Upregulated de novo lipogenesis (DNL) plays a pivotal role in the progress of the nonalcoholic fatty liver disease (NAFLD). Cytoplasmic citrate flux, mediated by plasma membrane citrate transporter (SLC13A5), mitochondrial citrate carrier (SLC25A1), and ATP-dependent citrate lyase (ACLY), determines the central carbon source for acetyl-CoA required in DNL. Curcumin, a widely accepted dietary polyphenol, can attenuate lipid accumulation in NAFLD. Here, we first investigated the lipid-lowering effect of curcumin against NAFLD in oleic and palmitic acid (OPA)-induced primary mouse hepatocytes and high-fat plus high-fructose diet (HFHFD)-induced mice. Curcumin profoundly attenuated OPA- or HFHFD-induced hyperlipidemia and aberrant hepatic lipid deposition via modulating the expression and function of SLC13A5 and ACLY. The possible mechanism of curcumin on the citrate pathway was investigated using HepG2 cells, HEK293T cells transfected with human SLC13A5, and recombinant human ACLY. In OPA-stimulated HepG2 cells, curcumin rectified the dysregulated expression of SLC13A5/ACLY possibly via the AMPK-mTOR signaling pathway. Besides, curcumin also functionally inhibited both citrate transport and metabolism mediated by SLC13A5 and ACLY, respectively. These findings confirm that curcumin improves the lipid accumulation in the liver by blocking citrate disposition and hence may be used to prevent NAFLD.

Photothermal therapy is a new type of tumor therapy with great potential. An ideal photothermal therapy agent should have high photothermal conversion effect, low biological toxicity, and degradability. The development of novel photothermal therapy agents with these properties is of great demand. In this study, we synthesized boron quantum dots (BQDs) with an ultrasmall hydrodynamic diameter. Both in vitro and in vivo studies show that the as-synthesized BQDs have good biological safety, high photoacoustic imaging performance, and photothermal conversion ability, which can be used for photoacoustic imaging-guided photothermal agents for tumor treatment. Our investigations confirm that the BQDs hold great promise in tumor theranostic applications.

Background Extracellular electron transfer (EET) is essential in improving the power generation performance of electrochemically active bacteria (EAB) in microbial fuel cells (MFCs). Currently, the EET mechanisms of dissimilatory metal-reducing (DMR) model bacteria Shewanella oneidensis and Geobacter sulfurreducens have been thoroughly studied. Klebsiella has also been proved to be an EAB capable of EET, but the EET mechanism has not been perfected. This study investigated the effects of biofilm transfer and electron mediators transfer on Klebsiella quasipneumoniae sp. 203 electricity generation performance in MFCs. Results Herein, we covered the anode of MFC with a layer of microfiltration membrane to block the effect of the biofilm mechanism, and then explore the EET of the electron mediator mechanism of K. quasipneumoniae sp. 203 and electricity generation performance. In the absence of short-range electron transfer, we found that K. quasipneumoniae sp. 203 can still produce a certain power generation performance, and coated-MFC reached 40.26 mW/m2 at a current density of 770.9 mA/m2, whereas the uncoated-MFC reached 90.69 mW/m2 at a current density of 1224.49 mA/m2. The difference in the electricity generation performance between coated-MFC and uncoated-MFC was probably due to the microfiltration membrane covered in anode, which inhibited the growth of EAB on the anode. Therefore, we speculated that K. quasipneumoniae sp. 203 can also perform EET through the biofilm mechanism. The protein content, the integrity of biofilm and the biofilm activity all proved that the difference in the electricity generation performance between coated-MFC and uncoated-MFC was due to the extremely little biomass of the anode biofilm. To further verify the effect of electron mediators on electricity generation performance of MFCs, 10 µM 2,6-DTBBQ, 2,6-DTBHQ and DHNA were added to coated-MFC and uncoated-MFC. Combining the time–voltage curve and CV curve, we found that 2,6-DTBBQ and 2,6-DTBHQ had high electrocatalytic activity toward the redox reaction of K. quasipneumoniae sp. 203-inoculated MFCs. It was also speculated that K. quasipneumoniae sp. 203 produced 2,6-DTBHQ and 2,6-DTBBQ. Conclusions To the best of our knowledge, the three modes of EET did not exist separately. K. quasipneumoniae sp.203 will adopt the corresponding electron transfer mode or multiple ways to realize EET according to the living environment to improve electricity generation performance.

3D structured flower-like bismuth tungstate/magnetic-graphene nanoplates (Bi2WO6/mag-GNPs) composite was first synthetised for degradation of ciprofloxacin (CIP). The morphological properties of Bi2WO6/mag-GNPs were characterized by SEM, XRD and DRS. The results revealed the synthesized Bi2WO6/mag-GNPs had the enhanced photocatalytic activity because of the synergistic effects of Bi2WO6, graphene and Fe3O4. The photocatalytic degradation rate of Bi2WO6/mag-GNPs toward CIP was almost 6.0 times of mag-GNPs, 2.0 times and 1.7 times of Bi2WO6 and Bi2WO6/GNPs, respectively. Moreover, the Bi2WO6/mag-GNPs particles could be easily separated and recycled. The construction of Bi2WO6/mag-GNPs effectively resolved the separation of photocatalyst and improved the photocatalysis activity of Bi2WO6.

The objective of this study is to assess bioelectricity generation, pollutant removal (COD, ammonium, nitrate) and the bacterial communities on anodes in constructed wetlands coupled with microbial fuel cells (CW-MFCs), through feeding the systems with three different types of synthetic wastewater (system 1: normal wastewater; system 2: ammonium-free wastewater; system 3: nitrate-free wastewater). Three CW-MFCs were operated with different wastewater concentrations and hydraulic retention times (HRTs) over a long time period (6 months). The results indicate that the maximum open circuit voltage (775.63 mV) and maximum power density (0.628 W m−3) were observed in system 3 (period 3), and that bioenergy production was inhibited in system 2, when feeding with ammonium-free wastewater continuously. COD removal rates in the three systems were similar during each period and ranged from 82.2 ± 6.8% to 98.3 ± 2.2%. Ammonium removal occurred at the air cathode of the CW-MFCs through nitrification, and a higher level of ammonium removal was found in system 1 (period 3) compared with the others. Meanwhile, denitrification occurred at the anaerobic anode of the CW-MFCs, and a large amount of nitrate was removed effectively. The highest nitrate removal rate was 98.8 ± 0.5% in system 2 (period 3). Additionally, four genera related to electricity generation were detected at the anode: Geothrix; Desulfovibrio; Desulfobulbus; and Geobacter. The relative abundances of Desulfovibrio, Desulfobulbus and Geothrix gradually increased during the three periods in system 3, which might be beneficial for bioelectricity generation. Further investigations are needed to optimize the CW-MFC performance and explain the mechanism behind the pollutant degradation and electron motion in the CW-MFCs.

This study aimed to investigate the removal performance and mechanisms of dexamethasone (DEX), a representative fluoroglucocorticoid (FGC), from micro-polluted oligotrophic groundwater in a bio-electrochemical system amended with polyaniline-loaded activated carbon (PANI@AC) as three-dimensional particle electrodes (BES-3D). The BES-3D achieved a DEX removal efficiency of 95.7%, which was 39.0% and 14.1% higher than that of a single biological system (SBIO) and two-dimensional bio-electrochemical system (BES-2D), respectively. The preliminary metabolic mechanism of defluorination accounted for 53.5%, 41.1%, and 16.3% in BES-3D, BES-2D, and SBIO, respectively, which was accompanied by demethylation, side-chain fracture, and hydroxyl oxidation for ketone formation and final-ring opening. The main mechanism by which removal was improved in BES-3D was the enrichment of functional microbes and enhancement of the expression of dehalogenation genes. The relative abundance of functional microbes with electron transfer ability and reductive dehalogenating genera, i.e., Pseudomonas, Methylotenera, Desulfuromonas, Sphingomonas, and Microbacterium, in BES-3D was 3.7–6.1 times higher and the copy number of functional genes was 1.9 times higher than those of SBIO, which contributed to the high DEX removal.

The main aim of this research was to study the biodegradation of polycyclic aromatic hydrocarbons (PAHs) and the influence of environmental factors during composting in order to realize the utilization of PAHs contaminated sewage sludge. A production scale compost experiment was conducted with three mixtures of sewage sludge (SS): green forest waste (GFW) at quality ratios of 3:1 (T1), 3:2 (T2) and 3:3 (T3). The residual concentration of PAHs in the three treatments met the permissible limit prescribed by the Agricultural Sludge Pollutant Control Standard (GB 4284-2018). The biodegradation rates of PAHs were 0.0280, 0.0281 and 0.0218 mg/d, and removal efficiencies were 70.7%, 75.2% and 62.4% in T1, T2, T3, respectively, which followed the first-order rate kinetics. The residual concentration of PAHs in T2 was the lowest (1.81 mg/kg), while the germination index (GI) of Tall Fescues (~0.9) was the highest during 50 days of co-composting. The bacterial diversity negatively correlated with the residual PAHs content. The main genus, Bacillus, Pseudomonas and Methylotenera, with cellulose-degrading and PAHs co-metabolizing functions contributed significantly to sludge maturation and PAHs degradation. The dominant microorganisms, Pseudomonas, Sphingobacterium and Chitinophagaceae_uncultured, could support the high removal rate of PAHs in T2. Temperature, pH, total organic carbon (TOC), total nitrogen (TN) and carbon/nitrogen (C/N) ratio had significant positive effect on the compost maturity and quality, and these parameters correlated with the PAHs biodegradation efficiency during composting, especially in T2.

Adenosine monophosphate-activated protein kinase (AMPK) plays a key role in maintaining whole-body homeostasis and has been regarded as a therapeutic target for the treatment of diabetic nephropathy (DN). Herein, a series of 1,2,4-oxadiazole-containing pyrazolo[3,4-b]pyridinone derivatives is reported as AMPKɑ1β1γ1 activators. The in vitro biological assay demonstrated that compounds 12k (EC50 [AMPKα1γ1β1] = 180 nM) and 13q (EC50 [AMPKα1γ1β1] = 2 nM) displayed significant enzyme activation. Mechanism studies indicated that both compounds reduced the levels of reactive oxygen species in a rat kidney fibroblast cell line (NRK-49F) stimulated by transforming growth factor-β and induced early apoptosis of NRK-49F cells at 10 μM. Molecular docking studies suggested that 13q exhibited critical hydrogen-bond interactions with the critical amino acid residues Lys29, Lys31, Asn111, and Asp88 at the binding site of the AMPK protein. These results enrich the structure pool of AMPK activators and provide novel lead compounds for the subsequent development of compounds with a promising therapeutic potential against DN.

Cholestasis has limited therapeutic options and is associated with high morbidity and mortality. The A(1) adenosine receptor (A(1)AR) was postulated to participate in the pathogenesis of hepatic fibrosis induced by experimental extrahepatic cholestasis; however, the contribution of A(1)AR to intrahepatic cholestatic liver injury remains unknown. Here, we found that mice lacking A(1)AR were resistant to alpha-naphthyl isothiocyanate (ANIT)-induced liver injury, as evidenced by lower serum liver enzyme levels and reduced extent of histological necrosis. Bile acid accumulation in liver and serum was markedly diminished in A(1)AR(-/-) mice compared with wild-type (WT) mice. However, biliary and urinary outputs of bile acids were significantly enhanced in A(1)AR(-/-) mice. In the liver, mRNA expression of genes related to bile acid transport (Bsep and Mdr2) and hydroxylation (Cyp3a11) was increased in A(1)AR(-/-) mice. In the kidney, A(1)AR deficiency prevented the decrease of glomerular filtration rate caused by ANIT. Treatment of WT mice with A(1)AR antagonist DPCPX also protected against ANIT hepatotoxicity. Our results indicated that lack of A(1)AR gene protects mice from ANIT-induced cholestasis by enhancing toxic biliary constituents efflux through biliary excretory route and renal elimination system and suggested a potential role of A(1)AR as therapeutic target for the treatment of intrahepatic cholestasis.

A systematic study using density functional theory has been performed for β-Ga2O3 doped with non-metal elements X (X = C, N, F, Si, P, S, Cl, Se, Br, and I) to evaluate the effect of doping on the band edges and photocatalytic activity of β-Ga2O3. The utilization of a more reliable hybrid density functional, as prescribed by Heyd, Scuseria and Ernzerhof, is found to be effective in predicting the band gap of β-Ga2O3 (4.5 eV), in agreement with the experimental result (4.59 eV). Based on the relaxed structures of X-doped systems, the defect formation energies and the plots of density of states have been calculated to analyze the band edges, the band gap states and the preferred doping sites. Our results show that the doping is energetically favored under Ga-rich growth conditions with respect to O-rich growth conditions. It is easier to replace the threefold coordinated O atom with non-metal elements compared to the fourfold coordinated O atom. X-doped systems (X = C, Si, P) show no change in the band gap, with the presence of discrete midgap states, which have adverse effect on the photocatalytic properties. The photocatalytic redox ability can be improved to a certain extent by doping with N, S, Cl, Se, Br, and I. The band alignments for Se-doped and I-doped β-Ga2O3 are well positioned for the feasibility of both photo-oxidation and photo-reduction of water, which are promising photocatalysts for water splitting in the visible region.
