Through an initial documentary analysis of the flipped classroom in the context of a pandemic and of the new pedagogical models based on active methodologies or pedagogies and cooperative learning, the experience presented below is part of an innovation project based on the design of cooperative activities focused on students to enhance their meaningful learning and with the aim of improving their satisfaction and motivation and thus, as a consequence, their academic performance. For the evaluation of this experience, in which 29 students from the theoretical-experimental subject of General Chemistry participated from the Technological Bases Engineering training plan of the Universidad Católica del Norte, three data collection instruments were administered based on the use of the questionnaire and the interview with the students during and at the end of the experience. The main results show high acceptance and satisfaction with the design of the innovation; an improvement in the development of teamwork competence; and a significant impact of the active methodologies and cooperative learning strategies implemented in increasing the academic performance of students.

The maritime pine (Pinus pinaster Ait.) is a highly valuable Mediterranean conifer. However, recurrent drought events threaten its propagation and conservation. P. pinaster populations exhibit remarkable differences in drought tolerance. To explore these differences, we analyzed stem transcriptional profiles of grafts combining genotypes with contrasting drought responses under well-watered and water-stress regimes. Our analysis underscored that P. pinaster drought tolerance is mainly associated with constitutively expressed genes, which vary based on genotype provenance. However, we identified key genes encoding proteins involved in water stress response, abscisic acid signaling, and growth control including a PHD chromatin regulator, a histone deubiquitinase, the ABI5-binding protein 3, and transcription factors from Myb-related, DOF NAC and LHY families. Additionally, we identified that drought-tolerant rootstock could enhance the drought tolerance of sensitive scions by regulating the accumulation of transcripts involved in carbon mobilization, osmolyte biosynthesis, flavonoid and terpenoid metabolism, and reactive oxygen species scavenging. These included genes encoding galactinol synthase, CBL-interacting serine/threonine protein kinase 5, BEL1-like homeodomain protein, dihydroflavonol 4-reductase, and 1-deoxy-D-xylulose-5-phosphate. Our results revealed several hub genes that could help us to understand the molecular and physiological response to drought of conifers. Based on all the above, grafting with selected drought-tolerant rootstocks is a promising method for propagating elite recalcitrant conifer species, such as P. pinaster.

In this study, a new and simple glassy carbon electrode modified with carbon nanohorns (SWCNH/GCE) was used for the determination of Cr(VI) in aqueous matrices via adsorptive cathodic stripping voltammetry (AdCSV). The modified electrode was characterized via field emission scanning electron microscopy and cyclic voltammetry, which revealed a homogeneous distribution of spherical agglomerates of SWCNH on the electrode surface. The modification increased the electrochemically active area from 0.10 cm2 ± 0.01 (GCE) to 0.16 cm2 ± 0.01 (SWCNH/GCE). The optimized analytical conditions were as follows: a supporting electrolyte (0.15 mol L−1 HCl), an accumulation potential of 0.8 V versus Ag/AgCl, and an accumulation time of 240 s. Validation of the analytical methodology was performed, obtaining a linear range between 20 and 100 µg L−1, a limit of detection of 3.5 µg L−1, and a limit of quantification of 11.6 µg L−1 with good accuracy and precision. The method was applied to the analysis of spiked tap water samples, and the results were compared using a flame atomic absorption spectrophotometer (FAAS) with no significant statistical differences.

The entertainment industry undergoes constant changes, and video games, along with the ways people enjoy them, evolve dynamically. In this context, there arises a need to innovate in areas such as user–game interaction and the design of conventional controllers to keep up with these transformations. This study has found that the design requirements of the external geometry of video game controllers significantly impact users’ purchasing decisions, who often rely on their sensations and emotions. The primary aim of this research has been to demonstrate that it is possible to develop a specific methodology to establish design guidelines that evaluate geometry and ergonomic aspects, considering users’ emotional needs. To achieve this goal, the Kansei methodology has been employed, an engineering discipline that focuses on product design based on emotional interaction and user sensations when using them. Through advanced statistical techniques such as principal component analysis and linear regression, the set of observations has been reduced and a robust predictive model has been developed. Following this methodological approach, surveys and field studies have been conducted, thus differentiating the project from a conventional engineering approach in various stages. Special attention has been paid to the tactile part of the controller, especially its casing. After thorough analysis, the redesigned casing, incorporating improvements derived from statistical analyses, has been compared to existing market models. The results obtained show a significant improvement compared to existing market models, particularly highlighting enhancements in functional characteristics and ergonomic design, enabling comfortable use for individuals with different hand sizes and preventing the grip surface from being slippery. In summary, this work supports the effectiveness of the Kansei methodology as an approach to adapting product design to users’ perceptions and emotional needs.

The measurement of computational thinking ability among computer science (CS) university students is of paramount importance. This study introduces UniCTCheck, a novel method designed to assess the main components of computational thinking in CS students. Utilising two key instruments, namely, the web application CTScore and the psychometric scale CTProg, this research aims to precisely evaluate seven core components of computational thinking and six programming concepts skills essential for CS students. The study, conducted at Rey Juan Carlos University and Atlantic Technological University Galway, involved a diverse sample of students from different year levels and programme specialisations. Through a rigorous research design, including sampling strategies and data collection tools, this study seeks to address critical research questions related to the measurement of variations in students’ computational thinking and programming skills by gender, university level, and location. By shedding light on the significance of computational thinking and programming in the educational realm, this research contributes to the existing literature and underscores the essential role of computational skills in the modern era.

A new curcuminoid molecule (3) has been designed and synthesized, containing a central -(CH2)2-COOH chain at the α carbon of the keto-enol moiety in the structure. The carboxylic acid group is added to react with exposed amino groups on silica oxide nanoparticles (nSiO2), forming an amide bond to attach the curcuminoid moiety to the nSiO2 covalently. The Kaiser test quantifies the functionalization degree, yielding 222 μmol of curcuminoid per gram of nanoparticles. The synthesized hybrid nanosystem, nSiO2-NHCO-CCM, displays significant emission properties, with a maximum emission at 538 nm in dichloromethane, similar to curcuminoid 1 (without the central chain), which emits at 565 nm in the same solvent. Solvent-induced spectral effects on the absorption and emission bands of the new hybrid nanosystem are confirmed, similar to those observed for the free curcuminoid (1). The new nanosystem is evaluated in the presence of kerosene in water, showing an emission band at 525 nm as a detection response. The ability of nSiO2-NHCO-CCM to change its fluorescence when interacting with kerosene in water is notable, as it overcomes the limitation caused by the insolubility of free curcuminoid 1 in water, allowing for the exploitation of its properties when connected to the water-stable nanosystem for future detection studies.

The p.Arg301Gln variant in the α -galactosidase A gene (GLA) has been poorly described in the literature. The few reports show controversial information, with both classical and nonclassical Anderson–Fabry Disease (AFD) presentation patterns. The aim of this study was to analyze the penetrance, clinical phenotype, and biochemical profile of an international cohort of patients carrying the p.Arg301Gln genetic variant in the GLA gene. This was an observational, international, and retrospective cohort case series study of patients carrying the p.Arg301Gln variant in the GLA gene associated with AFD disease. Forty-nine p.Arg301Gln GLA carriers, 41% male, were analyzed. The penetrance was 63% in the entire cohort and 1.5 times higher in men. The mean age of symptoms onset was 41 years; compared to women, men presented symptoms earlier and with a shorter delay to diagnosis. The typical clinical triad—cornea verticillate, neuropathic pain, and angiokeratomas—affected only 20% of the cohort, with no differences between genders. During follow-up, almost 20% of the patients presented some type of nonfatal cardiovascular and renal event (stroke, need for dialysis, heart failure, and arrhythmias requiring intracardiac devices), predominantly affecting men. Residual levels were the most common finding of α-GAL A enzyme activity, only a few women had a normal level; a small proportion of men had undetectable levels. The incidence of combined outcomes including all causes of death was 33%, and the cumulative incidence of all-cause mortality was 9% at the follow-up. Patients carrying the p.Arg301Gln GLA variant have a high penetrance, with predominantly cardiorenal involvement and clinical onset of the disease in middle age. Only a small proportion showed the classic clinical presentation of AFD. As in other X-linked diseases, males were more affected by severe cardiovascular and renal events. This genotype–phenotype correlation could be useful from a practical clinical point of view and for future decision making.

In the context of the current environmental issues and the need to consolidate effective efforts according to the criteria of a circular economy, adsorption of copper from water with functionalized materials has shown high performance at low cost. However, desorption requires high amounts of acid and base to regenerate the functionalized material. In order to overcome this drawback, this work proposes an alternative desorbing phase based on a specific extractant for copper 1,1,1-trifluoro-2,4-pentanodione (TFA) dissolved in the ionic liquid (IL) 1-butyl-3-methylimidazolium bis(trifluoromethylsulfonyl)imide. In this way, adsorption experiments were carried out using adsorbent fly ash, modified with a mesoporous siliceous material, and functionalized with 3-aminopropyl-triethoxysilane (APS). Under the operating conditions of this study, the best desorption performance was achieved using the lowest functionalization percentage of the absorbent material. The desorption percentage reached 45% after 200 min of contact with a stirred solution of TFA 0.2 [M] in IL. This percentage can be increased to around 70% through a second contact with fresh desorbing phase. Meanwhile, the conventional desorption technique with nitric acid showed faster kinetics and a slightly higher desorption percentage close to 50%. In turn, the final pH of the adsorbent slightly decreased when the proposed desorbing phase was used, representing an advantage over the use of acid. Additionally, it was possible to decrease the amount of desorbing phase allowing the concentration of copper. The COSMO-RS calculations indicate that a more hydrophobic IL could increase desorption percentage.

Early stages are under-represented in studies on the molecular and immune features of high-grade serous ovarian carcinoma (HGSOC), and specific studies focused on early-stage HGSOC are required for a better prognostic stratification and to personalize chemotherapy. The aim of this study was to determine the prognostic significance of CD8+ and CD4+ tumor-infiltrating lymphocytes (TILs), tumoral cell PD-L1 expression, BRCA mutational status and tumor mutation burden (TMB) in early-stage HGSOC. A retrospective study was performed on stage I and II HGSOC from the Molecular Reclassification of Early Stages of Ovarian Cancer (RECLAMO) cohort from the Spanish Group of Ovarian Cancer Research (GEICO). Centralized histological typing was performed based on morphological and immunohistochemical features. Intraepithelial (i) and stromal (s) CD8+ and CD4+ T cells and PD-L1 were evaluated on tissue microarrays by immunohistochemistry. BRCA1 and BRCA2 mutation status and TMB were analyzed in tumor DNA using next-generation sequencing. The study included 124 tumors. High iCD8+ (>20 TILs/core), low/intermediate CD4+ (35/core) were associated with favorable outcomes. Tumor cell PD-L1 expression (TPS ≥ 1) was present in only 8% of tumors. In total, 11 (16%) and 6 (9%) out of 69 HGSOC tested carried pathogenic or likely pathogenic BRCA1 or BRCA2 mutations, respectively. Median TMB of 40 tumors analyzed was 5.04 mutations/Mb and only 6 tumors had 10 or more mutations/Mb. BRCA status and TMB were not associated with TILs or prognosis. When compared with studies on advanced HGSOC, our results suggested that prognostic variables differed according to stage and that more studies focused on early stages of HGSOC are needed to better stratify these tumors.

The genetically related assemblages of the intestinal protozoa parasite Giardia lamblia are morphologically indistinguishable and are often derived from specific hosts. The Giardia assemblages are separated by large genetic distances, which might account for their relevant biological and pathogenic differences. In this work, we analyzed the RNAs cargo released into exosomal-like vesicles (ElVs) by the assemblages A and B, which differentially infect humans, and the assemblage E, which infects hoofed animals. The RNA sequencing analysis revealed that the ElVs of each assemblage contained distinct small RNA (sRNA) biotypes, suggesting a preference for specific packaging in each assemblage. These sRNAs were classified into three categories, ribosomal-small RNAs (rsRNAs), messenger-small RNAs (msRNAs), and transfer-small RNAs (tsRNAs), which may play a regulatory role in parasite communication and contribute to host-specificity and pathogenesis. Uptake experiments showed, for the first time, that ElVs were successfully internalized by the parasite trophozoites. Furthermore, we observed that the sRNAs contained inside these ElVs were first located below the plasma membrane but then distributed along the cytoplasm. Overall, the study provides new insights into the molecular mechanisms underlying the host-specificity and pathogenesis of G. lamblia and highlights the potential role of sRNAs in parasite communication and regulation.

Plants when exposed to toxic levels of metals can suffer morphological or physiological damage because toxic metals can interact with several vital molecules in the plant. One possibility to remove these contaminants from the environment is through the phytoremediation technique, since secondary metabolites produced by plants can reverse these damages. To evaluate the cytoprotective activity, the dry mass and possible damage to the membranes of Lactuca sativa (lettuce) seedlings subjected to different concentrations of mercury chloride in association with catechin and quercetin in suballelopathic concentration were determined. The coordination of mercury chloride with substances was also evaluated using vibrational spectroscopy (Raman and FTIR). The interaction of the mentioned flavonoids with mercury chloride was evidenced through vibrational spectroscopy. When the metal was associated with catechin and quercetin, there was an increase in dry mass of almost 3 times when compared with the HgCl2 alone, demonstrating that these flavonoids act as cytoprotective agents. However, in the presence of catechin and quercetin, membrane damage caused by mercury chloride has a level similar to that observed in control plants, demonstrating none statistical difference. Comparing the highest concentration with the lowest concentration of the metal associated with quercetin, it can be seen that the intensity of the peaks in this region decreases when the concentration of the metal increases, indicating an interaction between the metallic compound and the flavonoid. In this context, the use of secondary metabolites can be an alternative in the process of remediation of areas contaminated by mercury chloride, as they mitigate the effects of mercury chloride on lettuce seedlings.

Poly(ethylene glycol) diacrylate (PEGDA) of different molecular weights (Mn = 575 and 700) was used as crosslinking agent for the photopolymerization of 2-hydroxyethyl methacrylate (HEMA) in order to obtain HEMA/PEGDA-based hydrogels. Composites were synthesized in situ employing a new methodology that implies the addition of different quantities of silver nitrate aqueous solution to the monomer mixture with the finality to obtain hydrogels with different silver nanoparticles’ spatial density and distribution. Samples were characterized by thermal, optical, spectroscopic and structural/morphological methods. Thermal studies showed that the increase of PEGDA molecular weight and the AgNO3 concentration in the reaction mixture enhance the glass transition temperature and the thermal stability of the composites. This behavior could be related to the silver coordination with the polymer network. Infrared spectroscopy with Fourier transform and Raman analyses were realized in order to corroborate the sample chemical structure by the identification of specific functional groups. Surface hydrogel morphology was visualized with scanning electron microscopy analysis, detecting a homogeneous micro-porous surface for the samples obtained from high molecular weight PEGDA. Presence of silver nanoparticles was established by X-ray fluorescence spectroscopy and UV/Vis methods. In this last case, the characteristic silver nanoparticle plasmon was observed. Using Transmission Electron Microscopy it was possible to visualize a homogeneous spatial distribution of spherical silver nanoparticles with very narrow diameter distribution that rounds about 14–21 ± 5 nm. In general, the silver nanoparticle presence in the compounds enhances considerably their thermal/morphological characteristics. The underlying causes of Parkinson’s disease are complex, and besides recent advances in elucidating relevant disease mechanisms, no disease-modifying treatments are currently available. One proposed pathophysiological hallmark is mitochondrial dysfunction, and a plethora of evidence points toward the interconnected nature of mitochondria in neuronal homeostasis. This also extends to iron and neuromelanin metabolism, two biochemical processes highly relevant to individual disease manifestation and progression. Modern neuroimaging methods help to gain in vivo insights into these intertwined pathways and may pave the road to individualized medicine in this debilitating disorder. In this narrative review, we will highlight the biological rationale for studying these pathways, how distinct neuroimaging methods can be applied in patients, their respective limitations, and which challenges need to be overcome for successful implementation in clinical studies.

The present study optimised the ultrasound-assisted extraction (UAE) of bioactive compounds from Amaranthus hypochondriacus var. Nutrisol. Influence of temperature (25.86–54.14 °C) and ultrasonic power densities (UPD) (76.01–273.99 mW/mL) on total betalains (BT), betacyanins (BC), betaxanthins (BX), total polyphenols (TP), antioxidant activity (AA), colour parameters (L*, a*, and b*), amaranthine (A), and isoamaranthine (IA) were evaluated using response surface methodology. In this work, the rheological behavior of passion fruit peel extract was determined at different temperatures (5–40 °C) and peel content in the extract (40–55% w/w). The extract was obtained after blanching the passion fruit peels at 95 °C for 5 min, then they were crushed to reduce their size, water was added, and finally, they were subjected to liquefaction and subsequent filtration. Rheological measurements were made using a rheometer with a plate and plate geometry. Extract samples were adequately described by the power-law model exhibiting pseudoplastic behavior, without the presence of thixotropy. The temperature did not influence the flow behavior index, but the consistency coefficient did. The dynamic study (the temperature sweep test) showed that passion fruit peel extract exhibits a more elastic than viscous behavior, typical of a gel.

Debido a que los fenómenos de adsorción de tinturas sobre óxidos metálicos son de gran importancia en la búsqueda de sistemas de conversión de energía solar, en este trabajo se presenta el comportamiento espectroscópico y electroquímico del complejo [Fe(pytpy2)](PF6)2 (donde pytpy es 4'-(piridin-4-il)-2,2':6',2''-terpiridina) en solución fluida y anclado sobre una película delgada de nanocristales de dióxido de titanio. La constante de formación del aducto se estimó utilizando el modelo isotérmico de Langmuir y se encontró un valor de 1,03x105 M-1 para dicho complejo. El anclaje del complejo sobre la superficie del óxido metálico semiconductor se debe a la interacción del residuo de piridina con los sitios ácidos de Lewis presentes en la superficie del TiO2. Se observó que las propiedades espectroscópicas y electroquímicas del complejo no se ven modificadas por la adsorción.

The effects of pH (3.6 and 7.0) and irradiation UV-C dose irradiation (0, 9.81, 15.13, and 31.87 mJ/cm2) on the physicochemical properties and natural microbiota of red prickly pear juice were evaluated during processing and storage. Thermal treatments were used as the control applying high temperatures for a short time (HTST 80 °C/30 s) or ultra-high temperature (UHT 130 °C/3 s). UV-C treatments applied to juices with both pHs inactivated coliforms and mesophiles with the same efficacy as thermal treatments. Yeasts and molds were inactivated at a dose of >15.13 mJ/cm2 at both pHs. The UV-C doses showed no differences in betalains, polyphenols, or antioxidant activity. However, a decrease in these compounds was observed during storage. The lowest reductions in betacyanins (11.1–16.7%) and betaxanthins (2.38–10.22%) were obtained by UV-C treatment at pH 3.6. Thermal treatments (HTST and UHT) caused a reduction greater than UV-C irradiation in betacyanins, betaxanthins, polyphenols, and antioxidant activity after treatment. However, after storage at pH 3.6, the contents of these compounds reached those of the UV-C treatments, except for polyphenols. In specific pigments, betanin retention was highest at pH 3.6 (62.26–87.24%), and its retention decreases with UV-C dose increase and storage. The indicaxanthin retentions were higher (75.85–92.27%) than those of betanin, and the reduction was mainly due to storage. The physical properties (pH, acidity, and °Brix) were not affected by treatments, except for the color. The results suggest that a dose of 15.13 mJ/cm2 of a continuous UV-C system is a non-thermal alternative for the processing of red prickly pear juice at pH 3.6, preserving its properties.

This article shows how to create a robust thermal face recognition system based on the FaceNet architecture. We propose a method for generating thermal images to create a thermal face database with six different attributes (frown, glasses, rotation, normal, vocal, and smile) based on various deep learning models. First, we use StyleCLIP, which oversees manipulating the latent space of the input visible image to add the desired attributes to the visible face. Second, we use the GANs N’ Roses (GNR) model, a multimodal image-to-image framework. It uses maps of style and content to generate thermal imaging from visible images, using generative adversarial approaches. Using the proposed generator system, we create a database of synthetic thermal faces composed of more than 100k images corresponding to 3227 individuals. When trained and tested using the synthetic database, the Thermal-FaceNet model obtained a 99.98% accuracy. Furthermore, when tested with a real database, the accuracy was more than 98%, validating the proposed thermal images generator system.

Whilst avoidance of chemical modifications of DNA bases is essential to maintain genome stability, during evolution eukaryotic cells have evolved a chemically reversible modification of the cytosine base. These dynamic methylation and demethylation reactions on carbon-5 of cytosine regulate several cellular and developmental processes such as embryonic stem cell pluripotency, cell identity, differentiation or tumourgenesis. Whereas these physiological processes are well characterized, very little is known about the toxicity of these cytosine analogues when they incorporate during replication. Here, we report a role of the base excision repair factor XRCC1 in protecting replication fork upon incorporation of 5-hydroxymethyl-2′-deoxycytosine (5hmC) and its deamination product 5-hydroxymethyl-2′-deoxyuridine (5hmU) during DNA synthesis. In the absence of XRCC1, 5hmC exposure leads to increased genomic instability, replication fork impairment and cell lethality. Moreover, the 5hmC deamination product 5hmU recapitulated the genomic instability phenotypes observed by 5hmC exposure, suggesting that 5hmU accounts for the observed by 5hmC exposure. Remarkably, 5hmC-dependent genomic instability and replication fork impairment seen in Xrcc1−/− cells were exacerbated by the trapping of Parp1 on chromatin, indicating that XRCC1 maintains replication fork stability during processing of 5hmC and 5hmU by the base excision repair pathway. Our findings uncover natural epigenetic DNA bases 5hmC and 5hmU as genotoxic nucleosides that threaten replication dynamics and genome integrity in the absence of XRCC1.

Biodiesel is considered an important substitute of diesel oil. Traditionally, it is obtained by the transesterification of vegetable oils with methanol or ethanol, catalyzed by NaOH or KOH. Heterogeneous catalysts have been studied with the aim at facilitating and diminishing costs with purification stages. In the present work, the Na/Nb2O5 system was evaluated on the reaction of soybean oil with ethanol. It was verified, by DRX and IR, that the niobia calcined at 500ºC and impregnated with sodium underwent greater structural alterations than that treated at 300°C. These modifications allowed the generation of basic properties on niobia surface (Hammett and CO2 adsorption /IR). This catalyst showed the highest conversion (30%) among the used materials. The method chosen for evaluating the catalysts yield was the 1H NMR spectroscopy.

This article presents a wireless sensor for pest detection, specifically the Lobesia botrana moth or vineyard moth. The wireless sensor consists of an acoustic-based detection of the sound generated by a flying Lobesia botrana moth. Once a Lobesia botrana moth is detected, the information about the time, geographical location of the sensor and the number of detection events is sent to a server that gathers the detection statistics in real-time. To detect the Lobesia botrana, its acoustic signal was previously characterized in a controlled environment, obtaining its power spectral density for the acoustic filter design. The sensor is tested in a controlled laboratory environment where the detection of the flying moths is successfully achieved in the presence of all types of environmental noises. Finally, the sensor is installed on a vineyard in a region where the moth has already been detected. The device is able to detect flying Lobesia botrana moths during its flying period, giving results that agree with traditional field traps.

In this study, we report a low cost, fast and unexplored electrochemical synthesis strategy of copper oxide nanoneedles films as well as their morphological and chemical characterization. The nanostructured films were prepared using electrochemical anodization in alkaline electrolyte solutions of ethylene glycol, water and fluoride ions. The film morphology shows nanoneedle-shaped structures, with lengths up to 1–2 μm; meanwhile, high-resolution X-ray photoelectron spectroscopy (HRXPS) and spectroscopy Raman analyses indicate that a mixture of Cu(II) and Cu(I) oxides, or only Cu(I) oxide, is obtained as the percentage of water in the electrolyte solution decreases. A preliminary study was also carried out for the photocatalytic degradation of the methylene blue (MB) dye under irradiation with simulated sunlight in the presence of the nanoneedles obtained, presenting a maximum degradation value of 88% of MB and, thus, demonstrating the potential characteristics of the material investigated in the degradation of organic dyes.

This article presents a parametric study using full-wave simulations about the potential use of cold plasma discharges to achieve frequency reconfiguration on a Sievenpiper mushroom metasurface. The study was done by inserting plasma tubes in between the patches of the mushroom structure, in three different positions with respect to the top of the metasurface, and varying the electronic density while keeping the plasma collision frequency. The obtained results show that it is possible to shift the stop-band generated by the metasurface around 25% towards lower frequencies for an electron density value inside the tubes of 1014 cm−3, when they are placed in between the top patches of the metasurface. Additional insertion losses are exhibited when operating near the plasma frequency.

In order to analyze the mechanisms involved in copper accumulation in Ulva compressa, algae were collected at control sites of central and northern Chile, and at two copper-polluted sites of northern Chile. The level of intracellular copper, reduced glutathione (GSH), phytochelatins (PCs), PC2 and PC4, and transcripts encoding metallothioneins (MTs) of U. compressa, UcMT1, UcMT2 and UcMT3, were determined. Algae of control sites contained around 20 μg of copper g−1 of dry tissue (DT) whereas algae of copper-polluted sites contained 260 and 272 μg of copper g−1 of DT. Algae of control sites and copper-polluted sites did not show detectable amounts of GSH, the level of PC2 did not change among sites whereas PC4 was increased in one of the copper-polluted sites. The level of transcripts of UcMT1 and UcMT2 were increased in algae of copper-polluted sites, but the level of UcMT3 did not change. Algae of a control site and a copper-polluted site were visualized by transmission electron microscopy (TEM) and the existence of copper in electrodense particles was analyzed using energy dispersive x-ray spectroscopy (EDXS). Algae of copper-polluted sites showed electrodense nanoparticles containing copper in the chloroplasts, whereas algae of control sites did not. Algae of a control site, Cachagua, were cultivated without copper (control) and with 10 μM copper for 5 days and they were analyzed by TEM-EDXS. Algae cultivated with copper showed copper-containing nanoparticles in the chloroplast whereas control algae did not. Thus, U. compressa from copper-polluted sites exhibits intracellular copper accumulation, an increase in the level of PC4 and expression of UcMTs, and the accumulation of copper-containing particles in chloroplasts.

Metaplastic breast carcinoma (MBC) is a heterogeneous group of infrequent triple negative (TN) invasive carcinomas with poor prognosis. MBCs have a different clinical behavior from other types of triple negative breast cancer (TNBC), being more resistant to standard chemotherapy. MBCs are an example of tumors with activation of epithelial–mesenchymal transition (EMT). The mechanisms involved in EMT could be responsible for the increase in the infiltrative and metastatic capacity of MBCs and resistance to treatments. In addition, a relationship between EMT and the immune response has been seen in these tumors. In this sense, MBC differ from other TN tumors showing a lower number of tumor-infiltrating lymphocytes (TILS) and a higher percentage of tumor cells expressing programmed death-ligand 1 (PD-L1). A better understanding of the relationship between the immune system and EMT could provide new therapeutic approaches in MBC.

Changes in lifestyle in developed countries have triggered the prevalence of obesity and type 2 diabetes mellitus (T2DM) in the latest years. Consequently, these metabolic diseases associated to insulin resistance, and the morbidity associated with them, accounts for enormous costs for the health systems. The best way to face this problem is to identify potential therapeutic targets and/or early biomarkers to help in the treatment and in the early detection. In the insulin receptor signaling cascade, the activities of protein tyrosine kinases and phosphatases are coordinated, thus, protein tyrosine kinases amplify the insulin signaling response, whereas phosphatases are required for the regulation of the rate and duration of that response. The focus of this review is to summarize the impact of transmembrane receptor protein tyrosine phosphatase (RPTPs) in the insulin signaling cascade and secretion, and their implication in metabolic diseases such as obesity and T2DM.

The fabrication of porous-structured polymer films with patterned surface structures has recently attracted increased interest within the material science field. In this work, a series of microstructure scale patterned polymer films were obtained via breath figure methods (BF) with hydrophobic surface films based on self-assembled diblock copolymers by atom-transfer radical polymerization (ATRP). The surface characteristics and morphological properties, pore size, roughness, thickness, and wettability of the block copolymer films was studied in response to variation of the hydrophilic co-monomer structures. A significant improvement of the quality and order of the hydrophobic films was observed in response to thermal annealing and a consequent optimization of the assembly process.

Square wave voltammetry (SWV) analysis was used to assess the antioxidant interactions (synergism, addition, and antagonism) of fruit mixture extracts from grape (G), lemon (L), and blueberry (B) obtained by conventional extraction, ultrasound-assisted extraction, and high hydrostatic pressure extraction. The experimental results showed antagonistic antioxidant effects in all binary mixture extracts (L-G, L-B, and G-B). In DPPH and FRAP assays, the greatest antioxidant capacity was found in the G-B mixture (108.7 and 108.8 μmol TE g−1 dry extract, respectively) obtained by high hydrostatic pressure extraction; however, there were no significant differences when measured by ultrasound-assisted extraction. For TPC and TFC assays, the greatest values were for G-B (6.67 mg GA g−1 dry extract) and L-G (1.63 mg QE g−1 dry extract), respectively. SWV experiments showed antagonistic behavior in the mixtures. Among the different ratios of the fruit mixture extracts evaluated by SWV, 1 : 1 (w/w) combination showed the greatest antagonistic antioxidant effects. SWV suggests the components of the mixture with the highest antioxidant capacity oxidize after mixing. The results indicate that the presence of natural bioactive antioxidants in fruit mixtures does not guarantee that the interactions are synergistic.

Circular Economy (CE) models are increasingly referenced in international fora and on many countries’ climate action agendas. This emphasis is associated with the international environmental and climate crisis. The case of the electronics industry remains particularly relevant, given its background in the use of CE models and its potential to enhance their use. This work focuses on consumer behavior regarding electronics products in Mexico. This is a largely under-researched topic not only in Mexico but also in Latin America. This study demonstrates that, by trying to extend the lifespan of their electronic products, consumers have awareness of product circularity. However, there is a lack of incentives to capitalize on this consumer interest, compounded by the limited participation of manufacturers and distributors. It is concluded that with a well-designed public policy, the electronics industry (including the mobile phone sector) in Mexico can move toward a CE model more rapidly. A stronger initiative for CE by design should be part of these policies, not only in Mexico but also internationally. The role of better eco-labelling in promoting consumers’ environmental awareness is essential. The lessons of this case study might be of interest to other countries as well.

Clostridioides difficile is a Gram-positive, spore-forming bacterium that causes a severe intestinal infection. Spores of this pathogen enter in the human body through the oral route, interact with intestinal epithelial cells and persist in the gut. Once germinated, the vegetative cells colonize the intestine and produce toxins that enhance an immune response that perpetuate the disease. Therefore, spores are major players of the infection and ideal targets for new therapies. In this context, spore surface proteins of C. difficile, are potential antigens for the development of vaccines targeting C. difficile spores. Here, we report that the C-terminal domain of the spore surface protein BclA3, BclA3CTD, was identified as an antigenic epitope, over-produced in Escherichia coli and tested as an immunogen in mice. To increase antigen stability and efficiency, BclA3CTD was also exposed on the surface of B. subtilis spores, a mucosal vaccine delivery system. In the experimental conditions used in this study, free BclA3CTD induced antibody production in mice and attenuated some C. difficile infection symptoms after a challenge with the pathogen, while the spore-displayed antigen resulted less effective. Although dose regimen and immunization routes need to be optimized, our results suggest BclA3CTD as a potentially effective antigen to develop a new vaccination strategy targeting C. difficile spores.

(1) Background: Colorectal cancer (CRC) is among the best examples of the relationship between inflammation and increased cancer risk. (2) Methods: To examine the effects of spontaneous low-grade chronic inflammation on the pathogenesis of CRC, we developed a new murine model of colitis-associated cancer (CAC) by crossing Mucin 2 mutated mice (Winnie) with ApcMin/+ mice. (3) Results: The resulting Winnie-ApcMin/+ model combines an inflammatory background with a genetic predisposition to small intestinal polyposis. Winnie-ApcMin/+ mice show an early occurrence of inflammatory signs and dysplastic lesions in the distal colon with a specific molecular signature. (4) Conclusion: The Winnie-ApcMin/+ model is a perfect model to demonstrate that chronic inflammation represents a crucial risk factor for the onset and progression of tumoral lesions in individuals genetically predisposed to CRC.

Clostridioides difficile, formerly known as Clostridium difficile, is a spore-forming bacterium considered as the most common cause of nosocomial infections in developed countries. The spore of C. difficile is involved in the transmission of the pathogen and in its first interaction with the host; therefore, a therapeutic approach able to control C. difficile spores would improve the clearance of the infection. The C-terminal (CTD) end of BclA2, a spore surface protein of C. difficile responsible of the interaction with the host intestinal cells, was selected as a putative mucosal antigen. The BclA2 fragment, BclA2CTD, was purified and used to nasally immunize mice both as a free protein and after adsorption to the spore of Bacillus subtilis, a well-established mucosal delivery vehicle. While the adsorption to spores increased the in vitro stability of BclA2CTD, in vivo both free and spore-adsorbed BclA2CTD were able to induce a similar, specific humoral immune response in a murine model. Although in the experimental conditions utilized the immune response was not protective, the induction of specific IgG indicates that free or spore-bound BclA2CTD could act as a putative mucosal antigen targeting C. difficile spores.

β-Cyclodextrin (βCD), the less water soluble of the cyclodextrins, has been used as a capping agent in the preparation of semiconductor nanocrystals or quantum dots (QDs). Nevertheless, no reports have been found in the use of the highly water-soluble polymer of this, prepared by the crosslinking of the βCD units with epichlorohydrin in basic medium (βCDP). This polymer, besides to overcome the low solubility of the βCD, increases the inclusion constant of the guest; two parameters that deserve its use as capping agent, instead of the native cyclodextrin. In the present manuscript, we afforded the in-situ aqueous preparation of cadmium telluride (CdTe) QDs capped with βCDP. The polymer influence on the photoluminescent properties of the nanocrystals was analyzed. The βCDP controls the nanocrystals growth during the Oswald ripening stage. Consequently, the CdTe capped βCDP QDs showed lower Stokes-shift values, higher photoluminescent efficiency, and narrower size distribution than for nanocrystals obtained in the absence of polymer. Transmission electron microscopy (TEM) micrographs and energy dispersive X-ray spectroscopy (EDS) analysis revealed the composition and crystallinity of the CdTe QDs. This βCDP capped CdTe QDs is a potential scaffold for the supramolecular modification of QDs surface.
