Indonesian black rice cv Cempo Ireng was evaluated for cancer prevention using a T47D cell line model. Methanolic extract of black rice bran (BRB) showed cytotoxicity with an IC 50 of 522.13 µg/ml. This result was in contrast to water extract of BRB, which yielded an IC 50 of more than 6000 µg/ml. From the methanolic extract of BRB, its 6 fractions were found from the preparative TLC performance, with Rf values ranging from 0 to 0.97. According to the MTT assay of all fractions, fraction 3 (F 3 ) had the lowest IC 50 (60.17 ± 1.72µg/ml), while fraction 2 (F 2 ) had an IC 50 of 64.3 ± 0.61µg/ml. However, F 2 showed the highest induction of apoptosis, i.e. about 75.39 ± 0.43% compared to the T47D cells without treatment (control) (about 8.39 ± 0.16%) and with doxorubicin treatment (about 41.30 ± 0.08%). Furthermore, fraction 5 (F 5 ) not only induced apoptosis but also increased the S-phase arrest percentage (60.36 ± 2.07%), which was significantly higher than for the other treatments. Therefore, each fraction of BRB showed different cytotoxic responses to T47D cells.

The grapefruit (Citrus maxima L.) is a plant known by the public as a fruit consumed with various properties. This plant's use is well known, such as antioxidants, enhancing immunity, anti-aging, and antibacterial properties. This study aimed to identify and analyze the chemical compounds contained in C. maxima rind. The extract was obtained by the maceration method using ethanol and ethyl acetate as solvents. The fractionation process was carried out by Column Chromatography. Observation of thin-layer chromatography profiles with UV lamps 254 and 366 nm. Analysis of chemical compound components using GC-MS and data interpretation based on the Wiley 7.0 data library. The interpretation results of the EF1 fraction are β-copaen-4-α-ol; pentadecanoic acid; hexadecanoic acid; tetradecanoic acid; dotriacontane; osthol; 2H-1-benzopyran-2-one, 7-methoxy-8-(3-methyl-2-oxobutyl); furfural; 6-(2,3-Dihydroxy-3-methylbutyl)-7-methoxycoumarin; and 6-(iodomethyl)-5-methyl-4-oxahexanolide. The chemical compounds identified in EAF2 are 1-octadecanol; decane; tetracosane; hexacosane; and 1,2-benzenedicarboxylic acid (2-ethylhexyl) ester. It can be concluded that these compounds have biological and pharmacological activities.

The research aims to analyse the antioxidant, anti-tyrosinase and anti-wrinkle activities from grapefruit (Citrus Maxima L) and strawberry extracts. Samples were extracted by maceration using 96% ethanol and ethyl acetate, subsequently. The Ferric Reducing Antioxidant Power (FRAP) and β-carotene bleaching (BCB) were used to measure the antioxidant activity. The effect of anti-wrinkle was determined by testing the inhibition of elastase and collagenase enzyme, while anti-tyrosinase activity was analysed using mushroom tyrosinase enzyme. The results showed that strawberry extracts in ethanolic (SE) and ethyl acetate (SEA) have antioxidant activity in FRAP (EC50 = 404.39 ± 3.27 µg / mL and 1978.65 ± 37.25 µg/mL) and BCB (IC50 = 292.30 ± 4.69 µg/mL and 671.11 ± 6.74 µg/mL). Whereas the grapefruit peel extracts both in ethanolic (GPE) and ethyl acetate (GPEA) have antioxidant activity in FRAP (EC50 219.47 ± 71.96 µg / ml and 309.44 ± 95.76 µg/ml) and BCB (EC50 245.19 ± 162.47 µg/ml and 567.54 ± 95.31 µg/ml). As positive standards for FRAP antioxidant analysis were quercetin and vitamin C which has IC50 respectively 18.97 ± 4.50 µg/mL and 24.47 ± 1.44 µg/mL. While in BCB analysis, Butylated Hydroxy Toluene (BHT) used as positive standard (IC50 38.68 ± 5.70 µg/mL). The samples of SE, SEA, GPE and GPEA showed tyrosinase inhibitory activity which the IC50 values were respectively 492.68 ± 1.43; 2658 ± 48.08; 3312.5 ± 222.74; 2985.17 ± 122.80 µg/ml. Kojic acid (IC50 111.52 ± 0.42 µg/ml) is used as positive standard in this study. In addition, SE, SEA, GPE and GPEA were able to inhibit elastase and collagenase enzymes, although their activities were still lower than the positive standard used in this study. Elastastinal in concentration 50 µg/mL giving elastase inhibition about 71.71 ± 0.81 µg/mL, while vitamin C in the same concentration showed collagenase inhibition about 66.79 ± 1.23 µg/mL. It can be concluded that the extract of strawberry and grapefruit peel has antioxidant, anti-tyrosinase and anti-wrinkle activity through inhibition of elastase and collagenase enzymes; thus, they can be used as antiaging cosmetic ingredients.

The use of natural products has been widely used as a resource of new bioactive chemical compounds. One of them is the Paku Atai Merah (Angiopteris ferox Copel) tuber which has long been used empirically by the Dayak tribe of East Kalimantan as an anti-cancer. The purpose of this study was to determine the anticancer cytotoxic activity of the extract and fractions of Paku Atai Merah tuber against T47D breast cancer cells in vitro. Extract of Paku Atai Merah tubers was obtained by maceration method using ethanol solvent until obtained the ethanolic extract then fractionated using various solvents to obtain n-hexane, ethyl acetate, and aqueous-ethanol fractions. The cytotoxic effect was carried out based on the MTT assay. Phytochemical screening tests showed positive results for the presence of flavonoid, phenolic, tannin, saponin and steroid compounds. The results of the cytotoxic activity study showed that the ethyl acetate fraction had moderate cytotoxic activity in T47D cancer cells with an IC50 value of 84.8 µg/ml. Ethanol extract (513.06 µg/ml) and n-Hexane frsction (881.97 µg/ ml) were also included in the weak category. This study indicates that ethyl acetate fraction can be developed as a supportive therapy for breast cancer treatment.

This study aims to determine the bioactivity of L.domesticum as cosmetic ingredient. The extraction was prepared by maceration with ethanol 96 % and ethyl acetate to obtain ethanolic extract of fruit flesh (BLE), ethanolic extract of fruit peel (KLE), ethyl acetate extract of fruit flesh (KLE) and fruit peel (KLEA). Then, determination of compounds was conducted by thin layer chromatography (TLC) and analysed of total phenolic and flavonoid content with colorimetric method. The antioxidant capacity was measured using radical scavenging assay of 1,2-diphenyl-2-picrylhydrazyl (DPPH), β-carotene bleaching assay (BCB) and ferric reducing antioxidant power assay (FRAP). The inhibitory effect of skin degradation enzymes (anti-aging) was carried out using elastase and collagenase assay. The tyrosinase inhibitory effect was determined using mushroom tyrosinase. Based on TLC result, extract of L.domesticum containing phenolic, flavonoids, terpenoids and steroids compounds. The largest of total flavonoid content was obtained in KLE (4,76±0,26 %b/b EQ) and phenolic total content in BLEA (6,4±0,15 %b/b EG). Sequence of antioxidant activity measured using DPPH method was BLEA> KLE> BLE> KLEA; while using BCB method and FRAP method were respectively KLEA>BLEA> KLE> BLE; and BLEA> KLE> BLE> KLEA. KLE and KLEA extracts showed the most potent to remaining activity of the enzyme elastase and collagenase. In addition, BLE extract has the higest tyrosinase inhibition than others. Based on these results it can be concluded that the extracts of the fruit flesh and fruit peel of L. domesticum can be used as a cosmetic active ingredient for anti-aging and anti-tyrosinase.

Studies have suggested that both carrot ( Daucus carota L.) and tomato ( Solanum lycopersicum L.) callus extracts contain antoxidant compounds that might have the potental to protect cells from free radicals such as H 2 O 2 that contribute to cell damage. The other sources of free radical exposure in human cells, such as UV‐B, should also be examined. UV‐B exposure can trigger increased expression of inflammatory cytokines such as cyclooxygenase‐2 (COX‐2) and tumor necrosis factor‐α (TNF‐α) and the antinflammatory cytokine interleukin‐10 (IL‐10), which causes photoaging. This study was conducted to investgate the cytoprotectve actvity of carrot and tomato callus aqueous extracts by observing cell viability using the MTT assay. Immunocytochemistry methods were used to examine the effects of carrot and tomato callus aqueous extracts on the expression of COX‐2, TNF‐α, and IL‐10 in human dermal fbroblast adult (HDFa) cells exposed to UV‐B light. Carrot and tomato callus aqueous extracts were obtained by the maceraton method using aqua bidistlled solvent. Results showed that both carrot and tomato callus aqueous extracts at 0.5 mg/mL exhibited the highest cytoprotectve effect in HDFa cells compared to that at other concentratons. Both carrot and tomato callus aqueous extracts could also decrease the expression of COX‐2 and TNF‐α, whereas carrot callus aqueous extract increased the expression of the ant‐inflammatory cytokine IL‐10 in HDFa cells.

Summary Germinated Australian sweet lupin (ASL) flour (after germination for 7 days) was incorporated into muffin formulation at 0%, 2%, 4%, 6% and 8% of dried muffin weight. The effect on the physical characteristics of muffins including height, diameter, colour and texture (hardness, cohesiveness, springiness and chewiness) was measured. The stability of phenolic, phytosterol compounds and the antiradical activity of the muffins before and after baking (190 °C, 25 min) were determined. The level of incorporation of germinated ASL flour affected the volume, colour, hardness and muffin texture. Muffins incorporated with germinated ASL flour at the levels used in this research exhibited higher concentrations of total phenolic compounds, phytosterol and antiradical activity than those in control muffins. The baking process did not significantly reduce the phenolic compounds, phytosterol and antiradical activity.

The chemical contents and health benefits of black rice bran of some rice cultivars have been investigated. However, there has been little research on the ‘Cempo Ireng’ cultivar from Sleman, Yogyakarta. The aim of this present study was to determine the anthocyanin, antioxidant activity, and macro- and micronutrients contents of black rice bran from this local cultivar. The anthocyanin in the black rice bran was extracted using the maceration method with methanol as a solvent. The extract obtained was separated through a preparative thin layer chromatography (TLC) of silica GF254 and a mobile phase composed of n-butanol, acetic acid, and water. Two fractions were collected and analyzed for the anthocyanin content. The preparative TLC spots were separated for further detection and measurement of cyanidin 3-O-glucoside using HPLC followed by LC-MS. The antioxidant activity of the fractions were measured using the DPPH free radical scavenging method. The results showed that the anthocyanin in fraction 1 was identified as cyanidin 3-O-glucoside (66.1 ± 10.6 µg/g). The IC50 of fractions 1 and 2 were 200.96 and 218.36 µg/mL, respectively. Analysis of the macro- and micronutrients revealed that the black rice bran of ‘Cempo Ireng’ had nutrient contents comparable with other rice cultivars. Therefore, this local black rice bran can be used as a source of antioxidants and macro-- and micronutrients.

Anthocyanin of pigmented rice inhibits the growth of cancer cells. The cytotoxicity and apoptosisinducing properties of local black rice (cv Cempo Ireng) extracts and fractions, which contain anthocyanin including cyanidin 3-glucoside and peonidin 3-glucoside, on human cervical cancer cell line (HeLa cells) has been evaluated. The pigmented rice bran was extracted and fractionated using methanol-HCl. The MTT test was performed on HeLa cell cultures to observe the IC 50 value. Preparative TLC was performed to obtain the fractions of black rice bran. Cyanidin 3-glucoside and peonidin 3-glucoside were identified in the pigmented rice bran extract and fractions using UHPLC. Flowcytometry analysis was performed to measure the percentage of apoptotic cells. Our results suggest that the fractions are more toxic than the methanolic crude extract with IC 50 values of 85.95 ± 5.56 μg/mL (the lowest one) and 408.13 ± 51.9 μg/mL, respectively. The concentration of cyanidin 3-glucoside and peonidin 3-glucoside in the methanolic extract were 1.89 and 0.84 μg/mg, respectively. The apoptosis induction by fractions F2 and F4 (52 and 55%) were significantly higher compared to fraction F3 and F5 (30 and 33%) and doxorubicin (21%). Cyanidin 3-glucoside was detected in F4 (0.14 μg/ml) while peonidin 3-glucoside in F2 (0.012 μg/ml), however both were not detected in F3 and F5.
