leaf vacuole sequestrated higher proportions of Cd. Cd in the form of pectate/protein complexes (extracted by 1 mol·L−1 NaCl) played a decisive role in Cd translocation from root to shoot, which might be one of the mechanisms that led to the differences in shoot Cd accumulation between the two types of cultivars. Furthermore, the formation of Cdphosphate complexes (extracted by 2% HAc) was also involved in Cd detoxification within the roots of pakchoi under high Cd stress, suggesting that the mechanisms of Cd detoxification might be different between low- and high-Cd cultivars.

Mechanisms of low cadmium (Cd) accumulations in cultivars of water spinach are poorly investigated. We aimed to improve understanding of the subcellular biochemical properties of the mechanisms involved. A pot experiment was conducted to investigate the subcellular distributions of Cd in lateral and main roots, stems, and young and old leaves of a high-Cd (T308) and a low-Cd cultivar (QLQ). The ratio of main root:lateral roots Cd concentration in QLQ was lower (0.34–0.35) than that in T308 (0.39–0.55). The ratio of stem:main root Cd concentration in QLQ was much lower (0.60–0.73) than that in T308 (1.19–1.58). QLQ has higher capacity to sequester Cd in cell wall fractions of main and lateral roots than T308. The difference in shoot Cd concentration between QLQ and T308 is attributable to the difference in Cd translocation from lateral to main roots and from roots to the stem. Fixation of large amounts of Cd in old leaves is beneficial to protect young leaves from Cd toxicity. Cadmium immobilization by the cell wall is important in Cd detoxification, especially in main and lateral roots of QLQ and the shoot of T308.

We analyzed the polychlorinated biphenyls (PCBs) in human hair collected from an electronic waste (e-waste) recycling area in southern China and compared their composition profiles and chiral signatures to those of workplace and domestic dust. The PCB concentrations showed significant age dependence in dismantling workers' hair but not in residents' hair. Among residents, PCB concentrations decreased in the following order: elderly people>students>pre-school children>adults. The PCB homologue and congener profiles of the workers' hair were similar to those of the workplace dust. However, the PCB homologue profile of the residents' hair was clearly different from that of the domestic dust. The chiral congener CB95 generally exhibited a racemic or near-racemic composition in both hair and dust, with enantiomer fractions (EFs) ranging from 0.485 to 0.525 in hair and from 0.479 to 0.504 in dust. The EFs of CB132 in dust (0.477–0.513) were closer to a racemic chiral signature than those in hair (0.378–0.521), but this difference was not significant. Our results suggest that the chiral signature of PCBs may be a better tool than the PCB composition profile for identifying the external and internal sources of organic contaminants in human hair. Further measurements of chiral PCB signatures in hair and blood from the same individuals are needed to identify the external and internal sources of PCBs in human hair. Dechlorane Plus (DP) and its dechlorinated product, anti-Cl11-DP, were measured in serum of 70 occupationally exposed workers in an e-waste recycling region and 13 residents of an urban area in South China. The DP levels were significantly higher in the workers (22–2200 ng/g with median of 150 ng/g lipid) than in the urban residents (2.7–91 ng/g with median of 4.6 ng/g lipid). The DP concentrations in females were found to be associated with their age but such relation was not found for males. Significant differences in DP levels and DP isomer composition were found between genders. The females had remarkably higher DP levels and fanti values (fraction of anti-DP to total DPs) in serum than the males. Anti-Cl11-DP was significantly correlated with anti-DP for both genders but with different slope of regression line. The ratios of anti-Cl11-DP to anti-DP (mean of 0.017) in males were significantly higher than those (mean of 0.010) in females. Combining with the lower fanti values in males, it is likely that males have higher metabolic potential for DPs than females which resulted in the lower DP loading in serum. However, the different patterns of selective uptake and/or excretion of different compounds between genders cannot be eliminated as a possible reason for the observed gender differences. This study is the first to report on the gender difference in DP accumulation in human, and its mechanism is worth further investigation. In plants, microRNAs (miRNAs) play regulatory roles in response to various environmental stresses. In order to illustrate the regulation mechanisms of miRNAs involving the different Cd accumulation abilities between a low-shoot-Cd cultivar (QLQ) and a high-shoot-Cd cultivar (T308) of water spinach (Ipomoea aquatic Forsk.), six sRNA libraries at 3 different time points were constructed. Only 5 miRNAs were exclusively regulated in QLQ, among them, miRNA395 was up-regulated, which was supposed to enhance the Cd retention and detoxification in root. Also, the alterations of miRNA5139, miRNA1511 and miRNA8155 contributed to the attenuation of Cd translocation into the shoot of QLQ. More differentially expressed miRNAs were observed in T308, indicating more complex response was adopted by T308 under Cd stress. miRNA397 exclusively regulated in T308 has enhanced the Cd influx of T308 under Cd treatments. Besides, the Cd translocation of T308 was strengthened due to the up-regulation of MATE efflux family, which was targeted by miRNA3627. Our results unraveled the effects of the cultivar-dependent expression of these specific miRNAs on the different Cd accumulation and translocation abilities of QLQ and T308. These findings provide a new perspective for the molecular assisted breeding of low-Cd cultivars for leaf-vegetables.

Cultivars of water spinach (Ipomoea aquatica Forsk.) differ widely in their shoot cadmium (Cd) concentration. Previously, we suggested that low-Cd cultivars are better able to retain Cd in their roots and thus prevent root-to-shoot Cd translocation. In this study, we explored the roles of roots and shoots in Cd accumulation in a high-Cd (T308) and low-Cd cultivar (QLQ). We used reciprocal grafting to determine the importance of roots and shoots in Cd accumulation, and a dithizone histochemical method to investigate Cd distribution in the roots. The T308 scion with QLQ rootstock accumulated less Cd than the shoot of non-grafted T308. The QLQ scion with T308 rootstock showed a significantly higher Cd concentration than that in the shoot of non-grafted QLQ. Cadmium induced thicker phellem formation in the main roots of QLQ than in those of T308 and only QLQ showed thickening of the outer cortex cell walls in lateral roots. Shoot Cd accumulation was primarily determined by root-to-shoot Cd translocation, not root Cd uptake. The thicker phellem and outer cortex cell walls in QLQ than in T308 may be one reason why QLQ roots were able to retain more Cd, and thus reducing Cd translocation to shoots.

To help reduce risks of heavy metal pollution, two pot experiments were conducted to investigate the variations, transfer potential, and stability of Cadmium (Cd) and Lead (Pb) accumulations in celery (Apium graveolens L.) and to screen for low Cd and Pb accumulative cultivars. The maximum differences in shoot Cd concentration were 4.7-fold under low-Cd exposure and 3.3-fold under high-Cd exposure. These genotype variations in Cd accumulation are sufficiently large to help reduce Cd contamination risk in soil by using the Low-Cd-Accumulative genotypes. Cd accumulation of the Low-Cd-Accumulative genotypes is significantly positive correlated with Pb accumulation. Evidence obtained proves that Cd and Pb accumulations in celery are stable and genotype-dependent at the cultivar level. The presence of high-Pb contamination in soil promoted Cd accumulation in shoots of celery. Celery is considered a species with high risks in Cd pollution and low risks in Pb pollution. Among the tested cultivars, cv. Shuanggangkangbing (SGKB) had the lowest shoot Cd and Pb accumulating abilities, and thus is the most important material for breeding of pollution-safe cultivars (PSCs) to minimize Cd and Pb accumulations in celery.

The chemical form and subcellular and molecular distribution of cadmium (Cd) in the roots and leaves was investigated in two rice genotypes: the low Cd-accumulating ‘‘Guangyuanzhan No. 3’, and the high Cd-accumulating ‘‘Zhenguiai.’’ Elevated Cd level in the medium caused a significant increase in Cd concentration in all parts of the roots and leaves. The soluble fractions in leaves and roots of ‘Zhenguiai’ contained a higher proportion of the Cd than those in ‘Guangyuanzhan No. 3’. Furthermore, ‘Zhenguiai’ had a higher proportion of de-ionized water and 80% ethanol-extractable Cd, which is the most mobile Cd among its various chemical forms. Following gel filtration of the soluble fraction of leaf and root extracts of both genotypes, a compound with a molecular weight of ∼3 kDa, probably comprising phytochelatins (PCs), was the major Cd-binding component. To reduce the potential risks of cadmium (Cd) and lead (Pb) entering the human food chain in vegetables, two pot experiments (Exp. 1 and Exp. 2) were carried out to screen for Cd and Pb pollution-safe cultivars (PSCs) of Chinese flowering cabbage (Brassica parachinensis L.). The three Cd treatments in Exp. 1 (0.114, 0.667, and 1.127 mg kg−1) showed that Chinese flowering cabbage could easily take up Cd from polluted soils, and there were wide variations in Cd accumulation among different cultivars. The Cd accumulation trait at cultivar level was rather stable under different soil Cd treatments. In Exp. 2, seven cultivars that had been shown in Exp. 1 to be typical high or low accumulators of Cd were selected and six Cd + Pb joint exposure treatments were applied to them. Differences in the subcellular distribution and chemical speciation of Cd between two Chinese flowering cabbage (Brassica parachinensis L.) cultivars, Lubao70 (LB70, low-Cd cultivar) and ChixinNO.4 (CX4, high-Cd cultivar) were investigated under various soil Cd and P treatments. Subcellular fractionation of Cd-containing tissues showed that a higher proportion of Cd was bound to the cell wall fraction of LB70 than that of CX4, indicating that Cd compartment functioned better in LB70. Compared to CX4, LB70 had lower proportions of Cd in inorganic form and water-soluble form, but higher proportions of Cd in proteins/pectates integrated form, implying that the low Cd accumulation in LB70 is associated with the low in vivo mobility of Cd. In both cultivars, shoot and root Cd concentration and translocation of Cd from the roots to the shoots obviously decreased with increasing soil P level. It was found that phosphorus (P) played important roles in Cd uptake and translocation via the processes involved in bonding Cd to the cell wall fraction and forming Cd-phosphate complexes. It is suggested that use of low-Cd cultivars in conjunction with P supply is a much useful way to reduce the pollution risk of Cd in the food chain.

Dechlorane Plus (DP) and a dechlorination product, 1,6,7,8,9,14,15,16,17,17,18-octadeca-7,15-diene (anti-Cl(11)-DP), were measured in human hair and indoor dust collected from an e-waste recycling area and two control areas (rural and urban) in South China. DP was detected in hair and dust samples at concentrations ranging from 0.02-58.32 ng/g and 2.78-4197 ng/g, respectively. anti-Cl(11)-DP, mainly detected in human hair and dust samples from the e-waste recycling area, ranged from nd (nondetected) to 0.23 ng/g in hair and from nd to 20.22 ng/g in dust. Average values of anti-DP fractional abundance (f(anti) ratio) in hair of e-waste dismantling workers (0.55 ± 0.11) and dust from e-waste recycling workshops (0.54 ± 0.15) were significantly lower than those in other groups (0.62-0.76 means for hair and 0.66-0.76 means for dust). Significantly positive correlation between DP concentrations in dust and hair and similarity in f(anti) ratios between hair and dust suggest that ingestion of dust comprise one of the major routes for DP exposure. Significantly positive relationships were also observed between anti-Cl(11)-DP and anti-DP for both hair and dust samples with similar regression line slopes. The ratios of anti-Cl(11)-DP to anti-DP between hair and dust show no significant difference. These results suggest that anti-Cl(11)-DP in the human body is likely accumulated from the environmental matrix and not formed from biotransformation of the parent DP.

A maize variety, Huatian-1, had an unusually low translocation rate of cadmium (Cd) (59.6 mg°kg−1 in the roots and 0.093 mg°kg−1 in the grain) compared to 24 other varieties while being grown in soils with 16.50 mg°kg−1 Cd. This indicates that this particular species may have special mechanisms that affect the absorption and translocation pattern of Cd. In this paper, the technique of suppression subtractive hybridization (SSH) was used to isolate and identify Cd-induced genes from Huatian-1 hydroponically exposed to 0.1mM CdCl2 for 1 h, 12 h, 24 h, and 48 h. We found a total of 15 differentially expressed genes in the four groups; 2, 3, 4, and 6 genes were from the groups of 1 h, 12 h, 24 h, and 48 h treatment, respectively. Phospholipase PLDb1 mRNA, adenosine triphosphate (ATP) phosphoribosyl transferase 2, and Sp17 were turned on in the maize in response to Cd stress, and it might provide new clues to explain the mechanism of maize tolerance to Cd.

To evaluate the biomagnification extent of polybrominated diphenyls ethers (PBDEs) and polychlorinated biphenyls (PCBs) in a highly contaminated freshwater food web from South China, trophic magnification factors (TMFs) for 18 PBDE congeners and 53 PCB congeners were calculated. The TMF values ranged 0.26-4.47 for PBDEs and 0.75-5.10 for PCBs. Forty-five of 53 PCBs and BDEs 47, 100 and 154 had TMFs greater than one, suggesting their biomagnification in the present food web. The TMFs for PBDEs were generally smaller than those for PCBs with the same degree of halogenation, indicating a lower biomagnification potential for PBDEs compared to PCBs. For PCBs, it followed a parabolic relationship between TMFs and logK(OW) (octanol-water partition coefficient). However, this relationship was not significant for PBDEs, possibly due to the more complex behaviors of PBDEs in the food web (e.g., metabolism), compared to that of PCBs.

Sesbania rostrata, an annual tropical legume, has been found to be tolerant to heavy metals, with an unknown mechanism. It is a promising candidate species for revegetation at mine tailings. In this study, sequential extractions with five buffers and strong acids were used to extract various chemical forms of cadmium and copper in S. rostrata, with or without Cd or Cu treatments, so that the mechanisms of tolerance and detoxification could be inferred. Both metals had low transition rates from roots to the aboveground of S. rostrata. The transition ratio of Cd (4.00%) was higher than that of Cu (1.46%). The proportion of NaCl extracted Cd (mostly in protein-binding forms) increased drastically in Cd treated plants from being undetectable in untreated plants. This suggests that Cd induced biochemical processes producing protein-like phytochelatins that served as a major mechanism for the high Cd tolerance of S. rostrata. The case for Cu was quite different, indicating that the mechanism for metal tolerance in S. rostrata is metal-specific. The proportion of water-insoluble Cu (e.g. oxalate and phosphate) in roots increased significantly with Cu treatment, which partially explains the tolerance of S. rostrata to Cu. However, how S. rostrata copes with the high biotic activity of inorganic salts of Cu, which increased in all parts of the plant under Cu stress, is a question for future studies. Sesbania rostrata is among the very few N-fixing plants tolerant to heavy metals. This study provides evidence for the detoxification mechanism of metals in Sesbania rostrata.

Persistent halogenated compounds (PHCs), such as dichlorodiphenyltrichloroethane and its metabolites (DDTs), hexachlorocyclohexane isomers (HCHs), polychlorinated biphenyls (PCBs), polybrominated diphenyl ethers (PBDEs), decabromodiphenylethane (DBDPE), and polybrominated biphenyl 153 (PBB 153), were quantified in muscles of five waterbird species collected from an extensive e-waste recycling region in the Pearl River Delta, South China. PCBs, at concentrations up to 1,400,000 ng/g lipid, were the dominant contaminants contributing to 80%-90% of PHCs. PBDEs and organochlorine pesticides (sum of DDTs and HCHs) contributed approximately equally to total PHCs with median concentrations ranging from 37-2200 and 530-4300 ng/g lipid, respectively. This contaminant distribution pattern was different from those acquired by most studies conducted in other regions. The concentrations of PCBs and PBDEs in Chinese-pond heron from the present study were higher than those from most other previous studies with birds having similar trophic levels. The extensive e-waste recycling activities were probably the cause of the elevated PCB and PBDE levels in the bird samples. The median concentrations of PBB 153 and DBDPE ranged from 3-140 and 10-176 ng/g lipid, respectively. The frequent detection and high concentrations of DBDPE in piscivorous birds implicate a potential environmental concern for this "new" brominated flame retardant. Additionally, the interspecies differences in the levels of contaminants and species-specific PBDE congener patterns were also elucidated in the present study.

The association between arbuscular mycorrhizal fungi (AMF) and Rhynchrelyrum repens was investigated. In six abandoned quarries in the Pearl River Delta area, R. repens was found to be associated with nine AMF species, including Glomus versiforme, G. brohultii, G. microaggregatum, G. clarum and G. claroideum, Acaulospora delicata, A. mellea, A. mollowae and Entrophospora infrequens. The genus Glomus and the species G. brohultii were recorded at the highest frequencies. Three typical arbuscular mycorrhizal (AM) structures, i.e. hyphae, vesicles and arbuscules, were found in the roots of the R. repens specimens collected from all the quarries investigated. Vesicles were the most frequently recorded structure. Results of a container-based experiment showed that R. repens had very high mortality (83.3%) in the absence of AMF in soil containing sufficient P (phosphorus); this indicates that R. repens is an obligate mycotroph. The presence of AMF significantly increased the biomass accumulation of R. repens seedlings (p < 0.01). It was also observed that AMF colonization was related to soil P and K (potassium) utilization by R. repens seedlings. It is, therefore, important to inoculate with AMF when using R. repens for the restoration of damaged ecosystems.

Bare rock slopes often deteriorate microclimates and drive water erosion and landslides. Moreover, they impose hidden troubles on humans and have increasingly been brought to the attention of the public. In order to optimize synthetic soils (SS) for improving the outside soil spray seeding (OSSS) technique used for the restoration of rock slopes, we investigated physico-chemical properties and plant growth of a series of SS designed with orthogonal array OA25 (55). Five additive components, cement, inorganic compound fertilizers, peat soil, wood chip and water–absorbent polymers at five levels, were applied to optimize the synthetic soils. In the present study, we only investigated the effects of a single additive component on all the indicators measured. Primarily, results showed that the contributions of cement to plant biomass, pH and water holding content; the contributions of inorganic compound fertilizers to plant biomass and total nitrogen, available nitrogen, total phosphorus and available phosphorus; the contributions of both peat soil and wood chip to plant biomass, water holding content and organic carbon; and the contributions of water absorbent polymers to the plant biomass, water holding content and available nitrogen were all significant. According to the plant growth and physico-chemical properties of the synthetic soils, Treatment-12 consisting of 1.5% cement, 0.4% inorganic compound fertilizers, 1.5% peat soil, 0.3% water absorbent agent and 2.0% wood chip, was the optimum combination among the 25 treatments for the restoration of rock slopes. The properties of the optimized SS to support growth of plants and to resist erosion were verified by an experiment using an actual rock slope.

Crops grown in heavy metal contaminated soils are an important avenue for these toxic pollutants entering the human food chain. Information on how crops respond to soil contaminations of single versus multiple metals is scarce and much needed. This study investigated the accumulation of Cd by 24 cultivars of asparagus bean (Vigna unguiculata subsp. Sesquipedalis L., family Fabaceae) under a low level (0.8 mg kg-1) and a high level (11.8 mg kg-1) of Cd exposure in a garden experiment, and that in a field experiment with Cd, Pb, and Zn (1.2, 486, and 1114 mg kg-1, respectively) contaminated soil. Both experiments showed that there were highly significant variations among the tested cultivars in Cd accumulation by roots, stems, leaves, and fruits of asparagus bean. In the garden experiment, all cultivars under the low Cd exposure and 41.7% of the tested cultivars under the high Cd exposure bore fruits (pods) whose Cd concentrations were lower than 0.05 mg kg-1 fw and therefore were safe for consumption. In addition, the fruit Cd concentrations of cultivars with black seed coats were significantly lower than those with red or spotted seed coats. These results suggest that asparagus bean is a hypo-accumulator to Cd pollutant and the trait of Cd accumulation is genetic-dependent among cultivars. In the field experiment, correlation between fruit Cd and Pb concentrations was significantly positive (p0.05). Additional correlation analyses between two experiments showed that fruit Cd concentrations in the field experiment were significantly correlated with those exposed to the high level of Cd stress, instead of to the low level of Cd stress in the garden experiment. This suggests that the presence of other toxic heavy metals in the soil might have facilitated the accumulation of Cd in fruits, and the selection of pollution-safe-cultivars (PSC) in multi-metal polluted condition could refer to the PSCs selected under a high level exposure of a single heavy metal.

In order to clarify how cadmium (Cd) chemical forms in planta relate to the genotype difference in Cd accumulation of spinach (Spinacia oleracea L.), two low-Cd and two high-Cd cultivars were compared under a hydroponic experiment with two concentrations of Cd (8.98 or 44.71 μmol Cd L(-1)). The concentrations of phosphorus in the hydroponic system were also adjusted to two levels (0.5 and 1.0 mmol L(-1)) to investigate the influence of phosphorus on the forms and accumulation of Cd in the tested cultivars. Average Cd concentrations in shoots were 8.50-10.06 mg kg(-1) for high-Cd cultivars and 6.11-6.64 mg kg(-1) for low-Cd cultivars a under lower Cd treatment and were as high as 24.41-31.35 mg kg(-1) and 19.65-25.76 mg kg(-1), respectively, under a higher treatment. Phosphorus significantly decreased Cd accumulation in the tested cultivars, and the effect had superiority over the cultivar alternation under higher Cd stress. Cadmium in the NaCl-extractable fraction of the plant tissues showed the greatest relationship to genotype difference of Cd accumulation. The difference in the capacity to binding Cd into F HAc, F HCl, or F Residue was another important mechanism involving in the genotype difference in Cd accumulation of spinach. Among them, average proportion of Cd in F HAc in low-Cd cultivars was higher than that in high-Cd cultivars in association with the effect of phosphorus.

Dechlorane Plus (DP) and a dechlorinated product of DP were measured in 34 matched human hair and serum samples (19 males and 15 females) collected from e-waste recycling workers in South China. The DP (sum of syn- and anti-DP) concentrations in hair and serum samples ranged from 6.3 to 1100 ng g(-1) dry weight and from 22 to 1400 ng g(-1) lipid weight (lw). The levels of anti-Cl11-DP ranged from 0.02 to 1.8 ng g(-1) in hair and from not detected to 7.9 ng g(-1) lw in serum. Significant positive correlations for both DP and anti-Cl11-DP concentrations between hair and serum samples were found (p0.05), indicating hair to be a suitable matrix for human DP exposure. However, a significant difference was found in the DP isomer composition between hair and serum, suggesting stereoselective bioaccumulation during the absorption of DP into hair. A sharp gender difference was found in the levels of DP in hair. Moreover, syn-DP, anti-DP and anti-Cl11-DP in hair significantly correlated with those in serum for male samples, but not for female samples. The observed gender differences in the present study may be, in part, ascribed to the much longer hair exposure time for females than males due to the difference in sampling distance from the scalp.

Legumes are ideal for revegetation of metal-mined wastelands which lack nitrogen (N). A greenhouse study was conducted to investigate the feasibility of using Sesbania rostrata and S. cannabina for the reclamation of lead/zinc (Pb/Zn) mine tailings and to evaluate the effects of organic amendment using sewage sludge (0%, 25%, 50%, and 75%, v/v). The results showed that both species could continue to grow on the highly toxic tailings substrata for at least 80 days, although their growth suffered from adverse effects. That S. rostrata with stem and root nodules had better growth (biomass, growth rates, and biomass of nodules) than S. cannabina suggested that S. rostrata is a better choice as a pioneer species for revegetation of the mine tailings. Stem nodules had less obvious adverse effects imposed by tailings than root nodules. Application of sewage sludge increased contents of total carbon (C), N, phosphorus (P), and potassium (K), and reduced total Zn, Pb, Cd, and DTPA-extractable Pb and Cd in tailings substrata. These, in turn, reduced metal (Zn, Pb, and Cd) uptake and accumulation in plant tissues, and improved plant growth performance, including biomass, growth rates, stem nodulation. Concentrations of six heavy metals (Cu, Ni, Zn, Cd, Cr, and Pb) in sediments and fine roots, thick roots, branches, and leaves of six mangrove plant species collected from the Futian mangrove forest, South China were measured. The results show that both the sediments and plants in Futian mangrove ecosystem are moderately contaminated by heavy metals, with the main contaminants being Zn and Cu. All investigated metals showed very similar distribution patterns in the sediments, implying that they had the same anthropogenic source(s). High accumulations of the heavy metals were observed in the root tissues, especially the fine roots, and much lower concentrations in the other organs. This indicates that the roots strongly immobilize the heavy metals and (hence) that mangrove plants possess mechanisms that limit the upward transport of heavy metals and exclude them from sensitive tissues. The growth performance of propagules and 6-month-old seedlings of Bruguiera gymnorhiza in the presence of contaminating Cu and Cd was also examined. The results show that this plant is not sufficiently sensitive to heavy metals after its propagule stage for its regeneration and growth to be significantly affected by heavy metal contamination in the Futian mangrove ecosystem. However, older mangrove seedlings appeared to be more metal-tolerant than the younger seedlings due to their more efficient exclusion mechanism. Thus, the effects of metal contamination on young seedlings should be assessed when evaluating the risks posed by heavy metals in an ecosystem.

(R)-Ethyl-3-hydroxyglutarate, (R)-3, is an intermediate in the synthesis of the statin side chain. Here, a new two-step, indirect biotransformation pathway involving the formation of ethyl (R)-4-carbamoyl-3-hydroxybutanoate, (R)-2, as an intermediate for (R)-3 production was developed using Rhodococcus boritolerans with ethyl (R)-4-cyano-3-hydroxybutyate, (R)-1, as substrate. Maximum conversion was with 10 g (R)-1/l, 7 g cells/l (dry wt), pH 7.5 and 25°C. A yield of 98 ± 0.5% (w/w) was attained within 8 h.
